Recently, we displayed an Iberian shrew species (Sorex granarius) with telomere structures unusual for mammals. Long telomeres on the short acrocentric arms contain an average of 213 kb of telomere repeats, whereas the other chromosomal ends have only 3.8 kb (Zhdanova et al., 2005; 2007). However, it is not clear whether these telomeres are typical for all shrew species or only for S. granarius. S. granarius and common shrew Sorex araneus are sibling species. In this study, using modified Q-FISH we demonstrated that telomeres in S. araneus from various chromosomal races distinguished by their number of metacentrics contain 6.8–15.2 kb of telomeric tracts. The S. araneus telomere lengths appear to correspond to telomere lengths in the majority of both shrew species and wild mammals, whereas S. granarius has telomeres with unique or rare structures. Using DNA and RNA high-specific modified probes to telomeric repeats (PNA and LNA), we showed that interstitial telomeric sites in S. araneus chromosomes contain mainly telomeric DNA and that their localization coincide with some evolutionary breakpoints. Interstitial telomeric DNA in S. granarius chromosomes was not revealed. Thus, the distribution of telomeric DNA may be significantly different, even in closely related species whose chromosomes are composed of almost identical chromosomal arms.  相似文献

The ultrastructure of polytene chromosomes of Drosophila and Stylonychia were compared in whole-mount spread preparations. In Drosophila the chromomeres appear as dense, unresolvable structures interconnected by 10-nm interband fibers. In contrast, chromomeres of Stylonychia polytene chromosomes are formed by aggregates of 30-nm loops laterally attached to 10-nm interband fibers. It is suggested that the polytene chromosomes in these two species are analogous rather than homologous structures.  相似文献

The largely Mexican genus Echeveria is characterized by an extensive series of dysploid chromosome numbers, with every gametic number from 12 to 34 known in at least one species. Within this nearly three-fold range of numbers, the boundary between diploidy and tetraploidy is not immediately apparent. However, species of Echeveria can be hybridized in an extraordinary number of combinations, both among themselves and with related genera, and study of the morphology of the hybrids and the pairing of their chromosomes provides information that helps to identify the ploidy of the parents. This paper reports observations from study of 80 hybrids between E. ciliata (n = 25) and 73 other species and/or cytotypes. Hybrids between E. ciliata and definite diploids are all nicely intermediate morphologically, whatever the chromosome numbers. In these same hybrids, most chromosomes become involved in pairing at meiosis, and the number of paired elements (bivalents and multivalents) approaches or equals, but never exceeds, the number of chromosomes received from the lower-numbered parent. In most cells, relatively few univalents are present, sometimes none. These observations are considered to indicate that all paired elements include at least one chromosome from each parent and therefore that pairing occurs between chromosomes of different parents only (allosyndesis). Since none of the 25 gametic chromosomes of E. ciliata is able to pair with any other, although they do pair very extensively with chromosomes from many other species having a wide range of numbers, E. ciliata is considered to be diploid in spite of its relatively high chromosome number. On the other hand, hybrids of E. ciliata with definite polyploids resemble the latter much more closely in their morphology, and at meiosis most or all pairing occurs by autosyndesis between chromosomes received from the polyploid parent, while the chromosomes from E. ciliata generally remain unpaired. In these respects most, but not all, species of Echeveria having as many as 34 gametic chromosomes have the same properties as E. ciliata and also are considered to be diploid. The ancestral chromosome number in the genus is not clear, but it is probably near the upper end of the series of dysploid numbers.  相似文献

