Loss of luteal progesterone secretion at the end of the ovine estrous cycle is via uterine PGF2α secretion. However, uterine PGF2α secretion is not decreased during early pregnancy in ewes. Instead, the embryo imparts a resistance to PGF2α. Prostaglandins E (PGE; PGE1 + PGE2) are increased in endometrium and uterine venous blood during early pregnancy in ewes to prevent luteolysis. Chronic intrauterine infusion of PGE1 or PGE2 prevents spontaneous or IUD, estradiol-17β, or PGF2α-induced premature luteolysis in nonbred ewes. The objective was to determine whether chronic intrauterine infusion of PGE1 or PGE2 affected mRNA for LH receptors, occupied and unoccupied receptors for LH in luteal and caruncular endometrium, and luteal function. Ewes received Vehicle, PGE1, or PGE2 every 4 h from days 10 to 16 of the estrous cycle via a cathether installed in the uterine lumen ipsilateral to the luteal-containing ovary.Jugular venous blood was collected daily for analysis of progesterone and uterine venous blood was collected on day-16 for analysis of PGF2α and PGE. Corpora lutea and caruncular endometrium were collected from day-10 preluteolytic control ewes and day-16 ewes treated with Vehicle, PGE1 or PGE2 for analysis of the mRNA for LH receptors and occupied and unoccupied receptors for LH. Luteal weights on day-16 in ewes treated with PGE1 or PGE2 and day-10 control ewes were similar (P  0.05), but were greater (P  0.05) than in day-16 Vehicle-treated ewes. Progesterone profiles on days 10–16 differed (P  0.05) among treatment groups: PGE1 > PGE2 > Vehicle-treated ewes. Concentrations of PGF2α and PGE in uterine venous plasma on day-16 were similar (P  0.05) in the three treatment groups. Luteal mRNA for LH receptors and unoccupied and occupied LH receptors were similar (P  0.05) in day-10 control ewes and day-16 ewes treated with PGE2 and were lower (P  0.05) in day-16 Vehicle-treated ewes. PGE2 prevented loss (P  0.05) of day-16 luteal mRNA for LH receptors and occupied and unoccupied LH receptors. Luteal and caruncular tissue mRNA for LH receptors and occupied and unoccupied LH receptors were greater (P  0.05) on day-16 of PGE1-treated ewes than any treatment group. mRNA for LH receptors and occupied and unoccupied receptors for LH in caruncules were greater (P  0.05) in day-16 Vehicle or PGE2-treated ewes than in day-10 control ewes. It is concluded that PGE1 and PGE2 share some common mechanisms to prevent luteolysis; however, only PGE1 increased luteal and endometrial mRNA for LH receptors and occupied and unoccupied LH receptors. PGE2 prevents a decrease in luteal mRNA for LH receptors and occupied and unoccupied receptors for LH without altering endometrial mRNA for LH receptors or occupied and unoccupied receptors for LH.  相似文献

In pregnant sheep at 120–128 days of gestational age, a study was done to evaluate the effect of the co-administration of verapamil (Vpm) and ivermectin (IVM) on the maternal and fetal disposition kinetics after intravenous administration of IVM. Ten pregnant Suffolk Down sheep of 63.5 ± 6.6 kg body weight (bw) were surgically prepared to insert polyvinyl catheters in the fetal femoral artery and vein and amniotic sac. The ewes were randomly assigned to two experimental groups. In Group 1 (control), 5 ewes were treated with an intravenous bolus of 0.2 mg IVM/kg bw. In Group 2 (Vpm-IVM), 5 ewes were subcutaneously treated with Vpm (2.5 mg/kg × 3 doses at 12 h intervals) and 0.2 mg/kg IVM by intravenous route. Maternal and fetal blood samples were taken before and after IVM administration during a 144 h post-treatment period. Samples were analyzed by liquid chromatography (HPLC). A non-compartmental pharmacokinetic analysis was performed and statistical differences were determined using the Mann–Whitney U-test.Significantly higher IVM levels in maternal plasma concentrations were determined after co-administration of Vpm/IVM compared with the group treated with IVM alone. Significant decreases in the volume of distribution and in the half-life of elimination (t½β) were observed in the Vpm/IVM treated group. A significantly faster (Tmax) and higher (Cmax) (P < 0.05) increase in IVM fetal plasma concentrations were observed in the group of fetuses from pregnant ewes treated with Vpm/IVM. The results of our study support the hypothesis that pharmacological blockage of P-glycoprotein with Vpm can increase the transfer of IVM to fetal circulation.  相似文献

