正常细胞的朊蛋白(PrPC)代谢和构象的改变是引发动物和人类可传播性海绵状脑病(transmissiblespongiformencephalopathies,TSEs)的根本原因。将羊瘙痒病(scrapie)仓鼠适应株263K颅内接种仓鼠,在接种后的第20、40、50、60、70、80天,通过Westernblot动态检测仓鼠脑中PrP存在的形式。结果在接种后第40天,在感染动物脑组织中即检测到PrPSc分子,比临床症状出现的时间早(平均潜伏期为66 7±1 1天),且无糖基化形式的PrP分子所占百分比在接种后期增加明显。除了标准分子量大小(30kD~35kD)的PrP分子外,在感染动物脑中存在着高分子量和低分子量形式的PrP分子。定量分析显示,随着接种潜伏期的延长,不同形式PrP分子的含量也在增加,其中低分子量形式的PrP分子与临床症状的出现密切相关。蛋白去糖基化实验表明,在感染动物脑组织中,除了标准分子量大小的PrP蛋白外,还存在一条更小分子量的PrP条带,而正常动物脑组织仅存在标准大小的PrP分子。低分子量形式的PrP分子具有与全长PrP分子相类似的糖基化模式。结果提示,scrapie263K感染的仓鼠脑组织中存在不同分子形式的PrPSc,其PrP分子的代谢可能不同于正常动物。  相似文献

为了探讨羊瘙痒因子263K感染仓鼠脑组织中星形胶质细胞和神经元的数量及功能改变,利用免疫印迹、免疫组化方法研究了胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)和神经元特异性烯醇化酶(neuron-specific enolase,NSE)在受染仓鼠各脑区中表达变化的特点,同时比较其与神经病理学改变及PrPSc沉积的关系.结果表明,感染终末期仓鼠的脑组织中GFAP表达量明显增高,与正常对照相比,大脑皮质、脑干和小脑区域GFAP着色细胞数量分别增高3.69、2.41和1.56倍.感染仓鼠脑组织NSE表达量低于正常对照,小脑、海马CA1区和大脑皮质NSE着色细胞数量分别仅为正常对照相应区域的22.5%、54.2%和53.9%.这些变化与PrPSc在脑组织中的沉积程度和空泡样变性程度相吻合.结果提示,GFAP和NSE的检测可分别很好地反映星形胶质细胞和神经元的数量及功能状态,成为在朊病毒病发病过程中重要的病理变化指标.  相似文献

In November 2004 a catastrophic windstorm destroyed a large part of the spruce forest in the Tatra National Park (Slovakia). The majority of the windthrown area was cleared; only a small part was left uncleared, thereby allowing regeneration by natural succession. The aim of the present study was to assess the impact of the different forestry practices on soil Oribatida. Three different stands were selected for the study, where sampling took place in June and October 2006: control forest stands (REF), windthrown stands left for natural development (NEX) and clear-cut windthrown stands (EXT). The mean abundance of Oribatida recorded in REF and NEX stands was significantly higher than in EXT stands. Kruskal-Wallis test of mean abundance of adults as well as juveniles confirmed significant influence of treatment and date. The highest abundance of adults was found in control forest stands (REF). Post hoc multiple comparison proved significantly lower abundance of adults in clear-cut stands (EXT) compared with REF. The mean abundance of adults and juveniles was several times higher in stands left for natural development (NEX) than in EXT stands. The highest species richness was observed in REF, followed by NEX and EXT stands. Ordination method showed differences in species composition between studied treatments. Furthermore, a much lower abundance of Hermannia gibba, a dweller of leaf litter and upper soil layers, was recorded in cleared stands compared to the other stands. Indeed, windthrown stands had an obvious lower species richness than control stands. The ordination method used demonstrated a significant influence of both treatment and sampling date on the abundance and species richness of Oribatida. The present study showed that clear-cutting of wind-damaged spruce forest markedly decreases the abundance of soil Oribatida compared with windthrown forest stands left to natural succession.  相似文献

A closed Lutzomyia longipalpis colony, from Ceará has been used to transmit Leishmania chagasi isolated from a fox in Pará state. The last time this colony was successfully used in similar transmission experiments was eight years (64 generations) ago indicating that this colony of Lu. longipalpis has fully maintained its vectorial capacity in spite of such a long period of maintainance in the laboratory.  相似文献

