A novel biotinylated gradient surface was designed to bind a streptavidin-FITC. The gradient polyethylene (PE) surface was prepared by a corona treatment with a power increase along the PE length, and successively, biotinylated gradient PE surface was prepared by the reaction of polyethylenimine (PEI) and biotin. Surface properties of the gradient PE surface changed according to PE length and chemical modification. The biotinylated gradient PE surface provided a continuous gradient of streptavidin-FITC through binding of avidin-biotin. The luminosity of fluorescence that corresponds to a streptavidin-bound FITC on the biotinylated gradient PE surface increased gradually on the surface along the PE length.  相似文献

低磷胁迫下磷高效基因型大麦的根系形态特征   总被引:1,自引:0,他引:1  
在根袋土培盆栽条件下,以磷高效基因型DH110+、DH147和低效基因型DH49大麦为试验材料,利用根系分析系统分析不同施磷(P2O5)水平(极低磷25 mg·kg-1、低磷50 mg·kg-1和正常磷75 mg·kg-1)下,磷高效基因型大麦的根系形态特征及其与植株磷素吸收的关系.结果表明: 低磷胁迫显著降低大麦生物量和磷吸收量,其中磷高效基因型的生物量和磷吸收量在各施磷水平下分别为低效基因型的1.24~1.70和1.18~1.83倍;大麦的总根长、总根表面积、平均根系直径、不定根长及其根表面积、侧根长及其根表面积均随施磷水平的降低而显著降低,其中磷高效基因型大麦在各施磷水平下的总根长、总根表面积、比根长、侧根长及根表面积分别为低效基因型的1.46~2.06、1.12~1.51、1.35~1.72、1.69~2.42和1.40~1.78倍,而平均根系直径为低效基因型的70.6%~90.2%;主成分分析表明,平均根系直径、比根表面积和比根长受基因型差异的影响较为明显,是区分两类磷效率基因型大麦根系形态差异的主要指标;偏最小二乘回归分析表明,各施磷水平下,总根长、总根表面积对大麦植株磷素吸收贡献均较大,随施磷水平降低,不定根长、不定根表面积对大麦植株磷素吸收的贡献明显降低,而平均根系直径、比根长、侧根长及其根表面积的贡献明显增加.磷高效基因型大麦可通过维持侧根的生长、根细度和比根长的增加来适应低磷胁迫.  相似文献

Many soft biological tissues possess a considerable surface stress, which plays a significant role in their biophysical functions, but most previous methods for characterizing their mechanical properties have neglected the effects of surface stress. In this work, we investigate the micropipette aspiration method to measure the mechanical properties of soft tissues and cells with surface effects. The neo-Hookean constitutive model is adopted to describe the hyperelasticity of the measured biological material, and the surface effect is taken into account by the finite element method. It is found that when the pipette radius or aspiration length is comparable to the elastocapillary length, surface energy may distinctly alter the aspiration response. Generally, both the aspiration length and the bulk normal stress decrease with increasing surface energy, and thus neglecting the surface energy would lead to an overestimation of elastic modulus. Through dimensional analysis and numerical simulations, we provide an explicit relation between the imposed pressure and the aspiration length. This method can be applied to determine the mechanical properties of soft biological tissues and organs, e.g., livers, tumors and embryos.  相似文献

为揭示黍(Panicum miliaceum L.)根系对异质养分环境的生长反应,作研究了黍根系从起始斑块向目标斑块水平生长时,时始斑块和目标斑块养分水平根生长的影响,就低养分起始珏块而言,粗根生物量,粗根长度,粗根表面积和细极长度在高养分目标斑块中的分配比例均小于其在低养分目标斑块中的分配比例,而细根长度及其密度,细根表面积指及其密度的变化恰好相反,就高养分起始斑块而言,高养分目标斑块的细根长度,细根长度密度,细根表面积指数和细根表面积密均不于低养分目标斑块,而粗根对目标斑块中养分状的反应不明显。当黍根系从桢的起始斑块进入不同的目标斑块后,目标斑块的养分状况对细根生物量及其分配无影响,而显影响细根长度和表现积,这指示细根是通过长度和表面积可塑性而不是生物量变化响应目标斑块中的养分差异。  相似文献

Static adult intraspecific allometry of jaws and teeth was investigated in a sample of 100 Negro crania. The relations between tooth area, postcanine surface, incisor surface, and four viscerocranial measures were examined separately for males and females. Our results indicate a marked lack of morphological integration between P-sets within the orofacial subregion and a similar lack of correspondence between jaw size and tooth size. Allometric analyses indicate that mandibular length scales negatively allometric to maxilloalveolar length and to bigonial width, that canine base area scales positively to upper and lower jaw length, and that all the other teeth scale negatively to jaw length. The postcanine surface area was found to be negatively allometric to the canine base area, which in turn scaled isometrically to incisor surface. Hence, any lengthening of the mandible will tend to be associated with a relative shortening of the maxilla, with relatively larger canines and a relative reduction of the cheek teeth.  相似文献

