When carrot cell cultures, after growth in semicontinuous culture, were transferred to media containing excess nutrients, they grew at different rates. The growth rates were generally higher after semicontinuous culture at higher dilution rates. There appears to be a limit on dilution rate above which growth rate does not increase. These changes were also displayed by clones from the parental culture. The possibility that these changes in growth rate reflect a need for the cultures to adapt to their new conditions is discussed. The growth rates of the cultures is markedly increased at 25 degrees C compared with 22 degrees C with little further increase at 28 degrees C. Growth rate is altered by less than 20% when pH is changed from 4.5 to either 5.5 or 4.2. The rates of anthocyanin accumulation by the cultures were similar under all conditions tested except at 22 degrees C. They were larger than the rates of dry weight accumulation. In contrast, the amounts of anthocyanin accumulated in the clones and in the parental cultures grown at pH 5.5 instead of pH 4.5 were very different. The observations were interpreted as showing that the clones differ in the rate of metabolism but not in the rate of synthesis of anthocyanins and that at pH 5.5 the rate of metabolism of anthocyanins but not the rate of synthesis is higher than it is at pH 4.5. The use of semicontinuous cultures as a source of inoculum for batch cultures rather than as a source of biomass for extraction of chemicals is discussed.  相似文献

Chemostat cultures of carrot suspension cultures, where growth was limited by the concentration of phosphate in the input medium, were achieved by replacing a fixed proportion of the culture with fresh medium at daily intervals. In the range 0.05–0.30mM phosphate in the input medium and at a specific growth rate of 0.357 days?1, steady-state culture density but not anthocyanin in the cells was strictly proportional to the input phosphate concentration with no intercept. At a phosphate concentration of 0.10mM and growth rates from 0.105 to 0.430 days?1, the steady-state culture density could not be described by Monod's model of chemostat cultures, but could be described by Nyholm's model. The steady-state levels of anthocyanin were not strictly proportional to the steady-state biomass under all conditions, showing that anthocyanin production is not completely growth associated.  相似文献

Streptomyces thermoviolaceus was grown in a chemostat under conditions of glutamate limitation. The effects of growth rate on production of the antibiotic granaticin, extracellular protein and protease activity as components of secondary metabolism were studied at 37, 45 and 50 degrees C. The amount of each secondary metabolite synthesized was highly dependent on growth rate and temperature. Granaticin yields were highest at growth rates of 0.1 to 0.15 h-1 at 37 degrees C, 0.175 h-1 at 45 degrees C and 0.045 h-1 at 50 degrees C. Protease activity of culture supernatants responded to low nutrient concentration and/or low growth rate. Measurements of extracellular protein revealed complex changes in amount which were dependent on growth rate and temperature. At 45 degrees C and a growth rate of 0.15 h-1, biomass yield was highest between pH 5.5 to 6.5 whereas granaticin synthesis was low at pH 5.5 and rose to highest values at between pH 6.5 and 7.5.  相似文献

The medium used for the growth of anthocyanin-accumulating wild carrot (D. carota) suspension cultures contained ammonia as a sole nitrogen source and was buffered with succinate. Ammonia was the first nutrient to be completely utilized.The uptake of carbohydrate, phosphate and succinate continued after ammonia depletion. Biomass accumulation was faster and greater when sucrose was initially present in the medium than when glucose was present. When sucrose was provided in the medium it was rapidly hydrolysed to glucose and fructose and the fructose was used preferentially to glucose. Summary The fatty acid, polysaccharide and pigment contents ofPorphyridium cruentum at different growth temperatures, light intensities and dilution rates were studied in light-limited chemostat. The highest biomass yield was obtained at 25°C over the range of dilution rate studied (0.079 to 0.30 day–1). The production of fatty acids increased with increasing dilution rates. At low dilution rates, the contents of fatty acids at 20°C, 25°C and 30°C were comparable. Temperature did not affect the content of linolenic acid (182) though fatty acids with less than two double bonds predominated at higher temperatures, those with more than two double bonds accumulated at lower temperatures. The yields of arachidonic acid (204) increased under light of higher intensity. The maximum production of extracellular and cellular polysaccharides occured at low dilution rates under high light at 25°C. The amounts of chlorophylla, carotenoids and phycoerythrin all increased in the same proportions with dilution rate, and were highest at 25°C.
Resumen Se estudió el contenido en polisaccaridos, pigmentos y ácidos grasos dePorphyridium cruentum a distintas temperaturas, intensidades lumínicas y porcentajes de dilución en un quimiostato con luz limitada. La mayor cantidad de biomasa para todas las diluciones estudiadas (0.079 a 0.30 día–1) se obtuvo a 25°C. A bajas diluciones los contenidos en ácidos grasos a 20°C, 25°C y 30°C eran comparables. La temperatura no afectó al contenido en ácido linolénico (182), sin embargo los ácidos grasos con menos de dos dobles enlaces se acumularon a temperaturas más bajas. La producción de ácido arachidónico (204) se incrementó bajo la mayor intensidad lumínica ensayada. La producción máxima de polisaccaridos celulares y extracelulares se dió a bajas diluciones en condiciones de alta iluminación a 25°C. Las cantidades de clorofila a, carotenoides y ficoeritrina se incrementaron paralelamente al porcentaje de dilución siendo más elevadas a 25°C.

