Atriplex spongiosa was grown in hydroponic culture with additionsof 0, 200, 400 and 600 mM NaCl. Frozen, hydrated cells and cellcompartments of young and mature leaf tissue were analysed byX-ray micro-analysis. Evidence for low K + Na and Cl content,and high K selectivity in the bundle sheath cytoplasm was obtainedfrom data on X-ray count ratios and on total X-ray counts. Vacuolesof the major cell types of the mature leaf had either high Kor high Na and Cl contents when grown in the absence or presenceof NaCl. Comparison of K, Na and Cl content of different cell types inthe mature leaf showed gradients in selectivity for K. relativeto Na between the bundle sheath cells and the bladder cells.In the young expanding leaves salt was sequestered in the numeroussalt bladders on the leaf surface, while the cytoplasm and developingvacuoles of undifferentiated cells contained largely K and littleNa or Cl. The results support general views on the compartmentation ofsalt in plants cells in relation to osmotic or saline stress. Bean plants (Phaseolus vulgarisL. ‘Zlota Saxa’)were cultured on complete (+P) or phosphate-deficient (-P) nutrientmedium. A large increase in glucose concentration was foundin the meristematic zone of -P roots compared to control roots.The increased glucose concentration in the meristematic zonedid not influence total respiration rate. Glucose or uncoupler(carbonyl cyanide m-chlorophenylhydrazone) failed to increasethe respiration rate in -P root segments, but stimulated respirationin +P roots. The ultrastructure of cortical cells from the meristematicroot zone showed marked differences between +P and -P plants.Large vacuoles, invaginations of the plasmalemma and condensedforms of mitochondria were dominating features in cortical cellsof -P roots. Analysis of extracts after treating roots withdimethylsulfoxide (DMSO) indicated different localization ofsugars in the cell compartments. In roots of -P plants, mostof the reducing sugars were detected in the cytoplasm fractionwhile most sucrose was in the vacuole. Observations of the effectof 10% DMSO on cell ultrastructure indicated partial destructionof the plasmalemma but not the tonoplast. The localization ofreducing sugars in secondary vacuoles or plasmalemma invaginationsin the cells from the meristematic region of -P roots is discussed.Copyright1998 Annals of Botany Company. The ion content of compartments within cortical cells of mature roots of the halophyte Suaeda maritima grown at 200 mol·m-3 NaCl has been studied by X-ray microanalysis of freeze-substituted thin sections. Sodium and Cl were found in the vacuoles at about four-times the concentration in the cytoplasm or cell walls, whereas K was more concentrated in the cell walls and cytoplasm than in vacuoles. The vacuolar Na concentration was 12- to 13-times higher than that of K. The Na concentration of cell walls of cortical cells was about 95 mol·m-3 of analysed volume. The cytoplasmic K concentration within the mature cortical cells was estimated to be 55 mol·m-3 of analysed volume.  相似文献

Driss-Ecole, D. and Perbal, G. 1987. Intracellular localizationof 3H-IAA in the apical bud of Lycopersicon esculentum.—J.exp. Bot. 38: 1362–1372. High resolution autoradiography of 3H-IAA was performed on ultra-thinsections of the apical bud of Lycopersicon esculentum treatedby DCC or glutaraldehyde. A quantitative study of the localizationof the labelling in the compartments (cell wall, cytoplasm,vacuoles and nucleus) of four types of cells (cells of the lateralzone of the apex, cells of the pith meristem, proximal and distalcells of the upper part of the pith) was made. The statisticalanalysis of the results has proved that the variability of thelabelling of the cell compartments in the four cell types wassimilar after treatment by DCC or glutaraldehyde. The densityof labelling is higher in each compartment of the meristematiccells compared with those of the differentiating cells. Thecells of the pith meristem can be distinguished from the meristematiccells of the lateral zone of the apex by a greater density oflabelling in the cytoplasm and the nucleus. These two cellulartypes contain a high density of radioactivity in vacuoles. Byconsidering the percentage of labelling of each compartmentrelative to the total labelling in the cell, it can be shownthat the meristematic cells are characterized by a high percentagein the nucleus whereas the vacuoles of the differentiating cellscontain the highest percentage of radioactivity. Using excised low-salt roots of barley and Atriplex hortenslsthe transport of endogenous potassium through the xylem vesselswas studied It was enhanced by nitrate and additionally by sodiumions which apparently replaced vacuolar potassium which wasthen available in the symplasm of root cells for transport tothe shoot Vacuolar Na/K exchange also has been