Binding properties of submaxillary gland muscarinic receptors and agonist-induced saliva secretion were studied in rats subjected to heat acclimation. The maximal binding capacity for the muscarinic antagonist N-[3H]methyl-4-piperidyl benzilate was increased from control value of 0.21 to 0.40 pmol/mg protein within 1-2 days of heat acclimation. The increase in the number of muscarinic receptors per gland (100%) was by far higher than the increase in tissue weight (20%), indicating higher density of receptors in the acinar cells of the treated rats. High levels of receptors coincided with the appearance of high-affinity binding sites for muscarinic agonists (oxotremorine, pilocarpine and carbamylcholine), and with reduced tissue sensitivity to pilocarpine. After 4-8 weeks of heat acclimation, the number of receptors as well as tissue response to pilocarpine returned to control levels. These results suggest a functional correlation between the transient upregulation muscarinic receptors in the submaxillary gland and the physiological activity in salivary secretion, and indicate that the high-affinity muscarinic receptors may attenuate saliva secretion during the initial phase of heat acclimation.  相似文献

The response of eccrine sweat glands of the forearms of four females was investigated during acclimation to heat. The sweat rate increased progressively during ten exposures. There was no significant change in the number of active sweat glands. The increased sweat rate could, therefore, be attributed to enhanced secretory activity of individual glands.
Zusammenfassung Die Schweissdrüsenreaktion des Vorderarms von 4 Frauen wurde während der Akklimation an Hitze untersucht. Die Schweisssekretion stieg progressiv während 10 Sitzungen an. Eine signifikante Änderung in der Zahl der aktiven Schweissdrüsen wurde nicht beobachtet. Die gesteigerte Schweisssekretion beruht danach auf einer vermehrten Schweissbildung der einzelnen Drüsen.

Resume On a étudié les réactions des glandes sudoripares de quatre femmes pendant la période d'acclimation à la chaleur. La sécrétion de sueur a augmenté progressivement durant 10 heures. On n'a cependant pas observé de modification sensible du nombre des glandes actives. L'augmentation de la sueur sécrétée provient donc d'une augmentation de la sécrétion de chacune des glandes en particulier.

An important target tissue for murine cytomegalovirus (CMV) infection is the submaxillary salivary gland. Submaxillary salivary gland biopsy specimens from BALB/c mice latently infected with murine CMV were examined for murine CMV DNA by in vitro enzymatic amplification using the polymerase chain reaction preceding oligonucleotide hybridization. The amplified sequence was a 152-base pair segment from within the immediate early gene of murine CMV. Biopsy and whole gland specimens from acutely infected BALB/c mice and latently infected, immunosuppressed BALB/c mice were compared for active murine CMV infection. After acute infection with murine CMV, virus was recovered in all cultures of both biopsy and whole salivary gland specimens but from none of the latently infected animals. Reactivated virus was detected by culture of both biopsy (90%) and whole salivary gland specimens (100%) from latently infected mice that received antithymocyte serum. Viral nucleic acid was detected in 90% of biopsy specimens from latently infected animals. Hence, active murine CMV infection can be detected in biopsy specimens from mice with acute and reactivated infection and murine CMV DNA can be amplified and detected in salivary gland biopsy specimens from latently infected animals. Biopsy of this or other target tissues can be useful for obtaining tissue for viral studies where the survival of the animal is important and it is useful to distinguish latent from acute or reactivated infection.  相似文献

Hypersalivation is an important mechanism for heat dissipation by animals without sweat glands. The water content and conductivity (at 20 kHz) in sub-maxillary salivary gland (SSG) and in other tissues were investigated in adult male rats exposed to microwaves (2880 MHz, 1.5 μs pulses at 1000 Hz) or to conventional heat at 40 °C. Eighty rats in one series were exposed, one at a time, for 30 min to microwaves producing a specific absorption rate (SAR) of 4.2,6.3,6.8,8.4,10.8 or 12.6 W/kg. Fifty rats were sham-exposed under similar environmental conditions. In the second series, ten rats were sham-exposed, 33 rats were exposed, one at time, for 15, 30 or 60 min to microwaves at a SAR of 9.5 W/kg, and 32 rats were exposed for similar periods to conventional heat at 40 °C. In rats of the first series colonic temperatures were elevated significantly at a SAR of 4.2 W/kg, while SSG water content and conductivity increased significantly at SAR values of 6.3 W/kg and higher. In the second series of experiments increases in colonic temperature and SSG water content were greater after 15 and 30 min of microwave exposure than after exposure to heat. Also, SSG conductivity was significantly depressed by heat and significantly increased by microwaves after exposure for 15 or 30 min. The results support the hypothesis that water content and conductivity of SSG of rats can be used as a sensitive specific test of a microwave induced thermal response.  相似文献

