The change in the body and cerebellar weights, together with the quantity of RNA, DNA and proteins in the cerebellum were studied for the first 10 days and on the 12th, 14th, and 17th days of postnatal life in normal, hypo- and hyperthyroid rats. In the normal animals: (1) The average cellular protein content decreases from the first to the second day, increases to a maximum at 4 days, then decreases. (2) The specific radioactivity of the RNA, 14 h after an intravenous injection of [6-14C]orotic acid, varies distinctly from birth to 9 days and reaches two maxima at 4 and 6 days. After 9 days it decreases markedly. (3) Mitotic activity (number of replicating cells) increases, reaches a maximum at 9 days, then decreases. (4) The specific radioactivity of the DNA (used as a measure of the percentage of the cellular population in division) reaches a maximum at 6 days. (5) Mitotic efficiency (number of replicating cells in mitotic activity) decreases from 2 to 7 days, and subsequently increases. In the hypothyroid animals: (1) The average cellular protein content increases from the first to the second day and then decreases. (2) The specific radioactivity of the RNA, always significantly higher than that of normal animals, varies from birth to 9 days, reaches two maxima at 4 and 6 days, then decreases after 9 days. (3) Mitotic activity, always significantly lower than that of normal animals, increases from birth, reaches a maximum at 9 days, then decreases. (4) The specific radioactivity of the DNA reaches a maximum at 6 days and the mitotic efficiency a minimum at 7 days. Neither are significantly different from that of normal animals. In the hyperthyroid animals: (1) The average cellular protein content, is maximal at 2 days, then decreases. (2) The specific radioactivity of the RNA, always significantly lower from that of normal animals, decreases from birth. (3) Mitotic activity is similar to that of normal animals, increases from birth up to 6 days, then decreases. (4) The specific radioactivity of the DNA increases from birth up to 5 days, then decreases. It is significantly lower than that of normal animals. (5) Mitotic efficiency is significantly higher than that of normal animals. In the different groups, the maximum of the average cell size, always precedes the maximum of the cellular division. In the hypothyroid animals, the rate of cell death is higher than that of normal animals, and the average cell size is higher during the first fourteen days. In the hyperthyroid animals, the rate of cell death is lower than that of normal animals, and the average cell size is higher at 14 and 17 days.  相似文献

Weight, RNA, DNA and protein content of the oviduct, vaginal cul-de-sac, lateral vagina and urogenital sinus and oestradiol and progesterone cytosol receptor concentrations in vaginal cul-de-sac, lateral vagina and urogenital sinus were examined after administration of oestradiol to ovariectomized animals and on days 0, 5, 9 and 13 of the non-pregnant cycle and on day 13 of the pregnant cycle. In ovariectomized animals, oestradiol induced an increase in weight, RNA:DNA and protein:DNA ratios and a decrease in DNA:tissue weight ratio for each organ and in addition an increase in total DNA in vaginal cul-de-sac and urogenital sinus. There was no effect of oestradiol on oestradiol cytosol receptor concentration but there was a significant increase in progesterone cytosol receptor concentration in all organs that were examined. During the oestrous cycle, changes in the wet weight of each organ showed a common pattern with maximum weight at day 0 followed by variable rates of decline until day 13. In oviduct and vaginal cul-de-sac, the decrease in weight was paralleled by a decrease in RNA:DNA and protein:DNA ratios whereas the DNA: tissue weight ratio showed the opposite pattern and total DNA remained unchanged. The changes in the lateral vagina and urogenital sinus were similar except that a significant decline in total DNA was also seen after day 0 and the DNA:tissue weight and protein:DNA ratios in the urogenital sinus and the lateral vagina respectively showed no significant changes. Progesterone cytosol receptor concentration in the lateral vagina and urogenital sinus were high on day 0 and then declined until day 13. In contrast, there were no consistent effects on oestradiol receptor concentration.  相似文献

