A greenhouse population of the sting nematode, Belonolaimus longicaudatus, obtained from an infested golf course in California''s Coachella Valley, was surface-decontaminated and cuhured on excised roots of Zea mays supported by Gamborg''s B5 medium. At 26-27 °C the females laid eggs, and newly emerged juveniles of the second generation completed three molts within 29 days after egg deposition. Sixty days after inoculation with 60 females and 40 males, an average of 529 nematodes and 83 eggs were recovered from the culture. The feeding process consisted of probing, stylet penetration, ingestion, and stylet retraction. Feeding seemed to be necessary before egg deposition or molting occurred. The sting nematode was observed feeding exclusively as an ectoparasite and preferably at the region of cell division and elongation. Vigorous feeding by many nematodes usually caused discoloration of root tips and termination of growth.  相似文献

Postinfection development of Meloidogyne chitwoodi from second-stage juveniles (J2) to mature females and egg deposition on ''Nugaines'' winter wheat required 105, 51, 36, and 21 days at 10, 15, 20, and 25 C. At 25 C, the J2 induced cavities and hyperplasia in the cortex and apical meristem of root tips with hypertrophy of cortical and apical meristem cell nuclei, 2 and 5 days after inoculation. Giant cells induced by late J2 were observed in the stele 10 days after inoculation. Clusters of egg-laying females were common on wheat root galls 25 days after inoculation. Juveniles penetrated wheat roots at 4 C and above, but not at 2 C, when inoculum was obtained from cultures grown at 20 C, but no penetration occurred at 4 C when inoculum was stored for 12 hours at 4 C before inoculation. In northern Utah, J2 penetrated Nugaines wheat roots in the field in mid-May, about 5 months after seedling emergence. M. chitwoodi eggs were first observed on wheat roots in mid-July when plants were in blossom. Only 40% of overwintered M. chitwoodi eggs hatched at 25 C.  相似文献

The embryogenic development of Rolylenchulus borealis, at 24-26 C, was completed on corn, in 12-15 days, and the life-cycle of the nematode from egg to egg required 35-40 days at 20-25 C. Juveniles remained in the soil as preinfective stages for 17-19 days before becoming adults. Only immature vermiform and swollen egg-laying females were found attached to corn roots. Eggs were laid in a gelatinous matrix on the root surface; the number of eggs per egg mass was 45 ± 28 on corn roots. Bean, green pea, potato, sorghum, and sweet potato were also found to be hosts of R. borealis. The nematode established a permanent feeding site on corn root in an endodermal cell that became hypertrophied. Pericyclic cells close to the feeding site showed granular cytoplasm and nuclei with hypertrophied nucleoli. A cell wall ingrowth was also noted around the area of stylet penetration into the endodermal cell.  相似文献

Two experiments were conducted to assess preference by sheep for endophyte-infected tall fescue growing in monoculture at least 5 m away from alfalfa (fescue-middle (FM)) over endophyte-infected tall fescue growing adjacent (0.2 to 1 m; fescue–alfalfa (FA)) to alfalfa (FA), and the effect of legume scent on preference for endophyte-infected tall fescue. In Experiment 1, 10 six-month-old lambs were offered for 12 days a choice of freshly harvested FA and FM. On days 13 and 14, lambs were offered the same choice, except cages (to allow access only to scent) containing freshly harvested alfalfa were put in the feeders containing FA, whereas cages containing freshly harvested FM were included with the feeders containing FM. Forage intake was measured 1 h after feeding and at three consecutive 2-h intervals thereafter. FA contained greater (P<0.002) concentrations of the alkaloid ergovaline (360±27 ppm) and CP (8±0.4%) than FM (219±27 ppm and 6±0.4%, respectively). Lambs preferred (P<0.05) FA to FM during the 1st hour of feeding, but the differences became smaller and disappeared in later feeding periods (P<0.005). Lambs offered FA with alfalfa scent or FM with FM scent preferred (P<0.05) FA but only on the 2nd day. In Experiment 2, 10 six-month-old lambs were offered a choice of FM with cages (to allow access only to scent) containing freshly harvested alfalfa or FM for 8 days. During the following 4 days, FM in the cages was replaced with freshly harvested sainfoin. Preference was greater (P<0.05) for FM offered with alfalfa scent than for FM offered with FM scent only on days 4 and 8. When lambs were offered FM with alfalfa or sainfoin in cages, they preferred (P<0.05) tall fescue with sainfoin scent over fescue with alfalfa scent, but intake was variable across hours and days (P<0.001). It is concluded that (1) lambs adjusted their intake of and preference for FA and FM over successive feeding bouts within each day, likely owing to an attempt to balance intakes of nutrients and alkaloids and (2) olfactory cues influenced preference, but to a lesser extent than nutrients and alkaloids in endophyte-infected tall fescue.  相似文献

