Speleological, paleomagnetic, mineral magnetic, and biochronological analyses have been undertaken at the Gondolin hominin-bearing paleocave, North West Province, South Africa. Two fossiliferous but stratigraphically separate sequences, GD2 and GD1/3, which were once part of a large cavern system, have been identified. Although some comparative paleomagnetic samples were taken from the GD 1, 3, and 4 localities that are currently under investigation, the research presented here focuses on the fossil-rich, in situ deposits at locality GD 2, excavated by E.S. Vrba in 1979. The GD 2 deposits are dominated by normal-polarity calcified clastic deposits that are sandwiched between clastic-free flowstone speleothems. The lower flowstone has a sharp contact with the red siltstone deposits and is of reversed polarity. The capping flowstone shows a change from normal to reversed polarity, thereby preserving a polarity reversal. While the paleomagnetic work indicates that the GD 2 fossil material was deposited during a normal-polarity period, the shortness of the sequence made matching of the magnetostratigraphy to the geomagnetic polarity time scale (GPTS) impossible without the aid of biochronology. While lacking multiple time-sensitive taxa, the recovery of specimens attributable to Stage III Metridiochoerus andrewsi is consistent with a deposition date between 1.9 and 1.5 Ma. A comparison of the magnetostratigraphy with the GPTS therefore suggests that the fauna-bearing siltstone of GD 2 date to the Olduvai normal-polarity event, which occurred between 1.95 and 1.78 Ma, and that the reversal from normal to reversed polarity identified in the capping flowstone dates to 1.78 Ma. The main faunal layers therefore date to slightly older than 1.78 Ma. Deposits from the GD 1 locality are dominated by reversed directions of magnetization, which show that this deposit is not of the same age as the faunal layers from the GD 2 locality.  相似文献

We have analyzed the histone genes from the sea urchin Lytechinus pictus. Examination of native DNA from individuals reveals four major Eco RI restriction endonuclease histone gene DNA fragments which have been labeled A (6.0 kb), B (4.1 kb), C (3.1 kb) and D (1.2 kb). The fragments A, B and C have been cloned into E. coli plasmids (pLpA, pLpB and pLpC). These histone gene fragments display length and sequence heterogeneity in different individuals. The plasmid pLpA contains the coding regions for H1, H4, H2B and H3 histones, and we determined that the DNA fragment D is tandem to A in native DNA and that it contains the H2A gene. The plasmids pLpB and pLpC contain the histone genes H2A-H1-H4 and H2B-H3, respectively, and together contain the sequences for the five major histones. Restriction analysis of native L. pictus DNA reveals that B and C are tandem to each other but not intermingled with the A-D-type repeat units, and are thus in separate clusters with a repeat length of 7.2 kb. Since the two cluster types do not segregate, they are not alleles. Hybridization of histone mRNA to exonuclease III-digested linear DNA demonstrated an identical polarity of the histone genes in the A-D- and B-C-type repeat units. This result revealed that the L. pictus histone genes have a polarity which is the same as other sea urchin histone genes examined to date—that is, 3′ H1-H4-H2B-H3-H2A 5′. Restriction endonuclease cleavage patterns of the cloned segments indicate that considerable sequence heterogeneity exists between the two types of histone gene repeat units.  相似文献

Further investigations were carried out at Ain Hanech, Algeria in 1998 and 1999 to explore its potential for investigating early hominid behavioral patterns and adaptation. Research concentrated on the stratigraphy and dating, identifying new archaeological deposits, and excavating the Ain Hanech and El-Kherba localities. To enhance the chronological control within a biostratigraphic framework, the Ain Boucherit fossil-bearing stratum, yielding a Plio-Pleistocene fauna, is correlated with the regional stratigraphy. In the stratigraphic sequence, the Ain Boucherit stratum, located 13m below the Ain Hanech Oldowan occurrences, is found in Unit Q of the Ain Hanech Formation. Unit Q shows a paleomagnetically reversed polarity, which may be correlated with an age earlier than the Olduvai normal subchron (1.95-1.77Ma). Based on test trenches and stratigraphic analyses, additional Oldowan deposits A, B, and C are identified at Ain Hanech. All three deposits and the El-Kherba site contain Mode I technology artefacts associated with an Early Pleistocene fauna. El-Kherba is stratigraphically equivalent to Ain Hanech. These two archaeological sites are estimated to be dated to about 1.8Ma.  相似文献

