An O2-evolving Photosystem II subchloroplast preparation was obtained from spinach chloroplasts, using low concentrations of digitonin and Triton X-100. The preparation showed an O2 evolution activity equivalent to 20% of the uncoupled rate of fresh broken chloroplasts, but had no significant Photosystem-I-dependent O2 uptake activity. The preparation showed a chlorophyll ab ratio of 1.9 and a P-700chlorophyll ratio of 12400. Absorption spectra at room temperature and fluorescence emission spectra of chlorophyll at 77 K suggested a significant decrease in Photosystem I antenna chlorophylls in the O2-evolving Photosystem II preparation.  相似文献

The mineral helvite, (Mn4S)(BeSiO4)3, contains discrete tetrahedral Mn4S+6 clusters in which the S?2 is tetrahedrally coordinated and each Mn(II) is in a distorted tetrahedron of one S?2 and three oxygens; the cluster is situated within an encompassing lattice of SiO4?4 and BeO4?6 tetrahedra. Mn4S+6 centers provide an interesting model for comparison to the polynuclear manganese center that is associated with photosynthetic water oxidation. Magnetic susceptibility data between 77 and 298 K have been measured for a natural helvite sample containing principally Mn4S+6 centers but with significant contamination from Mn3FeS+6 and Mn3CaS+6. The data exhibited Curie-Weiss behavior with μeff = 5.969 B.M. and θ = 178.3 K. An analysis of the magnetic susceptibility, based on Van Vleck's formalism, demonstrated the presence of antiferromagnetic coupling, with a coupling constant J = ?5.83 cm?1. Mössbauer spectra of Mn3FeS centers in helvite and of Fe4S centers in the related mineral danalite have also been recorded. Isomer shifts show little temperature dependence and lie in the range 1.23–1.43 mmsec.. This range is typical of tetrahedrally coordinated Fe(II) in several ionic crystals but is significantly above that of Fe(II) in ferredoxins and below that in the [quinone-Fe(II)-quinone] complex of the photosynthetic bacterium,Rhodopseudomonas sphaeroides. Quadrupole splittings are highly temperature dependent, ranging from 2.4 mmsec at 4.2 K to less than 0.5 mmsec at 248 K.  相似文献

Exposure of cobalt (II) carboxypeptidase Aα, [(CPD)Co(II)], to small molar excesses of the oxidizing agent m-chloroperbenzoate rapidly destroys (< 30 sec) both its peptidase and esterase activities in parallel. Concomitantly, the characteristic Co(II) electron paramagentic resonance (EPR) signal is abolished. [(CPD)Co(III)], isolated from the reaction mixture, has the same molecular weight and amino acid composition as [(CPD)Co(II)], contains 0.95 g-atom of Co and 0.01 g-atom of Zn per mole of protein, does not exhibit an EPR spectrum and is catalytically completely inactive towards both peptide and ester substrates. Identical treatment of the native zinc enzyme affects neither its catalytic activity nor its metal content. The reaction of m-chloroperbenzoate with [(CPD)Co(II)] follows saturation kinetics and is prevented by the inhibitor β-phenylpropionate. Furthermore, under the conditions found to oxidize [(CPD)Co(II)] effectively, there is no reaction with Co(II) E. coli alkaline phosphatase. Thus, m-chloroperbenzoate has the characteristics of an active-site directed oxidizing reagent for [(CPD)Co(II)].  相似文献

