White light strongly promotes dormancy in freshly harvested cereal grains, whereas dark and after-ripening have the opposite effect. We have analyzed the interaction of light and after-ripening on abscisic acid (ABA) and gibberellin (GA) metabolism genes and dormancy in barley (Hordeum vulgare 'Betzes'). Analysis of gene expression in imbibed barley grains shows that different ABA metabolism genes are targeted by white light and after-ripening. Of the genes examined, white light promotes the expression of an ABA biosynthetic gene, HvNCED1, in embryos. Consistent with this result, enzyme-linked immunosorbent assays show that dormant grains imbibed under white light have higher embryo ABA content than grains imbibed in the dark. After-ripening has no effect on expression of ABA biosynthesis genes, but promotes expression of an ABA catabolism gene (HvABA8'OH1), a GA biosynthetic gene (HvGA3ox2), and a GA catabolic gene (HvGA2ox3) following imbibition. Blue light mimics the effects of white light on germination, ABA levels, and expression of GA and ABA metabolism genes. Red and far-red light have no effect on germination, ABA levels, or HvNCED1. RNA interference experiments in transgenic barley plants support a role of HvABA8'OH1 in dormancy release. Reduced HvABA8'OH1 expression in transgenic HvABA8'OH1 RNAi grains results in higher levels of ABA and increased dormancy compared to nontransgenic grains.  相似文献

A quantitative model for change in germination rate of barley(Hordeum vulgare L.) during and after loss of primary dormancyis presented. Change in mean germination time on a logarithmicscale is normally distributed within the period of after-ripeningand the standard deviation of this distribution is shown tobe a quantitative function of after-ripening temperature. Therate of change of mean germination time is in inverse proportionto the product of the standard deviation and a parameter whichis characteristic of the seed population. The latter parameteris the rate constant for change in probit cumulative germinationas a negative linear function of the logarithm of mean germinationtime. A model based solely on dormancy loss is combined withan existing model of change in probit viability as a functionof mean germination time to produce a model which predicts thetime to and optimum value of mean germination rate of a populationas it after-ripens. The model provides a quantitative link betweenthe effect of pre-germination and germination environments ontotal and rate of germination of an initially dormant population.Experimental data from dormant barley (cv. Triumph) stored at27, 38, 45, 50 and 60 °C, and germinated at 18 °C wereused to validate the model. The data show that germination ratecontinues to increase after primary dormancy is lost until itreaches an upper limit determined by the intrinsic germinativevigour of the seed lot. Wheat (Triticum aestivum L.) embryos form in dynamically-regulatedovular environments. Our objectives were to improve developmentof cultured immature wheat embryos by simulating, in vitro,abscisic acid (ABA) levels and O2 tensions as found in wheatovules during zygotic embryogenesis. We characterized from intactwheat kernels embryo respiration, embryo morphology and embryoand endosperm + ABA levels at 13, 19 and 25 d post-anthesis(DPA). Young (13 DPA) embryos were then excised and culturedin vitro, where they were exposed to 0·2 or 2·Ommol m–3 ±ABA and 2.·1, 2·5 or 7·4mol m–3 (6, 7 and 21%, respectively) gaseous O2. At 6and 12 d in culture, + ABA levels, embryo respiration and embryomorphology were characterized by treatment. Thirteen-day-oldembryos from two different plant populations differed by 17-foldin initial ABA content. However, this difference did not affectprecocious germination in vitro, nor did it affect the amountof exogenous ABA required to reduce precocious germination by40%. In this respect, embryos from both populations were equallysensitive to exogenous ABA. Cavity sap O2 levels (2·1to 2·5 mol m–3) were much more effective in preventingprecocious germination of cultured embryos than were cavitysap levels of ABA (0·2 to 2·0 mmol m–3).The combination of physiological levels of both ABA and O2 largelynormalized DW accumulation and embryo morphology without alteringendogenous + ABA levels. Carbon dioxide appears to modulate the growth rates of the barley(Hordeum vulgare L.) coleoptile and the enclosed leaves duringearly germination. Its effect is to stimulate the coleoptileto elongate at a rate equal to or greater than that of the enclosedleaves. Such stimulation was observed directly in barley coleoptilesfloated on CO2-sarurated water or indirectly using a DCMU solution.This mechanism ensures that the tender leaves remain enclosedwithin the structurally strong coleoptile sheath until the shootpushes through the soil surface. It is suggested that CO2 respiredby the enclosed leaves effects this modulation: the CO2 couldpass into the coleoptile chamber and thence into the coleoptilevia stomata present on coleoptile inner walls.  相似文献

