Anthracnose caused by Elsinoë ampelina is one of the most important table grape diseases in humid regions in Brazil and Australia. The objective of this study was to characterize E. ampelina isolates from Brazil and Australia by means of phylogenetic analyses, morphological features and pathogenicity tests. Phylogenetic relationships among 35 isolates were determined based on a data set of internal transcribed spacer (ITS), histone H3 (HIS3) and elongation factor 1‐α (TEF) sequences. In phylogenetic tree analyses, using a combined ITS and TEF sequence alignment, all E. ampelina isolates were clustered together in a single well‐supported clade. In contrast to the absence of genetic variability within ITS and TEF sequences, HIS3 sequences showed 54 polymorphic sites. The haplotype network generated from HIS3 data set showed four distinct haplotypes. EA1 was the predominant haplotype including 29 isolates from both countries. High genetic variability was observed in two Brazilian isolates, haplotype EA4, which may have lost the intron region during species evolution. Colony colours differed between Brazilian and Australian isolates, but showed similar wrinkled colony texture, absence of spores, sparse‐to‐absent white aerial mycelium and slow growth (0.049–0.060 mm/day). Brazilian isolates produced conidia of 5.65 × 2.65 μm, larger than conidia from Australian isolates, which measured 5.14 × 2.30 μm. In pathogenicity tests, all nine Australian isolates inoculated were pathogenic on detached canes and potted vines of table grape.  相似文献

Temperature‐dependent development, parasitism and longevity of the braconid parasitoids, Fopius arisanus Sonan and Diachasmimorpha longicaudata Ashmed on Bactorcera invadens Drew Tsuruta & White, was evaluated across five constant temperatures (15, 20, 25, 30 and 35°C). Developmental rate decreased linearly with increasing temperature for both the parasitoid species. Linear and Brière‐2 nonlinear models were used to determine the lower temperature threshold at which the developmental rate (1/D) approached zero. For F. arisanus, lower thresholds to complete development estimated with the linear and nonlinear models were 10.1 and 6.9°C, respectively. The total degree‐days (DD) required to complete the development estimated by the linear model for F. arisanus was 360. In D. longicaudata, the linear and nonlinear models estimated lower thresholds of 10.4 and 7.3°C, respectively, and the total DD estimated was 282. In F. arisanus, percentage parasitism differed significantly across all temperatures tested and was highest at 25°C (71.1 ± 2.5) and lowest at 15°C (46.4 ± 1.4). Parasitoid progeny sex ratio was female biased at all temperatures except at 20°C. In D. longicaudata, percentage parasitism was highest at 20°C (52.2 ± 4.0) and lowest at 15°C (27.7 ± 2.5). Parasitoid progeny sex ratio was female biased and similar for all temperatures. Adult longevity of both parasitoids was shortest at 35°C and longest at 15°C, and females lived significantly longer than males at all temperatures tested. Our findings provide some guidance for future mass rearing and field releases of the two parasitoids for the management of B. invadens in Africa.  相似文献

The rust and brown eye spot, caused by Hemileia vastatrix and Cercospora coffeicola, respectively, are the most important fungal diseases on coffee in South America. Their management is mainly by chemical treatment, and there is no genetic resistance to brown eye spot known so far. Considering the need for developing alternative products for their control, the goal of this work was to evaluate the effects of phosphites and by‐products of coffee and citrus industries on rust and brown eye spot. Formulations of coffee and citrus industry by‐products, phosphites and their combination with fungicide were evaluated in field experiments, and their effect on fungal urediniospores and conidia was evaluated in vitro. In the field, treatments were applied individually or in combination and the in vitro assays were performed with manganese phosphite (Reforce Mn), potassium phosphite and citrus industry by‐product (Fortaleza), copper phosphite and coffee industry by‐product (Fitoforce Full), and fungicide. The severity and incidence of rust and brown eye spot on coffee leaves, yield, and leaf retention were evaluated in the field. Percentage of spore germination was evaluated in vitro for both fungi, whereas mycelial growth was evaluated for C. coffeicola only. The treatments Fortaleza, Reforce Mn and Fitoforce Full suppressed both diseases with a reduction in defoliation. In the year 2012, the plants treated with Reforce Mn and Reforce Mn + Fortaleza showed a yield increase of 72 and 88%, respectively, which was similar to the results shown by the fungicide treatment. In vitro inhibition of germination of H. vastatrix urediniospores and of C. coffeicola conidia was observed and suggests that the products exert some toxic effects to both fungi. Finally, the results observed indicate that the combined use of by‐products of plant‐processing industries and phosphites is an alternative and can be added efficiently to the management of coffee diseases.  相似文献