Consistent with a recent literature report (Repine, J. E. etal. (1981) Proc.Nat.Acad.Sci.USA7?8?, 1001–1003), the release of [3H]-thymine from PM-2 DNA by Fe(II)-H2O2-generated ·OH was suppressed by dimethyl sulfoxide. In contrast, DMSO did not affect [3H]-thymine release mediated by Fe(II)-bleomycin. Under aerobic conditions in the presence of t-butyl phenylnitrone, Fe(II)-BLM produces an epr signal that has been presumed to arise by transfer of ·OH or O2? from the “active complex” of bleomycin to the spin trap. Remarkably, high concentrations (80 mM) of PBN had no effect on the ability of Fe(II)-BLM to solubilize [3H]-thymine, although the ability of authentic ·OH to degrade DNA was completely suppressed under these condition. The suproxide dismutase catalyst tetrakis(4-N-methylpyridyl)porphineiron(III) also failed to suppress BLM-mediated DNA degradation. Moreover, the epr signal observed with 1.6 mM Fe(II)-BLM in the presence of 80 mM PBN was found to be much less intense than that produced by 1.6 mM Fe(II) and 290 mM H2O2, but equivalent in intensity to that obtained with 45 mM Fe(II) and exoess H2O2. We conclude that the fragmentation of DNA produced by Fe(II)-BLM can be due neither to free ·OH nor to O2?. We suggest that DNA degradation is initiated by an “active complex” consisting of BLM, metal and oxygen that functions by abstracting H· from susceptible sites on DNA.  相似文献

The effects of Na+ and Mg2+ on the “dark” level (O level) and light-induced (P level) fluorescence in sucrose-washed spinach chloroplasts were studied. Low concentrations of NaCl (2–10 mM) cause a significant decrease in both the O and P levels in the chlorophyll fluorescence transient. The effect on the O level may reflect changes in the bulk chlorophyll a. At 77 °K NaCl increases the F735F685 emission peak ratio in dark-adapted and preilluminated chloroplasts, but has no significant effect on this ratio in sucrose-washed Photosystem II particles. This evidence is consistent with a sodium-induced excitation-energy distribution in favor of Photosystem I.In the presence of MgCl2, with or without NaCl, there is a slight decrease in the O and P level fluorescence as compared with the salt-free control, but an increase as compared with the NaCl-treated sample. Magnesium appears to override the sodium-induced changes. At low temperatures in chloroplasts and Photosystem II particles, MgCl2 has different effects on the F735F685 ratio apparently depending on the state of the membrane. Magnesium, however, always induces an increase in the F695F685 ratio. These results suggest that magnesium may influence Photosystem II reaction centers as well as energy distribution between the two photosystems.  相似文献

EPR data are used to describe the conformation and identity of the atoms coordinated to Cu(II) in Cu(II)-bleomycin bound to oriented DNA fibers. The fibers were slowly drawn from viscous solutions of Cu(II)-bleomycin-DNA containing one Cu(II)-bleomycin to 200 basepairs. EPR measurements were made at room temperature and 90 K for different orientations of the external magnetic field with respect to the helical axes of the fibers. The g-values (g =2.21, g =2.04) and the hyperfine constant (A =175 G) are consistent with values expected for Cu(II) chelated to a square planar array of ligands. In the oriented fibers, the square planar arrays do not all have the same orientations with respect to the fiber axes. At room temperature the chelated ions have rotational freedom in which the normal to the planar array has almost complete freedom of rotation about axes perpendicular to the DNA fiber axes. The normal maintains an angle of 75° with respect to the axis, in the plane of the basepair, about which it rotates. Nine superhyperfine peaks on the high field side of the EPR spectrum were partially resolved. The number and splitting (12 G) of these superhyperfine peaks indicate that four nitrogen atoms are chelated to Cu(II) in a square planar array. These data on Cu(II)-bleomycin bound to DNA give information on the orientation of the metal-containing portion of bleomycin which lies outside the double helix.  相似文献