Large populations of Criconemella curvata and extended experimental periods were required to adversely affect yield of ''Moapa 69'' alfalfa. C. curvata ectoparasitic feeding caused a reduction in feeder root numbers and tap root size and small lesions on tap and secondary roots. Greater reproduction occurred at 27 C than at 22 C, but the effect of the nematode on alfalfa growth was the same at both temperatures.  相似文献

The effect o f soil temperature on parasitism and development of Rotylenchulus reniformis on resistant (''Peking'' and ''Custer'') and susceptible (''Hood'' and ''Lee'') soybean (Glycine max) cultivars was studied. Soil temperatures of 15, 21.5, 25, 29.5 and 36 C ± 1 C were maintained in temperature tanks in a greenhouse. R. reniformis developed best at 25 and 29.5 C. The female life cycle can be completed within 19 days after inoculation under favorable conditions at 29.5 C. Plant root growth was best at 21.5 C. During a 27-day period, no egg masses were present on nematodes feeding on roots grown at 15 and 36 C. Egg masses developed on Hood but not on Lee when nematodes were introduced into soil and maintained at 29.5 C for 2 days before raising the temperature to 36 C.  相似文献

The reproductive tract of the parasitoidMicroctonus hyperodae was found to comprise 6.1±0.2 ovarioles containing a total of 40–60 oöcytes. After oviposition into its hostListronotus bonariensis, the parasitoid's egg volume increased by 205 times prior to hatching. At 19.1°C ovipositingM. hyperodae survived for a mean 21±4 days and laid a mean of 48±8 eggs.M. hyperodae collected from Colonia, Uruguay laid a mean of 62±15 eggs which was significantly more than the other ecotypes. Under caging conditions with an ample supply of hosts,M. hyperodae laid 51% of its eggs in the first 72 hours and on average 41% of the species' life-span occurred after the exhaustion of its egg supply. Minimum temperature forM. hyperodae oviposition was found to be c. 5°C; beyond this the rate of increase in egg-laying was approximately linear until 30°C whereafter the rate fell abruptly. There was effectively no egg-laying at 39°C. The parasitoid showed no preference for one host sex or the other either in the laboratory or under field conditions. Compared to otherMicroctonus spp.,M. hyperodae appeared to show low fecundity and high longevity. The adaptive implications of this are discussed.  相似文献

Studies were conducted to examine under differing temperatures (12, 16, 20, 24, 28, and 32 C) the penetration anti development of Meloidogyne hapla in resistant lines ''298'' and ''Nev. Syn XX'', and susceptible ''Lahontan'' and ''Ranger'' hardy-type alfalfas. The results indicated that resistance to M. hapla was similar to that previously described for M. incognita in nonhardy alfalfa. Although initial penetration in resistant seedlings was similar to that of susceptible seedlings, nematode larvae failed to establish and develop in root tissues and nematode numbers subsequently declined. In susceptible seedlings, nematode development proceeded rapidly, and egg production began after 5 weeks. Temperature had little influence on the nematode development except to slow the response at the lower temperatures. Other studies were conducted to verify a previously reported immune (no penetration) reaction to M. hapla by the ''Vernal'' selection ''M-4''. When compared to the resistant (penetration without nematode development) Vernal selection ''M-9'' under differing temperatures (20, 24, 28, and 32 C), each selection was equally penetrated by M. hapla but at a lower level than in susceptible Ranger cuttings. Generally, no root galling was observed in either M-4 or M-9; however, very slight galling was found 35 days after inoculation on about 50% of these cuttings when grown at 32 C.  相似文献

Gossypium arboreum ''Nanking CB 1402'' possessed a high level of resistance to Rotylenchulus reniformis. Within 16 h, the nematode penetrated roots of resistant and susceptible cottons equally. After 36 h, significantly fewer nematodes were found in resistant roots. Larvae fed in either an endodermal or pericyclic cell and had no specificity for root tissue of a particular age. In roots of resistant G. arboreum ''1402,'' wall breakdown of pericyclic cells was evident after 3 d, endodermal and cortical cells collapsed, and the hypertrophied pericyclic cells disintegrated within 12 d. Cell walls immediately adjacent to the nematode''s head were thickened and more safranin positive in resistant than in susceptible cotton cultivars. Several other cultivars of G. arboreum were also resistant to R. reniformis, based on nematode fecundity and percent egg reduction.  相似文献

