Based on the ability of phytosiderophores to chelate other heavy metals besides iron (Fe), phytosiderophores were suggested to prevent graminaceous plants from cadmium (Cd) toxicity. To assess interactions between Cd and phytosiderophore-mediated Fe acquisition, maize (Zea mays) plants were grown hydroponically under limiting Fe supply. Exposure to Cd decreased uptake rates of 59Fe(III)-phytosiderophores and enhanced the expression of the Fe-phytosiderophore transporter gene ZmYS1 in roots as well as the release of the phytosiderophore 2'-deoxymugineic acid (DMA) from roots under Fe deficiency. However, DMA hardly mobilized Cd from soil or from a Cd-loaded resin in comparison to the synthetic chelators diaminetriaminepentaacetic acid and HEDTA. While nano-electrospray-high resolution mass spectrometry revealed the formation of an intact Cd(II)-DMA complex in aqueous solutions, competition studies with Fe(III) and zinc(II) showed that the formed Cd(II)-DMA complex was weak. Unlike HEDTA, DMA did not protect yeast (Saccharomyces cerevisiae) cells from Cd toxicity but improved yeast growth in the presence of Cd when yeast cells expressed ZmYS1. When supplied with Fe-DMA as a Fe source, transgenic Arabidopsis (Arabidopsis thaliana) plants expressing a cauliflower mosaic virus 35S-ZmYS1 gene construct showed less growth depression than wild-type plants in response to Cd. These results indicate that inhibition of ZmYS1-mediated Fe-DMA transport by Cd is not related to Cd-DMA complex formation and that Cd-induced phytosiderophore release cannot protect maize plants from Cd toxicity. Instead, phytosiderophore-mediated Fe acquisition can improve Fe uptake in the presence of Cd and thereby provides an advantage under Cd stress relative to Fe acquisition via ferrous Fe.  相似文献

? The direct analysis of phytosiderophores (PSs) and their metal complexes in plants is critical to understanding the biological functions of different PSs. Here we report on a rapid and highly sensitive liquid chromatography-electrospray ionization-quadrupole-time of flight-mass spectrometry (LC-ESI-Q-TOF-MS) method for the direct and simultaneous determination of free PSs and their ferric complexes in plants. ? In addition to previously reported PSs - deoxymugineic acid (DMA), mugineic acid (MA) and epihydroxymugineic acid (epi-HMA) - two more PSs, avenic acid (AVA) and hydroxyavenic acid (HAVA), were identified by this method in roots of Hordeum vulgare cv Himalaya and in root exudates under iron (Fe) deficiency. ? The two identified PSs could be responsible for Fe acquisition under Fe deficiency because of their relative abundance and ability to form ferric complexes in secreted root exudates. ? This LC-ESI-Q-TOF-MS method greatly facilitates the identification of free PSs and PS-Fe complexes in one plant sample.  相似文献

Peanut/maize intercropping is a sustainable and effective agroecosystem that evidently enhances the Fe nutrition of peanuts in calcareous soils. So far, the mechanism involved in this process has not been elucidated. In this study, we unravel the effects of phytosiderophores in improving Fe nutrition of intercropped peanuts in peanut/maize intercropping. The maize ys3 mutant, which cannot release phytosiderophores, did not improve Fe nutrition of peanut, whereas the maize ys1 mutant, which can release phytosiderophores, prevented Fe deficiency, indicating an important role of phytosiderophores in improving the Fe nutrition of intercropped peanut. Hydroponic experiments were performed to simplify the intercropping system, which revealed that phytosiderophores released by Fe‐deficient wheat promoted Fe acquisition in nearby peanuts and thus improved their Fe nutrition. Moreover, the phytosiderophore deoxymugineic acid (DMA) was detected in the roots of intercropped peanuts. The yellow stripe1‐like (YSL) family of genes, which are homologous to maize yellow stripe 1 (ZmYS1), were identified in peanut roots. Further characterization indicated that among five AhYSL genes, AhYSL1, which was localized in the epidermis of peanut roots, transported Fe(III)–DMA. These results imply that in alkaline soil, Fe(III)–DMA dissolved by maize might be absorbed directly by neighbouring peanuts in the peanut/maize intercropping system.  相似文献

