We have developed seven microsatellite primers for the solitary wasp Ancistrocerus adiabatus. The number of alleles per locus ranged from three to 14. The markers were developed to study population structure, inbreeding and sex determination of A. adiabatus. For each locus, we genotyped 30 female wasps collected from Southwest Michigan. The observed heterozygosity ranges from 0.067 to 0.933. We have also determined that primers for these microsatellite loci amplify in three other species in the genus: Ancistrocerus antilope, Ancistrocerus catskill, and Ancistrocerus campesterus.  相似文献

Background  

The major lineages of eusocial insects, the ants, termites, stingless bees, honeybees and vespid wasps, all have ancient origins (≥ 65 mya) with no reversions to solitary behaviour. This has prompted the notion of a 'point of no return' whereby the evolutionary elaboration and integration of behavioural, genetic and morphological traits over a very long period of time leads to a situation where reversion to solitary living is no longer an evolutionary option.  相似文献

To evaluate the role that a trap‐nest cover might have on sampling methodologies, the abundance of each species of trap‐nesting Hymenoptera and the parasitism rate in a Canadian forest were compared between artificially covered and uncovered traps. Of trap tubes exposed at eight forest sites in six trap‐nest boxes, 531 trap tubes were occupied and 1216 individuals of 12 wasp species of four predatory families, Vespidae (Eumeninae), Crabronidae, Sphecidae and Pompilidae emerged over 2 years, and no bee species were found. Results indicated that artificial covering led to a significant increase in the number of nested tubes of Ancistrocerus adiabatus, Ancistrocerus antilope, Ancistrocerus campestris and Auplopus mellipes, and significant effects of covering were not found for the other species. No significant difference in the overall parasitism rate between covered and uncovered traps was noted. These suggested that the covering technique could provide more opportunities for some wasp species to colonize trap nests.  相似文献

White-faced hornet, yellow hornet, and yellow jacket venoms have very similar protein compositions; each contains mainly three basic proteins. Two of these proteins have hyaluronidase and phospholipase activities and the third one, designated as antigen 5, is of as yet unidentified biochemical function. These three proteins have molecular weights of about 45 000, 35 000, and 25 000, respectively. The three proteins of white-faced hornet venom have been purified to near homogeneity, while this is the case only for antigen 5 of yellow hornet and yellow jacket venoms. Strong antigenic cross-reaction of the hyaluronidase from these three vespid venoms was observed using specific rabbit anti-venom sera, while weak cross-reactions of phospholipases and of antigen 5s were observed. All three proteins are active as allergens to varying degrees in vespid sensitive individuals. With each vespid venom its antigen 5 seems to be the major allergen. The results help to clarify the commonly observed varying degrees of multiple sensitivity of people to different vespids.  相似文献

Vespid wasps (Vespula vulgaris L. and V. germanica Fab. Hymenoptera; Vespidae) are highly abundant in 1 million ha of New Zealand's indigenous beech forests (Nothofagus spp.) and have had detrimental effects on the New Zealand native fauna. This hyperabundance is due in part to the vast supply of carbohydrate‐rich honeydew produced by scale insects Ultracoelostoma spp. native to New Zealand. Current control methods include the use of wet cat food as a protein source with insecticide as a lure‐and‐kill‐based system, but there are problems with fresh baits degrading rapidly, and a more durable formulation would enable the expansion and longevity of wasp control. Four crude protein baits were tested for vespid attraction. Green‐lipped mussels had the highest vespid catch of the crude baits tested, and aged and fresh mussels were equally attractive. From headspace analysis of the green‐lipped mussel volatiles, a series of butanoate esters, 3‐octanone and 1‐octen‐3‐ol were identified as possible attractants. These compounds were tested individually and in various blend combinations for the attraction of Vespula wasps in matagouri vegetation at the edge of beech forests. We found synergistic effects between single attractive compounds when tested in various combinations, and the multicomponent lures were more attractive to these wasps than heptyl and octyl butanoate, previously identified attractants for vespid species. The new multicomponent lures could form the basis for a new generation of attractants for social wasps that can provide sustained control methods for invasive vespid wasps.  相似文献