To evaluate the potential of Bacillus thuringiensis (Bt) Cry1F protein accumulation in soil, transgenic corn containing event DAS-01507-1 encoding the cry1F gene was grown in three field sites for 3 consecutive yr, and the corn plants were incorporated into the soil through postseason tillage or no tillage each year. Soil samples were collected from these fields, and the level of Cry1F protein in these samples was determined using an enzyme-linked immunosorbent assay (ELISA) with a synthetic invertebrate gut fluid as an extraction buffer. The ELISA was validated in soil matrices over the concentration range of 18-180 ng/g dry weight, with a limit of detection of 4.5 ng/g dry weight. The assay was shown to have good accuracy and precision. No detectable Cry1F protein was found in any of the soil samples collected from the Cry1F corn fields. Soil also was bioassayed, and no biological activity was observed against Heliothis virescens neonates. These results indicate that the level of Cry1F protein accumulated in soil after 3-yr continuous planting of transgenic Cry1F corn is negligible.  相似文献

Expression of recombinant protein HSA-AX15(R13K) in Pichia pastoris GS115 strain produced both the intact protein and its two degradation products with molecular weights of around 43kDa and 66.2kDa, respectively. To reduce or avoid the degradation, a modified P. pastoris GS115 stain, in which YPS1 gene was disrupted, was constructed via homologous recombination and used as a host strain for the HSA-AX15(R13K) expression. After 60h of induction during culture, it was found that the degradation product of around 66.2kDa was reduced significantly in the supernatant of yps1-disrupted strain compared with that in the supernatant of wild-type strain. By the Western blot analysis of culture supernatants from wild-type and yps1-disrupted strains expressing HSA-AX15(R13K), the significant improvement was also seen in the degradation product of around 43kDa. Comparison of cell growth between the two strains demonstrated a similar growth tendency, thereby indicating that the disruption of YPS1 gene has no effect on the normal physiology of GS115 strain. Following induction for 60h, the yield of intact HSA-AX15(R13K) in the yps1 disruptant was three-fold higher than that in the wild-type strain. Therefore, such a P. pastoris mutant deficient in YPS1 activity is suitable for the high-level expression of recombinant protein HSA-AX15(R13K).  相似文献

Net pen fish farms generally enrich the surrounding waters and the underlying sediments with nutrients and organic matter, and these loadings can cause a variety of environmental problems, such as algal blooms and sediment anoxia. In this study we test the potential of biofiltration by artificial reefs for reducing the negative environmental impacts surrounding fish farms in the Gulf of Aqaba, Red Sea. Two triangular-shaped artificial reefs (reef volume 8.2 m3) constructed from porous durable polyethylene were deployed at 20 m; one below a commercial fish farm and the other 500 m west of this farm in order to monitor the colonization of these reefs by the local fauna and to determine whether the reef community can remove fish farm effluents from the water. Both reefs became rapidly colonized by a wide variety of organisms with potential for the removal of compounds released from the farms. Within the first year of this study fish abundances and the number of species reached 518–1185 individuals per reef and 25–42 species per reef. Moreover, numerous benthic algae; small sessile invertebrates (bryozoa, tunicates, bivalves, polychaetes, sponges, anemones) and large motile macrofauna (crustaceans, sea urchins, gastropods) settled on the reef surfaces. Depletion of chlorophyll a was measured in the water traversing the artificial reefs in order to assess the biofiltration capacity of the associated fauna. Chlorophyll a was significantly reduced to a level 15–35% lower than ambient concentrations. This reduction was greatest at intermediate current speeds (3–10 cm s–1), but was not influenced by current direction. The reef structures served as a successful base for colonization by natural fauna and flora, thereby boosting the local benthic biodiversity, and also served as effective biofilters of phytoplankton.  相似文献

Ecological research has been repeatedly criticized for its general lack of progress. While it has proved difficult to reveal scientific progress, from a subjective view trends in ecology seems evident. In order to objectively detect principal trends in this broad discipline during the past 25 years (1978–2002), we performed several word and co-word analyses on titles and abstracts of articles that appeared in five core journals of ecology: «Ecology>>, «Oecologia>>, «Oikos>>, «Journal of Animal Ecology>> and «Journal of Ecology>>. A general time gradient in the use of words was evident and words with a positive trend dominated. Trend words derived from article titles were used to perform co-word analyses on abstracts available since 1991. Besides a trend towards the study of processes most important ecological fields found are landscape ecology, biodiversity, ecosystem research, modelling, and food-web/predator–prey interactions. A semantic map of trend words was built by ordination. In the map, words formed a continuous space with the three thematic directions «biotic>>, «abiotic>> and «temporal & spatial>> representing the classic aspects of ecological research. With respect to the obvious trends and features derived from our analyses, we suggest that the results represent real scientific progress in ecology as a pluralistic, multi-aspect and multiple-causation discipline. Such a progress could hardly be observed from details in sub-disciplines.  相似文献

Background

Lassa and Junín viruses are the most prominent members of the Arenaviridae family of viruses that cause viral hemorrhagic fever syndromes Lassa fever and Argentine hemorrhagic fever, respectively. At present, ribavirin is the only antiviral drug indicated for use in treatment of these diseases, but because of its limited efficacy in advanced cases of disease and its toxicity, safer and more effective antivirals are needed.

