Drinking water contaminated with arsenic poses serious threat to the human health. The present study was aimed for quality assessment of the groundwater of Khairpur Mir's in respect with arsenic and other elemental contamination like Fe, Cu, Co, and Ni. The presence of the trace elements in groundwater from different sources in the study area was measured by using atomic absorption spectroscopy. For arsenic analysis hydride generation technique (MHS-15) was used with detection limit of 0.02 µg l?1. Elevated level of arsenic was observed in most of the samples as compared to recommended value of World Health Organization (WHO) guidelines (10 µg l?1). However, levels of Fe, Cu, Co, and Ni in hand pump (HP) water samples was found in the range of 4–1610 µg l?1, 0–556 µg l?1, 0–230 µg l?1, and 0–700 µg l?1, respectively. Whereas in tube well (TW) water samples the observed values are 5–1620 µg l?1, 0–50 µg l?1, 4–110 µg l?1, and 0–360 µg l?1 for Fe, Cu, Co, and Ni, respectively. Significant difference was observed between TW and HP water samples. It was concluded that the level of arsenic found was very high up to 13 fold more than the WHO recommended limit in study area. While the levels of other elements was noted within the safe limit.  相似文献

Field-measured grazing rates (ml/animal/d) of cladocerans (mostly daphniids) and diaptomids were assembled from various published studies and plotted as a function of corresponding phytoplankton concentration (μg l−1 f.w.). Filtering rates of both zooplankton groups initially increased with seston concentration until maximal grazing rates were observed at approximately 4 × 102 and 1 × 102 μg l−1 for cladocerans and copepods, respectively; at higher algal concentrations, filtering rates of both declined as a function of food concentration. The shape of these curves are most consistent with Holling's (1966) Type 3 functional response. We found little support for the Type 3 functional response in published laboratory studies of Daphnia; most investigators report either a Type 1 or Type 2 response. The one study in which the Type 3 response was observed involved experiments where animals were acclimated at low food concentrations for 24 h, whereas those studies associated with response Types 1 or 2 had acclimation periods of only 1 to 3 h. We therefore assembled relevant data from the literature to examine the effect of acclimation period on the feeding rates of Daphnia at low food concentrations. In the absence of any acclimation, animals filtered at extremely low rates. After 2 h of acclimation, however, filtering rates increased 4 to 5-fold but declined again with longer durations; after > 70 h of pre-conditioning, filtering rates were almost as low as they had been with no acclimation. We also found little support for the Type 3 functional response in published studies of copepods. The only study associated with a Type 3 response involved a marine copepod that had been subjected to a starvation period of 48 h; however, an analysis of the effects of acclimation period did not yield conclusive evidence that filtering rates of freshwater copepods (Diaptomus and Eudiaptomus) decrease significantly with acclimation duration. The low filtering rates associated with long acclimation periods in laboratory experiments appears to be a direct result of animals becoming emaciated from prolonged exposure to low food concentrations, a situation which renders them incapable of high filtering rates. This may explain the Type 3 functional response for field cladocerans, since zooplankton in food-limiting situations are constantly exposed to low food concentrations, and would therefore have low body carbon and consequently less energy to filter-feed. We cannot, however, use this to explain the Type 3 response for field diaptomids, since copepods in the laboratory did not appear to lose body carbon even after 72 h of feeding at very low food levels, and there was inconclusive evidence that either Diaptomus or Eudiaptomus decrease their filtering rates with acclimation period. Although Incipient Limiting Concentrations (ILC) for Daphnia ranged from 1 to 8.5 × 103 μg 1−1, more than half of these fell between 1 and 3 × 103 μg l−1, bracketing the value of 2.7 × 102 μg l−1 for field cladocerans. There was, however, a great deal of variation in reported maximum ingestion rates (MIR), maximum filtering rates (MFR) and ILC values for Daphnia magna. ILC values from the few laboratory studies of freshwater copepods ranged between 0.5 to 2.8 × 103 μg 1−1, and was higher than the ILC value of approximately 0.2 × 103 μg l−1 calculated for field populations of D. minutus. Generally, there was considerable agreement among laboratory studies regarding the shape of grazing-rate and ingestion-rate curves when data were converted to similar units and presented on standardized scales.  相似文献