Plants require minimal amounts of certain metals (Zn,Fe,Cu,etc) for optimal growth and productivity, but excess of these metals leads to cell death. When growth is limited by metal excess or metal deficiency plants respond by synthesizing nonproteinogenic chelating substances. Phytosiderophores are secreted by roots of iron deficient grasses and are important in providing sufficient Fe for normal growth. In response to growth-inhibitory levels of heavy metals plants synthesize metal-binding phytochelatins which detoxify excess metals. Biostimulants such as humic substances and oligomers of lactic acid have properties in common with both phytosiderophores and phytochelatins. The word phytochelates is proposed as a generic term to cover substances that affect plant growth by acting as chelating agents.  相似文献

Roots of grasses in response to iron deficiency markedly increase the release of chelating substances (`phytosiderophores') which are highly effective in solubilization of sparingly soluble inorganic FeIII compounds by formation of FeIIIphytosiderophores. In barley (Hordeum vulgare L.), the rate of iron uptake from FeIIIphytosiderophores is 100 to 1000 times faster than the rate from synthetic Fe chelates (e.g. Fe ethylenediaminetetraacetate) or microbial Fe siderophores (e.g. ferrichrome). Reduction of FeIII is not involved in the preferential iron uptake from FeIIIphytosiderophores by barley. This is indicated by experiments with varied pH, addition of bicarbonate or of a strong chelator for FeII (e.g. batho-phenanthrolinedisulfonate). The results indicate the existence of a specific uptake system for FeIIIphytosiderophores in roots of barley and all other graminaceous species. In contrast to grasses, cucumber plants (Cucumis sativus L.) take up iron from FeIIIphytosiderophores at rates similar to those from synthetic Fe chelates. Furthermore, under Fe deficiency in cucumber, increased rates of uptake of FeIIIphytosiderophores are based on the same mechanism as for synthetic Fe chelates, namely enhanced FeIII reduction and chelate splitting. Two strategies are evident from the experiments for the acquisition of iron by plants under iron deficiency. Strategy I (in most nongraminaceous species) is characterized by an inducible plasma membrane-bound reductase and enhancement of H+ release. Strategy II (in grasses) is characterized by enhanced release of phytosiderophores and by a highly specific uptake system for FeIIIphytosiderophores. Strategy II seems to have several ecological advantages over Strategy I such as solubilization of sparingly soluble inorganic FeIII compounds in the rhizosphere, and less inhibition by high pH. The principal differences in the two strategies have to be taken into account in screening methods for resistance to `lime chlorosis'.  相似文献

Deoxymugineic acid (DMA) is a member of the mugineic acid family phytosiderophores (MAs), which are natural metal chelators produced by graminaceous plants. Rice secretes DMA in response to Fe deficiency to take up Fe in the form of Fe(III)–MAs complex. In contrast with barley, the roots of which secrete MAs in response to Zn deficiency, the amount of DMA secreted by rice roots was slightly decreased under conditions of low Zn supply. There was a concomitant increase in endogenous DMA in rice shoots, suggesting that DMA plays a role in the translocation of Zn within Zn-deficient rice plants. The expression of OsNAS1 and OsNAS2 was not increased in Zn-deficient roots but that of OsNAS3 was increased in Zn-deficient roots and shoots. The expression of OsNAAT1 was also increased in Zn-deficient roots and dramatically increased in shoots; correspondingly, HPLC analysis was unable to detect nicotianamine in Zn-deficient shoots. The expression of OsDMAS1 was increased in Zn-deficient shoots. Analyses using the positron-emitting tracer imaging system (PETIS) showed that Zn-deficient rice roots absorbed less 62Zn-DMA than 62Zn2+. Importantly, supply of 62Zn-DMA rather than 62Zn2+ increased the translocation of 62Zn into the leaves of Zn-deficient plants. This was especially evident in the discrimination center (DC). These results suggest that DMA in Zn-deficient rice plants has an important role in the distribution of Zn within the plant rather than in the absorption of Zn from the soil. To determine the Fe inefficiency factors in the maize mutant ys1 (Zea mays L. cv Yellow Stripe), root exudates of Fe-inefficient ys1 and of two Fe-efficient maize cultivars (Alice, WF9) were collected in axenic nutrient solution cultures. Analysis by thin-layer chromatography and high-performance liquid chromatography revealed that under Fe deficiency ys1 released the phytosiderophore 2[prime]-deoxymugineic acid (DMA) in quantities similar to those of Alice and WF9. Under nonaxenic conditions, DMA released by plants of all three cultivars was rapidly decomposed by microorganisms in the nutrient solution. Uptake experiments with 59Fe-labeled DMA, purified from root exudates of either Fe-deficient Alice or ys1 plants, showed up to 20 times lower uptake and translocation of 59Fe in ys1 than in Alice or WF9 plants. The presence of microorganisms during preculture and short-term uptake experiments had no significant effect on uptake and translocation rates of 59Fe in Alice and ys1 plants. We conclude that Fe inefficiency in the maize mutant ys1 is the result of a defect in the uptake system for Fe-phytosiderophores.  相似文献

