Phenylalanine is the precursor of the cinnamic acids and coumarins in the stems and leafs of P. graeca L. Esterification of p-coumaric acid by quinic acid is required before oxidation to chlorogenic acid. In our experiments, we did not obtain radioactive flavonols from 14C phenylalanine. PAL activity varies as a result of light and temperature in the same manner as the level of flavonoids (especially the phenolic acids). This enzyme, therefore, plays a regulatory role in the synthesis of these phenolic substances. The variation in PAL activity during illumination does not follow the same course as described for other plants.  相似文献

Measurement of protein synthesis in rat lungs perfused in situ   总被引:6,自引:6,他引:0  
Compartmentalization of amino acid was investigated to define conditions required for accurate measurements of rates of protein synthesis in rat lungs perfused in situ. Lungs were perfused with Krebs–Henseleit bicarbonate buffer containing 4.5% (w/v) bovine serum albumin, 5.6mm-glucose, normal plasma concentrations of 19 amino acids, and 8.6–690μm-[U-14C]phenylalanine. The perfusate was equilibrated with the same humidified gas mixture used to ventilate the lungs [O2/CO2 (19:1) or O2/N2/CO2 (4:15:1)]. [U-14C]Phenylalanine was shown to be a suitable precursor for studies of protein synthesis in perfused lungs: it entered the tissue rapidly (t½, 81s) and was not converted to other compounds. As perfusate phenylalanine was decreased below 5 times the normal plasma concentration, the specific radioactivity of the pool of phenylalanine serving as precursor for protein synthesis, and thus [14C]phenylalanine incorporation into protein, declined. In contrast, incorporation of [14C]histidine into lung protein was unaffected. At low perfusate phenylalanine concentrations, rates of protein synthesis that were based on the specific radioactivity of phenylalanyl-tRNA were between rates calculated from the specific radioactivity of phenylalanine in the extracellular or intracellular pools. Rates based on the specific radioactivities of these three pools of phenylalanine were the same when extracellular phenylalanine was increased. These observations suggested that: (1) phenylalanine was compartmentalized in lung tissue; (2) neither the extracellular nor the total intracellular pool of phenylalanine served as the sole source of precursor for protein; (3) at low extracellular phenylalanine concentrations, rates of protein synthesis were in error if calculated from the specific radioactivity of the free amino acid; (4) at high extracellular phenylalanine concentrations, the effects of compartmentalization were negligible and protein synthesis could be calculated accurately from the specific radioactivity of the free or tRNA-bound phenylalanine pool.  相似文献

Specific incorporation of l-[U-14C]phenylalanine, [U-14C]cinnamic acid and p[2-14C]coumaric acid into bakuchiol has been observed in Psoralea corylifolia. Our findings show that the aromatic moiety along with two carbon atoms of the side chain are biosynthetically derived via phenylpropane pathway and not by the alternate pathway proposed earlier.  相似文献

When grown on glucose as principal carbon source the culture medium of Polyporus hispidus was found to contain phenolic acids, including p-coumaric and caffeic acids. 14C-Studies indicated that phenylalanine is converted to cinnamic acid as well as to phenylpyruvic acid and tyrosine in cultures. Cell-free preparations of mycelium contained phenylalanine and tyrosine ammonia-lyse activities and were capable of effecting the hydroxylation of cinnamic, p-coumaric and benzoic acids.  相似文献

An investigation was conducted to study the interrelation of free amino acid metabolism and root formation in etiolated pea stem sections as dependent on time and on inhibition of root formation by kinetin and ethionine. The rise in the level of aspartic acid and increase in the rate of conversion of14C-labeled glucose to free amino acids were found to be characteristic features of the formation of foci of meristematic cells in pericyclo region. The formation of roots was reflected, in general, much more in the rate of conversion of labeled glucose to free amino acids than in the levels of corresponding amino acids. The total amount of free amino acids was not significantly changed during incubation of stem sections in a solution of kinetin (5×10?5 m). A rapid fall in their level was recorded in the next 24 hours. The incorporation of14C from glucose into a precursor of lignin, phenylalanine, was completely inhibited by kinetin which stimulated simultanously the growth of adjacent buds. Stimulation of secondary xylem formation, which appeared later, was accompanied by the resumption of14C-incorporation into phenylalanine. Inhibition of root formation by ethionine resulted in the rapid fall of the level of most amino acids and in a significant decrease in the rate of incorporation of14C from glucose into amino acids. A decreasing level of ethionine in tissues during cultivation of ethionine-treated stem sections was accompanied by a gradual rise in the individual amino acids and in the rate of conversion of glucose into free amino acids.  相似文献

