Pluronic F-68, PEG 8000, or PEG 20 000 added to cell suspension cultures of transgenic Nicotiana tabacum promoted cell growth and the production of the recombinant murine granulocyte macrophage-colony stimulating factor (mGM-CSF) in a 5-l stirred tank bioreactor. The specific growth rates were enhanced from 0.27 d–1 to 0.47 d–1, 0.37 d–1 and 0.4 d–1 when Pluronic F-68, PEG 8000, or PEG 20 000 was added, respectively. The maximum cell density was also increased most to 13.6 g l–1 when Pluronic F-68 was added (11.3 g l–1 in the control culture). In terms of mGM-CSF production, PEG 8000 gave the greatest stimulation and with 2 g PEG 8000 l–1, mGM-CSF increased from 1.6 to 6.6 ng ml–1.  相似文献

Brevibacterium linens forms hydrolytic enzymes which can be used to accelerate the ripening of cheese without causing bitterness. B. linens ATCC 9172 was grown to a high cell density (50 g dry wt l–1 after 60 h) in a mineral medium containing lactic acid, soy-peptone and ammonium sulphate by applying a continuous feed of nutrients. The maximal activities of l-leucine aminopeptidase and cell-associated proteinase were 286 U l–1 and 202 U l–1, respectively. The cell-associated lipolytic activity exhibited a strong and sudden increase at 46 h, resulting in a maximum of 9.5 U g–1 dry wt; thus the volumetric productivity of proteolytic and lipolytic activity was 4220 U l–1 h–1 and 7.3 U l–1 h–1, respectively.  相似文献

Preliminary attempts to define a completely synthetic medium able to support divisions of haploid tobacco mesophyll protoplasts at low initial densities have failed. High protoplast concentrations together with large amounts of naphtaleneacetic acid in the medium (3 mg l-1 NAA) were required for maximal induction of protoplast division. However, cell suspensions derived from haploid protoplasts after four days of preculture at high initial cell densities could be diluted to densities as low as 1–4 cells ml-1, provided the concentration of NAA in the medium was lowered to below 0.3 mg l-1. The optimal NAA supply for low cell density growth was affected by the nature of the nitrogen source.A simple minimal medium which supports the growth of these haploid cells with a plating efficiency of 30–40%, independent of the cell density in the range of 1–4 to 3·104 cells ml-1, has been established. In this medium inositol was the only vitamin stringently required for growth.Growth of cells at low densities was also possible in a medium initially containing 3 mg l-1 NAA, provided it was conditioned by the growth of protoplasts at high densities. Preliminary experiments with [14C]NAA showed that the amount of free NAA remaining in the medium after preincubation at high densities was drastically reduced. Simultaneously, NAA conjugates accumulated in the medium. Candida parapsilosis was grown for 59 h in a medium containing corn cob hydrolysate consisting of 50 g xylose l–1, 3.0 g glucose l–1, 2.0 g arabinose l–1, and 0.9 g acetic acid l–1. A biomass of 9.1 g l–1 was produced with 36 g xylitol l–1 and 2.5 g ethanol l–1. In a medium containing 50 g xylose l–1 instead of corn cob hydrolysate, the concentrations of cells, xylitol, and ethanol were 8.6 g l–1, 33 g l–1, and 0.2 g l–1, respectively. The differences between two cultures were due to the glucose and arabinose in the corn cob hydrolysate stimulating growth and the low concentration of acetic acid stimulating xylitol production.  相似文献

Callus of Orthosiphon stamineus could be induced successfully from petiole, leaf and stem tissues but not roots when cultured on MS medium containing different concentration of NAA (0–4.0 mg l–1) and 2,4-D (0–2.0 mg l–1). Highest fresh weight callus production was obtained from leaf explants and those with best friability were obtained on MS medium plus 1.0 mg l–1 2,4-D plus 1.0 mg l–1 NAA. Cell suspension cultures were established from these cultures. The appropriate cell inoculum size for the best cell growth was 0.75 g of cells in 20 ml culture medium. Cell suspension culture using MS medium supplemented with 1.0 mg l–1 2,4-D promoted the best cell growth with maximum biomass of 8.609 g fresh weight and 0.309 g dry weight 24 days after inoculation. Cells that grew in MS medium supplemented with 1.0 mg l–1 2,4-D reached the stationary growth phase in 15 days as compared to the cells that grew in MS medium supplemented with 1.0 mg l–1 2,4-D + 1.0 mg l–1 NAA reached the stationary phase in 24 days. MS medium supplemented with 1.0 mg l–1 2,4-D was considered as the maintenance medium for maintaining the optimum cell growth of O. stamineus in the cell suspension cultures with 2-week interval subculture.  相似文献

