Two strains of sulfate-reducing bacteria of the genus Desulfovibrio (A2 and TomC) isolated from metal mining waste were able to grow on agar Postgate C nutrient medium under microaerobic conditions. Since their growth in liquid nutrient medium was just slightly affected by 1% O2 (initial concentration in the gas phase) and 0.05–0.1 mM H2O2, these strains were relatively oxygen-tolerant. Only the presence of oxidants in high concentrations (5–10% О2 or 0.3–1.0 mM H2O2) resulted in practically complete inhibition of their growth. Strain A2 was more resistant to oxidative stresses than strain TomC. Activities of the key enzymes of antioxidant defense—superoxide dismutase (SOD), catalase, and peroxidase—were revealed in the cell-free extracts of strain A2 grown under strict anaerobic conditions. While strain TomC was found to possess no peroxidase activity, its catalase activity was much higher than that of strain A2 (36 and 2 U/mg protein, respectively). SOD activity of both strains was almost the same (5 U/mg protein). Sublethal H2O2 doses (concentration of 0.05–0.15 mM and exposure for 45–240 min) resulted in a drastic increase of catalase activity, especially in strain A2. Sublethal О2 doses (1–2% in the gas phase) had no significant effect on activities of the antioxidant enzymes of both strains. The cytochrome composition determined from the absolute absorption spectra of the whole cells of strains TomC and A2 revealed the presence of the c heme (438 and 831 pmol/mg protein) and the d heme (336 and 303 pmol/mg protein, respectively). The presence of the d heme indicated the presence of the bd heme–heme quinol oxidase, which together with the c heme may provide for the functioning of the electron transport segment of the antioxidant defensive system, which is responsible for aerotolerance of sulfate-reducing bacteria.  相似文献

The objective of this study was to investigate the effect of different concentrations of methyl salicylate (MeSA) on direct defense and indirect defense in poplar cuttings (Populus × euramericana ‘Nanlin 895’). Four defense-related enzyme activities, such as superoxide dismutase (SOD, EC1.15.1.1), oxydoreductases peroxidase (POD, EC1.11.1.7), catalase (CAT, EC1.11.1.6), and polyphenol oxidase (PPO, EC 1.14.18.1), were measured. The results showed that the SOD activities were induced by 1.0 and 10.0 mM MeSA and the POD activities were induced by MeSA. The CAT activity was induced at low concentrations of MeSA but was inhibited by high concentrations, and the PPO activity was affected by 0.1, 1.0, and 10.0 mM MeSA. Furthermore, the volatiles were detected using gas chromatography–mass spectrometry and gas chromatography. Twenty-one volatile compounds were tentatively identified in the emissions from plant leaves. Of these volatile compounds, (Z)-3-hexen-1-ol and cis-3-hexenyl acetate emissions were the highest. (Z)-3-Nonen-1-ol, (E)-2-hexen-1-ol, 1-octanol, (E,Z)-3,6-nonadien-1-ol, β-ionone, and hexadecanamide were six compounds that were only produced after treatment by 10.0 mM MeSA. These results showed that direct and indirect defense mechanisms in poplars were induced by MeSA.  相似文献

In this study, a relationship between lipid peroxidation, the antioxidant defense system and salt stress in salt-sensitive cultivated tomato (Lycopersicon esculentum) and its salt-tolerant wild relative (L. pennellii) was established. Superoxide dismutase (SOD) activities were significantly higher in the leaves of L. pennellii than those of L. esculentum after 12 and 84 d. POX activity showed a gradual increase in both cultivars under 70 mM NaCl. POX activity in L. pennellii significantly increased after 6 and 84 d whereas showed no remarkable change in leaves of L. esculentum under 140 mM NaCl. A higher salinity tolerance of L. pennellii was also correlated with a lower lipid peroxidation, which might be due to a higher content of antioxidant enzymes studied.  相似文献

