Melanin plays an important role in protecting organisms from ultraviolet radiation (UVR). Therefore, it is possible that differently colored strains can show different sensitivities to UVR. In the present work, life span, fertility and courtship behavior of wild type (w), ebony (e) and yellow (y) strains of Drosophila melanogaster were studied to evaluate their sensitivity to ultraviolet (UV). Because a range of photo- toxic effects of UVR are mediated through generation of free radicals, levels of free radicals, lipid per- oxide (malondialdehyde, MDA) and superoxide dismutase (SOD) activity of three strains were examined to indicate their antioxidant defending ability and oxidative status. It was shown that w always had the highest lifespan and fertility not only in the control but also in UV-exposed groups. Moreover, lifespan and fertility of e were significantly higher (P<0.0001) than those of y in the UV-exposed groups, but not for the control. On the other hand, UV exposure had an adverse effect on courtship of flies. Stronger electron paramagnetic resonance (EPR) signals could be detected in w, e and y exposed to 5 min UV. And there were more significant changes of EPR signals in y than in w and e. UVR had no significant (P=0.1782) effect on the SOD activities. After pooling data from the control and UV-exposed groups, we found that w had a significantly (P<0.05) higher level of SOD activity, but e and y were nearly at the same levels (P>0.05). MDA levels were increased in the UV dose-dependent manner (P=0.0495). In con- clusion, our results suggested that UVR can decrease life span and fertility of flies and do harm to courtship, which may be due to oxidative damage to flies tissues (e.g. central nervous system) induced by free radicals. w had the highest tolerance to UVR, which may be ascribed to its advantage of survival under the natural condition and at high level of SOD activity. Then differences of pigment between e and y in absorbing UV, shielding against UV and scavenging free radicals produced by UVR should be responsible for their different sensitivity to UVR.  相似文献

Male-male courtship behavior was recently reported to be induced in large populations of Drosophila (e.g., 600–1500 flies) by ectopic expression of the white (w) gene. Little is known about the basis of this behavior; in male-female courtship, sensory cues are believed to play an important role. Previous data are consistent with the possibility that misexpression of w causes abnormal reception or processing of sensory information. We show here that w-induced male-male courtship occurs in isolated pairs of flies. Thus the behavior does not depend on sensory cues found only among large populations of flies, or on cues produced only by a small subset of such populations. This finding enabled quantitative analysis of mechanisms that underlie the behavior. Specifically, male-male courtship does not depend on the reception of olfactory information, nor on the reception or generation of auditory cues, as determined by surgical ablation of antennae, maxillary palps, or wings. Although the rapid onset of the behavior following w induction suggested that its basis could lie in a modulation of sensory physiology, we found visual, olfactory, and gustatory function to be normal in physiological or behavioral tests. The only sensory deprivation to produce an effect on male-male courtship was testing under dim red light; the percentage of flies courting another male was reduced to one-fourth of control values. A striking age dependence of the behavior is also documented: courtship between paired male mini-w+ flies was not observed in tests of very young (1-day-old) flies, but occurs at high levels between the ages of 1 and 4 weeks. © 1996 John Wiley & Sons, Inc.  相似文献

Summary Unequal crossing over occurs in females heterozygousw a /w a4 such that a deficiency and duplication can be recovered regularly. A triplication was also found following crossing over in the homozygous duplication.Genetic analysis of the duplication showed that of the several knownw loci only thew e andsp-w loci are duplicated.By means of crossing overw e andsp-w were introduced separately into the left half of the duplication andw e into the right half. The phenotypic consequences of such substitutions are interpreted in terms of the loci which are included in specific functional units or genes. They suggest thatw e andsp-w are in distinctive functional units.The fact that these duplications and deficiencies are without obvious affect on salivary gland band morphology suggests that the gene loci constitute only a small portion of the band structure.  相似文献

