The polytene chromosomes of 512 larvae of the Simulium nobile species group collected from 16 stream sites in northern, central and southern Thailand were examined. Band-by- band comparisons relative to the established standard chromosome map for the subgenus Simulium distinguished the 2 species of this group, S. nobile and S. nodosum. The differences are mainly due to the position of the nucleolar organizer, pseudochromocenter, and fixed and polymorphic inversions. S. nobile is a monomorphic species, while S. nodosum is a polymorphic species. The morphology of these taxa is described based on the gill filaments and the color of the antennae and female genitalia. S. nobile is distributed in southern Thailand at low altitudes, whereas the distribution of S. nodosum is in the north at higher altitudes.  相似文献

Simulium cauchense Floch & Abonnenc and Simulium quadrifidum Lutz are widely distributed in the Amazon region and are morphologically similar at the larval and pupal stages. Chromosomally, these species are readily distinguished by the position of the nucleolar organizer, which is in the short arm of chromosome I in S. cauchense and in the long arm of chromosomes III in S. quadrifidum. They also differ by three fixed inversions. Sex chromosomes are undifferentiated in both species. Chromosomal resolution of the two species allowed us to evaluate four structural features previously used as diagnostic aids at the larval stage. Characters that distinguish larvae of the two species are the number of branches and branching patterns of the dorsal abdominal setae and the dark band on each primary fan. Branching patterns of the gill histoblasts were often diagnostic, with S. quadrifidum exhibiting more proximal branching and S. cauchense more distal branching. Sites where both species occurred sometimes had larvae with one petiole branching proximally and the other distally; in these cases examination of the chromosomes permitted assignment of the specimen to species. Pigmentation patterns of larvae, on the other hand, are highly variable. Color typically is sex linked in both species.  相似文献

Polytene chromosomes of four members of the Simulium perflavum species group in Brazil are described, and a standard map for the species group is presented. Simulium rorotaense Floch & Abonnenc, S. perflavum Roubaud and an undescribed species (S.‘X’) are chromosomally conservative, each representing a single species with a unique preimaginal habitat. Chromosomal, morphological and ecological evidence indicates that S. maroniense Floch & Abonnenc, previously considered synonymous with S. rorotaense, is a good species. Independent morphological and chromosomal analyses yielded 99.4% agreement in separating larvae of S. rorotaense and S. maroniense. The two species can be distinguished by gill morphology or by a subterminal inversion on the long arm of chromosome III. Simulium maroniense consists of at least four cytotypes, each with different sex chromosomes and autosomal polymorphism profiles and associated with a particular landscape type, altitude, temperature regime or geographical location. Simulium rorotaense and S. maroniense share one unique inversion, as do S. perflavum and S. ‘X’, indicating two pairs of sister species. The anthropogenic S. perflavum probably dispersed into Central Amazonia sometime after the mid-1970s.  相似文献

It is noted that the chromosomal inversion 2L-7, which has been used in the past to separate S. Sanetipauli from S. soubrense, occurs as an intraspecific polymorphism and hence cannot be considered diagnostic, although in some populations 2L-7 can still strogly indicate the presence of two species. However, two newly recognised inversions, 1L-A and 2L-A, can be used in combination to identify S. sanctipauli, S. soubrense and a new species S. soubrense B. The absence of the relevant heterozygotes for these two new inversion confirms the separate specific status of S. sanctipauli from S. soubrense from S. soubrense B as well as providing a reliable means of larval identification. The misuse of 2L-7 as a species diagnostic inversion has undoubtedly led to past misidentifications of S. sanctipauli and S. soubrense, and it is possible, for example, that only S. sanctipauli is resistant to organophosphate insecticides in Ivory Coast and not S. soubrense. Beffa form appears to be a distinctive geographic race of S. soubrense, but forme konkouré remains as yet unassigned. A cytotaxonomic key for the identification of members of the S. sanctipauli sub-complex is presented.  相似文献

Chromosomal analysis of more than 500 larval black flies from 19 sites in Armenia and Turkey revealed five taxa in the Prosimulium hirtipes group: Prosimulium frontatum Terteryan, Prosimulium rachiliense Djafarov cytoform ‘A,’ P. rachiliense Djafarov cytoform ‘B,’ Prosimulium tomosvaryi (Enderlein), and a new species of Prosimulium. To associate the names rufipes (Meigen) and tomosvaryi with cytological entities, larvae from the respective type localities in Germany were characterized chromosomally. Prosimulium frontatum is restricted to the Caucasus Mountains where studied populations have five unique inversions. It is most closely related to cytoforms ‘A’ and ‘B’ of P. rachiliense, the three taxa sharing one unique inversion. The two cytoforms of P. rachiliense are separated by about 1200 km, obscuring decisions about their reproductive isolation. The names rachiliense and possibly pronevitschae Rubtsov apply to cytoform ‘A’ in Armenia, whereas the name fulvipes (Edwards) might apply to cytoform ‘B’ in Turkey and to material formerly identified in Turkey as P. rufipes. Populations of P. tomosvaryi in Armenia are chromosomally distinct from previously studied populations in Europe and Morocco, although tied to European and Turkish populations by a shared X‐linked inversion. We conservatively view Armenian, European, and Turkish populations of P. tomosvaryi as a single, polymorphic species. A new species, chromosomally related to P. hirtipes (Fries) and P. tomosvaryi by two uniquely shared inversions, was discovered in Turkey. © 2014 The Linnean Society of London  相似文献

