The interactions of N‐acetyl‐L‐cysteine‐capped CdTe quantum dots (QDs) with bovine serum albumin (BSA) and bovine hemoglobin (BHb) were investigated by isothermal titration calorimetry (ITC), fluorescence, synchronous fluorescence, fluorescence lifetime, ultraviolet–visible absorption, and circular dichroism techniques. Fluorescence data of BSA–QDs and BHb–QDs revealed that the quenching was static in every system. While CdTe QDs changed the microenvironment of tryptophan in BHb, the microenvironment of BSA kept unchanged. Adding CdTe QDs affected the skeleton and secondary structure of the protein (BSA and BHb). The ITC results indicated that the interaction between the protein (BSA and BHb) and QDs‐612 was spontaneous and the predominant force was hydrophobic interaction. In addition, the binding constants were determined to be 1.19 × 105 L mol?1 (BSA–QDs) and 2.19 × 105 L mol?1 (BHb–QDs) at 298 K. From these results, we conclude that CdTe QDs have a larger impact on the structure of BHb than BSA.  相似文献

本研究旨在对罗丹明类荧光探针ZM-6与人血清白蛋白(HSA)的相互作用进行研究。采用了荧光光谱法、三维荧光光谱法、同步荧光光谱法以及CD光谱法在模拟生理条件下对二者的相互作用以及HSA的构象进行了研究。研究结果表明,探针与ZM-6之间的猝灭机理主要是静态猝灭方式。根据热力学数据确定了二者之间的作用力,类型为范德华力和氢键。二者之间的结合距离为4.45 nm。同时得出,ZM-6对HSA的构象产生了影响。此研究对于探针分子的设计以及修饰提供有效的数据以及理论支持。  相似文献

采用紫外和荧光光谱研究了水溶性壳聚糖(CS)与牛血清白蛋白(BSA)之间的相互作用。结果表明:随CS浓度的增加,BSA的紫外吸收光谱表现出明显的增色效应和较小的紫移;CS可以猝灭BSA的内源荧光,其猝灭机理是CS与BSA形成复合物的静态猝灭。并且测定了在不同温度下,该反应的结合常数KA分别为6.92×106(298 K),5.01×106(308 K),3.31×106(318 K),CS与BSA以摩尔比1∶1结合。同时采用同步荧光光谱法探讨了CS对BSA构象的影响。  相似文献

The interaction of meso-tetra(4-N-hydroxyethylpyridyl)porphyrin, meso-tetra(3-N-hydroxyethylpyridyl)porphyrin, and their zinc complexes with bovine serum albumin (BSA) was studied by electronic spectroscopy, CD, and equilibrium dialysis at pH 7.2. The titration of the porphyrins with BSA was accompanied by a decrease in light absorption and a bathochromic shift of the Soret band, as well as by the appearance of an isobestic point. The porphyrin interaction with BSA also led to the induction of positive CD spectra in the visible region, which is explained by the porphyrin sorption on the protein globule. The equilibrium dialysis helped in determining the stoichiometry of binding and the binding constants of the porphyrins under study with BSA using Scatchard plots. This interaction is nonspecific and reversible.  相似文献

To research the mechanism of dimercaptosuccinic acid coated‐superparamagnetic iron oxide nanoparticles (SPION) with human serum albumin (HSA), the methods of spectroscopy, molecular modeling calculation, and calorimetry were used in this paper. The inner filter effect of the fluorescence intensity was corrected to obtain the accurate results. Ultraviolet–visible absorption and circular dichroism spectra reflect that SPION changed the secondary structure with a loss of α‐helix and loosened the protein skeleton of HSA; the activity of the protein was also affected by the increasing exposure of SPION. Fluorescence lifetime measurement indicates that the quenching mechanism type of this system was static quenching. The isothermal titration calorimetry measurement and molecular docking calculations prove that the predominant force of this system was the combination of Van der Waals’ force and hydrogen bonds.  相似文献

在模拟动物体生理条件下,用荧光猝灭、荧光偏振和紫外-可见吸收光谱法研究了槐定碱与牛血清白蛋白(BSA)结合作用。荧光猝灭数据显示,槐定碱与BSA发生反应生成了新的复合物,属于静态荧光猝灭。求出了不同温度(19、25、31、37℃)下槐定碱与BSA作用的结合常数分别为1.219×106,1.164×106,1.110×106和1.057×106L/mol,由van’tHoff方程式计算槐定碱与BSA反应的热力学参数:焓变ΔH和熵变ΔS值分别为-5.97kJ/mol和96.11J/(mol.K),表明槐定碱与BSA间的作用力以静电引力为主。以华法林和布洛芬(分别为siteI和siteII探针)为标记药物研究槐定碱在BSA上的结合位点,结果表明,槐定碱结合在BSA疏水空腔的siteI位点。  相似文献