Morphometric karyotype characters were studied in 25Angelica spp. (Umbelliferae, Apioideae) and in one species of the related genusTommasinia. For three species the chromosome numbers are new. In our study the majority of the species investigated are diploids with 2n = 22, some are tetraploids with 2n = 44 (for these tetraploids also diploid cytotypes are reported in the literature). Among the diploid species,A. miqueliana has a distinct karyotype consisting of submetacentric and acrocentric chromosomes only, the remaining diploids with 2n = 22 as well as tetraploids with 2n = 44 have rather symmetrical karyotypes, consisting of metacentric and submetacentric chromosomes. The very different chromosome number 2n = 28 has been found inA. gmelinii. Its karyotype includes two distinct groups of chromosomes: 8 pairs of rather large metacentrics and submetacentrics and 6 pairs of very short and asymmetrical chromosomes. Chromosome numbers and structures appear to be useful in the taxonomy of some intrageneric taxa inAngelica.  相似文献

Earlier karyotypical work on Nodipecten nodosus embryos indicated that this species has a diploid number of 38, with six pairs respectively of metacentric and submetacentric chromosomes and seven pairs of subtelocentric chromosomes, although there were some difficulties in obtaining complete metaphases. The present work provides additional results on specific regions of the chromosomes in N. nodosus and, by meiotic studies, confirms the chromosome number with more reliability. Active nucleolar organizer regions (NOR), detected in mitotic metaphases from embryos, can be characterized in N. nodosus by a high level of heteromorphism of NOR-sites, indicating that these regions are not appropriate as chromosomal markers in this species. The procedure for detecting constitutive heterochromatin of chromosomes allowed us to observe most of the heterochromatic blocks at a pericentromeric position and some at telomeric and interstitial positions. The analysis of meiotic chromosomes from gonad tissue revealed the presence of 19 bivalents during metaphase I, all homomorphic and isopicnotic, confirming the previously described diploid chromosomal number of 38 for N. nodosus. From these results, some evolutionary aspects of the Pectinidae are briefly discussed.  相似文献

Acetocarmine squashes of root tips have demonstrated that 2n = 20 and 38 in Saxifraga virginiensis. These contrast with the earlier reported count of 2n = 28 for this species. In several populations supernumerary chromosomes were detected. Both intrapopulational and interpopulational variation in supernumerary chromosome number were detected, with the largest number of supernumerary chromosomes observed being six. Because these supernumerary chromosomes are equal in size to many of the smaller A chromosomes during mitotic metaphase, the presence of supernumerary chromosomes in this species could not be ascertained by analysis of mitotic metaphase preparations alone. During mitotic prophase, however, the supernumerary chromosomes of S. virginiensis are highly heterochromatic, appearing more densely coiled and darkly stained than the A chromosomes. This characteristic facilitated the recognition of supernumerary chromosomes in this species. The similarity in size of A and supernumerary chromosomes during mitotic metaphase and the observation of six supernumerary chromosomes in one population suggest that the count of 2n = 28 reported earlier for S. virginiensis may actually be a misinterpretation of 2n = 20 plus 8 supernumerary chromosomes. Furthermore, these findings and the observation of this same supernumerary chromosome phenomenon in other species of Saxifraga raise the possibility that some of the many disparate chromosome counts attributed to aneuploidy in the large genus Saxifraga may also be the result of misinterpretations of supernumerary chromosomes as A chromosomes.  相似文献

Photo-maps of the fat body and midgut polytene chromosomes of Drosophila auraria were constructed. These photo-maps are compared with a new, more detailed photo-map of the salivary gland chromosomes of the same species. Seven, not previously described inverted tandem-duplications were detected, raising the number of such structures found in this species to 31. The constancy of the banding pattern based on the analysis of the above chromosomes is discussed.  相似文献

Of 21 allotriploid males that possessed two genomes of Rana nigromaculata and one genome of Rana lessonae 10 produced a large number of spermatozoa in their testes. When 4 of these males were backcrossed with a female of R. nigromaculata, all of the resulting froglets were diploid in chromosome number and were completely R. nigromaculata type in appearance. These allotriploid males proved to have produced spermatozoa with one R. nigromaculata genome hybridogentically. Therefore, their germ line cells were investigated for the mechanism of elimination of their R. lessonae chromosomes. In histologicla sections of testes, the great majority of spermatogonia (approximately 104 cells) between mitotic prometaphase and anaphase appeared normal in chromosome behavior, whereas 17 spermatogonia showed several chromosomes whose behavior deviated from the normal course during the same period. These deviant chromosomes concentrated together near the equatorial plate and remained stationary at anaphase. In metaphase chromosome preparations made from spermatogonia, 67 and 185 of the 477 chromosome spreads were diploid and triploid, respectively. The rest were aneuploid. Notably, 8 triploid spreads consisted of 26 or more normal chromosomes and 13 or fewer degenerate chromosomes. From these results it is concluded that a set of R. lessonae chromosomes is eliminated from some, but not all spermatogonia by becoming degenerate during the mitotic period.by H.C. Abstract

