Glycollate Formation during the Photorespiration of Acetate by Chlorella   总被引:1,自引:0,他引:1  
WhenChlorella pyrenoidosa photoassimilates 3H-14C-acetate theglycollic acid formed shows a high 3H/14C ratio, the only othercompounds showing similar ratios being glycerate and serine.The 3H/14C ratio of glycollate was unaffected by the TCA cycleinhibitors MFA, diethylmalonate and arsenite showing that 3Hin glycollate does not result from the oxidation of acetatevia the TCA cycle, the resulting NADP3H2 or NAD3H2 being usedfor the reduction of the glycollate precursor. Although DCMUdecreased the 3H/14C ratio, complete inhibition of glycollatelabelling was not observed with 10–6 M DCMU, at whichconcentration complete inhibition of the Hill reaction is achieved.Although the 3H/14C ratio was unaltered, total dpm of both 14Cand 3H in glycollate were increased by INH. The 3H/14C ratiosof glycerate and serine were decreased by INH, as were the totaldpm of 3H and 14C incorporated into these compounds. Thus, INHinhibits the further metabolism of glycollate to glycerate andserine. The effect of INH on incorporation of 14C-I-acetateinto various cell fractions was investigated. The incorporationof 14C into polysaccharide and lipid was decreased, while theincorporation of 14C into the water-soluble fraction of cellsand therelease of 14CO2 were little affected. Although glycollicacid was an early product of acetate photoassimilation in Chlorellapyrenoidosa, glycollate excretion does not take place undera wide range of environmental conditions shown to favour glycollateexcretion by other algae. However, small amounts of labelledglycollate were detected in the supernatant from the cells duringthe photoassimilation of 3H-14C-acetate, but this glycollatedid not show the high 3H/14C ratio of glycollate present withinthe cell. The failure of Chlorella pyrenoidosa to excrete appreciableamounts of glycollate when photoassimilating acetate or carbondioxide was considered to result from the presence of glycollateoxidase (EC 1.1.3.1 [EC] ) which allowed the further metabolism ofglycollate. Besides glycollate oxidase, glyoxylate reductasewas also demonstrated in Chlorella pyrenoidosa so that glycollatecould function in hydrogen transfer during the photoassimilationof acetate.  相似文献

Measurement of carbonic anhydrase (CA) activity in homogenatesof accessory boring organs of the muricid gastropod,Purpuralapillus, by Meldrum and Roughton's manometric method showedthat CA is always present in boring as well as in inactive ABOs,but in variable amounts. Tests by the same method on whole ABOsin an isotonic solution were negative, proving that CA remainsintracellularly. Experiments in vivo on inhibition and activation demonstratedclearly that CA is responsible for demineralization of the valvesof lamellibranchs by Purpura:(a) at low concentrations of Diamox,partial inhibition of the enzymeoccurred, that is, the numberof complete holes decreased or disappeared and etchings increased;and at higher concentrations full inhibition took place. Inhibitionis reversed when snails are replaced in normal sea water; (b)action of pure CO2 or mixtures of CO2 and O2 accelerated boring:in the optimal mixture, three times more boreholes were producedby snails than in the controls, and in about half the time.Under these conditions the reaction catalyzed by CA goes tothe right with hydration of CO2 and is accompanied by releaseof H+ ions; in the presence of an increased concentration ofCO2, the reaction is intensified and results in an additionalrelease of H+ ions. Consequently, destruction of CaCO3 by theABO of Purpura in sea water enriched with CO2 is accelerated. To identify the nature of the exchanged ion, we tested the effectof NaCl and KC1 on the boring mechanism in vivo by increasingthe amount of these salts in the seawater of snails with prey.In this case augmentation of the boring activity was noticed.These results suggest that the boring activity is accompaniedby complex ionic exchanges.  相似文献