To study growth responses of the roots of Panicum miliaceum L. to heterogeneous supply of nutrients. The authors analyzed the effects of the nutrient levels in both original patches (O) and destination patches (D) on the root growth of P. miliaceum when its roots were allowed to extend from original patch into destination patch. When the nutrient levels in the original patches were low, coarse root biomass ratio (coarse root biomass in the D/total coarse root biomass), coarse root length ratio (coarse root length in the D/total coarse root length), coarse root surface area ratio (coarse root surface area in the D/total coarse root surface area) and fine root length ratio (fine root length in the D/total fine root length) were greater in the destination patches with lower nutrient levels than in the destination patches with higher nutrient levels, while fine root length, fine root length density, fine root surface index, and fine root surface area density were smaller in the former than in the latter. When the nutrient levels in the original patches were high, fine root length, fine root length density, fine root surface area index and fine root surface density were greater in the destination patches with lower nutrient levels than in the destination patches with higher nutrient levels, coarse roots did not respond to the nutrient levels in the destination patches significantly. When the roots extended from the original patches with the same nutrient level into the destination patches with contrasting nutrient levels, fine root biomass and its percentage allocation did not respond to the nutrient levels in the destination patches significantly, whereas both root length and root surface area did. This indicates that the fine roots of P. miliaceum responded to difference in nutrient supply by plasticity in their length and surface area, rather than in their root biomass.  相似文献

Perinatal changes in the apical surface of the colonic absorptive cells in the rat were studied morphometrically. Cell microvilli length increased from day 20 through neonatal day 3, during which a maximum incremental growth rate was noted between fetal day 22 and neonatal day 1. Microvilli width remained almost constant throughout the period. Enlargement of the apical surface of the microvilli showed a similar developmental pattern as was seen from the measurement of length and surface area of any one of the microvilli. Fetal oral administration of milk in utero caused incremental growth in length and surface area, as well as an associated apical surface enlargement. The present study indicates that the function of the colonic absorptive cells, which is acquired later on in utero, is activated by ingestion of maternal milk after birth.  相似文献

对轻度、中度、重度和极度退化的草原群落中星毛委陵菜(Potentilla acaulis)根系构型参数及相应的土壤水分、容重和硬度等指标进行了分析, 以研究星毛委陵菜根系构型对草原退化的生态适应性。结果表明: 1)在以大针茅(Stipa grandis)为建群种的典型草原中, 随着退化程度的加剧, 星毛委陵菜在群落中的作用逐渐增强, 其根幅、根深、一级垂向根数、分蘖子株数和水平分蘖根长度显著增加; 2)根表面积、二级侧根长度、总根长和根分叉数4个根系构型参数是解释星毛委陵菜根系构型对草原退化生态适应的首选指标, 解释力依次减小, 累计贡献率为92.34%; 3)直径2 mm以下的根系对单株系星毛委陵菜的根表面积和总根长影响显著; 4)阔腰倒锥体三维根系构型是星毛委陵菜适应草原退化并使之成为建群种的优势构型。  相似文献

Methods are described for measuring the colonisation of a radish (Raphanus sativus) root system by seedlings with rifampicin resistant fluorescent pseudomonads by dilution plating, and which would take account of differences in root morphology. Differences in the levels of pseudomonad colonisation was highly dependent on the units in which surface areas was expressed. Population levels are expressed using estimates of surface area based on root length, tap-root length and root weight. The best estimate of surface area was root length, but the most practical method was surface area calculated as a function of dry weight. This method could differentiate differences in the levels of root colonisation independent of differences in root morphology and was efficient enough to allow the routine processing of a large number of replicate root samples.  相似文献

INTRODUCTION: Long-term stability of cementless acetabular cups depends on osseointegration, which requires primary stability of the implant. The aim of this study was to determine the influence of different surface treatments on the primary stability of press-fit acetabular cups. Mechanical lever-out tests were performed to quantify the stability in vitro. MATERIALS AND METHODS: A hemispherical press-fit cup design with a flattened pole was used and different surface modifications were applied: smooth, corundum-blasted, titanium plasma spray, rough titan plasma spray, and titanium plasma spray with a rim. The outer diameter of all cups was kept constant. Polyurethane foam was selected as the test material and cup insertion was performed with a maximal force of 6000 N. The excess length between the cup and the surface of the foam blocks was measured. The maximum lever-out force was measured and the lever-out torque was calculated. RESULTS: The excess length of cups with a smooth surface was significantly less (p<0.001) than for the other cups, with no significant differences among the other surface modifications. The lever-out torque for cups with a smooth surface was significantly less (p<0.001) than for the other cups, with no significant differences among the other surface modifications. CONCLUSION: Only the cup with a smooth surface showed significant differences for excess length and lever-out torque. The other surface modifications exhibited the same stability. As long as a rough surface is chosen, cup design seems to have a greater influence on stability than surface modification. Although the study did not mimic real in vivo conditions and the lever-out-torques cannot be transferred to clinical situations, initial stability before bony ingrowth occurred could be clearly analysed.  相似文献