Vespid venoms contain Antigen 5, an important allergen whose primary structure and immunological behavior have been extensively studied from venoms of vespids of the Northern Hemisphere. We report herein structural and immunological aspects of Antigen 5 from Polybia scutellaris subspecies rioplatensis (vulgar name: camoati) found in South America. Mast cell degranulation, histamine release, and IgE induction experiments performed in mice allow us to suggest that P. scutellaris Antigen 5 is a variant with reduced IgE response and anaphylactic activity. Sequence data indicate that the protein has a 72.5-90.3% similarity to that of members of the vespid Antigen 5 family with an already known primary structure. Moreover, results suggest that the protein-a new member of an extracellular protein superfamily-could be a good candidate for immunotherapy related to vespid allergy.  相似文献

Introduced vespid wasps (Vespula germanica and V. vulgaris) are highly efficient predators of native invertebrates. They have the potential to reduce populations of threatened species and change ecosystem dynamics, yet their impact is largely unknown in Australia. The introduction of vespid wasps has coincided with a decline in numbers of threatened Ptunarra brown butterflies (Oreixenica ptunarra) in Tasmania, Australia. The Ptunarra brown butterfly is endemic to Tasmania, where its habitat has been fragmented by clearance for agriculture and forestry. Local extinctions of the species were previously thought to be principally due to its inability to fly the long distances between habitat patches in this disjointed landscape. We investigate the importance of the new threat of vespid wasp predation in the decline of O. ptunarra in the highland grasslands of northwest Tasmania. Numbers of O. ptunarra analysed over a period of 15 years dramatically declined after the arrival of vespid wasps. Wasp control was trialled to determine whether it affected butterfly numbers. Current control methods decreased wasp numbers considerably, resulting in a small increase in butterfly numbers, indicating that wasp predation is keeping O. ptunarra at low densities. Without ongoing conservation measures, it is likely that butterfly numbers will stay low, potentially leading to genetic bottlenecks and more local extinctions. An increase in the intensity of wasp control, in combination with other conservation management methods, is required for the protection and recovery of O. ptunarra.  相似文献

The multiple independent origins of eusociality in the insect order Hymenoptera are clustered in only four of more than 80 families, and those four families are two pairs of closely related taxa in a single part of the order. Therefore, although ordinal-level characteristics can contribute to hymenopteran eusocial evolution, more important roles have been played by traits of infraordinal taxa that contain the eusocial forms. Many factors have been proposed and discussed, but assessments of traits' salience to eusocial evolution have heretofore not been joined to phylogenetics. In the present analysis, cladograms of superfamilies and families of Hymenoptera and of the family Vespidae are used to ordinate the appearance of traits that play roles in vespid eusociality. Proximity of traits' first appearance to the origin of eusocial Vespidae is taken as one measure of traits' salience to vespid eusocial evolution. Traits that subtend only eusocial taxa and that are uniquely associated with eusociality have foundations in more general traits that subtend more inclusive taxa. No single trait is uniquely causative of vespid eusocial evolution. High-salience traits that closely subtend vespid eusociality include nesting, oviposition into an empty nest cell, progressive provisioning of larvae, adult nourishment during larval provision malaxation, and inequitable food distribution among nestmates. The threshold characteristic of Polistes-grade eusociality is life-long alloparental brood care by first female offspring who remain, uninseminated, at their natal nest. Traits directly associated with occurrence of such workers are larva-adult trophallaxis, which can foster relatively low larval nourishment early in a colony cycle, and protogyny and direct larval development, which combine to yield restricted mating opportunities for female offspring that are the first to emerge in the colony cycle. Trait mapping suggests no role for asymmetry of relatedness due to haplodiploidy, but it suggests high salience for haplodiploidy as a mechanism enabling the production of all-female clutches of first offspring.  相似文献