A murine hybridoma cell line producing a monoclonal antibody against penicillin-G-amidase and a murine transfectoma cell line secreting a monovalent chimeric human/mouse Fab-antibody fragment were cultivated in three different media (serum-containing, low protein serum-free, and iron-rich protein-free) in flask cultures, stirred reactors and a fixed bed reactor. In static batch cultures in flasks both cell lines showed similar good growth in all three media.In suspension in a stirred reactor, the hybridoma cell line could be cultivated satisfactory only in serum-containing medium. In low protein serum-free medium, Pluronic F68 had to be added to protect the hybridoma cells against shear stress. But even with this supplement only batch, not chemostat mode was possible. In iron-rich protein-free medium the hybridoma cells grew also in continuous chemostat mode, but the stability of the culture was low. The transfectoma cell line did not grow in stirred reactors in any of the three media.Good results with both cell lines were obtained in fixed bed experiments, where the cells were immobilized in macroporous Siran®-carriers. The media, which were optimized in flask cultures, could be used without any further adaptation in the fixed bed reactor. Immobilization improved the stability and reliability of cultures of non-adherent animal cells in serum-free media tremendously compared to suspension cultures in stirred reactors. The volume-specific glucose uptake rate, an, indicator of the activity of the immobilized cells, was similar in all three media. Poly(3-hydroxybutyrate) (PHB) was produced by fed-batch cultures of Ralstonia eutropha with phosphate limitation under different glucose concentrations. When glucose was kept at 2.5 g l–1, cell growth and PHB synthesis were limited due to the shortage of carbon source but a higher PHB content occurred in the cell-growth stage. This shows that a low glucose concentration is favorable for PHB accumulation in R. eutropha. PHB obtained with glucose at 9 g l–1 is 1.6 times that obtained with 40 g l–1. When glucose was in the range of 9 to 40 g l–1, PHB concentration and productivity decreased significantly with the increase of glucose concentration. The highest PHB productivity was obtained with glucose at 9 g l–1.  相似文献

Trichoderma reesei Rut-C30 is a highly derepressed mutant which synthesised active cellulases in culture media containing glucose and lactose as the only carbon sources. The maximum biomass, filter paper and specific filter paper activities for cell growth on 20 g glucose l–1 were 20 g dry cell wt l–1, 1.9 FPU ml–1 and 4.8 FPU mg–1 protein respectively, while on 40 g glucose l–1 were 25 g dry cell wt l–1, 4.5 FPU ml–1 and 6.2 FPU mg–1 protein, respectively. This strain had a higher specific filter paper activity (6.2 FPU mg–1 protein) than was produced by other T. reesei mutants (3.6 FPU mg–1 protein).  相似文献

Freely suspended and Ca-alginate-immobilized cells of Pimelobacter sp. were used for degradation of pyridine. When the pyridine concentration was up to 2 g l–1, freely suspended cells completely degraded pyridine regardless of the initial cell concentrations used. However, when the pyridine concentration increased to 4 g l–1, the initial cell concentration in freely suspended cell culture should be higher than 1.5 g dry cell weight l–1 for complete degradation of pyridine. In addition, a freely suspended cell culture with a high initial cell concentration resulted in a high volumetric pyridine-degradation rate, suggesting the potential use of immobilized cells for pyridine-degradation. When the immobilized cells were used for pyridine-degradation, neither specific pyridine-degradation rate nor tolerance against pyridine was improved. However, a high volumetric pyridine-degradation rate in the range 0.082–0.129 g l–1 hr–1 could be achieved by the immobilized cells because of the high cell concentration. Furthermore, when the immobilized cells were reused in degrading pyridine at a concentration of 2–4 g l–1 they did not lose their pyridine-degrading activity for 2 weeks. Taken together, the data obtained here showed the feasibility of using immobilized cells for pyridine-degradation.  相似文献

Summary Direct alcoholic fermentation of dextrin or soluble starch with selected amylolytic yeasts was studied in both batch and immobilized cell systems. In batch fermentations, Saccharomyces diastaticus was capable of fermenting high dextrin concentrations much more efficiently than Schwanniomyces castellii. From 200 g·l–1 of dextrin S. diastaticus produced 77 g·l–1 of ethanol (75% conversion efficiency). The conversion efficiency decreased to 59% but a higher final ethanol concentration of 120 g·l–1 was obtained with a medium containing 400 g·l–1 of dextrin. With a mixed culture of S. diastaticus and Schw. castellii 136 g·l–1 of ethanol was produced from 400 g·l–1 of dextrin (67% conversion efficiency). S. diastaticus cells attached well to polyurethane foam cubes and a S. diastaticus immobilized cell reactor produced 69 g·l–1 of ethanol from 200 g·l–1 of dextrin, corresponding to an ethanol productivity of 7.6g·l–1·h–1. The effluent from a two-stage immobilized cell reactor with S. diastaticus and Endomycopsis fibuligera contained 70 g·l–1 and 80 g·l–1 of ethanol using initial dextrin concentrations of 200 and 250 g·l–1 respectively. The corresponding values for ethanol productivity were 12.7 and 9.6 g·l–1·h–1. The productivity of the immobilized cell systems was higher than for the batch systems, but much lower than for glucose fermentation.  相似文献