Datura innoxia plants were fed via the roots with cinnamic acid-[214C], (±)-phenyllactic (2-hydroxy-3-phenylpropanoic) acid-[214C] and phenylalanine-[2-14C]. In each case apohyoscine, hyoscine, hyoscyamine and littorine were isolated from the aerial parts, and hyoscine, hyoscyamine and littorine from the roots. Cinnamic acid was not incorporated into the acid moieties of the alkaloids. Phenyllactic acid served as a better precursor than phenylalanine for tropic acid (hyoscine and hyoscyamine) and atropic acid (apohyoscine). Phenylalanine served as an effective precursor for the phenyllactic acid moiety of Littorine.  相似文献

An enzymatic method using phenylalanine ammonia-lyase (l-phenylalanine ammonia-lyase, EC 4.3.1.5) for the rapid conversion of l-[U-14C]phenylalanine to the deaminated lignin precursor trans-[U-14C]cinnamic acid is described.   • 2 The GABA effect required the presence of Na+ and occurred at GABA concentrations which are thought to be physiological (1–5 mM).
  • 3 Of all other amino acids tested at tissue extract concentrations in the system, only glycine had a similar effect. No analogues of GABA tested had a significant stimulatory effect upon leucine incorporation into protein, with the exception of homocarnosine which was mildly stimulatory.
  • 4 The effect of GABA upon the incorporation of L-[U-14C]leucine was examined in the presence of added amino acid substrates, both individually and as mixtures. Also, the incorporation of L-[U-14C]leucine was compared with incorporation of L-[U-14C]Iysine and L-[U-14C]phenylalanine. The results are discussed in terms of GABA interaction with activating, transfer and transport mechanisms of other amino acids, inhibition of proteinase activity, and the possibility that GABA is stimulating the synthesis or turnover of specific proteins in the brain ribosomal system.

Specifically radiolabeled [14C-lignin]lignocelluloses were prepared from the aquatic macrophytes Spartina alterniflora, Juncus roemerianus, Rhizophora mangle, and Carex walteriana by using [14C]phenylalanine, [14C]tyrosine, and [14C]cinnamic acid as precursors. Specifically radiolabeled [14C-polysaccharide]lignocelluloses were prepared by using [14C]glucose as precursor. The rates of microbial degradation varied among [14C-lignin]lignocelluloses labeled with different lignin precursors within the same plant species. To determine the causes of these differential rates, [14C-lignin]lignocelluloses were thoroughly characterized for the distribution of radioactivity in nonlignin contaminants and within the lignin macromolecule. In herbaceous plants, significant amounts (8 to 24%) of radioactivity from [14C]phenylalanine and [14C]tyrosine were found associated with protein, although very little (3%) radioactivity from [14C]cinnamic acid was associated with protein. Microbial degradation of radiolabeled protein resulted in overestimation of lignin degradation rates in lignocelluloses derived from herbaceous aquatic plants. Other differences in degradation rates among [14C-lignin]lignocelluloses from the same plant species were attributable to differences in the amount of label being associated with ester-linked subunits of peripheral lignin. After acid hydrolysis of [14C-polysaccharide]lignocelluloses, radioactivity was detected in several sugars, although most of the radioactivity was distributed between glucose and xylose. After 576 h of incubation with salt marsh sediments, 38% of the polysaccharide component and between 6 and 16% of the lignin component (depending on the precursor) of J. roemerianus lignocellulose was mineralized to 14CO2; during the same incubation period, 30% of the polysaccharide component and between 12 and 18% of the lignin component of S. alterniflora lignocellulose was mineralized.  相似文献

DL-Phenylalanine-[3-14C] and cinnamic acid-[3-14C] were fed to this plant and the label from cinnamic acid was incorporated into gallic acid, phyllodulcin and quercetin. By feeding p- coumaric acid-[U-3H], caffeic acid-[U-3H] and hydrangea glucoside A-[U-3H], it was possible to show that hydroxylation at C-3′in phyllodulcin occurs after the ring closure of dihydroisocoumarin. The biosynthetic pathway of phyllodulcin in this plant is thus: phenylalanine → cinnamic acid → p- coumaric acid → hydrangenol → phyllodulcin.  相似文献