An effective visual signal elicits a strong receiver response. The visual receptors of most insects are sensitive to ultraviolet (UV), blue and green light. The decorations of certain orb web spiders may be described as a type of visual signal, a sensory trap, as they exploit visual biases in insects. We filtered UV and blue light from the decorations of Argiope keyserlingi , under field conditions, using plastic sheets to test if the UV and blue light reflected affects the type of prey caught. We found that houseflies, blowflies, stingless bees, honeybees and vespid wasps were caught more frequently in webs with decorations than webs without, while ichneumonid wasps were caught less frequently. Blowflies, stingless bees, honeybees and vespid wasps were caught more often in unfiltered decorated webs. These insects also have receptor sensitivities in the blue and UV. We showed that exploiting visual sensory biases plays an integral role in attracting insects to orb web decorations. Allergic reactions to vespid stings are one of the major causes of IgE-mediated anaphylaxis. Vespa and Vespula venoms are closely related; Polistes venom is more distantly related and its allergens are less well studied. There is limited cross-reactivity between Polistes and the other vespid venoms because of differences in the epitopes on the allergen molecules.In this study, the major allergens of Polistes gallicus are isolated and characterized. P. gallicus venom contains four major allergens: phospholipase, antigen 5 (Ag5), hyaluronidase and protease that were characterized by mass spectrometry and specific binding to IgE. The complete amino acid sequence of Ag5 and the sequence of the N-terminal region of phospholipase were also determined. The alignment of Ag5 from P. gallicus (European species) and Polistes annularis (American species) shows an 85% identity that increases to 98% within the same subgenus. This could suggest the presence of specific epitopes on Ag5 molecule being the variations on the superficial loops. The features of the P. gallicus allergens could explain the partial cross-reactivity found between the American and European Polistes venoms, and suggest that the use of European Polistes venoms would improve the diagnostic specificity and the therapy of European patients and of North American patients sensitized by European Polistes.  相似文献

1. The action of crude venoms of four aculeate species: Apis mellifera, Vespa crabro, Vespula germanica and Vespula vulgaris on human erythrocytes was investigated in order to determine the lytic and phospholipase activity of different aculeate venoms and their ability to induce red blood cell hemolysis. 2. Bee venom was the only extract to completely lyse red blood cells at the concentration of 2-3 micrograms/ml. 3. Phospholipase activity in all of the examined vespid venoms was similar and the highest value was recorded in V. germanica. 4. Vespid venoms exhibited phospholipase B activity, which is lacking in honeybee venom. 5. In all membrane phospholipids but lecithin, lysophospholipase activity of vespid venoms was 2-6 times lower than the relevant phospholipase activity. 6. The incubation of red blood cells with purified bee venom phospholipase A2 was not accompanied by lysis and, when supplemented with purified melittin, the increase of red blood cell lysis was approximately 30%.  相似文献

Hornets possess highly toxic venoms, which are rich in toxins, enzymes and biologically active peptides. Many bioactive substances have been identified from wasp venoms. Vespa mastoparan (MP-VBs) and Vespa chemotatic peptide presenting antimicrobial action (VESP-VBs) were purified and characterized from the venom of the wasp, Vespa bicolor Fabricius. The precursors encoding VESP-VBs and MP-VBs were cloned from the cDNA library of the venomous glands. Analyzed by FAB-MS, the amino acid sequence and molecular mass for VESP-VB1 were FMPIIGRLMSGSL and 1420.6, for MP-VB1 were INMKASAAVAKKLL and 1456.5, respectively. The primary structures of these peptides are homologous to those of chemotactic peptides and mastoparans isolated from other vespid venoms. These peptides showed strong antimicrobial activities against bacteria and fungi and induced mast cell degranulation, but displayed almost no hemolytic activity towards human blood red cells.  相似文献

Background

Double sensitization (DS) to bee and vespid venom is frequently observed in the diagnosis of hymenoptera venom allergy, but clinically relevant DS is rare. Therefore it is sophisticated to choose the relevant venom for specific immunotherapy and overtreatment with both venoms may occur. We aimed to compare currently available routine diagnostic tests as well as experimental tests to identify the most accurate diagnostic tool.

Methods

117 patients with a history of a bee or vespid allergy were included in the study. Initially, IgE determination by the ImmunoCAP, by the Immulite, and by the ADVIA Centaur, as well as the intradermal test (IDT) and the basophil activation test (BAT) were performed. In 72 CAP double positive patients, individual IgE patterns were determined by western blot inhibition and component resolved diagnosis (CRD) with rApi m 1, nVes v 1, and nVes v 5.

Results

Among 117 patients, DS was observed in 63.7% by the Immulite, in 61.5% by the CAP, in 47.9% by the IDT, in 20.5% by the ADVIA, and in 17.1% by the BAT. In CAP double positive patients, western blot inhibition revealed CCD-based DS in 50.8%, and the CRD showed 41.7% of patients with true DS. Generally, agreement between the tests was only fair and inconsistent results were common.