In vitro experiments indicated that boric acid, monopotassium phosphate, sodium metabisulfite and synthetic fungicide fluopyram + tebuconazole were effective in inhibiting conidia germination and germ‐tube elongation of Venturia inaequalis. Monopotassium phosphate even at the highest concentration used in the study reduced conidia germination and germ‐tube elongation of V. inaequalis by 22.1% and 28.8%, respectively; however, the difference between two compounds at lower concentrations except 0.05% (for conidia germination) and 0.1% (for germ‐tube elongation) of boric acid was statistically significant (P ≤ 0.05). Complete inhibition was achieved by 0.01% sodium metabisulfite, 0.035% fluopyram + tebuconazole and 0.2% boric acid. Two orchard trials were conducted on the highly susceptible cv. Mutsu to apple scab to ascertain the efficacy of 0.2% boric acid, 0.5% monopotassium phosphate, 0.5% sodium metabisulfite and 0.035% fluopyram + tebuconazole for the control of apple scab. In both 2013 and 2014, except for the applications of monopotassium phosphate and sodium metabisulfite, the applications of boric acid and fluopyram + tebuconazole to trees at 10‐day intervals significantly reduced disease incidence and severity on leaves and fruit compared to the water‐treated control. In both years, the efficacy of boric acid and fluopyram + tebuconazole treatments was similar in reducing both disease incidence and severity on leaves and fruit in all monthly assessments from July to September. All treatments were neither phytotoxic to leaves and fruit nor did they adversely affect quality parameters of harvested fruit. These results show that boric acid treatment may be applied as an alternative chemical for the control of apple scab.  相似文献

Several species of cyanobacteria biomineralizing intracellular amorphous calcium carbonates (ACC) were recently discovered. However, the mechanisms involved in this biomineralization process and the determinants discriminating species forming intracellular ACC from those not forming intracellular ACC remain unknown. Recently, it was hypothesized that the intensity of Ca uptake (i.e., how much Ca was scavenged from the extracellular solution) might be a major parameter controlling the capability of a cyanobacterium to form intracellular ACC. Here, we tested this hypothesis by systematically measuring the Ca uptake by a set of 52 cyanobacterial strains cultured in the same growth medium. The results evidenced a dichotomy among cyanobacteria regarding Ca sequestration capabilities, with all strains forming intracellular ACC incorporating significantly more calcium than strains not forming ACC. Moreover, Ca provided at a concentration of 50 μM in BG‐11 was shown to be limiting for the growth of some of the strains forming intracellular ACC, suggesting an overlooked quantitative role of Ca for these strains. All cyanobacteria forming intracellular ACC contained at least one gene coding for a mechanosensitive channel, which might be involved in Ca influx, as well as at least one gene coding for a Ca2+/H+ exchanger and membrane proteins of the UPF0016 family, which might be involved in active Ca transport either from the cytosol to the extracellular solution or the cytosol toward an intracellular compartment. Overall, massive Ca sequestration may have an indirect role by allowing the formation of intracellular ACC. The latter may be beneficial to the growth of the cells as a storage of inorganic C and/or a buffer of intracellular pH. Moreover, high Ca scavenging by cyanobacteria biomineralizing intracellular ACC, a trait shared with endolithic cyanobacteria, suggests that these cyanobacteria should be considered as potentially significant geochemical reservoirs of Ca.  相似文献

In vitro experiments showed that ammonium bicarbonate and aqueous extracts of oregano were effective in inhibiting conidia germination and germ‐tube elongation of Venturia inaequalis. Complete inhibition was achieved by 1% ammonium bicarbonate, 2% oregano extract and 0.01% synthetic fungicide difenoconazole. Two orchard experiments were conducted on the highly susceptible cv. Mutsu to apple scab to investigate the efficacy of ammonium bicarbonate alone or in combination with an aqueous extract of oregano for the control of apple scab. In 2008 and 2009, except for the applications of 1% aqueous extract of oregano, the applications of ammonium bicarbonate (0.5 and 1%) and difenoconazole (0.01%) to trees at 10‐day intervals significantly reduced disease incidence and severity on leaves and fruit compared to the water‐treated control. In both years, the efficacy of 0.5 and 1% ammonium bicarbonate in inhibiting both disease incidence and severity on leaves and fruit was equally effective in all monthly assessments from June to September. Combining 0.5 and 1% ammonium bicarbonate with 1% aqueous extract of oregano did not significantly improve the efficacy of stand‐alone applications of treatments in the final assessment in 2008 and 2009. All treatments were neither phytotoxic to leaves and fruit nor did they adversely affect quality parameters of fruit including physiological disorders and taste both at harvest and after storage. These results indicate that ammonium bicarbonate treatment may be applied as an alternative chemical for the control of apple scab.  相似文献

