Two plant growth-promoting bacteria, Bacillus subtilis and Pseudomonas corrugata, immobilized in a sodium alginate based formulation were evaluated for their survival, viability and plant growth-promoting ability after 3 years of storage at 4 °C. Populations of both of the bacterial isolates recovered from the immobilized sodium alginate beads were in the order of 108 cfu g−1. The plant-based bioassay indicated that the plant growth promotion ability of both of the bacterial isolates was equal to those of fresh broth-based formulations. The bacterial isolates retained the root colonization, and antifungal and enzyme activities in the alginate-based formulation during storage.  相似文献

The influence of inoculation with phosphate-solubilizing bacteria (Enterobacter agglomerans) on maize growth, P uptake and root exudation was studied. Plants were grown in an axenic culture device where P was supplied as soluble phosphate at different contents (0, 5, 15 or 25 ppm) in the nutrient solution and as insoluble rock phosphate added to the culture sand. Experimental device was successfully used to obtain axenic root systems or good establishment of the inoculated strain in the rhizosphere of maize (109 bact. g−1 dry rhizospheric material). Plant growth was promoted by inoculation only for 5 or 15 ppm of soluble P in the nutrient medium without any significant effect on P uptake by the plant, suggesting that the quantities of P released by bacterial rhizospheric activity were very small. Amounts of organic compounds (total C and water-soluble C) exuded were relatively low (3.0 to 6.4% of the total plant biomass) and were reduced by bacterial inoculation when plant growth was largely promoted. Carbon balance modification and plant growth hormone production by the inoculated strain were suspected and discussed.  相似文献

Thirty-day-old plants of mustard (Brassica juncea L.) were sprayed with 10−10, 10−8, or 10−6 M aqueous solution of 28-homobrassinolide (HBR). The HBR-treated plants were healthier than those treated with water and yielded more. Maximum increase over control was found in 60-d-old, 10−8 M-HBR-treated plants in fresh and dry mass per plant, carbonic anhydrase (CA, E.C. 4.2.1.1) activity, and net photosynthetic rate (P N), at harvest in number of pods per plant and seed yield per plant (the respective values were 25, 30, 34, 69, 24, and 29 %). A further increase in the concentration of HBR (10−6 M) did not make any additional impact on the growth and yield. Increased CA activity and P N were correlated with growth and seed yield. Summary The content of endogenous gibberellin (GA)-like substances of roots and root nodules of SOya, and GA production byRhizobium japonicum cultures, were investigated by a combined thin layer chromatographic (TLC)-dwarf pea epicotyl bioassay technique. GAs were more concentrated in root nodules than in the roots, totalling 1.34 and 0.16 nM GA3 equivalents g−1 dry wt. respectively. GA production byR. japonicum cultures was demonstrated (1.00 nM GA3 equivalentsl −1) and comparison of the GA components of plant and bacterial culture medium extracts, suggested that rhizobial GA production may contribute to the nodule GA content. Cis-trans abscisic acid (ABA) was identified in root and nodule extracts by TLC-gas liquid chromatography (GLC), and amounted to 0.18 and 2.21 nM g−1 dry wt. respectively, whereas 0.30 and 4.63 nM ABA equivalents g−1 dry wt. were detected by a TLC-wheat embryo bioassay technique. Sphingomonas spp possess unique abilities to degrade refractory contaminants and are found ubiquitously in the environment. We developed Sphingomonas genus-specific PCR primers (SPf-190 and SPr1-852) which showed specific amplification of a 627-bp 16S rDNA fragment from Sphingomonas spp. A PCR assay using these Sphingomonas specific primers was developed to detect Sphingomonas aromaticivorans B0695R in three texturally distinct soil types, showing detection limits between 1.3–2.2 × 103 CFU g−1 dry soil. A sphingolipid extraction protocol was also developed to monitor Sphingomonas populations in soil quantitatively. The detection limit of the assay was 20 pmol g−1 dry soil, equivalent to about 3 × 105 cells g−1 dry soil. Survival of S. aromaticivorans B0695R was monitored in the three different soils by antibiotic selective plate counting, PCR and sphingolipid analysis. All three approaches showed that the B0695R cells persisted in the low biomass Sequatchie sub-soil at about 3–5 × 107cells g−1 dry soil. In comparison to the plate counting assay, both the PCR and sphingolipid analysis detected a significantly higher level of B0695R cells in the clay soil and Sequatchie top-soil, indicating the possibility of the presence of viable but non-culturable B0695R cells in the soils. The combination of PCR and sphingolipid analysis may provide a more realistic estimation of Sphingomonas population in the environment. Hairy roots of Rhamnus fallax Boiss. were induced using Agrobacterium rhizogenes strain A4M70GUS. The culture established on Woody plant media (WPM) showed a typical hairy root phenotype: rapid growth, reduced apical dominance and root plagiotropism. Seven clones of R. fallax were selected on the basis of their differences in colour and the root branching. The growth of hairy root culture, measured through gain in fresh mass, was done under 16-h photoperiod or in the dark. An increase in anthraquinone (AQ) content was obtained in clones with yellow and less branched roots, like clone 1 [16.43 mg g−1(d.m.)] and clone 7 [14.21 mg g−1(d.m.)], compared with other analysed transformed and non-transformed tissue. This study presents the first report of successful transformation of any species from family Rhamnaceae by A. rhizogenes and analysis of AQ production in transformed tissue.  相似文献