We investigated the possible existence of superoxide dismutase (SOD; EC 1.15.1.1) isoenzymes in the pollen of Nicotiana tabacum (Petit Havana SR-1 cultivar). To detect SOD activity, crude extracts from tobacco pollen were subjected to native polyacrylamide gel electrophoresis followed by staining with nitroblue tetrazolium (NBT). The presence of six SOD isoenzymes was detected in tobacco pollen. Treatment with SOD inhibitors indicated the presence of one manganese SOD (Mn SOD), five copper-zinc SOD (Cu/Zn SOD) isoenzymes, and the absence of iron SOD (Fe SOD).  相似文献

In order to assess the role of the antioxidant defense system against salt treatment, the activities of some antioxidative enzymes and levels of some nonenzymatic antioxidants were estimated in Azolla caroliniana subjected to NaCl treatment (50 mM) for 10 days in absence or presence of nitrate. In A. caroliniana, salt treatment in absence of nitrate preferentially enhanced electrolyte leakage, lipid peroxidation, and H2O2 content. Also, the specific activitiy of guaiacol peroxidase (POX), glutathione reductase (GR), catalase (CAT), ascorbate peroxidase (APX), and superoxide dismutase (SOD) increased. In addition, reduced glutathione level increased and consequently, glutathione/oxidized glutathione (GSH/GSSG) ratio increased. Accumulation of Na+ increased significantly by salinity stress which resulted in a significant decrease in K+ accumulation, accordingly, K+/Na+ ratio decreased. Replacement of potassium chloride by potassium nitrate in nutrient solution under salt stress (50 mM NaCl) exhibited a reduction in electrolyte leakage, lipid peroxidation, and H2O2 contents. Conversely, the specific activity of APX, POX, GR, CAT, and SOD increased. The content of total ascorbate decreased, in contrast, reduced and GSSG increased and the ratio of GSH/GSSG increased 2.3-fold compared to the control value. Sodium ion accumulation was minimized in the presence of nitrate, potassium ion accumulation increased and as a result, K+/Na+ ratio increased when compared with the corresponding salinized plants. The differential changes in the specific activity of antioxidant enzymes due to NaCl treatment and nitrate may be useful as markers for recognizing salt tolerance in A. caroliniana.  相似文献

Seedlings of Camellia sinensis (L.) were grown hydroponically to study the effect of aluminium (Al) on leaf antioxidant defence system and cell ultrastructure. We found that malondialdehyde (MDA) content decreased at 0–0.32 mM Al, but increased significantly at 0.53 mM Al. Like MDA, hydrogen peroxide (H2O2) content increased at 0.53 mM Al; however, no differences were observed at 0–0.32 mM Al. Superoxide dismutase (SOD, EC1.15.1.1) activity remained practically constant at 0–0.32 mM Al, but increased sharply at 0.53 mM Al; catalase (CAT, EC1.11.1.6) and guaiacol peroxidase (GPX, EC1.11.1.7) activities decreased following an initial increase, reaching their peaks at 0.32 mM Al. Ascorbate peroxidase (APX, EC 1.11.1.11) activity increased and glutathione (GR, EC 1.6.4.2) level fluctuated with increasing Al concentrations. Transmission electron microscope analysis of Al-treated leaves showed that although cell ultrastructural integrity was maintained at 0–0.32 mM, significant membrane damage was observed at 0.53 mM. Our results suggest that at low Al concentrations, the leaf antioxidant defence system can scavenge reactive oxygen species and sufficiently protect cells from free radical injury. However, at higher Al concentrations (0.53 mM), the balance between formation and detoxification of ROS is lost, resulting in the destruction of cell ultrastructure.  相似文献

