The amino acids of four species of Microsporidia parasitizing crustaceans were investigated: three species — Thelohania maenadis, Ormieresia carcini and Ameson pulvis — are parasites of Carcinus mediterraneus; Inodosporus sp. is a parasite of Palaemon serratus. Seventeen protein amino acids were identified of which aspartic acid, glutamic acid and lysine were quantitatively the most important, being quantitatively similar in all four parasites. The relative amounts of the next most abundant amino acids were found to vary and might serve as a systematic criterion at the genus level. For example, phenylalanine is predominant in Ormieresia and leucine in Inodosporus: serine in Thelohania and glycine in Ameson are also predominant, but to a lesser extent. The free amino acids composition shows little qualitative variation among the four genera, but quantitative differences are found in the composition of Microsporidia parasitizing the same host species; this may reflect variations in the amino acid metabolism of the parasite. The urea cycle in Ormeresia is most remarkable in this connection. The free amino acid level in the parasites was generally found to be in inverse proportion to the level in the host; the amino acids which are found to exist at high levels in the parasites correspond to essential amino acids of the Crustacea. Crystalloids accumulate in the vacuoles of the giant sporangiophores of Phycomyces blakesleeanus Burgeff during growth. On the basis of solubility in alkaline solutions, cytochemical staining reactions, trypsin sensitivity, optical absorption and response in the Lowry protein test, the crystalloids have been judged to consist principally of an acidic protein. In assays by Lowry test and by reference to optical absorption at 280 mμ, dark-grown sporangiophores were consistently found to contain from 2 to 4 times as much crystalloid material as light-grown counterparts. Concurrent assays of soluble phenolic materials revealed no significant effect of culture illumination, while carotene content of sporangiophores and mycelium was found to be raised from 2 to 4-fold by illumination during growth.  相似文献

Two hydroxybenzoic acids, gallic and protocatechuic acids, are abundant in the giant sporangiophores of the zygomycetePhycomyces blakesleeanus,much more so than in the basal mycelium or the culture medium. The actual concentrations vary with illumination, age of the culture, and composition of the medium. We devised a simple screening procedure to isolatehbamutants whose sporangiophores contained less gallic acid than the wild type. The most useful mutant had very low concentrations of hydroxybenzoic acids in the sporangiophores, but about the same as the wild type in the basal mycelium and the medium. The mutant was only slightly different from the wild type in growth and morphology. Mutant and wild-type sporangiophores grew away from ultraviolet C sources (260 nm) equally well. Contrary to previous conjectures, ultraviolet tropism does not depend on the ultraviolet absorption of gallic acid or other free hydroxybenzoic acids in the sporangiophore. Against expectations, phenols did not impair DNA extraction: sporangiophores produced better DNA preparations than basal mycelia and thehbamutant only slightly better than the wild type.  相似文献

A cell line of Eschscholtzia californica selected for meta-fluorotyrosine (MFT) tolerance was found to have 10-fold increased levels of phenylalanine and tyrosine compared to the parent line, while most other amino acids were only increased 2-fold. Tracer experiments with shikimic acid in the presence of MFT showed that the biosynthesis of the aromatic amino acids was not impaired in the tolerant line. Feeding experiments with phenylalanine, tyrosine, or shikimic acid also revealed a reduced turnover of the pools of the aromatic amino acids in the variant. Thus undisturbed de novo biosynthesis of the aromatic amino acids and dilution of toxic effects of MFT by the enlarged pool sizes seemed to be the main reason for the acquired tolerance. Despite the enlarged availability of the precursor tyrosine, formation of the benzophenanthridine alkaloids was enhanced neither in the growth nor in the production medium.  相似文献

A p-fluorophenylalanine- (PFP) resistant cell line of Nicotiana tabacum and wild type cells accumulating high and low levels of cinnamoyl putrescines, respectively, were used to study the formation of putrescine in the biosynthesis of cinnamoyl putrescines. Labelled arginine and ornithine were equally well incorporated into the main conjugates caffeoyl and feruloyl putrescine. Trapping experiments indicated that both amino acids were decarboxylated for putrescine biosynthesis. Nearly all alcohol-extractable radioactivity from the labelled amino acids was found as cinnamoyl putrescines in the PFP-resistant cell line, whereas wild type cells retained significant radioactivity in the amino acids. The enzyme activities of arginine and ornithine decarboxylases in the resistant cell line were increased 3- to 6-fold.  相似文献

Under the circumstances of experimentation described, the sporangiophores of Phycomyces are found to be most sensitive to stimulation by light in the violet between 400 and 430 mµ. Toward the red, sensitivity falls to nearly zero near 580 mµ, while in the near ultra-violet around 370 mµ, sensitivity is still high. The previous experiments of Blaauw had placed the point of greatest sensitivity some 80 mµ nearer the red end of the spectrum. Because of the known presence in the sporangiophores of Phycomyces of "accessory" pigments, care must be taken in identifying such results with the absorption spectrum of the photosensitive substance.  相似文献

