Neonicotinoid insecticides, such as imidacloprid, are selective agonists of insect nicotinic acetylcholine receptors (nAChRs) and are used extensively in areas of crop protection and animal health to control a variety of insect pest species. Here, we describe studies performed with nAChR subunits Nlα1 and Nlα2 cloned from the brown planthopper Nilaparvata  lugens , a major insect pest of rice crops in many parts of Asia. The influence of Nlα1 and Nlα2 subunits upon the functional properties of recombinant nAChRs has been examined by expression in Xenopus oocytes. In addition, the influence of a Nlα1 mutation (Y151S), which has been linked to neonicotinoid lab generated resistance in N. lugens , has been examined. As in previous studies of insect α subunits, functional expression has been achieved by co-expression with the mammalian β2 subunit. This approach has revealed a significantly higher apparent affinity of imidacloprid for Nlα1/β2 than for Nlα2/β2 nAChRs. In addition, evidence has been obtained for the co-assembly of Nlα1 and Nlα2 subunits into 'triplet' nAChRs of subunit composition Nlα1/Nlα2/β2. Evidence has also been obtained which demonstrates that the resistance-associated Y151S mutation has a significantly reduced effect on neonicotinoid agonist activity when Nlα1 is co-assembled with Nlα2 than when expressed as the sole α subunit in a heteromeric nAChR. These findings may be of importance in assessing the likely impact of the target-site mutations such as Y151S upon neonicotinoid insecticide resistance in insect field populations.  相似文献

An active intracellular to extracellular Ca2+ efflux has been proposed in heart muscle. A myocardial sarcolemmal ATPase stimulated by an intracellular pCa and serving as a Ca2+ pump has been postulated. A recently developed myocardial sarcolemmal preparation has not permitter a search for such an enzymatic activity. In these studies, an ATPase has been demonstrated in the sarcolemma which is activated by Mg2+, stimulated as the Ca2+ is raised to a pCa of 6, and is inhibited by ouabain. These findings suggest a mechanism by which Ca2+ within the myocardium may be modulated and thus how the force of contraction may be altered by cardiac glycosides.  相似文献

采用常规骨髓细胞空气干燥法,研究了贵州6种蝙蝠的核型。白腹管鼻蝠(Murina leucogaster)2n=44,染色体臂数(FN)为58;普通长翼蝠(Miniopterus schreibersi)染色体数是2n=46,FN为50,黄大蹄蝠(Hipposideros pratti)2n=32,FN为60;角菊头蝠(Rhinolophus cornutus)2n=62,FN为60;云南菊头蝠(R.yunnanensis)2n=44,FN是60;犬蝠(Cynopterus sphinx)2n=34,FN=58。其中白腹管鼻蝠、云南菊头蝠和犬蝠为国内首次报道。  相似文献

Rhodopseudomonas palustris is a species of purple photosynthetic bacteria that has a multigene family of puc genes that encode the alpha and beta apoproteins, which form the LH2 complexes. A genetic dissection strategy has been adopted in order to try and understand which spectroscopic form of LH2 these different genes produce. This paper presents a characterisation of one of the deletion mutants generated in this program, the pucBAd only mutant. This mutant produces an unusual spectroscopic form of LH2 that only has a single large NIR absorption band at 800 nm. Spectroscopic and pigment analyses on this complex suggest that it has basically a similar overall structure as that of the wild-type HL LH2 complex. The mutant has the unique phenotype where the mutant LH2 complex is only produced when cells are grown at LL. At HL the mutant only produces the LH1-RC core complex.  相似文献

Membrane mass spectrometry (MMS) with reduced sample withdrawal has been used to investigate the metabolic activity of yeast cells immobilised in porous glass. An adapted MS membrane inlet reactor with a polyethylene terephthalate barrier membrane has been constructed for this purpose. In a first experiment, the mass transport of O2 in a porous glass disc under well-defined experimental conditions has been studied by determining the apparent effective diffusion coefficient. The behaviour of immobilised Saccharomyces cerevisiae has been monitored by the MMS measurement of O2 and CO2 after applying a step in glucose concentration. Free-cell kinetic parameters were used in a dynamic reaction-diffusion model to simulate the O2 consumption curve. The theoretical and experimental curve showed comparable behaviour, which means that the immobilisation of yeast cells in porous glass has no substantial effect on its growth kinetics.  相似文献

