In order to investigate the effects of acute heat stress and subsequent stress removal on function of hepatic mitochondrial respiration, production of reactive oxygen species (ROS) and lipid peroxidation in broiler chickens, 128 six-week-old broiler chickens were kept in a controlled-environment chamber. The broiler chickens were initially kept at 25 °C (relative humidity, RH, 70 ± 5%) for 6 d and subsequently exposed to 35 °C (RH, 70 ± 5%) for 3 h, then the heat stress was removed and the temperature returned to 25 °C (RH, 70 ± 5%). Blood and liver samples were obtained before heat exposure and at 0 (at the end of the three-hour heating episode, this group is also abbreviated as the HT group), 1, 2, 4, 8, 12 h after the stress was removed. The results showed that acute heat stress induced a significant production of ROS, function of the mitochondrial respiratory chain, antioxidative enzymes [superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px)] activity, and formation of malondialdehybe (MDA). Within the first 12 h after removal of the heat stress, the acute modification of the above parameters induced by heat stress gradually approached to pre-heat levels. The results of the present study suggest that acute exposure to high temperatures may depress the activity of the mitochondrial respiratory chain. This leads to over-production of ROS, which ultimately results in lipid peroxidation and oxidative stress. When the high temperature was removed, the production of ROS, mitochondrial respiratory function and oxidative injury that were induced by acute heat exposure gradually approached the levels observed before heating, in a time-dependent manner.  相似文献

The pioneering work by Professor Soller et al., among others, on the use of genetic markers to analyze quantitative traits has provided opportunities to discover their genetic architecture in livestock by identifying quantitative trait loci (QTL). The recent availability of high-density single nucleotide polymorphism (SNP) panels has advanced such studies by capitalizing on population-wide linkage disequilibrium at positions across the genome. In this study, genomic prediction model Bayes-B was used to identify genomic regions associated with the mean and standard deviation of egg weight at three ages in a commercial brown egg layer line. A total of 24,425 segregating SNPs were evaluated simultaneously using over 2900 genotyped individuals or families. The corresponding phenotypic records were represented as individual measurements or family means from full-sib progeny. A novel approach using the posterior distribution of window variances from the Monte Carlo Markov Chain samples was used to describe genetic architecture and to make statistical inferences about regions with the largest effects. A QTL region on chromosome 4 was found to explain a large proportion of the genetic variance for the mean (30%) and standard deviation (up to 16%) of the weight of eggs laid at specific ages. Additional regions with smaller effects on chromosomes 2, 5, 6, 8, 20, 23, 28 and Z showed suggestive associations with mean egg weight and a region on chromosome 13 with the standard deviation of egg weight at 26-28 weeks of age. The genetic architecture of the analyzed traits was characterized by a limited number of genes or genomic regions with large effects and many regions with small polygenic effects. The region on chromosome 4 can be used to improve both the mean and standard deviation of egg weight by marker-assisted selection.  相似文献

Several blood parameters were studied in the dogfish Scyliorhinus canicula subjected to different confinement conditions (transport and maintenance) and zinc contamination. Confinement stress significantly decreased erythrocyte counts (RBCC), haematocrit (Ht), haemoglobin (Hb), leucocrit (Lt), mean corpuscular haemoglobin (MCH) and mean corpuscular haemoglobin concentration (MCHC). Mean corpuscular volume (MCV) did not change with this stress and plasma glucose concentration significantly increased. In fish subjected to confinement stress conditions, Zn treatment significantly decreased Hb, MCH and MCHC and plasma glucose concentration. Ht and MCV did not change and Lt and RBCC significantly increased. Results are discussed related to other stress effects and effects of zinc treatment on fish confined in non-stressful conditions.  相似文献