A genetic transformation method using Agrobacterium rhizogenes was developed for Harpagophytum procumbens. The influence of three factors on hairy root formation was tested: bacterial strains (A4 and ATCC 15834), various types of explants and acetosyringone (AS) (200 μM). The highest frequency of transformation (over 50% of explants forming roots at the infected sites after 6 weeks of culture on Lloyd and McCown (WP) medium) was achieved using a combination of nodal stem explants and A. rhizogenes strain A4. The addition of 200 μM AS to root induction medium was found to enhance hairy root induction but its effect varied depending on bacterial strain and explant type. Three of the most vigorously growing hairy root clones of H. procumbens were chosen and analyzed for accumulation of iridoid and phenylethanoid glycosides. The transgenic nature of these root clones was confirmed by PCR amplification; they were positive for rolB and rolC genes. Harpagoside, verbascoside and isoverbascoside were identified by HPLC and LC–ESI-MS as the major compounds from all analyzed hairy root clones. The Hp-3 root clone showed the higher harpagoside content (0.32 mg g−1 dry wt.) compared with other analyzed transformed and non-tuberized untransformed roots of H. procumbens. However, the level of the compound in the hairy root clone was lower than that detected in a sample of commercially available root tubers of H. procumbens. The Hp-3 root clone also produced high amounts of verbascoside and isoverbascoside (8.12 mg g−1 dry wt. and 9.97 mg g−1 dry wt., respectively) comparable to those found in root tubers.  相似文献

Enhanced nitrogen (N) levels accelerate expansion of Calamagrostis epigejos and Arrhenatherum elatius, highly aggressive expanders displacing original dry acidophilous grassland vegetation in the Podyjí National Park (Czech Republic). We compared the capability of Calamagrostis and Arrhenatherum under control and N enhanced treatments to (i) accumulate N and phosphorus (P) in plant tissues, (ii) remove N and P from above-ground biomass during senescence and (iii) release N and P from plant material during decomposition of fresh formed litter. In control treatment, significantly higher amounts of total biomass and fresh aboveground litter were observed in Calamagrostis than in Arrhenatherum. Contrariwise, nutrient concentrations were significantly higher (11.6–14.3 mg N g−1 and 2.3 mg P g−1) in Arrhenatherum peak aboveground biomass than in Calamagrostis (8.4–10.3 mg N g−1 and 1.6–1.7 mg P g−1). Substantial differences between species were found in resorption of nutrients, mainly P, at the ends of growing seasons. While P concentrations in Arrhenatherum fresh litter were twice and three times higher (1.6–2.5 mg P g−1) than in Calamagrostis (0.7–0.8 mg P g−1), N concentrations were nearly doubled in Arrhenatherum (13.1–15.6 mg N g−1) in comparison with Calamagrostis (7.4–8.7 mg N g−1). Thus, the nutrients (N and mainly P) were retranslocated from the aboveground biomass of Calamagrostis probably more effectively in comparison with Arrhenatherum at the end of the growing season. On the other hand, Arrhenatherum litter was decomposed faster and consequently nutrient release (mainly N and P) was higher in comparison with Calamagrostis which pointed to different growth and nutrient use strategies of studied grass species.  相似文献

