Regression of 4 intermolt and induction of 14 early ecdysone puffs in two inbred stocks of Drosophila with high (HA) and low (LA) activity and hybrid F1 HA x LA were studied. Experiments were performed in vitro with ecdysone-containing media. Differences in developmental rate and fitness of stocks HA, LA and their hybrid were shown to correlate with the rate of change in polythene chromosomes puffing activity. Adaptive heterosis and ontogenetic homeostasis mechanisms are under discussion in connection with the results obtained.  相似文献

The influence of long-term heating on the puffing activity of polytene chromosomes in the early prepupa salivary glands was investigated. The activity of puffs was estimated by two criteria: size and frequency. The rearing of insects at a temperature of 29 degrees resulted in puff changes: the activity of some puffs increased or depressed, some puffs were inhibited, other puffs were induced newly. The differential response of each chromosome was observed. A possible mechanism of the effect of heating on the puff activity of polytene chromosomes is discussed.  相似文献

The effects of inbreeding and low temperature on the pattern of homologous chromosome synapsis in ovarian nurse cell nuclei of Drosophila melanogaster strains were studied. Exposure to decreased temperature (16 degrees C) caused a noticeable increase in the rate of asynapses of homologous chromosomes, whereas this effect was insignificant for F30 inbreeding generation. Long-term inbreeding has a substantial effect on the relative positions of chromosomes in the nurse cell nuclei. This is visually evident only in the interstrain hybrids between highly inbred strains LA (F923) and HA (F923) or between either strain and laboratory strain Canton S or the flies from the natural population, where abnormalities in homologous chromosome synapsis are observed in virtually all nuclei.  相似文献

Polymorphism of heterochromatin C-blocks in chromosomes of rye genome has been studied in the F1BC1 hybrids and the D/A substitution lines of rye-wheat amphidiploids (Verasen x L374, Novosibirskaya x x L246)--secalotriticum for revealing cytogenetic markers of rye chromosomes. An increase in polymorphism for the presence and value of heterochromatin blocks in chromosomes of rye genome was shown in the F1BC1 hybrids ((Verasen x L374) x L145 x L145) and unstable genotypes of the F6BC1 chromosome substitution lines (line 118 (Novosibirskaya x L246) x Reso x Reso), line 104 ((Verasen x x L374) x Garmoniya x Garmoniya) that was related to activation of mobile genetic elements present in cereals. Heterochromatin markers of all seven chromosomes in rye genome were revealed. The polymorphism system of heterochromatin blocks may serve as a marker for specificity of the linear chromosome structure during reconstruction of synthetic cereal genomes, and as a test for cytological and morphogenetic stability of hybrid polygenomes in a series of generations.  相似文献

We studied common wheat alloplasmic lines differing in fertility traits, which had been obtained from the backcross progeny of barley-wheat hybrids Hordeum vulgare L. (2n = 14) x Triticum aestivum L. (2n = 42), using molecular analysis and chromosome C-banding. It was found that the nuclei of all alloplasmic lines studied, regardless of their fertility traits, contained only the common wheat chromosomes (2n = 42). The formation of line L-79(10)(3)F6, stable for self-fertility, from line L-79(10)(3)F6 was accompanied by changes of the proportions of simple sequence repeats of the parental common wheat varieties in the nuclear genome. The presence of barley genome fragments in line accessions with incomplete self-fertility was shown by RAPD. Heteroplasmy for mitochondrial genome loci was detected in these lines with the use of primers specific to the tMet-18S-5S repeat of mitochondrial ribosomal genes.  相似文献

Wheat-aegilops hybrid plants Triticum aestivum L. (2n = 42) x Aegilops cylindrica Host (2n = 28) were investigated with using microsatellite markers. In two BC1F9 lines some genome modifications connected with losing DNA fragments of initial variety or appearing of Aegilops genome elements were detected. In some investigated hybrids new amplicons lacking in parental plants were found. Substitution of wheat chromosomes for aegilops chromosomes was not revealed. Analysis of microsatellite loci in BC2F5 plants showed stable introgression of aegilops genetic elements into wheat; elimination of some transferred aegilops DNA fragments in the course of backcrossing; decreasing size of introgressive elements after backcrossing. Introgressive lines were classified according to genome changes.  相似文献

Summary Two F5 strains of tetraploid triticale (2n= 4x=28), obtained from 6x triticaleX2 rye progenies, were crossed with diploid and tetraploid rye, some durum and bread wheats, and various 8x and 6x triticale lines. Meiosis in the different hybrid combinations was studied. The results showed that the haploid complement of these triticales consists of seven chromosomes from rye and seven chromosomes from wheat. High frequencies of PMCs showing trivalents were observed in hybrids involving the reference genotypes of wheat and triticale. These findings proved that several chromosomes from the wheat component have chromosome segments coming from two parental wheat chromosomes. The origin of these heterogeneous chromosomes probably lies in homoeologous pairing occurring at meiosis in the 6x triticaleX2x rye hybrids from which 4x triticale lines were isolated. A comparison among different hybrids combinations indicated that the involvement of D-genome chromosomes in homoeologous pairing is quite limited. In contrast, meiotic patterns in 4x triticale X 2x rye hybrids showed a quite high pairing frequency between some R chromosomes and their A and B homoeologues.  相似文献