investigatedby measurements of longitudinal ion profiles in single rootsof both species. In Atriplex roots a change in the externalsolution from K+ to Na+ induced an exchange of vacuolar K+ forNa+, in particular in the subapical root tissues and led toincreased K+ transport and loss of K+ from the cortex. In inverseexperiments a change from Na+ to K+ did not induce an exchangeof vacuolar Na+; merely in meristematic tissues Na+—apparentlyfrom the cytoplasm—was extruded in exchange for K+. Inroots of barley seedlings without caryopsis, as in excised roots,a massive exchange of K+ for Na+ was observed in the continuouspresence of external 1.0 mM Na and 0.2 mM K. This exchange alsowas attributed to the vacuole and was most pronounced in theyoung subapical tissues. It did not occur, however, in the correspondingtissues in roots of fully intact barley seedlings. In these,the young tissues retained a relatively high K/Na ratio alsoin their vacuoles. Similarly, contrasting results were obtainedwith intact and excised roots of Zea mays L. Based on theseresults a scheme of the events that lead to selective cationuptake in intact barley roots is proposed. In this scheme acrucial factor of selectivity is sufficient phloem recirculationof K+ by the aid of which K+ rich cortical cells are formednear the root tip. When matured these cells are suggested tomaintain a high cytoplasmic K/Na ratio due to K+ dependent sodiumextrusion at the plasmalemma and due to recovery of vacuolarK+ by Na/K exchange across the tonoplast. Hajibagheri, M. A., Gilmour, D. J., Collins, J. C. and Flowers,T. J. 1986. X-ray microanalysis and ultrastructural studiesof cell compartments of Dunaliella parva. -J. exp. Bot. 37:1725–1732. Ultrastructural studies of the unicellular green alga Dunaliellaparva showed the presence of cytoplasmic vacuoles. X-ray microanalysiswas performed on sections of cells which had been freeze substitutedin acetone. It was found that the concentrations of both Naand Cl were much higher in the vacuoles than in the cytoplasm.When cells were grown in 0·4 kmol m–3 NaCl theNa and Cl concentrations in the vacuoles were 349 and 283 molm –3 respectively, whilst cytoplasmic Na and Cl concentrationswere 37 and 26 mol m–3. Corresponding values for cellsgrown in 1·5 kmol m–3 NaCl were 392 mol m–3Na and 325 mol m–3 Cl in the vacuoles and 36 mol m–3Na and 30 mol m–3 Cl in the cytoplasm. Immediately afterexposure to an increase in external salinity Na and Q concentrationsincreased in both vacuoles and cytoplasm. The distribution of ions, particularly K and Na, was studied in roots of barley seedlings grown on various ionic solutions. Analyses were made by means of electron probe x-ray microanalysis using frozen, fractured bulk specimens. By this technique, it was demonstrated that there can be variability in the ratio K/Na measured in the vacuoles of cortical cells, with this ratio often being lower in epidermal cells of the root than in the inner cortex. A sharp difference in the K/Na ratio was also found between cells of the endodermis and those of the adjacent cortex, and generally higher ratios of K/Na occurred in the stele than in the cortex. Estimation of the concentrations in the cytoplasm was at the limit of resolution of this technique, but it can be shown that the K/Na ratio in the cytoplasm was higher than that in the vacuole. In low salt roots, the K concentration in the cytoplasm was higher than that in the vacuoles. The results with the x-ray microprobe confirm other measurements based on flux analysis or analysis of small samples of the root.  相似文献

A study was made of ultrastructural aspects and ion distributionin roots of Phaseolus coccineus as affected by NaCl and Na2SO4salinity. In the proximal region of the root, xylem parenchymacells are differentiated as transfer cells with well developedwall protuberances adjacent to the half-bordered pits of thevessels. The cytoplasm of these transfer cells contains cisternaeof rough endoplasmic reticulum, the number of which was increasedgreatly when the plants were grown in the presence of NaCl orNa2SO4. The cisternae of the endoplasmic reticulum are oftenassociated closely with the plasmalemma and interconnected withit by fibrillar bridges. Wall protuberances occur also in the exodermis and epidermisof the more apical region of the root. Their function is stillunknown. P. coccineus excludes Na, but not Cl, from the leaves by retainingit particularly in the proximal region of the root. X-ray microanalysisof unfixed, frozen, hydrated specimens revealed that the transfercell-type xylem parenchyma cells in salt-treated roots accumulatedNa relative to both the adjoining xylem vessels and the corticalcells and showed very high Na/K and Na/Cl ratios. It is suggestedthat the xylem parenchyma cells can reabsorb Na from the vessels,probably in exchange for K, and that Na exclusion from the shootis at least partly mediated by this process. The implicationof this for regulation of salt transport in salt sensitive glycophytesis discussed.  相似文献