The protein-surfactant system constituted by bovine serum albumin (BSA) and N-decanoyl-N-methylglucamide (MEGA-10) has been studied by using surface tension, steady-state fluorescence, and dynamic light scattering measurements. It was found that the presence of protein delays the surfactant aggregation, which was interpreted as a sign of binding between surfactant and protein. Binding studies were carried out by two different methods. First, a treatment based on surface tension measurements was used to obtain information on the number of surfactant molecules bound per protein molecule under saturation conditions. Second, the binding curve for the BSA/MEGA-10 system was determined by examining the behavior of the intrinsic BSA fluorescence upon the surfactant addition. Both approaches indicate that the binding process is essentially cooperative in nature. The results of the aggregation numbers of MEGA-10 micelles, as well as those of resonance energy transfer from tryptophan residues to 8-anilinonaphthalene-1-sulfonate, corroborate the formation of micelle-like aggregates of surfactants, smaller than the free micelles, adsorbed on the protein surface. The dynamic light scattering results were not conclusive, in the sense that it was not possible to discriminate between protein-surfactant complexes and free micelles. However, the overall results suggest the formation of "pearl necklace" complexes in equilibrium with the free micelles of the surfactant.  相似文献

合成了壳寡糖和稀土离子La3+的配合物,利用红外光谱、紫外光谱和差热-热重手段对其结构和性质进行了表征。采用抑菌圈法考察了壳寡糖、壳寡糖-La对革兰氏阳性菌金黄色葡萄球菌和革兰氏阴性菌大肠杆菌的抑菌活性。此外,紫外光谱、荧光光谱和循环伏安曲线法研究壳寡糖-La与牛血清白蛋白(BSA)相互作用。结果表明壳寡糖、壳寡糖-La对两种细菌均具有较强的抑菌活性,且壳寡糖-La的抑菌活性强于壳寡糖;壳寡糖-La使BSA的内源荧光猝灭,猝灭机制为静态猝灭,并计算了室温下壳寡糖-La与BSA的结合常数和结合位点数分别为6.35×104L/mol和1.29。  相似文献

利用光谱探针技术研究在酸性溶液中考马斯亮蓝G-250(Coomassie brilliant blue G-250,CBBG)与牛血清白蛋白(bovine serum albumin,BSA)相互作用机理,考察了不同实验条件对CBBG-BSA复合物吸收光谱的影响。实验结果表明:CBBG与BSA相互作用产生光谱蓝移主要是由CBBG与BSA间的疏水相互作用引起,而静电作用则是形成CBBG-BSA蓝色复合物的必要条件。同时,CBBG聚集体的聚集程度是影响CBBG-BSA蓝色复合物形成的重要因素。  相似文献

The interaction of ceftriaxone sodium (CS), a cephalosporin antibiotic, with the major transport protein, bovine serum albumin (BSA), was investigated using different spectroscopic techniques such as fluorescence, circular dichroism (CD), and UV–vis spectroscopy. Values of binding parameters for BSA–CS interaction in terms of binding constant and number of binding sides were found to be 9.00 × 103, 3.24 × 103, and 2.30 × 103 M?1 at 281, 301, and 321 K, respectively. Thermodynamic analysis of the binding data obtained at different temperatures showed that the binding process was spontaneous and was primarily mediated by van der Waals force or hydrogen bonding. CS binding to BSA caused secondary structural alterations in the protein as revealed by CD results. The distance between CS and Trp of BSA was determined as 3.23 nm according to the Förster resonance energy transfer theory. © 2012 Wiley Periodicals, Inc. The interaction between copper (II) 2-oxo-propionic acid salicyloyl hydrazone (CuIIL) and bovine serum albumin (BSA) under physiological conditions was investigated by the methods of fluorescence spectroscopy, UV-Vis absorption, and circular dichroism spectroscopy. Fluorescence data showed that the fluorescence quenching of BSA by CuIIL was the result of the formation of the BSA–CuIIL complex. The apparent binding constants (K a) between CuIIL and BSA at four different temperatures were obtained according to the modified Stern–Volmer equation. The thermodynamic parameters, enthalpy change (ΔH) and entropy change (ΔS), for the reaction were calculated to be ?80.79 kJ mol?1 and ?175.48 J mol?1 K?1 according to van’t Hoff equation. The results indicated that van der Waals force and hydrogen bonds were the dominant intermolecular force in stabilizing the complex. The binding distance (r) between CuIIL and the tryptophan residue of BSA was obtained to be 4.1 nm according to Förster’s nonradioactive energy transfer theory. The conformational investigation showed that the application of CuIIL increased the hydrophobicity of amino acid residues and decreased the α-helical content of BSA (from 62.71% to 37.31%), which confirmed some microenvironmental and conformational changes of BSA molecules.  相似文献