The effects of three selected agrochemicals on bacterial diversity in cultivated soil have been studied. The selected agrochemicals are Cerox (an insecticide), Ceresate and Paraquat (both herbicides). The effect on bacterial population was studied by looking at the total heterotrophic bacteria presence and the effect of the agrochemicals on some selected soil microbes. The soil type used was loamy with pH of 6.0–7.0. The soil was placed in opaque pots and bambara bean (Vigna subterranean) seeds cultivated in them. The agrochemicals were applied two weeks after germination of seeds at concentrations based on manufacturer’s recommendation. Plant growth was assessed by weekly measurement of plant height, foliage appearance and number of nodules formed after one month. The results indicated that the diversity index (Di) among the bacteria populations in untreated soil and that of Cerox-treated soils were high with mean diversity index above 0.95. Mean Di for Ceresate-treated soil was 0.88, and that for Paraquattreated soil was 0.85 indicating low bacterial populations in these treatment-type soils. The study also showed that application of the agrochemicals caused reduction in the number of total heterotrophic bacteria population sizes in the soil. Ceresate caused 82.50% reduction in bacteria number from a mean of 40 × 105 cfu g−1 of soil sample to 70 × 104 cfu g−1. Paraquat-treated soil showed 92.86% reduction, from a mean of 56 × 105 cfu g−1 to 40 × 104 cfu g−1. Application of Cerox to the soil did not have any remarkable reduction in bacterial population number. Total viable cell count studies using Congo red yeast-extract mannitol agar indicated reduction in the number of Rhizobium spp. after application of the agrochemicals. Mean number of Rhizobium population numbers per gram of soil was 180 × 104 for the untreated soil. Cerox-treated soil recorded mean number of 138 × 104 rhizobial cfu g−1 of soil, a 23.33% reduction. Ceresate- and Paraquat-treated soils recorded 20 × 104 and 12 × 104 cfu g−1 of soil, respectively, representing 88.89% and 93.33% reduction in Rhizobium population numbers. Correspondingly, the mean number of nodules per plant was 44 for the growth in untreated soil, 30 for the plant in the Cerox-treated soil, 8 for the plant in Paraquat-treated soil and 3 for the plant in Ceresate-treated soil. The study has confirmed detrimental effect of insecticide on bacterial populations in the soil. Total heterotrophic counts, rhizobial counts as well as the number of nodules of all samples taken from the chemically treated soils were all low as compared to values obtained for the untreated soil. However, the effect of the insecticide was minimal in all cases as compared to the effects of the herbicides on the soil fauna. Indiscriminate use of agrochemicals on farms can therefore affect soil flora and subsequently food production.  相似文献

Murashige and Skoog’s (MS) medium was supplemented with supernatant of Halomonas desiderata RE1 in different combinations to observe the impact of bacterial auxin on in vitro growth of Brassica oleracea L. Three groups of combinations MS + BS (Bacterial supernatant), MS + BS + 10% CW (coconut water) and MS + BS + 4 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) were considered. Different amounts of BS used in each combination were 50, 100, 150 and 200 μl in 5 ml MS medium. Media combinations inoculated with seeds, internodal explants and callus of B. oleracea L. were incubated in a growth chamber at 25 ± 1°C and exposed to 16-h cool fluorescent light. Seeds inoculated on MS + BS and MS + BS + 10% CW, shoot elongation was observed over control whereas this response was suppressed in 2,4-D-containing media. In explants inoculated on MS + BS, MS + BS + 10% CW and MS + BS + 4 mg l−1 2,4-D different responses such as callus induction, adventitious shoot induction and hypertrophy were observed at different supernatant treatments. In callus inoculation, callus proliferation was observed in most of the treatments at different media combinations.  相似文献

Nitrogen-fixing enterobacteria (Enterobacter agglomerans and Citrobacter freundii) were commonly found associated with the microflora of stained Sassafras (Atherosperma moschatum Labill.) in forests of SE Tasmania. However, their populations never exceeded 3× 104 cells g−1 dry wood and comprised at most 3% of the total bacterial flora. Bacterial colonization of the wood appeared to coincide with that by non-hymenocetous fungi: bacteria were never isolated in significant numbers from wood not infected with fungi. The contribution of the N2-fixing flora to the N economy of the habitat is considered to be negligible.  相似文献