We used quantitative electron-probe energy-dispersive x-raymicroanalysis to localize endogenous Na, Cl, K, P, S, Mg andCa in cryofixed and freeze-dried cryosections of the cap (i.e.the putative site of graviperception) and elongating zone (i.e.site of gravicurvature) of horizontally oriented roots of Zeamays. Ca, Na, Cl, K and Mg accumulate along the lower side ofcaps of horizontally oriented roots. The most dramatic asymmetriesof these ions occur in the apoplast, especially the mucilage.We could not detect any significant differences in the concentrationsof these ions in the central cytoplasm of columella cells alongthe upper and lower sides of caps of horizontally-oriented roots.However, the increased amounts of Na, Cl, K and Mg in the longitudinalwalls of columella cells along the lower side of the cap suggestthat these ions may move down through the columella tissue ofhorizontally-oriented roots. Ca also accumulates (largely inthe mucilage) along the lower side of the elongating zone ofhorizontally-oriented roots, while Na, P, Cl and K tend to accumulatealong the upper side of the elongating zone. Of these ions,only K increases in concentration in the cytoplasm and longitudinalwalls of cortical cells in the upper vs lower sides of the elongatingzone. These results indicate that (1) gravity-induced asymmetriesof ions differ significantly in the cap and elongating zoneof graviresponding roots, (2) Ca accumulates along the lowerside of the cap and elongating zone of graviresponding roots,(3) increased growth of the upper side of the elongating zoneof horizontally-oriented roots correlates positively with increasedamounts of K in the cytoplasm and longitudinal walls of corticalcells, and (4) the apoplast (especially the mucilage) may bean important component of the pathway via which ions move ingraviresponding rots of Zea mays. Summary The elemental composition of the morula cell of Halocynthia papillosa blood was studied by X-ray microanalysis with respect to the possible iron accumulation in this cell type. We found various amounts of Na, Mg, P, S, Cl, K, Ca, Fe and Br in the cytoplasm, nucleus and vacuoles. With the exception of a few cells, Ca, Fe and Br were not detected. Thus, the morula cells of the studied species are not iron-rich cells.  相似文献

Tannin coenocytes develop from mononucleate tannin mother cells.The process occurs within the whole of the first (youngest)internode and its development can be divided into three stages.In stage I the MTC is isodiametric and similar to the surroundingcells of the flank meristem, being present in the ninth celllayer from the apex surface. The nucleus becomes lanceolateand elongates, and large cytoplasmic vacuoles appear. A twofoldelongation of both the cell and nucleus continues in the secondstage, the cell-nucleus ratio indicating that it is due to theenlarged vacuole, which pushes a thin layer of cytoplasm closeto the cell wall. In this layer of cytoplasm dilated ER cisternaoccur together with small and large vacuoles, a fusion of thevacuoles increasing their volume. The method of the modified compartmental analysis for excisedroots has been adopted for measuring K+-fluxes and compartmentationin barley (Hordeum distichon) roots. Efflux of 42K and 86Rbindicated that more than two intracellular compartments wereinvolved in the tracer exchange; the 42K data clearly showedthe components. On the basis of the efflux behaviour of theapical and more basal tissues of the roots, the three componentsof efflux were attributed to the cytoplasm of differentiated(fast) and meristematic tissues (intermediate) and to the vacuoles(slow exchange) of the roots. A model is proposed on the basisof which, the fluxes corresponding to the meristematic and differentiatedtissues of the root can be estimated. Additionally, fluxes ofthe differentiated root tissues were determined by using effluxdata obtained with root segments without apical tissues. Thedata obtained in both ways compare reasonably well and agreeto independent chemical measurements. Comparison of the 42K and 86Rb efflux data show strong discriminationof K in favour of Rb+ and indicate that 86Rb is not suitableas a tracer for K+ in efflux measurements, at least with barleyroots.  相似文献