The influence of a water stress or foliar ABA spraying pretreatmenton stomatal responses to water loss, exogenous ABA, IAA, Ca2+,and CO2 were studied using excised leaves of Solanum melongena.Both pretreatments increased stomatal sensitivity of water loss,in the presence and absence of CO2, but decreased stomatal sensitivityto exogenous ABA. CO2 greatly reduced the effect of exogenouslyapplied ABA. IAA decreased leaf diffusion resistance for controland ABA sprayed leaves, but did not influence the LDR of previouslywater-stressed leaves. Eight cvs of barley (Hordeum vulgare L.) were separately plantedwith Wild Oats (Avena fatua L., genetically pure line CS40)in a sand culture with two external K+ concentrations. Substantialdifferences were observed among barley cvs in their abilityto compete with wild oat. The variety Fergus was highly competitiveat both high and low [K+]e, whereas Steptoe was competitiveonly at high [K+]e, and Compana was only weakly competitivewith wild oat. The differences between barley cvs were relatedto their previously reported efficiencies of K+ uptake and utilization. Barley was grown at a range of oxygen concentrations (0.5–9mg l–1), in nutrient solutions. Growth of both shootsand seminal roots was restricted by O2 concentrations lowerthan 2–3 mg l–1) but nodal root growth was not. Root porosities were increased even at those O2 concentrationswhich did not restrict growth, and were inversely proportionalto the protein levels of the roots. Barley and rice, at the early tillering stage, were grown inaerated nutrient solutions (> 7 mg O2 l–1) and transferredto solutions of low O2 concentrations (< 0.5 mg l –1). For barley, low O2 concentrations during the first 5 days severelyinhibited growth of seminal roots had less effect on nodal roots,and did not reduce shoot growth. Longer exposure to low O2 concentrationsreduced shoot as well as root growth. Sugar concentrations inroots and shoots increased within 7 h after transfer of plantsto low O2 concentrations. After 5 days at low O2 concentrationssugar concentrations were very high in fast growing nodal rootsand in shoots, as well as in the slower growing seminal roots. In rice, low O2 concentrations increased sugar levels of rootsduring summer, but not during winter. In summer, the highersugar levels at low O2 concentrations persisted throughout adiurnal cycle. In root apices, sugar concentrations were increasedby low O2 concentrations, even though the experiment was donein winter and the bulk of the root system showed no differencein sugar levels. The data indicate that sugar accumulation, at low O2 concentrations,is caused by reduced growth and also that even apices of rootsgrown at low O2 concentrations have sufficient substrates forrespiration. The potential role of xyloglucan endotransglycosylase (XET)in GA-stimulated cell elongation was investigated during leafexpansion in barley (Hordeum vulgare L.). XET activity in aqueousextracts of leaves was detected in all segments along the elongatingblade of leaf 1 of seedlings, but was at highest levels in basalsegments. Leaf 1 elongation rates of gibberellin (GA)-responsivedwarf mutants were lower than the wild type, and accompaniedby reduced levels of XET activity. Leaf elongation rates ofthe dwarfs increased following treatment with gibberellic acid(GA3) associated with higher levels of XET activity. The slendermutant, crossed into a dwarfing background, exhibited high ratesof leaf 1 elongation and high levels of XET activity withoutadded GA3. The elongation of leaf 3 in a GA-responsive dwarfmutant was also studied. Treatment with GA3 resulted in bladeand sheath lengths being 5-fold and 7-fold (respectively) thelengths of controls, and again there were increases in bladeand sheath XET activities. To investigate the basis for changesin XET activity levels two XET-related cDNA clones were isolated.RNAs detected by the two clones occurred at the highest levelsin basal segments of rapidly elongating leaves, but they haddifferent distribution