Stratigraphic, sedimentological and paleomagnetic studies were conducted on the travertine from Denizli Basin, near Kocabas village, in the Denizli region in Turkey, following the paleontological discovery in 2002. The stratigraphic and sedimentological studies show at least two main cycles of mass travertine, separated by a fluvial deposit and overlain by a fluvio-lacustrine deposit. These travertines must have formed in environments with strong hydrodynamics (streams or waterfalls) and are preferentially located at breaks of slopes. The paleomagnetic study shows that all the quarry travertine presents reverse magnetic polarity. On the other hand, the detrital fluvio-lacustrine deposit above the travertine presents normal geomagnetic polarity, except at the top, where it is reversed. Given the presence of an archaic Homo erectus skull and Villafranchian paleontological remains in the upper travertine unit, the whole travertine dates from the upper Matuyama, and is more recent than the Olduvai event (1.78 Ma). The normal polarity recorded in the upper fluvio-lacustrine deposit could correspond to the Cobb Mountain excursion, dated to 1.22 Ma.  相似文献

The isolation of the two hybrid plasmids 56H8 and 132E3, which contain D. melanogaster (Dm) DNA sequences complementary to the mRNA coding for the 70,000 dalton heat shock protein, has been reported (Schedl et al., 1978). Here we compare the sequence arrangement in the two cloned Dm DNA segments by restriction, cross-hybridization and heteroduplex analysis. The results show that the two cloned DNA segments derive from nonoverlapping regions of the Dm genome; that they contain homologous regions present once in 56H8 and twice in 132E3; and that each homologous region is composed of three distinct contiguous sequence elements, x, y and z, which together define a 3 kb common unit. While the 2.5 kb z elements show a high degree of sequence homology in all three common units, the three x and y elements display an intriguing relationship. The localization of the mRNA coding sequences within each of these common units is presented in the accompanying paper (Artavanis-Tsakonas et al., 1979).  相似文献

The genome of equine herpesvirus 1 (EHV-1) defective interfering (DI) particle DNA originates from discrete regions within the standard (STD) EHV-1 genome: the left terminus (0.0 to 0.04 map units) and the inverted repeats (0.78 to 0.79 and 0.83 to 0.87 map units of the internal inverted repeat; 0.91 to 0.95 and 0.99 to 1.00 map units of the terminal inverted repeat). Since DI DNA must contain cis-acting DNA sequences, such as replication origins, which cannot be supplied in trans by the STD EHV-1 virus, regions of the EHV-1 genome shown to be in DI DNA were assayed for the presence of a viral origin of DNA replication. Specifically, STD EHV-1 DNA fragments encompassing the genomic regions present in DI particle DNA were inserted into the vector pAT153, and individual clones were tested by transfection assays for the ability to support the amplification and replication of plasmid DNA in EHV-1-infected cells. The Sma-1 subfragment of the internal inverted repeat sequence (0.83 to 0.85 map units) was shown to contain origin of replication activity. Subcloning and BAL 31 deletion analysis of the 2.35-kilobase-pair (kbp) Sma-1 fragment delineated a 200-bp fragment that contained origin activity. The origin activities of all EHV-1 clones which were positive by the transfection assay were confirmed by methylation analysis by using the methylation-sensitive restriction enzymes DpnI and MboI. DNA sequencing of the 200-bp fragment which contained an EHV-1 origin of replication indicated that this region has significant homology to previously characterized origins of replication of human herpesviruses. Furthermore, comparison of known origin sequences demonstrated that a 9-bp sequence, CGTTCGCAC, which is conserved among all origins of replication of human lytic herpesviruses and which is contained within the 18-bp region in herpes simplex virus type 1 origins shown by others to be protected by an origin-binding protein (P. Elias, M. E. O'Donnell, E. S. Mocarski, and I. R. Lehman, Proc. Natl. Acad. Sci. USA 83:6322-6326) is also conserved across species in the EHV-1 origin of replication.  相似文献

