We had previously reported on HytaNPV infection in the looper caterpillars of Hyposidra talaca. Studying the pattern of cumulative daily mortality of looper caterpillars and comparing the post-looper crop production in microbial-control plots and insecticide-treated plots, we report that HytaNPV is a potential microbial agent of H. talaca.  相似文献

In order to replace the conventional chemical pesticides, extensive researches have been done on entomopathogenic fungi. Entomopathogenic fungus Beauveria bassiana is an important biocontrol agent against major economic pests and is being employed in Integrated pest management (IPM) along with synthetic pesticides. Cabbage aphid Brevicoryne brassicae L. is one of the important pests of Brassicaceae family. Therefore, in this research, the virulence isolate of B. brassicae (IRAN 429C) was investigated on adults of cabbage aphid under laboratory conditions. The experiments were conducted at 25 ± 2 °C, 60 ± 10 R. H. and a photoperiod of 16:8 (L: D). After preliminary experiments, the adult aphids were treated with fungal concentrations of 1 × 103 to 1 × 107 spores/ml. Probit analysis was conducted to calculate LC50 and LC95 values for the isolate. Positive correlation was observed between concentrations and pest mortality. LC50 and LC95 values calculated for IRAN 429C isolate are 2.04 × 105 and 1.82 × 108, respectively. The mortality was counted one day after the treatment and then continued for 14 days. Cumulative mortality for 14 days after treatment varied from 54% for IRAN 429C at low concentration (103 conidia/ml) to 83% at high concentration (107 conidia/ml). The lowest LT50 was obtained at 7.67 days for IRAN 429C isolate at concentration 1 × 107 spore/ml. According to the insecticidal activity of mentioned fungi on cabbage aphid, it can be used in biocontrol programmes of B. brassicae.  相似文献

The common pistachio psylla, Agonoscena pistaciae, Burckhardt and Lauterer (Hemiptera: Psyllidae) is the key pest of pistachio orchards in Iran. Chemical control is the common method for the control of this pest. In this study, the effects of pyriproxyfen (an insect growth regulator), neemarin (a botanical insecticide), acetamiprid (a neonicotinoid) and Ferula assafoetida essential oil were investigated on the fifth instar nymphs of A. pistaciae. Probit analysis of concentration-mortality data was conducted to estimate the LC50 values. The LC50 values for neemarin, pyriproxyfen, acetamiprid and F. assafoetida essential oil were estimated to be 0.22, 0.52, 290 and 5.62 ppm, respectively. Our results showed that the fifth instar nymphs of A. pistaciae are very sensitive to neemarin and pyriproxyfen.  相似文献

Soil samples were collected from different rice fields of Singur, Hooghly, West Bengal, India. Spore forming bacteria were isolated from the soil samples and among them, two isolates (BUSNC25 and BUSNC26) were larvicidal against third, fourth and fifth instar larvae of rice leaf folder, Cnaphalocrocis medinalis. The phenotypic, biochemical characterization and 16S rDNA analysis of the two isolates were done. On the basis of phenotypic, biochemical and phylogenetic analysis, the selected bacterial isolates (BUSNC25 and BUSNC26) were identified as Bacillus thuringiensis. The antibiotic sensitivity tests of these two isolates against selected doses of some standard antibiotics were done. Against the 3rd, 4th and 5th instar larvae of C. medinalis, the LC50 values of BUSNC25 were 2.45 × 104, 1.325 × 104 and 2.35 × 104 cfu/ml and of BUSNC26 were 3.375 × 104, 1.9 × 104 and 3.325 × 104 cfu/ml, respectively.  相似文献