The distribution of radioactivity in Chlorella during dark 14CO2fixation was investigated either (a) in normal cells with andwithout added ammonium chloride, or (b) in nitrogen-starvedcells supplied with intermediates of the Krebs-Henseleit ureacycle. In the control experiments almost all the activity was presentin compounds of or associated with, the tricarboxylic acid cycle. The amino-acids citrulline and arginine became radioactive onlyin the presence of ammonia or ornithine where initially theycomprised 40–60 per cent. of the total activity, reactionsof the Krebs–Henseleit urea cycle being implicated intheir formation. No evidence could be found for a complete ureacycle. Unidentified compounds deriving their radioactivity fromthe C4 carbon of citrulline and/or arginine were detected andformed up to 40 per cent. of the total 14CO2 incorporated after25 min.  相似文献

The effect of oxygen on photosynthetic 14CO2 fixation in theair-grown freshwater flagellate Chroomonas sp. (Cryptophyta)was studied. Considerable inhibition by anaerobiosis was observedonly under light-saturated conditions with no effect from theCO2 concentration. This inhibition was reversed by 2% O2. With,more than 2% O2, the rate of 14CO2 fixation was inhibited; 100%O2 caused about 20% inhibition which could be reversed by 2%O2. The degree of inhibition was only slightly higher at lowconcentrations (less than 0.43 mM NaHCO3) than at high CO2 concentrations,indicating that photorespiration is not a main cause of thisinhibition. Significant injuries to the plasma membrane were detected inEuglena gracilis cells during ozone exposure (240 µ1.liter1,delivery rate of l µmol.min–1), as assessed by measuringthe alterations of vitamin B12 and acetate uptakes and the leakageof intracellular K+ (Rb+). A rapid decrease in the uptake ofvitamin B12 and acetate was observed within 15 min of treatment,indicating that both transport systems are very sensitive toO3. On the other hand, the leakage of intracellular K+ ions,as measured by the efflux of 86Rb+ from prelabelled cells, couldonly be detected after 30 min of O3 exposure. These resultssuggest that the initial metabolic symptoms of injury is atthe level of the two transport systems examined and that thealteration of the membrane permeability to K+ ions appears asa second step in the cascade of oxidative events at the plasmamembrane level. When Euglena cells were allowed to recover underautotrophic growth conditions following O3 treatment, vitaminB12 and 86Rb+ (K+) ions uptakes returned gradually to controllevel within 5 h of the recovery period. Acetate uptake returnedto control level at a slower rate and needed 20 h for completerecovery. These results indicate that the cells were able toactively repair most of the initial oxidative damages inducedby O3. The metabolic significance of the repair mechanism(s)is discussed. Barley (Hordeum vulgare L. cv. Golf) plants were grown at twodifferent relative addition rates; 0.1 and 0.2 d–1 ofnitrate. Three to five days before measurements started theplants were transferred to a nutrient solution with 2 mM nitrateor ammonium. The ammonium-grown plants showed increased ammoniumlevels in both shoots and roots and also increased ammoniumconcentrations in xylem sap. Ammonia emission measured in cuvettes connected to an automaticNH3 monitor was close to zero for nitrate-grown plants but increasedto 0.59 and 0.88 nmol NH3 m–2 S–1 for plants transferredto ammonium after growing at RA=0.2 and 0.1 d–1, respectively.In darkness, NH3 emission decreased together with photosynthesisand transpiration, but increased rapidly when the light wasturned on again. Addition of 0.5 mM methionine sulphoximine (MSO) to the plantscaused an almost complete inhibition of both root and shootglutamine synthetase (GS) activity after 24 h. Ammonia emissionincreased dramatically and photosynthesis and transpirationdecreased in both nitrate- and ammonium-grown plants as a resultof the GS inhibition. At the same time plant tissue and xylemsap ammonium concentrations increased, indicating the importanceof GS in controlling plant ammonium levels and thereby NH3 emissionfrom the leaves. Bean plants (Phaseolus vulgaris L. cv. Fardenlosa Shiny) werelabelled with carbon-11 via their first trifoliate leaves when3-weeks-old and the transient