Geographic and vertical variations of size-fractionated (0.2–1μm, 1–10 μm, and >10 μm) Chlorophyll a (Chl.a) concentration, cyanobacteria abundance and heterotrophic bacteria abundance were investigated at 13 stations from 4°S, 160°W to 30°N, 140°E in November 1993. The results indicated a geographic distribution pattern of these parameters with instances of high values occurring in the equatorial region and offshore areas, and with instance of low values occurring in the oligotrophic regions where nutrients were almost undetectable. Cyanobacteria showed the highest geographic variation(ranging from 27×103 to 16,582×103cell l-1), followed by Chl.a (ranging from 0.048 to 0.178μg l-1), and heterotrophic bacteria (ranging from2.84×103 to 6.50 ×105 cell l-1). Positive correlations were observed between nutrients and Chl.a abundance. Correspondences of cyanobacteria and heterotrophic bacteria abundances to nutrients were less significant than that of Chl.a. The total Chl.a was accounted for 1.0–30.9%, 35.9–53.7%, and 28.1–57.3% by the >10μm, 1–10 μm and 0.2–1 μm fractions respectively. Correlation between size-fractionated Chl.a and nutrients suggest that the larger the cell size, the more nutrient-dependent growth and production of the organism. The ratio of pheophytin to chlorophyll implys that more than half of the >10 μm and about one third of the 1–10 μm pigment-containing particles in the oligotrophic region were non-living fragments, while most of the 1–10 μm fraction was living cells. In the depth profiles, cyanobacteria were distributed mainly in the surface layer, whereas heterotrophic bacteria were abundant from surface to below the euphotic zone. Chl.a peaked at the surface layer (0–20 m) in the equatorial area and at the nitracline (75–100 m) in the oligotrophic regions. Cyanobacteria were not the principle component of the picoplankton. The carbon biomass ratio of heterotroph to phytoplankton was greater than 1 in the eutrophic area and lower than 1 in oligotrophic waters. Environmental relevant concentrations of glyphosate‐based herbicide as 50 µg l?1, 300 µg l?1 and 1800 µg l?1 can affect sperm quality of yellowtail tetra fish Astyanax lacustris . Viability of sperm cells was impaired at 300 µg l?1, a concentration that is within legal limits in U.S.A. waterbodies, while motility was impaired at 50 µg l?1, which is the more stringent limit set in Brazilian law. Concentrations of total and inorganic phosphate were measured in forty-seven sediments of the River Garonne (France) using an anion resin, NaOH 0.1 M, Ca-NTA and Na-EDTA. Algal available P was measured using Scenedesmus crassus. The sediments presented a wide range of Tot-P concentrations (226 to 923 μg g−1). Resin P varied between 1.1 to 55.5 μg g−1 and NaOH-P between 3.5 to 262 μg g−1. NTA-P and EDTA-P varied between 13.9 and 178 μg g−1 and between 14.4 to 261 μg g−1 respectively. Algal available P varied between 8.8 to 262 μg g−1/ All forms of P are highly correlated and specially with algal available P (r = 0.70, 0.77, 0.88 and 0.77 for resin-P, NaOH-P, NTA-P and EDTA-P respectively). Nevertheless the partial correlation between EDTA-P and algal available P dropped to 0.05 when the concentration of NTA-P was considered as constant, indicating a spurious correlation with apatitic phosphate (extracted by Na-EDTA). Because of the strongest R 2 and the lowest standard error of estimate, phosphate extracted by Ca-NTA can be considered as the best predictor of algal available phosphate.  相似文献

Effects and accumulation of cadmium were studied in unialgal 10-1 batch-culture experiments with the dinoflagellateProrocentrum micans Ehrenberg. Tests were made using sterile filtered North Sea water enriched with nitrate and phosphate only in order to avoid disturbances by complex formation. Cadmium was added to the cultures in amounts of 100 to 0.13μg l−1. In one series it was added at the start of the experiments and in a second one after a growth period of 1 week. Addition of only 1.2μg Cd l−1 reduces multiplication rates and maximum cell densities of the algae. Not until 0.4μg Cd−1 does growth correspond to that of the controls. Cadmium concentrations were measured, after filtration, in the culture medium and in the biomass by means of flameless AAS. The cadmium content in algae increased from 2.7μg g−1 (dry weight) in controls to 500μg g−1 (dry weight) in media containing 100μg Cd l−1. Uptake occurred rapidly during the first few days of the experiments, slowed down somewhat during exponential growth stage, and increased during decay of the cultures. Cadmium content of culture media remained nearly constant (Series 1) or decreased only slowly during experimental time (Series 2). The highest concentration factor was measured in the controls. It decreased with increasing metal concentration in the medium and increased with experimental time. Structural modifications of the cells were visible after Lugol fixation only, indicating brittleness of the cell walls.P. micans has shown to be extremely sensitive to cadmium and to accumulate this metal.  相似文献

