A free-ranging eastern box turtle (Terrapene carolina carolina) was referred to the Wildlife Center of Virginia with a three-month history of marked swelling of the right hind limb initially diagnosed as chromomycosis by histopathology. Hematology revealed severe anemia (9%), leukocytosis (12.8 cells x 10(3)/microl), heterophilia (6.14 cells x 10(3)/microl), and monocytosis (0.51 cells x 10(3)/microl). Gross necropsy revealed a firm, encapsulated 3 x 1 cm subcutaneous mass filled with dark brown-black, friable necrotic material of the distal right hind limb. Microscopically, the mass was characterized by a granulomatous inflammatory process with numerous multinucleated histiocytic giant cells. Fungal elements were present within necrotic centers and associated with multinucleated cells. Special stains revealed numerous phaeoid hyphae and yeast; Exophiala jeanselmei was isolated by routine mycologic culture. Phaeohyphomycosis was diagnosed based on the histologic appearance of the fungal elements within the mass and culture results. There was no histopathological evidence of systemic infection. This is the first report of phaeohyphomycosis caused by fungi of the genus Exophiala in free-living reptiles.  相似文献

Aural abscesses are a common health problem in free-ranging eastern box turtles (Terrapene carolina carolina), and they have been associated with high body burdens of organochlorine (OC) compounds, which are known disruptors of vitamin A. The objective of this study was to determine if the presence of pathologic lesions in box turtles were correlated with increased and decreased levels of hepatic OC compounds and vitamin A, respectively. A graded scale for the pathologic changes observed in tissue samples collected from abscessed and nonabscessed box turtles over a 2-yr period (2003-04) was developed, and the levels of OC compounds and vitamin A in livers collected from the same turtles were determined through chemical analysis. Epidemiologic determinants of 46 cases of aural abscessation in free-living eastern box turtles (Terrapene carolina) admitted to the Wildlife Center of Virginia (Virginia, USA) from 1991 to 2000 were evaluated. County human population density, year and season of admission, weight, and sex did not affect the risk for box turtles to develop aural abscessation. Counties with cases of aural abscessation were not randomly distributed, but rather were clustered into two multi-county regions. Geographic location was the only risk factor associated with aural abscessation in box turtles found in this study. Possible etiologies could include chronic infectious disease, malnutrition, or chronic exposure to environmental contamination with organochlorine compounds.  相似文献

Aural abscess or abscess of the middle ear is common in free-living Eastern box turtles (Terrapene carolina carolina) of Virginia (USA) and elsewhere. Although its etiology remains unknown, hypovitaminosis A has been suggested on the basis of similar lesions occurring in captive chelonians fed diets that are deficient in vitamin A. This hypothesis was supported by significantly greater body burdens of organochlorine compounds (reported disruptors of vitamin A metabolism) and a nonsignificant trend toward lower serum and hepatic vitamin A levels in free-living box turtles with this lesion. The tympanic epithelium was evaluated in 27 box turtles (10 with aural abscesses and 17 without). Lesions of the tympanic epithelium of box turtles with aural abscesses included hyperplasia, squamous metaplasia, hyperemia, cellular sloughing, granulomatous inflammation, and bacterial infection. These changes were more severe in turtles with aural abscesses than in those without and were more severe in tympanic cavities that had an abscess compared to those without when the lesion was unilateral. Organs from 21 box turtles (10 with aural abscesses and 11 without) from the study population were examined for microscopic lesions, and minimal histopathologic changes were found, none of which were similar to those found in the tympanic epithelium. Histopathologic changes in box turtles with aural abscesses were consistent with a syndrome that may involve hypovitaminosis A.  相似文献

We performed a retrospective review of medical records for 574 eastern box turtles (Terrapene carolina carolina) that presented to the University of Tennessee College of Veterinary Medicine (UTCVM) wildlife clinic between 1995 and 2007. These cases were reviewed to document and investigate the presenting complaints, as identified by a good Samaritan, final diagnoses, treatment modalities, days in hospital, and final disposition. Presenting complaints were consistent with the final diagnoses in 81% of cases. Trauma was both the most common presentation and final diagnosis. Associations between final diagnosis and either environmental temperature or precipitation were not significant (P>0.05). However, precipitation had a statistically significant positive association (P<0.05) with aural abscessation. Approximately 5% (28/574) of the box turtles were evaluated for packed cell volume total protein and white blood cell count. No differences were observed between sick and healthy animals for any of these variables. Multiple treatment modalities were used in these cases and were often used in combination, rather than as individual therapies. The number of days spent in the hospital ranged from 0 to 66 days, with a median of 2 days. The wildlife clinic of UTCVM should continue to evaluate the health status of eastern box turtles. Our findings indicate that the high caseload, as well as case variety of eastern box turtles in eastern Tennessee, suggests potential use as a biomonitor for this species.  相似文献