inhibitions of translocation causedby the application of ammonium chloride solutions (10 mol m–3)to a peeled region of stem were studied. At pH 6·5 theammonium was without effect. At pH 11·0 even a briefapplication inhibited translocation for many minutes, whilelonger applications inhibited translocation for considerablylonger. Solutions of 10 mol m–3 sodium chloride were withouteffect at either pH. At pH 6·5 ammonium chloride solution contains predominantlyammonium ions (NH4+) and at pH 11·0 predominantly dissolvedammonia gas (NH3). Hence we conclude that phloem transport withinbean stems is inhibited by dissolved ammonia gas but not ammoniumions. Sucrose inhibition of flowering in Lemna perpusilla 6746 wasat least partially reversed by 5'-AMP, cyclic 3',5'-AMP, 5'-ADP,5'-ATP and K2HPO4. Experiments have examined the effect of phenylmercuric acetate(PMA) on the guard cells of Commelina communis. In one series,PMA was supplied to the leaf surface; after different time intervalsthe epidermis was removed and the ability of the stomata toopen was determined. In the other series, different concentrationsof PMA were included in the medium used for inccubating epidermalstrips with which ion-stimulated stomatal opening was assayed.At concentrations of 10-54 M and above the effect of PMA wassevere and the structural integrity of the guard cells was affected;they were unable to accumulate neutral red. At concentrationsarpound 10-6 M the guard cells were less affected and PMA broughtabout a transient stimulation of stomatal opening by releasingsubsidiary-cell turgor pressure. A solution of 5 x 10-4 M PMA applied to leaves reduced by halfthe photosynthetic 14CO2 incorporation into C. communis mesophyll.In Zea mays it increased the CO2 compensation point and alsothe resistance to diffusion in the gas phase (RA, but therewas a proportionately greater increase in the apparent liquidphase resistance (Rt). This direct inhibition of mesophyll photosynthesisundermines one of the major objectives of applying anatitranspirants,and for this reason it is suggested that PMA is unsuitable forgeneral application to crops.  相似文献

In order to express selectivity of ion channels, it is commonto determine the reversal voltages, Vr in the presence of differention species, and to convert these voltages to permeability ratiosby constant field equations, which are based on independentelectro-diffusion. In the case of individual ion channels, competitionrather than independence can be expected. This situation hasbeen described here in terms of a catalytic reaction cycle fora channel which may translocate K+ as well as Ca2+ . An exampleis given, where the fundamental rate constants for the K+ cycleand for the Ca2+ cycle are the same, and opposing 1:10 concentrationgradients for Ca2+ and for K+ are applied. In this case, thereversal voltage of the resultant total current is so closeto the equilibrium voltage of Ca2+, that constant field calculationwould yield an overestimated permeability ratio PCa:PK of about10. Two limiting cases are pointed out. In the case of dilutesolution, the ions move independently and the system can obeythe constant field theory. At high concentrations, the systemapproaches complete dependence and will consequently behaveas an antiporter. The effect of Ca2+ and ammonia on mitochondrial NADH-glutamatedehydrogenase (GDH: EC 1.4.1.2 [EC] ) isolated from turnip root (Brassicarapa L.) activity was examined. Increasing the ammonia [(NH4)2SO4]concentration led to significant substrate inhibition whichcould be reversed by micromolar levels of Ca2+. The sensitivityof the enzyme to ammonia inhibition and its reversal by Ca2+was affected by proteolysis. After treatment with various proteases,lower concentrations of Ca2+ were capable of fully activatingthe enzyme or overcoming the inhibitory effects of high ammonium,compared to non-treated enzyme. However, the protease-treatedenzyme was still sensitive to ethylene glycol-bis(ß-aminoethylether) N,N,N',N'-tetraacetate (EGTA). In contrast, NADH-GDHactivity was inhibited approx. 