A further phytochemical investigation on the whole plants of Ypsilandra thibetica yielded one sapogenin and 12 spirostanol saponins. Five of these are new compounds, designated as ypsilandrosides C-G (26). Their structures were determined by detailed spectroscopic analysis, including extensive 1D and 2D NMR data, and by the result of a hydrolytic reaction. Compounds 25 were rare steroidal saponins that an apiofuranosyl unit was directly linked at C-3 of the aglycone. Selected spirostanol saponins (26, 9) were tested for their cytotoxic activities against K562, SPC-A-1, BGC-823, Eca-109, and AGS cell lines. Compounds 6 and 9 showed moderate inhibitory activity against all five cell lines. The antifungal properties of the new spirostanol saponins (26) against Candida albicans were also determined.  相似文献

Seven triterpenoid saponins, including four new compounds, catunarosides A–D (14), and three known compounds, swartziatrioside (5), aralia-saponin V (6), araliasaponin IV (7) were isolated from the stem bark of Catunaregam spinosa, a Chinese mangrove associate. Their structures were elucidated on the basis of their spectral data and hydrolysis experiments. The antifeedant activities of compounds 17 against Plutella xylostella were also evaluated.  相似文献

Tribulus terrestris is a valuable herb known for its application in the folk medicine in many parts of the world. Furostanol and spirostanol saponins of tigogenin, neotigogenin, gitogenin, neogitogenin, hecogenin, neohecogenin, diosgenin, chlorogenin, ruscogenin and sarsasapogenin type are frequently found in this plant. Four sulphated saponins of tigogenin and diosgenin type are also isolated. Extracts and steroidal saponins have been found to possess various pharmacological activities. Preparations based on the saponin fraction of T. terrestris are used for treatment of infertility and libido disorders in men and women, as well as for treatment of cardiac diseases. Food supplements containing T. terrestris extracts are on sale in USA and Europe with claim of a general stimulating action. In this review the steroidal saponins found in T. terrestris are presented, covering the literature up to 2004. The data reveal clear difference in the saponin composition of samples from this plant species collected from different geographical regions. A comprehensive account on the biological activity of extracts and individual saponins is also included.  相似文献

The genus Chlorophytum (Liliaceae) owing to the presence of pharmacologically important saponins has attracted interest of the scientific community to investigate the chemistry of the saponins and study their cytotoxicity. Chloromaloside-A having cytotoxicity against cancer cell lines has been isolated from C. malayense, while saponins from C. borivilianum are gaining popularity as substitute for viagra. Leveillula on monocotyledonous plants have been recorded as L. taurica by several authors, whereas the fungus on Allium has been described as an independent species, namely L. allii, by some authors. We sequenced ca 600 bp of the rDNA ITS region for two Leveillula specimens from Allium and Polianthes (both from monocotyledons) and compared them with several already published sequences from Leveillula isolates from dicotyledons. Pair-wise percentages of sequence divergences were calculated for all Leveillula isolates. The ITS sequence of the Polianthes isolate was identical to L. taurica on Helianthus and Vicia. The sequence of the Allium isolate was 99.5 % identical to L. taurica on Euphorbia, Haplophylum, Peganum, etc. These results suggest close relationships between monocot and dicot pathogenic Leveillula species. The identity between two monocot isolates was 98.4 %. Phylogenetic analysis revealed that the two monocot isolates do not group into a clade together. This result suggests that Leveillula acquired parasitism to monocots at least twice independently.  相似文献