Eastern box turtle (Terrapene carolina carolina) populations are in decline, likely due to anthropogenic forces and disease, necessitating hematologic and biochemical data from healthy individuals for evaluation of wild populations. We repeatedly sampled 21 free-ranging eastern box turtles from May to September 2009 in the spring, summer, and fall to establish temporal hematologic and biochemical reference intervals. Packed cell volume, aspartate aminotransferase, and potassium levels declined significantly as the active season progressed. High levels of albumin, globulin, and calcium coincided with the presence of eggs in females. These reference intervals should provide baseline data for the clinical evaluation of wild box turtles presented for veterinary care or for studies of wild populations.  相似文献

Hematology is a common tool for wildlife health assessments. Manual leukocyte counts are required in reptiles, however, disagreement between quantification methods has been observed in some chelonians. This study determined agreement between two methods of leukocyte quantification, eosinophilic leukopet, and blood film white blood cell (WBC) estimates, in free-living eastern box turtles (Terrapene carolina carolina). Simultaneously, we assessed the impact of handling duration on both leukocyte quantity and corticosterone levels. We collected blood at capture (<2 min from disturbance) and again before release 30–150 min later from 92 box turtles at six sites in Illinois and Tennessee. Constant and proportional error was present in the leukopet results for WBC, lymphocytes, and basophils compared to the estimate method. Both methods were in agreement for heterophils, monocytes, and eosinophils. Agreement between the methods was significantly more likely at WBC counts below 23,241/µl. All hematologic parameters were significantly higher at the final blood draw compared to the initial blood draw using both WBC determination methods, except relative eosinophil and basophil counts. Corticosterone levels varied with time, with maximum concentrations reached at 54 min postcapture, followed by a rapid return to baseline levels. Corticosterone level was not significantly associated with any hematologic parameter or sex. This study provides a framework for understanding the effects of animal handling methodology and diagnostic modality when evaluating hematologic health in box turtles.  相似文献

Ranaviruses (genus Ranavirus) have been observed in disease epidemics and mass mortality events in free-ranging amphibian, turtle, and tortoise populations worldwide. Infection is highly fatal in turtles, and the potential impact on endangered populations could be devastating. Our objectives were to determine the prevalence of ranavirus DNA in blood and oral swabs, report associated clinical signs of infection, and determine spatial distribution of infected turtles. Blood and oral swabs were taken from 140 eastern box turtles (Terrapene carolina carolina) that were presented to the wildlife centers at the University of Tennessee (UT; n=39), Wildlife Center of Virginia (WCV; n=34), and North Carolina State University (NCSU; n=36), as well as a free-ranging nonrehabilitation population near Oak Ridge, Tennessee (OR; n=39) March-November 2007. Samples were evaluated for ranavirus infection using polymerase chain reaction (PCR) targeting a conserved portion of the major capsid protein. Two turtles, one from UT and one from NCSU, had evidence of ranavirus infection; sequences of PCR products were 100% homologous to Frog Virus 3. Prevalence of ranavirus DNA in blood was 3, 0, 3, and 0% for UT, WCV, NCSU, and OR, respectively. Prevalence in oral swab samples was 3, 0, and 0% for UT, WCV, and NCSU, respectively. Wildlife centers may be useful in detection of Ranavirus infection and may serve as a useful early monitoring point for regional disease outbreaks.  相似文献

A free-ranging adult female eastern box turtle (Terrapene carolina carolina) was presented to the University of Tennessee in October 2003 because of suspected trauma and blindness. Physical examination revealed lethargy, clear ocular and nasal discharges, and white oral and laryngeal plaques. Intracytoplasmic inclusions within heterophils and large mononuclear leukocytes were observed on routine blood smear examination. Postmortem findings included necrosis of epithelial and parenchymal cells with intracytoplasmic inclusions. Ultrastructurally, the leukocyte inclusions consisted of variably electron-dense granular material and viral particles consistent with the Iridoviridae family of viruses. The virus shared 100% sequence identity to a 420-base pair sequence of frog virus 3 (family Iridoviridae, genus Ranavirus) as determined by polymerase chain reaction and gene sequencing targeting a portion of the Ranavirus major capsid protein gene.  相似文献