The influence of temperature on the rate of reduction of P-680+, the primary donor of Photosystem II, has been studied in the range 5–294 K, in chloroplasts and subchloroplasts particles. P-680 was oxidized by a short laser flash. Its oxidation state was followed by the absorption level at 820 nm, and its reduction attributed to two mechanisms: electron donation from electron donor D1 and electron return from the primary plastoquinone (back-reaction).Between 294 and approx. 200 K, the rate of the back-reaction, on a logarithmic scale, is a linear function of the reciprocal of the absolute temperature, corresponding to an activation energy between 3.3 and 3.7 kcal · mol?1, in all of the materials examined (chloroplasts treated at low pH or with Tris; particles prepared with digitonin). Between approx. 200 K and 5 K the rate of the back-reaction is temperature independent, with t12 = 1.6 ms. In untreated chloroplasts we measured a t12 of 1.7 ms for the back-reaction at 77 and 5 K.The rate of electron donation from the donor D1 has been measured in darkadapted Tris-treated chloroplasts, in the range 294–260 K. This rate is strongly affected by temperature. An activation energy of 11 kcal · mol?1 was determined for this reaction.In subchloroplast particles prepared with Triton X-100 the signals due to P-680 were contaminated by absorption changes due to the triplet state of chlorophyll a. This triplet state has been examined with pure chlorophyll a in Triton X-100. An Arrhenius plot of its rate of decay shows a temperature-dependent region (292–220 K) with an activation energy of 9 kcal · mol?1, and a temperature-independent region (below 200 K) with t12 = 1.1 ms.  相似文献

A cobalt-porphyrin containing protein has been isolated from the sulfate-reducer Desulfovibrio desulfuricans (Norway). This violet-colored protein has a molecular weight of approx. 13,000 daltons and contains 1 cobalt atom/molecule. The apo-protein was estimated to contain 104 amino-acid residues giving a molecular weight of 11,000 daltons. The UV-visible absorption spectrum of the protein exhibiting maxima at 588,418 and 280 nm with a shoulder at 550 nm is characteristic of metalloporphyrin proteins. The molar extinction coefficients of the cobalt-protein at 588, 418 and 280 nm are 31,330 , 64,670 and 17,200 respectively and its absorbance ratio A280A588 is 0.54. The protein is reduced by dithionite giving a blue-colored reduced form. Important spectral modifications of the chromophore occurred during the reduction including a shift of the Soret peak from 418 to 381 nm and a shift of the α band in the opposite direction from 588 to 593.5 nm. The Co-protein was slowly reduced by the hydrogenase from D.desulfuricans under hydrogen in the presence of cytochrome C3. The reported data suggest that the redox states of the cobalt center of this new electron carrier correspond to the Co(III) and Co(II) states.  相似文献

Phosphorylation in vitro of the light-harvesting chlorophyll ab protein complex associated with Photosystem II (LHCII) resulted in the lateral migration of a subpopulation of LHCII from the grana to the stroma lamellae. This movement was characterized by a decrease in the chlorophyll ab ratio and an increase in the 77 K fluorescence emission at 681 nm in the stroma lamellae following phosphorylation. Polyacrylamide gel electrophoresis indicated that the principal phosphoproteins under these conditions were polypeptides of 26–27 kDa. These polypeptides increased in relative amount in the stroma lamellae and decreased in the grana during phosphorylation. Pulse/chase experiments confirmed that the polypeptides were labelled in the grana and moved to the stroma lamellae in the subsequent chase period. A fraction at the phospho-LHCII, however, was unable to move and remained associated with the grana fraction. LHCII which moved out into the stroma lamellae effectively sensitized Photosystem I (PS I), since the ability to excite fluorescence emission at 735 nm (at 77 K) by chlorophyll b was increased following phosphorylation. These data support the ‘mobile antenna’ hypothesis proposed by Kyle, Staehelin and Arntzen (Arch. Biochem. Biophys. (1983) 222, 527–541) which states that the alterations in the excitation-energy distribution induced by LHCII phosphorylation are, in part, due to the change in absorptive cross-section of PS II and PS I, resulting specifically from the movement of LHCII antennae chlorophylls from the PS-II-enriched grana to the PS-I-enriched stroma lamellae.  相似文献

