为了明确来自湖北武穴、孝感、随州3个不同地区的转cry1Ab/cry1Ac、cry2A、cry1C基因明恢63(分别命名为TT51、T2A-1和T1C-19)对赤拟谷盗Tribolium castaneumHerbst生长发育的影响,在室内以不同的转Bt基因稻谷继代饲养赤拟谷盗6代,结果表明:各处理赤拟谷盗的卵期3~4d,卵孵化率85%~100%,幼虫期22~27d,化蛹率85%~97%,蛹期6~7d,百蛹重0.25~0.31g,性比0.8~1.4,羽化率82%~97%,产卵前期5~7d。不同抗虫转基因水稻对赤拟谷盗各发育历期及生命表参数没有显著的影响,同一转基因事件,没有因为种植地区的不同造成对赤拟谷盗生长发育的差异。3种Bt蛋白在继代饲养的赤拟谷盗幼虫体内检测均呈阳性反应,但积累量很小,继代饲养后,没有发现在赤拟谷盗体内明显的累积。  相似文献

害虫细胞色素P450基因可被杀虫剂迅速诱导,然而当前对不同杀虫剂处理下赤拟谷盗P450基因诱导表达特性的研究较少。本研究首先通过序列比对选取了来自不同家族的8个赤拟谷盗P450基因CYP4G7、CYP4Q4、CYP4BR3、CYP12H1、CYP6BK11、CYP9D4、CYP9Z5和CYP345A1,然后采用四种不同杀虫剂氯氰菊酯、氟氯氰菊酯、氯菊酯和吡虫啉对赤拟谷盗20 d幼虫进行生物测定,再根据生测结果以四个药剂亚致死剂量分别处理幼虫,并采用荧光定量PCR分析8个P450基因的表达特性。结果表明,CYP4G7和CYP345A1可以分别被氯氰菊酯(分别上调1.97倍和2.06倍)、氟氯氰菊酯(2.00倍和2.03倍)和氯菊酯(1.73倍和1.81倍)显著诱导,而CYP4BR3和CYP345A1可以被吡虫啉(分别上调1.99倍和1.93倍)显著诱导。本研究结果表明赤拟谷盗P450基因的显著诱导与基因家族类型以及农药品种有关。  相似文献

在30 ℃、75 %相对湿度条件下研究种间竞争对玉米象(Sitophilus zeamais)、谷蠹(Rhizopertha domini-ca)、赤拟谷盗(Triboliumcastaneum)和锈赤扁谷盗(Cryptolestes ferrugineus)4种主要储粮害虫种群动态的影响,并对种群动态进行回归分析。结果表明,玉米象与谷蠹、赤拟谷盗与锈赤扁谷盗混合饲养种群增长均受到显著抑制,玉米象和谷蠹对赤拟谷盗和锈赤扁谷盗的种群增长具有明显的促进作用,赤拟谷盗和锈赤扁谷盗对玉米象和谷蠹的种群增长具有一定的抑制作用。回归分析结果表明玉米象种群最大增长潜能最大,锈赤扁谷盗最小,种群增长率变化规律不明显。  相似文献

研究结果表明,应用二氯苯醚菊酯、氯氰菊酯和氰戊菊酯的3种微胶囊剂对赤拟谷盗、豆蚜、小菜粉蝶和粘虫室内测定及大田试验药效均为显著。处理后6天,死亡率90%左右,各天药效相当或略高于乳油。其残效期对赤拟谷盗24天,对其他3种试虫15天左右,相当于该类药剂乳油残效期的2倍。3种微胶囊剂的药效高低顺序依次为氯氰菊酯>二氯苯醚菊酯>氰戊菊酯,和其乳油效果高低的顺序基本相同。  相似文献

应用粉剂药膜法,一种食品级惰性粉——4号粉相应剂量处理赤拟谷盗Tribolium castaneum(Herbst)、烟草甲Lasioderma serricorne(Fabricius)、锯谷盗Oryzaephilus surinamensis(Linnaeus)的卵和1龄幼虫,赤拟谷盗、锯谷盗、烟草甲的卵平均孵化率均在97%以上,处理后卵孵化的1龄幼虫平均死亡率均在96%以上;处理3种害虫的1龄幼虫其平均死亡率均在99%以上。结果表明食品级惰性粉对3种试虫卵的孵化率几乎没有影响,但对其1龄幼虫有较好的防治效果。通过混粮法应用正交试验研究发现,惰性粉剂量和小麦含水量对赤拟谷盗F1代防治效果均有显著影响。当小麦处于安全水分时(含水量12%14%),10014%),100150 mg/kg 4号粉能有效防治赤拟谷盗F1代。  相似文献

