Gas chromatography combustion isotope ratio mass spectrometry (GC-C-IRMS) was used to measure the (13)C/(12)C ratios of PLFAs at natural abundance levels from a temperate grassland nitrogen (N) and phosphorus (P) factorial fertilization experiment in northern Greece. In each plot two rhizosphere samples were derived centred around individual Agrostis capillaris and Prunella vulgaris plants. It was hypothesized that the isotopic signal of microbes that preferentially feed on recalcitrant litter such as fungi would be modified by fertilization more strongly than that of opportunistic microbes using labile C. Microbial community δ(13)C was affected by both P and N fertilization regime and plant species identity. However, we have been unable to detect significant nutrient effects on individual groups of microbes when analyzed separately in contrast to our original hypothesis. Intra-treatment variability, as evaluated from Hartley's F(max) tests in the five first PCA components axes as well as the size of the convex hulls in PCA scoreplots and Mahalanobis distances, was considerably higher in the non-fertilized controls. Moreover, a significant relationship was established between the change in PLFA abundances and their respective changes in δ(13)C for the aggregate of samples and those simultaneously fertilized with N and P. We conclude that use of compound specific isotope analysis in the absence of labelling represents a valuable and overlooked tool in obtaining an insight of microbial community functioning.  相似文献

For many years, the physiological significance of Ca(2+) oscillations has been a matter of debate, but the potential to encode and transduce information in the pattern of an oscillation is obvious. In this review, we only consider transients and oscillations observed during fertilization in plants with the major focus on flowering plants. After presenting data related to algae, fertilization mechanisms in flowering plants are defined as a multi-step phenomenon, starting with pollination during which calcium plays a key role, especially during pollen-stigma interactions (compatible and incompatible reactions). The pollen tube serves as a guide and a pathway for the sperm cells on their course towards their female target cells. For many years, the pollen tube has also been studied as an easily accessible in vitro model to elucidate the role of calcium on tip growth. Finally, in flowering plants, a unique double fertilization system is present. Interesting data obtained from an in vitro fertilization system in maize are presented and discussed. In addition, the new approaches made possible by Arabidopsis and Torenia and their potential limitations are covered.  相似文献

Integrins are involved in a wide variety of cell adhesion processes, and have roles in gamete binding and fusion in mammals. Integrins have been also discovered in the scleractinian coral Acropora millepora (Cnidaria: Anthozoa). As a first step toward understanding the molecular basis of fertilization in corals, we examined the effect of polyclonal antisera raised against recombinant coral integrins on gamete interactions in A. millepora. Antiserum raised against integrin βcn1 dramatically decreased the binding of Acropora sperm to eggs and significantly decreased fertilization rates relative to preimmune serum and seawater controls. However, the antiserum against AmIntegrin α1 did not affect significantly either sperm–egg binding or fertilization. One possible explanation for this is that AmIntegrin α1 may preferentially mediate interactions with RGD-containing ligands, whereas mammalian α6 integrin (which is most directly implicated in gamete interactions) preferentially interacts with laminin-related ligands. Our results suggest that β1 type integrins are involved in the fertilization process in Acropora and that some functions of these molecules may have been conserved between corals and mammals. A. The effects of liquid and solid fertilizers on fine-root development were studied in a 130-year-old Scots pine (Pinus sylvestris L.) stand. Ingrowth cores,viz. initially root-free mesh bags with sieved mineral soil taken outside the plots and driven to a depth of 30 cm from the soil surface, were subsequently resampled and the amount of fine roots was estimated. The total accretion of both fine-root length and dry weight was comparatively high in the liquid fertilization plot. The most substantial net accretion, however, during the 1984 period was in the control plot. The results of the study is that the ramification pattern of the fine roots was strongly influenced by fertilization. The average number of root tips per unit length was 9.9 cm−1 in the control plot cm−1, compared with both the solid (A and B) and liquid fertilization plots (2.3, 3.2 and 3.3 cm−1, respectively) due to a greater occurrence of mycorrhizal aggregates (‘ball mycorrhiza’). The effects of fertilization on the mineral nutrient concentrations in the fine roots were limited—the strongest effects were in the liquid-fertilized area. The observed increase in the concentration of most mineral nutrients in the latter experimental area, however, may be due to a change in the growth pattern of the fine roots rather than an effect of the fertilizers themselves.  相似文献