The pool sizes of the common amino acids in purified intact chloroplasts from Vicia faba L. were measured (nanomoles per milligram chlorophyll). The three amino acids present in the highest concentrations were glutamate, aspartate, and threonine. Alanine, serine, and glycine were each present at levels between 15 and 20 nanomoles per milligram chlorophyll and 13 other amino acids were detectable at levels below 10.  相似文献

Gravitropic response of sporangiophores ofPilobolus crystallinus was studied by successive microscopic observation of the sporangiophores horizontally placed in the dark (red light) and by analysis of sporangiophore response to centrifugal stimulation. Negative tropism against the gravitative and also centrifugal stimulation was found only in mature sporangiophores after development of sporangium and after the resumption of elongation beneath the fully-developed subsporangial vescle, but there was no response in younger sporangiophores, implying that the gravitative perception system of the sporangiophores is dependent on their developmental stages.  相似文献

By incorporating various secondary compounds in the normal diet of larval Callosobruchus maculatus bruchids, we show that the effects of any particular compound are dosage-dependent. Alkaloids are generally the most toxic of the compounds tested. Non-protein amino acids are more toxic than protein amino acids but the latter can be toxic at 1 and 5% incorporation in the diet. The non-protein amino acid homoarginine has a salutary effect on larval survival at low concentrations. A variety of other secondary compounds found in seeds are toxic at various levels representative of those levels found in seeds in nature, and for all secondary compounds tested a 0.1–5% incorporation in the diet often has a detrimental effect on production of adult beetles. We conclude that many of the secondary compounds found in seeds are likely to be toxic to at least some animal, and thus are likely to be responsible at least in part for the extreme host-specifity shown by seed-eating insects.  相似文献

A comparison of barley (Hordeum vulgare L.) leaves was made between the cytosolic content of amino acids and sucrose as determined by subcellular fractionation and the corresponding concentration in phloem sap, which was collected continuously for up to 6 days from severed aphid stylets. Because amino acids were found to be almost absent from the vacuoles, and because the amino acid patterns in the stroma and cytosol are similar, whole leaf contents could be taken as a measure of cytosolic amino acid levels for a comparison of data during a diurnal cycle. The results show that the pattern of amino acids in the phloem sap was very similar to the pattern in the cytosol. Therefore, we concluded that the overall process of transfer of amino acids from the cytosol of the source cells into the sieve tubes, although carrier mediated, may be a passive process and that the translocation of amino acids via the sieve tubes requires the mass flow of sucrose driven by the active sucrose transport involved by the phloem loading.  相似文献

Candida albicans cells have low levels of ergosterol when grown in ascorbic acid-supplemented media. When cells are grown in hydroquinone-supplemented media, the ergosterol levels became higher as compared to normal cells. The uptake of lysine, glycine, glutamic acid, proline, methionine and serine is reduced in hydroquinone-supplemented cells. In contrast to hydroquinone-supplemented cells, the rate and level of accumulation of these amino acids are higher in ascorbic acid-supplemented cells. Nystatin-resistant isolates of C. albicans with low ergosterol contents also exhibit an increased rate and level of accumulation of these amino acids. The uptake of phenylalanine and leucine remained unaffected by such a change in ergosterol levels brought about by different supplementation of the media. The results demonstrate a correlation between ergosterol levels and amino acids uptake. Contrary to various reports, the rate of K+ efflux does not seem to correlate with the amino acid uptake in C. albicans cells.  相似文献

Callus cultures of peanut (Arachis hypogaea L. cv Valencia Tennessee Red) cotyledons grown on Linsmaier and Skoog medium containing normal levels of auxin and cytokinin do not synthesize either 4-methyl-eneglutamic acid or 4-methyleneglutamine, which nonprotein amino acids are normally found in significant amounts in peanut plants. If mature peanut embryos (with cotyledons removed) are germinated and grown on a similar medium containing no added phytohormone, normal levels of these two amino acids accumulate. The addition of an auxin, however, prevents formation of 4-methyleneglutamic acid and 4-methyleneglutamine; typical levels of other free amino acids are seen and excised embryos so cultured develop into apparently otherwise normal plants. Kinetin addition to embryo cultures has little or no effect. 4-Methyleneglutamine is formed when 4-methyleneglutamic acid is added to embryo cultures maintained on auxin-containing medium, indicating that the phytohormone does not block amidation but rather the biosynthesis of 4-methyleneglutamic acid.  相似文献

Using a partially purified bovine brain extract, our lab identified three novel endogenous acyl amino acids in mammalian tissues. The presence of numerous amino acids in the body and their ability to form amides with several saturated and unsaturated fatty acids indicated the potential existence of a large number of heretofore unidentified acyl amino acids. Reports of several additional acyl amino acids that activate G-protein coupled receptors (e.g., N-arachidonoyl glycine, N-arachidonoyl serine) and transient receptor potential channels (e.g., N-arachidonoyl dopamine, N-acyl taurines) suggested that some or many novel acyl amino acids could serve as signaling molecules. Here, we used a targeted lipidomics approach including specific enrichment steps, nano-LC/MS/MS, high-throughput screening of the datasets with a potent search algorithm based on fragment ion analysis, and quantification using the multiple reaction monitoring mode in Analyst software to measure the biological levels of acyl amino acids in rat brain. We successfully identified 50 novel endogenous acyl amino acids present at 0.2 to 69 pmol g−1 wet rat brain.  相似文献

