Data are presented on the isolation, biosynthesis, and identification of a small peptide (H) from parathyroid gland. Under our experimental conditions this peptide (H) represents, in addition to secretory protein-I and proparathyroid hormone, the other major protein which is rapidly synthesized during shorterm incubations of tissue slices. N-terminal sequence analysis was performed on samples of peptide H and the resulting data used to conduct a search of the sequence data bank. The search established the identity of peptide H as ubiquitin. These findings establish parathyroid gland as another system which rapidly produces ubiquitin invitro, in addition to the systems employing hypothalamus and pituitary where ubiquitin biosynthesis was initially observed by Seidah etal and Scherrer etal.  相似文献

The amino acid sequences of pyridoxal-binding tetrapeptide and the NH2-terminal portion of aspartate transaminase from E.coli B were analyzed and compared with those of the corresponding parts of the cytosolic and mitochondrial isozymes from pig heart. After borohydride reduction and chymotryptic digestion of the E.coli enzyme, a pyridoxal-containing peptide was isolated, showing the sequence, Ser-Lys(Pxy)-Asn-Phe, identical with that of the cytosolic isozyme. The NH2-terminal sequence was determined up to 33 residues with a liquid phase sequence analyzer. Nearly the same degree of homology was observed among the NH2-terminal sequences of the three aspartate transaminases.  相似文献

Three chromophoric peptides have been isolated and characterized from tryptic digests of the α subunit of C-phycocyanin from Oscillatoria,agardhii. The amino acid sequences revealed that one phycocyanobilin was ester bond by a tyrosine residue, and another was most probably attached by a thioether linkage. Structural studies of the third chromophoric peptide gave no evidence of how the phycocyanobilin was attached.  相似文献

Proton and 13C magnetic resonance studies are reported on the synthetic polypentapeptide of elastin, HCO-(Val(1)-Pro(2)-Gly(3)-Val(4)-Gly(5))n-Val-OMe, where n ∼- 18. Temperature and solvent dependence of peptide NH chemical shift and solvent dependence of peptide carbonyl chemical shift were used to delineate these moieties preliminary to identification of secondary structure.Based on these studies it is proposed, for the organic solvents of dimethyl sulfoxide, methanol, and low-temperature trifluoroethanol, that dynamic hydrogen bonds form in order of decreasing frequency of occurrence between the Val(1)CO and the Val(4) NH (a β-turn), between the Gly(3) NH and the Gly(5)CO (an 11-atom, hydrogen-bonded ring), and a more limited interaction between the Gly(3)CO and the Gly(5) NH (a γ-turn).Arguments are presented that relate the conformational features proposed above to the coacervate, which is a filamentous state.  相似文献

Chemically formylated Met-tRNAmMet and Met-tRNAfMet species from E.coli and yeast were tested for their capacity to serve as chain-initiators in a cell-free system from E.coli. In the presence of R 17 mRNA, initiation factors and E.coli ribosomes, all four Met-tRNAs could form functional initiation complexes as measured by ribosomal binding kinetics, fMet-puromycin formation and synthesis of a dipeptide fMet-Ala. Unformylated Met-tRNAfMet from E.coli displayed significantly less activity as a peptide chain-initiator than the formylated Met-tRNAmMet species from E.coli and yeast. Although the latter tRNAs were less effective initiators than the “physiological” initiator tRNAs, the data seem to indicate that a blocked α-amino group represents the major token of identification by which Met-tRNA is admitted to function in E.coli peptide chain initiation.  相似文献

The pharmacologic activity of the hexapeptide fragment corresponding to the amino acid fragment 60–65 in β-lipotropin, (β-LPH-(60–65)) was studied in vitro and in vivo. In binding assays on synaptosomal plasma membrane the peptide was found to be equipotent to met-enkephalin, but behaved differently to cations; in contrast to met-enkephalin both Mn+2 and Na+ enhanced the binding of β-LPH-(60–65) to synaptosomal plasma membrane. On both the quinea pig ileum and mouse vas deferens β-LPH-(60–65) inhibited contractions elicited by electrical stimulation and each effect was reversible by naloxone. On the guinea pig ileum β-LPH-(60–65) was equipotent to met-enkephalin and 0.5 as potent as normorphine but on the vas deferens it was 4.6 times more potent than normorphine. The activities of β-LPH-(60–65) appear to be due to the intact compound rather than to its conversion to met-enkephalin, since the peptide extracted from the ileum assay was found to behave identically as β-LPH-(60–65) with high pressure liquid chromatography. When β-LPH-(60–65) was administered centrally to mice and rats, no overt central actions were observed and an antinociceptive effect could not be demonstrated. Nor did β-LPH-(60–65) antagonize morphine action or precipitate the withdrawal syndrome in morphine dependent animals. It is concluded that the good agreement which generally exists between in vitro and in vivo assay procedures for opiate-like activity of morphine and its surrogates does not necessarily hold for the endogenous peptides with similar actions.  相似文献