The aqueous, ethyl acetate, methanolic and Total Oligomer Flavonoids (TOF) enriched extracts, obtained from the aerial parts of Cyperus rotundus, were investigated for their contents in phenolic compounds. Antioxidative activity using the NBT/riboflavin assay system, antimicrobial activity against Gram positive and Gram negative bacterial reference strains as well as antigenotoxic activity tested with the SOS chromotest assay were also studied. Significant antibacterial activity against reference strains; Staphylococcus aureus, Enterococcus faecalis, Salmonella enteritidis and Salmonella typhimurium, was detected in the presence of ethyl acetate and TOF enriched extracts. In addition to their antimicrobial activity, the same extracts showed a significant ability to inhibit nitroblue tetrazolium reduction by the superoxide radical in a non enzymatic O2.− generating system, and were also able to reduce significantly the genotoxicity induced by nifuroxazide and Aflatoxin B1. The antioxidant, antimicrobial and antigenotoxic activities exhibited by C. rotundus depend on the chemical composition of the tested extracts.  相似文献

The changes in lipid peroxidation and the involvement of the antioxidant system in relation to salt stress tolerance were investigated in the callus of Acanthophyllum glandulosum and Acanthophyllum sordidum. The callus was subjected to NaCl stress (50–200 mM) for 40 d. The callus of A. glandulosum was less sensitive to NaCl stress than that of A. sordidum. Increasing concentrations of NaCl from 50 to 200 mM correlated to increased proline content in A. glandulosum. Total protein content was higher in extracts of A. glandulosum than in extracts of A. sordidum under both control and salinity treatments. Compared with A. sordidum, lipid peroxidation and H2O2 content were lower and the activities of superoxide dismutase (SOD), catalase (CAT), guaiacol peroxidase (GPX), ascorbate peroxidase, and glutathione reductase were higher in A. glandulosum under salt stress. Activity staining of antioxidant enzymes separated by native polyacrylamide gel electrophoresis (PAGE) revealed that callus of A. sordidum had five Fe-SOD isoforms and one Mn-SOD isoform, all of which were reduced by salinity. In A. glandulosum, two Mn-SOD, three Fe-SOD, and one Cu/Zn-SOD isoforms were detected. Out of these six SOD isoforms, expression of the Mn-SOD and Fe-SOD isoforms was enhanced at 100 mM and higher NaCl concentrations. Two and six GPX isoforms were detected in A. sordidum and A. glandulosum, respectively. Expression of the single CAT isoform in A. sordidum was preferentially reduced by salinity. In A. glandulosum, the two CAT isoforms showed differential down regulation under NaCl stress, with the CAT2 isoform detected only under control condition. These results suggest that A. glandulosum callus is better protected against salinity-induced oxidative damage by maintaining higher activities of antioxidant enzymes than the callus of A. sordidum.  相似文献

Aspartokinase (EC 2.7.2.4), homoserine-dehydrogenase (EC 1.1.1.3) and dihydrodipicolinic-acid-synthase (EC 4.2.1.52) activities were examined in extracts from 1-year-old and 11-year-old cell suspension cultures and whole roots of garden carrot (Daucus carota L.). Aspartokinase activity from suspension cultures was inhibited 85% by 10 mM L-lysine and 15% by 10mM L-threonine. In contrast, aspartokinase activity from whole roots was inhibited 45% by 10 mM lysine and 55% by 10 mM threonine. This difference may be based upon alterations in the ratios of the two forms (lysine-and threonine-sensitive) of aspartokinase, since the activity is consistently inhibited 100% by lysine+threonine. Only one form each of homoserine dehydrogenase and of dihydrodipicolinic acid synthase was found in extracts from cell suspension cultures and whole roots. The regulatory properties of either enzyme were identical from the two sources. In both the direction of homoserine formation and aspartic--semialdehyde formation, homoserine dehydrogenase activities were inhibited by 10mM threonine and 10 mM L-cysteine in the presence of NADH or NADPH. KCl increased homoserine dehydrogenase activity to 185% of control values and increased the inhibitory effect of threonine. Dihydrodipicolinic acid synthase activities from both sources were inhibited over 80% by 0.5 mM lysine. Aspartokinase was less sensitive to inhibition by low concentrations of lysine and threonine than were dihydrodipicolinic acid synthase and homoserine dehydrogenase to inhibition by the respective inhibitors.  相似文献