A developmental time-table has been established for the embryos of viviparous Diploptera punctata. The percentage of gestation time occupied by pre-dorsal closure stages is only 19 per cent compared to 40 or 50 per cent in non-viviparous species. The increase in wet weight of the embryos begins several days before dorsal closure and continues throughout gestation. The increase in dry weight, protein, carbohydrate, and uric acid does not begin until shortly after dorsal closure but thereafter parallels the increase in wet weight.The increase in oöcyte protein during vitellogenesis is tenfold, less than in oviparous or ovoviviparous species. But the increase in embryo protein during gestation is sixtyfold; this characterizes viviparity in D. punctata.Although total lipid increases during gestation, lipid as a percentage of wet weight decreases most rapidly before dorsal closure and less rapidly there-after. It is suggested that before dorsal closure lipid is the major energy source for development. After dorsal closure the embryos are able to drink through their mouths the fluid nutrient provided by the mother.The ultimate source of nutrients required by developing embryos is the maternal diet during gestation. Females starved from ecdysis do not produce young; females fed only sugar from ecdysis produce viable larvae but suffer loss in body weight. Embryos from such females, although normal in length, are deficient in protein.  相似文献

Changes in fr. and dry wt, soluble reducing sugars, protein. total carbohydrate, DNA, RNA, sucrose synthetase activity and invertase activity were recorded for the developing embryo of Hordeum distichum var Julia over the period 18–60 days after anthesis. Fresh wt increased until 45 days whereupon rapid dehydration commenced. Reducing sugar concentration remained low throughout development but total carbohydrate and protein accumulated rapidly over the initial period to reach maximum values at around 50 days. DNA concentration remained relatively constant throughout the middle and later stages of development, but RNA, on the other hand, increased rapidly to reach a maximum value at maturity. Sucrose synthetase (assayed in the direction of sucrose cleavage) was considerably more active with UDP than ADP and reached a maximum value around 35 days after anthesis. When assayed in the direction of sucrose synthesis the peak of activity was slightly later in development and doubled in value. Invertase activity was appreciable and was still present at maturity.  相似文献

1. After treatment of immature rats with diethylstilboestrol, the wet weight and RNA content of uterine tissue increased rapidly, reaching a peak at 40hr. After an initial lag of a few hours, the acid-soluble ribose and protein contents also rose to maxima at 40hr. No increase in DNA content occurred until at least 24hr. after treatment. 2. The RNA from immature rat uterus isolated at various times up to 6hr. after administration of oestradiol-17beta was labelled by injecting [(3)H]uridine and [(3)H]guanosine intraperitoneally 30min. before the animals were killed. It was fractionated on columns of kieselguhr coated with methylated serum albumin and the radioactivity in fractions corresponding to transfer RNA, 7s RNA, ribosomal RNA, Q(1)-RNA, Q(2)-RNA and DNA-like RNA was determined. 3. The radioactivity of the whole RNA increased steadily for 6hr. after hormone treatment. The earliest changes occurred in the Q(1)-RNA (ribosomal RNA precursor), whereas at longer time-intervals the radioactivity of the ribosomal RNA, 7s RNA and transfer RNA increased by four- to five-fold. The radioactivity of the DNA-like RNA increased by about 50%, but only at the longer time-intervals. 4. It is concluded that one of the earliest changes in response to oestradiol treatment is a major increase in synthesis of ribosomal RNA followed later by a similar increase in synthesis of transfer RNA and by a much smaller increase in synthesis of DNA-like RNA. The change in synthesis of ribosomal RNA in immature rat uterus may represent one of the most important responses to oestradiol treatment.  相似文献

The brood sac of viviparous Diploptera punctata is a typical insect integumentary gland which secretes a ‘milk’ containing protein and carbohydrate to nourish the developing embryos. During gestation the secretory cells proliferate organelles of protein synthesis and secretion and brood sac wet weight, protein content, synthetic activity and secretory output increase five- to six-fold ; a maximum of 0.4 mg protein was collected in 24 hr from one brood sac in a later stage of gestation. Following parturition, when secretory activity ceases, these parameters fall markedly, and the secretory cells decrease their mass by autophagic regression. Acid phosphatase has been located histochemically in autolysomes and assayed in brood sac homogenates; activity reaches a maximum five days after parturition.  相似文献