30% by organic mercurials (200µm), but the residual activity was not affected by thesubsequent additions of EGTA. NADH-GDH activity could also bestimulated by additions of high concentrations of NaCl (300mM) in the absence of added Ca2+. Previous papers have shown that abscisic acid can inhibit transportof ions across the root to the xylem vessels, resulting in reducedexudation from excised roots or inhibiting guttation from intactplants. However, it has not been established whether the inhibitionwas due to a reduction in salt transport (Js) or in permeabilityof the roots to water (Lp). This paper investigates the effectof ABA on Lp and Js separately. It is shown that Lp increasedin ABA and then fell, but was about the same as in control rootswhen transport was inhibited. The effect of ABA on exudationtherefore appeared to be mainly due to reduction in Js. Inhibitionof Js was also present in intact, transpiring plants and sowas not due to reduced water flow. The inhibition of ion releaseto the xylem affected Na+, Mg2+, Ca2+, and phosphate as wellas the major ion in the exudate, K+. It is concluded that ABAinhibits salt transport to the shoot by acting on ion transportinto the xylem, and not by reducing water flow coupled withsalt transport.  相似文献

Phagotrophic protists are major components of pelagic food webs,both as consumers of bacterial and phytoplankton cells, andas regenerators of inorganic nutrients. In this study, we estimatedthe efficiency of ammonium regeneration by protists feedingon bacteria within natural plank-tonic assemblages, using a15N tracer method, in which the excretion of 15N-labeled ammoniumdue to grazing on 15N pre-labeled bacteria was followed overtime. We tested this approach in experiments based on the additionof heat-killed 15N-labeled bacteria to laboratory cultures andto samples of coastal seawater. During two experiments, variationin abundance of bacterivores and bacterioplankton resulted innon-constant grazing rates. Deterministic computer models thatused abundance of bacteria and protists as variables were developedto estimate best-fit values of grazing mortality (g, h–1)and of ammonium regeneration efficiency (RE, fraction of theinitial 15N label in added bacteria which is released as ammonium).Estimated ammonium RE were 0.30–0.35 for one trophic linksystems with both a monospecific culture and a mixed speciesassemblage of bacterivorous flagellates. RE was higher for multi-trophicstep food webs: 0.60 for 5 µm pre-screened coastal seawaterand 0.90 for whole coastal seawater.  相似文献

Seedlings of Ricinus communis L. cultivated in quartz sand weresupplied with a nutrient solution containing either 1 mol m–3NO3 or 1 mol m–3 NH+4 as the nitrogen source. Duringthe period between 41 and 51 d after sowing, the flows of N,C and inorganic ions between root and shoot were modelled andexpressed on a fresh weight basis. Plant growth was clearlyinhibited in the presence of NH+4. In the xylem sap the majornitrogenous solutes were nitrate (74%) or glutamine (78%) innitrate or ammonium-fed plants, respectively. The pattern ofamino acids was not markedly influenced by nitrogen nutrition;glutamine was the dominant compound in both cases. NH+4 wasnot transported in significant amounts in both treatments. Inthe phloem, nitrogen was transported almost exclusively in organicform, glutamine being the dominant nitrogenous solute, but theN-source affected the amino acids transported. Uptake of nitrogenand carbon per unit fresh weight was only slightly decreasedby ammonium. The partitioning of nitrogen was independent ofthe form of N-nutrition, although the flow of nitrogen and carbonin the phloem was enhanced in ammonium-fed plants. Cation uptakerates were halved in the presence of ammonium and lower quantitiesof K+, Na+ and Ca2+ but not of Mg2+ were transported to theshoot. As NH+4 was balanced by a 30-fold increase in chloride in thesolution, chloride uptake was increased 6-fold under ammoniumnutrition. We concluded that ammonium was predominantly assimilated inthe root. Nitrate reduction and assimilation occurred in bothshoot and root. The assimilation of ammonium in roots of ammonium-fedplants was associated with a higher respiration rate. The NADP$-specific isocitrate