A phosphatidyl choline (PC) exchange between apoHDL/PC micellar complexes in solution and the same complexes immobilized on Sepharose was studied. The PC exchange in buffer was represented in terms of pseudo first order reversible process. The first order constants for the unidirectional efflux of PC from apoHDL/PC-Sepharose (k1) and for the unidirectional efflux of PC from apoHDL/PC complexes (k2) were equal to (0.45 +/- 0.2) x 10(-3) and (1.35 +/- 0.2) x 10(-3) min-1, respectively. The k1 values showed the Arrhenius dependence on the temperature within range 278-323 K. Plasma serum proteins facilitated the PC efflux from apoHDL/PC-Sepharose being additional acceptors of PC. These data allow use of apoHDL/PC-Sepharose for correcting lipid plasma composition in vitro.  相似文献

Summary The lipid content and composition from an axolemma-rich preparation isolated from squid retinal axons was analyzed.The lipids, which accounted for 45.5% of the dry weight of this membrane, were composed of 22% cholesterol, 66.7% phospholipids and 5.2% free fatty acids. The negatively charged species phosphatidyl ethanolamine (37%), phosphatidyl serine (10%) and lysophosphatidyl ethanolamine (4%) made up 51% of the phospholipids. The amphoteric phosphatidyl choline and sphingomyelin accounted for 39% and 4%, respectively.The relative distribution of fatty acids in each of the isolated phospholipids was studied. The most remarkable feature of these phospholipids was the large proportion of long-chain polyunsaturated fatty acids. The 226 acyl chain accounted for 37% in phosphatidyl ethanolamine, 21.7% in phosphatidyl choline, 17.5% on phosphatidyl serine and 20.3% in sphingomyelin (all expressed as area %).The molar fraction of unsaturated fatty acids reached 65% in phosphatidyl ethanolamine and 42.0 and 44.8% in phosphatidyl choline and phosphatidyl serine, respectively. The double bond index in these species varied between 1.0 and 2.6.The lipid composition of the axolemma-rich preparation isolated from squid retinal axons appears to be similar to other excitable plasma membranes in two important features: (a) a low cholesterol/phospholipid molar ratio of 0.61; and (b) the polyunsaturated nature of the fatty acid of their phospholipids.This particular chemical composition may contribute a great deal to the molecular unstability of excitable membranes.The preceding papers of this series were published inArchives of Biochemistry and Biophysics.  相似文献

The lipid composition of the leaves of hardy Vernal and cold-sensitive Caliverde alfalfa plants, grown at different temperatures, was determined. Phosphatidyl glycerol, phosphatidyl inositol, and the sulfolipid content were directly related to growth temperature. Mono- and digalactose diglyceride and phosphatidyl choline and ethanolamine were inversely related to temperature. At corresponding growth temperatures Vernal plants showed higher percentages of mono- and digalactose diglyceride and phosphatidyl choline and ethanolamine than Caliverde plants, while the opposite was true for phosphatidyl glycerol and inositol and sulfolipid. Differences in fatty acid composition of corresponding leaf lipid fractions of plants grown at different temperatures or differences in fatty acid composition between lipid fractions of plants of different varieties in general were negligible.  相似文献

Exposure of adult rats to hypobaric hypoxia caused hypolipidemia, hypotriglyceridemia and hypophospholipidemia. Hypobaric hypoxia produced an increase in liver triglyceride and cholesterol levels and a decrease in lung triglyceride, total phospholipid and phosphatidyl choline. The proportion of phosphatidyl choline in the pulmonary surfactant fraction I phospholipids (responsible for reducing surface tension) decreased (55.2% as compared to 80.4% in control animals). Incorporation of 32-P into liver phosphatidyl ethanolamine was significantly increased, incorporation into lung phosphatidyl choline and phosphatidyl ethanolamine was increased whereas a decreased incorporation into plasma phosphatidyl choline was observed. The data suggest an enhanced lipid synthesis in liver with a probable impairment of mobilization into plasma.  相似文献