Soybean (Glycine max [L.] Merr.) ovary growth was measured from anthesis to 6 days after anthesis (DAA) to establish a timetable of biochemical events that might be useful in identifying processes that initiate abscission. Two procedures were developed to provide samples with either high or low percent pod set for `IX93-100,' a semideterminate line having long racemes. Characteristics measured were fresh and dry weight, soluble and insoluble protein, soluble carbohydrate, starch, RNA, and DNA. Setting ovaries grew more rapidly than abscising ovaries. Since there was a daily increase in ovary weight in both groups, all measured characteristics showed daily increases when expressed on perovary basis. Statistically significant differences between groups were detected between 2 and 5 DAA for most characteristics. When chemical composition was expressed on concentration basis, starch level was significantly higher in setting ovaries at 5 and 6 DAA. Regression analysis showed that these deviations between setting and abscising samples started between anthesis and 1 DAA. We conclude that processes leading to eventual shedding of fertilized ovaries (called flower abortion in soybeans) commence soon after anthesis of the shed flower, and that setting and abscising ovaries do not differ in protein, soluble carbohydrate, starch, or nucleic acid content when abscission processes begin.  相似文献

1. Changes in dry weight, DNA, RNA, protein and reserve and structural carbohydrate were measured during the aerobic growth of yeast on 0.9% glucose in an aerobic synthetic medium. 2. After glucose had been consumed and during the growth of yeast on ethanol and acetate, the rate of formation of DNA remained about the same but the rate of increase of dry weight was greatly diminished. 3. During the second stage of growth the ratios dry weight/DNA, protein/DNA, RNA/DNA and carbohydrate/DNA decreased to about 30% of the corresponding values during the first stage of growth. 4. A higher fraction of the dry weight of the yeast cells could be accounted for by the reserve carbohydrate content of the cells during the second stage of growth. 5. By the end of the first stage of growth an increase in the reserve carbohydrate content of the cells was observed. Part of this reserve carbohydrate was consumed by the cells in the beginning of the second stage of growth. The possibility of adaptation of cells at the expense of their reserves is discussed.  相似文献

Examinations of the macromolecular components of the protein synthesizing system (RNA, DNA and protein) have been made in the marine cyanobacterium, Synechococcus sp. WH 7803. Slowly growing, irradiance limited cells have less RNA and lower rates of RNA synthesis than do those growing at rapid rates. RNA content and synthesis increase in conjunction with division rate. Protein content is variable. Protein synthesis increases up to a plateau at division rates less the maximum observed. The results imply that there is extra protein synthetic capacity produced at high, irradiance limited growth rates. Synechococcus sp. WH 7803 responds to an increase in irradiance through a rapid shiftup in macromolecular synthesis. RNA, protein and DNA increase in a sequential fashion which precedes the onset of cell division. After decreases in irradiance, protein synthesis is maintained despite reductions in RNA. This suggests that there is some degree of physiological buffering which occurs in this species. These studies indicate that, as in more extensively studied procaryotic models, the protein synthesizing system plays a central role in the global mechanisms regulating growth in Synechococcus sp. The development of DNA and RNA synthesis in the germ cell population was studied after a 3H-thymidine or 3H-uridine pulse at each stage of spermatogenesis. The autoradiographic results show that the first sign (after 3 days in vitro) of cellular changes is an increase in RNA synthesis which reaches a maximum at day 5. DNA replication (premeiotic S phase) occurred at day 7, then cells entered meiotic prophase (day 9). Meiotic divisions and spermiogenesis occurred after 11 days. Silver grain counts permit the conclusion that RNA synthesis is clearly higher during premeiotic interphase (days 3-7) than during spermatogonial proliferation (day 0). It appears therefore that male meiotic differentiation in Nereidae is accompanied by increased RNA synthesis.  相似文献

