25名运动员在相同的时间里随3种不同类型的负荷和不同的时间里承受相同的负荷,测定负荷后血浆TCA循环产物,FFA组成和尿液尿素氮,尿肌酐,游离氨基酸组分含量。结果表明:同一时间里采用运动量相近但类型不同的负荷,会产生不同的生理效应,同一负荷内容安排在下午比安排在早晨或上午更容易导致人体内蛋白质分解代谢加强;耐力负荷以早晨训练对TCA循环和FFA及其组分影响较大;下午训练利于发展速度,力量等项目的素  相似文献

探讨几种一半因素对产毒性大肠杆菌(ETEC0两种主要害居因子CFAⅠ和CFAⅡ表达的影响。结果表明细菌的生长期以及细胞外渗透压、PH、气体组成的不同均可不同程度地影响CFAⅠ和CFAⅡ的表达,其中渗透压对两种定居因子的影响有所不同。研究结果提示ETEC的CFAⅠ和CFAⅡ的表达与环境适应有关。  相似文献

孤生受体COUP-TF和nur77的功能及其作用机理仍未阐明.以DNA瞬时转染和测定氯霉素乙酰转移酶(CAT)活性,以及凝胶阻抑测定,分析COUP-TF和nur77的相互作用对视黄酸应答元件(RAREs)的影响.实验表明,COUP-TF通过降低RAREs的基础活性,来增强RARE对视黄酸(RA)的敏感性,而nur77则拮抗COUP-TF的作用.nur77能够加强不同RAREs的转录活性,并且与RA的诱导无关.结果证实,nur77通过与COUP-TF的直接作用而对RAREs产生影响,从而抑制COUP-TF与RAREs结合和COUP-TF的转录活性  相似文献

表皮生长因子(EGF)、转化生长因子-α(TGFα)、表皮生长因子受体(EGFR)和蛋白激酶C(PKC)与细胞生长、增殖分化调节和细胞癌变有密切关系。作者用免疫组织化学方法检测了细支气管肺泡细胞增生(BAH)和细支气管肺泡细胞癌(BAC)的EGF、TGFα、EGFR和PKC表达。结果表明:BAC中的EGF、TGFα阳性率和阳性强度以及EGFR、PKC阳性强度均明显高于BAH。BAH的重度不典型增生病例,其EGF、TGFα、EGFR和PKC均呈高表达。TGFα、EGFR和PKC三者在BAC和BAH中的表达存在明显相关性。提示:TGFα及其受体EGFR和PKC是细支气管肺泡细胞增生、恶性转化和肺泡癌细胞失控生长的重要因素。  相似文献

神经生长抑制因子( G I F)是一种特异存在于哺乳动物脑中的金属硫蛋白(m etallothionein, M T)类似物,又称 M T Ⅲ.它与 M T 有相同的结合 Zn(Ⅱ), Cd(Ⅱ), Cu(Ⅰ)等金属的能力,但与 M T 不同的是它能够抑制神经细胞的生长,并发现在患 Alzheim er disease( A D 症)病人的大脑中 G I F蛋白量和m R N A 的量均显著下降,研究证明 G I F对神经细胞的抑制活性主要存在于其 β结构域中.为进一步研究 β结构域结构和功能的关系,将 β结构域的 c D N A 克隆入融合表达载体p G E X 4 T 1 中, I P T G 诱导并高效表达了 β结构域蛋白,通过氨基酸组成和质谱的测定,证明得到了目的蛋白.利用金属重组的方法,分别得到了结合 Cd 和 Zn 的 G I F 的 β结构域,并测定了其巯基和金属含量对蛋白量的比值,证明所得 G I F β与 M T β在结合金属能力上十分相似.用紫外光谱学的研究表明, Cd M T 的 β结构域在250 nm 处比 Cd G I F 的 β结构域有一明显肩峰,从而表明二者的金属—巯基结合簇的结构有明显不同,而这种结构上的差异有可能导致二者在功能上的不同.  相似文献

