Trichophyton rubrum (T. rubrum) is the most common of the superficial fungi. In an effort to better understand the genetic and biochemical makeup ofT. rubrum, we generated cDNA libraries from 3 growth stages and used these to isolate 4002 unique expressed sequence tags (ESTs). Sequence comparisons with the Genbank database allowed 1226 of the ESTs to be assigned putative functions or matched with homologs from other organisms. Of the remaining ESTs, 989 were only weakly similar to known sequences and 1787 had no identifiable functions, suggesting that they represent novel genes. We further analyzed the presence of several important genes involved in the growth, metabolism, signal transduction, pathogenesis and drug resistance inT. rubrum. This information was used to newly elucidate important metabolic pathways inT. rubrum. Taken together, our results should form the molecular basis for continued research on the physiological processes and pathogenic mechanisms ofT. rubrum, and may lead to a better understanding of fungal drug resistance and identification of new drug targets.  相似文献

Lipolytic activity of dermatophytes was tested by the method generally used forCandida lipolytica. Most of the freshly isolated strains ofMicrosporum canis, M. gypseum, Epidermophyton floccosum andTrichophyton mentagrophytes gave positive reactions, whereas, only few strains ofT. schoenleini, T. violaceum, T. megnini, T. rubrum andT. tonsurans yielded such reactions. The mechanism of the aerobic dark assimilation of acetate in the photoheterotrophically grown purple nonsulfur bacteriumRhodospirillum rubrum was studied. Both in the light and in the dark, acetate assimilation inRsp. rubrum cells, which lack the glyoxylate pathway, was accompanied by the excretion of glyoxylate into the growth medium. The assimilation of propionate was accompanied by the excretion of pyruvate. Acetate assimilation was found to be stimulated by bicarbonate, pyruvate, the C4-dicarboxylic acids of the Krebs cycle, and glyoxylate, but not by propionate. These data implied that the citramalate (CM) cycle inRsp. rubrum cells can function as an anaplerotic pathway under aerobic dark conditions. This supposition was confirmed by respiration measurements. The respiration of cells oxidizing acetate depended on the presence of CO2 in the medium. The fact that the intermediates of the CM cycle (citramalate and mesaconate) markedly inhibited acetate assimilation but had almost no effect on cell respiration indicated that citramalate and mesaconate were intermediates of the acetate assimilation pathway. The inhibition of acetate assimilation and cell respiration by itaconate was due to its inhibitory effect on propionyl-CoA carboxylase, an enzyme of the CM cycle. The addition of 5 mM itaconate to extracts ofRsp. rubrum cells inhibited the activity of this enzyme by 85%. The data obtained suggest that the CM cycle continues to function inRsp. rubrum cells that have been grown anaerobically in the light and then transferred to the dark and incubated aerobically.  相似文献

Summary WhenTrichophyton rubrum is grown in a minimal medium containing glucose, the carbon skeleton of fungal phenylalanine and tyrosine is derived from the glucose carbon. Tracer experiments with variously labeled glucose-C14 indicate that phenylalanine synthesis is linked to glycolysis, but suggest that the pentose phosphate pathway is not involved. These findings suggest that aromatic amino acid biosynthesis may not be linked to the shikimic acid pathway inT. rubrum.  相似文献

Summary and Conclusions A hitherto unknown variant ofT. rubrum var.flava is described which besides its yellow pigment has the following characteristics different fromT. rubrum (Castellani). Analysis of the fatty acid content ofT. rubrum showed that the organism is capable of synthesizing a variety of saturated and unsaturated long-chain fatty acids (Kostiw, Vicher &Lyon, 1966). The present study was undertaken to determine the mechanism of synthesis of these fatty acids. Experimental data point to the presence of two mechanisms of fatty acid synthesis inT. rubrum:de novo synthesis and chain elongation. Secretion of CMCase byTermitomyces clypeatus was only observed in the presence of a gluconeogenic amino acid, a citrate-cycle acid, maleate, subinhibitory concentrations of glucosamine, or fluoride in the medium. The enzyme was not secreted in the presence of caffeine or IBMX or theophylline, and these phosphodiesterase inhibitors lowered the secretion of CMCase by glutamate. The presence of both glucosamine and glutamate in a cellulose medium were, however, antagonistic to CMCase secretion. In a growth medium, xylose and glucose were equivalent carbon source for the fungus while succinate was a poor source and strongly repressed growth at higher concentrations. Growth ofT. clypeatus was highly favored in media containing xylose/glucose with succinate/glutamate. During growth ofT. clypeatus in a glucose medium, the intracellular glucose level was stabilized by the presence of succinate, glutamate or glucosamine in the medium. All these observation suggested that a negative cellular regulation, mediated by carbon catabolic product(s), existed inT. clypeatus which regulated the secretion of CMCase. A transient but significant increase of intracellular cAMP and cGMP levels was observed at the onset of mycelial growth in glucose and glucose/maleate media, respectively.  相似文献