Trivalent chromium (Cr3+) is essential for animal and human health, whereas hexavalent Cr (CrO4 2−) is a potent carcinogen and extremely toxic to animals and humans. Thus, the accumulated Cr in food plants may represent potential health hazards to animals and humans if the element is accumulated in the hexavalent form or in high concentrations. This study was conducted to determine the extent to which various vegetable crops absorb and accumulate Cr3+ and CrO4 2− into roots and shoots and to ascertain the different chemical forms of Cr in these tissues. Two greenhouse hydroponic experiments were performed using a recirculating-nutrient culture technique that allowed all plants to be equally supplied with Cr at all times. In the first experiment, 1 mg L−1 Cr was supplied to 11 vegetable plant species as Cr3+ or CrO4 2−, and the accumulation of Cr in roots and shoots was compared. The crops tested included cabbage (Brassica oleracea L. var. capitata L.), cauliflower (Brassica oleracea L. var. botrytis L.), celery (Apium graveolens L. var. dulce (Mill.) Pers.), chive (Allium schoenoprasum L.), collard (Brassica oleracea L. var. acephala DC.), garden pea (Pisum sativum L.), kale (Brassica oleracea L. var. acephala DC.), lettuce (Lactuca sativa L.), onion (Allium cepa L.), spinach (Spinacia oleracea L.), and strawberry (Fragaria ×  ananassaDuch.). In the second experiment, X-ray absorption spectroscopy (XAS) analysis on Cr in plant tissues was performed in roots and shoots of various vegetable plants treated with CrO4 2− at either 2 mg Cr L−1 for 7 d or 10 mg Cr L−1 for 2, 4 or 7 d. The crops used in this experiment included beet (Beta vulgaris L. var. crassa (Alef.) J. Helm), broccoli (Brassica oleracea L. var. Italica Plenck), cantaloupe (Cucumis melo L. gp. Cantalupensis), cucumber (Cucumis sativus L.), lettuce, radish (Raphanus sativus L.), spinach, tomato (Lycopersicon lycopersicum (L.) Karsten), and turnip (Brassica rapa L. var. rapifera Bailey). The XAS speciation analysis indicates that CrO4 2− is converted in the root to Cr3+ by all plants tested. Translocation of both Cr forms from roots to shoots was extremely limited and accumulation of Cr by roots was 100-fold higher than that by shoots, regardless of the Cr species supplied. Highest Cr concentrations were detected in members of the Brassicaceae family such as cauliflower, kale, and cabbage. Based on our observations and previous findings by other researchers, a hypothesis for the differential accumulation and identical translocation patterns of the two Cr ions is proposed. The current study was undertaken to enumerate Gram-positive bacteria in fresh sub-tropical marine fish and determine the effect of ambient storage (25°C) on the Gram-positive bacterial count. Total and Gram-positive bacteria were enumerated in the muscles, gills and gut of fresh and stored Pseudocaranx dentex, Pagrus auratus and Mugil cephalus on tryptone soya agar (TSA) and TSA with 0.25% phenylethyl alcohol (PEA), respectively. Initial studies indicated that PEA significantly reduced total aerobic bacterial count (TABC) whereas control Gram-positive bacteria were not affected by 0.25% PEA. TABC significantly increased in all fish body parts, whereas Gram-positive aerobic bacterial count (GABC) significantly increased only in the muscles and gills during ambient storage for 15 h. The TABC of the fish species increased from 4.00, 6.13 and 4.58 log cfu g−1, respectively in the muscles, gills, and gut to 6.31, 7.31 and 7.23 log cfu g−1 by the end of storage. GABC increased from 2.00, 3.52 and 2.20 log cfu g−1 to 4.70, 5.85 and 3.36 log cfu g−1. Within each species, TABC were significantly higher in the gills compared to that of muscles and gut; however, no significant differences were found in GABC between muscles and gills. This study demonstrated the potential importance of Gram-positive bacteria in sub-tropical marine fish and their spoilage.  相似文献

Eriophorum vaginatum L. subsp.spissum (Fern.) Hult., a dominant plant in arctic tundra ecosystems, has acid phosphatase activity evenly distributed along its root surface from the root tip to a distance at least 16 cm from the tip. These root surface phosphatases have optimal activity from pH 3.5 to 4.0; mean soil pH for soil samples collected with roots was 3.69. Apparent energy of activation and Q10 values (14.0 kcal mol−1 and 2.2, respectively) do not provide evidence for temperature acclimation, but substantial phosphatase activity was measured at 1°C. Kinetic parameters determined for this root surface phosphatase were as follows: Km=9.23 mM, Vmax=1.61×10−3 μmoles mm−2h−1. The presence of inorganic phosphorus in the assay medium did not inhibit root surface phosphatase activity except at very high concentrations (100 mM); even then, only slight inhibition was detected (7 to 19%). A comparison of hydrolysis rates with inorganic phosphate assimilation rates measured forE. vaginatum indicates that organic phosphate hydrolysis may occur at approximately one third the rate of inorganic phosphate absorption. Calculations show that inorganic phosphate produced by root surface phosphatase activity may satisfy 65% of the annual phosphate demand ofE. vaginatum. Since arctic tundra soils are typically higher in dissolved organic phosphorus compounds than in inorganic phosphate, root surface phosphatase activity may make a considerable contribution to the phosphate nutrition of this widespread and abundant arctic plant.  相似文献