水菖蒲活性物质β-细辛醚对四种储粮害虫的熏蒸活性   总被引:2,自引:0,他引:2  
植物性次生物质在植物-害虫的关系中起着非常重要的作用, 植物中的一些成分对害虫具有熏蒸、触杀和驱避等作用。水菖蒲Acorus calamus L.是一种常用中药, 它的主要杀虫活性成分为β-细辛醚。本研究通过室内生测试验研究了水菖蒲根茎提取物β-细辛醚对玉米象Sitophilus zeamais Motschulsky、谷蠹Rhyzopertha dominica (Fabricius)、赤拟谷盗Tribolium castaneum (Herbst)和四纹豆象Callosobruchus maculatus (Fabricius) 4种储粮害虫的熏蒸击倒和致死作用。结果表明: β-细辛醚对4种试虫的熏蒸击倒和致死作用明显。以50 μL/L的浓度处理120 h后, 对玉米象、谷蠹和四纹豆象的击倒作用均达到100%, 而对赤拟谷盗击倒率为50%; 玉米象、谷蠹和四纹豆象的死亡率分别为81.23%, 97.78%和100%, 而赤拟谷盗死亡率仅为8.89%。处理24 h, β-细辛醚对玉米象、谷蠹、赤拟谷盗和四纹豆象的KC50分别为49.38, 102.96, 124.04和1.07 μL/L; 处理120 h, β-细辛醚对玉米象、谷蠹、赤拟谷盗和四纹豆象的LC50分别为17.82, 4.42, 116.48和0.73 μL/L。结果显示水菖蒲根茎提取物β-细辛醚对4种储粮害虫均具有明显的熏蒸效果, 具有开发为储粮害虫熏蒸剂的潜力。  相似文献

赤拟谷盗和杂拟谷盗是两个同域发生的近缘储粮害虫。为明确它们的生殖隔离程度和机制,本研究比较分析了赤拟谷盗和杂拟谷盗雄虫对同种和异种雌虫的交配选择;赤拟谷盗与杂拟谷盗交配后,测定了精子在异种雌体内的存活情况;将赤拟谷盗和杂拟谷盗进行正反杂交,研究其F1代、F2代和回交代杂种是否产生及其雌雄比。结果显示,赤拟谷盗和杂拟谷盗雄虫对同种雌虫的爬跨率和交配率高于其对异种雌虫的爬跨率和交配率,表明赤拟谷盗和杂拟谷盗种间性隔离不完全;赤拟谷盗和杂拟谷盗交配后精子在异种雌体内是存活的,杂交所产的F1代卵为受精卵,说明交配后完成了精子传送和受精过程,表明种间机械隔离和配子隔离不完全;赤拟谷盗和杂拟谷盗的杂种F1代自交和回交产生了F2代和回交代,表明种间不存在杂种不育;一些杂交组合产生的F2代和回交代数量少且存在雌雄性比偏离,表明种间存在部分杂种不活和杂种衰败。本研究明确了赤拟谷盗和杂拟谷盗的生殖隔离机制,这对于揭示它们的物种进化关系有着重要意义。  相似文献

储粮书虱Liposcelis spp.和锈赤扁谷盗Cryptolestes ferrugineus(Stephens)由于对磷化氢具有较高的抗性而成为目前粮食储藏过程最难防治的两类害虫,研究储粮防护剂和磷化氢联用对储粮书虱和锈赤扁谷盗的防治效果,对于缓解其对磷化氢的抗性以及其在实仓中综合防治具有重要的经济意义。本文用55%甲基嘧啶磷乳油和2%溴氰·甲嘧磷粉剂分别和磷化氢联用对广东、广西、安徽和河南4个不同气候条件的粮库中的书虱和锈赤扁谷盗进行防治效果的研究。结果表明,在磷化氢熏蒸粮食之前用55%甲基嘧啶磷乳油或2%溴氰·甲嘧磷粉剂处理粮面表层的粮食,在4个粮库的试验粮仓内书虱和锈赤扁谷盗的防治效果有显著差异:在北方干燥少雨的河南汤阴库防治效果最好,可达100%,其次是安徽舒城库、广西柳州库,防治效果分别为71.8%和71.9%。效果最差的是高温高湿的是广东湛江库,防治效果为50.4%。用55%甲基嘧啶磷乳油或2%溴氰·甲嘧磷粉剂和磷化氢联用对书虱和锈赤扁谷盗具有很好的防治效果,说明储粮防护剂和磷化氢联用对于储粮书虱和锈赤扁谷盗具有更好的防治效果。  相似文献