The synthesis of the cation-binding cyclic octapeptide, cyclo(Glu-Sar-Gly-(N-decyl)Gly)2 is reported. This peptide, containing two ionizable Glu carboxyl side chain protons per molecule, can form neutral cation complexes with divalent ions via protonmetal exchange. Solubilized in chloroform solution, the peptide has been found to extract calcium from an aqueous phase (pH 8.5, 100 mM Tris) generally on a 11 molar basis. By contrast, under comparable conditions but with other metal chlorides, the peptide does not extract magnesium, sodium, or potassium. That the extraction proceeds via proton-metal exchange was demonstrated by the absence of (radioactive) chloride ion from the organic phase. Parallel sets of experiments performed with the naturally-occurring ionophore A23187 reaffirmed that the latter substance extracts calcium and magnesium with nearly equal propensity.  相似文献

The synthetic replicate of a 44 amino acid peptide isolated from a human pancreatic tumor which had caused acromegaly possesses high specific activity to release growth hormone (GH) in anesthetized male rats. The GH secretion induced by this peptide is dose-dependent from 50 ng to 1 μg, with plasma GH concentrations increasing more than 10-fold within 5 min of iv administration at the higher doses. Two enzymatic degradation products of the 44 residue peptide were also isolated and consist of the first 37 and 40 amino acids. All three peptides appear to possess similar potency, on a molar basis, invivo, contrary to invitro results. The specificity of these peptides on GH release was shown by their failure to alter plasma concentrations of prolactin (PRL), thyroid-stimulating hormone (TSH), luteinizing hormone (LH), follicle-stimulating hormone (FSH) and corticosterone. Based on these invivo results, the three peptides with serve as powerful tools with which to investigate the mechanisms of GH secretion.  相似文献

Ternary complex formation between eukaryotic initiation factor 2 (eIF-2), initiator Met-tRNA and guanosine 5′-(β, γ-imino) triphosphate [GMP-P(NH)P] is strongly inhibited by mRNA in the Artemia salina system. Developing A. salina embryos contain a factor which displays a novel activity, namely the ability to counteract the mRNA-induced inhibition of ternary complex formation. This factor is heat-labile. It is proposed that the factor may play an important role in protein biosynthesis by preventing mRNA from inhibiting an early step of peptide chain initiation.  相似文献

The amino acid sequence of a 27-residue peptide released during the early stages of the plasmin digestion of human fibrinogen has been determined. The corresponding cyanogen bromide fragment has also been isolated from the purified α-chains of fibrinogen, although a separable fraction of those chains lack the fragment, evidently because of invivo degradation. The insoluble acrosome granule content of sea urchin sperm consists of a single 30,500 dalton protein named bindin. Bindin mediates species-specific recognition and adhesion of sperm to the egg surface. Bindin from Strongylocentrotus purpuratus (Sp) and Strongylocentrotus franciscanus (Sf) have tyrosine as their single N-terminal amino acid. The pI of Sp bindin is 6.62 and of Sf 6.59. Amino acid analysis reveals almost identical composition between the two species for 16 amino acids. Only two (or three) amino acids, Pro and Asx, show large species differences. Tryptic peptide maps of the two species of bindin show very similar patterns with 24 spots of identical correspondence.  相似文献

The mechanism of activation of ε-prototoxin to ε-toxin has been ascertained from partial amino acid sequences of both ε-prototoxin and ε-toxin. The activation of ε-prototoxin from Clostridium perfringens type D by brief exposure to trypsin is caused by scission of a peptide bond between Lys14 - Ala15. A small peptide (14 amino acid residues) is split from the NH2-terminus of the ε-prototoxin to give the active ε-toxin.  相似文献

Cd-binding peptide 1 (Cd-BP1) from S.pombe (1) has been purified and characterized. Cd-BP1 has a characteristic shoulder at 265 nm in UV absorption spectrum, and shows two marked Cotton bands at 257 nm (negative) and 275 nm (positive). These are not found in the other Cd-thioneins (2) and are unique properties of Cd-BP1 from S.pombe. With acidification (pH 2) and successive neutralization, a shoulder at 265 nm in UV spectrum and a Cotton band at 275 nm disappeared, and the molecular weight changed from 4 000 to 1 800. In connection with these changes, two molecular forms of Cd-binding peptide are considered.  相似文献

