SAS-6 is required for centriole biogenesis in diverse eukaryotes. Here, we describe a novel family of SAS-6-like (SAS6L) proteins that share an N-terminal domain with SAS-6 but lack coiled-coil tails. SAS6L proteins are found in a subset of eukaryotes that contain SAS-6, including diverse protozoa and green algae. In the apicomplexan parasite Toxoplasma gondii, SAS-6 localizes to the centriole but SAS6L is found above the conoid, an enigmatic tubulin-containing structure found at the apex of a subset of alveolate organisms. Loss of SAS6L causes reduced fitness in Toxoplasma. The Trypanosoma brucei homolog of SAS6L localizes to the basal-plate region, the site in the axoneme where the central-pair microtubules are nucleated. When endogenous SAS6L is overexpressed in Toxoplasma tachyzoites or Trypanosoma trypomastigotes, it forms prominent filaments that extend through the cell cytoplasm, indicating that it retains a capacity to form higher-order structures despite lacking a coiled-coil domain. We conclude that although SAS6L proteins share a conserved domain with SAS-6, they are a functionally distinct family that predates the last common ancestor of eukaryotes. Moreover, the distinct localization of the SAS6L protein in Trypanosoma and Toxoplasma adds weight to the hypothesis that the conoid complex evolved from flagellar components.  相似文献

SYNOPSIS. An electron microscope study of sporozoites of Eimeria nieschulzi Dieben, 1924 revealed that they have a pellicle which is thickened at the anterior end to form 2 polar rings. Radiating posteriorly from the rings, directly beneath the pellicle, are approximately 25 microtubules which may aid in support and locomotion of the sporozoite. Within the polar ring is a dense conoid. Numerous toxonemes extend posteriorly from the area of the conoid. Two paranuclear bodies are present and some toxonemes are closely associated with the anterior body. Numerous ribosomes, bodies containing granular material, and osmiophilic vesicle bounded bodies are also present. Each sporozoite has a single nucleus with a diffuse karyosome and distinct nuclear double membrane.  相似文献

E. F. v. Schlotheim described 46 species of Jurassic and Cretaceous belemnites in 1813,1820, and 1832. Their type specimens are in part housed at the Humboldt University of Berlin. The holotype ofBelemnites irregularis is here refigured. Lectotypes forB. acuarius, B. canaliculatus, B. clavatus, B. giganteus, andB. tripartitus are designated and figured for the first time. The lectotype ofBelemnites giganteus is designated to be the neotype ofBelemnites suevicus Klein 1773. 36 species are nomina nuda and nomina dubia, because none of their type specimens could be detected, among them the invalidB. paxillosus v. Schlotheim 1820, which is, according to its lectotype designated herein, likeBelemnites giganteus, a junior synonym ofMegateuthis suevica (Klein 1773). An original type specimenof Belemnites mucronatusv. Schlotheim 1813, for which a neotype had already been ruled by the ICZN in 1985, was re-discovered in thev. Schlotheim collection in 1996.v. Summary It is possible to obtain a permanent breeding ofOpius concolor by use of third instar larvae ofCeratitis capitata as host. TheOpius obtained by such breeding will accept to parasitize successfully the larvae ofDacus oleae. This breeding will allow the study of many obscure points of the biology of the parasite, mainly conditions by which the host is chosen by the laying female and the reason of the endoparasitical development in the two hosts. It should be useful to know if the ecological strains are differenciated by similar experimentation withO. siculus Monastero. It should be aslo interesting to extend this experience to other Trypetid species (for instance to the hosts ofOpius humilis Silv.: Braconid which is spread in Hawaii might be a synonym ofconcolor asFischer supposes). This breeding could be eventually used to start the study and realisation of industrial multiplications in view of the biological control applications.   相似文献

The present communication deals with the observations made on the morphology, reproduction and cytology ofUlothrix zonata (Weber & Mohr)Kütz. in culture. The alga displays a remarkable phenotypic plasticity in nature as well as in culture. The present study provides additional evidence of karyology in support ofLokhorst andVroman's treatment ofU. zonata which merges a number of earlier described species in it. The hypothesis ofHenriques andFernandes that several Iberian species ofNarcissus (Amaryllidaceae) are tristylous is reconsidered. Contrary to the opinion ofBateman and most subsequent authors, we believe that the available evidence indicates that some populations ofN. triandrus andN. fernandesii, at least, are tristylous; other populations ofN. triandrus are distylous.Hugonia cf.penicillanthemum (Linaceae) from new Caledonia is distylous, but it remains possible that other species ofHugonia are tristylous. The disputed occurrence of heterostyly in S. African species ofBauhinia (Leguminosae),Cleome (Capparaceae) andAneilema (Commelinaceae), and inAgelaea (Connaraceae) is discussed.  相似文献