Synopsis Application of Baker's acid Haematein test for the histochemical detection of phospholipids to rat hypothalamus reveals striking colour differences between white and grey matter. It was found previously that these differences could not be attributed to corresponding differences in the acid Haematein-positive lipid content in various regions of the hypothalamus. As the staining reaction might depend on the degree of unsaturation of lipids, the fatty acid compositions of phosphatidyl choline and the polar lipid fraction from the optic chiasma and some regions of rat hypothalamus were investigated.The polar lipid fraction of the optic chiasma contained relatively more mono-unsaturated and less saturated fatty acids than the polar lipid fractions from the area anterior, and the suprnoptic and paraventricular nuclei of the hypothalamus. The same trend was observed for the fatty acid composition of phosphatidyl choline isolated from these regions. Both the polar lipid fraction and phosphatidyl choline showed a lower content of poly-unsaturated fatty acids in the optic chiasma than in the area anterior. It is argued that the colour differences between myclinated and unmyelinated regions of the hypothalamus demonstrable after application of Baker's acid Haematein test cannot be ascribed merely to corresponding differences in the amounts of unsaturated lipids in these regions. According to the author the particular structure of myelin might be held responsible for the stronger staining of white matter compared with grey matter.  相似文献

Significant differences in lipid composition have been found between normal human lymphocytes and polymorphonuclear leukocytes (isolated from blood by means of glass-bead columns), abnormal leukocytes from patients with acute and chronic leukemia, and leukocytes from peritoneal exudates. Lipid extracts of isolated leukocytes were analyzed for total lipid, phosphorus, cholesterol, and plasmalogens. Individual phospholipids and neutral lipids were separated by thin-layer chromatography. The major phospholipids were phosphatidyl choline, ethanolamine glycerophosphatides, sphingomyelin, phosphatidyl serine, and phosphatidyl inositol. Plasmalogen was found mainly as phosphatidal ethanolamine. The neutral lipid fractions contained free cholesterol and various amounts of triglyceride, but little esterified cholesterol. Normal lymphocytes contained about half as much total lipid per cell as normal polymorphonuclear leukocytes, with a similar cholesterol:-lipid-P ratio but relatively more lecithin and less ethanolamine glycerophosphatide. Normal mature leukocytes, compared with immature cells of the same morphological series, had a higher total lipid content per cell, more cholesterol, and a higher ratio of cholesterol to lipid-P. Little difference was found in total lipid-P per cell, but mature cells contained relatively less lecithin and more sphingomyelin. These findings may reflect differences in the relative content of various intracellular organelles as well as possible differences in the quantity and composition of the plasma membrane.  相似文献

Summary A study was made of the concentrations and fatty acid compositions of the major phosphoglycerides in the yolk of the alligator egg on the 8th and 75th days of incubation. The major phosphoglycerides were phosphatidyl choline and phosphatidyl ethanolamine which at the start of incubation accounted for 72 and 18%, respectively, of total phosphoglyceride. Phosphoglycerides were characterised by low levels of linoleic acid but extremely high levels of C20 and C22 polyunsaturates. The extensive absorption of phosphoglyceride over the incubation period was accompanied by a reduction in percentage of phosphoglyceride within the residual yolk lipid, a reduction in the proportion of phosphatidyl choline within the total phosphoglyceride, and increases in the proportions of phosphatidyl ethanolamine and lyso-phosphatidyl choline. There were small but wide-spread changes in the fatty acid composition of the phosphatidyl choline during incubation. Within the phosphatidyl ethanolamine and phosphatidyl serine fractions there were very large reductions in the C20 and C22 polyunsaturated fatty acid levels. The phosphoglyceride changes are discussed with respect to the unique role of yolk lipid absorption in the nutrition of the developing embryo.  相似文献

The lipid composition of leaves has been investigated in different genotypes of cucumber ( Cucumis sativus L.), which differ in temperature requirement for cultivation. In addition the effects of hardening by low but non-chilling temperature, soil heating and grafting (on the chilling-resistant C. ficifolia L.) on lipid composition have been studied. Content and composition of phospholipids and sterols were determined as well as phospholipid/sterol ratio, and fatty acid composition of total lipids and the different phospholipids.
The effects of genetic differentiation and of the various culture treatments on lipid composition of the leaves were very different. Genetic differentiation was evident as higher levels of Iinolenic acid in several phospholipids in the more cold-tolerant cultivars. Hardening the plants by low temperature resulted in a higher phospholipid level (especially phosphatidyl choline), more unsaturated phospholipid, and lowering of the sterol/phiospholipid ratio, all properties which may contribute to a higher membrane fluidity and lower growth temperature limit. Soil healing reduced the phospholipid level of the leaves slightly, and a higher content of 3- trans -hexadece-noic acid in phosphatidyl glycerol was observed. Grafting cucumber on the cold-resistant rootstock of C. ficifolia also raised the level of trans -hexadecenoic acid in phosphatidyl glycerol. The role of this fatty acid in the functioning of the chloroplast is discussed.  相似文献