Cyanobacterial toxin production is allied to some unknown trigger resulting in the production of toxins such as microcystin. We hypothesize that microcystins serve as metal ligands to control bioavailability and toxicity of ambient metals. Since ultraviolet radiation (UVR) promotes photo-oxidation of organic metal ligands and influences trace metal bioavailability, the present study aimed to investigate the influence of UVR, Cu, and Zn on specific growth rates, biomass, photosynthetic capacity, and microcystin content in Microcystis aeruginosa. Two toxigenic strains of Microcystis were cultivated using either Lake Erie filtered water or a chemically defined medium, with realistic concentrations of Cu and Zn combined with natural or artificial UVR exposure. Cu was more toxic than Zn on the basis of free ion concentration of trace metals in synthetic medium, although in Lake Erie water total added Zn (10 nM) or Zn plus Cu (10 nM) had a more detrimental effect on biomass and specific growth rate. Natural UVR delivered at 25% ambient levels caused no decrease on the parameters measured (chlorophyll-a, photosynthetic rate), yet artificial levels of UVR (up to 5.9 μmol UVB photons m−2 s−1) negatively affected biomass and specific growth rate. Cellular levels of microcystin (per unit chlorophyll-a) were concomitant with specific growth rather than being triggered in response either of these stressors (UVR, Zn, and Cu) alone or in combination, in agreement with a purported constitutive production of microcystins.  相似文献

海滨滨麦叶片和根对不同厚度沙埋的生理响应差异分析   总被引:1,自引:0,他引:1  
以烟台海岸抗风沙植物滨麦为研究材料,通过对不同厚度沙埋下其叶片和根部抗氧化酶活力(超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT))、丙二醛(MDA)含量和渗透调节物含量变化的分析,探讨了叶片和根部对沙埋生理响应的差异。试验按滨麦成株株高(约40 cm)对其进行了轻度沙埋(在株高1/4处)、中度沙埋(2/4处)和重度沙埋(3/4处)。在沙埋第6天,分别测定了不同厚度沙埋处理下,植株各段叶片和根抗氧化酶活力、MDA和渗透调节物含量。结果表明,轻度和中度沙埋均加速植株生长。与对照相比,经轻度、中度沙埋处理6 d,叶片平均MDA含量增加,在重度沙埋下降低。不同厚度沙埋6 d,叶片平均SOD活力和脯氨酸含量增加,而CAT活力、可溶性糖和可溶性蛋白质含量下降。但不同厚度沙埋均使沙上叶片MDA、脯氨酸、可溶性蛋白质含量和SOD和CAT活力增加,尤其是叶片顶部增加最为明显,使沙下叶片MDA、可溶性糖、可溶性蛋白质含量和CAT活力下降,导致同株沙上和沙下叶片MDA、脯氨酸、可溶性糖、可溶性蛋白质含量和SOD和CAT活力差异显著(P0.05)。与叶片相比,根中MDA、可溶性蛋白质含量和SOD和CAT活力较低,而POD活力和可溶性糖含量较高并与叶片差异显著(P0.05)。不同厚度沙埋6 d,滨麦根中MDA和可溶性蛋白质含量变化较小,可溶性糖含量和CAT、POD、SOD活力略有降低。研究表明,滨麦根和叶片对不同厚度沙埋的生理响应不同。沙埋直接作用于叶片并诱发叶内氧自由基积累,但叶片通过快速激活的抗氧化酶保护系统(CAT、SOD)维持氧自由基代谢平衡,以及渗透调节物(脯氨酸、可溶性糖)的积累维护细胞水分代谢平衡,并满足能量的需求和快速生长。但在不同厚度沙埋下,由于根系不受沙埋直接影响而生理变化较小,并且还维持较低的膜脂过氧化水平,这可能是根能维持正常的吸水输水功能并在沙埋处理过程中和沙埋后地上叶片快速生长摆脱沙埋的重要物质基础。  相似文献

Juvenile hormone (JH) involvement in male reproduction is poorly understood. In Drosophila melanogaster adults, JH deficiency has been shown to result in lowered protein synthesis in male accessory glands. To probe additional roles, we have examined males homozygous for a null allele of Methoprene-tolerant (Met). This gene is involved in the action of JH, possibly at the JH receptor level, and Met27 null mutants reflect a diminution of JH action. Met27 males were found to have reduced protein accumulation in male accessory glands and to court and mate wild-type females much less avidly than do either Met+ or Met27; Met+ transgenic males. Exposure of Met27 males to methoprene partially rescued the courtship deficiency. However, sperm transfer as reflected by fertility of Met27 fathers was found to be similar to that of Met+. Taken together with previous work examining the JH-deficient mutant apterous, these results corroborate JH involvement in protein synthesis in the male accessory glands and suggest a role for JH in promoting male mating behavior in these flies.  相似文献