The growing demand for alternative strategies for plant disease management has sought a reduction in the use of fungicides via the employment of resistance inducers and foliar fertilizers, among others. Therefore, this study aimed at evaluating the following foliar fertilizers for the management of phoma leaf spot: a foliar fertilizer based on macro‐ and micronutrients (Fmm: 10% N, 4% S, B, 5% Fe and 5% Zn); one based on cobalt and molybdenum (Fcm: 2% Co and 3% Mo); manganese phosphite (FMn: 30% P2O5 and 9% Mn); and the FMn+Fmm, Fcm+Fmm and FMn+Fcm+Fmm associations compared to a boscalid fungicide and a control with no treatment. The disease severity, the chlorophyll a and b contents, the net photosynthetic rate (LPR), the phenylalanine ammonia lyase (PAL) activity and the lignin content in leaves were assessment. Based on the severity, the area under the disease severity progress curve (AUSPC) and the efficiency of disease control in relation to untreated plants were calculated. The experimental design was in randomized blocks with eight treatments and three replications. Treatments FMn+Fmm and Fcm were the most effective in reducing the AUPSC in comparison with the control and promoted an increased activity of PAL. FMn was the treatment that promoted the highest increase in the LPR. There were no effects of the treatments on the lignin content compared to the control.  相似文献

AIMS: This study compares the effect of temperature (4-37 degrees C) and water activity (aw: 0.99-0.87) and their interactions on the germination rates, lag times prior to germination and mycelial growth 'in vitro' of Penicillium digitatum, P. italicum and Geotrichum candidum, the main postharvest pathogens affecting citrus fruits. METHODS AND RESULTS: Germination and growth were markedly influenced by temperature and aw. Generally, lag times were longer and germination and growth rates were slower when conditions of temperature and aw were far from optimum. All the studied species were able to germinate over a range of 4-30 degrees C at 0.995 aw, although in non-optimal conditions P. digitatum only reached 40-60% of germinated conidia. At low temperatures, P. italicum germinated and grew faster than P. digitatum and G. candidum, particularly at 0.95 aw. Penicillium italicum was also able to germinate and grow in the driest studied conditions (0.87 aw), while G. candidum did not germinate under 0.95 aw. CONCLUSIONS: Knowledge of the ecological requirements of these fungi is important in order to understand their behaviour in natural situations and to predict fungal spoilage on citrus fruits.  相似文献

Light scattering and viscometric studies have been carried out on two preparations, A and B, of rooster comb hyaluronate. Sedimentation rate studies have also been performed with A. Light scattering measurements in 0.2 m KCl for preparation A gave a molecular weight of 3.3 × 106 and for B, 1.0 × 106. In (0.1–0.3) M NaCl similar measurements gave a particle weight for A of (4.4–6.4 × 106 and for B (1.7–2.8 × 106. In 0.066 m CaCl2 molecular weight values of 9.5 × 106 for A and 1.7 × 106 for B were obtained. Thus in the presence of Na+ and Ca2+ ions aggregates of chains persisted into dilute solution. Measurements by light scattering on A and B in 4 m guanidinium chloride gave values in the same range as those obtained in 0.2 m KCl. Sedimentation rate studies on A gave values of 10.3 Svedbergs in 0.2 m KCl and 12.2 Svedbergs in 0.2 m NaCl and 0.066m CaCl2. The shear dependence of the viscosity was studied using a conicylindrical viscometer at shear rates between 0.5 and 20 s?1. Preparation A in 0.2 m KCl and NaCl yielded values for (νsp/cc→0 of 5000 and 7100 ml g?1 respectively in keeping with the tendency to aggregate. The behaviour for preparation B was similar. In 0.066 m CaCl2 there was a marked dependence of viscosity on shear speed below 10 s?1 for all concentrations and the value of (νsp/c)→0 at 0 s?1 for preparation A was 7700 ml g?1 while at a shear rate of 8 s?1 (νsp/c)c→0 ? 5000 ml g ?1. Similar effects were found for preparation B and the data suggest associations of chains disruptable by weak shear forces. The increase in viscosity with concentration in the presence of 0.066 m CaCl2 was much less than in the presence of KCl or NaCl, suggesting that the Ca2+ had a marked effect on the ”rigidity’ of the molecules in solution. A viscometric titration experiment with Ca2? showed that a level of 0.02 m CaCl2 in 0.2 m NaCl was sufficient to produce the change in viscosity presented above and that significant perturbations of the viscosity were present at 0.005?0.01 m CaCl2.  相似文献