Clinical signs of upper respiratory tract disease-like syndrome (URTD-LS) were observed in free-ranging eastern box turtles (Terrapene carolina carolina) from Virginia, USA (May 2001-August 2003), some of which also had aural abscesses. After a Mycoplasma sp. was detected by polymerase chain reaction (PCR), a study was undertaken to better define the range of clinical signs of disease and to distinguish mycoplasma-associated URTD-LS from other suspected causes of URTD-LS and aural abscessation in box turtles. Nasal and/or ocular swabs (from turtles possessing URTD-LS) or nasal washes (from asymptomatic turtles) were collected from turtles May 2001-August 2003; samples were assayed for Mycoplasma spp., chelonian herpesvirus, and iridoviruses by PCR testing. A partial DNA sequence (933 bases) of the small ribosomal subunit (16S rRNA) of the box turtle Mycoplasma sp. was analyzed to determine its phylogenetic relatedness to other Mycoplasma spp. of veterinary interest. Mycoplasma sp. was detected in seven (six with clinical signs of URTD-LS; one asymptomatic) of 23 fortuitously collected animals from six of 11 Virginia counties. Clinical signs in Mycoplasma sp.-infected animals included unilateral to bilateral serous to mucopurulent nasal discharge, epiphora, ocular edema, and conjunctival injection. Five Mycoplasma sp.-positive animals possessed aural abscesses; two did not. Analysis of the mycoplasma 16S rRNA gene sequence from one asymptomatic and three symptomatic animals representing four counties revealed a consensus Mycoplasma sp. sequence closely related to, but distinct from, M. agassizii. None of the samples collected contained viral DNA of chelonian herpesviruses or invertebrate and vertebrate (including FV3) iridoviruses. In conclusion, a new Mycoplasma sp. was associated with URTD-LS in native box turtles from Virginia that was not codetected with other suspected causes of chelonian upper respiratory disease; there was no proof of a direct relationship between aural abscessation and the Mycoplasma sp.  相似文献

We measured microclimate, field metabolic rates (FMRs), water flux, and activity patterns of telemetered box turtles (Terrapene carolina) in South Carolina from September 1987 to October 1988. Turtles were inactive for most of the winter and were active only sporadically during the rest of the year. Using the doubly labeled water method, we found that water flux averaged 8.8, 18.9, and 26.4 mL kg(-1) d(-1) in winter, spring, and summer/fall, respectively. FMR for the same periods averaged 0.028, 0.065, and 0.124 mL CO(2) g(-1) h(-1). Differences in FMR among seasons were significant but not between sexes. Using operative temperatures, we predicted standard and maximum metabolic rates of turtles. In winter, FMR was elevated above standard metabolic rates and close to maximum metabolic rates, whereas in spring and summer/fall, FMR fell midway between standard and maximum metabolic rates. We used a model to predict metabolic rates, geographical distribution, and potential reproductive output of box turtles across latitudes in eastern North America. Low FMR and low annual reproductive output may allow box turtles to survive and flourish in unpredictable resource environments by minimizing costs and risks, thereby maintaining greater lifetime reproductive success.  相似文献

We have performed the first study of neuronal activity in freely-moving reptiles. 23 brainstem units were recorded from areas throughout the reticular formation, during wakefulness and quiescence in the box turtle. These units responded to various sensory stimuli and increased firing rates in relation to motor activity during wakefulness. All but one unit showed significant decreases in discharge during quiescence. Group I cells (32%) fired mostly during active movements and exhibited silent periods of 5 min or longer during quiescence while group II cells (68%) maintained slow tonic activity in quiescence. Polygraphic data showed no consistent, cyclically occurring phasic events during quiescence.  相似文献

We isolated and characterized eight polymorphic microsatellite loci for a Texas population of three-toed box turtle, Terrapene carolina triunguis, using a refined hybridization capture procedure. All eight primer pairs amplified successfully at all loci in seven Texas ornate box turtles (T. ornata ornata). Due to the decline and conservation concerns of North American box turtles, these isolated microsatellites may be a most valuable tool for evaluating baseline population genetic structure for threatened box turtle populations.  相似文献