patterns along the leaf. Overall, thedata indicate that an XET-like activity is detectable in barleyleaves, that the activity level and related. During a screen for mutants with defective germination, a newphenotype was observed consisting of red pigmentation of theembryonic axis in the dormant seed. Segregation ratios, as determinedin F2 and back-crossed progeny, indicate that the phenotypeis due to a recessive single gene mutation that has been symbolizedrea to denote red embryonic axis. A closer inspection of therea phenotype revealed that the mutant is occasionally viviparous,indicating a defect in abscisic acid (ABA) metabolism. The mutationprobably affects ABA sensitivity since no difference in ABAcontent was detected in mutant versus normal tissues. Moreover,when immature mutant and wild-type embryos were incubated onmedia containing 10 M ABA, only the mutants germinated. ABA-regulatedgene expression in rea embryos differed from that of embryosof the viviparous mutant vp1 which does not respond to the inhibitoryaction of ABA at the level of immature embryo germination. Barley (Hordeum vulgare L. C.I.666) was shown to be susceptibleto the growth retardant (2-chloroethyl)-trimethylammonium chloride(CCC). The estimation of cell number in the dwarfed third leafblade indicated that a decrease in mitotic activity had occurredin treated plants. There was also a decrease in cell size intreated plants. The dwarfing action of CCC was reversed by exogenousgibberellic acid (GA3) but this was shown to be the result ofincreased cell elongation only. GA3 did not promote cell divisionin healthy or CCC-treated plants. Assay of endogenous gibberellinsshowed a significant reduction in the level of a substance correspondingto GA3 in CCC-treated plants. It is suggested that CCC-induceddwarfing of barley is largely the result of a reduction in meristematicactivity. This may be related to an effect on gibberellin biosynthesisbut is not reversed by the application of exogenous GA 3.  相似文献

Imbibed seeds of Arabidopsis thaliana (L.) Heynh., passed annuallythrough a pattern of changes in dormancy. Dormancy was brokenin summer and re-induced in autumn-winter. A second small germinationflush occurred in early spring. The role of sensitivity to light,nitrate and gibberellins (GAs) in regulating annual dormancypatterns and germination was studied with the use of GA-deficient(gal-2) and wild-type seeds. Dark-incubated seeds were exposedto a natural temperature regime for periods up to 18 monthsand at regular intervals germination capacity of portions ofseeds was tested at laboratory conditions. Germination datafitted as logistic dose response curves showed that sensitivityto light varied with the seasons in both genotypes. From interpretationof curve parameters, it is proposed that the observed sensitivitychanges involve alterations in the number of receptors, in thebinding characteristics of the receptors and/or in the responsechain initiated by ligand-receptor interaction. In this responsechain GA biosynthesis is stimulated (wild type) and sensitivityto GAs is enhanced (wild type, gal -2). GA sensitivity is alsodirectly influenced by temperature, thus without the interferenceof light. However, the significance of direct regulation ofGA requirement seemed to diminish with prolonged incubationoutdoors, whereas reversible changes in light sensitivity remainedclear. Therefore, we propose that seasonal dormancy patternsare mainly regulated by changes in sensitivity to light. GAsensitivity contributes to this pattern but is not primarilycontrolling dormancy. The GA requirement for germination isobvious as gal-2 seeds did not germinate at any time of theyear when deprived of applied GAs. However, GA biosynthesisis not required for dormancy control, as a dormancy patternwas also observed in the absence of the capacity to synthesizeGAs. Relationships between nitrate (NO-3) supply, uptake and assimilation,water uptake and the rate of mobilization of seed reserves wereexamined for the five main temperate cereals prior to emergencefrom the substrate. For all species, 21 d after sowing (DAS),residual seed dry weight (d.wt) decreased while shoot plus rootd.wt increased (15–30%) with increased applied NO-3concentrationfrom 0 to 5–20 mM . Nitrogen (N) uptake and assimilationwere as great with addition of 5 mM ammonium (NH+4) or 5 mMNO-3but NH+4did not affect the rate of mobilization of seedreserves. Chloride (Cl-) was similar to NO-3in its effect onmobilization of seed reserves of barley (Hordeum vulgare