Summary Isolates ofT. mentagrophytes andT. rubrum are encountered that cannot be distinguished from each other solely on the basis of morphological criteria.Since these two species fundamentally differ in the manner in which they attack hairin vitro, this property can be used as a diagnostic aid, when correlated with their morphologic characteristics as developed on a variety of media. T. mentagrophytes radially penetrates hair segments immersed in water forming wedge-shaped perforations.T. rubrum does not perforate hair.The method of determining the ability or inability ofT. mentagrophytes orT. rubrum to perforate hair and the use of this test in identifying those species are described.  相似文献

The capitula ofTaraxacum officinale andT. japonicum open in response to temperature rise at lower temperatures (thermonasty), and in response to light at higher temperatures (photonasty), as was the case inT. albidum. The capitula ofT. officinale could respond to the same temperature rise more sensitively than those ofT. albidum orT. japonicum. The minimum temperature for photonastic opening is as low as 13 C forT. officinale, while that forT. albidum andT. japonicum is about 18 C. That is why the capitula ofT. officinale opened earlier than those ofT. albidum andT. japonicum in the morning in April under natural conditions. The capitulum continued to be open for about 13–14 hr inT. officinale and about 8–11 hr inT. japonicum and inT. albidum both under natural conditions in April and even under constant light-temperature conditions, suggesting that the time of capitula-closing in these three species is not controlled by changes in environmental factors (light and/or temperature).  相似文献

Summary A mutual antagonism (in vitro) has been shown betweenTrichophyton rubrum andCandida albicans by means of two membered pure cultures and by means of cultures filtrates from each organism grown singly. The growth ofT. rubrum was inhibited by metabolic products ofC. albicans. The pigment ofT. rubrum has been shown to be a pH indicator in vivo. Metabolic products ofT. rubrum completely inhibit the development of mycelia byC. albicans, but have little effect on growth in the yeast phase. Chemical fractionation experiments indicated that there are two diffusible metabolic products ofT. rubrum affecting mycelia development: 1) soluble in water and in acetone, heat stable, and adsorbed by activated charcoal, 2) soluble in water, insoluble in acetone, heat labile, and not adsorbed by activated charcoal. The general phenomenon of yeast to mycelia conversions has been considered in detail. A notation system has been developed in this connection; yeast (Y) to mycelia (M) transformations may be expressed as YM; interconversions of the type exhibited byBlastomyces may be written Y:M. The relationships between these processes and the analogous bacteria (B) to filament (F) conversions, B F, has been pointed out. Evidence that Y..M, YM, and BF may result from the inhibition of a common unit enzymatic mechanism has been presented. Converging evidence from such diverse fields as the physicochemical study of the kinetics of bacterial growth (Hinshelwood), genetics of irradiated bacteria (Witkin; Eisenstark andClark), cytochemical mechanism of penicillin action (Pratt andDufrenoy), and dimorphism of pathogenic fungi (this paper;Nickerson andEdwards) are all interpreted as pointing in this direction.  相似文献

Summary Aldolase activity in the cell-free extracts of two dermatophytes,T. mentagrophytes andT. rubrum, was investigated. The kinetics of the enzyme and the effects of metal ions and metal-binders are also reported. The enzyme was more active inT. mentagrophytes than inT. rubrum. The optimum pH for the enzyme action was 7.2 and it was completely inactivated at 60° C. Cobalt and magnesium ions and cysteine activated the enzyme. Inhibition caused by EDTA and o-phenanthroline was partially reversed by cobalt ions. The dermatophyte aldolase resembles bacterial aldolase in its properties.  相似文献