Fluvio-lacustrine post-Miocene deposits of the Bolivian Altiplano have been sampled in 400 m of section in the La Paz and Avo Avo basins in order to provide a magnetostratigraphic scheme for chronostratigraphic interpretations. Stable characteristics components were isolated after stepwise AF or thermal demagnetizations. The directions of cleaned magnetization were compared to the normal and reverse dipole field directions at the site. The plot of the mean directions calculated for each stratigraphic level define successive magnetozones which can be identified on the standard geomagnetic polarity scale with the help of the K/Ar ages obtained on tuff layers. The deposition of the middle and upper part of the La Paz Formation covers the Gauss epoch. The Gauss/Matuyama (2.48 Ma) and the Gilbert/Gauss (3.4 Ma) limits are located in the top and the middle of the formation. The occurrence of the first glacial/interglacial oscillation can be placed in the early Matuyama (ca 2.2 Ma) in agreement with the oxygen isotopic record from the equatorial ocean.  相似文献

A combined magnetostratigraphic and biostratigraphic study has been performed on the Maastrichtian Senpohshi Formation in eastern Hokkaido Island, northern Japan, which is an approximately 1300 m thick section mainly composed of hemipelagic mudstone. The identification of magnetic polarity was possible at 51 horizons, whereby four magnetozones were recognized. These magnetozones were correlatable to geomagnetic polarity chrons C31r to C30n, suggesting that the age of the Senpohshi Formation is spanning from middle to upper part of the Maastrichtian (ca. 69–67 Ma).The magnetostratigraphy of the Senpohshi Formation established in this study enables a direct age correlation to the Maastrichtian successions in other regions. Thus, this detailed chronology of the formation contributes to paleontological studies of the Maastrichtian in the North Pacific region. For instance, this magnetostratigraphic age assessment implies the following: (1) the stratigraphic range of the ammonite Pachydiscus flexuosus contains polarity chrons from the lower part of C31r to the lower part of C31n, (2) the first occurrence (FO) of the calcareous nannofossil Nephrolithus frequens in the North Pacific region is correlatable to polarity chron C30n or below, and (3) the FO of the bivalve “Inoceramusawajiensis is located within polarity chrons from C31r to the upper part of C31n. This suggests that the inoceramid extinction event in the North Pacific region might have occurred during polarity chrons from C31r to the upper part of C31n (ca. 70.5–67.8 Ma), which is 2.3–5.0 Myr prior to the Cretaceous/Paleogene boundary. The trend of the Maastrichtian faunal turnover in the North Pacific is well consistent with those of other regions, brings a new evidence for understanding the global faunal turnover in the Maastrichtian, just before Cretaceous/Paleogene mass extinction.  相似文献

Palaeomagnetic analysis was conducted on speleothems from Members 1-5 at Sterkfontein Cave, South Africa. Palaeomagnetic analysis of siltstone and speleothem from the bulk of Member 4 indicate a reversed magnetic polarity that dates the deposits and its Australopithecus africanus fossils to between 2.58 and ∼2.16 Ma. Further confirmation of this age comes in the form of two short normal polarity events correlated to the Rèunion (∼2.16 Ma) and Huckleberry Ridge (∼2.05 Ma) events in speleothem capping the bulk of Member 4 and coeval with deposition of the final phase of Member 4, including A. africanus fossil Sts 5. At ∼2.16-2.05 Ma, Sts 5 is the youngest representative of A. africanus yet discovered. Palaeomagnetic analysis of the Silberberg Grotto deposits identifies a single short geomagnetic field event in flowstone overlying the StW 573 Australopithecus fossil, which is suggested to represent the Rèunion event at ∼2.16 Ma. This further supports the uranium lead age estimates of 2.3-2.2 Ma for the StW 573 fossil. Based on a reversed polarity for the deposits below the skeleton it cannot be older than 2.58 Ma. If StW 573 is considered to be a second species of Australopithecus then this indicates that two species of Australopithecus are present at Sterkfontein between 2.6 and 2.0 Ma. All of the Member 5 deposits date to less than 1.8 Ma based on a comparison of palaeomagnetic, faunal, and electron spin resonance age estimates. The StW 53 fossil bearing infill (M5A) is intermediate in age between Member 4 and the rest of Member 5 (B-C) at around 1.78-1.49 Ma. The rest of Member 5 (B-C) containing Oldowan and Acheulian stone tools and Homo and Paranthropus fossils was deposited gradually between 1.40 and 1.07 Ma, much younger than previously suggested.