1. Adaptation of salivary cooling mechanism during acclimation to heat (34 degrees C) and its role in thermoregulation of the rats was studied on conscious rats with either one submaxillary gland chronically cannulated or both submaxillaries ligated. 2. During heat stress (40 degrees C) acclimated rats showed a decrease both in rectal temperature threshold for salivation (Tre-TS), in salivary flow rate and in Tre (hyperthermic plateau). Animals survived for extended periods and rats with ligated glands survived 40% less than non-ligated rats. 3. For both cannulated and ligated rats short term acclimation (5 days) was the most effective. 4. It is suggested that earlier activation of salivation mechanism is associated with the decreased hyperthermic plateau and that the decreased salivary flow rate allows better control of water balance of the animals. Consequently, survival period during heat stress is extended.  相似文献

Submaxillary salivary gland responsiveness during the heat acclimation procedure (34 +/- 1 degree C) was studied in the rat. Gland responsiveness was evaluated by measuring saliva flow rate of the anesthetized animals following either parasympathetic nerve stimulation or i.v. application of pilocarpine. A thirty percent decrease in glandular responsiveness for the two modes of stimulation was measured during the first 10 days of acclimation. Following 60 days of acclimation recovery was observed. It is concluded that decreased responsiveness of the heat-acclimated gland is a glandular event. Increased saliva flow occurring at the initial phase of acclimation is apparently due to changes in the thermoregulatory center.  相似文献

A significant amount of pyroglutamate aminopeptidase (PGAP) activity was found to be present in 27,000 x g supernatant of rat submaxillary gland, maximum activity being at pH 6.5. EDTA stimulated the enzyme activity by 95% at pH 8.0 while at pH 6.5 it did not have any significant effect. On comparison of its properties submaxillary PGAP appears to be different from brain, pituitary and other reported PGAPs. Submaxillary PGAP could also catalyze efficiently the formation of cyclo (His-Pro) from TRH. Cyclo (His-Pro) formation by submaxillary enzyme was more pronounced than that by liver PGAP.  相似文献

Effect of beta-adrenoceptor blockade on the changes in rectal temperature (RT), heat production (HP) and noradrenaline (NA) concentration in the submaxillary salivary glands (SMSG) of rats exposed to heat (45 degrees C) was studied. Propranolol (P) (15 mg/kg i.p.) decreased the tolerance to heat. The survival time of propranolol-treated (PT) rats was 30 min shorter. The temperature curve of control rats exposed to heat can be divided into three phases: a rapid rise in RT; a plateau (TP) and a prelethal increase. In PT animals, under identical conditions, TP disappears and RT further rises accelerating death. In the initial phase of heat exposure, HP was markedly decreased to the same extent in both experimental groups; but after 20 min HP increases in PT rats. The content of NA from SMSG both in the initial phase and in the TP phase is modified in PT rats.  相似文献

1. The enzymic activity which catalyzes transfer of sulfate ester group from 3'-phosphoadenosine-5'-phosphosulfate to mucus glycoprotein was found associated with Golgi-rich membrane fraction of rat submandibular salivary gland. 2. Optimum enzyme activity was obtained with 0.5% Triton X-100, 4 mM MgCl2 and 25 mM NaF at a pH of 6.8 using desulfated submandibular salivary mucus glycoprotein. The apparent Km of the enzyme for mucus glycoprotein was 11.1 mg/ml. 3. Alkaline borohydride reductive cleavage of the synthesized 35S-labeled glycoprotein led to the liberation of the label into reduced oligosaccharides. A 75.4% of the label was found incorporated in four oligosaccharides. These were identified in order of abundance as sulfated penta-, tri-, hepta- and nonsaccharides. 4. Based on the results of chemical and enzymatic analyses of the intact and desulfated compounds the pentasaccharide was characterized as SO3H----GlcNAc beta----Gal beta----GlcNAc(NeuAc alpha----)GalNAc-ol and the trisaccharide as SO3H----GlcNAc beta----Gal beta----GalNAc-ol.  相似文献