The spatial and temporal variability of phytoplankton abundance (in terms of chlorophyll a and cell number), inorganic nitrogen, suspended particulate matter (SPM), and light availability was determined throughout one year in the Tagus estuary, Portugal. Chlorophyll a concentrations showed a strong seasonal variation with values ranging from 1 to 32 μg l?1(average 5.4 μg l?1). Chlorophyll patterns were unimodal for sites 1, 2, and 3 and bimodal for site 4. Diatoms and cryptophytes were, throughout the year, the dominant groups in this shallow and unstratified estuarine system. Nitrate concentrations were seasonally related to river flow and ammonium concentrations spatially related to sources of sewage input. Lower river inputs and long water residence times during summer initially promoted the accumulation of phytoplankton, but the resulting low dissolved inorganic nitrogen (DIN) concentrations lead to limitation of phytoplankton growth. Chlorophyll a and DIN values obtained in the present study were comparable to those reported 20 years ago for the Tagus estuary.  相似文献

The permanently eutrophic Sundays Estuary experiences recurrent harmful algal blooms (HABs) of Heterosigma akashiwo (Raphidophyceae). This study aimed to identify the environmental variables shaping phytoplankton community composition and succession patterns during a typical spring/summer harmful algal bloom (HAB) period. Monitoring of abiotic and phytoplankton variables was undertaken over the period of a month in 2016. Surface water salinity corresponding to mesohaline conditions (9 to 12) was a prerequisite for site selection. During the study, two HABs (>550 µg Chl a l?1) of H. akashiwo occurred, each lasting for approximately a week in duration. Analyses highlighted nutrient depletion (i.e. nitrate and phosphate concentrations) as the key constraint on bloom duration. When the density of H. akashiwo decreased, the community composition became more diverse with species belonging to Bacillariophyceae and Dinophyceae becoming more abundant; albeit to a lesser degree (<180 µg Chl a l?1). Dissolved oxygen shifted from super-saturated conditions (>14 mg l?1) during peak HAB conditions, to instances of bottom water oxygen depletion (2–4 mg l?1) during the decay phase. These findings highlight the potential severity of transforming a catchment from natural to one that is highly regulated by agricultural practices, while also emphasising the need for management intervention.  相似文献

Using rainbow trout Oncorhynchus mykiss, the present study demonstrated that: (1) calcium (Ca) increased the range of copper (Cu) concentrations that O. mykiss avoided; (2) Ca conserved the maintenance of pre‐exposure swimming activity during inescapable acute (10 min) Cu exposure. Data showed that when presented with a choice of Cu‐contaminated water (ranging from 0 to 454 µg Cu l?1) and uncontaminated water in a choice tank, O. mykiss acclimated and tested at low Ca concentration (3 mg Ca l?1) avoided the 10 µg Cu l?1 only. By contrast, O. mykiss acclimated and tested at high Ca concentration (158 mg Ca l?1) avoided all the Cu concentrations ≥37 µg l?1. The Cu avoidance was connected with increased spontaneous swimming speed in the Cu‐contaminated water. When subjected to inescapable Cu exposure (35 µg Cu l?1), O. mykiss acclimated and tested at low Ca concentration reduced their spontaneous swimming speed, whereas no response was observed in O. mykiss acclimated and tested at high Ca concentration. Collectively, the data support the conclusion that in O. mykiss the behavioural responses to acute Cu exposure are Ca‐dependent.  相似文献

