Caloric excess has been postulated to disrupt cardiac function via (i) the generation of toxic intermediates, (ii) via protein glycosylation and (iii) through the generation of reactive oxygen species. It is now increasingly being recognized that the nutrient intermediates themselves may modulate metabolic pathways through the post-translational modifications of metabolic enzymes. In light of the high energy demand of the heart, these nutrient mediated modulations in metabolic pathway functioning may play an important role in cardiac function and in the capacity of the heart to adapt to biomechanical stressors. In this review the role of protein acetylation and deacetylation in the control of metabolic programs is explored. Although not extensively investigated directly in the heart, the emerging data support that these nutrient mediated post-translational regulatory events (i) modulate cardiac metabolic pathways, (ii) integrate nutrient flux mediated post-translational effects with cardiac function and (iii) may be important in the development of cardiac pathology. This review examines the influence of a reduction in caloric intake on cancer and longevity. Data indicating that rodents subjected to caloric restriction display lower tumor incidence, reduced susceptibility to carcinogens and extended lifespan is analyzed. The potential role of the growth hormone/insulin-like growth factor type 1 (IGF-I) axis in this effect is discussed as is the evidence that a reduction in growth hormone and/or IGF-I leads to a reduction in spontaneous tumors and susceptibility to carcinogens.  相似文献

A meta-analysis was used to examine the independent and interactive effects of dietary restriction, endurance exercise training and gender on resting metabolic rate (RMR). Sixty different group means (covering 650 subjects) were identified from the scientific literature and subjected to meta-analysis techniques. Collectively (i.e., all groups combined), body weight loss was greater (P < 0.05) for men ( 18 kg) than for women ( 12 kg). There were no statistically significant exercise training or gender effects on RMR during weight loss. Collectively (i.e., all groups combined), dietary restriction resulted in a – 0.59 kJ min–1 ( – 12%) decrease in RMR (P < 0.05). When normalized to body weight, RMR was reduced by less than 2% (P < 0.05). These data suggest that exercise training does not differentially affect RMR during diet-induced weight loss. In addition, decreases in resting metabolism appear to be proportional to the loss of the metabolically active tissue.  相似文献

The study analysed the responses of three metabolic parameters in five distinct adipose tissue depots to caloric restriction (4 weeks) in the rat. The aims were to evaluate whether specific adipose tissue depots were recruited for triacylglycerol (TAG) storage and/or mobilisation, and to determine to what extent specific adipose tissue depots exhibited preferences for the source of fatty acid (FA) for TAG storage. Caloric restriction led to a general enhancement of the response of lipoprotein lipase (LPL), FA synthesis and glucose utilisation to a meal. Effects were particularly marked in the parametrial, perirenal and interscapular depots compared with mesenteric and subcutaneous depots. There was no evidence that individual depots selectively expressed a preference for the pathways concerned with the generation of FA for storage (the exogenous (LPL) and the endogenous (synthesis) pathway). However, the temporal sequence of activation of these pathways differed in a manner consistent with a switch from preponderant use of FA produced via de novo synthesis during the very early phase of feeding towards later use of FA derived from circulating TAG. The overall excursions in insulin levels observed in the calorie-restricted rats were comparable to those found in free-feeding rats, but the magnitude and the rapidity of the individual metabolic responses of the adipocyte were augmented. The data are consistent with a general enhancement of insulin sensitivity and responsiveness in adipose tissue of calorie-restricted rats, together with adaptive regional specialisation of adipocyte function. These adaptations would be predicted to facilitate the immediate conservation of dietary nutrients by promoting their storage as the FA or glycerol moieties of adipose tissue TAG and thereby to ensure the regulated release of FA and glycerol from adipose tissue in accordance with the requirement for glucose conservation and/or production.  相似文献