Chlorella sorokiniana was cultured in heterotrophic or mixotrophic mode in outdoor enclosed tubular photobioreactor. The culture temperature was maintained at 32–35 °C. At night, theChlorella culture grew heterotrophically, and 0.1 M glucose was completely consumed. The biomass growth yield of glucose was 0.35 ± 0.001 g-biomass g-glucose–1. During the day, the algal culture grew mixotrophically and the biomass growth yield was 0.49 g-biomass g-glucose–1 in low density culture (initial biomass concentration, Xo = 2 g l–1), 0.56 g-biomass g-glucose–1 in medium density culture (Xo = 4 g l–1) and 0.46 g-biomass g-glucose–1 in high density culture (Xo = 7 g l–1). The daily area productivity of the culture, with Xo = 4 g l–1 corresponded to 127 g-biomass m–2 d–1 during the day and 79 g-biomass m–2 d–1 during the night. In all the cultures, the dissolved O2 concentration increased in the morning, reached the maximum value at noon, and then decreased in the afternoon. The dissolved CO2 concentration remained at 3 mBar in the morning and increased in the afternoon. Glycolate was not found to accumulate in culture medium.  相似文献

Summary Klebsiella oxytoca fermented 199 g·l–1 high test or invert molasses using batch fermentation with substrate shift to produce 95.2–98.6 g 2,3-butanediol·l–1 and 2,4–4.3 g acetoin·l–1 with a diol yield of 96–100% of the theoretical value and a diol productivity of 1.0–1.1 g·l–1·h–1. Fermentation was performed numerous times with molasses in repeated batch culture with cell recovery. Such repeated batch fermentation, in addition to a high product yield, also showed a very high product concentration. For example, 118 g 2,3-butanediol·l–1 and 2.3 g acetoin·l–1 were produced from 280 g·l–1 of high test molasses. The diol productivity in this fermentation amounted to 2.4 g·l–1·h–1 and can undoubtedly be further increased by increasing the cell concentration. Because the Klebsiella cultures ferment 2,3-butanediol at an extremely high rate once the sugar has been consumed, the culture was inhibited completely by the addition of 15 g ethanol·l–1 and switching off aeration. Summary Two physiological characteristics of butyric fermentation, inhibition by the acids produced, butyrate and acetate, and dependence on the growth rate of the distribution of these acids, prompted a study of butyrate production in a continuous fermentation system with cell recycle by microfiltration. The influence of the main operating parameters, glucose input (feed concentration and dilution rate) and bleed dilution rate on production of acids and biomass was studied. The performance of the system greatly exceeded the results obtained in batch and simple continuous fermentations as a high productivity for butyrate (9.5 g l–1 h–1) was achieved whilst retaining a satisfactory concentration of butyrate (29.7 g l–1) and low acetate production (0.6 g l–1) at a cell biomass concentration of 35 g l–1. The continuous bioconversion of xylose-containing solutions (obtained by acid hydrolysis of barley bran) into xylitol was carried out using the yeast Debaryomyces hansenii under microaerophilic conditions with or without cell recycle. In fermentations without cell recycle, the volumetric productivities ranged from 0.11–0.6 g l–1 h–1 were obtained for dilution rates of 0.008–0.088 h–1. In experiments performed with cell recycle after membrane separation, the optimum xylitol productivity (2.53 g l–1 h–1) was reached at a dilution rate of 0.284 h–1.  相似文献

Summary This paper presents a study of propionic acid and propionibacteria production from whey by usingPropionibacterium acidi-propionici in continuous fermentation with cell recycle. The highest propionic acid volumetric productivity achieved was 5 g.l–1.h–1 with no biomass bleeding. A maximal biomass concentration of 130 g.l–1 was achieved before initiating biomass bleeding to give a biomass volumetric productivity of 3.2 g.l–1.h–1 with a biomass of 75 g.l–1 and a propionic acid productivity of 3.6 g.l–1.h–1 (for about 100 hours i.e. more than 50 residence times).  相似文献