ObjectiveTo determine first-trimester thyroid function values and associations with thyroperoxidase antibody (TPO-Ab) status, smoking, emesis, and iodine-containing multivitamin use.MethodsWe collected information by interview, questionnaire, and blood draw at the initial obstetric visit in 668 pregnant women without known thyroid disease. We compared thyroid-stimulating hormone (TSH), total thyroxine (T4), and free T4 index (FT4I) values by TPO-Ab status. Multiple regression was used to identify characteristics associated with thyroid function values.ResultsThe following median (range containing 95% of the data points) thyroid function test values were obtained in 585 TPO-Ab–negative women: TSH, 1.1 mIU/L (0.04-3.6); FT4I, 2.1 (1.5-2.9); and T4, 9.9 μg/dL (7.0-14.0). The following median (range containing 95% of the data points) thyroid function test values were obtained in 83 TPO-Ab–positive women: TSH, 1.8 mIU/L (0.3-6.4) (P < .001); FT4I, 2.0 (1.4-2.7) (P = .06); and T4, 9.3 μg/dL (6.8-13.0) (P = .03) (P values denote statistically significant differences between TPO-Ab–positive and negative participants). Among TPO-Ab–negative participants, TSH level was not associated with use of iodine-containing multivitamins, smoking, or race. TSH increased 0.03 mIU/L for every year of maternal age (P = .03) and decreased by 0.3 mIU/L for every increase in parity (P < .001). T4 decreased 0.04 μg/dL for every year of maternal age (P = .04). Mean FT4I was 2.05 in smokers and 2.20 in nonsmokers (P < .01). There were no relationships between T4 or FT4I and parity, race, or iodine-containing multivitamin use.ConclusionTPO-Ab status of pregnant women should be considered when constructing trimester-specific reference ranges because elevated serum TPO-Ab levels are associated with higher TSH and lower T4 values. ObjectiveTo assess the relationship between serum thyrotropin (thyroid-stimulating hormone or TSH) on one hand and thyroid-stimulating immunoglobulin (TSI), free thyroxine (T4), and triiodothyronine (T3) levels on the other in Graves’ disease, inasmuch as TSH may be suppressed in the presence of TSI because TSI may bind to the TSH receptor on the thyroid gland membrane and thus eliminate the need for circulating TSH for stimulating the thyroid gland.MethodsWe determined serum TSI levels in 37 women and 13 men with Graves’ disease, stratified into 4 groups on the basis of serum TSH levels irrespective of serum free T4 and T3 levels. Our reference ranges were 0.72 to 1.74 ng/dL for free T4, 80 to 200 ng/dL for T3, and to 4.0 μU/mL for TSH.ResultsMean serum TSI concentrations were highest (215% ± 28%) in patients with undetectable TSH levels (< 0.03 μU/mL) and lowest (103% ± 9%) in those with supernormal TSH concentrations (> 4.0 μU/mL). TSI levels were intermediate in the other study groups: 157% ± 16% in patients with subnormal though detectable TSH levels (0.03 to 0.39 μU/mL) and 125% ± 12% in those with normal TSH levels (0.4 to 4.0 μU/mL). Moreover, a progressive decline in TSI levels with increasing serum TSH concentrations was noted, along with a significant negative correlation (r = -0.45; P < 0.01) between serum TSI and TSH concentrations. Finally, relationships between free T4 and T3 levels on one hand and TSI or TSH levels on the other were not significant, with a considerable variability in free T4 and T3 levels being noted in individual study groups.ConclusionSerum TSH is frequently suppressed after treatment with antithyroid drugs or radioiodine (131I), irrespective of clinical thyroid function as expressed by increased, normal, or decreased free T4 and T3 concentrations. In an individual patient with Graves’ disease, the serum TSH level may be more reflective of the circulating TSI concentration than is thyroid gland function as expressed by free T4 and T3 concentrations and therefore may be as reliable a predictor of remission as TSI. The fertility obtained in sheep after the use of intravaginal progesterone devices is related to the content of progesterone of the device. The hypothesis of this study was that the reproductive response of anoestrous ewes to the ram-effect could be improved by the administration of oestradiol-17β in conjunction with CIDRs treatment—using previously used CIDRs in a 5-day progestagen priming. Therefore, the objective was to determine if oestradiol-17β treatment increases fertility of anoestrous ewes primed with used CIDRs and stimulated by the ram-effect. The hypothesis was tested with CIDRs that had been previously used for 12 or 18 days. The trial was performed during the non-breeding season using 158 Corriedale ewes. Ewes had been isolated from rams since Day −35 (Day 0 = introduction of the rams). A CIDR (0.3 g progesterone, InterAg, Hamilton, New Zealand) was inserted on Day −5 in all ewes with CIDR that had been previously used for 12 days (n = 62) or 18 days (n = 96). Also on Day −5, 29 and 53 ewes that had received CIDRs of 12 or 18 days, respectively, received an intra-muscular treatment of 50 μg of oestradiol-17β (E groups). The ewes that did not receive the oestradiol-17β treatment remained as the control group (C group). Overall the treatment groups were thus: C12 (n = 33), C18 (n = 43), E12 (n = 29), and E18 (n = 53). On Day 0 all CIDRs were withdrawn, and ewes were placed with 18 rams and 20 ewes hormonally induced to exhibit oestrus. Sexual receptivity of ewes treated with CIDRs was estimated from marks on the rumps of the ewes daily from Day 0 to Day 5, and the pregnancy status diagnosed with transrectal ultrasonography on Day 40. The percentage of ewes exhibiting oestrus and pregnancy rates were lower in ewes synchronized with previously used CIDRs for 18 days, compared to those used for 12 days. The responses of ewes in oestrus were 39.4, 14.0, 65.5, and 32.1% for the C12, C18, E12, and E18 groups respectively, with pregnancy rates of 30.3, 14.0, 34.5, and 17.0%. Administration of oestradiol-17β increased the frequency of oestrous response in ewes that were treated with CIDRs previously used for 12 days (P < 0.05), but not in those treated with CIDRs used for 18 days. It could be concluded that the administration of oestradiol-17β only improved the percentage of ewes responding to oestrus when CIDRs previously used for 12 days were used for 5 days before the introduction of rams. No positive effect on fertility was observed irrespective of the period during which CIDR had been previously used.  相似文献