It has been shown previously (Treeby et al., 1989) that phytosiderophores, released by roots of iron deficient grasses (Gramineae), mobilize from calcareous soils not only iron (Fe) but also zinc (Zn), manganese (Mn) and copper (Cu). Mobilization of Fe may therefore be impaired by other micronutrient cations. This has been studied in both, model experiments with Fe hydroxide and with a calcareous soil (15% CaCO3, pH 8.6) amended with micronutrients as sulfate salts.Mobilization of Fe from Fe hydroxide by phytosiderophores (epi-3-hydroxymugineic acid) was not affected by the addition of CaCl2, MgSO4 and MnSO4, slightly inhibited by ZnSO4 and strongly inhibited by CuSO4. In a calcareous soil amended with increasing levels of ZnSO4, MnSO4 and CuSO4, mobilization of Fe by phytosiderophores remained uneffected by Zn and Mn amendments but was progressively impaired by increasing levels of Cu amendment, correlated with corresponding enhancement of Cu mobilization.High concentrations of ZnSO4 and MnSO4 and relatively high concentrations of CuSO4 were required for inhibition of Fe mobilization by phytosiderophores. It is therefore concluded that in most calcareous soils phytosiderophores efficiently mobilize Fe, and that phytosiderophores play an important role in Fe acquisition by grasses grown on calcareous soils.  相似文献

Under iron deficiency the release of so-called phytosiderophores by roots of barley plants ( Hordeum vulgare L. cv. Europa) was greater by a factor of 10 to 50 compared to iron-sufficient plants. This enhanced release occurred particularly in apical zones of the seminal roots and in the lateral root zones. Under iron deficiency, uptake rates for iron, supplied as FeIII phytosiderophore, increased by a factor of ca 5 as compared to iron-sufficient plants. This enhanced uptake rate for iron was also much more pronounced in apical than in basal root zones. In contrast, with supply of the synthetic iron chelate, FelII EDDHA (ferric diaminoethane-N, N-di- o -hydroxyphenyl acetic acid), the Fe deficiency-enhanced uptake rates for iron were only small and similar along the roots, except for the lateral root zones. The high selectivity of barley roots for uptake and translocation of FeIII phytosiderophores compared with FeIII EDDHA is reflected by the fact that, at the same external concentration (2 μ M ), rates of uptake and translocation of iron from FeIII phytosiderophores were between 100 (Fe-sufficient) and 1 000 times higher (Fe-deficient plants) than from FeIII EDDHA. The relatively high rates of uptake and particularly of translocation of iron supplied as FeIII EDDHA in the zone of lateral root formation strongly suggest an apoplastic pathway of radial transport of the synthetic iron chelate into the stele in this root zone.
The results demonstrate that apical root zones are the main sites both for Fe deficiency-enhanced release of phytosiderophores and for uptake and translocation of iron supplied as FeIII phytosiderophores.  相似文献

Iron (Fe) is an essential microelement for plant growth. Fe availability is particularly limited on calcareous soils, which have high pH. Approximately 30% of the world's soils are considered calcareous with low Fe availability, which results in extensive areas of Fe deficiency in plants. Some graminaceous plants are known to secrete high amounts of mugineic acid family phytosiderophores (MAs) under Fe deficiency. This Fe acquisition system is called the Strategy-II mechanism. Tolerance to Fe deficiency in graminaceous plants is thought to depend on the quantity of MAs secreted by plants under Fe deficiency stress. This system was utilized to enhance the tolerance of rice to low Fe availability. Transgenic rice expressing the barley naat genes, one of the genes for the enzymes on the biosynthetic pathway of MAs, showed tolerance to low Fe availability when grown in a calcareous soil.  相似文献