Gap junctions are intercellular channels formed by the serial, head to head arrangement of two hemichannels. Each hemichannel is an oligomer of six protein subunits, which in vertebrates are encoded by the connexin gene family. All intercellular channels formed by connexins are sensitive to the relative difference in the membrane potential between coupled cells, the transjunctional voltage (Vj), and gate by the separate action of their component hemichannels (Harris, A.L., D.C. Spray, and M.V. Bennett. 1981. J. Gen. Physiol. 77:95-117). We reported previously that the polarity of Vj dependence is opposite for hemichannels formed by two closely related connexins, Cx32 and Cx26, when they are paired to form intercellular channels (Verselis, V.K., C.S. Ginter, and T.A. Bargiello. 1994. Nature. 368:348-351). The opposite gating polarity is due to a difference in the charge of the second amino acid. Negative charge substitutions of the neutral asparagine residue present in wild-type Cx32 (Cx32N2E or Cx32N2D) reverse the gating polarity of Cx32 hemichannels from closure at negative Vj to closure at positive Vj. In this paper, we further examine the mechanism of polarity reversal by determining the gating polarity of a chimeric connexin, in which the first extracellular loop (E1) of Cx32 is replaced with that of Cx43 (Cx43E1). The resulting chimera, Cx32*Cx43E1, forms conductive hemichannels when expressed in single Xenopus oocytes and intercellular channels in pairs of oocytes (Pfahnl, A., X.W. Zhou, R. Werner, and G. Dahl. 1997. Pflügers Arch. 433:733-779). We demonstrate that the polarity of Vj dependence of Cx32*Cx43E1 hemichannels in intercellular pairings is the same as that of wild-type Cx32 hemichannels and is reversed by the N2E substitution. In records of single intercellular channels, Vj dependence is characterized by gating transitions between fully open and subconductance levels. Comparable transitions are observed in Cx32*Cx43E1 conductive hemichannels at negative membrane potentials and the polarity of these transitions is reversed by the N2E substitution. We conclude that the mechanism of Vj dependence of intercellular channels is conserved in conductive hemichannels and term the process Vj gating. Heteromeric conductive hemichannels comprised of Cx32*Cx43E1 and Cx32N2E*Cx43E1 subunits display bipolar Vj gating, closing to substates at both positive and negative membrane potentials. The number of bipolar hemichannels observed in cells expressing mixtures of the two connexin subunits coincides with the number of hemichannels that are expected to contain a single oppositely charged subunit. We conclude that the movement of the voltage sensor in a single connexin subunit is sufficient to initiate Vj gating. We further suggest that Vj gating results from conformational changes in individual connexin subunits rather than by a concerted change in the conformation of all six subunits.  相似文献