In the present work LC-MS/MS was applied to measure the concentrations of intermediates of glycolysis and TCA cycle during autonomous, cell-cycle synchronized oscillations in aerobic, glucose-limited chemostat cultures of Saccharomyces cerevisiae. This study complements previously reported oscillations in carbon dioxide production rate, intracellular concentrations of trehalose and various free amino acids, and extracellular acetate and pyruvate in the same culture. Of the glycolytic intermediates, fructose 1,6-bisphosphate, 2- and 3-phosphoglycerate, and phosphoenolpyruvate show the most pronounced oscillatory behavior, the latter three compounds oscillating out of phase with the former. This agrees with previously observed metabolic control by phosphofructokinase and pyruvate kinase. Although individually not clearly oscillating, several intermediates of the TCA cycle, i.e., alpha-ketoglutarate, succinate, fumarate, and malate, exhibited increasing concentration during the cell cycle phase with high carbon flux through glycolysis and TCA cycle. The average mass action ratios of beta-phosphoglucomutase and fumarase agreed well with previously determined in vitro equilibrium constants. Minor differences resulted for phosphoglucose isomerase and enolase. Together with the observed close correlation of the pool sizes of the involved metabolites, this might indicate that, in vivo, these reactions are operating close to equilibrium, whereby care must be taken due to possible differences between in vivo and in vitro conditions. Combining the data with previously determined intracellular amino acid levels from the same culture, a few clear correlations between catabolism and anabolism could be identified: phosphoglycerate/serine and alpha-ketoglutarate/lysine exhibited correlated oscillatory behavior, albeit with different phase shifts. Oscillations in intracellular amino acids might therefore be, at least partly, following oscillations of their anabolic precursors.  相似文献

The aim of the present study was to examine how combined strength and endurance training in the morning and evening influences the adaptations in strength and endurance performance, perception of time management, psychological well-being and sleep. The combined training period lasted for 24 weeks and the participants were divided into the morning training (MG, n = 18), evening training (EG, n = 24) and control groups (CG, n = 10). Isometric leg press force (iLP), maximal oxygen consumption (VO2max), sleep behavior, fatigue, time management, motivation, self-esteem and health-related quality of life (HRQoL) were assessed. Morning to evening difference in iLP was observed in both MG and EG at Pre and Post, with higher force values in the evening, but not for VO2max. iLP force increased significantly in EG in the morning (p < 0.001) and evening (p = 0.010). VO2max increased in MG and EG both in the morning (both p < 0.001) and in the evening (MG: p < 0.001; EG: p = 0.003). Participants of the present study slept 7–8 h per night and the self-reported sleep duration, get-up time and the average time to go to bed were similar between the groups and did not change from Pre to Post. From HRQoL dimensions, the score for bodily pain decreased in MG (p = 0.029) and significant between-group differences were observed for Pre-Post changes in MG and EG (p = 0.001) as well as between MG and CG (p < 0.001). In vitality, a significant between-group difference was observed for Pre to Post changes in MG and EG (p = 0.014). Perception of time management decreased in EG (p = 0.042) but stayed unchanged for MG and CG. For the intrinsic motivation to participate, significant between-group differences were observed for MG and EG (p = 0.033) and between MG and CG (p = 0.032) for Pre to Post changes. Self-esteem improved in MG (p = 0.029) and EG (p = 0.024). The present combined strength and endurance training program performed in the morning and in the evening led to similar improvements in strength and endurance performance. Training in the morning or in the evening did not disrupt the already good sleep behavior and it was able to further increase the self-esteem. Although training in the morning hours may leave more time for free time activities or social life (i.e. family and friends) compared to the evening training, it might be more challenging to stay motivated to participate in prolonged training programs in the morning hours.  相似文献