BackgroundIodine is an essential trace element for the synthesis of thyroid hormones, which are keys in maternal metabolism during pregnancy as well as in neurological development during fetal and postnatal life. This was a prospective study on iodine status and thyroid function in women during pregnancy in the Basque country to assess whether there was any relationship among maternal urinary iodine, maternal thyroid function and thyrotropin (TSH) in newborns, and to explore any difference in women experiencing miscarriages.MethodsWe analyzed TSH, free T4 (FT4), free T3 (FT3), thyroid peroxidase antibody (TPO-Ab) titers in serum and urinary iodine concentrations (UIC) in 2104 women in the first trimester of pregnancy and in 1322 of them in their second trimester. We obtained neonatal TSH levels in 1868 cases.ResultsIn the first (T1) and second trimesters (T2), the median UICs were 88.5 μg/L and 140 μg/L, respectively. No relationship was found between UIC and FT4, or maternal and neonatal TSH. In T1 and T2, 9.7% and 7.5% of women were TPO-Ab positive, respectively. The total miscarriage rate was 10%. The percentage of miscarriages in healthy women was 8.9%, lower than in women with overt hypothyroidism (21.2%; p < 0.001) and than in women with subclinical hypothyroidism (15.6%; p < 0.025). The miscarriage rate was not higher in TPO-Ab-positive women.ConclusionsIn this study most women had iodine deficiency during pregnancy. Neonatal TSH is not correlated with maternal UIC during pregnancy. Pregnant women with hypothyroidism have a higher rate of miscarriages.  相似文献