The new genus is characterized by the formation of brown to dark colored ascocarps bearing brown appendages with verticels of short branchlets. SYNOPSIS. The sporozoite of Lankesteria culicis was studied by light and electron microscopy, after excystation in the intestine of Aedes aegypti 1st stage larvae. The sporozoite was 9.5–10.0 μ long with a blunt anterior end and a tapered posterior region. The organism was enclosed by a typical pellicle consisting of an outer and an inner membrane with underlying subpellicular microtubules. The anterior end had a conoid with 2 associated rings, a polar ring which served as a termination of the subpellicular microtubules and a flask-shaped structure situated internal and posterior to the conoid. A micropyle consisting of a collar formed from the inner membrane and lacking an invagination of the outer membrane was present near the anterior end of the parasite. The nucleus was centrally located and had a peripheral concentration of chromatin and a central nucleolus. One or more mitochondria were observed in the vicinity of the nucleus.  相似文献

TheSimaroubaceae generally have no true stipules. The stipule-like appendages of some genera proved to be pseudo- or metastipules (Weberling & Leenhouts 1965). There seem to be some exceptions, however: the generaCadellia (incl.Guilfoylia) andRecchia on the one hand, and theIrvingioideae on the other. As these taxa, with exception ofRecchia, have simple leaves, there are no indications that their stipule-like appendages might be pseudo- or metastipules. In regard to their position and ontogeny these appendages behave completely like true stipules. Assuming the view ofForman, one could conceive a morphological line from the long, broadly inserted axillary stipules of mostIrvingioideae to the small scaly triangular stipules ofIxonanthoideae. The similarities between the stipules ofIrvingioideae andErythroxylaceae (already emphasized byHallier and others), become even more evident when their ontogeny is investigated. TheIrvingioideae, therefore, might be regarded as a separate family (perhaps with some relation to theErythroxylaceae,Hallier) or as a subfamily ofIxonanthaceae (Forman).—In addition to data on stipules some results on the palynology and shoot anatomy of the generaCadellia (incl.Guilfoylia) andRecchia are reported. Their relationship with theSimaroubaceae also appears doubtful. If they are to be included, they represent a somewhat isolated group near the base of the family which otherwise has lost its stipules.

Zoospore ultrastructure (incl. flagellar apparatus) has been investigated in three species ofTrebouxia (T. glomerata, T. erici, T. pyriformis) and one species ofPseudotrebouxia (P. impressa) using an absolute configuration analysis. Zoospores in all taxa studied are nearly identical in ultrastructure and exhibit a very distinctive disposition of cell organelles: cells are naked, biflagellate and considerably flattened along the plane of flagellar beat, the single contractile vacuole is located anteriorly in the ventral region of the cell, the nucleus is anteriorly to centrally located in the dorsal region of the cell. A single dictyosome is located close to the anterior, ventral edge of the nucleus. The chloroplast occupies a posterior position in the cell and usually has an anterior profile in the left region of the cell. There are two branched mitochondria per cell or a single mitochondrial reticulum with profiles anterior to the nucleus (in the dorsal region of the cell), and posterior to the nucleus. In zoospores ofTrebouxia spp. the posterior mitochondrial profile is associated with a microbody, inP. impressa zoospores the anterior mitochondrial profiles are associated with a microbody. The zoospores contain a distinctive system of three ER-cisternae: one system links to both basal bodies and extends to the nucleus, the other two systems subtend the plasmamembrane on the left and right broad cell surfaces and extend to the posterior region of the cell. The flagellar apparatus is structurally identical to that previously described for zoospores ofFriedmannia israelensis and exhibits basal body displacement by one basal body diameter into the 11/5 o'clock direction, a non-striated distal connecting fiber, a cruciate microtubular root system lacking system I fibers and presence of a single system II fiber which connects the basal bodies with the nucleus and runs parallel to one of the ER-strands. The left flagellar roots (X-roots) are subtended by a complex set of amorphous and striated material that connects each left root with both basal bodies.—This study demonstrates the close systematic relationship between the phycobiontsTrebouxia andPseudotrebouxia and the generaFriedmannia, Pleurastrum, andMicrothamnion and supports recent classification schemes which place all these taxa into a single order separate from otherChlorophyta. Summary Eight monocentric fungi are reported in two provisionally identified species ofRhizosolenia from sea-water. Only two species are assigned specific names. These areRhizophydium planktonicum Canter andR. fragilariae Canter. A fungus with depressed-globose sporangia is included as a form ofR. fragilariae. Two unidentified species ofChytridium are reported. Summary Growth response ofRhizoctonia solani Kühn, the damping-off fungus, to metabolites of selected antagonistic rhizospheric bacteria and fungi of some Egyptian cotton varieties, namely, two strains ofBacillus subtilis Cohn,Aspergillus terreus Thom, andAspergillus flavus Link produced in culture media containing nitrate- or ammonium-nitrogen sources, proved the potency ofB. subtilis metabolites in inhibitingR. solani mycelial growth whether from nitrate- or ammonium-nitrogen culture media. Metabolite filtrates ofB. subtilis (II) are more potent than those ofB. subtilis (I). Increasing concentration of bacterial metabolite filtrates resulted in a decreased mycelial dry weight ofR. solani. The bacterial inhibitory factor forR. solani mycelial growth is partially affected by heat. Metabolite filtrates ofA. terreus from nitrate-nitrogen are slightly more potent than from ammonium-nitrogen culture media while an opposite relation is evident withA. flavus metabolites. Growth responses ofR. solani to different experimental dilutions of metabolite filtrates ofA. terreus andA. flavus proved the intervention of the nutritive factor in witholding growth of the damping-off fungus.  相似文献