This study aimed to determine whether patients with aseptic and bacterial meningitis presented alterations in oxidative stress parameters of cerebrospinal fluid (CSF). A total of 30 patients were used in the research. The CSF oxidative stress status has been evaluated through many parameters, such as lipid peroxidation through thiobarbituric acid reactive substances (TBARS) and antioxidant defense systems such as superoxide dismutase (SOD), glutathione S-transferase (GST), reduced glutathione (GSH) and ascorbic acid. TBARS levels, SOD and GST activity increase in aseptic meningitis and in bacterial meningitis. The ascorbic acid concentration increased significantly in patients with both meningitis types. The reduced glutathione levels were reduced in CSF of patients with aseptic and bacterial meningitis. In present study we may conclude that oxidative stress contributes at least in part to the severe neurological dysfunction found in meningitis.  相似文献

Many secondary plant compounds are capable of photoactivation resulting in the production of toxic species of oxygen. One mechanism of defense for insects feeding on phototoxic plants may be the presence of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPOX), and glutathione reductase (GR). The activities of these enzymes were examined in larvae of three lepidoptera: Ostrinia nubilalis, Manduca sexta, and Anaitis plagiata. Highest levels of antioxidant enzyme activity were found in A. plagiata, a specialist feeder on Hypericum perforatum, which contains high levels of the phototoxin hypericin. Larvae of A. plagiata fed leaf discs treated with hypericin exhibited a short-term, concentration-dependent decline in enzyme activity. Longer term studies with A. palgiata fed either the photoxic H. perforatum, or the closely related but non-phototoxic H. calycinum, resulted in increased CAT and GR activity in larvae fed the phototoxic plant whereas SOD activity was not significantly different. These results suggest that CAT and GR may be inducible defenses against phototoxins.  相似文献

Antioxidant enzymes can modify cell response to nitrosative stress induced, for example, by nitric oxide or compounds decomposing with its formation. Therefore, we investigated the effects of S-nitrosoglutathione (GSNO) on cell survival, activity of antioxidant enzymes, and concentrations of reduced and oxidized glutathione in parental and isogenic strains defective in Cu,Zn- or Mn-superoxide dismutases (Cu,Zn-SOD and Mn-SOD, respectively), or in both of them. Stress was induced by incubation of the yeast with 1–20 mM GSNO. The strains used demonstrated different sensitivity to GSNO. A Cu,Zn-SOD-defective strain survived the stress better than the parental strain, while the double mutant was the most sensitive to GSNO. The ·NO-donor at low concentrations (1–5 mM) increased SOD activity, but its high concentrations (10 and 20 mM) decreased it. The activity of catalase in all strains was enhanced by GSNO. Inhibition of protein synthesis by cycloheximide did not prevent the activation of SOD, but it prevented the activation of catalase. These facts suggest that SOD was activated at a posttranslational level and catalase activity was enhanced via de novo synthesis. A GSNO-induced increase in oxidized glutathione level in the studied yeast strains might account for cell killing by GSNO due to the development of oxidative/nitrosative stress. Thermal stability of antioxidant defense enzymes superoxide dismutase (SOD, EC 1.15.1.1) and ascorbate peroxidase (APX, EC 1.11.1.11) was studied in chloroplasts and mitochondria of leaf and inflorescence in heat adaptive weed Chenopodium album. Leaf samples were taken in March (31 °C/14 °C) and young inflorescence (INF) was sampled at flowering in April (40 °C/21 °C). Leaf and INF chloroplast and mitochondrial fractions were subjected to elevated temperatures in vitro (5–100 °C) for 30′. SOD and APX showed activity even after boiling treatment in both chloroplast and mitochondria of leaf and INF. SOD was more heat stable than APX in both chloroplasts and mitochondria in both the tissues. Chloroplast contained more heat stable SOD and APX isozymes than mitochondria in both leaf and INF. To the best of our knowledge this is the first report showing presence of thermostable APX isozymes (100 °C for 30′) in chloroplasts and mitochondria in C. album. Heat stable isozymes of SOD and APX in chloroplasts and mitochondria in leaves and inflorescence may contribute to heat tolerance in C. album.  相似文献