Current thought is that proliferating cells undergo a shift from oxidative to glycolytic metabolism, where the energy requirements of the rapidly dividing cell are provided by ATP from glycolysis. Drawing on the hexokinase–mitochondrial acceptor theory of insulin action, this article presents evidence suggesting that the increased binding of hexokinase to porin on mitochondria of cancer cells not only accelerates glycolysis by providing hexokinase with better access to ATP, but also stimulates the TCA cycle by providing the mitochondrion with ADP that acts as an acceptor for phosphoryl groups. Furthermore, this acceleration of the TCA cycle stimulates protein synthesis via two mechanisms: first, by increasing ATP production, and second, by provision of certain amino acids required for protein synthesis, since the amino acids glutamate, alanine, and aspartate are either reduction products or partially oxidized products of the intermediates of glycolysis and the TCA cycle. The utilization of oxygen in the course of the TCA cycle turnover is relatively diminished even though TCA cycle intermediates are being consumed. With partial oxidation of TCA cycle intermediates into amino acids, there is necessarily a reduction in formation of CO2 from pyruvate, seen as a relative diminution in utilization of oxygen in relation to carbon utilization. This has been assumed to be an inhibition of oxygen uptake and therefore a diminution of TCA cycle activity. Therefore a switch from oxidative metabolism to glycolytic metabolism has been assumed (the Crabtree effect). By stimulating both ATP production and protein synthesis for the rapidly dividing cell, the binding of hexokinase to mitochondrial porin lies at the core of proliferative energy metabolism. This article further reviews literature on the binding of the isozymes of hexokinase to porin, and on the evolution of insulin, proposing that intracellular insulin-like proteins directly bind hexokinase to mitochondrial porin.  相似文献

Acute neuromuscular and endocrine adaptations to weight-lifting were investigated during two successive high intensity training sessions in the same day. Both the morning (I) (from 9.00 to 11.00 hours) and the afternoon (II) (from 15.00 hours to 17.00 hours) training sessions resulted in decreases in maximal isometric strength (p less than 0.01 and less than 0.05), shifts (worsening) in the force-time curve in the absolute scale (p less than 0.05 and ns.) and in decreases in the maximal integrated EMG (p less than 0.01 and less than 0.05) of the selected leg extensor muscles. Increases in serum total (p less than 0.05) and free testosterone (p less than 0.01) and in cortisol (p less than 0.01) concentrations were found during training session II. These were followed by decreases (p less than 0.001 and p less than 0.01 and ns.) in the levels of these hormones one hour after the termination of the session. The responses during the morning training session were different with regard to the decreases in serum total testosterone (p less than 0.05), free testosterone (ns.) and cortisol (p less than 0.05). Only slight changes were observed in the levels of luteinizing hormone and sex hormone-binding globulin during the training sessions. Increases (p less than 0.01) took place in somatotropin during both training sessions. The present findings suggest that high intensity strengthening exercises may result in acute adaptive responses in both the neuromuscular and endocrine systems. The tricarboxylic acid (TCA) cycle is the central hub of oxidative metabolism, running in the classic forward direction to provide carbon for biosynthesis and reducing agents for generation of ATP. Our metabolic tracer studies in melanoma cells showed that in hypoxic conditions the TCA cycle is largely disconnected from glycolysis. By studying the TCA branch point metabolites, acetyl CoA and citrate, as well as the metabolic endpoint glutamine and fatty acids, we developed a comprehensive picture of the rewiring of the TCA cycle that occurs in hypoxia. Hypoxic tumor cells maintain proliferation by running the TCA cycle in reverse. The source of carbon for acetyl CoA, citrate, and fatty acids switches from glucose in normoxia to glutamine in hypoxia. This hypoxic flux from glutamine into fatty acids is mediated by reductive carboxylation. This reductive carboxylation is catalyzed by two isocitrate dehydrogenases, IDH1 and IDH2. Their combined action is necessary and sufficient to effect the reverse TCA flux and maintain cellular viability.  相似文献