Summary Mitomycin C was injected into the abdomen of male flies of the y 2 sc1 waG strain of Drosophila melanogaster. They were mated with females bearing attached-X chromosomes, and the male offspring (F1) were analysed for the appearance of mutations in the X chromosome. We observed y 1 and sc + reversions induced either by excision of mdg4 (gypsy) with retention of one long terminal repeat (LTR) or by insertion of a foreign sequence into mdg4, partial reversion of the w aG mutation, w aGw aGd, and unstable f mutations. The overall mutation frequency was considerably higher than in control flies of the y 2 sc1 waG strain. Possible mechanisms of genomic rearrangements induced by Mitomycin C, in particular the role of homologous recombination, are discussed.  相似文献

The ref(2)P gene of Drosophila melanogaster is implicated in sigma rhabdovirus multiplication. Two common alleles of ref(2)P are known, ref(2)P 0 which permits sigma virus multiplication and ref(2)P pwhich is restrictive for most sigma virus strains. This gene maps to the cytogenetic region 37E3-F3. Using Df(2L)E55 (=Df(2L)37D2-El;37F5-38A1), we have screened for lethal, semi-lethal and visible mutations following diepoxybutane (DEB) or ethyl methanesulfonate (EMS) mutagenesis. Our data confirm than DEB is mor efficient than EMS at inducing deletions. The mutations obtained in this region define 14 complementation groups. One of them, l(2)37Dh, appears to be a general enhancer of Minute and Minute-like mutations. None of the mutations were allelic to the ref(2)P locus. Loss-of-function alleles of ref(2)P (called null) were selected following DEB mutagenesis. Homozygous or hemizygous ref(2)P nullflies are male sterile. These flies, like homozygous or hemizygous ref(2)P 0flies, are fully permissive for sigma virus replication. We suggest that the ref(2)P products interact with viral products, but that this interaction is not necessary for an efficient viral cycle.  相似文献

为了比较研究酸雨与紫外辐射对淡水水体常见藻华蓝藻的生理学影响,选取铜绿微囊藻(Microcystis aeruginosa)产毒(FACHB-905)与不产毒(FACHB-469)株系作为实验材料,通过人工模拟酸雨,研究了不同p H处理后2藻株的光合生理变化以及对紫外辐射的敏感性的异同。实验设置3个p H梯度,p H7.10为对照组(正常培养基培养的藻体),两模拟酸雨处理组(p H5.65和p H4.50);两种辐射处理,可见光处理(PAR)以及全波长辐射处理(PAB)。研究结果表明,905藻株细胞粒径在各p H处理下都要显著高于469藻株,模拟酸雨处理显著降低了两藻株细胞的平均粒径及体积,但叶绿素含量显著提高;酸雨处理同时也引起细胞死亡率的增加,表现为藻体有效光化学效率显著降低,生长速率显著受到抑制,低p H下呈负增长,且这种抑制程度在469下更为显著。高的可见光以及紫外辐射处理,使两株系有效光化学效率随p H的降低而呈降低趋势,其中469藻株降低至更低的水平,且高光辐射以及紫外诱导的抑制率要显著高于905藻体,这可能与469藻株较低的光保护色素有关(较低的类胡萝卜素以及紫外吸收物质)。在未来全球变化背景下,不同种类的浮游植物对环境变化的响应及适应能力不同,可改变水体的群落结构和种群丰度,铜绿微囊藻905较469较强的耐受酸雨以及紫外辐射的能力,可能会使该株系在竞争力上占据优势。  相似文献