A cladistic analysis of Simulium ( Trichodagmia ) sensu Crosskey and Howard, using 34 morphological characters of larvae (6 characters), pupa (5) and adults (23), yields nine most parsimonious trees under equal weights (length 101 steps CI 0.49 RI 0.73). Successive weighting based on the maximal rescaled consistency index preferred one of the nine (31.37 steps CI 0.62 RI 0.87 total fitcon3= 235.8), which was also one of two trees found under implicit weights with concavity values of 3–6. The cladogram justifies the recognition of two subgenera. Simulium ( Trichodagmia ) sensu stricto (containing S. muiscorum, sumapazense, S. wygodzinskyorum, S. nigrimanum, S. chalcocoma, S. huairayacu and S. lahillei ) is supported by the branchial tip sclerotization and the presence of cibarial teeth, larval body tegument covered with lanceolate hairs, female with simple claw, and gonapophysis size. Simulium ( Thyrsopelma ) (containing S. scutistriatum, S. hirtipupa, S. orbitale, S. guianense, S. perplexum and S. itaunense ) is supported by the hypostomial teeth.  相似文献

Simulium craigi Adler and Currie is a polymorphic species based on polytene chromosome banding patterns in the long arm of chromosome III (IIIL). Three cytotypes are described based on the predominant IIIL sequences. These correspond to three broad geographic areas: cytotype CC from Pennsylvania; cytotype AF from Ontario and Manitoba; and cytotype ACF/BCF from New Hampshire. In the absence of sympatric populations, these cytotype differences are best explained by clinal variation within a single species. The relationship of S. craigi to other described members of the S. vernum group is discussed.  相似文献

Simulium yahense black flies infected with microfilaria of Onchocerca volvulus were kept in a defined insectary environment in Liberia, West Africa. A daily sample of infected flies was dissected for larvae developing in the thoracic muscles and examined for growth in stadial development. Microfilariae ingested by black flies transformed to the L1 larval stage without molting. Successive larval development included molting to the L2 stage and, finally, to the L3 stage, which was infective in humans. The cephalic cap, consisting of a laterally located hook and central stoma, occurs in the first larval stage. The caudal appendix and the laterally located anal opening are apparent in the L1 larva. In the L2 stage, the cephalic cap is lost and the large circular stoma becomes surrounded with elevated flaps. The caudal appendix was lost after larvae molted to the L3 stage, and in its place, 3 terminal papillae developed. Sense organs, such as 2 opposing phasmids and 8 papillae that were arranged into 2 circles, developed in the cephalic region of the L3 larva. The evidence of pathological consequences due to the presence of the L3 larva in the fly host are illustrated and discussed.  相似文献

Recently, knowledge of Neotropical Simuliidae has been accumulating quickly. However, information about supra-specific relationships is scarce and diagnoses of Simulium subgenera are unsatisfactory. To investigate the relationships among Simulium (Chirostilbia) species and test the subgenus monophyly, we performed a cladistic analysis. The ingroup included all species of this subgenus and the outgroup included representatives of the 17 species groups of Neotropical Simulium and three Holarctic species. The study was based on a data matrix with 31 terminal taxa and 45 morphological characteristics of adult, pupa and larva. The phylogenetic analysis under equal weights resulted in eight most-parsimonious trees (length = 178, consistency index = 34, retention index = 67). The monophyly of the S. (Chirostilbia) was not supported in our analysis. The Simulium subpallidum species group was closer to Simulium (Psilopelmia) and Simulium (Ectemnaspis) than to the Simulium pertinax species group. Additionally, we describe the three-dimensional shape of the terminalia of male and female of Simulium (Chirostilbia) for the first time and provide comments about the taxonomic problems involving some species of the subgenus: Simulium acarayense, Simulium papaveroi, S. pertinax, Simulium serranum, Simulium striginotum and S. subpallidum.  相似文献