Chloride ions are necessary to compensate for the positively charged potassium ions imported into guard cells of Allium cepa L. during stomatal opening. Therefore an external Cl- supply of intact Allium plants is important. But high levels of chloride have been found to reduce the sensitivity of the starch-lacking stomata and isolated guard cell protoplasts (GCPs) from Allium to potassium ions, fusicoccin and abscisic acid. Furthermore, with high levels of chloride, malate anions disappear from the guard cells of Allium, a finding which contrasts with situation in Vicia where the stomatal sensitivity to K+ ions, fusicoccin and ABA is not influenced by Cl- ions and malate levels are unaffected. Phytochemical investigation of the methanol extract from the whole plants of Patrinia scabiosaefolia Fisch. resulted in the isolation of four new triterpenoid saponins (14) along with six known compounds (510). On the basis of spectroscopic and chemical methods, the structures of the new compounds were established as 3-O-β-d-xylopyranosyl-(1→3)-α-l-rhamnopyranosyl-(1→2)-β-d-xylopyranosyl-12β,30-dihydroxy-olean-28,13β-olide (1), 3-O-α-l-rhamnopyranosyl-(1→2)-β-d-xylopyranosyl-12β,30-dihydroxy-olean-28,13β-olide (2), 3-O-β-d-xylopyranosyl-(1→2)-β-d-glucopyranosyl-12β, 30-dihydroxy-olean-28,13β-olide (3), and 3-O-β-d-glucopyranosyl-(1→4)-β-d-xylopyranosyl-(1→3)-α-l-rhamnopyranosyl-(1→2)-β-d-xylopyranosyl-oleanolic acid 28-O-β-d-glucopyranoside (4), respectively. Compounds 1–3 possess a novel 12β,30-dihydroxy-olean-28,13β-lactone aglycone and a 12β-hydroxy substituent that is rarely found in this kind of triterpenoid saponin.  相似文献

Two new furostanol saponins, 3-O-[α-l-rhamnopyranosyl-(1→4)-β-d-glucopyranosyl]-26-O-β-d-glucopyranosyl-25(R)-furosta-5,22(23)-dien-3β,20α,26-triol (1), 3-O-[β-d-glucopyranosyl-(1→3)-O-α-l-rhamnopyranosyl-(1→2)-β-d-glucopyranosyl]-26-O-β-d-glucopyranosyl-20(R)-methoxyl-25(R)-furosta-5,22(23)-dien-3β,26-diol (2) were isolated from the Dioscorea panthaica along with five known steroidal saponins (37). The structures of the new saponins were determined by detailed analysis of spectral data (including 2D NMR spectroscopy). The inhibitory activities of the saponins against α-glucosidase were investigated, gracillin (4) and 3-O-[α-l-rhamnopyranosyl-(1→2)-β-d-glucopyranosyl]-26-O-β-d-glucopyranosyl-25(R)-furosta-5,20(22)-dien-3β,26-diol (5) were found to exhibit potent activities with IC50 values of 0.11 ± 0.04 mM and 0.09 ± 0.01 mM.  相似文献

Triterpenoid saponins, impatienosides A-G, together with 12 known saponins, were isolated from the whole plants of Impatiens siculifer. Their structures were established on the basis of extensive 1D and 2D NMR and MS analyses coupled with chemical degradation. Cytotoxic activities of the isolated saponins were evaluated against three human cancer cell lines: human myeloid leukemia HL-60 cells, human stomach KATO-III adenocarcinoma, and human lung A549 adenocarcinoma.  相似文献

The spread of multidrug-resistant (MDR) strains of bacteria necessitates the discovery of new classes of antibacterials and compounds that inhibit these resistance mechanisms. At present, there are no single chemical entity plant-derived antibacterials used clinically, and this chemically diverse group deserves consideration as a source for two major reasons. First, plants have exceptional ability to produce cytotoxic agents and second there is an ecological rationale that antimicrobial natural products should be present or synthesised de novo in plants following microbial attack to protect the producer from pathogenic microbes in its environment. We have been characterising plant-derived products that are either antibacterial in their own right, or modulators of resistance in bacterial strains possessing multidrug efflux mechanisms. These efflux transporters are responsible for resistance to certain antibiotics and antiseptics and occur in strains of methicillin-resistant Staphylococcus aureus (MRSA), a major clinical problem at present. We are also investigating plant sources for compounds with activity against mycobacteria with a view to discovering drug leads with potential activity toward tuberculosis (TB) producing species. This paper will briefly review the literature on plant derived bacterial resistance modifying agents and antibacterials. Examples in this area from our own work will be given. The activities of plant-derived antibacterials show that there are many potential new classes of antibacterial agents which should undergo further cytotoxicity, microbial specificity and preclinical studies.  相似文献

