Summary Spectral sensitivity curves for four sustaining neurons in the optic tracts of Procambarus clarkii were determined under dark-adapted and chromatic light-adapted conditions. The max in the dark-adapted state is at 570 to 575 nm, and shifts to longer wavelengths in the violet-light-adapted state (Fig. 4). Red-light-adaptation suppresses the sensitivity of the yellow-green receptors of the eye and alters the discharge pattern of the sustaining neurons, thereby exposing an input with a max at 445 nm from the blue-sensitive receptors (Fig. 5). The specificity of perch (Perca fluviatilis) roe fucolectin was studied using the protein dot blot technique, followed by detection with colloidal gold–labeled neoglycoproteins bearing human milk oligosaccharides. The strongest binding was noted with the H type 1 pentasaccharide lacto-N-fucopentaose (Fuc1-2Gal1-3GlcNAc1-3Gal1-4Glc); the interaction with the H type 6 trisaccharide 2"-fucosyllactose (Fuc1-2Gal1-4Glc) was weaker. Binding of the perch lectin to the Lewis antigens (associated with tumors and embryonic tissues) was also studied. It was found that the lectin weakly interacted with the hexasaccharide lacto-N-difucohexaose I, Leb (Fuc1-2Gal1-3[Fuc1-4]GlcNAc1-3Gal1-4Glc), but not with Lea, Lec, or Lex antigens. Thus, the perch roe lectin exhibited pronounced differences in carbohydrate specificity from other fucolectins—a feature that may be used in structural studies and isolation of fucose-containing glycoconjugates.  相似文献

Summary Ascorbate has been related to the differentiation of several mesenchymal cells including haematopoietic cells. We have previously demonstrated that ascorbate enhances the activity of 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) on monocytic differentiation of HL-60 cells. Here, we show that ascorbate-mediated modification of cellular redox state and AP-1 (activating protein-1) DNA binding during early phases are related to the enhancing effect of ascorbate on differentiation. Ascorbate, but not its fully oxidized form, dehydroascorbate, or an ascorbate analogue with a low rate of oxidation, ascorbate-2-phosphate, enhanced the differentiation induced by 1,25(OH)2D3, modified cytosolic reactive oxygen species levels and mitochondrial redox potential (m), and modulated AP-1 DNA binding in HL-60 cells. Ascorbate itself increased AP-1 binding to DNA in noninduced cells, whereas it inhibited AP-1 binding in 1,25(OH)2D3-induced cells. However, ascorbate increased the mRNA levels ofc-jun, junB. andc-fos in 1,25(OH)2D3-induced cells. The genes encoding three invariant components of the human T-cell antigen receptor, the CD3 , , and chains, are located on human chromosome 11 at band q23. We isolated cosmid clones containing the human CD3 and chain genes in vectors designed for rapid and efficient chromosome walking. The human CD3 gene was located in the region immediately downstream of the CD3 and genes using synthetic oligonucleotide probes and the localization of this gene confirmed by DNA sequencing. Detailed restriction mapping of the CD3 locus demonstrated that all three CD3 subunits are encoded within 60 kb of DNA with the CD3 gene located 26 kb downstream of the CD3 and genes. Analysis of genomic DNA on pulsed field gels using probes isolated from these cosmid clones defined a physical map of 750 kb spanning the CD3 locus on human chromosome 11g23. The CD3 genes thus comprise a multigene family encoding cell surface components important for transmembrane signaling on T lymphocytes. The arrangement of these genes suggest that they may share common regulatory elements for the control of gene expression during T-cell ontogeny.  相似文献