A Method to Estimate Practical Radial Oxygen Loss of Wetland Plant Roots   总被引:1,自引:0,他引:1  
The estimation of practical radial oxygen loss (ROL) of wetland plant roots was attempted in this study. We have devised a new method to measure ROL of wetland plant roots. The whole root system was bathed in an anoxic nutrient solution. Oxygen released from the root was removed immediately by introducing oxygen-free nitrogen gas (O2 < 4 nmol L−1) to mimic natural habitats where released oxygen is consumed rapidly due to chemical and biological oxidation processes. Oxygen removed from the root-bathing chamber was simultaneously detected colorimetrically by use of the highly oxygen-sensitive anthraquinone radical anion (AQ·) in a cell outside the root-bathing chamber, which decolorized by a rapid reaction with oxygen. An emergent macrophyte Typha latifolia L. was incubated, and its ROL was measured by both the new method and one of the conventional methods, the closed chamber/electrode method, by which the ROL of Typha latifolia L. had not yet been measured. The new method succeeded in detecting the ROL, whereas the conventional method was not able to detect oxygen, due to the level being below the detection limit of the oxygen electrode. The oxygen supply via the seedlings of Typha latifolia L. was ca. 10 times higher compared with control measurements without plant. Light illumination significantly enhanced the ROL of Typha latifolia L. (0.33 nmol O2 g−1 root dry weight s−1 under light and 0.18 nmol O2 g−1 root dry weight s−1 in the dark). Theses values fall between those previously reported by the closed chamber/titanium citrate method and the open chamber/electrode method.  相似文献

One rape (Brassica napus cv. Wesroona) plant and four cotton (Gossypium hirsutum cv. Sicot 3) plants were grown in plastic cells containing soil labelled with 407 kBq of33P g−1 soil. After 5–8 days of growth, the33P depletion zones of all plants were autoradiographed and33P uptake by plants was measured. The autoradiographs were scanned with a microdensitometer and the optical densities at several places within the33P depletion zones of roots were obtained. The volume of soil explored by root hairs was estimated from measurements of root diameters and lengths of roots and root hairs. About half of the total33P depleted by cotion roots came from outside the root hair cylinder whereas most of33P taken up by rape was from within the root hair cylinder. Plants grown in a macrostructured soil may have roots growing in voids, within aggregates or on the surfaces of aggregates. The results of this study demonstrate that root hairs have a strong influence on the accessibility of phosphorus to roots in such a soil, and thus on the phosphorus nutrition of plants.  相似文献