A cypovirus from the black fly Simulium ubiquitum (SuCPV) was isolated and examined using biological and molecular techniques. SuCPV produces small (typically 0.25mum), polyhedral shaped inclusion bodies (polyhedra), in which the virus particles become multiply embedded. SuCPV is the third cypovirus isolated from Diptera, but the first from Simuliidae that has been characterized using molecular analyses. SuCPV has a genome composed of 10 segments of dsRNA, with an electrophoretic migration pattern that is different from those of recent UsCPV-17 and CrCPV-17 isolates from the mosquitoes Uranotaenia sapphirina and Culex restuans, respectively. The SuCPV electropherotype appears to show significant differences from those of the previously characterized lepidopteran cypoviruses. Sequence analysis of SuCPV segment 10 shows that it is unrelated to either of the two CPV isolates from Diptera or to the CPV species for which Seg-10 has been previously characterized from Lepidoptera. A comparison of the terminal regions of SuCPV genome segments to those of CPV-1, 2, 4, 5 14, 15, 16, 17, 18, and 19 also revealed only low levels of conservation. We therefore, propose that SuCPV is classified within a new Cypovirus species, which we have tentatively identified as Cypovirus-20. We have therefore referred to this virus isolate as S. ubiquitum CPV-20 (SuCPV-20).  相似文献

Abstract.  Like many other organisms, Simuliidae often show a high degree of endemism on oceanic islands, and the Gulf of Guinea, including its islands, is a region of general biogeographical interest in view of the uniqueness of its flora and fauna. This paper presents a comprehensive review of the Simuliidae in the region, and is the first faunistic record of blackflies from Bioko. Simuliidae (larvae, pupae and neonate adults) were collected from Bioko and from around Mount Cameroon and compared with previous findings from Mount Cameroon, Príncipe and São Tomé. Twenty-seven species are known from the region. Twenty-five species have been recorded around Mount Cameroon, eight species on Bioko, three species on São Tomé and one species on Príncipe. Two taxonomic 'forms' (one on Bioko alone, and one on both Bioko and around Mount Cameroon) and one species (found only on São Tomé) are endemic to the region. A study of the variation in the morphology of Simulium cervicornutum revealed two morphotypes, one from Bioko and around Mount Cameroon and the other from the rest of Africa. The speciation and biogeography of the Simuliidae in the Gulf of Guinea are discussed.  相似文献

Double labelling of Simulium ornatipes polytene chromosomes with H3- and C14-thymidine shows that chromosome synthesis follows three distinct phases viz. a short phase of initiation in puffs and interbands spreading to more condensed regions; a long continuous labelling phase, then a discontinuously labelled end phase as bands complete their replication in temporal sequence. Analysis of H3 labelling patterns indicates that while heterochromatic bands replicate there is no clear correlation between heterochromatic or C-banding regions and band replication time. The major characteristic governing band replication time appears to be band size and density. However, in some bands this relationship is modified, perhaps it is suggested, by DNA organisation influencing the efficiency of replicons. The existence of great variability in homologous band replication times, even within a chromosome pair, indicates that the control of band replication is highly autonomous. It is suggested that polymorphisms at the molecular level determine this variation. Replication time of active nucleolar organisers is very long in contrast to the short replication of condensed inactive organisers. This may reflect differential polytenisation of ribosomal DNA as a result of a developmental polymorphism, or the amplification of ribosomal DNA by active nucleolar organisers.  相似文献

The polytene chromosomes of 3347 larvae of the Simulium tuberosum group in Asia were analysed, representing the largest ever cytogenetic study of black flies in the Oriental Region. Band‐by‐band comparisons, relative to the established standard chromosome map for the subgenus Simulium, revealed 17 cytogenetically distinct taxa in Thailand, plus an 18th in China. Six of these taxa correspond to morphologically described species (S. doipuiense, S. rufibasis, S. setsukoae, S. tani, S. yuphae and S. weji). Recognition of the 18 taxa is based largely on unique inversions, either fixed or sex linked, primarily in the long arm of chromosome III. The greatest cytological diversity was discovered in the S. tani lineage, with ten cytoforms. This marked chromosomal diversification within S. tani is based largely on two inversions that have assumed different roles over evolutionary time, variously functioning in different combinations as fixed inversions, sex‐linked inversions and autosomal polymorphisms. Shared unique chromosomal features, relative to the subgeneric standard chromosome map, allowed evolutionary relationships among the cytotaxa to be inferred. Simulium (Simulium) liubaense, sp. nov. is described based on the female specimens collected from Liuba, Shaanxi Province, China. This species is assigned to the variegotum-group of the subgenus Simulium, and is closely related to S. (S.) jingfui Cai and An, 2008, S. (S.) taiwanicum Takaoka, 1979, S. (S.) hackeri Edwards 1928, S. (S.) chamlongi Takaoka and Suzuki, 1984. However it is clearly differentiated from them by the structure of cibarium, genital fork, genital plate, paraproct and cercus of the female.