The ontogeny of the vegetative shoot apices of two importantvegetable plants, Solanum melongena L. and Capsicum annuum L.is described. Each shoot apex is studied at different stagesof seed germination. The relationship between time of germinationand (i) area of vacuoles in the cell, (ii) total area of thecell, (iii) area of the nucleus in the cell, and (iv) ratioof area of vacuoles in the cell to the total area of the cellin each apex is examined. The differentiation of cytohistologicalzonation in both apices is distinct only after one or two leavesare initiated and developed. At a certain stage heterogeneityin staining in the peripheral region of the shoot apex of S.melongena is found. The zonation in both plants differentiatesgradually in histological and cytological features. Vacuolationincreases or decreases in all the cells of the shoot apex orin the cells of a particular region of the shoot apex at differentstages of its ontogeny.  相似文献

The effects of changing the external concentrations of Na, K,Ca, and Cl on the potentials of the cytoplasm and the vacuolewith respect to the bathing medium of the internodal cells ofNitella translucens have been investigated. The potential differencebetween the vacuole and the cytoplasm is practically unaffectedby the concentration changes. The observed changes of potentialdifference are therefore attributed to the boundary separatingthe cytoplasm from the medium; this boundary is possibly a plasmalemma–cellwall complex. The difference of potential between the cell walland the medium has also been measured and, in the presence ofCa, shown to be markedly sensitive only to the external Ca concentration.The results are divided into two sections: (a) for cells pretreatedin 5 mM NaCl, the subsequent experiments being carried out inCa-free media, and (b) for cells initially immersed in a standardartificial pond water containing the chlorides of Na, K, Ca.With the pretreated cells the external Na/K ratio was variedwith the total NaCl+KCl concentration kept constant at 1.1 mM.The results suggest that over a limited range of concentrationsthe cytoplasm-medium potential difference can be described byan equation similar in form to a Goldman equation but containingonly terms for Na and K, the average value of the permeabilityratio (= PNa/PK) being 0.27. In the presence of Ca the effectsof Na and K on the cytoplasm-medium potential difference aregreatly reduced, while the effect of Ca is relatively large.The results cannot be fitted to any form of Goldman equationcontaining terms for the major ions. The possibility of a contributionto the plasmalemma potential from electrogenic pumps is brieflydiscussed. Measurements of the Na and K content of the cytoplasmand the vacuole have been made for the pretreated cells. TheNa concentration in the cytoplasm is 37 mM and in the vacuole73 mM; the K concentration is 93 mM in the cytoplasm and 67mM in the vacuole. The Nernst potentials for both ions are comparedwith the cytoplasm-medium and cytoplasm-vacuole potential differences.This analysis shows that Na is actively transported from thecytoplasm into the medium as well as into the Vacuole; K ispumped into the cytoplasm from the medium but appears to beclose to electrochemical equilibrium across the tonoplast. Early cellular events during secondary embryogenesis were studiedin a cork oak recurrent embryogenic system in which embryosarise either in a multicellular budding pathway from a compactmass of proliferation or from isolated single cells in friablecallus. The compact mass of proliferation originated from theepidermal cells at the hypocotyl whose growth and convolutionwas characterized by a decrease in the nucleus/cytoplasm ratioand a marked increase in storage products. The transition fromthe compact mass to meristematic primordia occurred at the peripheryand was accompanied by cell dedifferentiation and a drasticreduction of storage products. Meristematic primordia evolvedto globular embryos by the organization of a protodermis andtwo internal centres. Microscope analysis of friable callusshowed an hypothetical sequence from single cells to aggregatesof a few cells, meristematic cell clusters and globular embryos.Single cells showed typical features of embryogenic cells suchas rich cytoplasm and a large number of starch grains and lipidbodies. A progressive cell dedifferentiation and a drastic reductionof storage products was observed when aggregates of a few cellsand meristematic cell clusters were compared. Progressive bipolarizationin large meristematic cell clusters initiated globular embryoformation. The comparison of both embryogenic pathways at theultrastructural level showed that subcellular changes followa similar sequential pattern, especially with regard to thestorage products. The possible role of plastid extrusions andmultivesicular bodies in the changing pattern of starch metabolismduring embryogenesis is discussed. The growth of garden orache, A triplex hortensis was studiedunder conditions of mild NaCl or Na2SO4 salinity. Growth, drymatter production and leaf size were substantially stimulatedat 10 mM and 50 mM Na+ salts. Increased growth, however, appearedto be due to a K+-sparing effect of Na+ rather than to salinityper se. The distribution of K+ and Na+ in the plant