A series of experiments were conducted in ewes and wether (castrate male) lambs to evaluate the influence of prostaglandins on secretion of anabolic hormones and to determine if repeated injections of prostaglandin (PG) F2α would chronically influence the secretion of these hormones and perhaps growth rate as well.A single intravenous injection of PGA1 and PGB1 (100 μg/kg) exerted no significant (P > .10) influence on plasma concentrations of prolactin (PRL), growth hormone (GH) or thyrotropin (TSH) in ewes. PGA1, but not PGB1, stimulated an increase in the plasma concentration of insulin. Infusion of PGF2α for 5.5 hr into ewes resulted in increased (P < .05) plasma concentrations of both GH and PRL while TSH and insulin were not significantly influenced. Prostaglandin F2α, when injected subcutaneously into wether lambs (10 mg twice weekly) stimulated (P < .05) plasma GH concentrations after the first injection, but not after 3 weeks of treatment. Changes in plasma PRL or TSH were not observed consistently in the lambs treated chronically with PGF2α or TRH.Prostaglandin F2α, in the present studies, and PGE1 in previously reported studies (1–3), has been demonstrated to be stimulatory to the secretion of PRL and GH. In contrast, PGA1 and PGB1, which lack an 11-hydroxyl group, failed to influence the secretion of either PRL or GH. It would, therefore, appear that the 11-hydroxyl group is a structural requirement for prostaglandins to influence the secretion of these two hormones in sheep.Treatment with thyrotropin releasing hormone (TRH), alone or in combination with PGF2α, significantly (P < .05) increased growth rate (average daily gains) while PGF2α did not, despite the fact that both compounds exerted similar effects on plasma GH.  相似文献

The objective of this study was to evaluate the effect of crossbreeding Awassi ewes with either Charollais or Romanov sires, on pre-weaning lamb production as reflected by reproductive performance and milk production in the ewes. Two hundred and five, 3- to 6-year-old multiparous ewes of three breeds were allocated to three groups [Awassi (A; n = 56), F1 Romanov × Awassi (RA; n = 78) and F1 Charollais × Awassi (CA; n = 71)]. Upon lambing, ewes and their offspring were placed in a large pen in which they remained until the end of the trial. Body weight (BW) and body condition score (BCS) of the ewes and the BW of the lambs were recorded weekly from birth to weaning at 70 days of age. Milk production was recorded weekly from parturition to weaning. Pregnancy rates were not influenced by breed-type. Multiple births and the fecundity of ewes were greater (P < 0.05) in the RA group, compared to the A and CA groups—while the weaning percentage was similar between the breeds. Lamb birth and weaning weights were similar, while the kg lamb born per kg ewe lambed was greater (P < 0.05) in the RA group, compared to the CA group. The CA ewes recorded a higher BW (P < 0.01) and BCS (P < 0.01) than the other breed groups. Awassi ewes produced more milk (P < 0.01) throughout the observation period. Milk ash percentage was higher (P < 0.05) in the CA group, while the percentage of crude protein and dry matter was similar for the different groups. Results of the present study indicate that crossbreeding of Awassi ewes with Charollais and Romanov breeds decreased milk production in the progeny, without affecting lamb growth. The crossbreeding of Awassi with Charollais resulted in improved BW and BCS, while the crossbreeding of Awassi ewes with Romanov resulted in improved reproductive performance of the crossbreds. Crossbreeding Awassi ewes with exotic rams can thus be conducted to increase the number of lambs produced per ewe.  相似文献

The aim of this paper was studying the phenotypic and genetic relationships between udder morphology traits and udder health in dairy sheep. From 2000 to 2008 udder traits, somatic cell count (SCC) and clinical mastitis cases were recorded in an experimental flock consisting each year of around 900 ewes. A logistic regression was performed to evaluate the risk for a ewe of showing either a mastitis or at least 2 daily SCC records greater than 1 × 106 cells/ml in one of its lactations as function of udder traits scored in 1st lactation. 1587 individual udder scores and 39,950 SCC daily records were used for the analysis. Secondly, genetic correlation between lactation mean of somatic cell score and udder traits were estimated by a REML method applied to four bi-trait animal models, using data from 2251 primiparous ewes. Logistic regression results indicated that the risk of mastitis or high SCC values during the productive life increased as the cistern height increased and the degree of udder suspension and udder depth decreased. This suggests that the appraisal of the udder is a useful tool for culling decision aimed at increasing the sanitary status of the flock. The genetic correlation between lactation SCS and udder traits were favourable for udder depth (−0.50 ± 0.12), teat placement (0.39 ± 0.011) and degree of udder suspension (−0.42 ± 0.011) and closed to zero for the degree of separation of the two halves. Thus selection for udder morphology, already implemented in some dairy sheep breeds with the aim of improving machine milkability, will lead to a favourable correlated genetic response on SCC.  相似文献