The effect of repeated conidial sub-culturing of Metarhizium anisopliae on its virulence against Helicoverpa armigera (Hübner) was studied. The LT50 observed against third instar larvae of H. armigera for the first sub-culture was 3.4 days; it increased to 4.5 and 5.6 days for the 20th and the 40th sub-cultures, respectively. The LT50 values after passage of the 40th sub-culture on H. armigera decreased to 4.4 and 3.7 days for the 40th (first in vivo) and the 40th (fifth in vivo) passages, respectively. Similarly, the LC50 of M. anisopliae towards third instar larvae of H. armigera increased from the first sub-culture (0.17×104) to (3.0×104) for the 40th conidial transfers on potato dextrose agar and again decreased to 0.74×104 and 0.23×104 in the 40th (first in vivo) and the 40th (fifth in vivo) passage, respectively. Similar trends for LC50 and LT50 values were seen when sugarcane woolly aphid, Ceratovacuna lanigera Zehntner was used as a host. Significant variation in appressorium formation and cuticle-degrading enzyme production such as chitinase, chitin deacetylase, chitosanase and protease during subsequent sub-culturing and passage through H. armigera was observed. Though there was no effect on internal transcribed spacer (ITS) sequence pattern, interestingly, in randomly amplified polymorphic DNA (RAPD), significant differences in the band intensities and in the banding pattern for different sub-cultures of M. anisopliae were observed. As stable virulence towards the insect pest is desirable for commercialisation of a mycoinsecticide, such changes in virulence due to repeated in vitro transfer need to be monitored and minimised.  相似文献

The brown planthopper, Nilaparvata lugens, is one of the most harmful insect pests of rice crops in Asian countries. To find an effective biological control agent against this pest, we investigated the bacterial flora of field N. lugens collected from Jiangsu Province, China, in 2012 and tested its insecticidal activity. A novel bacterium strain, S-JS1, was isolated from N. lugens nymphs and adults and showed a high level of insecticidal activity. Based on its phenotypic, physiological and biochemical properties, and its 16S rRNA gene phylogeny, the isolate was assigned to Serratia marcescens; the name S. marcescens S-JS1 is proposed. The pathogenicity of S-JS1 against the third-instar nymphs, and the macropterous and the brachypterous adults of N. lugens were compared. The median lethal concentration (LC50) values of S-JS1 against the brachypterous adult were the lowest (LC50, 1.53?×?108 colony forming units (cfu)/ml), followed against the macropterous adult (LC50, 1.65?×?109?cfu/ml) and third-instar nymphs (LC50, 1.86?×?109?cfu/ml) at 5 days post-infection. The median lethal time values of 8?×?108?cfu/ml S. marcescens S-JS1 against the brachypterous adult, macropterous adult, and third-instar nymph were 4.5, 5.5, and 5.7 days, respectively. These results indicate that the S-JS1 isolate appears to be a promising S. marcescens strain with strong biocontrol potential against N. lugens.  相似文献

The diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae) is one of the most important pests of cruciferous plants throughout the world. In recent years, this insect has been a serious pest for cabbage fields in Tehran province. Resistance of P. xylostella to all main groups of insecticides has been recorded and it is ranked in the 20 most resistant pest species reported up to now. According to many researchers, to eliminate the problem of pest resistance to chemical pesticides, an integrated pest management programme should be used. In line with this, the uses of microbial control agents (MCAs) are discussed. The bacterium, Bacillus thuringiensis (Bt) is one of microbial control agents of pests. It is characterised by its ability to produce proteic crystalline inclusions during sporulation. Cry1 protein has insecticidal activity and is highly specific to certain insects and not toxic to unrelated insects, plants or vertebrates. In this work, the pathogenicity of some Bt isolates, including Dipel, 20, 29, 79 and 87, was tested against P. xylostella and the lethal concentrations (LC50) of their crystal proteins to P. xylostella third larval instar was determined. The experiment was designed in factorial in randomised complete design with 5 treatments (different concentrations including 104, 105, 106, 107, 108 CFU/ml and 5 replications and with 10 third larval instars. Spore–crystal complex was applied to the surface of natural diets (cabbage leaves) and the mortality of P. xylostella larvae was assessed 120?h after exposure of Bt toxin in each treatment. Results showed that percentage of survival was significantly higher for control treatment. Results also showed that after 5?days, LC50 for isolates of Dipel, 20, 29, 79 and 87 were equal to 1?×?106, 1?×?105, 5?×?105, 4?×?105 and 1?×?104 CFU/ml, respectively. LT50 were equal to 93.71, 48.04, 71, 40.49 and 75.28?h. Leptinotarsa decemlineata is a serious potato pest throughout the world that has developed resistance to many insecticides. This study investigated the virulence of two indigenous isolates of Beauveria bassiana, consisting of DEBI007 and IR1217C. Five different concentrations of each isolate, 1 × 107, 3.1 × 107, 1 × 108, 3.1 × 108 and 1 × 109 conidia ml?1, were applied in bioassay using dipping method on second instar larvae. Control insects were treated with distilled water containing 0.01% Tween-80 (Sigma, Germany) as surfactant. The results showed that mortality percentages due to DEBI007 and IR1217C isolates in the highest concentration were 60% and 57.77%, respectively. There was a significant difference between efficacies of different conidial concentrations; therefore, with rise in conidial concentration, the mortality percentage was increased. The DEBI007 isolate that was previously isolated from the soil was more effective than IR1217C isolate. Calculated LC50 for this effective isolate was 1.4 × 107 conidia ml?1, which, respectively, is the highest concentration.  相似文献

We tested the combined effect of the fungus Beauveria bassiana and the microsporidium Nosema pyrausta on the European corn borer larvae, Ostrinia nubilalis, in the laboratory. The first instar of O. nubilalis larvae was the most sensitive to the B. bassiana infection followed by the fifth, second, third, and fourth instar (LC50s were 4.91, 6.67, 7.13, 9.15, and 6.51 × 105 conidia/ml for the first to fifth instars, respectively). Mortality of each instar increases positively with concentration of conidia. When B. bassiana and N. pyrausta were used in combination, mortality increased significantly in all instars. Relative to the B. bassiana treatment alone, the B. bassiana + N. pyrausta treatment decreased the LC50s by 42.16%, 37.63%, 21.60%, 27.11%, and 33.95% for the first to fifth instars, respectively. The combined effects of the two pathogens were mostly additive. However, at the two highest concentrations the pathogens interacted synergistically in the first and second instar. Individuals that survived the B. bassiana and B. bassiana + N. pyrausta treatments and developed into adults had significantly shorter lifespans and females oviposited fewer eggs than non-exposed insects. The effects on the longevity and the egg production were most pronounced at high concentration of B. bassiana conidia.  相似文献

An indigenous betabaculovirus (PapyGV) of the Pandemis leafroller, Pandemis pyrusana (Kearfott), was studied in the laboratory and greenhouse to determine how the virus affected leafroller mortality and foliar damage. Probability of mortality increased with virus concentration as observed after 7 and 10 days of feeding on virus treated diet in neonates and second instar larvae. LC50 estimates for neonates at 7 and 10 days was 2743 and 389 occlusion bodies (OBs)/mm2. For second instars, LC50 was 139,487 and 813 OBs/mm2 at 7 and 10 days. There was no biologically significant mortality response to increasing virus concentrations by fourth instar larvae; however, when fourth instar larvae were infected with virus on diet and then fed apple leaves, the leaf area consumed declined up to 50% with higher virus concentrations. In a greenhouse study, neonate larvae that fed on seedlings treated with water showed >90% survival and 80% pupation rate of larvae after being transferred to diet. In contrast, larvae that fed on apple seedlings sprayed with 3×106 OBs/ml showed poor survival when transferred to diet after acquiring the virus and failed to reach the pupal stage. This virus shows promise for population regulation and can produce reduction in feeding damage.  相似文献