Résumé On a étudié le contenu dePorphyridium cruentum en acides gras, en polysaccharides et en pigments, en fonction de la température de croissance, de l'intensité lumineuse et du taux de dilution dans un chémostat limité par la lumière. Le rendement le plus élevé en biomasse a été obtenu à 25°C quel que soit le taux de dilution (compris entre 0.079 et 0.30 jours–1). La production d'acides gras augmente avec l'augmentation du taux de dilution. A des taux de dilution faibles, le contenu en acides gras est indépendant de la température (comprise entre 20 et 30°C). La température n'influence pas le contenu en acide linolénique (182) bien que les acides gras à deux ou plusieurs doubles liaisons s'accumulent à température plus basse. Les rendements en acide arachidonique (204) augmentent sous une lumière de plus forte intensité. La production maximum de polysaccharides tant extracellulaires que cellulaires a lieu à un taux faible de dilution sous une lumière intense à 25°C. Les teneurs en chlorophylle a, caroténoïdes et en phycoerythrine augmentent toutes les trois dans les mêmes proportions avec le taux de dilution et sont les plus élevées à 25°C.

Anthocyanin formation in a suspension culture of Daucus carota is induced by transfer from medium containing 2,4-dichlorophenoxyacetic acid (2,4-D) to one lacking 2,4-D. The specific yields were strongly influenced by the inoculum density. Inoculum density altered the effect of zeatin concentration on anthocyanin accumulation. The in part by increasing the sucrose levels. It was inferred from the results that sucrose was exhausted at a low concentration of sucrose and at a high cell density, resulting in the decrease of yield of anthocyanin.  相似文献

The effects of four pH steps and one dilution rate step are described for an ammonia-nitrogen-limited continuous culture of Escherichia coli B/r. Two of the pH steps, 6.06-5.49 and 5.96-5.60, led to prolonged transients in both cell density and rate-limiting nutrient concentration. The other two pH steps, 6.20-5.96 and 5.60-6.20, had almost no effect on the culture. The dilution rate step led to a sharp transition in the steady-state external pyruvate concentration. Monitoring of the external pyruvate concentration for the pH 5.96-5.60 step revealed that the transient phase continued long after the cell density and rate-limiting nutrient concentration returned to steady-state values. The implications for industrial and laboratory fermentations are discussed.  相似文献

Micrococcus sedentarius , an organism associated with pitted keratolysis, produced two proteinases in culture supernatant fluids, as shown by non-denaturing PAGE with overlaying with a casein substrate. A mixture had optimal activity at pH 10 with azocasein substrate. At pH 7.1 and 8.1 in continuous culture with varying dilution rates high proteinase production occurred at relative specific growth rates (μrels) 0.39 and 0.77 and biomass concentrations decreased with increasing dilution rate. One proteinase was constitutive and varied little in production with different growth rates. The other proteinase was under control with high production at low growth rates and no production at high growth rates. With varying pH at μrels, 0.39 and 0.77 maximum biomass concentration and proteinase production occurred between pH 8.0 and 9.0 as did the highest specific growth rate. These results support the hypothesis that Mic. sedentarus produces pitting in the stratum corneum when the skin is hydrated and the pH rises above neutrality.  相似文献

The effect of salinity on vacuolar pH was studied in carrot (Daucus carota L.) cells grown in liquid suspension culture either in the absence or presence of 150 mM NaCl. Both vacuolar and cytoplasmic pH were determined by several independent techniques. These techniques were NMR spectrometry, distribution of radioactive probes and spectrophotometric measurement of the absorbance changes of a naturally occurring vacuolar pH indicator. There was no difference in the cytoplasmic pH between cells grown in the presence or the absence of NaCl, but the vacuolar pH of cells grown in the presence of NaCl was higher by 0.38 to 1.05 pH units (depending on the technique that was used) than the vacuolar pH of cells grown in the absence of NaCl.  相似文献

The growth characteristics ofCandida blankii as a function of temperature and pH in a simulated bagasse hemicellulose hydrolysate were determined in chemostat culture. The highest maximum specific growth rate of 0.44h–1 was reached at 38°C and at pH 5.5, with a sharp decrease in growth rate on either side of this temperature. Growth occurred at 46°C but not at 48°C. The protein and cell yields varied little below 40°C and the respective values were 0.22 and 0.5 g/g at 38°C. At the lower pH values, a severe linear decrease in cell and protein yields occurred, whereas a small increase in these yields at decreasing pH values was found when acetic acid was omitted from the medium. In the presence of acetic acid, a very sharp decrease in the growth rate at pH values below pH 4.5 was noted, despite the very low residual acetic acid concentrations, of less than 50 mg/l, in the culture.  相似文献