Among Angiosperms, only six species are known to possess the lowest identified chromosome number, i.e. 2n = 4. These plants are the monocotyledons Zingeria biebersteiniana, Colpodium versicolor, Ornithogalum tenuifolium and Rhynchospora tenuis, and the dicotyledons Haplopappus gracilis and Brachyscome dichromosomatica. The low chromosome number may be cytogenetically derived from a different ancestral basic number, characteristic of each genus, by different processes, including tandem fusion or unequal reciprocal translocations with loss of centric fragments. All these plants possess low nuclear 4C DNA contents, ranging from 1.56 pg (R. tenuis) to 8.20 pg (H. gracilis), and they generally display a similar chromosome size and a similar position of the nucleolus organising region (NOR), that is often located in the terminal or subterminal region of the small chromosome pair. All these characteristics could be a consequence of common adaptative mechanisms. Peculiar characteristics within these karyotypes are the holocentric chromosomes of R. tenuis and the presence of B chromosomes in B. dichromosomatica. Plants with a very low chromosome number may be considered to constitute a fascinating “island of interest”; moreover, they represent simple systems helpful for the examination of the structural organisation and evolution of Angiosperm chromosomes.  相似文献

The lampbrush chromosomes of the long-toed salamander, Ambystoma macrodactylum Baird, have been analysed and a map of the oocyte genome prepared. The location of C-bands and cold-induced-constrictions has been established in mitotic chromosomes and compared with the location of marker structures and chiasmata in several lampbrush bivalents. In the lampbrush chromosomes, C-bands are tentatively correlated with sphere-organizing loci and with regions of low chiasma frequency; cold-induced-constrictions are tentatively correlated with regions of high chiasma frequency. In general, in this salamander, C-bands do not coincide in position with cold-induced-constrictions. We have compared our results with those obtained by Callan (1966) in his investigation of chromosomes of the axolotl, Ambystoma mexicanum, and we present an analysis of the similarities and differences that are visible in the chromosome sets of these two ambystomatid species.  相似文献

Every chromosome number from n = 12 to n =34 and also many higher numbers are known in one or more of the 130+ species of Echeveria, and the numerical boundary between diploids and tetraploids is not immediately apparent. Echeveria also is extraordinary for the number and diversity of hybrids that it can produce in cultivation, both within the genus and with species of several related genera. In 42 collections studied, the morphologically and cytologically variable E. secunda of central Mexico has n = 30-32, often with one or more B-chromosomes, and some quadrivalents are formed at meiosis in nearly every cell. Twenty-four hybrids of E. secunda, with 22 species or cytotypes considered diploids, resemble the former much more closely in appearance, and at meiosis 15-16 paired elements (bivalents and multivalents) are formed, never more, regardless of the number of chromosomes, 12 to 34, that were received from the other parent. It is concluded that the 15-16 paired elements in these hybrids are formed by the 30-32 chromosomes received from E. secunda, and that most chromosomes from the other parents occur as univalents, although usually a few associate with pairs from E. secunda to produce multivalents. Hybrids of E. secunda with 11 definitely tetraploid species having n = 34 to n = 68 are nicely intermediate in morphology between their parents, form mostly or entirely bivalents at meiosis, and most, probably all, including five intergeneric hybrids, are fertile. These observations are all consistent with the conclusion that E. secunda is an autotetraploid, even though no plants of the species having n = 15 or 16 have been found, and even though some other species of Echeveria having as many as 34 gametic chromosomes appear to be effectively diploid. Observations on pollen stainability and on second-generation hybrids are all compatible with this conclusion. The high chromosome numbers in many Mexican Crassulaceae that are now effectively diploid may have originated as polyploids that have become diploidized by mutation, loss, or suppression of duplicated chromosomes, segments, and genes. Hybrids of E. secunda, with three other species that appear to be tetraploids, have less regular meiosis, apparently because all of the chromosomes from the other parents do not regularly form pairs in the hybrids. A cultivar ofChlorophytum elatum var.variegatum (spider plant, 2n=28) from Agra, India, is characterized by one, two or three supernumerary divisions of the cells of the microspore tetrad. During these divisions the chromosomes neither synapse nor split, nor are any spindles formed. The distribution of chromosomes is random, with the result that the microspores vary considerably in chromosome number. Only a small percentage of the pollen produced has the full haploid set. Abstract