Summary The layings ofCassida deflorata Suffr. are parasited in southern France byMymaridae (Anaphoidea sp. andFulmekiella ovata soyka),Trichogrammatidae (Monorthochaeta nigra Blood.) andEulophidae (Foersterella flavipes Foerst. andTetrastichus rhosaces Walk.). In the Alpes-Maritimes, two successive generations ofMymaridae andTrichogrammatidae seem to attack the layings ofC. deflorata. In Corsica, parasitism byMymaridae andF. flavipes is very frequent and may be the essential regulating factor of the populations ofCassida. In the other regions that have been studied, although the large number ofCassida imagos appearing in spring show that mortality during the aestivation and hibernation is not the primary factor, it does not seem that the limitation of the populations is essentially caused by the activity of oophagous insects.   相似文献

The subspecies ofAustralopithecus africanus Dart, 1925 have been revised in a morphological and statistical analysis. Four subspecific names were previously proposed, but only one was found to be valid. The subspeciesA. africanus transvaalensis (Broom, 1936), from the Plio/Pleistocene of South Africa, cannot be sustained due to an insufficient sample, and is combined with the nominate race,A. a. africanus. The type ofA. africanus afarensis Tobias, 1980 is a mistake in identification and notA. africanus, but a pongid. The population ofA. africanus from the late Pliocene of Ethiopia does indeed represent a relatively small-toothed geographical race for which the nameA. africanus aethiopicus was conditionally proposed; and the lectotype for it, A.L. 288-1, is notA. africanus, but the type ofHomo antiquus Ferguson, 1984. The trinominalaethiopicus is thus unavailable for the Ethiopian race, which is redescribed as a new subspecies,A. africanus miodentatus n. ssp., and the mandible A.L. 266-1 is designated as the holotype.  相似文献

Molecular access to amino acid excretion by Corynebacterium glutamicum and Escherichia coli led to the identification of structurally novel carriers and novel carrier functions. The exporters LysE, RhtB, ThrE and BrnFE each represent the protoype of new transporter families, which are in part distributed throughout all of the kingdoms of life. LysE of C. glutamicum catalytes the export of basic amino acids. The expression of the carrier gene is regulated by the cell-internal concentration of basic amino acids. This serves, for example, to maintain homoeostasis if an excess of l-lysine or l-arginine inside the cell should arise during growth on complex media. RhtB is one of five paralogous systems in E. coli, of which at least two are relevant for l-threonine production. A third system is relevant for l-cysteine production. It is speculated that the physiological function of these paralogues is related to quorum sensing. ThrE of C. glutamicum exports l-threonine and l-serine. However, a ThrE domain with a putative hydrolytic function points to an as yet unknown role of this exporter. BrnFE in C. glutamicum is a two-component permease exporting branched-chained amino acids from the cell, and an orthologue in B. subtilis exports 4-azaleucine.  相似文献