Cadmium stress in sugar cane callus cultures: Effect on antioxidant enzymes   总被引:5,自引:0,他引:5  
Catalase (CAT) and superoxide dismutase (SOD) are antioxidant enzymes which are important in the metabolism of reactive oxygen species (ROS), and can be induced by environmental stresses including cadmium (Cd), a heavy metal toxic to living organisms. Sugar cane (Saccharum officinarumL.) in vitro callus cultures were exposed to CdCl2 and the activities of CAT and SOD were analysed. Lower concentrations of CdCl2, such as 0.01 and 0.1 mM caused a significant increase in callus growth, whereas 0.5 and 1 mM CdCl2 strongly inhibited growth of the callus cultures, but only after 9 days of CdCl2 treatment. Red-brown patches were also observed in calluses exposed to 0.5 and 1 mM CdCl2. Calluses grown in 0.01 and 0.1 mM CdCl2 did not exhibit any changes in CAT activity even after 15 days of growth in the presence of CdCl2. However, for calluses grown in higher concentrations of CdCl2 (0.5 and 1 mM), a rapid increase in CAT activity was detected, which was 14-fold after 15 days. Furthermore, up to five CAT isoforms were observed in callus tissue. Total SOD activity did not exhibit any major variation. One Mn-SOD and two Cu/Zn-SOD isoenzymes were observed in callus cultures and none exhibited any variation in response to the CdCl2 treatments. The results suggested that in sugar cane callus cultures, CAT may be the main antioxidant enzyme metabolizing H2O2.  相似文献

Mountain environmental stresses result in increased formation of hydrogen peroxide (H2O2) and accumulation of malondialdehyde (MDA) in leaves of Polygonum viviparum. The activities of several antioxidative system enzymes such as superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), peroxidase (POD, EC 1.11.1.7), glutathione reductase (GR, EC 1.6.4.2), dehydroascorbate reductase (DHAR, EC 1.8.5.1) and the contents of several non-enzymatic antioxidants such as reduced form of ascorbate (ASC), dehydroascorbate (DHA), reduced glutathione (GSH), and oxidized glutathione (GSSG) were investigated in leaves of P. viviparum, which were collected from three altitudes (2,200, 3,200, and 3,900 m) of Tianshan Mountain in China. The activities of these four antioxidative enzymes were accompanied by increases of H2O2 levels from 2,200 to 3,200 m. However, the activities of CAT and POD were decreased, whereas the activities of SOD and GR continually increased at 3,900 m. Analyses of isoforms of SOD, CAT, POD, and GR showed that the leaves of P. viviparum exposed different altitude conditions are capable of differentially altering the intensity. Additionally, two new isoforms of SOD were detected at 3900 m. A continual increase in the ASC, ASC to DHA ratio, GSH and GSH/[GSH + GSSG] ratio, and the activity of DHAR were observed in leaves of P. viviparum with the elevation of altitude. These results suggest that the higher contents of ASC, GSH as well as an increase in reduced redox state may be essential to antioxidation processes in the leaves of P. viviparum, whereas antioxidant enzymes system is a cofactor in the processes.  相似文献