Abstract

The main objective of the current research was to study the reproductive behaviour of artificial triploid potato hybrids between wild Solanum commersonii and the cultivated potato Solanum tuberosum. When used in 3x × 2x crosses, triploids gave aneuploid progenies with somatic chromosome number ranging from 29 to 36. Fertilization fitness data suggested that the survival rate of gametes produced by the triploid parents may be related to their chromosome number. In addition, consistent with molecular data, our results indicated that fitness of gametes and chromosome number of progenies are influenced by the genome dosage of interspecific triploids. Since a main route to polyploidy formation is via 2n gametes and triploids, our study may contribute to a better understanding of polyploid plant reproduction, evolution and breeding.  相似文献

Polyspermy is a common problem in bovine in vitro fertilization (IVF) and has a still unclear etiology. In this specie, after IVF, despite the lack of a biochemical post-fertilization hardening, the stiffness of the outer ZP layer is significantly increased. Therefore, polyspermy might be related to an incomplete or insufficient stiffening of the ZP. We obtained, by using atomic force spectroscopy in physiological conditions, a complete characterization of the biomechanical changes of the inner and outer ZP layers occurring during oocyte maturation/fertilization and correlated them to the ultrastructural changes observed by transmission electron microscopy using ruthenium red and saponin technique. In both the inner and outer ZP layers, stiffness decreased at maturation while, conversely, increased after fertilization. Contextually, at the nanoscale, during maturation both ZP layers displayed a fine filaments network whose length increased while thickness decreased. After fertilization, filaments partially recovered the immature features, appearing again shorter and thicker. Overall, the observed biomechanical modifications were substantiated by ultrastructural findings in the ZP filament mesh. In fertilized ZP, the calculated force necessary to displace ZP filaments resulted quite similar to that previously reported as generated by bovine sperm flagellum. Therefore, in bovine IVF biomechanical modifications of ZP appear ineffective in hindering sperm transit, highlighting the relevance of additional mechanisms operating in vivo.  相似文献

Restoration of riparian forest productivity lost as a consequence of flow regulation is a common management goal in dryland riverine ecosystems. In the northern hemisphere, dryland river floodplain trees often include one or another species of Populus, which are fast‐growing, nutrient‐demanding trees. Because the trees are phreatophytic in drylands, and have water needs met in whole or in part by a shallow water table, their productivity may be limited by nitrogen (N) availability, which commonly limits primary productivity in mesic environments. We added 20 g N m?2 in a 2‐m radius around the base of mature Populus fremontii along each of a regulated and free‐flowing river in semiarid northwest Colorado, USA (total n = 42) in order to test whether growth is constrained by low soil N. Twelve years after fertilization, we collected increment cores from these and matched unfertilized trees and compared radial growth ratios (growth in the 3‐year post‐fertilization period/growth in the 3‐year pre‐fertilization period) in paired t tests. We expected a higher mean ratio in the fertilized trees. No effect from fertilization was detected, nor was a trend evident on either river. An alternative test using analysis of covariance (ANCOVA) produced a similar result. Our results underscore the need for additional assessment of which and to what extent factors other than water control dryland riverine productivity. Positive confirmation of adequate soil nutrients at these and other dryland riparian sites would bolster the argument that flow management is necessary and sufficient to maximize productivity and enhance resilience in affected desert riverine forests.  相似文献