The purpose of this study was the elucidation of the chemical mechanism of an important process in iron acquisition by graminaceous plants: the dissolution of iron oxides in the presence of phytosiderophores. We were particularly interested in the effects of diurnal root exudation of phytosiderophores and of the presence of other organic ligands in the rhizosphere of graminaceous plants on the dissolution mechanism. Phytosiderophores of the type 2′-deoxymugineic acid (DMA) were purified from the root exudates of wheat plants (Triticum aestivum L. cv. Tamaro). DMA-promoted dissolution of goethite under steady-state and non-steady-state conditions and its dependence on pH, adsorbed DMA concentration, and the presence of the organic ligand oxalate were studied. We show that dissolution of goethite by phytosiderophores follows a surface controlled ligand promoted dissolution mechanism. We also found that oxalate, an organic ligand commonly found in rhizosphere soils, has a synergistic effect on the steady-state dissolution of goethite by DMA. Under non-steady-state addition of the phytosiderophore, mimicking the diurnal exudation pattern of phytosiderophore release, a fast dissolution of iron is triggered in the presence of oxalate. To investigate the efficiency of these mechanisms in plant iron acquisition, wheat plants were grown on a substrate amended with goethite as only iron source. The chlorophyll status of these plants was similar to iron-fertilized plants and significantly higher than in plants grown in iron free nutrient solutions. This demonstrates that wheat can efficiently mobilize iron, even from well crystalline goethite that is usually considered unavailable for plant nutrition.  相似文献

Two different types of root response to Fe deficiency (strategies) have been identified in species of the Plant Kingdom. In Strategy I which occurs in all plant species except grasses, a plasma membrane-bound reductase is induced with enhanced net excretion of protons. Often the release of reductants/chelators is also higher. In Strategy II which is confined to grasses, there is an increase in the biosynthesis and secretion of phytosiderophores which form chelates with FeIII. Uptake of FeIII phytosiderophores is mediated by a specific transporter in the plasma membrane of root cells of grasses. From results based mainly on long-term studies under non-axenic conditions this classification into two strategies has been questioned, and the utilization of Fe from microbial siderophores has been considered as an alternative strategy particularly in grasses. Possible reasons for controversial results are discussed in some detail. The numerous effects of microorganisms in non-axenic cultures, and the as yet inadequate characterization of the so-called standard (basic) reductase present major limitations to understanding different mechanisms of Fe acquisition. In comparison with the progress made in identifying the cellular mechanisms of root responses in Strategy I and Strategy II plants, our understanding is poor concerning the processes taking place in the apoplasm of root rhizodermal cells and of the role of low-molecular-weight root exudates and siderophores in Fe acquisition of plants growing in soils of differing Fe availability.  相似文献

Nicotianamine synthase (NAS) is an enzyme that is critical for the biosynthesis of the mugineic acid family of phytosiderophores in graminaceous plants, and for the homeostasis of metal ions in nongraminaceous plants. We isolated one genomic NAS clone, ZmNAS3, and two cDNA NAS clones, ZmNAS1 and ZmNAS2, from maize (Zea mays cv Alice). In agreement with the increased secretion of phytosiderophores with Fe deficiency, ZmNAS1 and ZmNAS2 were positively expressed only in Fe-deficient roots. In contrast, ZmNAS3 was expressed under Fe-sufficient conditions, and was negatively regulated by Fe deficiency. This is the first report describing down-regulation of NAS gene expression in response to Fe deficiency in plants, shedding light on the role of nicotianamine in graminaceous plants, other than as a precursor in phytosiderophore production. ZmNAS1-green fluorescent protein (sGFP) and ZmNAS2-sGFP were localized at spots in the cytoplasm of onion (Allium cepa) epidermal cells, whereas ZmNAS3-sGFP was distributed throughout the cytoplasm of these cells. ZmNAS1 and ZmNAS3 showed NAS activity in vitro, whereas ZmNAS2 showed none. Due to its duplicated structure, ZmNAS2 was much larger (65.8 kD) than ZmNAS1, ZmNAS3, and previously characterized NAS proteins (30-38 kD) from other plant species. Poaceae plants release phytosiderophores into the rhizosphere in order to chelate iron (Fe), which often exists in insoluble forms especially under high pH conditions. The impact of phytosiderophore treatment at the physiological and molecular levels in vivo remains largely elusive, although the biosynthesis of phytosiderophores and the transport of phytosiderophore-metal complexes have been well studied. We recently showed that the application of 30 μM of the chemically synthesized phytosiderophore 2′-deoxymugineic acid (DMA) was sufficient for apparent full recovery of otherwise considerably reduced growth of hydroponic rice seedlings at high pH. Moreover, unexpected induction of high-affinity nitrate transporter gene expression as well as nitrate reductase activity indicates that the nitrate response is linked to Fe homeostasis. These data shed light on the biological relevance of DMA not simply as a Fe chelator, but also as a trigger that promotes plant growth by reinforcing nitrate assimilation.  相似文献

Aims

To test if multi–surface models can provide a soil-specific prediction of metal mobilization by phytosiderophores (PS) based on the characteristics of individual soils.