To investigate the effect of fungal elicitors on biosynthesis of natamycin in the cultures of Streptomyces natalensis HW-2, the biomass and filtrate of the broth from Aspergillus niger AS 3.6472, Penicillium chrysogenum AS 3.5163, A. oryzae AS 3.2068, and Saccharomyces cerevisiae AS 2.2081 were used to induce natamycin production in S. natalensis HW-2. The results showed that the biomass of P. chrysogenum AS 3.5163 could enhance the yield of natamycin from 0.639 to 0.875 g?l?1. The elicitor from the fermentation broth of P. chrysogenum AS 3.5163 showed the highest inducing efficiency with the yield of natamycin enhanced from 0.632 to 1.84 g?l?1. The elicitor that was cultured for 2 days showed the strongest inducing activity during the fermentation of S. natalensis HW-2 for 24 h, and the yield of natamycin was enhanced from 0.637 to 2.12 g?l?1. The biochemical parameters were examined at the end of fermentation and the results demonstrated that both the growth of cells and the concentration of residual sugar could be influenced. The residual sugar decreased from 5.03 to 4.27 g?l?1, and the biomass decreased from 10.26 to 6.87 g?l?1. Finally, the elicitor was identified as a low molecular weight substance with a similar polarity to that of butyl alcohol by primary qualitative analysis.  相似文献

Lake Timsah is considered as the biggest water body at Ismailia City with a surface area of 14?km2. It is a saline shallow water basin lies approximately mid-way between the south city of Suez and the north city of Port Said at 30o35′46.55“N and 32o19′30.54″E. Because it receives water with high and low salinities, salinity stratification is producing in the Lake Timsah, with values of 14–40‰ for the surface water and over 40‰ for the bottom water. The temperature of the lake water decreased to below 19 °C in the winter and rose to above 29?°C in the summer; the concentration of dissolved oxygen ranged between 6.5 and 12.2?l?1 and the pH fluctuated between 7.9 in its lower value and 8.2 in its higher value. Water transparency was very low as indicated by Secchi disc readings recorded during this study and varied between 0.3 and 2.7?m. The main chemical nutrient (phosphorus) reached its highest levels of 96?µg?l?1 in winter and their lowest values of 24?µg?l?1 during summer. This nutrient concentration is high especially by comparing with those of unpolluted marine waters, but is typical of the more eutrophic coastal waters worldwide. The composition and abundance of phytoplankton with dominancy of diatoms and increased population density (20,986 cell l?1) reflect the eutrophic condition of the lake. The intensive growth of phytoplankton was enriched by high concentration of chlorophyll a with annual values ranged between 6.5 and 56?µg?l?1. The objective of the present work was quantitative assessment of the quality of the water of the Lake Timsah using different approaches. During the present study, three different approaches were applied for the quantitative assessment of Lake Timsah water quality: the trophic state index (TST); trophic level index (TLI) and water quality index (WQI). Application of the trophic state and trophic level indices (TSI & TLI) revealed that the Lake Timsah has trophic indices of 60 and 5.2 for TSI and TLI, respectively. Both indices reflected the eutrophic condition of the lake waters and confirmed that the eutrophication is a major threat in the Lake Timsah. On the other hand, the WQI calculated for the Lake Timsah during the present study with an average of 49 demonstrated that the water of the Lake Timsah is bad and unsuitable for main and/or several uses. Moreover, WQI allows accounting for several water resource uses and can serve a more robust than TSI and/or TLI and can be used effectively as a comprehensive tool for water quality quantification. In conclusion, the three subjective indices used for the assessment process for the lake water are more suitable and effective for needs of the sustainable water resources protection and management of the Lake Timsah.  相似文献

Recombinant Escherichia coli, expressing the oleate hydratase gene of Stenotrophomonas maltophilia, was permeabilized by sequential treatments with 0.125 M NaCl and 2 mM EDTA. The optimal conditions for the production of 10-hydroxy-12,15(Z,Z)-octadecadienoic acid from α-linolenic acid by permeabilized cells were 35 °C and pH 7.0 with 0.1 % (v/v) Tween 40, 50 g permeabilized cells l?1, and 17.5 g α-linolenic acid l?1. Under these conditions, permeabilized cells produced 14.3 g 10-hydroxy-12,15(Z,Z)-octadecadienoic acid l?1 after 18 h, with a conversion yield of 82 % (g/g) and a volumetric productivity of 0.79 g l?1 h?1. These values were 17 and 168 % higher than those obtained by nonpermeabilized cells, respectively. The concentration, yield, and productivity of 10-hydroxy-12,15(Z,Z)-octadecadienoic acid obtained by permeabilized cells are the highest reported thus far.  相似文献