This experiment was conducted to evaluate the effects of chromium (chromium picolinate, CrPic) and zinc (ZnSO4·H2O) on egg production, egg quality, and serum insulin, corticosterone, glucose, cholesterol, and total protein concentrations of laying hens reared under a low ambient temperature (6.8°C). One hundred twenty laying hens (Hy-Line; 32 wk old) were divided into 4 groups, 30 hens per group. The laying hens were fed the control diet (T1) or the control diet supplemented with either 400 μg of Cr/kg of diet (T2), 30 mg of Zn/kg of diet (T3), or 400 μg of Cr plus 30 mg of Zn/kg of diet (T4). Although the dry matter intake (DMI) was similar (p>0.05) for all treatment groups, supplemental chromium and zinc either individually or together increased live-weight change, egg production, and improved feed efficiency (p<0.05). However, no significant differences were observed between T4 and T2 or T3. Compared to T1, supplemental chromium and zinc increased egg weight, eggshell weight, eggshell thickness, egg specific gravity, and Haugh unit (p<0.05) in T2, T3, and T4 groups, among which there was no significant difference. Serum insulin concentration increased (p<0.05) and corticosterone concentration decreased (p<0.05) with dietary chromium and zinc supplementation. Serum glucose and cholesterol concentrations decreased (p<0.05) and protein concentrations increased (p<0.001) with dietary chromium and zinc supplementation in all treatment groups. The results of this study indicated that either supplemental dietary chromium or zinc increased plasma insulin and decreased corticosterone concentrations and that had a positive effect on performance of laying hens under low ambient temperature.  相似文献

The effects of repeated weekly blood sampling on blood parameters were studied over a period of 22 days in a salmonid, Salmo gairdneri , and cyprinid fish, Leuciscus idus . The procedure induced mild anaemia demonstrated by decrease of red blood cell counts, haemoglobin and PCV. The changes were more marked in L. idus , which showed signs of increased red cell destruction. No significant trends could be observed in the white blood picture. The results show that blood tests are possible in individual fish, especially S. gairdneri over a long period.  相似文献

AIMS: This work aims at studying the impact of competitive exclusion of Salmonella serotype Enteritidis infection in layer chickens, by microbiota of fresh and dried-modified yoghurt, on egg production and weight, protection against infection, and on yolk-antibody and cholesterol levels. METHODS AND RESULTS: Four groups of 27-week-old layer chickens were included in this study. After an initial enrofloxacin treatment, groups 1 and 2 were administered fresh or dry yoghurt (respectively) for 14 days. Groups 1, 2 and 3 were challenged intraoesophageally with Salm. Enteritidis, on the sixth day of yoghurt administration, while group 4 was left unchallenged and without yoghurt administration. No significant difference in percent infectivity of visceral organs with Salm. Enteritidis was observed between the groups. The yoghurt administered groups showed an early significant antibody response in their yolk on the seventh day postchallenge (P < 0.05) and highest egg production and weight. Finally, the egg yolk cholesterol concentration was higher in Salm. Enteritidis-challenged groups than that observed in the unchallenged group. CONCLUSIONS: The results point to the possible involvement of yoghurt administration in immunopotentiation and improvement of egg production and weight. SIGNIFICANCE AND IMPACT OF THE STUDY: These findings warrant further research that could improve immunity and production in layer chickens infected with Salm. Enteritidis.  相似文献

 An investigation was carried out to verify whether the heat stress hyperthermia response of broilers is prostaglandin-dependent. Male broiler chickens of the Hubbard-Petterson strain, aged 35–49 days, were used. Chickens were injected with indomethacin (1 mg/kg intraperitoneally ) 15 min before or 2 h after heat exposure (at 35°C for 4 h), and rectal temperature was measured before injection and up to 4 h thereafter. Birds were separated into two groups with and without access to water during heat stress. The increase in rectal temperature was lower (P<0.05) in birds with access to drinking water during heat exposure. All birds injected with indomethacin exhibited an increase in rectal temperature, irrespective of whether indomethacin was administered before or in the course of the rise in temperature. The results revealed that the increase in rectal temperature during heat exposure is not prostaglandin-dependent, and that the use of cyclooxigenase inhibitors is not recommended to attenuate heat stress hyperthermia in broiler chickens. The tolerance of chickens to acute heat stress, evaluated by the time required to reach the critical body temperature (Tr) of 44.5 degrees C, was markedly enhanced as the period of fasting was extended. 2. Fasting reduced the rates of heat-induced changes in blood acid-base and electrolyte status. 3. Changes in Tr were correlated with changes in blood pH, pCO2, [Cl-] and [Pi] but not with changes in [Na+] or [K+]. 4. Recent studies have shown that chromium (Cr) could alleviate the negative effects of heat stress on livestock and poultry, but there is little information available to laying ducks. This study aimed to investigate the effects of dietary addition of chromium propionate on laying performance, egg quality, serum biochemical parameters and antioxidant status of laying ducks under hot (average 32 °C) and humid (average 75% relative humidity) summer conditions. A total of 900 66-week-old weight- and laying-matched Shanma laying ducks were randomly divided into five treatments, each with 6 replicates of 30 individually caged birds. The birds were fed either a basal diet or the basal diet supplemented with either 200, 400, 600, or 800 μg/kg Cr as chromium propionate. All laying ducks were given feed and water ad libitum for 5 weeks. The results showed that dietary supplementation with chromium propionate significantly increased the laying rate and yolk colour score (P < 0.05). Treatment with 400 μg/kg Cr as chromium propionate significantly decreased the feed/egg ratio by 5.4% (P < 0.05). Increased supplemental Cr from 0 to 800 μg/kg resulted in an increase in albumen height and the Haugh unit linearly (P < 0.05). Increased supplemental Cr decreased serum cortisol (P < 0.001, linear; P = 0.008, quadratic), heat shock protein-70 (P < 0.001, linear; P = 0.007, quadratic) and glucose (P = 0.007, linear), whereas it increased serum insulin (P = 0.011, Linear), total protein (P = 0.006, linear; P = 0.048, quadratic) and albumin (P = 0.035, linear; P = 0.088, quadratic). Dietary Cr levels increased the activities of superoxide dismutase and glutathione peroxidase, the total antioxidant capacity linearly and quadratically (P < 0.05). A linear and quadratic (P < 0.05) decrease of the malondialdehyde concentrations in response to dietary Cr level was observed. These results indicated that dietary supplementation of Cr as chromium propionate, particularly at 800 μg/kg could beneficially affect the laying rate, egg quality and antioxidant function, as well as modulate the blood biochemical parameters of laying ducks under heat stress conditions.  相似文献