The purpose of the study was to investigate the effects of different doses of equine chorionic gonadotropin (eCG) treatment on follicular development, ovulation and pregnancy rate during the breeding season in fat-tailed Chall ewes. Seventy-two cycling (62.5 ± 2.5 kg), multiparous Iranian Chall ewes were used in the trial. The ewes were randomly allocated to 6 groups (n = 12/group). Estrus was synchronized with the aid of controlled intravaginal drug release (CIDR) devices, inserted for 14 days. At the time of CIDR removal (day 14), the ewes received i.m. either 0 (control group, G0), 450 (G450), 550 (G550), 650 (G650), 750 (G750) or 850 (G850) IU eCG. Vasectomized rams were used to detect estrus in the ewes from 24 h after CIDR removal. Ovarian follicular activity was monitored with the aid of transrectal ultrasonography on the day of CIDR insertion (day 0) and daily from the day of eCG treatment (day 14), until estrus (day 16). During these days, blood samples were collected for the determination of plasma progesterone and estradiol concentrations. Laparoscopic intrauterine inseminations were conducted 54–60 h after CIDR removal. The number of CL's and pregnancy diagnosis was recorded using ultrasonography 7 and 54 days following AI, respectively. Half of ewes in control group and most of the ewes treated with eCG showed signs of estrus within 36 h of CIDR removal. The ewes in groups G750 and G850 recorded the highest number of large follicles at estrus and CL's 7 days later. The pregnancy rate in groups G550 (75.0%) and G650 (75.0%) was higher (P < 0.05) than that in the other groups. The ovarian response and estradiol concentration, as well as pregnancy rate showed that 550 or 650 IU eCG treatment is the most effective doses in improving the pregnancy rate in Iranian Chall ewes.  相似文献

In the ewe, ovarian antral follicles emerge or grow in a wave-like pattern and each wave is preceded by a peak in the serum FSH level. The purpose of the current study was to investigate whether in anestrous Western White Face ewes, a combination of progesterone and estradiol affects the circulating FSH peak secretion and the number of small ovarian follicles. Five ewes were treated with subcutaneous silastic rubber implants (10 cm × 0.47 cm), containing 10% estradiol-17β w/w (controls) and 5 ewes were treated with the same estradiol implant, along with subcutaneous implants (11 cm × 0.48 cm) containing 10% progesterone w/w for 12 days. Daily transrectal ovarian ultrasonography and blood sampling was performed from 5 days before, to 9 days after the period of implantation. Blood samples were also taken every 12 min for a 6 h period on day −2, 6 and 13 prior to or after implant insertion (day 0, day of implant insertion). Pulsatility in the serum LH levels was eliminated by the implants (P < 0.05). During the implantation period, the serum FSH peak amplitude was lower in ewes treated with implants releasing estradiol and progesterone, compared to ewes treated with implants releasing only estradiol (P < 0.05). No follicular waves emerged during implant treatment in both groups (P < 0.05) and the number of serum FSH peaks did not differ during implantation, compared to before implantation. During the implantation period, the number of small follicles did not differ in ewes with implants releasing estradiol and progesterone, compared to ewes treated with implants releasing only estradiol. To conclude, supra-physiological concentrations of estradiol completely eliminated the serum LH pulsatality and suppressed the follicular wave emergence, while the FSH secretory peaks that preceded the follicular waves were not affected. Supra-physiological concentrations of estradiol-17β with physiological concentrations of progesterone decreased the serum FSH peak amplitude, eliminated the serum LH pulses, but did not decrease the size of the small follicle pool in anestrous ewes.  相似文献

This study describes the fluctuations in serum energy-related metabolites during a period of 2 weeks before, to 2 weeks after parturition in Lori-Bakhtiari ewes. The effect of parity was also studied. Blood profiles were determined in 60 healthy pregnant ewes with single (n = 30) and twin (n = 30) lambings. Blood was collected from each ewe on days 14 and 7 prepartum, and days 7 and 14 postpartum to determine the serum non-esterified fatty acid (NEFA), β-hydroxybutyrate (BHBA), glucose, cholesterol, blood urea nitrogen (BUN) and calcium (Ca) levels. The age of the ewes had no significant effect on the energy metabolism indicators. Serum NEFA, BHBA, glucose, BUN and calcium concentrations recorded peak levels 7 days before parturition. However, NEFA and BHBA recorded significant changes (P < 0.05) during the peri-parturition period. All metabolites changed significantly in ewes carrying twin-bearing ewes, compared to single-bearing ewes. Serum BHBA concentrations recorded positive correlations with the serum NEFA (P < 0.01) and cholesterol (P < 0.05), while blood glucose had negative correlations (P < 0.01) with NEFA, BHBA and Ca. Blood NEFA and BHBA recorded positive correlations (P < 0.05) with the BUN levels and negative correlations (P < 0.05) with Ca. The results showed that blood NEFA and BHBA levels are sensitive indicators of the energy balance during the peri-parturition period in ewes.  相似文献

