This study evaluated the effect of sludge age on simultaneous nitrification and denitrification in a membrane bioreactor treating black water. A membrane bioreactor with no separate anoxic volume was operated at a sludge age of 20 days under low dissolved oxygen concentration of 0.1-0.2 mg/L. Its performance was compared with the period when the sludge age was adjusted to 60 days. Floc size distribution, apparent viscosity, and nitrogen removal differed significantly, together with different biomass concentrations: nitrification was reduced to 40% while denitrification was almost complete. Modelling indicated that both nitrification and denitrification kinetics varied as a function of the sludge age. Calibrated values of half saturation coefficients were reduced when the sludge age was lowered to 20 days. Model simulation confirmed the validity of variable process kinetics for nitrogen removal, specifically set by the selected sludge age.  相似文献

The heterotrophic biomass has the capacity of utilizing substrate predominantly for growth or storage processes under steady-state conditions. In this study, the short-term variations in growth and storage kinetics of activated sludge under disturbed feeding conditions were analyzed using a multi-component biodegradation model. The variations in growth and storage kinetics were investigated with the aid of multi-response modeling and identifiability analysis. It was found that the heterotrophic biomass is able to increase its direct growth activity together with reducing the substrate storage capability under the availability of external substrate. Reducing the sludge age (SRT) from 10 to 2?days increased the maximum specific growth rate, μ (OHO,Max) from 3.9 to 7.0 day(-1), but did not considerably affected the maximum storage rate, k (Stor,OHO). The alteration of sludge age also elevated the half-saturation constant for growth (K (S,OHO)) from 5 to 25?mg COD/L. The increase in primary growth metabolism together with reduced storage rate was validated by model for two different sludge ages in the availability of external substrate. Aside from having a lower storage capability, the biomass had fast adaptation ability to direct growth process at low SRTs. The alteration of feed conditions was found to have different impacts on storage and growth kinetics. These results are significant and advance the field of activated sludge modeling under dynamic conditions by incorporation of short-term effects. Appropriate modifications including short-term effects in model structure may also reduce dynamic model recalibration efforts in the future.  相似文献

The influence of sludge age on granular sludge formation and microbial population dynamics in a methanol- and acetate-fed aerobic granular sludge system operated at 35°C was investigated. During anaerobic feeding of the reactor, methanol was initially converted to methane by methylotrophic methanogens. These methanogens were able to withstand the relatively long aeration periods. Lowering the anaerobic solid retention time (SRT) from 17 to 8 days enabled selective removal of the methanogens and prevented unwanted methane formation. In absence of methanogens, methanol was converted aerobically, while granule formation remained stable. At high SRT values (51 days), γ-Proteobacteria were responsible for acetate removal through anaerobic uptake and subsequent aerobic growth on storage polymers formed [so called metabolism of glycogen-accumulating organisms (GAO)]. When lowering the SRT (24 days), Defluviicoccus-related organisms (cluster II) belonging to the α-Proteobacteria outcompeted acetate consuming γ-Proteobacteria at 35°C. DNA from the Defluviicoccus-related organisms in cluster II was not extracted by the standard DNA extraction method but with liquid nitrogen, which showed to be more effective. Remarkably, the two GAO types of organisms grew separately in two clearly different types of granules. This work further highlights the potential of aerobic granular sludge systems to effectively influence the microbial communities through sludge age control in order to optimize the wastewater treatment processes.  相似文献

The sludge exchange process using two anaerobic digesters (CSTRs) in series was investigated under the mesophilic condition (36-38 °C). At first, the digesting sludge of the CSTRs in series with different TVFA/alkalinity ratios was tested in the laboratory by mixing the digesting sludge of two CSTRs from 6.5% to 50% based on volume. The sludge exchange test was then performed using the same CSTRs under batch and continuous processes. The change in the TVFA/alkalinity ratio was found to be linear with the digesting sludge exchange volume. The CSTR of TVFA/alkalinity ratio 1.970 recovered completely failed within 11 days for the batch process and the CSTR of TVFA/alkalinity ratio 1.514 within 3 weeks for the continuous feeding process at a sludge exchange volume of 13%. The reactor operation was stable when the TVFA/alkalinity ratio was less than 1.0 and when the TVFA concentration was lower than 10,000 mg L−1.  相似文献