The backcross progenies of the barley-wheat hybrids Hordeum vulgare L. (2n = 14) x Triticum aestivum L. (2n = 42) and two alloplasmic lines derived from them were studied using microsatellite markers of barley and wheat. The F1 hybrids and first backcross plants BC1 contained the genetic material of both cultivated barley and the cultivars of common wheat involved in developing of these hybrid genotypes. The genomes of BC3, BC4, and alloplasmic lines contained no microsatellite markers of the cultivated barley, whereas chromosomes of each homeologous group of common wheat were identified. In chromosomes of backcross progenies BC3, BC4, and alloplasmic lines yielded by backcrosses of hybrids and various common wheat cultivars, microsatellite markers of the parental wheat cultivars were shown to undergo recombination.  相似文献

Male hybrids of the cross D. azteca x D. athabasca are larger (hybrid giant males) than their parents, whereas hybrid females are of the same size as the parental species. Microspectrophotometric measurements have shown that the larval polytene salivary gland chromosomes of hybrid giant males undergo one more endoreplication than those of their sisters or parents. Replication patterns of the larval salivary gland chromosomes were compared after pulse labeling with 3H-thymidine and autoradiography. In females of either species as well as of hybrids X-chromosomes and autosomes are equally labeled, i.e. all chromosome arms replicate synchronously. In males, however, often fewer sites are labeled on the X-chromosome than on the autosomes. In addition, in a significant number of nuclei from D. athabasca males and also from hybrid giant males the converse can also be observed: i.e. more sites are labeled on the X-chromosome than on the autosomes. The modified labeling patterns are interpreted as an indication of a time-shift in the replication of hemizygous X-chromosomes in males, in relation to the autosomes.  相似文献

Summary The effect of inbreeding on the puffing patterns of polytene chromosomes of Drosophila subobscura was analysed. Puffing activity was studied in two strains of D. subobscura: one which had been subject to inbreeding for 288 generations, and in the hybrids from a cross between them. A strong overall decrease in puffing activity was found in the inbred line. In general, hybrids behaved in a similar way to the inbred line or showed activity intermediate between the two lines. The fertility and viability of the two homozygous lines and of the hybrids were also determined. These parameters of fitness are very low in the inbred line. Polytene chromosomes of Drosophila hydei have been employed to study the effect of bromodeoxyuridine (BrdU) on chromosome function and structure. Two media have been tested for their capacity to permit substitution of thymidine by BrdU in larval salivary glands. In medium L up to 80% of the polytene DNA is replicated with maximum substitution of 80% of the thymidine in one strand by BrdU. The degree of substitution achieved with this medium is uniform within individual nuclei but varies between nuclei. An alternate medium (S) results in more complete substitution but less total DNA synthesis. Autoradiographic analyses reveal that extensive BrdU substitution is correlated with an inhibition of chromosomal RNA synthesis. Nucleolar RNA synthesis is almost totally suppressed by exposure to BrdU. Chromosomes which exhibit extensive BrdU incorporation produce puffs in response to a temperature shock or in vitro incubation. Preliminary studies suggest that puffing at some loci is anomalous. The data, therefore, indicate that moderate BrdU incorporation completely suppresses nucleolar RNA synthesis and reduces chromosomal RNA synthesis but does not generally prevent the induction of new puffs. — Exposure of larvae to BrdU also stabilizes the morphology of their polytene chromosomes in the presence of high pH and high ionic strength. Chromosome morphology that is lost at 0.6 M NaCl is regained at 0.15 M NaCl in isolated nuclei derived from BrdU-fed larvae. No recovery is observed in chromosomes of unsubstituted control larvae.  相似文献

Sensitivity to mutagens was studied in mouse lines selectively bred for high analgesia (HA) and for low analgesia (LA) induced by 3-min swimming in 20 degrees C water. Apart from pain-related traits HA mice also manifest, as compared to the LA line, higher emotionality in various behavioural tests, and cope worse with the hypothermic challenge of swimming in cold water. In the present study HA mice appeared more susceptible to the mutagenic effect of whole-body gamma-radiation and mitomycin-C injection. Both treatments caused higher frequencies of chromosomal aberrations and micronucleus in bone marrow cells in the HA than in the LA line. The results are discussed in terms of a genetic correlation between animals' susceptibility to environmental stressors and the mechanism of mutagenesis. As shown by our recent molecular study, the selection for magnitude of swim analgesia has differentiated the parental outbred population into two distinct genotypes characterised by specific minisatellite and microsatellite sequences for each line, which may be genetic markers of particular traits. We conceive that the breeding strategy, along with the differentiation of stress-related phenomena, has altered the frequencies of genes controlling DNA repair in each line.  相似文献