New Late Triassic–earliest Jurassic magneto-biostratigraphic data have been obtained from three overlapping sections in the Southern Alps, Italy (Costa Imagna, Brumano, Italcementi Quarry), composed of ~ 520 m of shallow-marine carbonates outcropping in stratigraphic continuity. Characteristic magnetic components of presumed depositional age record a sequence of 9 normal and reverse polarity magnetozones (as defined by at least three stratigraphically superposed samples) linked by conodont and palynofloral evidence from this study and the literature to Rhaetian to Triassic–Jurassic boundary age. This represents a significantly larger number of polarity zones than previously recognized in more condensed Rhaetian sections from the literature, and by inference represents more time. These data are placed in a broader Late Triassic temporal framework by means of correlations to published magneto-biostratigraphic data from the Tethyan marine Pizzo Mondello section and the Newark astronomical polarity time scale (APTS). This framework is consistent with a position of the Norian–Rhaetian boundary (as defined at Brumano and Pizzo Mondello by the first appearance of Misikella posthernsteini) within Newark magnetozones E17r–E19r in the ~ 207–210 Ma time interval, in basic agreement with the position originally estimated in the Newark using pollen biostratigraphy (E18 at 208–209 Ma). This framework is also consistent with the position of the Triassic–Jurassic boundary interval (placed at Italcementi Quarry at the acme of Kraeuselisporites reissingeri coincident with a negative carbon isotope excursion) correlative to just above Newark magnetozone E23r and just below the oldest CAMP lavas dated at ~ 202 Ma. Hence, we estimate the duration of the Rhaetian to be ~ 5.5–8.5 Myr (or even longer if the Triassic–Jurassic boundary is instead placed above the negative carbon isotope excursion as at Kuhjoch, which is the designated GSSP for the base of the Hettangian), and encompassing 9 magnetozones. This duration contrasts with a duration of ~ 2 Myr and only ~ 4 magnetozones in several alternative published magneto-biostratigraphic schemes.  相似文献

Results are presented of an integrated stratigraphic (calcareous plankton biostratigraphy, cyclostratigraphy and magnetostratigraphy) study of the Serravallian/Tortonian (S/T) boundary section of Monte Gibliscemi (Sicily, Italy). Astronomical calibration of the sedimentary cycles provides absolute ages for calcareous plankton bio-events in the interval between 9.8 and 12.1 Ma. The first occurrence (FO) of Neogloboquadrina acostaensis, usually taken to delimit the S/T boundary, is dated astronomically at 11.781 Ma, pre-dating the migratory arrival of the species at low latitudes in the Atlantic by almost 2 million years. In contrast to delayed low-latitude arrival of N. acostaensis, Paragloborotalia mayeri shows a delayed low-latitude extinction of slightly more than 0.7 million years with respect to the Mediterranean (last occurrence (LO) at 10.49 Ma at Ceara Rise; LO at 11.205 Ma in the Mediterranean). The Discoaster hamatus FO, dated at 10.150 Ma, is clearly delayed with respect to the open ocean. The ages of D. kugleri first and last common occurrence (FCO and LCO), Catinaster coalitus FO, Coccolithus miopelagicus last (regular) occurrence (L(R)O) and the D. hamatus/neohamatus cross-over, however, are in good to excellent agreement with astronomically tuned ages for the same events at Ceara Rise (tropical Atlantic), suggesting that both independently established timescales are consistent with one another. The lack of a reliable magnetostratigraphy hampers a direct comparison with the geomagnetic polarity timescale of Cande and Kent (1995; CK95), but ages of calcareous nannofossil events suggests that CK95 is significantly younger over the studied time interval. Approximate astronomical ages for the polarity reversals were obtained by exporting astronomical ages of selected nannofossil events from Ceara Rise (and the Mediterranean) to eastern equatorial Pacific ODP Leg 138 Site 845, which has a reliable magnetostratigraphy. Our data from the Rio Mazzapiedi–Castellania section reveal that the base of the Tortonian stratotype corresponds almost exactly with the first regular occurrence (FRO) of N. acostaensis s.s. as defined in the present study, dated at 10.554 Ma. An extrapolated age of 11.8 Ma calculated for the top of the Serravallian stratotype indicates that there is a gap between the top of the Serravallian and the base of the Tortonian stratotype, potentially rendering all bio-events in the interval between 11.8 and 10.554 Ma suitable for delimiting the S/T boundary. Despite the tectonic deformation and the lack of a magnetostratigraphy, Gibliscemi remains a candidate to define the S/T boundary by means of the Tortonian global boundary stratotype section and point (GSSP).  相似文献