revealeda remarkable preference for K+ in the roots and the hypocotyl.In the shoot the K/Na ratio decreased strongly with leaf age.However, the inverse changes in K+ and Na+ content with leafage were dependent on the presence of bladder hairs, which removedalmost all of the Na+ from the young leaf lamina. Measurementsof net fluxes of K+ and Na+ into roots and shoots of growingAtriplex plants showed a higher K/Na selectivity of the netion flux to the root compared to the shoot. With increasingsalinity the selectivity ratio SK, Na* of net ion fluxes tothe roots and to the shoots was increased. The data suggestthat recirculation of K+ from leaves to roots is an importantlink in establishing the K/Na selectivity in A. hortensis plants.The importance of K+ recirculation and phloem transport forsalt tolerance is discussed. Root explants of Brassica napus cultured in vitro form adventitiousshoots. The root buds originated at the base of the newly initiatedlateral root. Cells in association with the differentiatingphloem of the developing lateral roots were the sites for rootbud formation. A nodular mass of cytoplasmic cells developedby day 7 at the base of the lateral root. This group of cellscontinued to divide an enlarge. The cells in the peripheralregion of the nodular cell mass differentiated further intoa meristematic zone. The meristematic cells grew towards theperiphery of the cortex by crushing the outer layer of corticalcells. The changes that took place in mitotic index (MI), labellingindex (LI) and the relative proportions of interphase nucleiwith different amounts of DNA have been investigated duringthe regeneration of meristematic activity at the apex of rootsof Vicia faba over the 144 h period following removal of thecap and apical mm of the meristem. Measurements were also madeof the corresponding changes that took place as cells were displacedbasally along the root from the apex over the experimental period.In both parts of the root, MI and the relative proportions ofnuclei with different DNA contents changed from levels similarto those at the apex of the controls at the start and end ofthe experiment to levels resembling those found in more matureparts of the root at 24 and 48 h. In contrast to these results,LI declined over the experimental period. These cytologicalchanges were aresult of the development of lateral root primordiain both the apical 2 mm of the decapitated roots and as cellswere displaced out of the meristem into more basal parts ofthe root. It was concluded that the events leading to the regenerationof meristematic activity at the apex of roots from which thecap and apical mm of the meristem were removed, are no differentfrom those which result in lateral formation as cells are displacedbasally along the primary root from the apex, and they takeplace over the same time interval in both systems.  相似文献

Procambium is commonly recognized as a vascular meristem inshoot apices of vascular plants. Prestelar tissue comprisingprovascular tissue (PVT) and pith mother cells (PMCs) immediatelysubjacent to the single cell layer of promeristem has been consideredto represent the initial stage of stelar differentiation precedingprocambium and rib meristem in ferns. In addition to characterizationof PVT and PMCs on the basis of cell morphology, cytologicalfeatures and developmental continuity with procambium and ribmeristem, four lines of evidence from studies of shoot apicesof Matteuccia struthiopteris and Osmunda cinnamomea supportthis interpretation of initial differentiation. (1) Differentialstaining by safranin-fast green and crystal violet-erythrosinshows that PVT and PMCs differ in colour reactions from promeristemand resemble procambium and pith meristem, respectively. (2)Comparative ultrastructural study reveals qualitative differencesin the cell membrane system, nuclei, cytoplasm, vacuoles andplastids between promeristem and PVT but similarity of PVT toprocambium. (3) Large droplets of tannins occur in promeristembut not in PVT, PMCs and procambium. (4) Cytochemical studyof the shoot apex of Osmunda shows that carboxylesterase activityis strongly demonstrated in PVT and procambial cells but notin promeristem cells and PMCs. Ultrastructural effects of a basic protein, lysozyme, on rootcells of Iasione montana have been related to cellular mechanismsof root growth inhibition. Lysozyme is found to disrupt cellwalls and to disintegrate cellular membranes of elongating cells,but not membranes of mature and meristematic cells. Lysozymeseems to penetrate roots only in the region of elongation anddoes not affect meristematic cells; meristematic cells of Iasionemontana possess very thick peripheral cell walls covered byan electronopaque layer. As the cells elongate, primary cellwalls become thinner, the microfibrillar network becomes looserand the dense layer breaks off.