We investigated allometric relationships between vertebral centrum cranial surface areas and body weight and skeletal lumbar length in extant platyrrhine and cercopithecid species. Platyrrhines have smaller lumbar vertebral centra regarding the cranial surface area relative to their body weight than extant catarrhines. However, the stress to the spine of quadrupeds is not only influenced by the body weight but also its length, which contributes to the amount of bending moment. Our results indicated that platyrrhines and cercopithecids have similar lumbar vertebral centrum surface areas when they are scaled on the product of the body weight and skeletal lumbar length. Platyrrhines generally tend to have relatively short lumbar columns for a given body weight. As a result of this tendency, their vertebral centra appear relatively small if only body weight is taken into account. The centrum surface area is rather constant relative to the product of the body weight and skeletal lumbar length within platyrrhines or cercopithecids, despite the fact that skeletal lumbar length is in itself rather variable relative to body weight. This result indicates that the vertebral centrum articular area, the lumbar column length and the body weight are strongly correlated with each other and that such relationships are similar between platyrrhines and cercopithecids. These relationships were observed using both the zygapophyseal and rib definitions of the lumbar vertebrae. However, they were more clearly observed when the zygapophyseal definition was adopted. It appeared that lumbar vertebrae of Proconsul nyanzae (KNM−MW 13142) had distinctively smaller surface areas relative to its body weight and lumbar length than for platyrrhines and cercopithecids, differing from extant hominoids, which have comparatively larger lumbar vertebrae. In the case of Morotopithecus, the lumbar vertebral surface area seems to be as large as in extant platyrrhines and cercopithecids if it had a reduced number of lumbar vertebrae. In this paper, a novel metal-dielectric waveguide structure is proposed to support hybrid long range surface plasmon polaritons (LRSPPs) with a highly confined mode field. The simulation results showed that our proposed structure has better mode confinement and propagation length compared to that of conventional dielectric-loaded surface plasmon polaritons (DLSPPs) waveguides. This structure offers greater flexibility for the design of surface plasmon polaritons (SPPs) waveguides by altering the trade-off between mode confinement and propagation length. The proposed structure has significant potential for application in highly integrated photonic circuits.  相似文献

林地利用被认为是污泥资源化利用的重要方式,但施用污泥对林木根系生长的影响报道较少。本研究通过根箱试验,分析表施和混施10%污泥对速生树种团花不同土层根系形态、土壤pH值和电导率动态变化及根系重金属含量的影响,并拟合土壤pH值、电导率和根系重金属含量与根长的关系。结果表明: 与不施污泥(对照)相比,混施污泥显著抑制了团花根长、根表面积和根体积增长,混施污泥120和240 d后,0~20 cm土层总根长分别为不施污泥的76.9%和67.4%;表施污泥对团花根长和根表面积的影响不显著,但显著提高了根体积。混施污泥显著提高了土壤pH值和电导率及根系重金属含量,混施污泥0~20和20~40 cm土层根系镉含量分别是不施污泥的11.5和10.0倍。线性回归拟合分析显示,不同处理0~20 cm土层的电导率与根长均呈显著负相关;表施和混施污泥根系镉含量与根长呈极显著负相关。上述结果表明,混施污泥抑制了团花根系生长,这可能是由于混施污泥提高了土壤电导率和根系镉含量所致,而表施污泥对团花根系生长的作用不明显。  相似文献

依托陇中旱农区长期的保护性耕作定位试验,对不同耕作方式下春小麦和豌豆根系空间分布特征及作物产量进行研究,以探索耕作措施影响作物产量的机制.结果表明: 随着生育期的推进,春小麦和豌豆的总根长、根表面积呈先增后减的趋势,开花期达到最大;春小麦根系苗期以0~10 cm最多,花期、成熟期10~30 cm最多;而豌豆根系苗期和成熟期均以0~10 cm最多,花期10~30 cm最多.免耕秸秆覆盖和免耕覆膜增加了根长和根表面积,春小麦和豌豆各生育时期的根长较传统耕作增加了35.9%~92.6%,根表面积增加了43.2%~162.4%.免耕秸秆覆盖和免耕覆膜优化了春小麦和豌豆根系分布,与传统耕作相比,增加了春小麦和豌豆苗期0~10 cm土层根长和根表面积分布比例,花期和成熟期深层次根系分布也显著增加,免耕秸秆覆盖在开花期30~80 cm土层根长和根表面积的分布比例分别比传统耕作提高了3.3%和9.7%.春小麦各生育期的总根长、根表面积与产量呈显著正相关,豌豆各生育期的总根长与豌豆产量呈极显著正相关.免耕秸秆覆盖和免耕覆膜较传统耕作春小麦和豌豆产量增加23.4%~38.7%,水分利用效率提高了13.7%~28.5%.在陇中旱农区,免耕秸秆覆盖和免耕覆膜可以增加作物根长和根表面积,优化了根系在土壤中的空间分布,增强作物根层吸收能力,从而提高作物产量和水分高效利用.  相似文献