A Photosystem II reaction centre protein complex was extracted from spinach chloroplasts using digitonin. This complex showed (i) high rates of dichloroindophenol and ferricyanide reduction in the presence of suitable donors, (ii) low-temperature fluorescence at 685 nm with a variable shoulder at 695 nm which increased as the complex aggregated due to depletion of digitonin and (iii) four major polypeptides of 47, 39, 31 and 6 kDa on dissociating polyacrylamide gels. The Photosystem II protein complex, together woth the P-700-chlorophylla protein complex and light-harvesting chlorophyll ab-protein complex (LHCP) also isolated using digitonin, were reconstituted with lipids from spinach chloroplasts to form proteoliposomes. The low-temperature (77 K) fluorescence properties of the various proteoliposomes were analysed. The F685F695 ratios of the Photosystem II reaction centre protein complex-liposomes decreased as the lipid to protein ratios were increased. The F681F697 ratios of LHCP-liposomes were found to behave similarly. Light excitation of chlorophyll b at 475 nm stimulated emission from both the Photosystem II protein complex (F685 and F695) and the P-700-chlorophyll a-protein complex (F735) when LHCP was reconstituted with either of these complexes, demonstrating energy transfer between LHCP and PS I or II complexes in liposomes. No evidence was found for energy transfer from the PS II complex to the P-700-chlorophyll a-protein complex reconstituted in the same proteoliposome preparation. Proteoliposome preparations containing all three chlorophyll-protein complexes showed fluorescence emission at 685, 700 and 735 nm.  相似文献

The protein neurotoxin II from the venom of the scorpion Androctonusaustralis Hector was labeled with 125I by the lactoperoxidase method to a specific radioactivity of about 100 μCi/μg without loss of biological activity. The labeled neurotoxin binds specifically to a single class of non intereacting binding sites of high affinity (KD = 0.3 – 0.6 nM) and low capacity (4000 – 8000 sites/cell) to electrically excitable neuroblastoma cells. Relation of these sites to the action potential Na+ channel is derived from identical concentration dependence of scorpion toxin binding and increase in duration and amplitude of action potential. The protein neurotoxin II from the sea anemone Anemona sulcata also affects the closing of the action potential Na+ ionophore in nerve axons. The unlabelled sea anemone toxin modifies 125I-labeled scorpion toxin II binding to neuroblastoma cells by increasing the apparent KD for labeled scorpion toxin without modification of the number of binding sites. It is concluded that both Androctonus scorpion toxin II and Anemona sea anemone toxin II interact competitively with a regulatory component of the action potential Na+ channel.  相似文献

Superior antitumor activity of 1-β-D-arabinofuranosylcytosine (ara-C) conjugates of prednisolone and prednisone against L1210 leukemic mice, based on ara-C content, has encouraged us to synthesize 5′-(cortisone-21-phosphoryl)-1-β-D-arabinofuranosylcytosine (I) and 5′-(cortisone-21-phosphoryl)-1-β-d-arabinofuranosylcytosine (II) by condensation of N4,2′,3′-triacetyl-1-β-d-arabinofuranosylcytosine 5′-monophosphate with cortisol and cortisone in the presence of N,N′-dicyclohexylcarbodiimide at room temperature followed by removing the acetyl groups in 2 N methanolic ammonia in 20% yield. The conjugates I and II inhibited the invitro growth of L1210 by 50% (ED50) at 0.25 μM and 0.07 μM, respectively, while ara-C showed ED50 0.1 μM. However, the conjugates I and II exhibited 287% and 238% of TC at 50 mg/kg/day × 5 doses against L1210 leukemic mice, respectively, while ara-C at 25 mg and 50 mg/kg/day × 5 gave the respective 127% and 110% of TC.  相似文献