Microbial degradation of phenol was studied using batch and fedbatch cultures of acclimatized activated sludge under a wide range of phenol (0-793 mg l−1) and biomass (0.74-6.7 g l−1) initial concentrations. As cell growth continued after total phenol removal, the production and later consumption of a main metabolic intermediate was considered the step governing the biodegradation kinetics. A model that takes explicitly into account the kinetics of the intermediate was developed by introducing a specific growth rate model associated with its consumption and the incorporation of a dual-substrate inhibitory effect on phenol degradation. Biomass growth and phenol removal were adequately predicted in all the cultures. Moreover, the model-based design of the fedbatch feeding strategies allowed driving separately the phenol degradation under substrate-limitation and substrate-inhibition modes. A sensitivity analysis was also performed in order to establish the importance of the parameters in the accuracy of model predictions.  相似文献

Cold enzymatic hydrolysis conditions for bioethanol production were optimized using multi-objective optimization. Response surface methodology was used to optimize the effects of α-amylase, glucoamylase, liquefaction temperature and liquefaction time on S. cerevisiae biomass, ethanol concentration and starch utilization ratio. The optimum hydrolysis conditions were: 224 IU/gstarch α-amylase, 694 IU/gstarch glucoamylase, 77 °C and 104 min for biomass; 264 IU/gstarch α-amylase, 392 IU/gstarch glucoamylase, 60 °C and 85 min for ethanol concentration; 214 IU/gstarch α-amylase, 398 IU/gstarch glucoamylase, 79 °C and 117 min for starch utilization ratio. The hydrolysis conditions were subsequently evaluated by multi-objectives optimization utilizing the weighted coefficient methods. The Pareto solutions for biomass (3.655-4.380 × 108 cells/ml), ethanol concentration (15.96-18.25 wt.%) and starch utilization ratio (92.50-94.64%) were obtained. The optimized conditions were shown to be feasible and reliable through verification tests. This kind of multi-objective optimization is of potential importance in industrial bioethanol production.  相似文献

An anoxic/oxic step feeding process was improved to enhance nutrient removal by reconfiguring the process into (1) anaerobic/anoxic/oxic step feeding process or (2) modified University of Capetown (UCT) step feeding process. Enhanced nitrogen and phosphorus removal and optimized organics utilization were obtained simultaneously in the modified UCT type with both internal and sludge recycle ratios of 75% as well as anaerobic/anoxic/oxic volume ratio of 1:3:6. Specifically, the UCT configuration and optimized operational conditions lead to the enrichment of denitrifying phosphorus removal microorganisms and achieved improved anaerobic P-release and anoxic P-uptake activities, which were beneficial to the denitrifying phosphorus removal activities and removal efficiencies. Due to high mixed liquor suspended solid and uneven distributed dissolved oxygen, 35% of total nitrogen was eliminated through simultaneous nitrification and denitrification process in aerobic zones. Moreover, 62 ± 6% of influent chemical oxygen demands was involved in the denitrification or phosphorus release processes.  相似文献

The effect of starch addition on the microbial composition and the biological conversion was investigated using two upflow anaerobic sludge bracket (UASB) reactors treating methanolic wastewater: one reactor was operated with starch addition, and another reactor was operated without starch addition. Approximately 300 days of operation were performed at 30 kg COD/m3/d, and then, the organic load of the reactors was gradually increased to 120 kg COD/m3/d. Successful operation was achieved at 30 kg COD/m3/d in both reactors; however, the methanol-fed reactor did not perform well at 120 kg COD/m3/d while the methanol-starch-fed reactor did. The granule analysis revealed the granule developed further only in the methanol-starch-fed reactor. The results of the microbial community analysis revealed more Methanosaeta cells were present in the methanol-starch-fed reactor, suggesting the degradation of starch produced acetate as an intermediate, which stimulated the growth of Methanosaeta cells responsible for the extension of granules.  相似文献

The present study investigates the formation of aerobic granular sludge in sequencing batch reactor (SBR) fed with palm oil mill effluent (POME). Stable granules were observed in the reactor with diameters between 2.0 and 4.0 mm at a chemical oxygen demand (COD) loading rate of 2.5 kg COD m−3 d−1. The biomass concentration was 7600 mg L−1 while the sludge volume index (SVI) was 31.3 mL g SS−1 indicating good biomass accumulation in the reactor and good settling properties of granular sludge, respectively. COD and ammonia removals were achieved at a maximum of 91.1% and 97.6%, respectively while color removal averaged at only 38%. This study provides insights on the development and the capabilities of aerobic granular sludge in POME treatment.  相似文献