Radioactive microspheres were used to measure cardiac output and blood flow to most major tissues, including those in the pregnant uterus, in late-pregnant ewes at rest and during treadmill exercise (approximately 3-fold increase in metabolic rate for 30 min) in thermoneutral (TN) (dry bulb temperature (Tdb) = 13 degrees C, wet bulb temperature (Twb) = 10 degrees C) and mildly hot (MH) (Tdb = 40 degrees C, Twb = 27 degrees C) environments. Exercise caused major increases in blood flow to respiratory muscles, nonrespiratory limb muscles, and adipose tissue, and flow was decreased to some gastrointestinal tissues, spleen, pancreas, and to placental and nonplacental tissues in the pregnant uterus. Heat exposure had relatively little effect on these exercise-induced changes, except that flow was further increased in the respiratory muscles. Results are compared with those of a similar study on nonpregnant sheep in which changes in muscle, skin, and visceral flows during exercise were attenuated by heat exposure. It is suggested that redistribution of blood flow from the pregnant uterus, which in resting ewes took 22% of cardiac output, is a significant buffer against the potentially deleterious effects of combined exercise and heat stress on blood flow to exercising muscles and thermoregulatory tissues.  相似文献

Thermal stress has been shown to result in decreased egg production, decreased eggshell quality, and ultimately millions of dollars in losses to the industry. Therefore, there are many factors to consider when implementing genetic selection programs aimed at improving egg production under tropical conditions. So, trial is trying to improve the productivity and eggshell quality traits of the Fayoumi chicken under high ambient temperatures via selection programs and gene expression. In the present study, day-old Fayoumi chicks were raised either under normal temperature (control) or conditions of thermal stress (the heated group). At 35 weeks, male and female chickens from the control group were mated randomly and females selected for higher egg production and eggshell strength were mated to male siblings to obtain the progeny of the first generation (F1). F1 birds were further selected and mated to obtain the progeny of the second generation. Our results show that egg production and eggshell strength traits improved over successive generations via selection under conditions of heat stress. Furthermore, the reduction in egg production and eggshell strength as a result of heat stress declined from one generation to the next in birds selected for good heat tolerance, and an inverse relationship was observed between the OC-17 and eggshell strength. Additionally, levels of HSP90 and gene expression increased in the two successive generations, indicating that both productivity and heat tolerance were enhanced due to selection in birds raised under conditions of thermal stress. Moreover, generation exerted an important effect on this trait. Thus, desirable traits such as improved heat tolerance in producing lines were observed in Fayoumi chickens exposed to conditions of thermal stress via selection. Therefore, modern advances in studies of poultry breeding and genetics, such as gene expression studies, should be examined.  相似文献

A marked increase in the rate of oxagen consumption and heat production, measured by the direct method, is observed in the chick embryogenesis. The intensity of respiration and heat production of the embryos decreases as the development proceeds. During development the data obtained by direct and indirect calorimetry diverge. This divergence, referred to as psi u-function, gradually decreases by the moment of hatching. Differences in the value of heat production and psi u-function were found in crosses of meat and egg directions, related to differences in the growth rate and productivity of adult fowl.  相似文献