Hybrids were constructed between E. coli K12 unc? mutants uncoupled in oxidative phosphorylation, and thus defective in ATP biosynthesis, and an F′ plasmid carrying nitrogen fixation genes from Klebsiella pneumoniae. Examination of these hybrids showed that expression of nif+Kp genes in E. coli K12 does not require coupling of oxidative phosphorylation but needs the contribution of an anaerobic electron transport system involving fumarate reduction. The nifKp cluster of genes does not contain functions able to complement a defective Mg2+-ATPase aggregate but does contain a function(s) which appears to interact with the uncB? mutant over the formation of a redox system.  相似文献

Stellacyanin, the single blue copper protein from Rhusvernicifera, is reduced stoichiometrically by Cr(II)aq ions yielding a 1:1 adduct between the Cr(III) produced and the reduced protein. This Cr(III)-labeled stellacyanin is substitution inert and no significant loss of the label occurs during extensive dialysis for more than a week. Oxidation by O2 of the Cr(III)-labeled Cu(I) stellacyanin does not cause the loss of Cr(III) either. Furthermore, reduction of the Cr(III)-labeled stellacyanin Cu(II) by a second equivalent of Cr(II) may be attained without any further labeling. Thus, the one mole of Cr ions binds to stellacyanin during the first reduction step and is most probably coordinated at a specific locus on that protein.  相似文献

Four procedures utilizing different detergent and salt conditions were used to isolate oxygen-evolving Photosystem II (PS II) preparations from spinach thylakoid membranes. These PS II preparations have been characterized by freeze-fracture electron microscopy, SDS-polyacrylamide gel electrophoresis, steady-state and pulsed oxygen evolution, 77 K fluorescence, and room-temperature electron paramagnetic resonance. All of the O2-evolving PS II samples were found to be highly purified grana membrane fractions composed of paired, appressed membrane fragments. The lumenal surfaces of the membranes and thus the O2-evolving enzyme complex, are directly exposed to the external environment. Biochemical and biophysical analyses indicated that all four preparations are enriched in the chlorophyll ab-light-harvesting complex and Photosystem II, and depleted to varying degrees in the stroma-associated components, Photosystem I and the CF1-ATPase. The four PS II samples also varied in their cytochrome f content. All preparations showed enhanced stability of oxygen production and oxygen-rate electrode activity compared to control thylakoids, apparently promoted by low concentrations of residual detergent in the PS II preparations. A model is presented which summarizes the effects of the salt and detergent treatments on thylakoid structure and, consequently, on the configuration and composition of the oxygen-evolving PS II samples.  相似文献

The evolution of H2 gas in an in vitro illuminated chloroplast plus hydrogenase system was shown to function for six and a half hours at a continuous rate of about 10 μmoles H2/mg. chlorophyll/hour. Chloroplasts from various plant species, using different isolation media, and storage in a fixed state (glutaraldehyde) at 4°, ?5° and ?196° were shown to catalyze H2 production. Both Clostridium and E. coli hydrogenase were used. From the use of the photosystem II inhibitors DCMU and DBMIB and heat inactivation of photosystem II, it was concluded that H2O was the source of electrons for H2 gas production.  相似文献

A cytochrome c - cytochrome c oxidase complex containing 0.8–1.0 moles of cytochrome c per mole of cytochrome c oxidase (heme a + a3) was isolated as described by Ferguson-Miller, S., Brautigan, D.L., and Margoliash E., J. Biol. Chem. 251, 1104 (1976). This complex was reacted with dithiobissuccinimidyl propionate, an 11 Å bridging bifunctional reagent, and the cross-linked products obtained were analyzed by two dimensional gel electrophoresis. Cytochrome c was cross-linked to subunit II of cytochrome c oxidase. Other cross-linked products were formed involving different subunits of cytochrome c oxidase. These included I+V, II+V, III+V, V+VII, IV+VI and IV+VII. Experiments are also described using N,N′-bis(3-succinimidyloxycarbonylpropyl) tartarate. The major product formed with this 18 Å bridging bifunctional reagent was a pair containing II+VI.  相似文献