The optimum dilution rate for 2, 3-butanediol (BDL) production by Paenibacillus polymyxa was 0.2 h1 and the optical purity of BDL remained above 98 % at all dilution rates. With decreasing culture pH, ethanol and BDL production increased, whereas the optical purity of BDL decreased to 94 % at pH 5.7. In the chemostat culture at pH 6.3 and a 0.1 h1 dilution rate, the optimum air supply for BDL production was 200 ml min–1 in which the O2 uptake rate was 6.7 mmol l–1 h–1. The physiology of a recombinant Chinese hamster ovary cell line in glucose-limited chemostat culture was studied over a range of dilution rates (D = 0.008 to 0.20 h(-1)). The specific growth rate (mu) deviated from D at low dilution rates due to an increased specific death rate. Extrapolation of these data suggested a minimum specific growth rate of 0.011 h(-1) (mu(max) = 0.025 h(-1)) The metabolism at each steady state was characterized by determining the metabolic quotients for glucose, lactate, ammonia, amino acids, and interferon-gamma (IFN-gamma). The specific rate of glucose uptake increased linearly with mu, and the saturation constant for glucose (K(s)) was calculated to be 59.6 muM. There was a linear increase in the rate of lactate production with a higher yield of lactate from glucose at high growth rates. The decline in the rate of production of lactate, alanine, and serine at low growth rate was consistent with the limitation of the glycolytic pathway by glucose. The specific rate of IFN-gamma production increased with mu in a manner indicative of a growth-related product. Despite changes in the IFN-gamma production rate and cell physiology, the pattern of IFN-gamma glycosylation was similar at all except the lowest growth rates where there was increased production of nonglycosylated IFN-gamma. The effect of growth retardants on anthocyanin production was studied in wild carrot (Daucus carota) cell suspension cultures. Paclobutrazol [(2RS,3RS) — 1 — (4-chlorophenyl) — 4,4 —dimethyl-2-(1,2,4-triazol-1-yl) pentan-3-ol], uniconazole [(E)-1-(4-chlorophenyl-4,4 —) dimethyl-2-(1,2,4-triazol-1-yl)-1-penten-3-ol], tetcyclacis [5-(4-chloro-phenyl) -3,4,5,9,10-pentaaza-tetracyclo-5, 4, 102,6, O8,11 — dodeca-3, 9-diene], ancymidol [-cyclopropyl — 4 — methoxy-(pyrimidine-5-yl)benzyl alcohol] and CCC (2-chloro-ethyltrimethylammonium chloride) increased anthocyanin accumulation. AMO-1618 [(2-isopropyl-5-methyl-4-trimethyl-ammonium-chloride)-phenyl-1-piperidinium carboxylate] did not increase anthocyanin accumulation in the first passage but did increase it during the second passage on medium for improved anthocyanin accumulation. Prohexadione (3,5-dioxo-4-propionylcyclohexane carboxylic acid) decreased anthocyanin accumulation by 10%–12.5%.The inhibitory effect of gibberellin on anthocyanin accumulation was reversed by paclobutrazol. Paclobutrazol together with 10–6M GA3 increased anthocyanin level from 33% of control in GA3 treated cell suspension to 76%. These results are consistent growth retardants increasing anthocyanin accumulation in carrot cell suspension cultures by inhibiting gibberellin biosynthesis.  相似文献

The influence of pH and temperature on the substrate yield coefficient for growth of Saccharomyces cerevisiae in a chemostat under limited organic substrate conditions was studied. Mathematical analysis of the substrate yield coefficient as a function of pH and temperature in the near-optimal area was made. It was shown that the location of pH and temperature optima were independent of each other. The maximum substrate yield coefficient had the following coordinates: pH = 4.1, temperature = 28.5°C.  相似文献

采用恒化培养的方法,考察了稀释率(D)和碳氮比(mol/mol)对圆红冬孢酵母Rhodosporidiumtoruloides AS 2.138 9积累油脂的影响。结果表明:稀释率增大,油脂含量和油脂得率降低。在D=0.02 h 1时油脂得率最大,为0.18 g油/g糖;D=0.14 h 1时油脂生成速率最大,为0.09 g/(L.h)。碳氮比增大,油脂含量略有增加。在C/N=92时油脂得率最大,为0.12 g油/g糖;C/N=32时油脂生成速率最大,为0.13 g/(L.h)。碳氮比对油脂的脂肪酸组成影响不明显,油脂的棕榈酸、硬脂酸和油酸总含量超过85%。  相似文献

Position of cells in their cell cycle was determined microscopically in chemostat cultures ofCandida utilis. Proportion of cells in phase G1 decreased in a linear manner from 86% to 58% with dilution rate. Proportion of cells in phase S increased in the same range ofD from 5.6 to 13.5% and in the (G2+M) phase from 8.4 to 28.5%, again linearly. Differential centrifugation was used to separate chemostat cultures to mother and daughter cells. Analyses showed that, relative to mother cells, daughter cells contain 2.1–11.9% more protein and 25.5–34.6% more RNA in dry matter. Their mass is 34.4–5.6% lower and volume is 154–19% smaller.  相似文献