Phytophthora root and crown rot (Phytophthora cryptogea) on gerbera is difficult to manage because most gerbera cultivars are susceptible to P. cryptogea. This study was conducted in order to determine the in vivo (pot experiment) efficacy of some fungicides and biofungicides. In pot experiments, fungicides were applied 7 days after inoculation with P. cryptogea, while biofungicide was applied 7 days before inoculation. In this study, soil drenches of five fungicides were tested. “Ametoctradin+dimethomorph (100 ml/day),” “mandipropamid+difenoconazole (60 ml/day),” “propamocarb+fosetyl‐Al (200 ml/day),” “mancozeb+metalaxyl‐M (250 g/day)” and “azoxystrobin+difenoconazole (100 ml/day)” active substances were used. Similarly, one biofungicide Bacillus amyloliquefaciens syn. MBI 600 (50 g/100 L) was applied by soil drenching. Efficacy of treatments was assessed according to the percentage of the root system which was visibly rotten at the end of the experiment. Root and crown rot severity was rated on a scale of 0 = 0% root system necrotic, 1 = 1%‐25% necrotic, 2 = 26%‐50% necrotic, 3 = 51%‐75% necrotic and 4 = 76%‐100% necrotic from 12 to 21 days. In this experiment, “azoxystrobin 200 g/L + difenoconazole 125 g/L” exhibited the highest efficacy against P. cryptogea with a ratio of 43.75%. The other fungicides and biofungicides ametoctradin 300 g/L + dimethomorph 225 g/L, mandipropamid 250 g/L + difenoconazole 250 g/L, propamocarb 530 g/L + fosety‐Al 310 g/L, mancozeb 64%+metalaxyl‐M 4% and Bacillus amyloliquefaciens syn. MBI 600 11% were ineffective. Importance should be given to management strategies of P. cryptogea of and more experiments should be carried out for a better understanding of the use of registered fungicides and biofungicides.  相似文献

The early floral development of Actinidia (A. arguta, A. callosa, A. chinensis and A. kolomikta; Actinidiaceae), Saurauia (S. montana, S. oldhamii, S. pittieri and S. subspinosa; Actinidiaceae), Roridula gorgonias (Roridulaceae) and Heliamphora nutans (Sarraceniaceae) was studied comparatively using scanning electron microscopy. Late stages of androecium development are additionally presented for Clematoclethra scandens (Actinidiaceae), Darlingtonia californica and Sarracenia leucophylla (Sarraceniaceae). Flowers are typically pentamerous and share a number of developmental features: perianth organs emerge in a clockwise or anticlockwise spiral sequence on the floral apex with relatively long plastochrons between successive organs, resulting in conspicuous size differences among perianth organs in early development; the perianth always consists of two differentiated whorls (unlike earlier interpretations of the perianth in Heliamphora); the androecium is polystemonous in most species and is initiated with leading stamens in alternipetalous positions; successive stamen primordia appear in a lateral succession until a ring‐like structure is formed; and the anthers become inverted shortly before anthesis. Later androecial development differs conspicuously between taxa and further proliferation may be centrifugal, centripetal and/or lateral. For Ericales, unusual features of floral development include: petals initiated in a spiral sequence (but later organized in a whorl) with comparatively long plastochrons between individual petals (except Saurauia); common occurrence of perianth organs in double positions in Actinidiaceae; and anthers that become inverted close to anthesis. The floral development in the sarracenioids is additionally compared with that of other families and clades in Ericales, further emphasizing the highly variable patterns of androecium development in the order.  相似文献