测定以植物提取物异硫氰酸酯为主要成分而复配的宏劲杀虫剂对玉米象Sitophilus zeamais(Motschulsky)、赤拟谷盗Tribolium castaneum(Herbst)和谷蠹Rhizopertha dominica(Fabricius)3种重要储粮害虫的熏蒸效果及其对菜青虫Pieris rapae(L.)、红蜘蛛Tetranychus cinnabarinus(Boisduval)的杀虫效果。结果表明,宏劲杀虫剂48h对玉米象、赤拟谷盗和谷蠹的LC50分别是0.016,0.009和0.009μL/mL,均明显低于常规熏蒸剂磷化铝的要求处理剂量;对菜青虫和红蜘蛛的LC50分别是0.0159和1.3738mg/mL,其防效均优于对照药剂3%阿菊乳油和3%新型水分散性颗粒剂。  相似文献

测定了杠柳 (PeriplocasepiumBunge .)其根皮乙醇粗提液对菜青虫Pierisrapae (L .)的拒食活性 ,并进行了乙醇粗取液及其与氯氰菊酯混用防治菜青虫药效试验。生测结果表明 ,乙醇粗提液对菜青虫 4龄和 5龄幼虫具有较强的拒食活性 ,2 4h拒食中浓度分别为 1 3 2 0 8mg L和 1 1 96 3mg L ,且对拒食后试虫生长发育有严重的影响。药效试验结果显示 ,乙醇粗提液 1 0 0倍液及其混配制剂 1 0 0 0倍液对菜青虫具有较好的防治效果。温室处理后 5d ,杠柳乙醇粗提液 1 0 0倍液、混剂 1 0 0 0倍液的防虫效果与 1 0 %氯氰菊酯 1 0 0 0倍液效果相当 ;田间处理后 5d ,杠柳粗提液防虫效果虽然较差 ,但保叶效果仍达 60 %以上 ,而混剂的防虫和保叶效果均与氯氰菊酯效果相当。  相似文献

分别在2005年和2006年的5、6月份,在Kamalpur和Hunterpara茶园、孟加拉西部以及印度地区对一种昆虫致病真菌球孢白僵菌对油桐尺蛾Buzura (=Biston) suppressaria Guen.的生物效力进行了评估。以农场主常用的化学杀虫剂40% SP灭多虫和25% EC氯氰菊酯作为标准检测物。试验中球孢白僵菌浓度为1.50 g,2.00 g和2.5 g/lit. of water;灭多虫和氯氰菊酯浓度分别为0.75 mL,1.00 mL和1.50 mL,和0.50 mL,1.00 mL和1.50 mL/lit. of water。在喷洒2.5 g/lit. of water 球孢白僵菌3天后,Kamalpur和Hunterpara茶园中油桐尺蛾种群个体数分别降低了88.00%和84.00%。死的毛虫变成黑色,垂悬在叶片上。并且发现球孢白僵菌的杀虫活性与高剂量的灭多虫和氯氰菊酯的相当。  相似文献

Larval packet test was used for detection of resistance levels against cypermethrin and deltamethrin, the most commonly used synthetic pyrethroids, in the multi-host tick Hyalomma anatolicum collected from district Moga, Punjab (India). Results indicated the presence of level I resistance against deltamethrin (RF = 2.81), whereas the tick isolate was susceptible to cypermethrin (RF = 0.2). The aqueous and ethanolic extracts of leaves of Cymbopogon winterianus, Vitex negundo and Withania somnifera along with roots of Vitex negundo were assessed for their acaricidal activity against the larvae of deltamethrin resistant H. anatolicum. The efficacy was assessed by measuring per cent larval mortality and determination of LC50 values. The various ethanolic extracts produced a concentration dependent increase in larval tick mortality, whereas the aqueous extracts exhibited a much lower mortality. The highest mortality (93.7 ± 0.66 %) was observed at the 5.0 % concentration of ethanolic extract of leaves of C. winterianus and the lowest LC50 value (0.011 %) was recorded for ethanolic extracts of leaves of V. negundo. The results indicated that these plant extracts have potential to be developed as herbal acaricides.  相似文献