为揭示丝栗栲(Castanopsis fargesii)细根功能性状对环境变化的适应机制,对郭岩山500、700、900 m海拔处丝栗栲细根功能性状及其与土壤因子的关系进行研究。结果表明,丝栗栲细根生物量与细根根长密度、表面积密度、组织密度及体积密度呈正相关,细根根长密度、体积密度、表面积密度和比根长4个性状间均呈极显著正相关关系,且均与细根组织密度呈显著负相关。根际土含水量、C和N含量与细根比根长、根长密度、体积密度、表面积密度均存在显著正相关关系,而土壤容重与细根组织密度呈正相关。海拔700 m的细根生物量、根长密度、表面积密度及体积密度显著大于海拔500和900 m的。500和900 m海拔的根长密度、表面积密度与土壤深度呈负相关,而500 m海拔细根的组织密度与土壤深度呈正相关。因此,郭岩山丝栗栲通过改变细根功能性状来适应海拔和土壤的变化。  相似文献

This report describes a novel ECTO-NOX protein with an oscillating activity having a period length of ca. 26 min encountered with buffy coat fractions and sera of aged individuals (70–100 years) that generates superoxide as measured by the reduction of ferricytochrome c. The oscillating, age-related reduction of ferricytochrome c is sensitive to superoxide dismutase, is inhibited by coenzyme Q and is reduced or absent from sera of younger individuals (20–40 years). An oscillating activity with a regular period length is a defining characteristic of ECTO-NOX proteins (a group of cell surface oxidases with enzymatic activities that oscillate). The period length of ca. 26 min is longer than the period length of 24 min for the usual constitutive (CNOX) ECTO-NOX proteins of the cell surface and sera which neither generate superoxide nor reduce ferricytochrome c. The aging-related ECTO-NOX protein (arNOX) provides a mechanism to transmit cell surface oxidative changes to surrounding cells and circulating lipoproteins potentially important to atherogenesis. Additionally, the findings provide a rational basis for the use of dietary coenzyme Q to retard aging-related arterial lesions.  相似文献

本文根据田间实测资料研究了冬小麦根系各种参数(长度、重量、活性表面和根径)的垂直分布。结果表明,在所观测地区的土壤条件下,冬小麦根系的长度、重量、表面积和体积随深度呈指数分布,而累积根系的长度、重量和表面积随深度呈双曲线型分布。根据作者的观测资料和国内外其它观测资料分析研究表明,对于不同土壤,根区各层土壤中累计根重及根长的百分比随相对深度的变化都符合下列双曲线函数形式:这一研究结果可为根系生态研究和作物对水分吸收的模拟工作提供参考。  相似文献

We have shown previously that truncating all of the variable cytoplasmic C-terminus of Kv1.1 potassium channels to G421stop had only a small inhibitory effect on their cell surface conductance density levels and cell surface protein levels. Here we investigated the role of a highly conserved cytoplasmic C-terminal charged region of five amino acids (HRETE) of the S6 transmembrane domain in the protein and conductance expression of Kv1.1, Kv1.2, and Kv1.4 channels. For Kv1.1 we found that E420stop, T419stop, and E418stop showed cell surface conductance densities and cell surface protein levels similar to full length control, whereas R417stop and H416stop exhibited essentially no conductance but their surface protein levels were similar to full length control. A bulky non-negatively charged hydrophilic amino acid at position 417 appeared to be critical for wild type gating of Kv1.1 because R417K and R417Q rescued conductance levels whereas R417A or R417E did not. The R417A mutation in the full length Kv1.1 also exhibited surface protein levels similar to control but it did not exhibit significant conductance. In contrast, mutation of the equivalent arginine to alanine in full length Kv1.2 and Kv1.4 appeared to have little or no effect on channel conductance but rather decreased cell surface protein levels by inducing partial high ER retention. These findings are consistent with the notion that the arginine amino acid in the HRETE region plays a different role in affecting conductance levels or cell surface protein levels of very closely related Kv1 potassium channels.  相似文献