Thirty male post-weaned Muzafarnagari lambs, of about 3 months of age and similar body weight (18.47 ± 1.31 kg), were divided into 5 similar groups in order to observe the effect of graded levels of palm oil supplementation on growth, nutrient utilization, cost of feeding, carcass characteristics and meat quality under feedlot regimen. Lambs in different treatment groups were fed with concentrate mash supplemented with 0% (T1), 2.5% (T2), 5.0% (T3), 7.5% (T4) and 10% (T5) palm oil and gram straw (Cicer arietinum) ad libitum. The growth trial was continued for 12 weeks. A metabolism trial of 6 days collection was conducted after 60 days of experimental feeding. After 90 days of feeding, randomly selected three animals from each group were slaughtered according to standard procedure for assessment of carcass traits and meat quality. Separated thoracic portion of Longissimus thoracis muscle from each carcass was collected for analysis of moisture, protein and fat. Accelerated growth in lambs under T3 (P < 0.05) was observed as compared to other treatments with similar DM intake. Similarly, DM intake (kg)/kg gain was found most efficient (P < 0.01) in T3 and least efficient in 10% palm oil supplemented T5. Due to this, feed cost (rupees)/kg weight gain was calculated less (44.23) in T3 than other palm oil supplemented treatments. DM, CP, Ca and P intakes were similar in all treatment groups during metabolism trial. Intake ratio of Ca and P was recorded 3.68:1.00, 3.42:1.00, 3.37:1.00, 3.69:1.00 and 3.44:1.00 in T1 to T5 treatments, respectively. Water intake and water intake/kg DM intake was also similar in different treatment groups. OM digestibility was significantly higher (P < 0.01) in 2.5% palm oil supplemented T2 ration as compared to other treatments. DM, CP, EE, CF, NFE and P digestibilities were found similar among different treatment groups. TDN percentage in the feed increased from 60.16 (T1) to 66.17 (T5), which was mainly due gradual increase of palm oil in the diet, although the difference was non-significant. Whereas, DCP percentage in feed gradually depressed (P < 0.05) due to higher level of palm oil supplementation. Nitrogen retention (%) as percentage of N-absorbed was significantly higher (P < 0.05) in palm oil supplemented groups than control T1, being highest in T3 and T4. This could be the reason for higher body weight gain in T3 and T4. Dressing and meat percentages (empty weight basis) ranged from 52.00 (T4) to 55.06 (T2) and 58.10 (T2) to 62.27 (T5), respectively, however, the differences were similar. Muscle fat% (DM basis) increased (P < 0.01) from 19.84 (T1) to 28.94 (T5) due to palm oil addition. Meat protein% (DM basis) remained unaffected due to these treatments, which ranged from 59.95 (T1) to 70.47 (T2). Result indicates that addition of 5% palm oil in concentrate mixture improved growth performance and feed conversion efficiency of weaned Muzafarnagari lambs keeping the quality of meat unaffected.  相似文献

Goat keeping is feasible for smallholder farmers in many world regions especially those best suited for extensive management. However, summertime grazing in arid zones entails major challenges to animal thermoregulation and well-being. An experiment was conducted to evaluate the thermoregulatory performance and selected hemogramic parameters in intensively (INT) or extensively (EXT) managed goat kids (N = 14). We applied a previously established technique to evaluate body thermal state of freely ranging animals, in which contemporaneous temperatures of the core (Tc) and periphery (Tp) are chronically recorded. Animals were initially kept for 12 days under INT management. Subsequently, seven animals were transferred to a grazing pasture and gradually transitioned over a four-day acclimatization period, then kept for the last 22 days under EXT conditions. Water drinking was limited to twice daily in both groups. Excessive solar radiation-induced heat load – with daytime black globe temperatures (Tbg) often exceeding 40 °C – under EXT was primarily responsible (r2 = 0.49; P < 0.05) for 0.57 and 1.72 °C rises in Tc and Tp, respectively, over INT kids. Unlike the typically biphasic pattern noticed for daily temperatures of both body sites in INT goats, that of EXT counterparts became rather polyphasic, whereby water drinking had drastic and prolonged thermolytic effect, inducing 0.40–0.41 and 0.79–1.45 °C declines in Tc and Tp, on midday and afternoon watering bouts, respectively. Despite indication for added daytime heat load, EXT goats displayed lower early morning Tc than INT. All animals exhibited hypohydration, as reflected by rises in hematocrit, serum osmolality, albumin, potassium, and sodium, being more pronounced in EXT conditions. Results emphasize the excessive thermophysiological strain facing grazing animals in arid zones during the summer.  相似文献