Summary With the aim of studying the possible utilization of brewery waste water activated sludge for animal feeding, the influence of the solids retention time (SRT) and nitrogen supplementation were investigated, especially with respect to biomass production and biomass composition. It was found that the SRT strongly influenced both parameters. At an SRT of from 4 to 6 days excellent biomass production was obtained. This biomass had the highest protein content and the daily protein production was four times higher than at a SRT of 20 days. Supplementation with urea doubled the protein production, lowered the carbohydrate and poly--hydroxybutyric acid content, but increased the nucleic acid content. The COD removal was better and phosphorus removal increased. In order to study these variables, a multi-channel laboratory system was designed. Because of its simplicity in operation and its versatility this system is described in detail.  相似文献

The study investigated acute impact of tetracycline and erythromycin on substrate storage under aerobic conditions. A fill and draw reactor fed with peptone mixture was maintained at steady-state at a sludge age of 10 days; the acclimated biomass was used in a series of batch runs. The first run served as control reactor with organic substrate alone and the others were started with antibiotic doses of 50 mg/L and 200 mg/L for assessing intracellular storage. Parallel batch reactors were also conducted for recording oxygen uptake rate profiles. Both antibiotics enhanced substrate storage, leading to higher levels of polyhydroxyalkanoates incorporated into biomass, but they impaired its internal utilization for microbial growth. The observed decrease in oxygen consumption under the acute effect of antibiotics could partially be related to substrate storage – except for 50 mg/L of erythromycin dosing – suggesting an additional substrate binding mechanism by antibiotics, leading to residual biodegradable substrate.  相似文献

The goal of the study was to determine the effectiveness of nitrification and denitrification and the kinetics of ammonia removal from a mixture of wastewater and anaerobic sludge digester supernatant in an SBR at limited oxygen concentration. In addition, the COD removal efficiency and sludge production were assessed.In the SBR cycle alternating aerobic and anaerobic phases occurred; in the aeration phase the dissolved oxygen (DO) concentration was below 0.7 mg O2/L. The low DO concentration did not inhibit ammonia oxidation-nitrification and the efficiency was ca. 96-98%. However, a relatively high COD concentration in the effluent was detected. The values of Km and Vmax, calculated from the Michaelis-Menten equation, were 43 mg N-NH4/L and 15.64 mg N-NH4/L h, respectively. Activated sludge production was almost stable (0.62-0.66 g MLVSS/g COD). A high net biomass production resulted from a low specific biomass decay rate of 0.0015 d−1.  相似文献

The potential use of activated sludge for the production of medium-chain-length polyhydroxyalkanoates (MCL-PHAs) was investigated. The enrichment of bacterial populations capable of producing MCL-PHAs was achieved by periodic feeding with nonanoic acid in a sequencing batch reactor (SBR). Denaturing gradient gel electrophoresis analysis revealed Pseudomonas aeruginosa strains to be predominant in the bacterial community during the SBR process. The composition of PHA synthesized by the enriched biomass from nonanoic acid consisted of a large concentration (>89 mol%) of MCL monomer units and a small amount of short-chain-length monomer units. Under fed-batch fermentation with continuous feeding of nonanoic acid at a flow rate of 0.225 g/L/h and a C/N ratio of 40, a maximum PHA content of 48.6% dry cell weight and a conversion yield (Yp/s) of 0.94 g/g were achieved. These results indicate that MCL-PHA production by activated sludge is a promising alternative to typical pure culture approaches.  相似文献

The anaerobic ammonium oxidation (anammox) process has attracted considerable attention in recent years as an alternative to conventional nitrogen removal technologies. In this study, an innovative hybrid reactor combining fluidized and fixed beds for anammox treatment was developed. The fluidized bed was mechanically stirred and the gaseous product could be rapidly released from the anammox sludge to prevent washout of the sludge caused by floatation. The fixed bed comprising a non-woven biomass carrier could efficiently catch sludge to reduce washout. During the operation, nitrogen loading rates to the reactor were increased to 27.3 kg N/m3/d, with total nitrogen removal efficiencies of 75%. The biomass concentration in the fluidized bed reached 26-g VSS/L. Anammox granules were observed in the reactors, with settling velocities and sludge volumetric index of 27.3 ± 6.5 m/h and 23 mL/g, respectively. Quantification of extracellular polymeric substances revealed the anammox granules contained a significant amount of extracellular proteins.  相似文献