Dear Editor, During recent decades, a novel mechanism of secretion has been identified in a wide range of mammalian cells. It involves the release of bioactive membrane nanovesicles (30-100 nm), termed exosomes, upon the fusion of multivesicular bodies with the plasma membrane (Thery et al., 2009). Exosomes are implicated in diverse functions, such as scavenging of archaic proteins, intercellular messengers delivering cell-specific signals, and vehicles for transmissible pathogens. The C4 perennial grasses Miscanthus × giganteus and Panicum virgatum (switchgrass) are emerging bioenergy crops. They are attractive because they are productive and recycle nutrients to the overwintering belowground rhizomes, before the dry shoots are harvested. They are therefore expected to require minimal fertilizer inputs, a desirable trait for any crop. Until now, Europe has had the only long-term trials of M. × giganteus, and these have either shown no or a small effect of nitrogen fertilization, while trials of P. virgatum in the USA have shown a clear positive effect of N fertilization. This study exploited the first long-term trials of M. × giganteus in the USA, and first side-by-side comparison with P. virgatum, to test the hypothesis that N fertilization would only improve yields of the latter. A split-plot N fertility treatment (0, 67, 134, and 202 kg(N)?ha?1) was added to >5-year-old replicated stands of the two crops at seven locations on contrasting soils in the US Midwest. Averaged across all locations, M. × giganteus yields increased significantly from 23.4 Mg ha?1 with no N fertilization to 28.9 Mg ha?1 (+25 %) at a N application rate of 202 kg ha?1. P. virgatum also showed significant yield increases from 10.33 Mg ha?1 at 0 kg(N)?ha?1 to 13.6 Mg ha?1 (+32 %) at 202 kg(N)?ha?1. Both species therefore responded to N fertilization and to a similar extent. The increase per unit of added N was small compared to crops such as Zea mays and unlikely to be economically worthwhile. Nitrogen fertilization arrested most of the long-term yield decline that would otherwise have occurred in P. virgatum, but eliminated only about 40 % of the decline observed in M. × giganteus, suggesting additional causal factors for long-term yield decline in this crop. While the crops responded to nitrogen addition at some locations, they did not at others. Therefore a one-case-fits-all optimum fertilization rate cannot be prescribed.  相似文献

The ferflization and its embryo fonnation of different types of embryo sacs were studied by using the technology of GMA half-section in the APIV strain of polyembryonic rice ( Oryza sativa L. ). In rare cases,all three egg cells in the embryo sac of 5-2-1 type could fertilize and develop into three embryos. But in most cases only one or two egg cells fertilized and developed into one or two embryos in the respective type of embryo sac. The frequency of poly-egg fertilization in total all was very low in the embryo sac of 6-2-0 type and 5-3-0 type. These results indicated that the polyembryos in APIV originated mainly from overall fertilization and develotment of the embryo sac with poly-egg apparatus. This was observed,for example, in 5-2-1 type in which three embryos were fertilized and developed from three egg cells and two embryos from two egg cells. The fertlization process of double set of embryo sac was most complicated, all often abnormal. Only in few ovaries the egg cells in both large and small embeyo sac could fertilize simultaneously. The fertilization and development of egg cells in the large embryo sac might be the main cause of the formation of the so called "mid-seated embryo" (the embryo far from the micropyle end).  相似文献

The concept of pilot pseudopodia is reconsidered 30 years after its inauguration (Gerisch, G., Hülser, D., Malchow, D., Wick, U., 1975. Cell communication by periodic cyclic-AMP pulses. Phil. Trans. R. Soc. Lond. B 272, 181-192). The original hypothesis stated that protruding pseudopodia serve as dynamic sensory organelles that aid a cell in perceiving variations of chemoattractant concentration and, consequently, in navigation during chemotaxis. This influential idea is reevaluated in the light of recent findings about the mechanisms governing chemotactic cell motility, morphology and dynamics of pseudopodia, and about molecular constituents and regulators of pseudopod extension and retraction. It is proposed that stimulation by a chemoattractant modulates speed of pseudopod protrusion and thereby increases cell elongation. Elongation further enhances chemotactic sensitivity of the cell to shallow chemoattractant gradients, reinforces cell polarization, and finally leads to suppression of lateral pseudopodia and continuation of cell migration in the gradient direction.  相似文献