The dimerization of 2 has been studied as a model for its activity in biological systems. Cation sensing behaviour of a pyrrole‐based derivative (2‐hydroxyl 3 methyl 6 isopropyl benzaldehyde}‐3,4‐dimethyl‐1H‐pyrrole‐2‐carbohydrazide (receptor 3) has been explored and is found to be selective towards Zn2+ over a variety of tested cations. The receptor 3 has shown high selectivity and sensitivity towards Zn2+ over the other alkali, alkaline earth and transition metal ions. In the presence of Zn2+, absorption band of receptor 3 has shown the red shift. The sensing behaviour has been suggested to continue via enhancement process which has further been supported by UV‐vis absorption and theoretical density functional theory (DFT) calculations indicating the formation of a 1:1 complex between the pyrrole based receptor 3 and Zn2+. The present work is presenting a highly selective dual channel colorimetric sensor for zinc with great sensitivity. The Sulfolobus solfataricus β-glycosidase (Sβgly) is a thermostable and thermophilic glycosyl-hydrolase with broad substrate specificity. The enzyme hydrolizes β-D-gluco-, fuco-, and galactosides, and a large number of /Winked glycoside dimers and oligomers, linked β1-3, β1-4, and β1-6, It is able to hydrolize oligosaccharides with up to 5 glucose residues. Furthermore, it is also able to promote transglycosylation reactions. The corresponding gene has been cloned and overexpressed both in yeast and Escherichia coli. Based on sequence and functional data, the Sβgly has been assigned to the so-called BGA family of glycosyl-hydrolases, including β-glycosidases, β-galactosidases and phosho-β-galactosidases from mesophilic and thermophilic organisms of the three domains. The Sβgly has been crystallized and the resolution of its structure is in progress. Because of its special properties, the enzymes has considerable biotechnological potential.  相似文献

The partial composition of O-antigens in Escherichia known as the causative agents of escherichioses localized in the intestine and other organs has been studied. Escherichia of groups 01, 02 and 03 are characterized by different partial composition of their O-antigens. This composition can be expressed by the formulae: 01a, 1b; 01a, 1b, 1c; 01a, 1b, 1c, 1d; 01a, 1e; 02a, 2b, 2c; 02a, 2b, 2c, 2d, 2e; 02a, 2b, 2d, 2e, 2g; 02a, 2c, 2e; 06a, 6b; 06a, 6c. No pronounced correlation between the partial composition of O-antigens within the O-group and the presence of different K- or H-antigens in the strain has been established. The scheme of adsorption has been developed and anti-Escherichia factor O-sera have been obtained. The use of these sera allows one to differentiate Escherichia strains, isolated from patients, within the O-groups according to the partial composition of their antigens, which is of both diagnostic and epidemiological importance.  相似文献

N-Nitroso-β-phenyl-β-lactam has been found to be a specific inhibitor of β-lactamase. N-Nitroso--phenyl-β-lactam, by contrast, was virtually ineffective although a transient inhibition of short duration was observed. The acyl enzyme derived from the β-phenyl isomer is presumably involved in a cross-linking reaction, whereas that from the -phenyl isomer was quenched by spontaneous hydrolysis without formation of a covalent bond. No inhibitory effect of the β-phenyl isomer on chymotrypsin has been observed.  相似文献

Summary A sarcoplasmic calcium-binding protein (SCP) has been purified from the muscle of the protochordate Amphioxus and shown to be more similar to invertebrate SCP's than to their counterpart found in vertebrates, i.e. parvalbumins. The Amphioxus protein has a pI of 4.9, is rich in tyrosine and tryptophan, has a molecular weight of 22,000 and binds strongly 2Ca2+ with a pK of 7.88. Magnesium competes with calcium for only one of the two metal-binding sites and induces positive cooperativity in Ca2+ binding.In cyclostome muscle (lamprey and hagfish), no protein with high affinity for Ca2+ or Mg2+ could be found, irrespective of molecular weight. Instead, a protein with moderate affinity for Ca2+ (105 m –1) was detected: it has a molecular weight of 60,000 and might be quite ubiquitous, as the presence of a similar protein has been reported both in red and white muscle of vertebrates such as chicken and rabbit.  相似文献