The study addressed to resolve the mechanism involved in cholesteroldependent regulation of giardia encystation process, revealed that (a) the trophozoites have the ability to express genes coding for receptorCk and sterol element binding protein (SREBP); (b) inhibition of cholesterol dependent activation of receptorCk results in the upregulation of CWP1 gene expression leading to encystation process. In the aquatic liverwort Riccia fluitans, membrane depolarization (m), change in membrane conductance (gm), and current-voltage (I-V) characteristics in the presence of different amino acids as well as the uptake of 14C-labeled amino acids were measured. L-isomers of the tested amino acids generate larger electrical effects (m, gm) than D-isomers, and the I-V characteristics show that the positive electrical inward-current of 20 mA m-2 generated by 0.5 mM D-serine is only about 50% of the current generated by adding 0.5 mM L-serine. Whereas - and -amino acids rapidly depolarize the membrane to the same extend, with -aminobutyric acid (-AB) and dipeptides no significant electrical effects have been measured. The uptake kinetics of 14C-labeled amino acids display three components: (I) A saturable high-affinity component with Ks-values of 48 M D-alanine, 12 M -aminoisobutyric acid (AIB), 9 M L-alanine, 8 M L-proline, and 6 M L-serine, respectively; (2) an apparently linear low-affinity component, and (3) an also linear but unspecific component at concentrations >20 times the given Ks-value. Uptake of 14C-labeled AIB can be inhibited competitively by all tested neutral amino acids, the L-isomers being more effective than the D-isomers, as well as by ammonium or methylamine. Vice versa, AIB competitively inhibits uptake of L-serine and L-alanine. It is concluded that an uncharged stereospecific carrier for the investigated amino acids exists in the plasmalemma of Riccia fluitans. Accumulation ratios of about 50 suggest secondary active transport driven by a transmembrane electro-chemical gradient (mainly m) which is generated by the electrogenic proton pump. The possibility of solving the mass balances to a multiplicity of substrates within a CSTR in the presence of a chemical reaction following Michaelis-Menten kinetics using the assumption that the discrete distribution of said substrates is well approximated by an equivalent continuous distribution on the molecular weight is explored. The applicability of such reasoning is tested with a convenient numerical example. In addition to providing the limiting behavior of the discrete formulation as the number of homologous substrates increases, the continuous formulation yields in general simpler functional forms for the final distribution of substrates than the discrete counterpart due to the recursive nature of the solution in the latter case.List of Symbols C{N. Summary The inheritance of the ability of barley anthers to produce microspore-derived callus in vitro was investigated. The genotypes selected were the two spring cultivars Dissa (D) and Sabarlis (S), the two F1 hybrids (DxS, SxD), the two backcross generations [Dx(DxS), Sx(DxS)], and an F2 generation derived from DxS. From a number of individuals of each generation, the first five spikes were harvested sequentially and after pre-treatment the anthers were removed and placed in culture. Cultures were scored for microspore callus production and plantlet differentiation. Although Dissa gave a significantly higher level of callus production than Sabarlis, the overall frequencies of green and albino plant production were higher from Sabarlis. There was no significant difference between reciprocal F1 hybrids. Analysis of variance revealed significant differences in response between the spikes sampled from the plants. This was the major source of variation in the experiment. Spike to spike variation also appeared to be a heritable character.  相似文献

As culture-bound syndromes, Japanese shinkeishitsu (constitutional neurasthenia) and taijinkyfush (anthropophobia) have received considerable attention in the Japanese literature. While these disorders are viewed as diagnostically distinct from Western psychiatric categories, recent studies by the Japanese suggest some affinity with Western social phobias, depression, and schizophrenia. The paper reviews this literature and offers suggestions for further cross-cultural research.This paper was accepted for publication prior to the compilation of Volume 13, No. 2, Neurasthenia in Asian Cultures. Consequently it does not refer to the papers in that volume, several of which are very relevant to the discussion in this paper.  相似文献

Phytotoxicity and inhibitory effects of the fusarial toxins fumonisin B1 (FB1) [m.p. 103–105 °C], fusaric acid [m.p. 106–107 °C], butenolide (4-acetamido-4-hydroxy-2-butenoic acid lactone) [116–117 °C], 9, 10-dihydroxyfusaric acid [m.p. 150–155 ° C], and moniliformin on chlorophyll synthesis in the aquatic macrophyte Lemna minor (duckweed) were examined. FB1 proved to be most active, reducing the growth of L. minor fronds and their ability to synthesize chlorophyll by 53% and 59%, respectively, at 0.7 g/ml. The growth rate of L. minor was reduced 59% by 6.7 g/ml fusaric acid, 62% by 66.7 g/ml butenolide, and 22% by 66.7 g/ml 9,10-dihydroxyfusaric acid. Moniliformin was the least phytotoxic to L. minor, with only a 16% suppression of growth rate and a 54% reduction in chlorophyll at 66.7 g/ml.The mention of firm names or trade products does not imply that they are endorsed or recommended by the US Department of Agriculture over other firms or similar products not mentioned.  相似文献