The aim of the experiment was to evaluate the effects of GnRH and/or eCG and progestin type (implant versus CIDR) on the induction of estrus and pregnancy rate following laparoscopic AI (LAI) with frozen semen. In the first trial, ewes (n = 129) were treated with norgestomet implants for 14 days. At implant removal ewes received eCG (400 IU) and/or GnRH (25 μg) 36 h after removal, resulting in control, eCG, GnRH, and eCG/GnRH groups (n = 30–34/group). In trial 2, ewes (n = 36) were treated with intravaginal fluorogestone acetate sponges (FGA) or CIDR for 12 days. After withdrawal, half of the ewes from each progestin group received eCG (400 IU), resulting in sponge, sponge/eCG, CIDR and CIDR/eCG groups (n = 8–10/group). In both trials, estrous activity was assessed using a vasectomized ram from the time of progestin removal to laparoscopic AI with frozen semen 58–60 h (trial 1) or 54–56 h (trial 2) following cessation of treatment. In trial 1, GnRH decreased (P < 0.05) the percentage of ewes in estrus (GnRH, 75.8% versus control, 93.8% versus eCG/GnRH, 94.1%), however pregnancy rates were similar in all groups (control, 53.1%; eCG, 70.0%; GnRH, 51.5%; eCG/GnRH, 55.9%, respectively). In trial 2, neither the type of progestin nor eCG treatment effected the percentage of ewes in estrus (sponge, 75.0%; sponge/eCG, 100.0%; CIDR, 100.0%; CIDR/eCG, 90.0%). However, pregnancy rates following LAI were higher (P < 0.05) when ewes were treated with eCG (progestin + eCG, 73.7% versus progestin alone, 41.2%). Results demonstrate that the source of progestin does not influence the expression of estrus or the proportion of ewes pregnant following LAI. When progestin treatment protocols are used in combination with eCG, pregnancy rates can be increased. A dose of GnRH near the end of progestin treatment may decrease the estrous response, by inducing ovulation before normal expression of estrus.  相似文献

Tensiomyography is a non-invasive method of neuromuscular assessment used to measure muscle action characteristics, muscle tone, and muscle fiber type, and provides information on acute and chronic responses of muscle to different training loads. The aims of the present study were: to analyse differences in muscle response and mechanical characteristics of two major muscles of the lower extremity in a large group of Spanish soccer players according to playing position, and to provide group norms against which clinical findings may be compared. Data were collected from 78 professional soccer players (age 26.6 ± 4.4 years; height: 179.2 ± 5.3 cm; body mass: 75.8 ± 5.3 kg). Tensiomyography was recorded from the rectus femoris (RF) and biceps femoris (BF) muscles after 2 days without take part in any strenuous exercise or training. Five tensiomyographic parameters were analyzed: maximal displacement (Dm), contraction time (Tc), sustain time (Ts), delay time (Td), and half-relaxation time (Tr). A good to excellent intra-session reliability was found for all contractile parameters (ICC ranged from 0.78 to 0.95). No significant differences between players of any position were observed in absolute values of BF. However, significant differences were observed for Tc, Tr and Ts between the different playing positions on RF (P < 0.05, effect size ranged from 1.3 to 1.6). Professional soccer players showed muscles with ability to rapidly generate force during contractions. The neuromuscular profile provided could help in identifying the normative data that are important for the different positions in order to optimize the training and recovery process of each individual player.  相似文献