The purpose of this study was to determine the effect of caffeine ingestion on physical performance after prolonged endurance exercise. Twenty three trained male volunteers participated in a 40-km march and were divided into two groups, matched for caffeine clearance rate and aerobic capacity. The experimental group ingested, prior to the march, a caffeinated drink at a dose of 5 mg.kg-1 body mass and at the 3rd and 5th h of marching an additional drink at a dose of 2.5 mg.kg-1 body mass. The control group ingested a drink of equal volume at the same times. Upon termination of the march each subject performed a cycle ergometer test at an intensity of 90% maximal oxygen consumption. Time to exhaustion and rate of perceived exertion (RPE) were recorded. Blood samples were drawn predrink, at the 3rd and 5th h of marching and immediately after the cycle ergometer test, and were analysed for caffeine, free fatty acids (FFA), lactate and glucose levels. Plasma FFA levels increased during the march (p less than 0.05), with no significant difference between groups. Lactate levels increased in the experimental group (p less than 0.05), with no significant change in the control group. Glucose levels did not change significantly in either group. After the cycle ergometer test, lactate levels were significantly higher in the experimental, as compared to the control group (3.77 +/- 0.33 vs 2.52 +/- 0.35 mmol.l-1, respectively). A global kinetic study of the central metabolism of Vero cells cultivated in a serum‐free medium is proposed in the present work. Central metabolism including glycolysis, glutaminolysis, and tricarboxylic acid cycle (TCA) was demonstrated to be saturated by high flow rates of consumption of the two major substrates, glucose, and glutamine. Saturation was reavealed by an accumulation of metabolic intermediates and amino acids, by a high production of lactate needed to balance the redox pathway, and by a low participation of the carbon flow to the TCA cycle supply. Different culture conditions were set up to reduce the central metabolism saturation and to better balance the metabolic flow rates between lactate production and energetic pathways. From these culture conditions, substitutions of glutamine by other carbon sources, which have lower transport rates such as asparagine, or pyruvate in order to shunt the glycolysis pathway, were successful to better balance the central metabolism. As a result, an increase of the cell growth with a concomitant decrease of cell death and a better distribution of the carbon flow between TCA cycle and lactate production occurred. We also demonstrated that glutamine was a major carbon source to supply the TCA cycle in Vero cells and that a reduction of lactate production did not necessary improve the efficiency of the Vero cell metabolism. Thus, to adapt the formulation of the medium to the Vero cell needs, it is important to provide carbon substrates inducing a regulated supply of carbon in the TCA cycle either through the glycolysis or through other pathways such as glutaminolysis. Finally, this study allowed to better understand the Vero cell behavior in serum‐free medium which is a valuable help for the implementation of this cell line in serum‐free industrial production processes. 1. Circulating levels of free fatty acids (FFA) and growth hormone (GH) were measured over a 24 hr period during the crop gland cycle of domestic pigeons (Columba livia). 2. Plasma FFA levels showed a marked circadian rhythm (p less than 0.004) with the peak level (considerably higher than that previously reported in nonbreeding pigeons) occurring at approximately 2400 hr. 3. It is suggested that these elevated levels reflect the increased demands for energy during incubation and crop "curd" production. 4. Diurnal fluctuations of plasma GH synchronizing with those of plasma FFA were not significant though peak concentrations were noted 4 hr previous to those of FFA. 5. Pigeons studied 12 days after hatching of young had the lowest levels of both plasma GH (p less than 0.06) and FFA (p less than 0.02) indicating their interacting relationship. 6. A consistently higher FFA level (p less than 0.02) was observed in the crop glands of male pigeons.  相似文献

A comparative study of the enzymes of tricarboxylic acid (TCA) and glyoxylate cycles in the mutant Yarrowia lipolytica strain N1 capable of producing -ketoglutaric acid (KGA) and citric acid showed that almost all enzymes of the TCA cycle are more active under conditions promoting the production of KGA. The only exception was citrate synthase, whose activity was higher in yeast cells producing citric acid. The production of both acids was accompanied by suppression of the glyoxylate cycle enzymes. The activities of malate dehydrogenase, aconitase, NADP-dependent isocitrate dehydrogenase, and fumarase were higher in cells producing KGA than in cells producing citric acid.  相似文献
