The efficacy of pyrimethamine-sulfadoxine in the treatment of uncomplicated falciparum malaria in young children of a malaria holoendemic area in rural Burkina Faso is reported. Of 28 children treated with a standard single dose of pyrimethamine-sulfadoxine and followed-up over 14 days, only one Late Treatment Failure and four Late Parasitological Failures were observed, all with low-grade parasitaemia. In this area of very restricted use of pyrimethamine-sulfadoxine, the drug appears to be still sufficiently effective in the treatment of malaria. These findings provide further evidence for the justification of continued use of pyrimethamine-sulfadoxine as a second-line treatment for malaria in Burkina Faso.  相似文献

The D14S7 locus defines the breakpoint on chromosome 14 of a t (8;14) (q24;q11) present in the T-cell line KE37-R in which DI4S7 sequences translocate 3' to the c-myc oncogene. It has been shown previously that DI4S7 rearranges specifically in some but not all T cell clones and in the present study we investigated the frequency and specificity of its rearrangements in human fresh lymphoma samples. DI4S7 rearrangements were extremely specific since they were detected in 3 out of 5 T-cell lymphoma samples positive for TCR beta and gamma but not in 17 miscellaneous non-T lymphomas, 4 non neoplastic lymphnodes as well as unstimulated and activated polyclonal T-cells. Most of the rearrangements were in the form of deletions that appear to involve large pieces of DNA since the segments detected by a V alpha probe were also deleted. Rearrangement of DI4S7 and V alpha regions were detected in lymphomas with a cortical thymocyte phenotype, demonstrating that they appear quite early in the differentiation of T cells.  相似文献

The present report describes clinical, hematological and biochemical studies of a 27-year old Egyptian woman in whom a fast moving Hb variant was found. The abnormal Hb constituted 48% of the total erythrocyte Hb of the propositus and her father. Structural studies demonstrated that in the abnormal Hb lysine beta 65 is replaced by glutamine. The new Hb mutant is designated hemoglobin J Cairo beta 65 (E9) Lys leads to Gln. This substitution results in only a moderate decrease in cooperativity. No evidence of Hb instability was found. A slight anemic state has been observed in the propositus since she reached adolescence.  相似文献

Abstract High-level resistance to glycopeptides in Enterococcus faecium is associated with an inducible 39-kDa cytoplasmic membrane protein. The present paper shows that such glycopeptide-resistant E. faecium strains can not only be isolated in a definite clinical setting but also from waste water of sewage treatment plants. Nearer characterization of these and of clinical isolates by resistance pattern, biotyping, and genotyping (DNA-fingerprinting with pulsed-field get electrophoresis) has shown that different glycopeptide-resistant E. faecium strains have been isolated from clinical sources and from waste water.  相似文献

The crystal structure of the homotetramer of a chimera beta alpha-subunit of human hemoglobin was refined at 2.5 A resolution. The chimera subunit was constructed by replacing an exon-encoded module M4 of the beta-subunit with that of the alpha-subunit, simulating an exon-shuffling event. The implanted module M4 retained the native alpha-subunit structure, while module M3 was disturbed around the site where a new type of intron was recently found. Some of the residues were found in alternative conformations that avoid steric hindrance at the subunit interface. The modules are modestly rigid in their backbone structures by using side-chains to compensate for interface incompatibility.  相似文献