Investigations have been carried out on the influence of the phospholipid composition of rat liver plasma membranes and of their physico-chemical properties on the activity of membrane-bound neutral sphingomyelinase. The membrane phospholipid composition was modified by the incorporation of different phospholipids into the membrane bilayer by means of lipid transfer proteins, n-butanol delipidation or exogenous sphingomyelinase (Staphylococcus aureus) treatment. The results indicate that the activity of neutral sphingomyelinase in liver plasma membranes depends upon phosphatidyl choline presence in the membrane bilayer and not upon membrane fluidity.  相似文献

Abstract— —The lipid composition of human, bovine and sheep pineal glands was determined. No characteristic species difference was found in lipid content and composition. The total lipid was 2·9–4·0 per cent of wet weight of which phospholipid comprised 58–71 per cent and cholesterol 13·9–15·8 per cent in the three species. The phospholipid composition was 45 per cent phosphatidyl choline, 22 per cent phosphatidyl ethanolamine, 14 per cent sphingomyelin, 9 per cent phosphatidyl serine, 8 per cent monophosphoinositide, and 2 per cent diphosphatidyl glycerol. The major fatty acids found were C14:0, C16:0, C18:0, C16:1, and C18:1. In contrast to other tissues, pineal sphingomyelin has a low C24:1 content. No significant amounts of polyphosphoinositides or gangliosides were detected. When its lipid composition is compared with that of a number of other tissues, pineal is found to be most similar to testes and unlike pituitary and brain.  相似文献

We have studied the effects of semisynthetic diets containing 5% by weight (12% of the energy) of either olive oil (70% oleic acid, OA) or corn oil (58% linoleic acid), or fish oil (Max EPA, containing about 30% eicosapentaenoic, EPA C 20:5 n-3, plus docosahexaenoic, DHA C 22:6 n-3, acids, and less than 2% linoleic acid), fed to male rabbits for a period of five weeks, on plasma and platelet fatty acids and platelet thromboxane formation. Aim of the study was to quantitate the absolute changes of n-6 and n-3 fatty acid levels in plasma and platelet lipid pools after dietary manipulations and to correlate the effects on eicosanoid-precursor fatty acids with those on platelet thromboxane formation. The major differences were found when comparing the group fed fish oil and depleted linoleic acid vs the other groups. The accumulation of n-3 fatty acids in various lipid classes was associated with modifications in the distribution of linoleic acid and arachidonic acid in different lipid pools. In platelets maximal incorporation of n-3 fatty acids occurred in phosphatidyl ethanolamine, which also participated in most of the total arachidonic acid reduction occurring in platelets, and linoleic acid, more than archidonic acid, was replaced by n-3 fatty acids in various phospholipids. The archidonic acid content of phosphatidyl choline was unaffected and that of phosphatidyl inositol only marginally reduced. Thromboxane formation by thrombin stimulated platelets did not differ among the three groups, and this may be related to the minimal changes of arachidonic acid in phosphatidyl choline and phosphatidyl inositol.  相似文献

In order to study the lipid dependence of glucose-6-phosphatase the lipid composition of microsomes from rat liver and hepatoma was modified using lipid exchange proteins. It was shown that the enzyme activity depends on the presence of phosphatidyl ethanolamine and phosphatidyl serine, but is unaffected by the enrichment of the microsomes with phosphatidyl choline. On the basis of the data obtained it was assumed that the aminophospholipids are required for the functioning of the protein carrier; however, they do not affect the activity of the catalytic component of the glucose-6-phosphatase system on the inner surface of the microsomal membrane.  相似文献