Alterations in the biochemical constituents of mycelia were studied during the growth, development, and starvation of Microsporum quinckeanum. On the basis of dry weight, growth of this dermatophyte could be divided into four phases: lag, log, stationary, and death. The percentage of total nitrogen, inorganic phosphorus, ribonucleic acid (RNA), and protein increased rapidly during the lag phase. The percentage of protein remained constant after the initial increase; however, inorganic phosphate and RNA decreased in older mycelia. Acid-soluble materials in the cells increased in concentration as the organism aged. Chitin was present in the spores at a much higher concentration than in the mycelia. The percentage of this compound decreased rapidly until the end of the lag phase. An increase and subsequent decrease in per cent chitin occurred during the log phase. Inorganic phosphorus in the mycelia increased from the value in the spore stage to a maximum in the early log phase, and then decreased rapidly during the remainder of the growth cycle. Compounds involved in protein synthesis increased rapidly during the lag phase of growth. Changes in chemical composition of the mold during starvation indicate that carbohydrate does not form the principal endogenous reserve of M. quinckeanum, whereas lipids may represent the primary reserve material.  相似文献

Abstract The vitellogenic period of gypsy moth ovarian development starts on day 3 of the pupal stage and continues through adulthood. During this period, rapid increases occur in follicle size, protein content, and wet weight of the ovary. Patency is observed on day 3 of the pupal stage.
Pre-vitellogenic follicles are formed in the last larval stadium. Newly formed follicles detach from the germarium on day 4, and increase rapidly to 140 per ovariole at the end of the last larval stadium. The pre-vitellogenic follicles are uniformly around 50 um in diameter. No vitellogenin is incorporated into the oocytes until the pupal stage.
Polyacrylamide gel electrophosesis (PAGE) in the presence of sodium dodecylsulphate (SDS) analysis of male and female haemolymph samples and vitellogenic ovaries demonstrates the presence of two female-specific subunits of vitellogenin of 180 kD and 160 kD. These proteins are detected only in haemolymph and ovarian extracts of vitellogenic females. The molecular weight of the native protein determined by size exclusion chromatography is approximately 400–420 kD.
A highly sensitive double antibody sandwich enzyme-linked immunosorbent assay (ELISA) was developed to monitor the temporal changes in vitellogenin titre in haemolymph. Vitellogenin production starts on day 2 of the last larval stadium, reaching a maximum level by day 6 of the last larval stadium, and decreasing in the late pupal stage as vitellogenin was internalized into the oocytes. This is the first report of vitellogenin production occurring in the larval stage of a holometabolous insect. The fact that vitellogenin production and uptake occur during different stages of development in the gypsy moth, opens up some interesting questions concerning the underlying regulatory mechanisms controlling each process.  相似文献

Mesophyll cells were isolated from primary leaves of 5- to 21-day Phaseolus vulgaris plants. The rate of photosynthesis and respiration, and RNA, protein, and lipid synthesis was determined for these cells. Appropriate 14C substrates and product purification procedures were used for each process prior to liquid scintillation counting. The size of the leaves increased about 5-fold between days 5 and 11, and then remained relatively constant. The greatest increase in size occurred between days 5 and 6. The age of the leaf from which the cells were isolated had a pronounced effect on the rate of all of these processes. The largest changes occurred during the period of leaf expansion (days 5-11). Initially the rate of RNA, protein, and lipid synthesis increased rapidly, maintained a maximum rate for only 1 day (day 6 or day 7), and then declined. The rate of photosynthesis increased more slowly reaching a maximum at day 9, remained relatively constant until day 15, and then declined. The rate of respiration decreased during the first 4 days to low level which was maintained throughout the experiment. The time course patterns of these biochemical processes in isolated cells were similar to those which have been reported for intact leaves. It seems that isolation of leaf cells does not modify their metabolic activity.  相似文献

