We have analysed the proteinase profiles of 11 species from 7 different genera of trypanosomatids by in situ detection of enzyme activities on SDS-PAGE gels containing co-polymerized gelatin as substrate, and the use of specific proteinase inhibitors. Our survey indicates that while cysteine- and metalloproteinases are distributed ubiquitously among trypanosomatids, there are marked differences between the enzyme profiles from the monogenetic (Crithidia, Herpetomonas, Leptomonas) and digenetic (Trypanosoma, Endotrypanum, Phytomonas, Leishmania) species. The detected metalloproteinase activities, ranging in size from 50–100 kDa, partitioned into the detergent-phase after Triton X-114 extraction, while most of cysteine proteinases, of three distinct molecular mass ranges (30–50 kDa, 80–100 kDa and 116–205 kDa), partitioned into the aqueous phase. Thus, within this group of organisms, the metalloproteinase activities seem to be predominantly membrane-associated proteins. We also show that the plant parasites of the genus Phytomonas exhibit a distinctive cysteine proteinase profile that might be exploited further as a criterion for taxonomy of the genus.  相似文献

SYNOPSIS. Five strains of trypanosomatids, Herpetomonas megaseliae, H. samuelpessoai, H. muscarum muscarum, H. muscarum ingenoplastis and a newly isolated Herpetomonas sp., were examined for the enzymes of arginine-ornithine metabolism. Ornithine carbamoyltransferase (E.C. 2.1.3.3) and argininosuccinate lyase (E.C. 4.3.2.1) were detected in cell extracts of H. megaseliae, H. samuelpessoai and H. muscarum muscarum but not of others. Both enzymes seemed repressible by arginine, which could account for their apparent absence in H. muscarum ingenoplastis and Herpetomonas sp., which grow in a complex, arginine-rich medium. Additionally, arginine deiminase (E.C. 3.5.3.6) and citrulline hydrolase were detected in cell extracts of the 5 strains examined. This latter enzyme, previously described only in Tetrahymena, effects the single-step hydrolysis of citrulline into ammonia, ornithine and CO2. Arginase (E.C. 3.5.3.1) and urease (E.C. 3.5.1.5) were not found in any of the strains examined. Some of the physicochemical characteristics of the enzymes encountered are described.  相似文献

To obtain additional information on the phylogenetic relationships within the family Trypanosomatidae (order Kinetoplastida), we have sequenced the small subunit ribosomal RNA genes from the endosymbiont containing species Herpetomonas roitmani TCC080, Herpetomonas sp. TCC263, Crithidia oncopelti ATCC 12982 and a partial large subunit rRNA gene from H. roitmani. The small subunit sequences in the two isolates of Herpetomonas are very similar but not identical, and so are their restriction digest profiles of kinetoplast DNA. The size of minicircles in both isolates is 4.2 kilobases. The inferred ribosomal RNA phylogenetic trees shows the genera Herpetomonas and Crithidia as polyphyletic. Endosymbiont-bearing herpetomonads cluster with the endosymbiont-bearing crithidias and a blastocrithidia to form a monophyletic clade, whereas the endosymbiont-free members of these genera are found elsewhere in the tree. These data support the hypothesis of a monophyletic origin of endosymbiosis in trypanosomatid evolution and also suggest that a taxonomic revision is needed in order to better describe the natural affinities in this family.  相似文献

In this study, we surveyed six species of cockroaches, two synanthropic (i.e. ecologically associated with humans) and four wild, for intestinal trypanosomatid infections. Only the wild cockroach species were found to be infected, with flagellates of the genus Herpetomonas. Two distinct genotypes were documented, one of which was described as a new species, Herpetomonas tarakana sp. n. We also propose a revision of the genus Herpetomonas and creation of a new subfamily, Phytomonadinae, to include Herpetomonas, Phytomonas, and a newly described genus Lafontella n. gen. (type species Lafontella mariadeanei comb. n.), which can be distinguished from others by morphological and molecular traits.  相似文献