We investigated the effect of monoamines and their agonists and antagonists upon responses of motoneurons medicated by NMDA-glutamate receptors. It was found that adrenalin strengthens the responses studied through activation of -and -adrenoreceptors. This effect of adrenalin is suppressed by an -adrenoblocker (phentolamine) and a -adrenoblocker (propranolol); it is restored by an -adrenomimetic (phenylephrine hydrochloride) and a -adrenomimetic (neoepinephrine). In 66% of the cases, dopamine inhibited motoneurons responses; in 34% of the cases, it strengthened such responses. In the presence of a selective blocker of D2-dopamine receptors (sulpiride), it only strengthened motoneuron responses. Serotonin strengthens responses of motoneurons; its action is suppressed by a blocker of type-II serotonin receptors (dezeril). A selective agonist of subtype-1A serotonin receptors, campirone, suppresses motoneuron responses. The effect of campirone is weakened by propranolol. The strengthening of NMDA-responses of motoneurons evoked by neoepinephrine and dopamine in the presence of sulpiride is weakened by a blocker of cAMP-dependent protein kinase, tolbutamide, and the strengthening of responses by phenylephrine hydrochloride and serotonin is weakened by an inhibitor of calmodulin, trifluoroperazine.M. Calorie restriction (CR) increases average and maximum lifespan and exhibits an apparent beneficial impact on age‐related diseases. Several studies have shown that CR initiated either in middle or old age could improve ischemic tolerance and rejuvenate the aging heart; however, the data are not uniform when initiated in young. The accurate time to initiate CR providing maximum benefits for cardiac remodeling and function during aging remains unclear. Thus, whether a similar degree of CR initiated in mice of different ages could exert a similar effect on myocardial protection was investigated in this study. C57BL/6 mice were subjected to a calorically restricted diet (40% less than the ad libitum diet) for 3 months initiated in 3, 12, and 19 months. It was found that CR significantly reversed the aging phenotypes of middle‐aged and old mice including cardiac remodeling (cardiomyocyte hypertrophy and cardiac fibrosis), inflammation, mitochondrial damage, telomere shortening, as well as senescence‐associated markers but accelerated in young mice. Furthermore, whole‐genome microarray demonstrated that the AMP‐activated protein kinase (AMPK)–Forkhead box subgroup ‘O’ (FOXO) pathway might be a major contributor to contrasting regulation by CR initiated in different ages; thus, increased autophagy was seen in middle‐aged and old mice but decreased in young mice. Together, the findings demonstrated promising myocardial protection by 40% CR should be initiated in middle or old age that may have vital implications for the practical nutritional regimen.  相似文献

A group of Bokoloji bulls (n=36), consisting of 18 young bulls (12 to 18 mo old) and 18 old bulls (24 to 30 mo old) were used to study the effects of feed restriction and realimentation on reproductive function. The bulls were placed either on a low (L). medium (M) or high (H) plane of nutrition during the 3 treatment periods. At the low and medium feeding levels, the animals received approximately 25 and 45%, respectively, of the intake of the bulls on the high plane diet. All the bulls were fed a low (L) plane of nutrition for 90 d in the first treatment period. The 3 treatments were designated as LL, LM and LH for the mid-period, and as LLH, LMH and LHH for the final period, according to the prescribed level of feeding. Body measurements and scrotal circumference were taken for all bulls before slaughter. A total of 18 bulls was slaughtered at the end of the mid-period, while the remaining 18 bulls were slaughtered at the end of the final period. Testicular weights as well as gonadal and epididymal sperm reserves were determined. At the end of the mid period, the bulls on a high plane of nutrition had significantly (P<0.05) higher live weights and chest girths than bulls on the medium and low planes of nutrition. Gonadal sperm/spermatid reserves of 5.2 x 10(9) and epididymal sperm reserves of 2.4 x 10(9) in bulls on the high plane of nutrition were significantly (P<0.05) higher than the sperm reserves for the bulls on either the low or the medium plane of nutrition. At the end of the final period, ho significant differences existed among the LHH, LMH and LLH treatments in liveweight, chest girth, scrotal circumference and sperm reserves (P>0.05). It is concluded that bulls which have undergone feed restriction for 90 d are capable of regaining their body growth and reproductive function if such bulls are placed on an adequate plane of nutrition.  相似文献

Background  

Most studies of molecular evolution are focused on individual genes and proteins. However, understanding the design principles and evolutionary properties of molecular networks requires a system-wide perspective. In the present work we connect molecular evolution on the gene level with system properties of a cellular metabolic network. In contrast to protein interaction networks, where several previous studies investigated the molecular evolution of proteins, metabolic networks have a relatively well-defined global function. The ability to consider fluxes in a metabolic network allows us to relate the functional role of each enzyme in a network to its rate of evolution.  相似文献