从生活垃圾堆肥中提取有益微生物菌种,配成不同浓度的复合微生物菌剂(CM),施入草坪基质,研究了复合微生物菌剂对干旱胁迫下草坪植物生理生态特性的影响。结果表明:在干旱胁迫下,接种过复合微生物菌剂的草坪植物叶片的丙二醛(MDA)含量显著低于未接种菌剂的对照,超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性显著高于对照。不同草坪植物品种生理生态特性在接种CM后对干旱胁迫的响应程度不同。即高羊茅(Festuca arundinacea L.),当接种200倍稀释液的菌剂,叶片SOD、POD和CAT活性达到最大,分别是对照的8.13、1.53和2.46倍;而黑麦草(Lolium perenne L.)则当接种100倍稀释液的菌剂,POD和CAT活性分别高出对照64.4%和56.1%,而SOD活性是对照的6.50倍。和对照相比,各接种菌剂处理的脯氨酸(Pro)含量、可溶性蛋白含量明显降低,离体叶片持水力却保持了较高的水平。上述结果表明,接种复合微生物菌剂后,植物能够通过自身的保护酶活性和渗透调节物质含量来减轻干旱伤害,维持植物体的正常生理代谢功能,从而有效缓解干旱胁迫对草坪植物的伤害,提高草坪植物的抗旱性,为干旱环境草坪植物的建植提供依据。  相似文献

The characteristics of sucrose-phosphate synthase (SPS; EC 2.4.1.14) activity in leaves of Phaseolus vulgaris L. cv. Linden was studied in plants subjected to water stress and various CO2 and light treatments. When water was withheld for 3 days causing mild water stress (–0.9 MPa), the activity of SPS measured in crude extracts was reduced ca 50%. The effect of water stress was most evident when the enzyme was assayed with saturating amounts of its substrates fructose 6-phosphate and UDP glucose. Placing a water-stressed plant in an atmosphere containing 1% CO2 reversed the effect of water stress on SPS activity over 5 h even though the water stress was not relieved. Holding unstressed leaves in low CO2 partial pressure reduced the extractable activity of SPS. After 1 h of low CO2 treatment the effect of low CO2 could be reversed by 20 min of 5% CO2. However, after 24 h of low CO2 treatment, less SPS activity was recovered by the 20 min treatment. The cytosolic protein synthesis inhibitor cycloheximide prevented the slow recovery of SPS activity, but did not affect the rapid recovery of SPS. We conclude that the effect of water stress on SPS activity was a consequence of the inhibition of photosynthesis caused by stomatal closure. Responses of Phaseolus vulgaris SPS to light were similar to the response to low CO2 in that the effects were most pronounced under Vmax assay conditions. This is the first report of this type of light response of SPS in a dicotyledonous species.  相似文献

The distribution of nitrite reductase (EC 1.7.7.1) and sulfite reductase (EC 1.8.7.1) between mesophyll ceils and bundle sheath cells of maize ( Zea mays L. cv. Seneca 60) leaves was examined. This examination was complicated by the fact that both of these enzymes can reduce both NO-2 and SO2-3 In crude extracts from whole leaves, nitrite reductase activity was 6 to 10 times higher than sulfite reductase activity. Heat treatment (10 min at 55°C) caused a 55% decrease in salfite reductase activity in extracts from bundle sheath cells and mesophyll cells, whereas the loss in nitrite reductase activity was 58 and 82% in bundle sheath cells and mesophyll cell extracts, respectively. This result was explained, together with results from the literature, by the hypothesis that sulfite reductase is present in both bundle sheath cells and mesophyll cells, and that nitrite reductase is restricted to the mesophyll cells. This hypothesis was tested i) by comparing the distribution of nitrite reductase activity and sulfite reductase activity between bundle sheath and mesophyll cells with the presence of the marker enzymes ribulose-l, 5-bisphosphate carboxylase (EC 4.1.1.39) and phosphoe-nolpyruvate carboxylase (EC 4.1.1.32), ii) by examining the effect of cultivation of maize plants in the dark without a nitrogen source on nitrite reductase activity and sulfite reductase activity in the two types of cells, and iii) by studying the action of S2-on the two enzyme activities in extracts from bundle sheath and mesophyll cells. The results from these experiments are consistent with the above hypothesis.  相似文献