走马胎是我国华南地区重要的民族药物,其体内的次生代谢物三萜皂苷具有多种药用功效。为了解三萜皂苷含量变化以及生物合成通路相关基因的调控规律,该研究对走马胎在不同组织部位以及不同外源激素处理下所呈现的表达模式进行了比较分析。结果表明:(1)三萜皂苷含量在不同组织部位间和外源激素处理下均存在显著差异,具体表现为根部组织的含量显著高于叶部组织的,而用外源水杨酸(SA)与茉莉酸甲酯(MeJA)喷施处理后其含量低于对照组(CK)。(2)对这两个差异比较组进行qRT-PCR分析结果显示,相比叶部组织,在根部组织中AkPMD、AkHDS、AkSS、AkSM以及与三萜皂苷合成相关的走马胎P450家族、UGT家族基因皆有不同程度的上调;外源SA与MeJA处理会导致合成途径上游的AkPMD、AkHDS、AkSS、AkSM上调,而下游的P450家族、UGT家族基因则有所下调。因此,通过对不同组织部位和不同外源激素处理这两种差异表达模式的比较研究,可推测在这两种表达模式下走马胎三萜皂苷的合成更多地与下游特异化修饰的酶相关。  相似文献

以野生百合渥丹、山丹和传统食用的兰州百合为研究对象,对其鳞茎中多酚类物质、11种单体酚的含量及抗氧化活性(ABTS自由基、超氧阴离子、羟自由基的清除能力,铜离子还原能力以及抑制脂质过氧化活性)进行了分析。结果表明:两种野生百合鳞茎中的多酚类物质含量及抗氧化活性均显著高于兰州百合。3种百合鳞茎中单体酚的种类也有所不同,但均含有没食子酸、矢车菊素-3-芸香糖苷、儿茶素、表儿茶素、杨梅酮、芦丁、对香豆酸、山奈酚。相关性分析显示,除对羟自由基的清除力外,各酚类物质总量与不同抗氧化指标之间呈显著正相关关系。试验结果认为,野生百合鳞茎可作为天然抗氧化资源应用于食品和医药业,具有一定的开发应用前景。  相似文献

Numerous studies have shown that large, herbivorous waterfowl can reduce quantity of aquatic plants during the breeding or wintering season, but relatively few document herbivory effects at staging areas. This study was done to determine if feeding activities of tundra swans (Cygnus columbianus columbianus) and Canada geese (Branta canadensis) had a measurable additive influence on the amount of aquatic plants, primarily muskgrass (Chara vulgaris), wild celery (Vallisneria americana), and sago pondweed (Potamogeton pectinatus), removed during the fall migration period at Long Point, Lake Erie, Ontario. Exclosure experiments done in fall 1998 and 1999 showed that, as compared to ducks and abiotic factors, these two large herbivorous waterfowl did not have any additional impact on above or below ground biomass of those aquatic plants. As expected, however, there were substantial seasonal reductions in above-ground and below-ground biomass of aquatic plants in wetlands that were heavily used by all waterfowl. We suggest that differences in large- and small-scale habitat use, feeding activity, and food preferences between tundra swans and other smaller waterfowl as well as compensatory herbivory contributed to our main finding that large waterfowl did not increase fall reductions of Chara spp, V. Americana, and P. pectinatus biomass.  相似文献

Continuing our investigations on medicinal plants of the Egyptian desert, two new triterpene glycoside derivatives, along with three known compounds have been isolated from the roots of Salsola imbricata, a shrub widely growing in Egypt. Their structures have been established as 3-O-β-d-xylopyranosyl-(1 → 2)-O-β-d-glucuronopyranosyl-akebonic acid 28-O-β-d-glucopyranoside and 3-O-β-d-xylopyranosyl-(1 → 2)-O-β-d-glucuronopyranosyl-29-hydroxyoleanolic acid 28-O-β-d-glucopyranoside on the basis of spectroscopic methods including 1D- (1H, 13C) and 2D-NMR (DQF-COSY, HSQC, HMBC) experiments as well as mass spectrometry analysis.  相似文献