Purpose of work

To establish pilot scale bioreactor cultures of somatic embryos of Siberian ginseng for the production of biomass and eleutherosides. Somatic embryos of Eleutherococcus senticosus were cultured in airlift bioreactors using Murashige and Skoog medium with 30 g sucrose l?1 for the production of biomass and eleutherosides. Various parameters including the type of bioreactor, aeration volume, and inoculum density were optimized for 3 l capacity bioreactors. Balloon-type airlift bioreactors, utilizing a variable aeration volume of 0.1–0.3 vvm and an inoculum of 5 g l?1, were suitable for biomass and eleutheroside production. In 500 l balloon-type airlift bioreactors, 11.3 g dry biomass l?1, 220 µg eleutheroside B l?1, 413 µg eleutheroside E l?1, and 262 µg eleutheroside E1 l?1 were produced.  相似文献

Coscinodiscus granii Gough was cultivated at low cell densities in aged Atlantic sea water containing very little dissolved organic carbon; the water was enriched with low levels of nutrients but no chelators were added. Cadmium additions provided final concentrations of 0.1 to 26.5 μg Cd l−1, zinc being kept constant at a level of 38 μg Zn l−1. Carrier-free109Cd and65Zn were used as tracers for the two metals. Growth in terms of cell numbers and primary productivity capacity, using the14C uptake rate, was followed during the exponential growth phase for the first 5 days of the experiment and then for a further 3 days during the stationary phase. On each day, the metal contents of the cells were determined. Cadmium concentrations of 20 μg Cd l−1 and more resulted in growth inhibition whereas 17.5 μg Cd l−1 reduced the growth only slightly. The physiological state of the cells influenced the heavy-metal uptake per cell at sublethal Cd levels. Dead cells had a higher heavy-metal concentration than living cells. Microscopical observations revealed that cells just about to divide were less sensitive to a given toxic heavy-metal concentration than cells which had recently divided. This might have been due to different surface/volume ratios.  相似文献

The winter dynamics of several chemical, physical, and biological variables of a shallow, polymictic lake (Opinicon) are compared to those of a deep, nearby dimictic lake (Upper Rock) during ice cover (January to early April) in 1990 and 1991. Both lakes were weakly inversely thermally stratified. Dissolved oxygen concentration was at saturation (11–15 mg l−1) in the top 3 m layer, but declined to near anoxic levels near the sediments. Dissolved oxygen concentrations in the deep lake were at saturation in most of the water column and approached anoxic levels near the sediments only. Nutrient concentrations in both lakes were fairly high, and similar in both lakes during ice cover. Total phosphorus concentrations generally ranged between 10–20 μg l−1, NH4-N between 16–100 μg l−1, and DSi between 0.9–1.9 mg l−1; these concentrations fell within summer ranges. NO3-N concentrations were between 51–135 μg l−1 during ice cover, but occurred at trace concentrations (<0.002 μg l−1) during the summer. The winter phytoplankton community of both lakes was dominated by flagellates (cryptophytes, chrysophytes) and occasionally diatoms. Dinoflagellates, Cyanobacteria and green algae were poorly represented. Cryptophytes often occurred in fairly high proportions (20–80%) throughout the water column, whereas chrysophytes were more abundant just beneath the ice. Zooplankton population densities were extremely low during ice cover (compared to maximum densities measured in spring or summer) in both lakes, and were comprised largely of copepods.  相似文献

Inter- and intra-population variation in the toxicity of the antifouling biocide copper pyrithione (CuPT) was examined for nauplius larvae of the barnacle Balanus amphitrite. Nauplii were collected from brooding adults from four sites within the Newport River estuary (NC), chosen based on an initial estimation of recent and historical human activities that affect local contamination levels. Each site was characterized for the presence of polycyclic aromatic hydrocarbons and for the frequency of gastropod imposex, an indicator of contamination by organotins. Sensitivity of nauplii to CuPT varied significantly across the sites/populations, with LC50 values ranging from 4.0 μg l?1 to 6.1 μg l?1. Larvae from the most contaminated site were the most sensitive to CuPT. Intrapopulation variation in toxicity was investigated by exposing nauplius larvae from 15 maternal families to a fixed concentration of CuPT (6.1 μg l?1). Variation in larval mortality among the families was significant, ranging from 15.1% to 98.9%.  相似文献

Objective

To produce δ-decalactone from linoleic acid by one-pot reaction using linoleate 13-hydratase with supplementation with whole Yarrowia lipolytica cells.