Animal personality can be defined as conspecific individuals consistently differing in behavioral tendencies. Personality is typically identified by behavioral repeatability, which occurs when within-individual variance is low relative to among-individual variance in the population. Intraspecific comparisons of behavioral repeatability in juveniles and adults within and across years are rare, but would be useful for testing hypotheses related to origins of animal personality and whether individuals exhibit stable or diverging behavior with ontogeny. To examine within- and across-year behavioral repeatability for eastern box turtles (Terrapene carolina), we assessed boldness (movement latency after brief confinement) of captive-born juveniles twice within three days when eight months old. We then repeated these tests for the same individuals one year later. Juveniles exhibited repeatable boldness within and across years. Although increasing body temperature was slightly associated with decreased movement latency, test year (1 or 2), or housing experience (being raised in an enriched or unenriched condition) had no effects on boldness. We also assessed across-year repeatability of boldness (head emergence from the shell after brief confinement) for wild adults at 1–3 year intervals. Adults also exhibited repeatable across-year boldness that was of similar magnitude to juveniles. We found no indication that sex class or whether adults had been radio-tracked influenced boldness. Our results suggest eastern box turtles demonstrate consistent individuality in boldness from an early age that is largely unaffected by temporal or environmental variation, and these behavioral differences can be maintained for multiple years in captivity and the wild, contrasting with theoretical expectations for personality development. These findings add to recent accumulating evidence demonstrating juvenile and adult box turtles exhibit multiple repeatable behaviors over the short- and long-term. We suggest this species is quickly gaining traction as a model organism for studying the proximate and ultimate causes of personality development within long-lived animals.  相似文献

Ectotherms can adjust their thermoregulatory set points in response to bacterial infection; the result may be similar to endothermic fever. We examined the influence of dose on the set point of body temperature (T(b)) in Terrapene carolina. After acclimating postprandial turtles to 20 degrees C, we injected them with two doses of bacterial endotoxin (LPS; lipopolysaccharide from Escherichia coli), 0.0025 or 0.025 mg LPS/g nonshell body mass, or with reptilian saline (control group). We placed the animals singly in linear thigmothermal gradients and recorded their T(b)'s for 48 h. The turtles showed dose-influenced thermal selection. Turtles injected with the high dose had T(b)'s significantly higher than control turtles, whereas low-dose turtles had T(b)'s significantly lower than control turtles. Also, there was a low daily effect on the T(b) of the turtles injected with the high dose. High-dose turtles had significantly higher T(b)'s than the control turtles during the first day but not during the second. Our results support the prediction of Romanovsky and Székely that an infectious agent may elicit opposite thermoregulatory responses depending on quality and quantity of the agent and the host health status.  相似文献

Genetic analyses of populations are essential to the conservation of threatened and cryptic taxa such as Chelonians. Turtles and tortoises are among the most imperiled vertebrate taxa worldwide, yet many of the natural history traits remain unknown leaving management decisions ill- or improperly informed. The eastern box turtle Terrapene c. carolina is no exception, with many gaps in our knowledge about traits such as juvenile dispersal and patterns of relatedness across the landscape, especially as it is a species not particularly tied to water and vulnerable to human disturbance. In addition, all long-term studies of this species have documented demographic population declines, even in protected habitats. In this study we explore finescale population structuring, gene flow, dispersal, and relatedness at four sites across the species range. These sites vary in habitat fragmentation and surrounding habitat quality. Many radiotelemtery and mark-recapture studies suggest that Terrapene spp. have a sedentary natural history, with small and temporally conserved home ranges and little propensity for dispersal. Based on these data we predicted that populations would be highly structured at fine geographic scales, closely related individuals (1st- and 2nd-degree relatives) would coexist in close proximity, and individuals exhibiting transient behavior would be true transients. All sites had low levels of population structuring, mean pairwise relatedness values were statistically zero, over 90 % of pairs of individuals were unrelated, 4.4–8.7 % were half-siblings, and fewer than 1.0 % were full siblings or parent-offspring pairs. These patterns were consistent across all four sites, regardless of habitat fragmentation. Furthermore, while some related pairs were found within a few meters of each other, others ranged up to 33 km apart. We found that one of two individuals with transient behavior was indeed a true genetic transient. These findings suggest that box turtles may be much more vagile than current management practices recognize. As most turtle species are strongly affected by anthropogenic disturbance, many may require much larger contiguous blocks of intact habitat for species persistence as the box turtle likely does. Management plans may therefore need to be updated to allow for safe and effective long-distance dispersal at the appropriate spatial scales in order to maintain genetic health of these species.  相似文献