Lead buffers (citrate and Tiron) were used to investigate the effects of low concentrations (0.1-6 microM) of Pb2+ on stimulus-secretion coupling in isolated bovine chromaffin cells. Nicotinic agonists and high K elicit secretion by enhancing Ca2+ influx into chromaffin cells. Pb2+ inhibited the catecholamine secretion in response to 500 microM carbachol and 77 mM K+ depolarization but was without significant effect on basal secretion. Pb2+ also inhibited the influx of 45Ca occurring in response to these agents. The K0.5 values for inhibition suggest that the carbachol-evoked flux is more sensitive to Pb2+ than influx in response to a direct depolarization. When extracellular calcium was lowered in the absence of Pb2+, both secretion and 45Ca entry were reduced. The effects of Pb2+ were comparable to those of lowered Ca2+. 22Na influx through nicotinic receptor-mediated channels, measured in the presence of tetrodotoxin (2 microM) and ouabain (50 microM), was inhibited by Pb2+. The results suggest that Pb2+ inhibits exocytotic catecholamine secretion by inhibiting Ca2+ influx. The differential sensitivity to Pb2+ of K- and carbachol-evoked 45Ca flux, coupled with the 22Na measurements, indicates that Pb2+ inhibits the movement of ions through acetylcholine-induced channels as well as through voltage-sensitive calcium channels.  相似文献

The binding of one fluorine including triazole (C10H9FN4S, FTZ) to bovine serum albumin (BSA) was studied by spectroscopic techniques including fluorescence spectroscopy, UV–Vis absorption, and circular dichroism (CD) spectroscopy under simulative physiological conditions. Fluorescence data revealed that the fluorescence quenching of BSA by FTZ was the result of forming a complex of BSA–FTZ, and the binding constants (K a) at three different temperatures (298, 304, and 310 K) were 1.516?×?104, 1.627?×?104, and 1.711?×?104?mol L?1, respectively, according to the modified Stern–Volmer equation. The thermodynamic parameters ΔH and ΔS were estimated to be 7.752 kJ mol?1 and 125.217 J?mol?1?K?1, respectively, indicating that hydrophobic interaction played a major role in stabilizing the BSA–FTZ complex. It was observed that site I was the main binding site for FTZ to BSA from the competitive experiments. The distance r between donor (BSA) and acceptor (FTZ) was calculated to be 7.42 nm based on the Förster theory of non-radioactive energy transfer. Furthermore, the analysis of fluorescence data and CD data revealed that the conformation of BSA changed upon the interaction with FTZ.  相似文献

本文采用荧光光谱法、紫外光谱法研究在生理条件(pH=7.4)下荷叶中紫云英苷(AST)与牛血清白蛋白(BSA)的相互作用。结果表明AST可与BSA结合并通过静态猝灭作用机制对BSA内源性荧光进行猝灭。在温度为298K及308K时,测得其猝灭速率常数(Kq)分别为4.31×1013L/mol/s和3.72×1013L/mol/s;结合常数(Kd)分别为2.009×105L/mol和0.927×105L/mol;结合位点数(n)分别为0.943和0.893。依据298K时测定的反应自由能变(△G0=-30.25kJ/mol),反应焓变(△H0=-59.02kJ/mol)及反应熵变(△S0=-96.54J/mol/K),结果发现AST与BSA间的结合反应可自发进行且其作用力主要表现为氢键和范德华力。此外,根据Frster非辐射能量转移理论得到AST与BSA之间的结合距离(r)为4.13nm,表明非辐射能量可从BSA转移至AST。  相似文献

由近红外光谱证实,十六烷基三甲基溴化铵(CTAB)/正己醇-正辛烷反胶束溶液是牛血清白蛋白(BSA)增溶于非极性有机溶剂的较理想中介。研究了反萃液酸度、离子强度和种类等参数对BSA反萃率的影响,过低的溶液酸度导致BSA变性。适宜的反萃液为:1.0~2.0mol/L KBr,pH4.3~4.9。蛋白质的紫外光谱表明,BSA分子在料液,反胶束溶液和反萃液中的构象大致相同。此外,探讨了反胶束溶液循环使用的效率问题。最后,通过采用适当的相比,成功地实现了蛋白质的回收和浓缩。  相似文献

从锌诱导的家兔肝脏中分离纯化得到金属硫蛋白两种亚型:ZnMT-Ⅰ和ZnMT-Ⅱ.在酸性条件下脱金属,经Sephadex G-25柱层析得到脱金属硫蛋白(apoMTs).用圆二色性(CD)光谱法研究,发现两种亚型apoMTs 与Pb2+ 的结合依赖于Pb2+ 的加入比例及pH 值.apoMT-Ⅰ在pH3~5之间,apoMT-Ⅱ在pH4~6之间与Pb2+ 结合形成特征簇合物Pb7MTs,其CD谱图特征峰位于316nm (- ),270 nm (+ ),245 nm (+ )及225 nm (- ),提示解铅中毒的最佳条件应控制在弱酸性环境.不同亚型apoMTs 与Pb2+ 的结合方式各不相同:Pb2+ 与apoMT-Ⅰ的结合采取平均分配的方式,而与apoMT-Ⅱ则为选择性结合方式,表明这两种亚型在解铅毒功能上存在差异.  相似文献