Plant species capable of hyper-accumulating heavy metals are of considerable interest for phytoremediation, and differ in their ability to accumulate metals from environment. Using two brassica species (Brassica juncea and Brassica napus), nutrient solution experiments were conducted to study variation in tolerance to cadmium (Cd) toxicity based on (1) lipid peroxidation and (2) changes in antioxidative defense system in leaves of both plants (i.e., superoxide dismutase (SOD EC 1.15.1.1), catalase (CAT EC 1.11.1.6), ascorbate peroxidase (APX EC 1.11.1.11), guaiacol peroxidase (GPX EC 1.11.1.7), glutathione reductase (GR EC 1.6.4.2), levels of phytochelatins (PCs), non-protein thiols (NP-SH), and glutathione. Plants were grown in nutrient solution under controlled environmental conditions, and subjected to increasing concentrations of Cd (0, 10, 25 and 50 μM) for 15 days. Results showed marked differences between both species. Brassica napus under Cd stress exhibited increased level of lipid peroxidation, as was evidenced by the increased malondialdehyde (MDA) content in leaves. However, in Brassica juncea treated plants, MDA content remained unchanged. In Brassica napus, with the exception of GPX, activity levels of some antioxidant enzymes involved in detoxification of reactive oxygen species (ROS), including SOD, CAT, GR, and APX, decreased drastically at high Cd concentrations. By contrast, in leaves of Brassica juncea treated plants, there was either only slight or no change in the activities of the antioxidative enzymes. Analysis of the profile of anionic isoenzymes of GPX revealed qualitative changes occurring during Cd exposure for both species. Moreover, levels of NP-SH and PCs, monitored as metal detoxifying responses, were much increased in leaves of Brassica juncea by increasing Cd supply, but did not change in Brassica napus. These results indicate that Brassica juncea plants possess the greater potential for Cd accumulation and tolerance than Brassica napus.  相似文献

Inhibition of root growth and modification of root morphology are the most sensitive responses of Lupinus luteus cv. Ventus L. to lead ions - Pb(NO3)2. Using electron paramagnetic resonance (EPR), we found that at the lead concentration of 150 mg.L–1, the level of free radicals remained at control level, whereas at the higher, sublethal concentration of 350 mg.L–1, they markedly increased. The EPR signal with the g-value at the maximum absorption of 2.0053 implied that the paramagnetic radical is derived from a quinone. The response of antioxidant enzymes, such as superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), peroxidase (POX, EC 1.11.1.7) and ascorbate peroxidase (APOX, EC 1.11.1.11), to exogenously applied lead ions was also examined. Enzyme activity was estimated as a function of time and concentration. Native polyacrylamide gel electrophoresis followed by specific staining revealed an increase in the activity of SOD, CAT, POX and APOX coinciding with the time of cultivation. A lead-dependent increase in activities of SOD and POX from root tip extracts was observed, whereas CAT and APOX activities decreased at the higher lead concentrations. These results suggest that at higher lead concentrations, the formation of both free radicals and reactive oxygen species is beyond the capacity of the antioxidant system, which in turn may contribute to the reduced root growth.  相似文献

The effects of varying concentrations of cadmium (Cd) on the development of Lycopersicon esculentum cv. Micro‐Tom (MT) plants were investigated after 40 days (vegetative growth) and 95 days (fruit production), corresponding to 20 days and 75 days of exposure to CdCl2, respectively. Inhibition of growth was clearly observed in the leaves after 20 days and was greater after 75 days of growth in 1 mM CdCl2, whereas the fruits exhibited reduced growth following the exposure to a concentration as low as 0.1 mM CdCl2. Cd was shown to accumulate in the roots after 75 days of growth but was mainly translocated to the upper parts of the plants accumulating to high concentrations in the fruits. Lipid peroxidation was more pronounced in the roots even at 0.05 mM CdCl2 after 75 days, whereas in leaves, there was a major increase after 20 days of exposure to 1 mM CdCl2, but the fruit only exhibited a slight significant increase in lipid peroxidation in plants subjected to 1 mM CdCl2 when compared with the control. Oxidative stress was also investigated by the analysis of four key antioxidant enzymes, which exhibited changes in response to the increasing concentrations of Cd tested. Catalase (EC 1.11.1.6) activity was shown to increase after 75 days of Cd treatment, but the major increases were observed at 0.1 and 0.2 mM CdCl2, whereas guaiacol peroxidase (EC 1.11.1.7) did not vary significantly from the control in leaves and roots apart from specific changes at 0.5 and 1 mM CdCl2. The other two enzymes tested, glutathione reductase (EC 1.6.4.2) and superoxide dismutase (SOD, EC 1.15.1.1), did not exhibit any significant changes in activity, apart from a slight decrease in SOD activity at concentrations above 0.2 mM CdCl2. However, the most striking results were obtained when an extra treatment was used in which a set of plants was subjected to a stepwise increase in CdCl2 from 0.05 to 1 mM, leading to tolerance of the Cd applied even at the final highest concentration of 1 mM. This apparent adaptation to the toxic effect of Cd was confirmed by biomass values being similar to the control, indicating a tolerance to Cd acquired by the MT plants.  相似文献