Twenty-eight moss species have been investigated cytologically. Mitotic karyograms have been presented for most of these species and these show a remarkable similarity in the n = 11 karyotype. Nine of the eleven chromosomes have terminal or sub-terminal centromeres. Lightly staining heterochromatic bands frequently occur along the axis of the chromosomes and very often these are the sites of chromosome bending; it has been suggested that these may be areas of neocentric activity. The karyotypes investigated show a reduction series of chromosome number which is paralleled by an increase in the number of long metacentrics; since the number of major chromosome arms is maintained in most of the species it has been suggested that Robertsonian fusion has been an important mechanism in the evolution of the Bracytheciaceae, and probably in all Diplolepidous mosses. Polyploidy has also played an important role in speciation. Finally it has been proposed that n = 11 is the primary basic number in the Diplolepideae.  相似文献

Lycopersicon esculentum accessions bearing fasciated (multiloculed) fruit were characterized based on their flower organ and locule number phenotypes. Greenhouse and field evaluations indicate that increases in locule number are associated with increases in the number of other floral organs (e.g., sepals, petals, stamens) in all stocks. F1 complementation, F2 segregation analysis, and genetic mapping indicate that at least four loci account for increases in the number of carpels/locules in these stocks. The most significant of these map to the bottoms of chromosomes 2 and 11 and correspond to the locule number and fasciated loci. All stocks tested were fixed for mutations at the fasciated locus, which maps to the 0.5-cM interval between the markers T302 and cLET24J2A and occurs in at least three allelic forms (wild type and two mutants). One of the fasciated mutant alleles is associated with nonfused carpels and repressed recombination and may be due to a small inversion or deletion. The other two loci controlling locule number correspond to the lcn1.1 and lcn2.2 loci located on chromosomes 1 and 2, respectively.  相似文献

该研究首次报道了滇羽叶菊和台湾刘寄奴的染色体数据。结果表明:两者的染色体数量都为48,核型公式均为2n=2x=36m+10sm+2st,与前人报道的刻裂羽叶菊的核型稍有不同。两者的染色体形态均由大到小逐渐变化,核型二型性不明显,但前者染色体明显比后者大。这说明羽叶菊属染色体基数确实应为x=24,且染色体大小在种间有较大差异。细胞学证据表明,该属与蒲儿根属中染色体基数为x=24的类群以及狗舌草属确实近缘。  相似文献

Southern blot analysis of the human acrocentric chromosomes that were flow-sorted from B-lymphoblastoid cell line GM130B revealed that the sensitivity of the ribosomal RNA (rDNA) gene clusters to the restriction enzyme NotI differs among these rDNA-containing chromosomes: the rDNA clusters of Chromosomes (Chr) 13, 14, and 15 are much more sensitive to NotI digestion than those of Chrs 21 and 22 in this particular cell line. Detailed analysis by use of methylation-sensitive enzymes HpaII and HhaI and methylation-insensitive enzyme MspI confirmed the significant variation in the methylation status of rDNA clusters among these chromosomes. Quantitative analysis by fluorescent in situ hybridization (FISH) indicated that copy number of rDNA varies among individual chromosomes, but the average copy number in the acrocentric Chrs 21 and 22 is significantly greater than that of the Chrs 13, 14, and 15 in GM130B cells. Similar analysis reveals that the methylation status of rDNA clusters in another B-lymphoblastoid cell line GM131 was different from that of GM130B. These data together indicate that the copy number and methylation patterns of rDNA clusters differ among individual acrocentric chromosomes in a given cell line, and they are different among cell lines.  相似文献