Aphids (Homoptera: Aphididae) are sap-sucking insect pests that feed on several plants of agronomical importance. Entomopathogenic fungi are valuable tools for potential aphid control. As part of a selection process, laboratory bioassays were carried with five different concentrations of Aspergillus clavatus (Desmazières), Aspergillus flavus (Link) and Metarhizium anisopliae ((Metschnikoff) Sorokin) spores against the pea aphid, Acyrthosiphon pisum (Harris). Aspergillus isolates induced higher mortalities than M. anisopliae, which is a well-known entomopathogen in the literature. Lethal concentrations (LC50 and LC90) were 1.23 × 103 and 1.34 × 107 spores/ml for A. flavus, 4.95 × 102 and 5.65 × 107 spores/ml for A. clavatus, and 3.67 × 103 and 9.71 × 107 spores/ml for M. anisopliae 5 days after treatment. Mycelia development and sporulation on adult cadavers were observed 48 h after incubation. The intrinsic growth rate of A. pisum decreased with increased spore concentration for all fungal strains, suggesting an increase in pathogen fitness related to a consumption of host resources. In conclusion, Aspergillus species could be useful in aphid control as pest control agents despite their saprophytic lifestyle. This is also to our knowledge the first report of A. clavatus and A. flavus strains pathogenic to aphids.  相似文献

Repellent, antifeedant and toxic effect of crude hexane extract of Ageratum conyzoides were investigated against Helicoverpa armigera. In orientation bioassay, the extract exhibited dose-dependent repellency against neonates. Extract significantly increased the mortality and decreased growth of different larval stages when administrated orally in artificial diet. EC50 value was at 0.11% for larval growth inhibition. Toxicity of the extract was manifested by high mortality of first instar larvae after 7 days of feeding on diet containing 0.05–0.4% of extract with LC50 of 0.17%. Under choice bioassay, extract showed strong antifeedant activity against fifth instar larvae with DI50 of 0.21%. In nutritional bioassay, extract significantly reduced RCR, RGR, ECI and ECD of fifth instar larvae with increased AD. When RGR were plotted against RCR, the growth efficiency of larvae fed on treated diet was significantly lower than the control fed larvae suggesting the antifeedant and toxic effect of extract.  相似文献

The biological activities of two species of bacteria isolated from soil of cotton fields identified as Bacillus subtilis strain NRC313 (BS NRC313) and Bacillus thuringiensis strain NRC335 (BT NRC335) were evaluated against the third larval instar of the cotton leafworm, Spodoptera littoralis (Boisd.). The different entomopathogenic bacteria of BS NRC313and BT NRC335 contained 10 × 108 cell/ml, and caused mortality of 100 and 97.3% for the above mentioned strains, respectively. Concentrations of 2.5 × 108 to 10 × 108 cell/ml of strains BS NRC313 and BT NRC335 were applied to the larvae: LC50 were 3.3 × 108 and 3.9 × 108 cell/ml respectively. The influence of exposure to toxin concentrations manifested in terms of decreasing the adult emergence and prolongation of the generation period. The percentage of larvae that survived and succeeded to pupate increased by decreasing the concentration. The longevity of adult emergence that resulted from larvae treated with Bacillus subtilis were 6.0 ± 0.51 and 9.0 ± 0.63 days at 5 × 108 and 2.5 × 108 cell/ml, respectively compared with 9.8 ± 0.47 in control. The results indicated that Bacillus subtilis was more potent than Bacillus thuringiensis. Field applications of B. thuringiensis, B. subtilis and Reldan achieved 55.6, 67.4 and 89.4% reduction of the cotton leafworm larvae Spodoptera littoralis in clover plants under field conditions.  相似文献