Fine root turnover is a major pathway for carbon and nutrient cycling in terrestrial ecosystems and is most likely sensitive to many global change factors. Despite the importance of fine root turnover in plant C allocation and nutrient cycling dynamics and the tremendous research efforts in the past, our understanding of it remains limited. This is because the dynamics processes associated with soil resources availability are still poorly understood. Soil moisture, temperature, and available nitrogen are the most important soil characteristics that impact fine root growth and mortality at both the individual root branch and at the ecosystem level. In temperate forest ecosystems, seasonal changes of soil resource availability will alter the pattern of carbon allocation to belowground. Therefore, fine root biomass, root length density (RLD) and specific root length (SRL) vary during the growing season. Studying seasonal changes of fine root biomass, RLD, and SRL associated with soil resource availability will help us understand the mechanistic controls of carbon to fine root longevity and turnover. The objective of this study was to understand whether seasonal variations of fine root biomass, RLD and SRL were associated with soil resource availability, such as moisture, temperature, and nitrogen, and to understand how these soil components impact fine root dynamics in Larix gmelinii plantation. We used a soil coring method to obtain fine root samples (⩽2 mm in diameter) every month from May to October in 2002 from a 17-year-old L. gmelinii plantation in Maoershan Experiment Station, Northeast Forestry University, China. Seventy-two soil cores (inside diameter 60 mm; depth intervals: 0–10 cm, 10–20 cm, 20–30 cm) were sampled randomly from three replicates 25 m × 30 m plots to estimate fine root biomass (live and dead), and calculate RLD and SRL. Soil moisture, temperature, and nitrogen (ammonia and nitrates) at three depth intervals were also analyzed in these plots. Results showed that the average standing fine root biomass (live and dead) was 189.1 g·m−2·a−1, 50% (95.4 g·m−2·a−1) in the surface soil layer (0–10 cm), 33% (61.5 g·m−2·a−1), 17% (32.2 g·m−2·a−1) in the middle (10–20 cm) and deep layer (20–30cm), respectively. Live and dead fine root biomass was the highest from May to July and in September, but lower in August and October. The live fine root biomass decreased and dead biomass increased during the growing season. Mean RLD (7,411.56 m·m−3·a−1) and SRL (10.83 m·g−1·a−1) in the surface layer were higher than RLD (1 474.68 m·m−3·a−1) and SRL (8.56 m·g−1·a−1) in the deep soil layer. RLD and SRL in May were the highest (10 621.45 m·m−3 and 14.83m·g−1) compared with those in the other months, and RLD was the lowest in September (2 198.20 m·m−3) and SRL in October (3.77 m·g−1). Seasonal dynamics of fine root biomass, RLD, and SRL showed a close relationship with changes in soil moisture, temperature, and nitrogen availability. To a lesser extent, the temperature could be determined by regression analysis. Fine roots in the upper soil layer have a function of absorbing moisture and nutrients, while the main function of deeper soil may be moisture uptake rather than nutrient acquisition. Therefore, carbon allocation to roots in the upper soil layer and deeper soil layer was different. Multiple regression analysis showed that variation in soil resource availability could explain 71–73% of the seasonal variation of RLD and SRL and 58% of the variation in fine root biomass. Ten accessions belonging to the Brassica oleracea subspecies alba and rubra, and to B. oleracea var. sabauda were used in this study. Protoplasts were isolated from leaves and hypocotyls of in vitro grown plants. The influence of selected factors on the yield, viability, and mitotic activity of protoplasts immobilized in calcium alginate layers was investigated. The efficiency of protoplast isolation from hypocotyls was lower (0.7 ± 0.1 × 106 ml−1) than for protoplasts isolated from leaf mesophyll tissue (2 ± 0.1 × 106 ml−1). High (70–90%) viabilities of immobilized protoplasts were recorded, independent of the explant sources. The highest proportion of protoplasts undergoing divisions was noted for cv. Reball F1, both from mesophyll (29.8 ± 2.2%) and hypocotyl (17.5 ± 0.3%) tissues. Potassium (K+) and chloride (Cl) are two essential elements for plant growth and development. While it is known that plants possess specific membrane transporters for transporting K+ and Cl, it remains unclear if they actively use K+-coupled Cl cotransporters (KCC), as used in animals, to transport K+ and Cl. We have cloned an Oryza sativa cDNA encoding for a member of the cation–Cl cotransporter (CCC) family. Phylogenetic analysis revealed that plant CCC proteins are highly conserved and that they have greater sequence similarity to the sub-family of animal K+–Cl cotransporters than to other cation–Cl cotransporters. Real-time PCR revealed that the O. sativa cDNA, which was named OsCCC1, can be induced by KCl in the shoot and root and that the expression level was higher in the leaf and root tips than in any other part of the rice plant. The OsCCC1 protein was located not only in onion plasma membrane but also in O. sativa plasma membrane. The OsCCC1 gene-silenced plants grow more slowly than wild-type (WT) plants, especially under the KCl treatment regime. After 1 month of KCl treatment, the leaf tips of the gene-silenced lines were necrosed. In addition, seed germination, root length, and fresh and dry weight were distinctly lower in the gene-silenced lines than in WT plants, especially after KCl treatment. Analysis of Na+, K+, and Cl contents of the gene-silenced lines and WT plants grown under the NaCl and KCl treatment regimes revealed that the former accumulated relatively less K+ and Cl than the latter but that they did not differ in terms of Na+ contents, suggesting OsCCC1 may be involved in K+ and Cl transport. Results from different tests indicated that the OsCCC1 plays a significant role in K+ and Cl homeostasis and rice plant development.  相似文献