When fed equal amounts of substrate, two Epidinium caudatum clone cultures of markedly different size produced similar volumes of microbial protoplasm. Addition of up to 50% volume of 72 h culture medium had no inhibitory effects on growth of Epidinium. Two clone cultures of Epidinium caudatum from Australia had longer generation times and showed less substrate attachment when compared to Ohio clones of this same species. Substitution of alfalfa for orchardgrass in the normal substrate increased Epidinium concentrations, while feeding only ground orchardgrass or alfalfa resulted in a marked decrease or disappearance of the protozoa. Eudiplodinium impalae, isolated from rumen contents of a steer in Australia, was successfully cultured, with generation times for this species averaging 11.3 h. Reducing particle size of the substrates by ball-milling was detrimental for growth of Entodinium and Epidinium; however, Eudiplodinium increased in concentration. Significant concentration differences were observed among six clone cultures of Epidinium obtained from Europe. A generation time of 18.7 h was measured for Enoploplastron triloricatum when the culture was transferred every 12 h. Lowering the incubation temperature to 34 °C completely inhibited protozoal growth of Epidinium and Entodinium exiguum after 12 days, but not for Entodinium caudatum.  相似文献

Tylenchorhynchus dubius was observed to feed on ''Toronto'' creeping bentgrass and ''Merion'' Kentucky bluegrass, and was a key participant in reducing the vegetative growth of both grass species. The severity of foliar and root macrosymptoms incited by T. dubius to ''Toronto'' bentgrass was greater on plants grown at 16 C than on plants maintained at 21, 27 and 32 C. These parasitized plants exhibited a suppression of secondary stolon formation, shortened internodes and premature inflorescence initiation. Initial inoculum densities of 500 and 1000 nematodes/test pot produced similar pathogenic effects on the host. Feeding was primarily on root hairs and epidermal cells immediately behind the meristematic region. No necrotic lesions or other diagnostic symptoms were visible at the feeding sites. Development of Tylenchorhynchus claytoni from unsegmented egg to hatching takes 135 hr at 22-25 C. The fourth molt lasts 5 to 6 days. During exsheathment the cast cuticle of the larva separated into two unequal parts, breaking near either the anterior or posterior end. The life cycle from egg to egg required from 31 to 38 days at 28 C on alfalfa seedlings and included four molts and four larval stages. Pathogenicity tests of Pratylenchus brachyurus on selected greenhouse-grown soybean cultivars indicated the nematode reduced seed yield of ''Hood'' but not that of ''Custer'', ''Bragg'', ''Dyer'', or ''Pickett''. Root weights of all cultivars were reduced. Damage and numbers of nematodes within soybean roots growing at 13, 21, and 29 C were greater at higher temperatures. At 29 C, root pruning was prominent in ''Hood'' and ''Pickett'' but limited in ''Custer'' and ''Hill''. Root pruning was not observed at 13 C and only ''Pickett'' showed pruning at 21 C. Plant height and foliage weight were not affected. P. brachyurus had no effect on the emergence of ''Pickett'' or ''Bragg'' soybeans. Nematode counts from roots of ''Pickett'' at intervals after inoculation indicated that hatching of second generation second-stage larvae occurred about 15 days after egg laying. An average of 68% of the initial inoculum penetrated the roots within five days of inoculation, the highest observed was 81% in five days. Details of structural damage in penetrated tissues were studied in sectioned roots. In soybean roots infected by P. brachyurus and/or Rhizoctonia solani greater damage occurred with nematode and fungus combined than with either acting alone.  相似文献

Nitrogen fixation (NF) by alfalfa and nitrogen transfer (NT) from alfalfa to associated timothy was studied under different environmental conditions in controlled growth chambers, using the15N dilution technique. Evidence was obtained of NT from alfalfa to the associated timothy. Conditions that favored NF by alfalfa resulted in an increase in its NT. Of 3 different temperature regimes (25/20, 16/14, and 12/9°C day/night), 16–25/14–20°C was the best range for NF by alfalfa and resulted in the greatest NT. High light intensity (550 uE.m−2.sec−1) and long days (16–20 h) also caused increased NF by alfalfa and benefitting timothy more than in a regime of low light intensity (by shading 50% or 75%) or short days (12/12 or 16/8 h day/night). When the inoculated (Rhizobium meliloti) root systems of plants were kept free from other microorganisms (axenic condition) to minimize possible decomposition of dead tissues, lower NT from alfalfa was observed, especially at later cuts, compared to non-axenic plants. This suggests that both direct excretion and decomposition of dead alfalfa tissues are sources of N benefit from alfalfa to associated timothy. The endoparasitic behavior of Pratylenchus penetrans was examined using video-enhanced contrast microscopy to observe living nematodes inside root tissue. Feeding behavior could be separated into phases of probing, cell penetration by the stytet, salivation, and food ingestion for brief and extended periods. After cell penetration, a small "salivation zone" was formed around the stylet tip. No feeding tubes were observed. Feeding and migration were interrupted by rest phases when a nematode became characteristically coiled inside a cell. Tissue damage was caused primarily by migration and extended feeding periods. A new aseptic culture system for studying interactions between tomato (Lycopersicon esculentum) and Meloidogyne incognita is described. Epidermal thin cell layer explants from peduncles of tomato produced up to 20 adventitious roots per culture in 4-9 days on Murashige &Scoog medium plus kinetin and indole acetic acid. Rooted cultures were transferred to Gamborg''s B-5 medium and inoculated with infective second-stage juveniles. Gall formation was apparent 5 days after inoculation and egg production by mature females occurred within 25 days at 25 C in the susceptible genotypes Rutgers and Red Alert. Resistant genotypes LA655, LA656, and LA1022 exhibited a characteristic hypersensitive response. This system provides large numbers of cultured root tips for studies on the molecular basis of the host-parasite relationship.  相似文献