Conclusions Of the several morphological and physiological criteria that have been proposed to identifyTrichophyton rubrum andTrichophyton mentagrophytes differentially, only the hair invasion test agrees with the immunological results reported here.A typical strain ofT. rubrum andT. mentagrophytes was obtained from the Centraalbureau voor Schimmelcultures (Netherlands) and was used as reference strain. All isolates that did not invade hair shafts (in vitro) and that produced three precipitate lines in agargel diffusion tests against anti-T. rubrum serum (refence strain, Tr-1) were placed in one group. These isolates all also produced a red undersurface pigment and macroconidia with long parallel sides. They were labledT. rubrum.Other isolates that did invade hair shafts, and produced only two precipitate lines against anti-T. rubrum serum were placed in another group and were labeledT. mentagrophytes. These isolates had variable characteristics of pigment production, colony and conidial morphology.The antigenic studies thus corroborate the validity of the hair invasion test. The hair invasion test will probably remain in wider use because of its simplicity.  相似文献

Aims: This study sought to develop a less expensive medium for growth of the polyhydroxyalkanoate-producing bacterium Rhodospirillum rubrum from the ethanol production coproduct, condensed corn solubles (CCS). Methods and Results: Small-scale trials using R. rubrum were performed in aerated or anaerobic stoppered serum bottles filled with media. The CCS (240 g l−1) achieved a maximum cell density and growth rate comparable with the defined supplemented malate-ammonium medium (mSMN) or tryptic soy broth. Microaerophilic solubles medium cultures exhibited significantly higher maximum cell densities and growth rates than did strictly anaerobic cultures; while illumination, nickel or biotin addition had no effect. Growth of R. rubrum in a pH controlled bioreactor was significantly better in CCS (240 g l−1) than in mSMN medium and supported production of 0·36% (cell dry weight) poly-(3-hydroxybutyrate-Co-3-hydroxyvalerate) after 24 h. Conclusions: A CCS medium was devised that supported R. rubrum growth for biopolymer production as effective as the defined medium. Significance and Impact of the Study: This study demonstrates that a more economical medium can be developed for biopolymer production using a low value coproduct from ethanol production. The impact is that this inexpensive solubles medium may make it more economical to produce the biopolymer on a commercial scale.  相似文献

Trichophyton rubrum is responsible for the majority of dermatophytosis. Current systemic and topical antifungals against dermatophytosis are often tedious and sometimes unsatisfactory. Antimicrobial photodynamic therapy (aPDT) is a non-invasive alternative suitable for the treatment of superficial fungal infections. This work investigated the photodynamic inactivation efficacy and effects of aloe-emodin (AE), a natural photosensitizer (PS) against T. rubrum microconidia in vitro, and evaluated the treatment effects of AE-mediated aPDT for T. rubrum-caused tinea corporis in vivo and tinea unguium ex vivo. The photodynamic antimicrobial efficacy of AE on T. rubrum microconidia was evaluated by MTT assay. The inhibition effect of AE-mediated aPDT on growth of T. rubrum was studied. Intracellular location of AE, damage induced by AE-mediated aPDT on cellular structure and surface of microconidia and generation of intracellular ROS were investigated by microscopy and flow cytometry. The therapeutic effects of AE-mediated aPDT against dermatophytosis were assessed in T. rubrum-caused tinea corporis guinea pig model and tinea unguium ex vivo model. AE-mediated aPDT effectively inactivated T. rubrum microconidia in a light energy dose-dependent manner and exhibited strong inhibitory effect on growth of T. rubrum. Microscope images indicated that AE is mainly targeted to the organelles and caused damage to the cytoplasm of microconidia after irradiation through generation of abundant intracellular ROS. AE-mediated aPDT demonstrated effective therapeutic effects for T. rubrum-caused tinea corporis on guinea pig model and tinea unguium in ex vivo model. The results obtained suggest that AE is a potential PS for the photodynamic treatment of dermatophytosis caused by T. rubrum, but its permeability in skin and nails needs to be improved.  相似文献