Phosphoinositol kinase (adenosine triphosphate-inositolmonophosphate phospho—tranferase) has been isolated from cotyledons; about 300-fold purification has been achieved, with a recovery of 11%. The enzyme has a pH optimum at 7·4. It can mediate phosphorylation of lower inositol phosphates to their corresponding higher homologues, ATP being the phosphate donor. ATP can be replaced partially by UTP and PEP. The enzyme requires divalent cations for the reaction. Mn2+ has been found to be twice as effective as Mg2+, Ca2+ being inhibitory. Phosphoinositol kinase has been found to be different from inositol kinase.  相似文献

The unusual catalytic network, revealed by the crystal structure of one of the two phospholipases A2 (PLA2) from the venom of the crotalid A.p.piscivorus has been probed using molecular dynamics. The catalytic network has been remodeled to a conformation similar to that found in all other PLA2, and the modeled structure has been submitted to energy minimization and molecular dynamics simulation, to explore the conformational space of the network. The calculations have yielded a large reorganization of the catalytic network, which gets a conformation close to that of the crystal structure. These results suggest that the unusual catalytic network observed in the studied PLA2 is a structural feature of the protein and not a crystal artifact.  相似文献

It has been demonstrated that the leaves of a range of foresttree species have responded to the rising concentration of atmosphericCO2 over the last 200 years by a decrease in both stomatal densityand stomatal index. This response has also been demonstratedexperimentally by growing plants under elevated CO2 concentrations.Investigation of Quaternary fossil leaves has shown a correspondingstomatal response to changing CO2 concentrations through a glacial-interglacialcycle, as revealed by ice core data. Tertiary leaves show asimilar pattern of stomatal density change, using palynologicalevidence of palaeo-temperature as a proxy measure of CO2 concentration.The present work extends this approach into the Palaeozoic fossilplant record. The stomatal density and index of Early Devonian,Carboniferous and Early Permian plants has been investigated,to test for any relationship that they may show with the changesin atmospheric CO2 concentration, derived from physical evidence,over that period. Observed changes in the stomatal data givesupport to the suggestion from physical evidence, that atmosphericCO2 concentrations fell from an Early Devonian high of 10-12times its present value, to one comparable to that of the presentday by the end of the Carboniferous. Excessive generation of reactive oxygen species (ROS) has been implicated in the pathogenesis of many diseases, including atherosclerosis, hypertension, and vascular complications of diabetes. However, the precise mechanisms by which ROS contribute to the development of these diseases are not fully characterized. Hydrogen peroxide (H2O2), a ROS, has been shown to activate several signaling protein kinases, such as extracellular signal-regulated kinase (ERK)1/2 and protein kinase B (PKB) in different cell types, notably in vascular smooth muscle cells. Because these pathways regulate cellular mitogenesis, migration, proliferation, survival, and death responses, their aberrant activtion has been suggested to be a potential mechanism of ROS-induced pathologies. The upstream elements responsible for H2O2-induced ERK1/2 and PKB activation remain poorly characterized, but a potential role of receptor and nonreceptor protein tyrosine kinases (PTKs) as triggers that initiate such events has been postulated. Therefore, the aim of this review is to highlight the involvement of receptor and nonreceptor PTKs in modulating H2O2-induced ERK1/2 and PKB signaling.  相似文献