Abstract Virus-like particles (VLPs) have been isolated from the yeast Endomyces magnusii . The VLPs measure 43 nm in diameter and contain six species of dsRNA (0.78, 0.83, 1.77, 1.84, 2.64, 4.30 kb respectively). E. magnusii produces a 'toxic' protein, which reduces the growth, and changes the colony morphology, of sensitive strains of Hansenula sp. growing on solid media. All strains of E. magnusii tested produced the 'toxin' and contained the VLPs. Current procedures of curing failed to destroy the ability to produce the 'toxin'.  相似文献

Correlations of Upper Triassic magnetic stratigraphies from Tethyan sections have been hampered by difficulties with conodont biostratigraphy and taxonomy, and discontinuous sedimentation, particularly in the ‘Hallstatt Limestones’ of Turkey and Austria. The magnetic stratigraphy and conodont biostratigraphy from the Upper Carnian to Upper Norian limestones exposed at Silická Brezová (Slovakia) can be correlated to other Tethyan sections and to the continental succession in the Newark Basin. The resulting correlations help to resolve some of the apparent discrepancies in existing conodont zonations, and result in a revised correlation to North American terrestrial vertebrate and palynological zones. The correlations imply that the Norian-Rhaetian boundary lies within Newark polarity zone E17r at ∼207 Ma. The Carnian-Norian boundary lies close to the base of Newark polarity zone E7r at ∼226 Ma. This implies durations for the Norian and Rhaetian stages of 19 Myr and 7 Myr, respectively.  相似文献

The sites of Barranco León D (BL-D) and Fuente Nueva 3 (FN-3) in the Guadix-Baza Basin (Granada, Spain), together with the site of Sima del Elefante in the Sierra de Atapuerca (Burgos, Spain), constitute one of the oldest records of the earliest hominid population in the European continent west of Dmanisi (Georgia, Lesser Caucasus). In the Guadix-Baza Basin, evidence of human occupation has been found to date in the form of lithic industry (Mode 1) and cut marks in large-mammal fossil remains (mainly of hippopotamuses and elephants), and recently a human tooth considered as the oldest in Europe has been discovered. Although in the case of Sima del Elefante there is unanimity among the scientific community regarding the chronology of the unit in which the hominid remains were found (Unit TE9c, 1.22 Ma), there is continuing debate on the chronology of the sites of the Guadix-Baza Basin (FN-3 and BL-D). This applies especially to BL-D, as the numerical datings published for this site have a very high error range (1.4 ± 0.38 Ma). In this paper, the chronology of these two sites is determined using as a marker the morphological and morphometric changes undergone by Mimomys savini in its first lower molar (m1) over the course of its evolutionary history. It has been possible to confirm that the oldest human presence in the Guadix-Baza Basin and at Sima del Elefante (Atapuerca) share a similar chronology, dated to between 1.1 and 1.4 Ma. Apparently, the oldest site with human remains in Europe is seen to be BL-D, dated to 1.26 ± 0.13 Ma, followed by Level TE9c, dated to 1.22 ± 0.16 Ma, and FN-3, dated to 1.20 ± 0.12 Ma.  相似文献

We present here the results of 44 paleomagnetic measurements, and single cosmogenic burial and optically stimulated luminescence ages for the Earlier Stone Age deposits from Wonderwerk Cave, Northern Cape, South Africa. The resulting paleomagnetic sequence: N>R>N>R>N constrains the Earlier Stone Age strata in this part of the site to between approximately 0.78-1.96 Ma. A single cosmogenic date of approximately 2.0 Ma from the base of the section offers some corroboration for the paleomagnetic sequence. Preliminary results indicate that the small lithic assemblage from the basal stratum may contain an Oldowan facies. This is overlain by several strata containing Acheulean industries. The preliminary radiometric dates reported here place the onset of the Acheulean at this site to approximately 1.6 Ma, which is roughly contemporaneous with that of East Africa.  相似文献