Candida utilis NCYC 321 was grown in steady-state culture in a chemostat under glucose limitation or NH4+ limitation at temperatures of 30, 25, 20, and 15 C and at dilution rates (equal to growth rates) in the range of 0.35 to 0.05 hr−1. Deoxyribonucleic acid contents of cells grown under the various conditions remained approximately constant, but the contents of several other cell components varied. Over the range of 30 to 15 C, the greatest differences were in the ribonucleic acid (RNA) and protein contents of cells grown under NH4+ limitation, which increased as the temperature was decreased. The contents of other components, particularly adenosine triphosphate in cells grown under glucose limitation, varied more when the cells were grown at different rates at a fixed temperature. Cells grown at a fixed rate under NH4+ limitation increased in volume as the temperature was decreased below 30 C. The increase in volume was closely correlated with increases in the proportions of RNA and protein in the dry weight of cells. Cells grown under glucose limitation showed much smaller increases in volume; these increases were poorly correlated with the increased RNA content and hardly at all with the increased protein content. Increases in volume with a decrease in growth temperature from 30 to 20 C were also demonstrated in cells grown under phosphate limitation and to a much smaller extent in cells grown under glycerol limitation. The increased RNA synthesized at low temperatures by cells grown under NH4+ limitation was found almost exclusively in the 40,000 × g supernatant fluid, but only about 40% of it sedimented at 100,000 × g. Cells grown at a fixed rate under NH4+ limitation synthesized less total carbohydrate when the temperature was decreased from 30 to 15 C. This decrease was mainly in the trichloroacetic acid-soluble fraction (probably trehalose) and in the intracellular hot alkali-soluble glucan (probably glycogen). Cells grown at a fixed rate under glucose limitation showed a small increase in carbohydrate content as the temperature was decreased from 30 to 15 C.  相似文献

Abstract— Changes in the activities of several specific enzymes were measured in the cerebellum during development. Early transient increases were found in both ornithine decarboxylase and S -adenosylmethionine decarboxylase, enzymes involved in the initial steps of polyamine synthesis. Different patterns of changes were found in neurotransmitter synthesizing enzymes. Tyrosine hydroxylase activity achieved adult levels very early, by 3 days after birth, and remained at this level. Glutamic acid decarboxylase activity, while very low at early stages, increased rapidly before birth and then after a lag period of 10 days started to increase rapidly, directly related to the general growth of cerebellar weight and protein content. Choline acetyltransferase activity started to increase rapidly, reaching a peak of about 100% of adult levels at 3-7 days after birth; the activity then gradually declined and at 20 days, after reaching a low of about 55% of adult values, gradually started to increase, reaching adult levels later than 40 days after birth. The development of protein carboxymethylase activity was similar to that of glutamic acid decarboxylase, directly related to the general growth of the cerebellum. Several interpretations of the results are discussed.  相似文献

Protein, carbohydrate, free amino acid, lipid, RNA levels, and electrophoretic changes in the protein profile were determined in the eggs of the water scorpion, Laccotrephes griseus, during normal embryonic development. The protein levels remain lower and relatively constant in the eggs of 0, 1 and 2 days of age, while in the eggs of older groups, i.e. between 2 and 6 days, a marked increase in the protein level occurs. Then its level declines. The RNA content shows a rise up to the day 6 stage, later it declines sharply, indicating an increase in the degree of synthetic activity that takes place during such period of embryonic development. Electrophoretic and densitometric analysis show the qualitative and quantitative changes of yolk protein reflecting the utilization of already existing proteins as well as the appearance of new proteins.Water content increases gradually as development proceeds. There is a steady depletion of carbohydrate and lipid during the course of embryonic development. The nature of yolk components as well as their preferential utilization during embryogenesis has been discussed in relation to the generally accepted view that protein serves as the source for the embryonic metabolism in aquatic insects does not hold good for L. griseus and other freshwater insects.  相似文献

Flowers of Digitalis purpurea L. cv. Foxy, take just over aweek to develop from the smallest buds and open. Both the anthersand the stigma are closed when the flower opens, the anterioranthers dehisce 2 days after flower opening and the posterioranthers a day later. The stigma opens 2 days later still andthe corolla remains attached for a further 2 days. The maximum size of the corolla is reached soon after openingand then remains constant. The fresh weight increases throughoutthe attached life of the corolla, whereas the dry weight reachesa maximum at flower opening and decreases during the remainderof the corolla's attached life. The content of antho-cyaninvaries greatly from spike to spike. In some spikes the maximumcontent is at flower opening, in others it is much later, butin all cases the loss of anthocyanin with ageing is very smalland the corollas are abcissed without visible wilting or fading.Maximum protein, RNA and glucose contents occur at, or soonafter, flower opening and the levels decrease in the corollasas they age. Fructose reaches a maximum at the same time asglucose but does not decrease as rapidly, so that fructose becomesthe major reducing sugar in the older corollas. Sucrose cannotbe detected in any flowers. Ethylene production increases steadilywith flower age, but the maximum production is recorded fromflowers which have recently ab-scissed the corolla regardlessof whether this occurs before detachment from the spike or withinthe collection tube.  相似文献