In response to reduced oxygen or nutrient supply, the fetus may redistribute cardiac output to conserve brain and heart growth, at the expense of the peripheral tissues; however, it is not known whether alterations in vascular function are maintained after birth or whether reduced fetal oxygen versus nutrient supply produces distinct effects. Using a pressure myograph, we examined isolated carotid and femoral artery responses to phenylephrine and endothelin-1 in neonatal rats, after either reduced maternal oxygen or global nutrient restriction during late gestation. Timed-pregnant Sprague-Dawley rats were randomly assigned to control (n = 10), hypoxia (12% O2, n = 9), or nutrient restriction (NR, 40% of control diet, n = 7) protocol and treated from day 15-21 of pregnancy. Pups were collected 3-12 h after birth. Neonatal weights (P < 0.001) and relative liver weights (P < 0.001) were lower in hypoxia and nutrient restriction treatments compared with control, while relative heart weights were greater in the hypoxia than in the control or nutrient restriction groups (P < 0.01). Constriction to phenylephrine was reduced in carotid arteries from the hypoxia and nutrient restriction groups compared with control (P < 0.001), while the femoral artery response was greater in hypoxia-treated neonates compared with control or nutrient-restricted neonates (P < 0.01). Only the hypoxia reduced carotid responses to endothelin-1, while no differences were observed in the endothelin-1 responses in femoral arteries. Maternal hypoxia and maternal nutrient restriction produced distinct effects on heart growth and neonatal vascular function, suggesting that regional changes in cardiovascular function after poor fetal growth are dependent on the nature of the insult in utero.  相似文献

The plasmid DNA scission by the restriction enzyme HaeIII was investigated in the presence of tetrakis(1-methylpyridinium-4-yl)porphyrin (H2TMPyP) and its manganese(III), iron(III), nickel(II), cobalt(III) and zinc(II) derivatives. The effect of metalloporphyrins on plasmid DNA cleavage was ascertained by gel electrophoresis. UV-Vis absorption spectroscopy and circular dichroism (CD) spectroscopy. In the absence of the metalloporphyrins, plasmid DNA scission did not occur in the presence of a low concentration of HaeIII (0.2 units microL(-1)) at 37 degrees C after 1 h incubation. However, DNA cleavage occurred in the presence of the metalloporphyrins and HaeIII (0.2 units microL(-1)) at 37 degrees C after 1 h incubation. Gel electrophoresis results indicate the catalytic effect of metalloporphyrins (Mn(III)-, Fe(III)-, Co(III)- and Zn(II)TMPyP) by binding to both DNA and the enzyme through electrostatic interaction, which was confirmed by the change in UV-Vis and CD spectra. A mechanism for the enhanced DNA cleavage is proposed.  相似文献

[目的]摸索葡萄溃疡病菌(Lasiodiplodia theobromae)限制性内切酶介导整合(Restriction enzyme mediated integration,REMI)的转化方法,并构建CSS-01s (L.theobromae)的REMI转化子库.[方法]利用REMI转化方法,将线性化的含有潮霉素抗性基因(Hygromycin phosphotransferase gene,Hyg)的pUCATPH质粒转化CSS-01s菌株的原生质体;测定其对潮霉素B的敏感浓度,摸索不同酶解液和酶解时间对原生质体制备的影响,统计不同限制性内切酶酶量对转化效率的影响,以摸索出的最优条件构建CSS-01s菌株的REMI转化子库,采用Southern blot的方法验证转化子.[结果]首次建立了一套L.theobromae的REMI转化方法,经转化得到包含6 000余个转化子的L.theobromaeCSS-01s转化子库,随机挑选的5个转化子经Southern blot分析,质粒均插入到了Hind Ⅲ相应的酶切位点处.[结论]浓度为2%崩溃酶+2%蜗牛酶酶混合液、酶解4h为原生质体获得的最优条件,每管转化体系中加入30 UHind Ⅲ时转化效率最高,该方法可以用来获得大量不同表型的REMI转化子.  相似文献

1. Plasma GH was greater (P less than 0.05) on day 12 in ad libitum-fed birds compared to restricted chicks. Conversely, maximum GH levels were found to occur in the nutrient restricted chicks during the period of accelerated growth (day 42). 2. A significant decline in circulating insulin concentrations with advancing age was evident in both ad libitum-fed and restricted chicks. 3. Feed restriction significantly suppressed circulating T3 in restricted chicks, with concentrations returning to control levels upon refeeding. 4. A significant increase in T4 with advancing age was evident in both treatment groups, with T4 being significantly greater in controls compared to restricted chicks at 54 days of age.  相似文献