该研究利用Gateway技术构建珠子参环阿屯醇合成酶基因(Panax japonicus cycloartenol synthase,PjCAS)的RNAi表达载体,利用农杆菌介导转化在珠子参细胞中成功实现了PjCAS 的RNA干扰;采用实时荧光定量PCR分析珠子参皂苷生物合成途径中关键酶基因的表达情况,同时检测转基因细胞中皂苷和植物甾醇含量的变化,探讨PjCAS基因对珠子参皂苷合成的调控作用。结果表明:(1)成功获得PjCAS基因的RNAi片段,并成功构建了PjCAS基因RNAi载体pHellsgate PjCAS。(2)经农杆菌遗传转化,获得6株实现PjCAS基因RNA干扰的转基因阳性细胞系。(3)与普通细胞系相比,转基因细胞系中PjCAS基因的表达量大约下降了85%,同时与珠子参皂苷合成直接相关的关键酶基因PjDSPjAS表达量最高分别上调了90%和150%。(4)转基因细胞系中6种单体皂苷的含量均显著高于对照组,其中达玛烷型单体皂苷Re、Rb1、Rd和齐墩果烷型单体皂苷R0、IV、IVa的平均含量比普通珠子参细胞系分别提高了28%、49%、40%、36%、59%、50%。说明珠子参皂苷含量的变化受PjCAS基因的间接调控。(5)6株转基因细胞系中植物甾醇含量较对照显著降低了53%~73%。研究发现,沉默PjCAS基因可促进珠子参皂苷合成的关键酶基因PjDSPjAS显著上调表达,并提高转PjCAS基因细胞系中单体皂苷的含量,从而促进了珠子参皂苷合成量的显著增加,证明通过抑制植物甾醇合成通路关键基因PjCAS的表达可以有效降低植物甾醇合成支路的代谢通量,使更多的代谢流朝着珠子参皂苷合成方向流动,最终促进了珠子参皂苷的生物合成。  相似文献

Three new triterpenoid saponins, elucidated as 3-O-β-d-glucopyranosyloleanolic acid 28-O-β-d-xylopyranosyl-(1→4)-α-l-rhamnopyranosyl-(1→2)-β-d-xylopyranoside (parkioside A, 1), 3-O-[β-d-apifuranosyl-(1→3)-β-d-glucopyranosyl]oleanolic acid 28-O-[β-d-apifuranosyl-(1→3)-β-d-xylopyranosyl-(1→4)-[α-l-rhamnopyranosyl-(1→3)]-α-l-rhamnopyranosyl-(1→2)β-d-xylopyranoside (parkioside B, 2) and 3-O-β-d-glucuronopyranosyl-16α-hydroxyprotobassic acid 28-O-α-l-rhamnopyranosyl-(1→3)-β-d-xylopyranosyl-(1→4)-α-l-rhamnopyranosyl-(1→2)-β-d-xylopyranoside (parkioside C, 3), were isolated from the n-BuOH extract of the root bark of Butyrospermum parkii, along with the known 3-O-β-d-glucopyranosyloleanolic acid (androseptoside A). The structures of the isolated compounds were established on the basis of chemical and spectroscopic methods, mainly 1D and 2D NMR data and mass spectrometry. The new compounds were tested for both radical scavenging and cytotoxic activities. Compound 2 showed cytotoxic activity against A375 and T98G cell lines, with IC50 values of 2.74 and 2.93 μM, respectively. Furthermore, it showed an antioxidant activity comparable to that of Trolox or butylated hydroxytoluene (BHT), used as controls, against 2,2-diphenyl-1-picryl hydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), oxygen and nitric oxide radicals.  相似文献

Bioactive natural compounds from garlic and onions have been the focus of researches for decades, firstly due to their pharmacological effects, and secondly due to their defence properties against plant diseases. In fact, garlic and onion, belonging to Allium genus, are among the oldest food plants known since ancient times and used as ingredient of many recipes and for therapeutic properties. These plants are well known to produce bioactive apolar sulphur compounds but less is known about their polar natural compounds, such as phenols, sapogenins and saponins, that are more stable to cooking, So, we continued our work on the discovery of polar bioactive metabolites from Allium with the isolation of a number of sapogenins and saponins from the wild onion species Allium elburzense, Allium hirtifolium, Allium atroviolaceum, and Allium minutiflorum, and, more recently, from the cultivated white onion, Allium cepa, and garlic, Allium sativum. In particular, the sapogenins and saponins isolated from A. elburzense and A. hirtifolium, named elburzensosides and hirtifoliosides respectively, exhibited significant antispasmodic properties. In addition, the saponins named minutosides isolated from A. minutiflorum showed promising antimicrobial activity. More recently the phytochemical analysis of A. cepa and A. sativum has been undertaken and afforded the characterization of saponins, phenols and N-cynnamic amides which showed significant antifungal activity.  相似文献