Batch production of xylitol from the hydrolysate of wheat straw hemicellulose using Candida guilliermondii was carried out in a stirred tank reactor (agitation speed of 300 rpm, aeration rate of 0.6 vvm and initial cell concentration of 0.5 g l–1). After 54 h, xylitol production from 30.5 g xylose l–1 reached 27.5 g l–1, resulting in a xylose-to-xylitol bioconversion yield of 0.9 g g–1 and a productivity of 0.5 g l–1 h–1.  相似文献

Hybridoma WuT3 secreting a monoclonal antibody against T lymphocytes was grown in RPMI 1640 medium supplemented with 1% human serum. The effect of the concentration of peptone, as an additive, was investigated on cell growth, monoclonal antibody formation, and cell metabolism over 0–10 g l–1 range. It was found that 1–5 g l–1 peptone can significantly promote the growth of cells and increase the formation of monoclonal antibody, especially at 3–5 g l–1, when both the accumulating level and secretion rate of monoclonal antibody are higher than that at other peptone concentrations. Based on glucose, lactate and ammonia analysis data, the efficiency of glycolysis was assessed and the utilization of amino acids was more efficient at 3–5 g l–1 peptone. Lake Texoma in southcentral Oklahoma was formed by the impoundment of the Washita and Red Rivers. The Red River is more highly saline than the Washita and creates a complex salinity gradient across the reservoir. Populations of chironomids were monitored with multiple-plate samplers in areas of high (34–113 mg l–1 Cl), intermediate (35–60 mg l–1 Cl) and low (4–27 mg l–1 Cl) salinity during the spring and summer of 1978. Food availability, temperature, salinity and wind direction influenced the distribution of the 14 genera and at least 22 species of chironomids which colonized the multiple-plate samplers. Filter-feeders attained their highest densities in the river-arm stations where levels of particulate organic matter (POM) were high. Algal grazers attained their highest densities in the clear intermediate area where the plates of the samplers were covered with algal mats. Most of the genera believed to be feeding primarily on POM decreased in density as the temperature and density of Glyptotendipes sp. rose. Certain species were restricted to either the Red River arm or the Washita River arm and this is probably a reflection of different salinity tolerances.  相似文献

The kinetics of continuous l-sorbose fermentation using Acetobacter suboxydans with and without cell recycle (100%) were investigated at dilution rates (D) of 0.05, 0.10, 0.15 and 0.3 h–1. The biomass and sorbose concentrations for continuous fermentation without recycle increased as the dilution rate was increased from 0.05 to 0.10 h–1. A maximum biomass concentration of 8.44 g l–1 and sorbose concentration of 176.90 g l–1 were obtained at D=0.10 h–1. The specific rate of sorbose production and volumetric sorbose productivity at this dilution rate were 2.09 g g–1 h–1 and 17.69 g l–1 h–1. However, on further increasing the dilution rate to 0.3 h–1, both biomass and sorbose concentrations decreased to 2.93 and 73.20 g l–1 respectively, mainly due to washout of the reactor contents. However, the specific rate of sorbose formation and volumetric sorbose productivity at this dilution rate increased to 7.49 g g–1 h–1 and 21.96 g l–1 h–1 respectively. Continuous fermentation with 100% cell recycle served to further enhance the concentration of biomass and sorbose to 28.27 and 184.32 g l–1 respectively (in the reactor at a dilution rate of 0.05 h–1). Even though, there was a decline in the biomass and sorbose concentrations to 6.8 and 83.40 g l–1 at a dilution rate of 0.3 h–1, the specific rates of sorbose formation and volumetric sorbose productivity increased to 3.67 g g–1h–1 and 25.02 g l–1 h–1.  相似文献

d-Ribose, a five-carbon sugar, is used as a key intermediate for the production of various biomaterials, such as riboflavin and inosine monophosphate. A high d-ribose-producing Bacillus subtilis SPK1 strain was constructed by the chemical mutation of the transketolase-deficient strain, B. subtilis JY1. Batch fermentation of B. subtilis SPK1 with 20 g l–1 xylose and 20 g l–1 glucose resulted in 4.78 g l–1 dry cell mass, 23.0 g l–1d-ribose concentration, and 0.72 g l–1 h–1 productivity, corresponding to a 1.5- to 1.7-fold increase when compared with values for the parental strain. A late-exponential phase was chosen as the best point for switching to a fed-batch process. Optimized fed-batch fermentation of B. subtilis SPK1, feeding a mixture of 200 g l–1 xylose and 50 g l–1 glucose after the late-exponential phase reduced the residual xylose and glucose concentrations to less than 7.0 g l–1 and gave the best results of 46.6 g l–1d-ribose concentration and 0.88 g l–1 h–1 productivity which were 2.0- and 1.2-fold higher than the corresponding values in a simple batch fermentation.  相似文献