The 0.5M KCl wash of rabbit reticulocyte ribosomes (I fraction) catalyzes the deacylation of Met-tRNAfMet. Upon DEAE-cellulose column chromatography, the deacylase activity elutes with the 0.1M KCl wash of the column (f1) and is well-resolved from the peptide chain initiation factors (1–3). The deacylase activity is specific for Met-tRNAfMet (retic., E.coli). Other aminoacyl tRNAs tested including fMet-tRNAfMet (retic., E.coli), Phe-tRNA (E.coli), Val-tRNA (retic.), and Arg-tRNA (retic.) are completely resistant to the action of the deacylase. In the presence of the peptide chain initiation factor (IF1) and GTP, retic. Met-tRNAfMet forms the initiation complex Met-tRNAfMet:IF1:GTP (2), and in this ternary complex Met-tRNAfMet is not degraded by the deacylase. E.coli Met-tRNAfMet binds to IF1 independent of GTP, and in this complex, this Met-tRNAfMet is degraded by the deacylase.Prior incubation of f1 with Met-tRNAfMet (retic.) strongly inhibited protein synthesis initiation, presumably due to deacylation of the initiator tRNA. This inhibition by f1 was completely prevented when Met-tRNAfMet (retic.) was pre-incubated with peptide chain initiation factors.  相似文献

The binding of magnesium ions to two tripeptides, L-Arg-D-Gla-D-Gla-OMe and Z-L-Arg(NO2)-D-Gla-D-Gla-OMe, and to bovine prothrombin fragment 1 as a function of pH has been monitored by 25Mg NMR spectroscopy. Binding to the tripeptide was dependent on peptide ionizations occurring at pH 4.6 – 4.8. The pH dependence of magnesium ion binding to fragment 1 reveals two inflection points 4.2 may be attributed to the deprotonation of the third side chain carboxylic acid group of the double γ-carboxyglutamic acid sequence. The origin of the increased binding of magnesium ions to fragment 1 at pH values above 7 is unknown.  相似文献

Monoamine oxidase, an intrinsic protein of outer mitochondrial membrane, was purified from bovine liver and rabbit antibody against the enzyme was prepared. The antibody could react with the monoamine oxidase of rat liver mitochondria. When rat liver RNA was translated invitro using rabbit reticulocyte lysate and monoamine oxidase peptide in the translation products was immunoprecipitated by the antibody, the peptide was detected in the products programmed by the messenger RNA's from total and free polysomes but not that from bound polysomes. The enzyme synthesized invitro had the same apparent molecular size as the mature protein in outer mitochondrial membrane.  相似文献

A soluble macromolecule-peptide conjugate, [(Met)3-OPEG] inhibited the uptake of Met-Met-[14C] Met into S.cerevisiae. Uptake of leucine into this strain was not affected by Met3-OPEG under identical conditions. Inhibition by the macromolecular inhibitor was competitive (KI = 5.1 × 10?5M)and followed the structural requirments of the peptide transport systems in S.cerevisiae and C.albicans. These findings give the first example of inhibition of metabolite transport by a synthetic macromolecular competitor.  相似文献

The purpose of this report is to review the current literature on cytochrome b5 in hepatic microsomes and to draw conclusions as to its role in microsomal electron transfer pathways. For details concerning the history of cytochrome b5 the reader is reffered to reviews by C. F. Strittmatter (1) and P. Strittmatter (2). For information on the chemistry of cytochrome b5 the reader is reffered to the papers by Ozols and Strittmatter (3), Kajihara and Hagihara (4), and Ehrenberg and Bois-Poltoratsky (5). For more recent studies on the isolation and properties of detergent solubilized cytochrome b5, which contains a hydrophobic peptide enabling reincorporation into membranes, the reader is referred to references 6-12.For simplicity, this minireview is divided into four parts, reflecting areas of study on the role of cytochrome b5 in the microsomes. One major area is in fatty acid 9 desaturation. Two other areas concern cytochrome b5 involvement in cytochrome P-450 mediated mixed function oxidations. The fourth section deals with other non-cytochrome P-450 pathways in which cytochrome b5 is suggested as being a component.  相似文献

Salmon and mammalian gonadotropin-releasing hormones (sGnRH, mGnRH) were tested for their ability to stimulate in vitro gonadotropin (GtH) release from superfused goldfish pituitary fragments. A two minute exposure to either peptide was sufficient to stimulate a dose-dependent increase in GtH release which reached maximum levels in 15 minutes and returned to baseline within one hour. Both peptides were approximately equipotent in stimulating GtH release, as was a superactive analog of mGnRH. These results demonstrate that sGnRH is capable of directly stimulating GtH release from teleost pituitary tissue, and that structural differences between the three peptides tested do not result in significant differences in in vitro bioactivity.  相似文献