The effect of basal media (Woody Plant Medium (WPM), Quoirin and Lepoivre (QL) and Olive Medium (OM)) and of various concentrations of cytokinins (6-benzyladenine (BA), zeatin, 6-(,-dimethylallyl-amino) purine (2iP)), solely or in combinations with each other and with gibberellic acid (GA3), on in vitro shoot proliferation of the greek olive cultivar `Chondrolia Chalkidikis' was investigated. WPM proved to be the most effective one, resulting in better morphological appearance of the microshoots produced. The highest number of new microshoots/explant (1.68), with a 3.0 cm shoot height and a 4.2 proliferation rate, was obtained when this medium was supplemented with 20 M zeatin. The number of microshoots/explant and proliferation rate increased to 1.85 and 6.8, respectively, by using the combination of 5–20 M zeatin with 1 M BA, but shoot height was reduced. 2iP was the least effective of the cytokinins tested. Combination of 20 M zeatin with 10 M GA3 affected positively shoot proliferation resulting in 1.80 microshoots/explant, 3.0 cm shoot height and 7.0 proliferation rate. However, the same concentration of GA3 in combination with 1 M BA reduced the number of new microshoots/explant (0.48) as well as the proliferation rate (2.4), although shoot height remained nearly the same (2.7). The effect of indole-3-butyric acid (IBA), -naphthaleneacetic acid (NAA) and putrescine on root induction was also studied. A rooting percentage of up to 70% and 2.3 roots/microshoot were achieved by the combination of 12 M IBA + 3 M NAA, however, abscission of shoot tips and leaves appeared. Both variables were increased, up to 93% and 4.0, respectively, by the addition of 30 M putrescine in the medium, without any undesired side effect. After acclimatization, survival of rooted microshoots was high (90%) while that of non-rooted ones was low (30%).  相似文献

Summary The enzymes -amylase (-1, 4-glucan 4-glucanohydrolase, 3.2.1.1), -amylase (-1,4-glucan maltohydrolase, 3.2.1.2) and phosphorylase (-1,4-glucan: orthophosphate glucosyltransferase, 2.4.1.1) were assayed in whole grains of barley throughout the maturation period. -amylase and phosphorylase had peaks of activity between 25 and 30 days after anthesis. On the other hand the activity of -amylase in both the available and latent forms reached a maximum value at 35 days after anthesis which did not decrease thereafter. -amylase activity was also assayed throughout development in the endosperm, aleurone, testa pericarp and embryo. Latent -amylase reached a constant maximum value in endosperm at 35 days but available -amylase reached a peak of activity at 25 days and then declined to zero at 45 days. Only latent -amylase was associated with the aleurone layer and activity rose to a maximum value at 35 days. The testa pericarp had mainly latent -amylase whose activity fell from an early maximum at 21 days to zero at 35 days. No hydrolytic activity was associated with the embryo. The phosphorylase activity was low and mainly associated with the endosperm fraction.  相似文献

Expression of a chimeric stilbene synthase gene in transgenic wheat lines   总被引:11,自引:0,他引:11  
A chimeric stilbene synthase (sts)gene was transferred into wheat. Stilbene synthases play a role in the defence against fungal diseases in some plant species (e.g. groundnut or grapevine) by producing stilbenetype phytoalexins like resveratrol. Resveratrol is also claimed to have positive effects to human health. Embryogenic scutellar calli derived from immature embryos of the two commercial German spring wheat cultivars Combi and Hanno were used as target tissue for cotransformation by microprojectile delivery. The selectable marker/reporter gene constructs contained the bargene either driven by the ubiquitinpromoter from maize (pAHC 25, also containing the uidAgene driven by the ubiquitinpromoter), or by the actinpromoter (pDM 302) from rice. The cotransferred plasmid pStil 2 consisted of a grapevine stscoding region driven by the ubiquitin promoter. Eight transgenic Combi and one Hanno TOplant were obtained and, except one Combi TOplant, found to be cotransformants due to the integration of both the stsgene and the selectable marker or reporter genes. Expression of the stsgene was proven by RTPCR, and, for the first time, by detection of the stilbene synthase product resveratrol by HPLC and mass spectrometry. The stsgene was expressed in four of the seven transgenic Combi T_oplants. Two of the respective T1progenies segregated in a Mendelian manner were still expressing the gene. Investigations into methylation of the stsgene showed that in three nonexpressing progenies inactivation was paralleled by methylation.  相似文献