Biosynthetic processes related to the production of vitellogenin (yolk precursor protein) have been examined in the fat body of adult female Locusta migratoria. Vitellogenin-producing capacity was assayed by incubation of fat body with [3H]leucine, followed by precipitation from the medium with specific antiserum. In normal development, vitellogenin synthesis began at about Day 7 after emergence and became maximal at about Day 13, when this protein accounted for 60% of the total fat body protein output. The production of other proteins increased to a lesser extent, becoming maximal at about Day 6. The incorporation of uridine into fat body RNA rose to a maximum at Day 8, which coincided with a marked increase in tissue RNA content. The DNA content in adult female fat body approximately doubled between Days 3 and 8. Vitellogenin synthesis, and the increases in RNA and DNA, were prevented by removal of the corpora allata (the source of juvenile hormone). In allatectomized locusts, vitellogenin synthesis was induced by JH or an analog, ZR-515. Applied topically in acetone, these gave steep dose-response curves, half-maximal at 75 and 150 μg, respectively. After a single treatment with ZR-515, fat body vitellogenin production rose slowly during 48 hr, then steeply to a maximum at 72 hr, but after decay of this effect during 10 days, a second application of ZR-515 induced renewed synthesis with little initial lag. Hormone treatment produced a smaller increase in the output of other proteins, and an increase in incorporation into RNA which preceded the major rise in vitellogenin synthesis. Male fat body produced little or no vitellogenin. These results are consistent with action of JH at the gene level.  相似文献

应用微量分析方法检测了山黧豆胚胎发育过程中ODAP毒素含量和核酸、蛋白质、糖类等大分子物质的含量变化。结果表明:每粒种子的ODAP含量随着胚的发育而增加。每粒种子DNA量随着细胞的迅速分裂而增加,R、蛋白质、淀粉含量随着胚的发育而成倍地增加,当进入心形胚时这些物质的增加更为迅速。如以每克干重中的含量来表示,那么ODAP、DNA及可溶性糖含量则随胚的发育而下降,其它大分子物质含量在胚发育前期升高,进入心形胚时,这些物质达到最高峰;到鱼雷胚时,这些物质含量开始下降,直到胚基本分化完全时,降到最低点;只有酸性蛋白质含量一直保持增长。  相似文献

1. The effect of simultaneous injections of oestrogen benzoate and progesterone (0.5mg/day each) on immature chicken oviduct tissue pretreated with oestrogen benzoate (0.5mg/day) was studied. 2. After 3 days of treatment with both hormones, the weight of the tissue doubles as compared with tissue treated only with oestradiol benzoate. 3. The progesterone-induced weight increase had no effect on total DNA content of the tissue, but greatly increased the protein/DNA and RNA/DNA ratios. 4. Amino acid incorporation in vivo after progesterone treatment was elevated as measured by using 2h pulses of amino acids; this effect could be accounted for by observed alterations in the concentrations of free amino acids in the tissue. 5. With longer pulses of amino acid the specific radioactivity of total protein remained high in tissue treated with progesterone, at times when specific radioactivity of protein in oestrogen-treated animals was diminishing. 6. From a knowledge of the specific radioactivity of labelled amino acids in the free amino acid pool and in newly synthesized protein, the rate of protein synthesis was estimated in differently treated tissues. 7. The results suggest that progesterone treatment does not cause an increase in protein synthesis. It is concluded that the observed accumulation of oviduct protein is achieved via an effect of progesterone on protein catabolism.  相似文献