2012年5~7月,应用e-Science信息技术和标记法,对陕西神木县红碱淖(N 38°13′~ 39°27′,E 109°42′~110°54′)遗鸥(Larus relictus)的育雏行为和雏鸟生长发育进行了研究.结果表明,雏鸟由双亲共同承担喂食.育雏前期,亲鸟采取直接喂食、食物呕吐于巢边和在巢中间断性喂食这3种喂食模式;亲鸟昼间平均喂食(0.706±0.036)次/h,夜间平均喂食(0.469±0.024)次/h,双亲在喂食频次上无显著差异(F=32.54,P>0.05).育雏后期,主要采取双亲直接喂食和亲鸟把食物呕吐于地面上,由雏鸟自己取食的喂食模式;亲鸟昼间平均喂食(0.416±0.021)次/h,夜间平均喂食(0.331±0.018)次/h,亲鸟喂食次数与雏鸟的日龄存在相关性(r =0.074,P<0.05).随着雏鸟日龄的增长,暖雏次数趋于减少,而在炎热晴天、降雨和大风等天气状况下,暖雏时间和护雏行为都增强.雏鸟20日龄后未再观察到暖雏行为.雏鸟体长及外部器官的形态学参数适合用Gompertz曲线方程拟合.同时,与其近缘种黑嘴鸥(L.saundersi)的育雏行为和雏鸟生长进行了比较.  相似文献

Immunizations of New Zealand White rabbits with specific macrophage migration inhibitory factor (MIF) tick peptide (PEP) produced circulating anti-tick PEP antibodies in the hosts. Antibody titers of greater than 1:5000 to tick MIF peptide were observed for crude sera from PEP-immunized rabbits. PEP- and BSA-vaccinated rabbits were infested with Amblyomma americanum adults. Feeding intervals, female weights, egg masses and percent egg hatch were measured for ticks feeding on control and immunized hosts. Feeding intervals were significantly lengthened to 13.3 days for PEP-vaccinated hosts compared to BSA-vaccinated controls at 12.4 days, while female engorgement weights and egg masses were unchanged. By immunizing hosts using specific tick PEP, we were able to alter the length of time the ticks fed on their hosts.  相似文献

Root gall induction and egg production by the four recognized host races and two cytological races of Meloidogyne incognita were compared on cotton Gossypium hirsutum cvs. Deltapine 16 (root-knot susceptible) and Auburn 634 (highly resistant). The 12 nematode populations included in the study were from various parts of the world. No population increases occurred on the highly resistant cultivar. After 45 days, populations of host races 1 and 2 induced slight root galling on both cuhivars with only limited reproduction. Host race 4 populations induced moderate root galling with higher reproduction on Deltapine 16 than that of race 1 or race 2 populations. Host race 3 populations induced severe root galling with population density increases of 7-30-fold. In a complementary study, 24 cotton cultivars or breeding lines were compared for suitability as hosts for a typical population of M. incognita race 3. The poorest hosts, ''Aubnru 623,'' ''Auburn 634,'' and ''McNair 220,'' yielded fewer eggs after 45 days than were added initially. The best hosts - ''M-8.'' ''DES 24-8,'' ''McNair 235,'' and ''Coker 20l'' - yielded > 5 times as many eggs as were added initially.  相似文献

The mermithid nematode, Filipjevimermis leipsandra, was successfully cultivated to the preadult stage in Schneider''s Drosophila medium supplemented with 20% fetal bovine serum. Upon transfer to a solid substrate the preadults continued to develop into ovipositing adult females. Four molts were observed. The first molt occurred in the egg. The second occurred after 6-8 days in culture during which the very thin cuticle was shed completely. The third molt occurred after 18-20 days in culture; the cuticle was retained by the third-stage nematode. This stage was considered comparable to the preadult stage that emerges from host larva, Diabrotica spp. The fourth molt occurred within 12 days after the preadult was transferred from the liquid medium to a solid substrate. Adult females began ovipositing viable eggs 1-3 days after the final molt.  相似文献