The lipid compositions of the gradient-purified gastric microsomal membranes from the fundic mucosa of pig, rabbit, and frog were determined. The total lipid content varied widely. Compared to the rabbit (21.6 ± 0.6 mg/100 mg protein), the pig had about twice as much and the frog about three times as much lipid. The levels of cholesterol were higher in both mammalian species (about 32% of the lipid) compared to frog (23%). Phospholipids accounted for about 45, 54, and 52% of the total microsomal lipids from pig, rabbit, and frog and the molar ratios of cholesterol to phospholipid in the three species were 1.95, 1.6, and 1.17, respectively. Phosphatidyl choline and phosphatidyl ethanolamine together constituted about 75% of the total phospholipids in pig and frog and 93% in rabbit gastric microsomes. Sphingomyelin comprised 19.3, 3.2, and 1.5% in pig, rabbit, and frog, respectively. Phosphatidyl inositol constituted 5, 2.7, and 23.6% in pig, rabbit, and frog, respectively. The ratios of phosphatidyl ethanolamine to phosphatidyl choline were 1.17, 1.1, and 0.85 in pig, rabbit, and frog, respectively. The saturated fatty acids 16:0 and 18:0 and the unsaturated fatty acid 18:1 and 18:2 were the predominant fatty acids in all phospholipids. The ratios of saturated to unsaturated fatty acids were between 0.8 and 0.9 in phosphatidyl choline and 0.27 and 0.5 in phosphatidyl ethanolamine from all three species. The contributions by saturated fatty acids were much more in phosphatidyl inositol and sphingomyelin than in phosphatidyl choline and phosphatidyl ethanolamine from all species. Position 1 of phosphatidyl choline had 63% saturated and 37% unsaturated fatty acids; while the reverse was true for position 2. Phosphatidyl ethanolamine, however, had 85% saturated fatty acids in position 1 compared to only 25% in position 2. Arachidonic acid (20:4) was present in significant amounts in all species located exclusively at position 2 of both phosphatidyl choline and phosphatidyl ethanolamine.  相似文献

Small unilamellar liposomes with an average diameter of 80 nm were prepared from phosphatidyl choline of various sources using the dialysis method with cholate as a detergent. When 14C-labeled soybean liposomes were intravenously injected into male NMRI mice, up to 10% of the total label was found in the liver lipid. The uptake was dose-dependent and reached an apparent saturation 4 h after injection. The liver maintained a constant radioactivity corresponding to 1.9 +/- 0.13 mg phospholipid/g liver until ten hours after injection of 850 mg labeled phosphatidyl choline/kg body wt. Little radioactivity was taken up by the spleen. Analogous doses of liposomes prepared from egg yolk phosphatidyl choline led to a radioactivity corresponding to 1.3 +/- 0.4 mg lipid/g liver 4 h after injection. Liposomes with a similar size were prepared from hydrated, i.e., saturated phosphatidyl choline. After intravenous administration of these liposomes, an amount of 5.3 +/- 0.5 mg labeled lipid was found per g liver after 4 h. In contrast to unsaturated liposomes, 5.8 +/- 0.8 mg lipid per gram spleen was trapped by the spleen. The pharmacodynamic effect of these different liposomes was studied in benzo[a]pyrene-pretreated mice intoxicated with 400 mg/kg paracetamol. Animals which received paracetamol exhibited serum alanine aminotransferase activities of 4220 +/- 1140 units/l after 4 h and exhaled 120 +/- 19 nmol ethane kg-1 h-1. When pretreated with 850 mg soybean phosphatidyl choline/kg body wt. (i.v.) 2 h prior to paracetamol, the increase in serum transaminase activity was reduced to 117 +/- 104 units/l and ethane exhalation amounted to 18 +/- 8 nmol kg-1 h-1. In contrast, similar pretreatment with egg yolk phosphatidyl choline or hydrated phosphatidyl choline failed to protect against paracetamol-induced hepatotoxicity. The different pharmacodynamic effects of the two phosphatidyl cholines of plant or animal origin cannot be explained on the basis of their different pharmacokinetics. In the case of soybean phosphatidyl choline liposomes, the amount of radioactive lipid found in the liver correlated with the hepatoprotective potency.  相似文献

Hydatid fluid collected from the lungs and livers of sheep and humans was analysed for protein and lipid composition. There were no marked differences in the composition of these parameters and the major lipids were triglycerides and diglycerides. The phospholipids, which formed the minor fraction, were mainly phosphatidyl choline and phosphatidyl inositol. Cholesterol present was in the free form.  相似文献

As one of the methods of finding out the structural change of lipid bilayers due to change of environmental solution, the capacitances of phosphatidyl choline (egg lecithin) and phosphatidyl serine (bovine brain) bilayer membranes in solutions of various pH and salt contents were measured. It was found that the capacitance of the bilayer depended upon pH and salt content. The capacitance had a minimum value around pH 4 for phosphatidyl choline and around pH 3-4 for phosphatidyl serine bilayers, respectively. The value of the capacitance increased as the pH of the solution became lower or higher. As the concentration of cholesterol in the phosphatidyl choline bilayer increased, the capacitance increased and reached a saturation value. A DC voltage across the phosphatidyl choline bilayer did not affect the value of the capacitance practically.  相似文献