The secondary structure of the toxin fromBacillus thuringiensis subsp.kurstaki (Btk) HD-73 was estimated by Raman, infrared, and circular dichroism spectroscopy, and by predictive methods. Circular dichroism and infrared spectroscopy gave an estimate of 33–40% -helix, whereas Raman and predictive methods gave approximately 20%. Raman and circular dichroism spectra, as well as predictive methods, indicated that the toxin contains 32–40% -sheet structure, whereas infrared spectroscopy gave a slightly lower estimate. Thus, all of these approaches are in agreement that the native conformation of Btk HD-73 toxin is highly folded and contains considerable amounts of both -helical and -sheet structures. No significant differences were detected in the secondary structure of the toxin either in solution or as a hydrated pellet.  相似文献

Three hemoglobin components in carp designated CI, CII, and CIII, were isolated by DEAE-Toyopearl ion-exchange chromatography. Constituent globin chains, 1, 2, 1 and 2, were analyzed by urea-Triton acid polyacrylamide gel electrophoresis and isolated by high performance liquid chromatography with a reversed-phase column. Tryptic peptide mapping indicated that the -globin chains of the three hemoglobin components have slightly different structures. In addition, N-terminal amino acid sequence analysis indicated that the 1-globin chain has a primary structure different from that of the 2-chain. A series of hybridization experiments between isolated hemoglobins, together with such structural properties of globin chains, suggested that the three hemoglobins have the following compositions: CI (1 2 2 1 ), CII (1 2 1 2), and CIII (1 2 2 2 ). Summary One hundred and twenty-two varieties, lines and wild accessions of Lycopersicon were screened under three different regimes during the autumn/winter season of 1982–83 and 1983–84 for resistance to tomato leaf curl virus (TLCV). L. hirsutum f. glabratum (B6013) and L. hirsutum f. typicum (A1904) proved to be highly resistant to TLCV in all three environments. Various accessions of L. peruvianum were also highly resistant. L. pimpinellifolium (A1921) exhibited no TLCV symptoms within 90 days. Of the cultivated varieties, Acc 99 exhibited the minimim score for susceptibility; AC 142, Collection No. 2, Kalyanpur Angurlata and HS 101 had a low rating for virus incidence. The inheritance of resistance was studied in the interspecific crosses between a TLCV resistant line of L. pimpinellifolium (A1921) and five (HS 101, HS 102, HS 110, Pusa Ruby and Punjab Chhuhara) susceptible cultivars of L. esculentum. Parents, F1, F2 and backcross progenies were artificially inoculated with local strains of TLCV using vector the viruliferious whitefly, Bemisia tabaci (Genn.). Data indicated that the resistance of L. pimpinellifolium (A 1921) is monogenic and incompletely dominant over susceptibility.  相似文献