The biosynthetic pathway for gallic and ellagic acids in young, mature and autumn leaves ofAcer buergerianum andRhus succedanea was examined by tracer experiments, and also by isotope competition, withd-shikimic acid-14C,l-phenylalanine-U-14C,l-phenyllactic acid-U-14C, gallic acid-G-14C and their unlabeled compounds. In young leaves of both plants, the incorporation rate of labeled shikimic acid into gallic acid was significantly higher than that of labeled phenylalanine, whereas in the mature and autumn leaves the latter was a good precursor rather than the former for the gallic acid biosynthesis. Therefore, two pathways for gallic acid formation, through β-oxidation of phenylpropanoid and through dehydrogenation of shikimic acid, could be operating inAcer andRhus leaves, and the preferential pathway is altered by leaf age. In both plants, the incorporation rate of labeled phenyllactic acid during a 24 hr metabolic period was almost the same as that of labeled phenylalanine. The incorporation ofd-skikimic acid-G-14C,l-phenylalanine-U-14C andl-phenyllactic acid-U-14C into ellagic acid was very similar to the case of the radioactive gallic acid formation. Furthermore, regardless of the presence of unlabeled shikimic acid and/or phenylalanine, incorporation of the radioactivity of labeled gallic acid into ellagic acid occurred at a very high rate, suggesting the reciprocal radical reaction of gallic acid for the ellagic acid formation. The incorporation of labeled compounds into ellagitanins was also examined and their biosynthesis discussed further.  相似文献

Effects of the administration of phenylalanine to rats on incorporation in vivo or in vitro of [U-14C]glucose into cerebral lipids were studied during the first 5–10 days of postnatal development. In addition, the effects of added phenylalanine and its deaminated metabolites on incorporation of [U-14C]glucose by homogenates into lipids of developing rat brain were investigated. Hyperphenylalaninaemia reduced incorporation both in vivo and in vitro of [U-14C]glucose into cerebral lipids. 2. Phenylalanine or tyrosine added in vitro at concentrations equivalent to those in the brain of the hyperphenylalaninaemic rat (0-1 μmole/ml incubation medium) did not inhibit incorporation of [U-14C)glucose into lipids, although at much higher concentrations of phenylalanine (36 μumoles/ml incubation medium) slight inhibition (10 per cent) of incorporation of [U-14C]glucose into lipids was observed. 3. In contrast, the deaminated metabolites in general exerted greater inhibitory effects at lower concentrations. Phenyllactic acid, in comparison to phenylpyruvic and phenyl-acetic acid, was the most potent inhibitor of the incorporation in vitro of [U-14C]glucose into cerebral lipids. These results indicated that these metabolites of phenylalanine were the more potent inhibitors of cerebral lipid metabolism in immature animals.  相似文献

Catabolism of flavonol glucosides was investigated in plant cell suspension cultures using kaempferol 3-O-β-d-glucoside and kaempferol 7-O-β-d-glucoside labelled with 14C either in the glucose or in the flavonol moiety. Catabolic rates of glucosides were compared with those of free glucose and kaempferol. All substrates were degraded efficiently by cell cultures of mungbean, soybean, garbanzo bean and parsley. Based on 14CO2-formation, glucose from position 3 of kaempferol is 3–5 times more rapidly metabolized than that from position 7. The flavonol nucleus from both isomers is, however, oxidized to the same extent with a considerable portion of the flavonol being incorporated into insoluble polymeric cell material.  相似文献

The relative efficiency of incorporation of the exogenously supplied primary precursors [2-14C]acetate, [U-14C]glucose and [U-14C]sucrose into essential oil was determined in the immature leaves of threeCymbopogon species(C. martinii,C. winterianus andC. flexuosus). Acetate was most efficiently incorporated into essential oil inC. winterianus andC. flexuosus, whereas glucose was the best precursor inC. martinii. The observations are consistent when expressed as radioactivity [Bq] of essential oil per leaf, as percent incorporation or moles of precursors utilized for essential oil synthesis. Thus, there is selectivity in the efficiency of precursor utilization for the oil synthesis inCymbopogon species.  相似文献