Methodology/Principal Findings

Here, we used a model of acute arenaviral infection in outbred guinea pigs based on challenge with an adapted strain of Pichindé virus (PICV) to further preclinical development of T-705 (Favipiravir), a promising broad-spectrum inhibitor of RNA virus infections. The guinea pig-adapted passage 19 PICV was uniformly lethal with an LD50 of ∼5 plaque-forming units and disease was associated with fever, weight loss, thrombocytopenia, coagulation defects, increases in serum aspartate aminotransferase (AST) concentrations, and pantropic viral infection. Favipiravir (300 mg/kg/day, twice daily orally for 14 days) was highly effective, as all animals recovered fully from PICV-induced disease even when therapy was initiated one week after virus challenge when animals were already significantly ill with marked fevers and thrombocytopenia. Antiviral activity and reduced disease severity was evidenced by dramatic reductions in peak serum virus titers and AST concentrations in favipiravir-treated animals. Moreover, a sharp decrease in body temperature was observed shortly after the start of treatment. Oral ribavirin was also evaluated, and although effective, the slower rate of recovery may be a sign of the drug''s known toxicity.

Conclusions/Significance

Our findings support further development of favipiravir for the treatment of severe arenaviral infections. The optimization of the experimental favipiravir treatment regimen in the PICV guinea pig model will inform critical future studies in the same species based on challenge with highly pathogenic arenaviruses such as Lassa and Junín.  相似文献

Abstract

The autecology of the Sardinian endemics Aquilegia barbaricina Arrigoni et Nardi and A. nugorensis Arrigoni et Nardi were investigated. Peaks of anthesis and seed dispersal were recorded for five populations occurring in two distinct habitats, one riparian and one rupicolous. Germination tests were carried out on seed lots belonging to each population by sowing seeds at 10, 15, 20, 25 and 25/15°C. In addition, seeds were incubated for 2 months at either 25°C (summer), 5°C (winter) or 25°C for 2 months plus 2 months at 5°C (summer followed by winter–SW), and then moved to the germination temperatures. Embryo measurements were taken during pre-treatments and germination. Experimental seed burials were carried out for two populations of each species. Both species dispersed in summer. The population of A. nugorensis occurring on rocky outcrops differed in phenology from both the other A. nugorensis population from riparian vegetation and from A. barbaricina. Both species showed morphophysiological seed dormancy, with <50% germination under laboratory conditions. All riparian populations germinated only after the SW pre-treatment, while the rupicolous population germinated at 25°C, without any pre-treatment. Low germination percentages were observed in the experimental seed burials, suggesting the ability for both species to form a persistent soil seed bank.  相似文献

Summary Food preference, metal avoidance behaviour and desiccation tolerance were investigated for two collembolan species, Folsomia fimetarioides and Isotomiella minor, in order to test some hypotheses for their different distributions in relation to a heavy metal gradient. F. fimetarioides showed a higher preference for metal tolerant fungi than I. minor when they were offered fungi of different metal tolerance. When they could choose between polluted and unpolluted fungi and substrate, F. fimetarioides significantly avoided the polluted source while I. minor did not. Both species were negatively affected by drought. I. minor was more tolerant than F. fimetarioides when exposed to drought in combination with metal pollution. High supply of preferred fungal species and a better ability to avoid heavy metals favour F. fimetarioides and determine its dominance over I. minor in polluted soils.  相似文献

Although theory suggests that hybrid zones can move or change structure over time, studies supported by direct empirical evidence for these changes are relatively limited. We present a spatiotemporal genetic study of a hybrid zone between Pseudacris nigrita and P. fouquettei across the Pearl River between Louisiana and Mississippi. This hybrid zone was initially characterized in 1980 as a narrow and steep “tension zone,” in which hybrid populations were inferior to parentals and were maintained through a balance between selection and dispersal. We reanalyzed historical tissue samples and compared them to samples of recently collected individuals using microsatellites. Clinal analyses indicate that the cline has not shifted in roughly 30 years but has widened significantly. Anthropogenic and natural changes may have affected selective pressure or dispersal, and our results suggest that the zone may no longer best be described as a tension zone. To the best of our knowledge, this study provides the first evidence of significant widening of a hybrid cline but stasis of its center. Continued empirical study of dynamic hybrid zones will provide insight into the forces shaping their structure and the evolutionary potential they possess for the elimination or generation of species.  相似文献