Zusammenfassung Der Einfluß des lichtempfindlichen Pinealkomplexes (Stirnorgan und Epiphysis cerebri) von Rana temporaria auf dorsale diencephale und mesencephale Hirnzentren breitet sich über ein größeres Gebiet aus als man bisher angenommen hat. Nach operativer Unterbrechung des Nervus- und Tractus pinealis wurde mit Punktzählverfahren die Dichte der Zell- und Zellkernvolumina sowie der Kapillaroberfläche im dorsalen Thalamus, mesencephalen (postthalamischen) zentralen Grau, Ganglion habenulae, Tectum opticum, in der Area praetectalis und im Subcommissuralorgan (Sco) bestimmt. Die morphometrischen Meßergebnisse lassen sich wie folgt interpretieren: 1. Der Pinealkomplex wirkt auf das Sco, das zentrale Grau und die Area praetectalis ein (vgl. Paul u. Mitarb., 1971); das Sco wird in erster Linie über das Stirnorgan beeinflußt. 2. Weitere Kerngebiete des Zwischenhirns (Ganglion habenulae und dorsaler Thalamus) gehören funktionell zu dem vom Pinealkomplex kontrollierten System. 3. Die von Kemali und Braitenberg (1970) beschriebene Asymmetrie der Ganglia habenularum zeigt sich auch im funktionellen Zusammenspiel dieser Kerngebiete mit dem Pinealkomplex, dem dorsalen Thalamus und dem dorsalen Abschnitt des zentralen Graus. Die in Stirnorgan und Epiphysis cerebri transformierten Lichtreize werden in Hirnabschnitte weitergeleitet, die olfaktorische und retinale Signalmuster verarbeiten. Über die biologische Bedeutung der nervösen Beziehungen des Stirnorgans zum sekretorischen Sco können noch keine Angaben gemacht werden.
Study of mesencephalic and diencephalic centers after transection of the centripetal nervous pathways of the pineal complex in Rana temporaria L.
Summary Nervous influences of the pineal complex of the frog, Rana temporaria, exerted upon mesencephalic and diencephalic centers were investigated using morphometric methods after the transection of the nervus or the tractus pinealis. Densities of the cell- and nuclear volumes and of the capillaries were measured by a point-counting method in the dorsal thalamus, the mesencephalic (postthalamic) central gray, the habenular ganglion, the optic tectum, the pretectal area, and the subcommissural organ (sco). The morphometric measurements showed: 1. A functional influence of the pineal complex on the sco, the central gray, and the pretectal area (see Paul et al., 1971); the frontal organ seems to be primarily related to the sco. 2. The habenular ganglia and the dorsal thalamus belong to a system which is controlled by the pineal complex. 3. The morphological asymmetry of the habenular ganglia (Kemali and Braitenberg, 1970) has an influence on the functional interactions between the pineal complex, the dorsal thalamus, and the central gray. The impulses arising in the light-sensitive pineal complex of the frog are conducted to brain regions which are known to integrate olfactory and retinal sensory inputs. The functional significance of the nervous connexion of the frontal organ with the secretory sco is still open to discussion.

Methyl jasmonate (JA-Me) at 10–3 Minhibited Amaranthus caudatus seed germination anddecreased -amylase activity. Exogenous gibberellin A3(GA3) and ethylene, but not benzyladenine (BA), increased activity ofthe enzyme in the presence of JA-Me, with ethylene being the most effective. Theinhibitor of ethylene action, 2,5-norbornadiene (NBD) inhibited seed germinationand decreased -amylase activity. The inhibitory action of JA-Me onAmaranthus caudatus seed germination is associated with theinhibition of -amylase activity. Summary The passive electrical properties and initiation of action potentials have been examined in the external epithelium of oikopleurid larvacean tunicates. The epithelial cells are electrically coupled, and are polygons up to 200 m across and up to 1.4–2.8 m thick. Membrane constants determined by a 2-electrode study were forO. dioica:: 922 m; Rm: 4.3 kcm2; Ri: 82.7 cm. Corresponding values for the largerO. longicauda were: 3350 m; 35.6 kcm2; and 104.5 cm. Mean resting potentials in both species were around 80 mV. Mechanical stimulation evokes over-shooting action potentials propagated (at 18 °C) at some 40 cm/s. These are rapid events, repolarisation being almost complete in 5 ms. There is no undershoot.When the recording electrode penetrates the epithelial cell from its inner surface distant mechanical stimulation may evoke similar action potentials arising from the stimulus site, but more often evokes graded small depolarisations which give rise to action potentials with increasing strength of mechanical stimulation. Reasons are given for considering these to be generator potentials resulting from deformation of the outer epithelial cell membrane by the tip of the recording electrode. The effects of epithelial action potentials upon the potentials recorded from the caudal muscle cells are briefly described.  相似文献