The pathogenicity and virulence of 10 isolates of entomopathogenic fungi from the soil of lodging pens of dairy production units in Aguascalientes, Mexico, on adults of Stomoxys calcitrans and Musca domestica were determined. All isolates were pathogenic when exposed by aspersion to a concentration of 1×108 conidia/ml, causing between 20.3 and 91.7% mortality in S. calcitrans and between 31 and 91.7% in M. domestica at 7 days post-exposure; in S. calcitrans isolates of Beauveria bassiana (Bb114) and Metarhizium anisopliae (Ma135), sensu lato, were the most noteworthy as mortality reached above 90% with an LC50 of 3.5×105 conidia/ml for Bb114, while for Ma135 reached 1.6×104 conidia/ml. In M. domestica Ma134 and Ma135 showed mortality above 90% with an LC50 of 4.3×104 and 1.4×105 conidia/ml, respectively.  相似文献

The Mediterranean fruit fly, Ceratitis capitata Wiedemann (Diptera: Tephritidae), is the major tephritid pest in Morocco. This pest survives in Moroccan forests Argania spinosa and continually invades the nearest agricultural areas. Entomopathogenic fungi are an interesting tool for fruit fly control and hold a useful alternative to conventional insecticides. However, primary selection of effective pathogens should be taken in laboratory condition prior to applying them in the field. Here, we used third late instar larvae of C. capitata to investigate the effectiveness of 15 local Beauveria bassiana isolates. Results showed that all isolates were able to infect the larval stage, producing a large mortality rate in puparia ranging from 65 to 95 % and caused significant reduction in adult emergence. The fungal treatments revealed that the mycosis occurred also in adults escaping infection as pupariating larvae. The percentage of mycosed puparia was highest in strain TAM6.2 (95 %) followed by ERS4.16 (90 %), therefore they were the most virulent. Median lethal concentration (LC50) was studied for five isolates at four concentrations ranging from 105 to 108 conidia ml?1. The results showed that the slopes of regression lines for B. bassiana ERS4.16 (slope = 0.386) and TAM6.2 (slope = 0.41) were the most important and had the lowest LC50 values (2.85 × 103 and 3.16 × 103 conidia ml?1 respectively). This investigation suggests that the soil of Argan forests contains pathogenic B. bassiana isolates and highlights for the first time their potential as biological control toward C. capitata larval stage in Morocco.  相似文献

The potato tuber moth (PTM) Phthorimaea operculella is a critical potato pest. Larvae infest both foliage and tubers and mature larvae pupate in the soil or other safe places. Cordyceps tenuipes, an entomopathogenic fungus, infect lepidopteran pupae. To determine the effectiveness of this fungus as a biocontrol agent for PTM, we evaluated the time-concentration-mortality (TCM) response of PTM pupae to C. tenuipes using the following bioassays: (1) direct immersion in conidial suspensions, (2) incubation in sterilised or (3) unsterilised soilpremixed with conidia, and (4) incubation in unsterilised soil drenched with conidial suspensions to simulate field conditions. Fungal infection caused 100%, 83.3%, 73.3%, and 85.0% mortality of PTM pupae in assays 1–4, respectively. At 108 conidia/mL or conidia/g concentration, assays 1 and 4 had short lethal times (LT50) of 2.2 and 2.6 days compared with 3.7 and 4.8 days for assays 2 and 3, respectively. On day 7 after inoculation, assays 1 and 4 also had low lethal concentrations (LC50) of 1.69 × 103 conidia/mL and 1.10 × 105 conidia/g compared with those of assays 2 and 3, which showed low virulence, with LC50 of 3.50 × 105 and 3.60 × 106 conidia/mL, respectively. Our results demonstrate that C. tenuipes is a promising candidate for PTM biocontrol at the pupal stage. Drenching the soil surface with conidial suspensions may be the most effective method of field application.  相似文献