ABSTRACT. Attention, perhaps overdue, is drawn to the extent and significance of endosymbionts (xenosomes sensu lato) in the cytoplasm and nuclei of many protozoa from diverse taxonomic groups. Even more importantly, recent advances in the study of such intimate associations are reviewed and discussed and their impact on broader problems of cell biology and evolution are stressed. Workers inside and especially outside the fields of protozoology and parasitology have often neglected such data, failing to appreciate their relevance to significant problems in their own fields of investigation. The major topics covered by speakers in the Symposium (to which this paper serves only as an introduction) include the following, in order of their presentation: terminology for the symbiont-host relationship and a brief overview of the field; the evolutionary problem of the origin of contemporary associations, including cell organelles such as mitochondria and plastids; the adaptive value of endosymbionts to their protozoan hosts; mechanisms of establishment, maintenance, and integration of such foreign bodies/invaders in their unicellular eukaryotic host cells; and the extent of algal and bacterial endosymbioses in diverse protozoan groups. In all papers, the principal relatively well studied complexes used as examples are the following: various kinds of algae in the larger foraminifera and in ciliates, radiolarians, and acantharians; the several types of bacteria in the cytoplasm of Amoeba and of Pelomyxa; the endonuclear bacterial symbionts of Paramecium; the cytoplasmic prokaryotes in Paramecium and in Parauronema; and the methanogenic bacteria of certain ciliates.  相似文献

Laboratory colonization of hematophagous insects must include an efficient method of blood feeding, preferably by artificial means. Strict rules for obtaining animal use permits, extensive animal maintenance costs, and indirect anesthesia effects on animal health warrant the development of an artificial membrane feeding technique for sand fly colonization in laboratories. An attempt was made to colonize Lutzomyia shannoni using an artificial blood feeding membrane to replace the use of live animals commonly used for sand fly blood‐feeding purposes. Lutzomyia shannoni readily fed through a pig intestine membrane exposed at an angle of 45°. However, it did not feed through a chicken skin membrane. Olfactory attractants were unable to improve blood‐feeding efficiency. Plaster of Paris was the most suitable oviposition substrate. Female L. shannoni adults laid no eggs on moist sand substrate. Sand fly adults held in groups of ten or more laid higher numbers of eggs than did individually maintained sand flies. Inclusion of the L. longipalpis oviposition hormone dodecanoic acid or the presence of previously laid eggs did not stimulate L. shannoni oviposition. The average L. shannoni egg, larval, and pupal duration were 9.3, 36.7, and 17.8 days, respectively. The addition of a 20% sugar solution improved adult female longevity. Females survived longer (14.8 days) than males (11.9 days). Lutzomyia shannoni was successfully colonized in the laboratory for up to four generations using this artificial membrane technique.  相似文献

ABSTRACT. Leishmania parasites, which afflict 12 million people in 88 countries, exist as promastigotes transmitted by insect vectors and as amastigotes residing in mammalian macrophages. Promastigote cells arranged in rosettes have also been described but universally disregarded as a distinct stage in the life cycle. We present evidence that only rosettes of Leishmania major promastigotes express cell surface poly‐α2,8 N‐acetyl neuraminic acid (PSA) and PSA containing de‐N‐acetyl neuraminic acid (NeuPSA). Expression of rosette‐specific PSA antigens was mosaic, with individual promastigotes expressing PSA, NeuPSA or both. A 50 kDa protein was detected by Western blot analysis of a detergent‐insoluble cell fraction with both PSA and NeuPSA‐reactive antibodies. Frequencies of rosette formation as well as cell surface PSA/NeuPSA expression were temperature dependent. Rosettes also engaged in an unusual swarming behavior, congregating into extended clusters. Distinct structures resembling cellular fusion bodies were formed in and released from rosettes. The results indicate that rosettes are an unrecognized stage in the life cycle of Leishmania. We hypothesize that rosettes initiate mating in Leishmania during which PSA/NeuPSA expression plays an important role. Recognizing rosettes as a distinct form of the Leishmania life cycle opens new possibilities for treatment or prevention of disease and, possibly, in vitro genetic recombination without passage of cells through insect vectors.  相似文献