Dietary modification ought to be the first line of strategy in prevention of the development of cardiac disease. The purpose of this study was to investigate whether dietary restriction, dietary-fibre-enriched diet, and their interactions might affect antioxidant capacity and oxidative stress in cardiac tissue. Male Wistar rats (180-200 g; n=10) were divided into four groups: control ad libitum diet (C), 50% restricted diet (DR), fed with fibre-enriched diet (F), and 50% restricted fibre-enriched diet (DR-F). After 35 days of the treatments, F, DR, and DR-F rats showed low cholesterol, LDL-cholesterol, and triacylglycerol, and high HDL-cholesterol in serum. The DR, DR-F, and F groups had decreased myocardial lipoperoxide and lipid hydroperoxide. The DR-F and F treatments increased superoxide dismutase and glutatione peroxidase (GSH-Px). The DR treatment increased GSH-Px and catalase activities. Dietary fibre beneficial effects were related to metabolic alterations. The F and DR-F groups showed high cardiac glycogen and low lactate dehydrogenase/citrate synthase ratios, indicating diminished anaerobic and elevated aerobic myocardial metabolism in these animals. There was no synergistic effect between dietary restriction and dietary fibre addition, since no differences were observed in markers of oxidative stress in the F and DR-F groups. Dietary fibre supplementation, rather than energy intake and dietary restriction, appears to be the main process retarding oxidative stress in cardiac tissue.  相似文献

The influence of feeding system on growth performance, carcass characteristics, and fatty acids profile of subcutaneous backfat and lipogenic enzymes activity was studied. The following feeding systems were applied: HH: group of 12 pigs fed with a high feeding level during growth and fattening periods; HL: group of 12 pigs fed with a high feeding level during growth period and a low feeding level during fattening period; LH: group of 12 pigs fed with a low feeding level during growth period and a high feeding level during fattening period; LL: group of 12 pigs fed with a low feeding system during growth and fattening periods. The high and low feeding levels applied during growth and fattening periods were 70 and 35 g feed/kg live weight0.75 (LW0.75) day and 121 and 87 g feed/kg LW0.75 day, respectively. The duration of the growth period was 163 days in all groups. The ages at slaughter of HH, HL, LH and LL pigs were approximately 383, 406, 439 and 503 days, respectively. The HH and LL pigs had the higher and lower average daily gain respectively, while the LH pigs presented the best feed conversion ratio during fattening period. At slaughter, the LL pigs had the higher value of loin width and the lower values of backfat thicknesses and Longissimus dorsi muscle intramuscular fat percentage (P<0.05). The subcutaneous backfat outer and inner layers from LH and LL pigs had a significantly (P<0.05) higher proportion of C18:1 n-9 and ΣMUFA (monounsaturated) acids than those from HH and HL pigs. The subcutaneous backfat inner layer from LL pigs had a lower C16:0 and ΣSFA (saturated) acids proportions than those from remaining groups of pigs. The activity of the fatty acid synthase (FAS) and glucose-6-phosphate dehydrogenase (G6PD) enzymes, measured in subcutaneous backfat, at the end of growing period was significantly (P<0.05) higher in pigs fed high feeding level during growing phase. An increase of feeding level during the fattening phase led to an increase in the activity of G6PD enzyme. In conclusion, a marked effect of feeding system was observed on pig productivity and fatty acid composition.  相似文献

以刀豆蛋白A(ConA)刺激的脾淋巴细胞增殖活性及自然杀伤细胞(NKcel)活性为细胞免疫功能检测指标,观察了阿片受体阻断剂纳洛酮(Naloxone,NLX)对大鼠海马内微量注射甲硫脑啡肽所致的免疫功能增强作用的影响。结果发现:(1)海马内微量注射白细胞介素1(IL1)诱导剂(细菌内毒素)脂多糖(lipopolysacharide,LPS,50ng/1μl)可降低机体免疫功能。(2)双侧海马内预先注射甲硫脑啡肽(MENK,浓度:10μg/μl)各1μl,可阻止脑内LPS降低免疫功能的作用。(3)脑啡肽的这种作用可被阿片受体阻断剂纳洛酮(10μg/1μl)阻断。(4)海马内单纯注射纳洛酮对机体免疫功能也起抑制作用。上述结果提示,海马内脑啡肽对免疫功能的增强作用是通过阿片受体介导的。  相似文献

Calcium release from the sarcoplasmic reticulum (SR) in cardiac muscle occurs through a specialised release channel, the ryanodine receptor, RyR, via the process of Ca-induced Ca release (CICR). The open probability of the RyR is increased by elevation of cytoplasmic Ca concentration ([Ca(2+)](i)). However, in addition to Ca, other modulators affect the RyR open probability. Agents which increase the RyR opening during systole produce a transient increase of systolic [Ca(2+)](i) followed by a return to the initial level due to a compensating decrease of SR Ca content. Increasing RyR opening during diastole decreases SR Ca content and thereby decreases systolic [Ca(2+)](i). We therefore conclude that potentiation of RyR opening will, if anything, decrease systolic [Ca(2+)](i). The effects of specific examples of modulators of the RyR, such as phosphorylation, metabolic changes, heart failure and polyunsaturated fatty acids, are discussed.  相似文献