Nutrient concentrations in the rhizosphere soil can be higher, lower or remain unchanged compared to the bulk soil, but relatively little is known about such changes for basic cations in the rhizosphere of tree roots. A modified root container technique of studying rhizosphere processes was employed. Plexiglas cylinders were horizontally split by a membrane with 30 M mesh size into an upper compartment for root growth and a root-free lower compartment, each with an inner diameter of 5 cm and a height of 10 cm. One 2-year-old Norway spruce (Picea abies) seedling was transplanted from a nursery into each cylinder. Plants were not specifically inoculated, but roots were colonised by a mix of ectomycorrhizal fungi originating from the nursery. The nutrient poor mineral soil used in the experiment was taken from a forest site in Bayerischer Wald, southern Germany. The soil was either supplied with a mix of Ca, Mg and K, or not supplied with these cations. Plants were harvested 30 weeks after transplanting. The nylon membrane between the root compartments restricted root growth to the upper compartment, so that by the end of the experiment a root mat was formed at the top side of the membrane. In the lower compartment, soil nearest to the root mat was regarded as rhizosphere soil while soil in a distance from the root mat was regarded as bulk soil. In the upper compartment, rhizosphere soil was obtained at the end of the experiment by gently shaking the roots. The soils were analysed for Ca, Mg and K contents following two different soil extraction methods. In the fertilised treatment, H2O-extractable Ca and Mg were accumulated in the rhizosphere. In contrast, K (NH4Cl-extraction) was depleted in the rhizosphere. In the bottom tube, the depletion of K (NH4Cl-extraction) was restricted to 1 cm distance from the root mat. In unfertilised soil, Ca, Mg and K concentrations did not differ clearly between rhizosphere and bulk soils. The results indicated that the occurrence of cation gradients in the rhizosphere depended on the level of soil nutrient supply. Distinct rhizosphere effects were measured by conventional soil extraction methods only when the soil was freshly fertilised with mineral elements prior to the experiment. In this case, K depletion in the rhizosphere reflected higher K uptake by the fertilised Norway spruce plants. For low-nutrient soils, novel techniques are required to follow subtle changes in the rhizosphere.  相似文献

A Burkard spore trap was used to monitor daily fungal spore counts in the atmosphere of Melbourne (Australia) between October 1991 and December 1994. Annual total spore counts varied widely (range 345 770 in 1994 to 1 106 037 in 1992), of which approximately half were identified asCladosporium sp. and only about 1%Alternaria sp. Highest daily total spore counts were recorded late-summer through to mid-winter, probably corresponding to senescence of annual grasses and leaf fall. Spore counts were negatively correlated with rainfall (P<0.05), significantly correlated with average temperature (P<0.001), and showed a highly significant linear relationship with cumulative temperature throughout the year.  相似文献

We retrospectively evaluated the epidemiology of onychomycosis and/or paronychia in 172 patients attending the Clinic of Dermatology and Venereology over a 5 year period. Although yeast isolates, belonging to the Candida species, represented the most frequent etiologic agents of these infections, an increasing prevalence of fungal infections due to emerging fungal pathogens (EFP) was noted throughout this time period. In particular, EFP as causative agents of these infections increased from 0 to 28.4% from 1998 to 2002.  相似文献

天然橡胶是重要的国防战略物质,巴西橡胶树( Hevea brasiliensis (Willd. ex A. Juss.) Muell. Arg)是天然橡胶的唯一来源,天然橡胶商业化形式极为单一,潜在的供给不足问题亟待解决。因此,寻找可替代巴西橡胶树的产胶植物一直受到全世界高度重视。蒲公英属橡胶草( Taraxacum kok-saghyz Rodin)根部含有与橡胶树橡胶类似的天然橡胶分子,该植物主要分布在温带和寒带地区,具有易于机械化收获、生长周期短、遗传转化相对容易等特点,是最具开发潜力的产胶植物。本文对橡胶草90年(1931 - 2018)来的研究历史和主要成果进行了概括,对近10年取得的最新成果进行了深度分析,并预测橡胶草在未来天然橡胶产业中的作用,期望为开展橡胶草商业化生产和橡胶生物合成相关基础研究提供一定的参考。  相似文献