通过对黑麦草(Lolium perenne L)在轻度、中度、重度、全割刈割处理6 d和12 d后,残留叶片和叶片再生部分生长速率,超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活力,丙二醛、可溶性糖、脯氨酸含量的分析以揭示在刈割胁迫后叶片抗氧护酶活力和渗透调节物含量与其补偿性生长的关系,以及牧草耐刈性的生理调控机理。结果表明,轻度和中度及全割后叶片生长速率均高于对照,重度刈割低于对照。全割后叶片补偿性生长最明显、轻度和中度次之,重度刈割无补偿性生长。对照黑麦草叶片各部位抗氧化酶和渗透调节物含量不同,叶片顶部MDA含量较高,伴随着较高的SOD、CAT活力和较高的脯氨酸含量;叶片基部MDA含量最低,SOD、CAT活力及脯氨酸含量也较低。与对照相比,不同强度刈割6 d黑麦草再生叶和叶片平均MDA含量、SOD和CAT活力、可溶性糖和脯氨酸含量均较低。而不同强度刈割12 d,黑麦草再生叶和叶片平均MDA含量仍较低,但SOD和CAT活力增高,脯氨酸含量增加,POD活力和可溶性糖含量低于对照。这表明刈割在减少了叶面积,降低光合能力的同时,刈割伤害胁迫启动了牧草补偿性生长使残留叶片快速生长,而且残留叶片面积与其补偿生长速率成正相关。另外,虽然不同强度刈割下叶片补偿性生长速率不同,但不同强度刈割(12 d)均激活残留叶片抗氧化保护酶系统和促进脯氨酸积累。在补偿生长过程中,CAT和SOD能及时清除残留叶片中积累的氧自由基,维持较低的膜脂过氧化和细胞膜完整性,积累的脯氨酸能维护细胞水分平衡。因此,抗氧化酶(SOD和CAT)和渗透调节物(脯氨酸)在黑麦草刈割后受伤部位快速自愈及残留叶片快速补偿生长中起重要生理保护作用。  相似文献

Summary The w m Co duplication of Drosophila hydei (Dp (1; Y) 16B2-17B1) contains 13–16 bands in salivary gland chromosomes. The duplication resides preferentially in the X heterochromatin or on the Y chromosome. In some stocks frequent (up to 4×10-3) exchanges of the duplication occur between different Y chromosomes (T(X; Y) and free Y) or between the X and the Y chromosome. About 60% of the T(X; Y)-Y exchanges induce mutations in the Y chromosomal male fertility genes of the recipient Y chromosome. From the mutational spectrum generated by the T(X; Y)-Y transpositions and from the variable efficiency as acceptor of different X-Y translocations it can be concluded that the exchanges show a remarkable site specificity: distal positions in the long arm of the Y chromosome are occupied preferentially. More proximal positions in the long arm of insertions into the short arm of the Y chromosome are found only with a lower frequency. No transpositions to the autosomes have been recovered. Duplications are lost with highly differing frequencies. The losses are not linked with insertions of the w m Co element into a new position and are more frequent than transpositions. Therefore, we regard the w m Co element as a giant transposon.  相似文献

D. birchii and D. serrata, two endemic Australian Drosophila species, have a copulatory courtship. The males of these species begin to court the female after mounting her and often go on with the courtship after the copulation is over. In the present paper we have described behavioral interactions between the male and the female and analyzed acoustic signals produced by the flies during courtship. Species differences were more pronounced in female than in male behavior. Variation within the species was obvious in the relative proportions of time the flies spent in different behaviors. Even though courtship took place nearly solely during copulation, some remains of precopulatory courtship were observed in both species. It is suggested that copulatory courtship exhibited by D. birchii and D. serrata flies is a derived rather than a primitive character.  相似文献

UV RESISTANCE LOCUS8 (UVR8) is a photoreceptor for ultraviolet‐B (UV‐B) light that initiates photomorphogenic responses in plants. UV‐B photoreception causes rapid dissociation of dimeric UVR8 into monomers that interact with CONSTITUTIVELY PHOTOMORPHOGENIC1 (COP1) to initiate signal transduction. Experiments with purified UVR8 show that the dimer is maintained by salt‐bridge interactions between specific charged amino acids across the dimer interface. However, little is known about the importance of these charged amino acids in determining dimer/monomer status and UVR8 function in plants. Here we evaluate the use of different methods to examine dimer/monomer status of UVR8 and show that mutations of several salt‐bridge amino acids affect dimer/monomer status, interaction with COP1 and photoreceptor function of UVR8 in vivo. In particular, the salt‐bridges formed between arginine 286 and aspartates 96 and 107 are key to dimer formation. Mutation of arginine 286 to alanine impairs dimer formation, interaction with COP1 and function in vivo, whereas mutation to lysine gives a weakened dimer that is functional in vivo, indicating the importance of the positive charge of the arginine/lysine residue for dimer formation. Notably, a UVR8 mutant in which aspartates 96 and 107 are conservatively mutated to asparagine is strongly impaired in dimer formation but mediates UV‐B responses in vivo with a similar dose–response relationship to wild‐type. The UV‐B responsiveness of this mutant does not correlate with dimer formation and monomerisation, indicating that monomeric UVR8 has the potential for UV‐B photoreception, initiating signal transduction and responses in plants.  相似文献