稻水象甲在福建省霞浦县长春镇田间种群发生动态的调查结果表明:5月中旬是稻水象甲越冬代成虫迁入稻田为害高峰期,5月下旬成虫种群数量迅速降低,并有少量幼虫开始为害水稻根部;6月初是幼虫发生为害高峰期,蛹高峰期出现在6月中旬,并开始有一代成虫羽化。药剂筛选试验结果表明:20%丁硫克百威乳油、48%毒死蜱乳油、40%三唑磷乳油、5%锐劲特悬浮剂对稻水象甲成虫具有良好的田间控制效果,药后7d防效均在92.31%—100%;5%丁硫克百威颗粒剂、15%乐斯本颗粒剂、3%呋喃丹颗粒剂施药后7d对稻水象甲幼虫的防效90.03%—93.56%。化学综合防治示范区水稻分蘖数和株高均显著大于对照区。  相似文献

棉田盲蝽Lygocoris lucorum已成为棉田的主要害虫。通过2年系统观察在不同时段、不同天气肓蝽的分布规律,结果显示,棉盲蝽有雨后群集在嫩尖危害的习性,建议在雨后的第2天早上选用40%毒死蜱乳剂1500倍液或2·5%的三氟氯氰菊酯乳剂2000倍液进行顶心扣打,可使防效提高58·3%。  相似文献

Forty-two extracts of folk medicinal plant organs from Pakistan were tested in competition binding assays for their interaction with the specific ligand recognition sites on the human alpha2-adrenoceptor subtypes alpha2A, alpha2B and alpha2C Strong binding of the extracts (40 mg/ml) from Acacia nilotica (L.) Delile leaves (88-98% displacement of radiolabel) and Peganum harmala seeds (89-96% displacement) on three subtypes prompted us to extract these plant materials with 40% and 80% methanol, ethanol, and acetone. The extraction results indicated an absence of alpha2-adrenoceptor binding activity in the stalk of A. nilotica and A. tortils, whereas the leaves of both plants contained activity. The extracts of A. nilotica leaves showed a slight, but consistent, preference for the alpha2C-adrenoceptor, whereas the leaves of A. tortils were slightly more active on the alpha2B subtype. The extract of P. harmala stalks was less active than that of its seeds. The binding activities of A. nilotica leaves and P. harmala seeds were mainly concentrated in the water and 30% methanol fractions and further sub-fractions. In a functional activity assay, the active fractions inhibited epinephrine-stimulated 35S-GTPyS binding, thus indicating a predominantly antagonistic nature of the compounds with alpha2-adrenoceptor affinity in these fractions. Among the known major alkaloids of P. harmala (demissidine, harmaline, harmine, 6-methoxyharmalan, and norharmane), only 6-methoxyharmalan showed moderate affinity (dissociation constant (Ki) of 530 +/- 40 nm for alpha2A subtype). This study is a first systematic attempt towards the discovery of potential drug candidates from these plant materials for treating alpha2-adrenoceptor related diseases.  相似文献

Seven Hawthorn extracts were tested in isolated guinea pig aorta rings. The effect on noradrenaline- (10 microM) induced contraction was investigated. The extracts were prepared using ethanol (40 to 70% v/v), methanol (40 to 70% v/v), and water as the extraction solvents. The aqueous-alcoholic extracts displayed similar spectra of constituents. They were characterised by similar procyanidin, flavonoid, total vitexin and total phenols content and by similar TLC fingerprint chromatograms. The aqueous extract, however, showed a different fingerprint and a noticeably lower concentration of procyanidins, flavonoids and total phenols but a similar total vitexin content. All 7 extracts had a relaxant effect on the aorta precontracted by noradrenaline and led to relaxations to 44 until 29% of the initial values. The EC50 values of the aqueous-alcoholic extracts varied between 4.16 and 9.8 mg/l. The aqueous extract produced a similarly strong maximal relaxation as the other extracts, but the EC50, at 22.39 mg/l, was markedly higher. The results show that Hawthorn extracts with comparable quality profiles were obtained by using aqueous-alcoholic extraction solvents (40 to 70% ethanol or methanol). The extracts exerted comparable pharmacological effects. When using water as the extraction solvent, both, the spectrum of constituents and the pharmacological effect, deviated remarkably. It is thus possible to obtain bioequivalent extracts with comparable effect profiles by using 40 to 70% ethanol or methanol as the extraction solvent.  相似文献