Chromosomes of five subantarctic species were studied from mitoticmetaphases using cell suspension techniques. Among the Protobranchia,Malletiagigan-lea has a diploid chromosome number of 2n = 38with five metacentric, seven submetacentric, five sub-telocentricand one telocentric pairs, and Yoldia (Aequiyoldia) woodwardialso shows 2n = 38 but with ten metacentric, six submetacentricand three sub-telocentric pairs. Among the Lamellibranchia,the pterioidan Limatula pygmaea has 2n = 38 with six metacentric,eleven submetacentric and two sub-telocentric pairs, the veneroidanCyclocardia astar-toides has 2n = 30 with five metacentric andten telocentric pairs and the anomalodesmatan Laternula ellipticahas 2n = 40 with two metacentric, one submetacentric, two subtelocentricand fifteen telocentric pairs. Our results indicate that in all the Nuculoidea studied so far,a diploid number of 2n = 38 has been found. In addition, thekaryotypes show a close overall appearance in relative lengthswith a majority of meta-centric-submetacentric chromosomes.The species differ in the proportion of the different morphologicaltypes of chromosomes. The interaction of a series of 2-phenylquinoline derivatives with RNA was investigated by means of viscometric, pKa, spectroscopic, binding, Tm, and kinetic methods. Compounds 1 , 2 , and 3 have a piperazyl substituent at the para, meta, or ortho position, respectively, while 4 has an unsubstituted phenyl ring. The pKa results suggest that 1 has three charges, 2 and 3 have more than two charges, and 4 has two charges at pH 6.2. Spectroscopic and Tm results indicate that 1 binds more strongly to RNA than 2–4 . Kinetic and modeling results indicate that 1 is a threading intercalator while 2 and 4 are classical intercalators. All experimental results indicate that 3 , which has a large twist between the phenyl and quinoline rings, binds weakly with RNA. © 1994 John Wiley & Sons, Inc.  相似文献

Complementary DNA clones encoding a DNA-binding factor have been obtained from Arabidopsis by DNA hybridization with a GT-2 factor cDNA clone from rice. The GT-2 gene appears to be present as a single copy in the Arabidopsis genome and is transcribed as a 2.1 kb mRNA which is not light-regulated. The longest open reading frame in the sequenced clones predicts a protein of 65 kDa, beginning with the first in-frame methionine. The protein contains basic, acidic, and proline/glutamine-rich motifs and has significant amino acid sequence homology to the rice GT-2 factor, including three regions of 50–75 amino acids each of greater than 60% identity. Two of these regions are predicted to form similar trihelix structures postulated to be involved in selective binding to specific variations of a GT-box motif DNA sequence found in the promoter regions of several plant genes. Except for weak similarity to a tobacco GT-box binding factor, GT-1a/B2F, Arabidopsis GT-2 has no similarity to other sequences in the databases. DNA-binding studies show that Arabidopsis GT-2 has binding characteristics similar to those of the rice GT-2 factor, but dissimilar to those of the tobacco GT-1a/B2F factor. The data indicate that a DNA-binding factor containing domains of similar structure and target-sequence specificity has been conserved between monocots and dicots.  相似文献

Hexokinase II, one member of a family of structurally similar enzymes that catalyze the phosphorylation of glucose in the 6-position, has been suggested to play a role in the pathophysiology of noninsulin-dependent diabetes mellitus (NIDDM). The gene for hexokinase II, HK2, has been previously mapped to human chromosome 2p13 by fluorescence in situ hybridization, and two-point linkage analysis has placed it near the locus for transforming growth factor α, TGFA. We now report the characterization of a (TA)n polymorphism in intron 12 of HK2. Using multipoint analysis of CEPH family genotypes, we have determined the most likely locus order to be cen-D2S169-[D2S286-HK2]-[D2S145-D2S291]-[D2S45- D2S101-TGFA]-tel. As HKII is a candidate gene that could contribute to the manifestation of insulin resistance and NIDDM, we genotyped 1152 Pima Indians, a Native American tribe that has the highest reported prevalence of NIDDM in the world. Although we did not detect any linkage or association of HK2 with insulin resistance or NIDDM in the Pima Indians, the polymorphism and detailed mapping of HK2 described in this report should prove useful in the assessment of the role of this gene in the predisposition to NIDDM in other populations. The ubiquitously present β-amyloid peptide plays an important role in the pathogenesis of Alzheimer’s disease. Its neurotoxicity has been blamed on its mal-activity to increase calcium-levels. In the present study, we demonstrate that treatment of fibroblasts with β-amyloid has, in deed, resulted in a transient rise in the calcium-concentration. Chronic exposition of cultures to the peptide, however, caused a fall in the calcium-level. Apparently, β-amyloid has biphasic effects: acutely, it increases the calcium-concentration of cells; in contrast, on the long-run, β-amyloid peptide acts as a calcium-antagonist. Therefore, the idea that β-amyloid peptide leads to neural degeneration solely by increasing cells’ calcium concentration must be replaced with a more complex view of its dual function in intracellular ionic homeostasis.  相似文献