The karyotype of chars from the Estikhed Lake (Eastern Chukotka) was examined. This karyotype comprises 78 chromosomes, NF = 98. Marker chromosomes include one pair of submetacentrics, one pair of large acrocentrics, and one pair of large subtelocentrics with very short second arms. Nucleolus organizer regions are located in telomeric regions of short arms of marker submetacentric chromosomes. Small heterochromatin blocks are observed in centromeric regions of most chromosomes. The Chukotka char karyotype is very similar to that of Taranetz charSalvelinus taranetzi from the Achchen Lake: these karyotypes differ only in stability of the chromosome number.  相似文献

Condensed and dispersed forms of the chromosomes of the dinoflagellate, Prorocentrum micans, deposited on grids by the microcentrifugation technique were studied by electron microscopy. In the normally condensed form, the chromosomes appear as banded rods surrounded by a peripheral cloud of partially dispersed fibers. Single fibers in these and in extensively dispersed preparations appear as smooth threads of uniform diameter (55–65 Å). The chromosome fibers are contrasted by positive-group-specific stains, indicating the presence of cationic moieties associated with the DNA. Occasionally Y-shaped chromosomes are seen; these may be replicating structures. These observations are in general agreement with studies of dinoflagellate chromosomes by other techniques, and provide support for the suggestion that these organisms possess a genome organization whose structure is typical of neither prokaryotes nor eukaryotes, and hence may be intermediate forms.  相似文献

The ends of eukaryotic chromosomes are capped with special nucleoprotein structures called telomeres. Telomere shortening due to telomerase inactivation may result in fusion of homologous or heterologous chromosomes, leading to their successive breakage during anaphase movement, followed by fusion of broken ends in the next cell cycle, i.e. the breakage-fusion-bridge (BFB) cycle. Using fluorescence in situ hybridization (FISH) with 25S rDNA and specific bacterial artificial chromosome (BAC) probes we demonstrate participation of chromosomes 2 and 4 of Arabidopsis thaliana AtTERT null plants in the formation of anaphase bridges. Both homologous and non-homologous chromosomes formed transient anaphase bridges whose breakage and unequal separation led to genome rearrangement, including non-reciprocal translocations and aneuploidy. The 45S rDNA regions located at the ends of chromosomes 2 and 4 were observed in chromosome bridges at a frequency approximately ten times higher than expected in the case of random fusion events. This outcome could result from a functional association of rDNA repeats at nucleoli. We also describe increased variation in the number of nucleoli in some interphase cells with supernumerary rDNA FISH signals. These data indicate that dysfunctional telomeres in Arabidopsis lead to massive genome instability, which is induced by multiple rounds of the BFB mechanism.  相似文献

The variation in DNA content of the micronucleus (germinal nucleus) of Stylonychia lemnae and its relation to the number of chromosomes was examined. Different populations possess similar amounts of micronuclear DNA but there are differences of ±30% between clones of the same population. However, the DNA content varies by about 100% in the micronuclei during the lifetime of a clone. The haploid micronucleus contains 35 or 36 chromosomes which persist in the developing macronucleus anlagen and grow to giant chromosomes. Besides this remaining subset, the micronucleus contains a variable number of germ line restricted chromosomes (mean about 140; range between 100 and 180). The somatic macronucleus eliminates these elements early in its development. The varying number of the germ line restricted chromosomes is responsible for the variation in the micronuclear DNA content.  相似文献