Two sets of reactors were operated at 15 °C and at sludge retention times (SRTs) of 65 days and 30 days, respectively. Each set was operated at six different mixing velocities. Shear forces provided by mixing affected particle size distribution and resulted in different sludge surface areas. The aim was to investigate the effect of increasing primary sludge surface area on anaerobic digestion at low temperature. The maximum surface areas at the applied mixing velocities were 5926 cm2/cm3of sludge and 4672 cm2/cm3 of sludge at SRTs of 65 days and 30 days, respectively. The corresponding calculated methanogenesis were 49% and 15% at SRTs of 65 days and 30 days, respectively. Maximum total solids (TS) reductions were 26% and 11% at 65 days and 30 days SRTs, respectively. Lipase activity increased with increasing SRT and sludge surface area. Results clearly showed that increasing sludge surface area improved sludge digestion at 15 °C.  相似文献

The biological phosphorus removal process is a process which depends basically on three internal storage compounds. Poly-beta-hydroxybutyrate (PHB) produced during the anaerobic phase is used as substrate for biomass, polyphosphate, and glycogen formation. The reaction rates of the aerobic processes are primarily determined by the PHB content of the cells. This PHB content is highly dynamic due to the conversions during the anaerobic and aerobic phase of the cycle and the ratio between substrate addition and biomass present in the reactor. The amount of biomass present in the reactor is determined by the sludge retention time and growth rate. A metabolic model of the biological phosphorus removal process was developed and verified over a wide range of growth rates. The effect of different growth rates on the internal fractions of stored components was determined and described mathematically. One set of kinetic parameters was capable of describing the measured conversions of all components observed in the reactor as a function of the sludge retention time. This research for the first time investigated hydrogen production from the fresh leachate originated from municipal solid wastes. We found that fermentation of the leachate generated H2 and was very much enhanced in the presence of extra phosphate in the batch reactor. The continuous expanded granular sludge bed (EGSB) reactor started to generate H2 at day 20 and continued to 176 days with 120 mg/l of extra phosphate present. The highest chemical oxygen demand (COD) removal efficiency (66.9%) was achieved at liquid up-flow velocity of 3.7 m/h and hydraulic retention time of 12 h. Under proposed optimal operation conditions, the mean H2 production rate reached up to 2155 ml/(l day). We also found that over 80% liquid metabolites were acetic acid and ethanol, suggesting the ethanol-type fermentation was dominant in the bioreactor. These findings indicate that the fresh leachate can be used as the source for continuous hydrogen production.  相似文献

A structured metabolic model is developed that describes the stoichiometry and kinetics of the biological P removal process. In this approach all relevant metabolic reactions underlying the metabolism, considering also components like adenosine triphosphate (ATP) and nic-otinamide-adenine dinucleotide (NADH(2)) are describedbased on biochemical pathways. As a consequence of the relations between the stoichiometry of the metabolic reactions and the reaction rates of components, the required number of kinetic relations to describe the process is reduced. The model describes the dynamics of the storage compounds which are considered separately from the active biomass. The model was validated in experiments at a constant sludge retention time of 8 days, over the anaerobic and aerobic phases in which the external oncentrations as well as the internal fractions of the relevant components involved in the P-removal process were monitored. These measurements include dissolved acetate, phosphate, and ammonium; oxygen consumption; poly-beta-hydroxybutyrate (PHB); glycogen; and active biomass. The effects of continuous and intermittent feeding strategies on nitrogen removal and N2O emission from surface flow and subsurface flow constructed wetlands were evaluated in this study. Microcosm wetlands planted with Phragmites australis were constructed and operated with different feeding strategies for the 4-month experiment. Results showed the intermittent feeding strategy could enhance the removal of ammonium effectively in the subsurface flow constructed wetlands, although it had no significant effect for the surface flow wetlands. And the intermittent feeding mode could promote the emission of N2O. The amount of N2O-N emission from the subsurface flow constructed wetlands with intermittent feeding mode was about 5 times higher than that with continuous feeding strategy and the emission rate ranged from 0.09 ± 0.03 to 7.33 ± 1.49 mg/m2/h. Compared with the surface flow constructed wetlands, the N2O emission in the subsurface flow constructed wetlands was affected significantly by the intermittent feeding mode.  相似文献