The activity of the shikimic acid pathway during shoot initiation in tobacco (Nicotiana tabacum L. Wisconsin 38) callus was examined. Enhancement of the activities of 3-deoxy-d-arabino-heptulosonic acid 7-phosphate synthase, shikimate kinase, chorismate mutase, and anthranilate synthase was observed during culture of tobacco callus under shootforming conditions in comparison to tissue cultured under non-organforming conditions. Confirmation of these findings was obtained by examining the incorporation of d-[14C]glucose into quinic and shikimic acids and of [14C]shikimic acid into tyrosine, phenylalanine, and tryptophan.  相似文献

Datura innoxia plants were fed the R- and S-isomers of [3-14C]-3-hydroxy-3-phenylpropanoic acid, and [3-14C]cinnamic acid along with dl-[4-3H]phenylalanine. The hyoscyamine and scopolamine isolated from the plants 7 days later were labeled with tritium, but devoid of 14C, indicating that 3-hydroxy-3-phenylpropanoic acid and cinnamic acid are not intermediates between phenylalanine and tropic acid. The [3H] tropic acid obtained by hydrolysis of the hyoscyamine was degraded and shown to have essentially all its tritium located at the para position of its phenyl group, a result consistent with previous work.  相似文献

The reactions leading to cinnamic acids from phenylalanine as only substrate were investigated in organelles from Quercus pedunculata Ehrh. roots. –“F 10 000′” fraction, including mitochondria and micro-bodies, catalyses the first reaction, i.e., cinnamate formation by deamination of phenylalanine. – Microsomal fraction catalyses all the steps from phenylalanine to caffeic acid via cinnamate and p-coumarate. These results suggest that microsomes are the intracellular site of the cinnamic units synthesis. The enzymes involved in these reactions, associated in the same cellular compartment, does not form a multienzyme system. The formation of caffeic acid by isolated microsomes is demonstrated for the first time; the reaction may be realised by an enzyme different from phenolase. – The free phenolic acids are the metabolically active forms.  相似文献

The effect of retinoic acid on glycosaminoglycan biosynthesis was investigated in rat costal cartilage chondrocytes in vitro. At levels of 10?9 to 10?8m retinoic acid, 35SO4 uptake into glycosaminoglycans was reduced 50%. At these low levels of retinoic acid there was no evidence of lysosomal enzyme release. The results are explained best in terms of modification of glycosaminoglycan synthesis, rather than accelerated degradation. Retinoic acid selectively modified the incorporation of 35SO4 or [14C]glucosamine into individual glycosaminoglycans fractions under the conditions studied. The relative incorporation of radiolabeled precursor into heparan sulfate (and/or) heparin increased three- to fourfold. The relative incorporation of radiolabeled precursor remained constant for chondroitin 6-sulfate, whereas incorporation into chondroitin 4-sulfate and chondroitin (and/or) hyaluronic acid decreased. Under the conditions studied, retinoic acid did not appear to be cytotoxic and did exhibit selective control over glycosaminoglycan biosynthesis. It is suggested that the decreased incorporation of 35SO4 into glycosaminoglycans at hypervitaminosis A levels of retinol may be accounted for by the presence of low levels of retinoic acid, a naturally occurring metabolite.  相似文献

1. Lipogenesis in vivo has been studied in mice given a 250mg. meal of [U-14C]glucose (2·5μc) or given an intraperitoneal injection of 25μg. of [U-14C]glucose (2·0μc). 2. The ability to convert a [U-14C]glucose meal into fatty acid was not significantly depressed by 6–7hr. of starvation. In contrast, incorporation of 14C into fatty acid in the liver after the intraperitoneal dose of [14C]glucose was depressed by 80% and by more than 90% by 1 and 2hr. of starvation respectively. Carcass fatty acid synthesis from the [U-14C]glucose meal was not depressed by 12hr. of starvation, whereas from the tracer dose of [U-14C]glucose the depression in incorporation was 80% after 6hr. of starvation. 3. Re-feeding for 3 days, after 3 days' starvation, raised fatty acid synthesis and cholesterol synthesis in the liver fivefold and tenfold respectively above the levels in non-starved control mice. These increases were associated with an increased amount of both fatty acid and cholesterol in the liver. 4. After 18hr. of starvation incorporation of a [U-14C]glucose meal into carcass and liver glycogen were both increased threefold.  相似文献