Temperature-dependent development of Spodoptera exigua (Hübner) were evaluated at eight constant temperatures of 12, 15, 20, 25, 30, 33, 34 and 36 °C with a variation of 0.5 °C on sugar beet leaves. No development occurred at 12 °C and 36 °C. Total developmental time varied from 120.50 days at 15 °C to 14.50 days at 33 °C. As temperature increased from 15 °C to 33 °C, developmental rate (1/developmental time) of S. exigua increased but declined at 34 °C. The lower temperature threshold (Tmin) was estimated to be 12.98 °C and 12.45 °C, and the thermal constant (K) was 294.99 DD and 311.76 DD, using the traditional and Ikemoto–Takai linear models, respectively. The slopes of the Ikemoto–Takai linear model for different immature stages were different, violating the assumption of rate isomorphy. Data were fitted to three nonlinear models to predict the developmental rate and estimate the critical temperatures. The Tmin values estimated by Lactin-2 (12.90 °C) and SSI (13.35 °C) were higher than the value estimated by Briere-2 (8.67 °C). The estimated fastest development temperatures (Tfast) by the Briere-2, Lactin-2 and SSI models for overall immature stages development of S. exigua were 33.4 °C, 33.9 °C and 32.4 °C, respectively. The intrinsic optimum temperature (TΦ) estimated from the SSI model was 28.5 °C, in which the probability of enzyme being in its native state is maximal. The upper temperature threshold (Tmax) values estimated by these three nonlinear models varied from 34.00 °C to 34.69 °C. These findings on thermal requirements can be used to predict the occurrence, number of generations and population dynamics of S. exigua.  相似文献

The primary goal of this study was to employ ultrasonography to examine the ovaries of ewes undergoing superovulatory treatment for correlations between antral follicular blood flow and ovarian responses/embryo yields. Five Santa Inês ewes were subjected to a short- (Days 0–6, Group 1) and five to a long-term progesterone-based protocol (Days 0–12, Group 2) to synchronize estrus and ovulations after the superovulatory treatment. Porcine FSH (pFSH, 200 mg) was administered in 8 decreasing doses over 4 days, starting on Days 4 and 10 in Groups 1 and 2, respectively. After CIDR removal, all ewes were bred by a ram and embryos were recovered surgically 7 days later. Transrectal ovarian ultrasonography was performed the day before and on all 4 days of the superovulatory treatment. Both an arbitrary-scale [(0) non-detectable; (1) small; (2) moderate; (3) intense blood flow] and quantitative analysis of the blood flow area were used to assess the follicular blood flow in color Doppler images. There were no significant correlations between the arbitrary blood flow scores and superovulatory responses in the ewes of the present study. However, there was a positive correlation between the quantitative estimates of follicular blood flow on the final day of the superovulatory treatment, and the number (DA: r = 0.68, P < 0.05; DA/TA × 100%: r = 0.85, P < 0.05) and percentage (DA: r = 0.65, P < 0.05; DA/TA × 100%: r = 0.91, P < 0.001) of unfertilized eggs (DA: Doppler area, TA: total area of the largest ovarian cross section). This experiment presents a commercially practical tool for predicting superovulatory outcomes in ewes and evidence for the existence of follicular blood flow threshold that may impinge negatively on oocyte quality when surpassed during hormonal ovarian superstimulation.  相似文献

The ability of sheep to consume big sagebrush (Artemisia tridentata Nutt.), a shrub that contains terpenes, may be improved with supplemental energy and protein needed for detoxification processes. We compared feeding behavior of supplemented and unsupplemented ewes grazing separately in three 0.5 ha plots of 40 m × 120 m during October 2001. Supplemented ewes were offered a choice of a high-energy (50% corn and 50% beet pulp—3.62 Mcal/kg DE and 9.5% CP) and a high-protein (85% alfalfa and 15% soybean meal—2.89 Mcal/kg DE and 15% CP) supplement for 15 min/day. Supplemented ewes consumed 492 g (S.E. = 24.3) of the high-energy supplement and 140 g (S.E. = 9.8) of the high-protein supplement. The incidence of feeding on sagebrush was recorded using scan sampling at 10-min intervals from 09:00 to 17:00 h every other day for 10 days. Incidence of feeding on sagebrush increased gradually for both treatments from 0.4% on day 1 to 28% on day 10 (P < 0.01), but supplemented ewes increased feeding frequency on sagebrush to nearly twice that of unsupplemented ewes by the end of the trial. Averaged throughout the trial, incidence of eating sagebrush was twice as high for supplemented compared to unsupplemented ewes (22% versus 11% of the total time feeding, P < 0.01). The results of this study show that supplemental macronutrients increase use of sagebrush by ewes. Thus, strategic supplementation during fall and winter may be a way to increase intake of sagebrush by sheep and enhance and maintain biodiversity in areas of the sagebrush steppe where sagebrush is deemed too abundant.  相似文献