Conclusion

BAT, CRD, and ADVIA showed a low rate of DS. However, the rate of DS is higher than expected by personal history, indicating that the matter of clinical relevance is still not solved even by novel tests. Furthermore, the lack of agreement between these tests makes it difficult to distinguish between bee and vespid venom allergy. At present, no routinely employed test can be regarded as gold standard to find the clinically relevant sensitization.  相似文献

Abstract 1. All larval instars of Epargyreus clarus , the silver-spotted skipper, construct and inhabit leaf shelters that are presumed to protect them from predator attack.
2. Shelters effectively protected the larvae against foraging Crematogaster opuntiae ants and naive Polistes spp. wasps in laboratory tests, but did not protect them from predators, largely vespid wasps, present in the field.
3. A range of factors, including type of predator, learning ability, and experience level, may determine the effectiveness of leaf shelters as protection from predators.  相似文献

Stings by insects from the Hymenoptera order are known to cause life-threatening allergic reactions and impair life quality. Despite the effectiveness of conventional vespid venom immunotherapy, more standardized and safer allergy vaccines are required and recombinant hypoallergenic variants are important clinical tools. Antigen 5 is a major allergen of vespid venoms and it was previously reported that Antigen 5 from Polybia scutellaris (Poly s 5) could be a hypoallergenic variant. In this work we assess the immunological behavior and allergenic activity of Poly s 5 in order to explore its suitability for specific immunotherapy. With this aim, recombinant Poly s 5 was expressed in Pichia pastoris and the presence of cross-reactive epitopes with Pol a 5, a known allergenic Antigen 5, was investigated both at the IgG and IgE levels, by ELISA assays and a basophil-mediator release assay respectively. A molecular model was also built to better understand the relationship between immunological and structural aspects. In mice, Poly s 5 induced IgG antibodies which cross-reacted with Pol a 5. However, Poly s 5 induced only minimal amounts of IgE and was a poor inducer of basophil-mediator release, even when the cells were sensitized with Pol a 5-specific IgE. Moreover, Poly s 5-specific serum showed a specific protective activity and was able to inhibit the Pol a 5-induced basophil degranulation. Structural analysis from the molecular model revealed that a few amino acid substitutions in the N-terminal region of Poly s 5 should lead to an alteration of the surface topography and electrostatic potential of the epitopes which could be responsible for its hypoallergenic behavior. These findings, taken as a whole, show that Poly s 5 is likely a naturally occurring hypoallergenic Antigen 5 variant.  相似文献

Pollination in the genus Pedilanthus is commonly effected by hummingbirds. Pollination by vespid wasps in Pedilanthus is documented for the first time based on observations of Pachodynerus nasidens and Eumenes americanus pollinating Pedilanthus diazlunanus. An hypothesis concerning the mechanism by which hummingbird pollination was replaced by insect pollination in Pedilanthus is advanced based on observations of insects on P. bracteatus.  相似文献

The narrow host range of insect herbivores is noted, and some of the reasons why specificity has evolved are reviewed. Works indicating the need for new approaches are pointed out including the possibility that generalist predators provide a suitable pressure. Experiments to test the hypothesis that generalists are more vulnerable than specialists to predators are described. They involved a vespid wasp and over thirty species pairs of caterpillars, matched for size and density. Overall, generalists were taken more readily than specialists: some but not all reasons could be detected. The results are discussed in ecological and evolutionary terms.  相似文献

A novel gene, RTVP-1, which shows significant sequence identity to the mammalian testis-specific proteins, a family of plant pathogenesis-related proteins and the vespid venom allergen, antigen-5, has been isolated from a cDNA library of the human glioblastoma brain tumor cell line, U-251 MG. The highest degree of sequence identity was with the human testis-specific protein, TPX1 (38.7% over 119 amino acids). Northern hybridization analysis revealed that in fetal tissue RTVP-1 RNA was detected only in the kidney, but its expression was ubiquitous in adult tissues including brain. Multiple mRNAs encoded by RTVP-1 were highly expressed in a panel of cell lines from nervous system tumors arising from glia, although expression was low or absent in non-glial-derived nervous system tumour cell lines. Summary The potential role of generalist natural enemies is presented as one of the important ecological pressures that select for narrow host range in phytophagous insects, and dominant relative to physiological bases for specialization. Experiments are described in three completely different systems indicating that generalist herbivores are more vulnerable to predation than specialist herbivores. The three predators were (a) the vespid waspMischocyttarus flavitarsus, (b) the Argentine antIridomyrmex humilis and (c) the coccinellid beetleHippodamia convergens. It is concluded the predators may provide strong selection pressure for maintenance and perhaps evolution of narrow host range in insect herbivores.  相似文献