dehydrogenase was partially purifiedfrom photosynthetically-grown Rhodospirillum rubrum. The pHoptimum is between 7.5 and 9.0 in phosphate buffer. The apparentKm is 3.1x10–5 M for isocitrate, 5.1x10–5 M forNADP$, 1.7x10–5 M for manganese, 1.5x10–4 M formagnesium, and 3.5x10–3 M for inorganic orthophosphate.Arsenate exerts a slight inhibition. The Q10 between 17.5°Cand 40°C is 1.62, and the energy of activation at 25°Cis 9.74 Kcal/mole. Glyoxylate and oxalacetate cause concertedinhibition of the enzyme activity. Various nucleotides inhibitthe activity. The kinetics of inhibition by ATP was found tobe mixed type with respect to NADP$ and isocitrate, the Ki valuesbeing 1.17x10–3 M and 1.10x10–3 M respectively.The inhibition between ATP and orthophosphate is competitivewith a Ki of 10–4M. In Daucus carota cells cultivated in vitro, the ammonium ionstimulates the incorporation of radioactivity from labelledglucose and labelled pyruvate into CO2 and into the residueinsoluble in 60 per cent (v/v) ethanol. There is a higher 14CO2production from [6-14C2] glucose than from [6-14C] glucose.These results suggest a possible stimulation of glycolysis bythe ammonium ion.  相似文献

A study has been made of photosynthetic 14CO2 fixation by isolated‘mature’ internodes of Nitella translucens. Experimentalconditions were similar to those used in studies of the ionicrelations of these cells. Maximum rates of photosynthesis were33–40µµmoles CO2, fixed per cm2 of surfacearea per second (equivalent to 12–15 /xmoles fixed permg chlorophyll per hour). l4CO2 fixation was inhibited to thedark level by 3(3,4,dichlorophenyl)-1, 1-dimethylurea (at 0-6µM or 10µM) and by the uncoupler carbonyl cyanide-m-chlorophenylhydrazone(SµM). The presence of imidazole or ammonium sulphate(both of which uncouple ATP production in vitro) did not resultin an inhibition of 14CO2 fixation. These results are discussedin relation to published work on solute uptake by Nitella translucens.During photosynthesis there was rapid movement of 14C-labelledorganic compounds out of the chloroplasts. 14C-labelled sucrose,ammo-acids, and sugar phosphates were found in samples of vacuolarsap.  相似文献

The activity of plasma membrane H+-ATPase was measured withmembrane fragments of guard-cell protoplasts isolated from Viciafaba L. ATP hydrolytic activity was slightly inhibited by oligomycinand ammonium molybdate, and markedly inhibited by NO3and vanadate. In the presence of oligomycin, ammonium molybdateand NO3, the ATP-hydrolyzing activity was strongly inhibitedby vanadate. It was also inhibited by diethylstilbestrol (DES),p-chloromercuribenzoic acid (PCMB) and Ca2+, but slightly stimulatedby carbonyl cyanide m-chlorophenylhydrazone (CCCP). The acitivityhad higher specificity for ATP as a substrate than other phosphoricesters such as ADP, AMP, GTP and p-nitrophenylphosphate; theKm was 0.5 mM for ATP. The activity required Mg2+ but was notaffected by K+, and it was maximal around pH 6.8. When guard-cellprotoplasts were used instead of membrane fragments, the ATPaseactivity reached up to 800µmol Pi.(mg Chl)–1.h–1in the presence of lysolecithin. The uptake rate of carbon and nitrogen (ammonium, nitrate andurea) by the Microcystis predominating among phytoplankton wasinvestigated in the summer of 1984 in Takahamaira Bay of LakeKasumigaura. The Vmax values of Microcystis for nitrate (0.025–0.046h–1) and ammonium (0.15–0.17 h–1) were considerablyhigher than other natural phytoplankton. The ammonium, nitrateand urea uptake by Microcystis was light dependent and was notinhibited with nigh light intensity. The K1 values were farlower than the Ik values. The carbon uptake was not influencedby nitrogen enrichment. Microcystis accelerated the uptake rateby changing Vmax/K s value when nitrogen versus carbon contentin cells declined. Nitrate was scarcely existent in TakahamairiBay during the summer, when Microcystis usually used ammoniumas the nitrogen source. However, the standing stock of ammoniumin the water was far lower than the daily ammonium uptake rates.Therefore, the ammonium in this water had to be supplied becauseof its rapid turn-over time (–0.7–2.6 h).  相似文献