铜锌复合污染对铜富集植物大聚藻抗氧化酶活性的影响   总被引:1,自引:0,他引:1  
以前期筛选的铜富集植物大聚藻为材料,采用两因素随机区组试验设计,通过盆栽试验研究了不同浓度铜锌复合污染对大聚藻抗氧化酶活性的影响,以揭示铜富集植物大聚藻对重金属的耐性机理,为芦溪河及其它类似污染河流的生态恢复与植被重建提供参考依据。结果表明:(1)铜锌复合污染条件下,大聚藻生物量都表现出低促高抑现象。(2)铜锌复合污染时,大聚藻MDA含量随铜锌浓度升高表现出先升高后降低的变化。(3)铜锌复合污染对大聚藻抗氧化酶系统活性均有不同程度的影响,低浓度铜锌复合污染对SOD(超氧化物歧化酶)、POD(过氧化物酶)和CAT(过氧化氢酶)有促进作用,而随浓度的升高则表现出不同的规律。研究发现,铜锌胁迫下,大聚藻细胞应急防御系统被启动,SOD、POD和CAT发挥作用,体内过量自由基及时被清除,使大聚藻能够保持高的耐性。  相似文献

The blue throat feathers of male bluethroats (Luscinia s. svecica) show a reflectance peak in the ultraviolet (UV) waveband (320 to 400 nm). The throat is actively displayed during courtship, suggesting a role for sexual selection on an ultraviolet signal. Indeed, a recent aviary experiment demonstrated that females discriminated against males with artificially reduced UV reflectance (Andersson and Amundsen 1997). Here, we report the results of a similar experimental manipulation applied on free-ranging males. UV-reduced (UVR) males had a lower success in attracting mates, as judged from a significantly later start of egg laying, compared with control (C) males. UVR males also spent significantly less time advertising for additional mates when their own mate was fertile, and they had a lower success in achieving extra-pair fertilizations. Furthermore, UVR males tended to guard their mates more closely and lose more paternity in their own brood than C males did. We conclude that the treatment affected both social and extra-pair mate choice. This is the first experimental evidence that UV signalling influences male mating success in free-ranging birds.  相似文献

Little is known about the UV photobiology of psychrophilic dinoflagellates, particularly in freshwater systems. We addressed the life strategies of Borghiella dodgei Moestrup, Gert. Hansen et Daugbjerg to cope with ambient levels of ultraviolet radiation (UVR) under cold conditions. Several physiological parameters related to growth, metabolism, and UVR protection were determined for 4 d in UVR‐exposed and control cells by applying stable isotope analysis, spectrophotometry, and liquid chromatography–mass spectrometry (LC/MS). In UVR‐exposed cells, assimilation of 15N and 13C and content of chl a and carotenoids, specifically diatoxanthin with respect to dinoxanthin and diadinoxanthin, were increased; furthermore, catalase activity showed a cyclic pattern with a strong increase after UVR exposure but a rapid return to preexposure levels. Both in UVR‐exposed and control cells, no lipid peroxidation of galactolipids was observed. However, in UVR‐exposed cells, content of galactolipids was higher and linked to an increase in monogalactosyldiacylglycerols (MGDGs). We concluded that Borghiella's adaptation to UVR depended on a general metabolic enhancement and efficient scavenging of oxygen radicals to mitigate and counteract damage. While Borghiella seemed to be well adapted to ambient UVR, the interactive effects of higher temperature and UVR on psychrophilic species in front of climate change merit further investigation.  相似文献