Sesamum indicum was grown in complete or potassium deficient nutrient solution and amino acids, amines, nitrogen and potassium were determined weekly in the leaves. The incorporation of l-arginine-[U-14C] into protein was also followed. The interconversions of the amino acids of the ordithine-urea cycle, and their contribution to the formation of amines, were studied in cell-free extracts and intact leaves using labelled amino acids. As the level of potassium in the leaves decreased, the levels of the amino acids ornithine, citrulline and arginine, and of the amines putrescine, N-carbamylputrescine and agmatine increased. Potassium deficiency also reduced the rate of protein synthesis. Putrescine appears to be formed preferentially from citrulline with N-carbamylputrescine as intermediate.  相似文献

Nitrogen is a macronutrient present in a wide range of cellular compounds including proteins, nucleic acids, amino acids and lipids. The levels of nitrogen often regulate many aspects of plant metabolism, growth and development. Extensive research has been conducted into the effects of N nutrition in model plants, however relatively little is known about the metabolic response of perennial ryegrass (Lolium perenne) grown under different N-supply conditions. This study aimed to identify key metabolic responses activated rapidly after challenging plants with different levels of N-supply. The metabolic response of the leaves of seven different L. perenne genotypes to three N treatments (low, medium and high levels of N) was characterized using a GC–MS approach. After 24 h it was observed that the levels of amino acids correlated with the levels of N-supply. Furthermore the results indicated that plants experiencing N-limitation accumulated very-long chain fatty acids and precursors of secondary aromatic metabolites while sugar levels were not significantly affected indicating a remobilization of carbon. Plants grown under high levels of N were found to have enhanced levels of inositol and threonic acid which could reflect an alteration of the redox potential under stress. Further analysis of Pearson’s correlation coefficient provided evidence that the chlorophyll metabolism may also be regulated in plants grown at high N concentrations.  相似文献

Precolumn formation of o-phthalaldehyde (OPA) derivatives of amino acids, followed by reverse-phase separation and fluorescent detection, provides rapid, sensitive amino acid analysis. Eighteen OPA-amino acid derivatives are resolved on a Micropack MCH 5 column and can be measured at picomole levels. Ease of derivative preparation and separation makes liquid chromatographic analysis of OPA-amino acids a convenient and improved technique for measuring or confirming the presence of low levels of amino acids in aqueous solutions. Use of the method was demonstrated by measuring low concentrations of amino acids released from zooplankters stressed by contaminants.  相似文献

Bacillus subtilis can perform chemotaxis toward all 20 l-amino acids normally found in proteins. Loss of a single chemoreceptor, McpC, was previously found to reduce chemotaxis to 19 of these amino acids. In this study, we investigated the amino acid-sensing mechanism of McpC. We show that McpC alone can support chemotaxis to 17 of these amino acids to varying degrees. Eleven amino acids were found to directly bind the amino-terminal sensing domain of McpC in vitro. Sequence analysis indicates that the McpC sensing domain exhibits a dual Per-Arnt-Sim (PAS) domain structure. Using this structure as a guide, we were able to isolate mutants that suggest that four amino acids (arginine, glutamine, lysine, and methionine) are sensed by an indirect mechanism. We identified four candidate binding lipoproteins associated with amino acid transporters that may function in indirect sensing: ArtP, GlnH, MetQ, and YckB. ArtP was found to bind arginine and lysine; GlnH, glutamine; MetQ, methionine; and YckB, tryptophan. In addition, we found that ArtP, MetQ, and YckB bind the sensing domain of McpC, suggesting that the three participate in the indirect sensing of arginine, lysine, methionine, and possibly tryptophan as well. Taken together, these results further our understanding of amino acid chemotaxis in B. subtilis and gain insight into how a single chemoreceptor is able to sense many amino acids.  相似文献

The objective of this study was to determine the role of a lactococcal branched-chain amino acid aminotransferase gene, ilvE, in the production of branched-chain fatty acids. Lactococcus lactis subsp. lactis LM0230 and an ilvE deletion mutant, JLS450, produced branched-chain fatty acids from amino and α-keto acids at levels above α-keto acid spontaneous degradation and the fatty acids' flavor thresholds. The deletion mutant produced the same amounts of branched-chain fatty acids from precursor amino acids as did the parent. This was not the case, however, for the production of branched-chain fatty acids from the corresponding precursor α-keto acids. The deletion mutant produced a set of fatty acids different from that produced by the parent. We concluded from these observations that ilvE plays a role in the specific type of fatty acids produced but has little influence on the total amount of fatty acids produced by lactococci.  相似文献