Results

Whole Y. lipolytica cells at 25 g l?1 produced1.9 g l?1 δ-decalactone from 7.5 g 13-hydroxy-9(Z)-octadecenoic acid l?1 at pH 7.5 and 30 °C for 21 h. Linoleate 13-hydratase from Lactobacillus acidophilus at 3.5 g l?1 with supplementation with 25 g Y. lipolytica cells l?1 in one pot at 3 h produced 1.9 g l?1 δ-decalactone from 10 g linoleic acid l?1 via 13-hydroxy-9(Z)-octadecenoic acid intermediate at pH 7.5 and 30°C after 18 h, with a molar conversion yield of 31 % and productivity of 106 mg l?1 h?1.

Conclusion

To the best of our knowledge, this is the first production of δ-decalactone using unsaturated fatty acid.

Optimum growth conditions for the fermentation of non-concentrated whey permeate by Kluyveromyces fragilis NRRL 665 have been defined. Use of 3.75 g yeast extract l?1, a growth temperature of 38°C and a pH of 4.0 allowed a maximum productivity of 5.23 g ethanol l?1 h?1 in continuous culture with a yield 91% of theoretical. Complete batch fermentation of permeate with 100 g lactose l?1 was possible with a maximum specific growth rate of 0.276 h?1 without any change in ethanol yield. Fermentation of concentrated permeate resulted, however, in a general decrease of specific substrate consumption rate, demonstrated by the inability to completely convert an initial 90 or 150 g lactose l?1 in continuous culture, even at dilution rates as low as 0.05 and 0.08 h?1, respectively. The decrease could be related to substrate inhibition, to an increase in osmotic pressure caused by lactose and salts, and to ethanol inhibition of both alcohol and biomass yield. The decrease in specific productivity could be counterbalanced by use of high cell density cultures, obtained by cell recycle of K. fragilis. Fermentation of a non-concentrated permeáte at a dilution rate of 1 h?1 resulted in a productivity of 22 g l?1 h?1 at 22 g ethanol l?1. Cell recycle using flocculating Kluyveromyces lactis NCYC 571 was also tested. With this strain a productivity of 9.3 g l?1 h?1 at 45 g product l?1 was attained at a dilution rate of 0.2 h?1, with an initial lactose concentration of 95 g l?1.  相似文献

Phytoplankton ecology of Loch Ard,Scotland   总被引:1,自引:1,他引:0  
The results of the ecological investigation discussed in this paper represent the first seasonal study of the phytoplankton to be carried out in Loch Ard. This body of water is a typical warm monomictic lake, with water temperatures during the period of study ranging from 18.6 to 4.4 °C and oxygen saturation from 68 to 112%. Stratification lasted from May to November. Phosphate was generally very low, being almost undetectable for about six months and not exceeding 0.125 µg.at.P 1–1 even at the winter maximum. Dissolved silica showed a regular seasonal pattern ranging from 9 to 21 µg.at.Si 1–1. Nitrate, like silicate, never showed signs of complete exhaustion, with a minimum of 4.0 and a maximum of 21.0 µg.at.N 1–1. The pH remained on the acid side of neutrality while the alkalinity never exceeded 7 mg CaCO3/1.From an examination of net phytoplankton Loch Ard could be described as diatom-desmid in nature. β-Glucosidase from Thermus thermophilus has specific hydrolytic activity for the outer glucose at the C-20 position in protopanaxadiol-type ginsenosides without hydrolysis of the inner glucose. The hydrolytic activity of the enzyme for gypenoside XVII was optimal at pH 6.5 and 90 °C, with a half-life of 1 h with 3 g enzyme l?1 and 4 g gypenoside XVII l?1. Under the optimized conditions, the enzyme converted the substrate gypenoside XVII to ginsenoside F2 with a molar yield of 100 % and a productivity of 4 g l?1 h?1. The conversion yield and productivity of ginsenoside F2 are the highest reported thus far among enzymatic transformations.  相似文献