Methods

Mechanistic multi-surface chemical equilibrium modeling was applied for obtaining soil-specific predictions of metal and PS speciation upon interaction of the PS 2’-deoxymugineic acid (DMA) with 6 soils differing in availability of Fe and other metals. Results from multi-surface modeling were compared with empirical data from soil interaction experiments.

Results

For soils in which equilibrium was reached during the interaction experiment, multi-surface models could well predict PS equilibrium speciation. However, in uncontaminated calcareous soils, equilibrium was not reached within a week, and experimental and modeled DMA speciation differed considerably. In soils with circum-neutral pH, on which Fe deficiency is likely to occur, no substantial Fe mobilization by DMA was predicted. However, in all but the contaminated soils, Fe mobilization by DMA was observed experimentally. Cu and Ni were the quantitatively most important metals competing with Fe for complexation and mobilization by DMA.

Conclusion

Thermodynamics are unable to explain the role of PS as Fe carrier in calcareous soils, and the kinetic aspects of metal mobilization by PS need to be closer examined in order to understand the mechanisms underlying strategy II Fe acquisition.  相似文献

Graminaceous plants have evolved a unique mechanism to acquire iron through the secretion of a family of small molecules, called mugineic acid family phytosiderophores (MAs). All MAs are synthesized from l-Met, sharing the same pathway from l-Met to 2'-deoxymugineic acid (DMA). DMA is synthesized through the reduction of a 3'-keto intermediate by deoxymugineic acid synthase (DMAS). We have isolated DMAS genes from rice (OsDMAS1), barley (HvDMAS1), wheat (TaD-MAS1), and maize (ZmDMAS1). Their nucleotide sequences indicate that OsDMAS1 encodes a predicted polypeptide of 318 amino acids, whereas the other three orthologs all encode predicted polypeptides of 314 amino acids and are highly homologous (82-97.5%) to each other. The DMAS proteins belong to the aldo-keto reductase superfamily 4 (AKR4) but do not fall within the existing subfamilies of AKR4 and appear to constitute a new subfamily within the AKR4 group. All of the proteins showed DMA synthesis activity in vitro. Their enzymatic activities were highest at pH 8-9, consistent with the hypothesis that DMA is synthesized in subcellular vesicles. Northern blot analysis revealed that the expression of each of the above DMAS genes is up-regulated under iron-deficient conditions in root tissue, and that of the genes OsDMAS1 and TaDMAS1 is up-regulated in shoot tissue. OsDMAS1 promoter-GUS analysis in iron-sufficient roots showed that its expression is restricted to cells participating in long distance transport and that it is highly up-regulated in the entire root under iron-deficient conditions. In shoot tissue, OsDMAS1 promoter drove expression in vascular bundles specifically under iron-deficient conditions.  相似文献

Nicotianamine is an intermediate for the biosynthesis of mugineic acid-family phytosiderophores (MAs) in the Gramineae and a key substance for iron metabolism in dicots. Nicotianamine synthase catalyzes the formation of nicotianamine from S-adenosylmethionine. Nicotianamine synthase activity was induced in barley roots at the 3rd day after withholding Fe supply and declined within one day followmg the supply of Fe3+-epihydroxymugineic acid. The induction of nicotianamine synthase activity by Fe-deficiency was observed also in sorghum, maize, and rye, and the level of nicotianamine synthase activity was highly associated with the MAs secreted among graminaceous plant tested. Phytosiderophores released by roots of iron-deficient grasses mobilise Fe, Zn, Mn and Cu in calcareous soils. Mobilisation of Fe, Zn and Cu can be explained as the chelation of these metal cations by phytosiderophores. Mobilisation of Mn could not be so explained because phytosiderophores have a much smaller affinity for Mn than for Fe, Cu and Zn. Model experiments have been made with freshly precipitated Fe(OH)3 and different soils to study the mobilisation of iron and manganese by plant-borne chelating phytosiderophores, the synthetic metal chelators DTPA and the microbial metal chelator sulphonated ferrioxamine B (FOB). Compared with the synthetic chelator DTPA, the plant-borne chelating phytosiderophores mobilised Fe very efficiently, but no change was observed in the Mn mobilisation by phytosiderophores.Different phytosiderophores, as well as the microbial metal chelator FOB, were used to compare the mobilisation of iron and manganese in a calcareous soil.  相似文献