The effect of transportation and confinement stress on Dicrocoelium dendriticum egg production was investigated. Sheep passing a minimum of 200 eggs g-1 of faeces were selected from a naturally infected flock. A group of six ewes (group A) was transferred to the laboratory premises and kept indoors for 28 days, while another group (B) of six ewes remained on pasture and was used as a control. Faecal examinations and egg counting were performed weekly, on all sheep, from one week before to 28 days after the transportation of the animals. Comparison of faecal egg counts between groups revealed higher (P < 0.01) counts in transported sheep sampled on days 7, 14 and 28 of the trial. Furthermore, egg counts obtained from sheep that were transferred remained consistently high while the ones from sheep that remained on pasture showed significant variation. Therefore, it is concluded that stress-inducing factors, such as transportation and confinement may enhance egg production of D. dendriticum.  相似文献

A study was conducted using 360 broiler chickens to evaluate the effects of dietary vitamin E (0, 125 and 250 mg/kg), selenium (Se, 0, 0.5 and 1 mg/kg), or their different combinations on immune response and blood biological parameters of broilers raised under either thermoneutral (TN, 23.9 °C constant) or heat stress (HS, 23.9 to 37 °C cycling) conditions. Humoral immunity was assessed by intravenous injection of 7 % sheep red blood cell (SRBC) followed by evaluation of serum for antibody titers in primary and secondary responses. Heterophil to lymphocyte (H/L) ratio also determined as an indicator of stress. Furthermore, at the end of the experiment, birds were bled for determination of some biological parameters. There was a significant reduction in body weight and feed intake, but the feed conversion ratio increased when the birds were exposed to HS (P?<?0.05). Body weight and feed intake were not influenced significantly by dietary vitamin E and Se (P?>?0.05), whereas feed conversion was improved significantly by 125 mg/kg vitamin E (P?<?0.05). The liver and lymphoid organ weights as well as IgM and IgG, antibody titers for primary and secondary antibody responses to SRBC were reduced significantly under HS (P?<?0.05). Heat stress also resulted in a significant increase in H/L ratio (P?<?0.05). Dietary vitamin E resulted in improvement of primary and secondary antibody responses both in TN and HS broilers (P?<?0.05). The HS birds also showed an improved antibody titer in secondary response with high concentration of Se (P?<?0.05). Vitamin E and Se had interactive effects on anti-SRBC titers; however, no consistent differences were found between dietary levels during the study. The H/L ratio decreased by feeding vitamin E at both levels either under HS or TN conditions (P?<?0.05). The serum concentrations of glucose, triglycerides, total cholesterol, and LDL-cholesterol were increased but serum HDL-cholesterol decreased in HS broilers (P?<?0.05).  相似文献

The influence of artificially induced anaemia on thermal strain was evaluated in trained males. Heat stress trials (38.6°C, water vapour pressure 2.74 kPa) performed at the same absolute work rates [20 min of seated rest, 20 min of cycling at 30% peak aerobic power (O2peak), and 20 min cycling at 45% O2peak] were completed before (HST1) and 3–5 days after 3 units of whole blood were withdrawn (HST2). Mild anaemia did not elevate thermal strain between trials, with auditory canal temperatures terminating at 38.5°C [(0.16), HST1] and 38.6°C [(0.13), HST2; P > 0.05]. Given that blood withdrawal reduced aerobic power by 16%, this observation deviates from the close association often observed between core temperature and relative exercise intensity. During HST2, the absolute and integrated forearm sweat rate ( sw) exceeded control levels during exercise (P < 0.05), while a suppression of forehead sw occurred (P < 0.05). These observations are consistent with a possible peripheral redistribution of sweat secretion. Selected blood parameters (arterial pH, O2 and CO2 tensions, oxygen content, bicarbonate and lactate concentrations, haematocrit, haemoglobin, red blood cell count, mean corpuscular haemoglobin, mean corpuscular volume and mean corpuscular haemoglobin concentration), oxygen consumption and ventilatory frequency were measured 0, 3 and 24 hours after cannulation of the dorsal aorta of 22 dogfish. Correlations were calculated between all pairs of variables along the post-operational recovery period. Results are discussed in terms of the integrated dynamics of the recovery process subsequent to handling, anaesthesia and surgery stress. It is suggested that erythrocyte swelling plays an important role in maintaining tissue oxygen supply during recovery from operational procedures.  相似文献