The chickpea pod borer, Helicoverpa armigera (Hübner) is one of the most destructive pests of India and most of the countries. In this experiment, sub-lethal effects of cypermethrin were evaluated against sixth instar larvae of this pest that had ingested insecticide-treated chickpea pods. The LC50, LC30 and LC10 values of cypermethrin were 80.38, 26.65 and 5.41?mg a.i.?L?1, respectively, against sixth instar. The sub-lethal effects on fitness, eggs, larvae, pupae, adult longevity and reproduction were observed in H. armigera population that survived exposure to the sub-lethal doses of LC50, LC30 and LC10 of cypermethrin. Survivorship was reduced to 36?days if H. armigera exposed to the sub-lethal dose of LC50, whereas it was 42?days in unexposed groups. Larvae treated with LC50 dose exhibit lowest intrinsic rate of increase (rm) (0.0413 females/female/day) and highest (0.0517) with the unexposed group. Time taken by H. armigera to complete one generation (Tc) was 40.09?days, while this time significantly lowered to 32.85?days when exposed to the sub-lethal dose of LC30. Hatching eggs were greatly reduced in the larvae exposed to cypermethrin at all the sub-lethal doses. About 430 eggs/female/generation were recorded after treatment with 80.38?mg/L as compared with untreated females (1390). Larvae that were exposed to sub-lethal concentrations of cypermethrin exhibited lower pupal weight and prolonged pupal developmental times compared control larvae. The overall fitness performance was better in the unexposed population as compared to the population exposed to sub-lethal doses.  相似文献

The virulence of 8 isolates of entomopathogenic hyphomycetes against adult and 5th instar nymph of Clavigralla tomentosicollis was evaluated in the laboratory at 4 different concentrations of inoculum. At all concentrations, Beauveria bassiana CPD 9 and Metarhizium anisopliae CPD 5 caused the highest mortality in adult bug ranging from 58 to 97% and 53 to 100%, respectively at 7 days post inoculation. The same isolates had the shortest LT50 (3.5 and 4.1 days, respectively) and the lowest LC50 (1.8 × 105 and 9.8 × 104 conidia ml-1 values in adult insects. In nymphs, M. anisopliae CPD 5 was the most virulent isolate causing mortality of between 43 to 92% with the shortest LT50 of 2.7 days and the lowest LC50of 4.6 × 105 conidia ml-1 which however did not differ significant from LC50 observed in B. bassiana CPD 9 isolate at 5 days post inoculation. A significant reduction in feeding in both developmental stages treated with fungi was observed at 2 days after treatment with the greatest reduction occurring in insects treated with B. bassiana CPD 9 and M. anisoplia CPD 5. In adult insects treated with these isolates, some bugs ceased feeding 24 h before death. When these two isolates were compared in caged experiment with an untreated control using a susceptible, tolerant and moderately resistant variety of cowpea, percentage pod and seed damage were significantly lower in fungal treated cages than in the control cages on all varieties tested. Grain yield per plant was also significantly higher in fungal treated cages than in the control cages on all varieties. The performance of M. anisopliae CPD 5 was however superior to B. bassiana CPD 9. Application of the fungi on moderately resistant variety of cowpea was found to enhance the performance of the pathogen. Effect of deltamethrin (Decis 2.8% EC) on the fitness of Helicoverpa armigera (Hübner) was determined using demographic studies. Survivorship (Ix) and expectancy (ex) was high with commencement of age and gradually decreased with progression of age in the exposed and unexposed groups. Highest number of unhatched eggs (~20%) was recorded with LC50 (79.0 mg/L) opposed to 1% in the control. Daily fecundity rate was significantly greater (200 females/day) in the control than 126 in LC20 (5.13 mg/L). Potential fecundity was lowest (179 females/female/generation) with LC50 treatment than 675 in the control. Smallest value of intrinsic rate of increase (rm) occurred at LC50 (0.0393 females/female/day) while the highest value (0.0539) was at control. Overall larval development was significantly reduced to 18.0 days with LC50 opposed to 24.1 days at control. Pupal period was shortened to 7.6 days with LC50, while it recorded 9.3 days in control.  相似文献