Marine planktonic algae are frequently exposed to metallic contaminants. Because heavy metals can be assimilated and accumulated by algal cells, they can then be transferred to higher trophic levels of food chains. We studied the effects of cadmium on protein production and the growth of the marine prasinophyte Tetraselmis gracilis (Kylin) Butcher. By means of toxicological assays, we estimated the LC50 of cadmium as 3.2 ppm and 1.8 ppm after 48 h and 96 h of exposure to this heavy metal, respectively. The growth curves and survival percentages of cell cultures in the presence of cadmium were determined, and a proportional reduction of both parameters with increasing metal concentrations was found. When chronically exposed to sublethal concentrations of cadmium, T. gracilis contained high levels of superoxide dismutase (SOD) activity, one of the main enzymes of the cell's antioxidant defense mechanism. Under these growth conditions, total SOD activity in crude extracts was increased by 41% (at 1.5 ppm) and 107% (at 3.0 ppm). Assays of SOD activity in nondenaturing polyacrylamide gels also showed a similar induction by cadmium. These results show that cadmium has potentially toxic properties since it significantly inhibited the growth of T. gracilis at low concentrations and promoted the induction of SOD activity, suggestive of an oxidative stress state. Besides being the first report of SOD in T. gracilis, this work describes experimental evidence of SOD induction by cadmium in this species.  相似文献

Salinity could damage cellular membranes through overproduction of reactive oxygen species (ROS), while antioxidant capacities play a vital role in protecting plants from salinity caused oxidative damages. The objective of this study was to investigate the toxic effect of salt on the antioxidant enzyme activities, isoforms and gene expressions in perennial ryegrass (Lolium perenne L.). Salt-tolerant ‘Quickstart II’ and salt-sensitive ‘DP1′ were subjected to 0 and 250 mM NaCl for 12 d. Salt stress increased the content of lipid peroxidation (MDA), electrolyte leakage (EL) and hydrogen peroxide (H2O2), to a greater extent in salt-sensitive genotype. Salt-stressed plant leaves exhibited a greater activity of superoxide dismutase (SOD, EC 1.15.1.1), peroxidase (POD, EC 1.11.1.7), ascorbate peroxidase (APX, EC 1.11.1.11) at 4 d after treatment (DAT), but a lower level of enzyme activity at 8 and 12 d, when compared to the control. Catalase (CAT, EC 1.11.1.6) activity was greater at 4 DAT and thereafter decreased in salt tolerant genotype relative to the control, whereas lower than the control during whole experiment period for salt-sensitive genotype. There were different patterns of five isoforms of SOD, POD and two isoforms of APX between two genotypes. Antioxidant gene expression was positively related to isoenzymatic and total enzymatic activities during 12-d salt-treated leaves of two genotypes, with a relatively higher level in salt-tolerant genotype. Thus, salt tolerance could be related to the constitutive/induced antioxidant gene, leading to more efficient enzyme stimulation and protection in perennial ryegrass.  相似文献