The brown macroalga Laminaria saccharina (L.) J. V. Lamour. was grown in large outdoor tanks at 50% ambient solar radiation for 3–4 weeks in July and August of 2000, 2001, and 2002, in either ambient or nitrogen (N)–enriched seawater and in either ambient light [PAR + ultraviolet radiation (UVR)] or ambient light minus UVR. Growth, N‐content, photosynthetic pigments, and RUBISCO content increased in N‐enriched seawater, indicating N‐limitation. UVR inhibited growth, but this inhibition was ameliorated by N‐enrichment. The response of growth to UVR could not be explained by changes in respiration and photosynthesis. Gross light‐saturated photosynthesis (Pmax) remained unaffected by UVR but was significantly higher under N‐enrichment, as was dark respiration (Rd). UVR had no effect on pigments or N content. However, RUBISCO contents were low in the presence of UVR, reflecting the overall change in soluble cellular protein. Overall, our data indicate that the response to UVR in L. saccharina depends on other environmental factors, such as N, and these effects need to be considered when evaluating the response of macroalgae to increased UVR.  相似文献

马麝(Moschus sifanicus)分布于我国青藏高原及周边区域,雄麝可分泌麝香。因历史上的过度利用及生境丧失等原因,马麝已极度濒危。马麝驯养是保育野生马麝资源及可持续生产麝香的有效方式。我国于1990年在甘肃兴隆山保护区开始马麝驯养,并于1996年实现了可持续的活体取香。分析了兴隆山麝场1996—2006年间的麝香生产及与泌香雄麝种群增长和种群结构的关系,结果表明:兴隆山麝场11a间共取香430头次,麝香总产量达3846.6 g,年均麝香产量为(349.69±84.69)g(n=11)。泌香雄麝的种群数与麝香年产量存在极显著的相关(R=0.638,P0.01),增长模型y=e(6.4285-3.6578/t)(R2=0.735,df=9,F=24.94,P=0.0010.01)和y=e(4.2049-3.4523/t)(R2=0.700,df=9,F=21.02,P=0.0010.01)可分别模拟该麝场的麝香年产量及泌香雄麝种群的增长。泌香雄麝的平均泌香量为(8.93±0.56)g/头(n=68),各年度的雄麝平均泌香量与麝香年产量相关极显著(R=0.442,P0.01),其增长模式呈指数式增长(y=7.5126e0.0244t)(R2=0.373,df=9,F=5.36,P=0.0460.05)。各年龄组雄麝间的平均泌香量差异显著(ANOVA,F7,59=2.522,P=0.0240.05),1.5岁马麝的泌香(2.00±1.82)g,(n=10)显著高于其他各年龄组马麝(P0.05),其余年龄组间的平均泌香量无显著差异(P0.05)。雄麝的年龄组与麝香年产量呈极显著的负相关(R=-0.936,P=0.0010.01),1.5—6.5岁雄麝占种群比例91.16%,生产麝香(3560.1g)占总产量的92.55%。以麝香生产为主的麝场,其驯养雄麝种群配置应以6.5岁龄以下雄麝为主。  相似文献

Contact chemosensation is required for several behaviors that promote insect survival. These include evasive behaviors such as suppression of feeding on repellent compounds, known as antifeedants, and inhibition of male-to-male courtship. However, the gustatory receptors (GRs) required for responding to nonvolatile avoidance chemicals are largely unknown. Exceptions include Drosophila GR66a and GR93a, which are required to prevent ingestion of caffeine [1] and [2], and GR32a, which is necessary for inhibiting male-to-male courtship [3]. However, GR32a is dispensable for normal taste. Thus, distinct GRs may function in sensing avoidance pheromones and antifeedants. Here, we describe the requirements for GR33a, which is expressed widely in gustatory receptor neurons (GRNs) that respond to aversive chemicals. Gr33a mutant flies were impaired in avoiding all nonvolatile repellents tested, ranging from quinine to denatonium, lobeline, and caffeine. Gr33a mutant males also displayed increased male-to-male courtship, implying that it functioned in the detection of a repulsive male pheromone. In contrast to the broadly required olfactory receptor (OR) OR83b, which is essential for trafficking other ORs [4], GR66a and GR93a are localized normally in Gr33a mutant GRNs. Thus, rather than regulating GR trafficking, GR33a may be a coreceptor required for sensing all nonvolatile repulsive chemicals, including tastants and pheromones.  相似文献