L.).Increased rate of mobilization of seed reserves with additionalNO-3or Cl-was associated with increases in shoot, root and residualseed anion content, total seedling water and residual seed watercontent (% water) 21 DAS. Addition of NH+4did not affect totalseedling water or residual seed water content. For barley suppliedwith different concentrations of NO-3or mannitol, the rate ofmobilization of seed reserves was positively correlated (r >0.95)with total seedling water and residual seed water content. Therate of mobilization of seed reserves of barley was greaterfor high N content seed than for low N content seed. Seed watercontent was greater for high N seed than for low N seed, 2 DAS.Additional NO-3did not affect total seedling water or residualseed water content until 10–14 DAS. The effects of seedN and NO-3on mobilization of seed reserves were detected 10and 14 DAS, respectively. It is proposed that the increasedrate of mobilization of seed reserves of temperate cereals withadditional NO-3is due to increased water uptake by the seedlingwhile the seed N effect is due to increased water uptake bythe seed directly. L-Tyrosine carboxy-lyase (E.C. 4. 1. 1. 25) was isolated fromroots of germinating barley (Hordeum vulgare). The enzyme requirespyridoxal phosphate for maximum activity. The optimum pH foractivity is about 7.0. The enzyme is inhibited by p-chloromercuribenzoateand hydroxylamine at 10–3 M. Enzyme activity is foundin extracts from young roots, especially from those in earlystages of development, but not in extracts from shoots of thesame plant. Localization and changes in the amounts of L-tyrosinecarboxy-lyase and aromatic amines in developing barley seedlingswere measured. Early seed development was studied in 17 genotypes of barley,Hordeum vulgare L., and 11 genotypes of rye, Secale cerealeL. The numbers of cells and nuclei in the embryos and endospermsof developing seeds were scored daily for 5 days after selfpollination. For embryos, the mean cell doubling times variedfrom 9.2–12.9 h for barley and 15.7–22.7 h for rye.Endosperm mitotic cycle times of both species were shortestover the first 24 h after pollination but then became longer.A non-linear correlation was found between the number of embryocells and the number of endosperm nuclei in barely and rye andis similar to that for other members of the Triticeae. The effects of guazatine, synthalins A and B and a homologousseries of aliphatic monoguanidines on the growth of cress, barleyand oat seedlings, and apple cell suspension cultures have beenstudied. In the homologous series of aliphatic monoguanidines[NH2C(=NH)NH(CH2)x–1CH3] greatest inhibition was foundwith x = 8-10 for cress, barley and oats and x= 10–14for apple cells. Spermine partially reversed the inhibitionin the light for cress and barley but in the dark no reversalwas found. Technical guazatine inhibited growth to a greaterextent than pure guazatine, and was comparable in toxicity tosynthalin B in cress, barley and oats. Reversal by spermineof inhibition due to guazatine and synthalin B was greater inthe light than in the dark in these plants. Calcium ions didnot reverse the toxicity of guazatine, synthalin B or dodine.Reversal of the inhibition by guazatine, synthalin B and dodineof the growth of the apple cells was considerably greater withspermine than with spermidine. Three wheat (Triticum aestivum L.) mutants that lacked dormancyat maturity were isolated from an ethylmethane sulphonate-treatedpopulation of a dormant red-grained line, Kitakei-1354 (Kitakei).The three mutants (EH47-1, EH47-2-5 and EH47-2-6) were selectedin segregating generations derived from one M2 plant. They differin morphological and physiological characteristics, showingthat these mutants contained several mutations besides non-dormancy.Despite these differences, embryos of all the mutants rapidlylost sensitivity to abscisic acid (ABA) during the later halfof seed maturation while Kitakei embryos maintained the sensitivityeven after maturity. These results suggest that embryo sensitivityto ABA plays a key role in seed dormancy. The profile of ABAcontent of EH47-1 embryos during seed development was similarto that of Kitakei, except for a significantly lower level at30 d after pollination (DAP). This reduced level of ABA at DAP30is discussed in relation to the development of seed dormancyand ABA sensitivity of the embryos. Segregation ratios for non-dormancyin progeny of EH47-1Kitakei crosses suggest that the non-dormancyof EH47-1 is a single dominant mutation.