The fatty acid composition of wheat seedling roots changed in response to temperature. As temperature declined, the level of linolenic acid increased and the level of linoleic acid decreased. The distribution of phospholipid classes was not influenced by temperature. Phosphatidyl choline and phosphatidyl ethanolamine were the predominant phospholipids isolated and comprised 85% of the total lipid phosphorus. Smaller quantities of phosphatidyl glycerol, phosphatidyl inositol, phosphatidic acid, and phosphatidyl serine were isolated. The fatty acid composition of phosphatidyl choline and phosphatidyl ethanolamine were the same and temperature affected the fatty acid composition of both phospholipids in the same manner.Growth in the presence of the substituted pyridazinone, BASF 13 338 (4-chloro-5-dimethylamino-2-phenyl-3(2H)pyridazinone), reduced the level of linolenic acid and increased the level of linoleic acid in the phosphatidyl choline, phosphatidyl ethanolamine, and total polar lipid fractions. BASF 13 338 did not affect the levels of palmitate, stearate, and oleate or the distribution of phospholipid classes.Respiration rates of wheat root tips were measured over a range of temperatures. The respiration rate declined as the temperature decreased. Neither the temperature at which the tissue was grown nor BASF 13 338 treatment influenced the ability of root tips to respire at any temperature from 4 to 30 C. The results indicated that the relative proportion of linolenic acid to linoleic acid did not influence the plants ability to grow and respire over the range of temperatures tested.  相似文献

The effect of pretreatment of cucumber (Cucumis sativus L.) roots with choline chloride or ethanolamine on leaf phospholipid composition and light-induced leaf damage during chilling was studied. Photooxidative chlorophyll degradation was similarly inhibited by both amino alcohols. The decrease of the chlorophyll a/chlorophyll b ratio and the increase of polyunsaturated-fatty-acid degradation during chilling in the light were equally inhibited by pretreatment with choline chloride or ethanolamine. Treatment with choline chloride and ethanolamine caused, respectively, 43% and 26% increases in the total phospholipid contents of the leaves. After treatment with choline chloride, the phosphatidylcholine content was higher than the content of phosphatidylethanolamine; the reverse was true after treatment with ethanolamine. The chlorophyll concentration increased less than the phospholipid concentration, resulting in a decreased chlorophyll/phospholipid ratio of treated leaves. During chilling in the light, degradation of phosphatidylcholine, ethanolamine and phosphatidyl glycerol occurred. Phosphatidyl glycerol was less sensitive than phosphatidylcholine and ethanolamine. The degradation was equally inhibited by pretreatment with either amino alcohol. Freezing and thawing of winter rape leaves was found to cause a marked decrease of total phospholipid content in general, and that of phosphatidyl choline in particular. In the highly injured leaves the decrease of phosphatidyl choline was irreversible, and it was accompanied by a marked increase of phosphatidic acid. In the slightly damaged leaves no phosphatidic acid accumulation was detected, irrespective of phosphatidyl choline decrease. Instead of phosphatidic acid, the phosphatidyl glycerol or an unidentified P-containing lipid increased after thawing in leaves hardened to the first (Tk50=?8°C) or to the second (Tk50=?13.5°C) level, respectively. Frost hardening was found to promote phospholipid recovery after freezing and to increase phospholipid content, including that of phosphatidyl choline and phosphatidyl ethanolamine. Possible association of frost hardening with modified phospholipase-D properties is suggested.  相似文献

Phospholipid composition of sheep blood platelets and its various plasma membrane fractions have been analyzed. Based on their flotation rates in discontinuous sucrose density gradient centrifugation, three membrane fractions were isolated. 5'-Nucelotidase and alkaline phosphatase were distributed nearly equally in all the three membrane fractions. However these membrane fractions showed differences in the distribution of phosphatidyl ethanolamine, phosphatidyl choline and phosphoinositides. Phosphatidyl ethanolamine was predominant in fraction I (11.05 micrograms PLP/mg protein) while phosphatidyl choline was predominant in fractions II and III (110.10 and 68.30 micrograms PLP/mg protein respectively). Phosphatidyl inositol (Ptd-InsP) was equally distributed in all three membrane fractions. However, both Ptd-InsP and phosphatidyl inositol 4,5-bisphosphate were about 4-fold higher in fraction II (73.55 and 89.89 micrograms PLP/mg protein respectively).  相似文献