The blue-green algaNostochopsis lobatus released amino acids (glycine, serine, arginine and glutamic acid), sugars (glucose, fructose and sucrose), organic acids (oxaloacetic acid and oxalic acid) and protein in the culture medium. The quantity of extracellular amino acids and protein increased with age of culture from 30 to 120 d. All culture filtrates ofN. lobatus were highly toxic to spore formation and germination ofWestiellopsis prolifica and growth and conjugation ofSpirogyra decimina, while old-age culture filtrates inhibited total chlorophyll, heterocyst and akinete formation in the same alga and total chlorophyll and zoospore formation inChaetophora attenuata. Summary The present study is a morphological, biochemical and spectrophotometric characterization of the eye lens pigmentation in 45 specimens (11–88 mm in standard length) of the deep-sea hatchetfish,Argyropelecus affinis (Stomiiformes: Sternoptychidae). For comparison, we also examined available lenses of other members of the family Sternoptychidae, including three other species of the genusArgyropelecus, and two species of the genusSternoptyx. Lens pigmentation was observed in all specimens ofArgyropelecus spp. larger than about 36 mm in standard length, but was absent in allArgyropelecus spp. individuals less than 36 mm. However, lens pigmentation was not observed inSternoptyx specimens of any size. Detailed studies ofA. affinis indicated that (1) at 36 mm the nascent lens fiber cells, which are continually laid down over preexisting, unpigmented cells, begin incorporating pigment, and (2) the pigment concentration increases steadily as pigmented cells are added during lens growth. Spectrophotometric and biochemical data suggested that the pigment is a carotenoprotein complex, the carotenoid-like chromophore being strongly associated with a specific soluble lens protein, alpha crystallin. While the lens coloration in these fishes is age-related, analyses of the retinal visual pigment revealed no concomitant age-related change in the peak wavelength of retinal sensitivity in these fishes. Our data on the spectral absorbance of the lens and visual pigment of these fishes suggest that the lens pigmentation acts as a short-wave filter to improve acuity of the visual system.  相似文献

Jensen, Altschuler &Bard (1957) have surveyed the respiratory enzymes ofTrichophyton mentagrophytes. Enzymes of the hexosediphosphate, hexosemonophosphate, and tricarboxylic acid pathways were found plus a group of miscellaneous enzymes. In view of the close morphological, immunological, and nutritional similarities ofT. mentagrophytes toTrichophyton rubrum, we felt that a study of the enzymology of the latter organism had potential value in forming a basis for the comparison of the two dermatophytes. Our results showed that cell-free extracts ofT. rubrum contain glucokinase, TPN-dependent glucose and fructose dehydrogenases and separate TPN- and DPN-linked glycerol dehydrogenases. These results are in accord with findings of other investigators. ATPase activity was minimal, and shikimic acid kinase, and shikimic and quinic acid dehydrohydrogenases were not detected.  相似文献

Clinical and mycological data on 1000 cases of dermatophytosis seen from 1970 to 1975 are reported. Tinea pedis was present in 44%, T. rubrum being the main causative agent; nail involvement was present in 41% of these cases. Tineainguinalis was present in 13%, 38% of them females, withT. rubrum isolated in 87% and the swimming-pool incriminated as the main source of contamination. There were various manifestations of tinea corporis in 128 patients, withT. rubrum responsible in 91%. Among the 21 cases of tinea capitis, there was one adult female with a kerion-type due toT. violaceum, 4 cases due toM. gypseum and 2 toM. canis. Tinea favosa was seen in 6 patients, all of them females and one with a spreading fungus on the soles and palms. Amonh the 10 cases with tinea barbae,T. rubrum was found in 4,T. tonsurans in 3,T. mentagrophytes in 2 andM. gypseum in one. There were widespread lesions ofT. rubrum located on large areas of the body and including the legs, arms, groins and nails, in 11 patients with other disorders, including diabetes, Cushing's syndrome and lymphoma.It was apparent that the dermatophytic flora of Israel comprises 10 dermatophytes,T. rubrum being the predominant agent of infection (in more than 80%), with a gradually increasing incidence evident. The other agents found in our survey wereT. mentagrophytes, T. violaceum, T. schoenleinii, T. tonsurans (found sporadically),E. floccosum andMicrosporum spp.M. canis andM. gypseum, the latter being reported for the first time in this country.T. verruccosum andT. megnini, mentioned in former publications, were not found in this survey.  相似文献

Summary Addition of various concentrations (0.5–20 mM) of acetylsalicylic acid (ASA) to tumor lines ofCatharanthus roseus cultivatedin vitro and requiring corn starch as carbon source, produced remarkable effects on secondary metabolite production. An increase of 505% total alkaloids per culture (cells plus liquid medium), 1587% total phenolics (liquid medium), 612% total furanocoumarins (liquid medium) and 1476% total anthocyanins (liquid medium) was detected. 1 mM ASA in combination with other elicitors, such as homogenates ofAspergillus fumigatus or trans-cinnamic acid, did not further increase the metabolite content substantially. The results suggest that ASA could act as a new biotic elicitor of metabolite production inC. roseus cell suspension culture.  相似文献
