[5-Br]5′-UMP 钠盐经催化氚化脱卤,纸层析分离得到[5-T]5′-UMP 钠盐。[5-T]5′-UMP 钠盐经酶促磷酸化后,纸层析分离得到[5-T]5′-UTP 钠盐(同时得到[5-T]5′-UDP 钠盐)。经鉴定紫外吸收光谱性质与标准样品一致,比放射性达10居里/毫克分子,放化纯度在98%以上。

Characterization of Imido [8-3H] Guanosine 5′-Triphosphate Binding Sites to Rat Brain Membranes

Besides their well-defined intracellular roles in transmembrane signals transduction, guanine derivatives play important roles by acting from the outside of neural cell membranes. These roles are mediated by two different pool sites in cell membranes: G proteins, which bind to specific (GDP and GTP) intracellular guanine derivatives, and sites that bind to extracellular guanine derivatives. In this study we investigated some methodological characteristics of both guanine derivatives binding sites (intracellular and extracellular) in rat brain neural membranes. By investigating the binding of a poorly hydrolyzed GTP analogue and the adenylate cyclase activity in neural membranes, we observed some distinctiveness of guanine derivatives binding sites: stability to washing procedures (extracellular) and modulation of adenylate cyclase activity (intracellular). These results allow dealing with each site separately, which could be useful for discriminating the roles of extracellular and intracellular guanine derivatives in the central nervous system.     

5-碘吲哚酚[3]苯基膦酸酯的合成及应用

本文利用苯基氧氯化膦和N-乙酰-5-碘吲哚酚为原料,建立了5-碘吲哚酚[3]苯基膦酸酯铵盐的合成和纯化方法。并报道该产品的紫外光谱、红外光谱、元素分析等理化性质,以及作为酶底物的应用情况。实验结果说明,合成的产品经提取和重结晶可达到层析纯。氢、氮、碳元素分析数据与理论结构计算值相符。产品在常温条件下保存两年以上无结构变化,受酶催化的专一性好,能用于磷酸二酯酶分析和组织化学研究。     

[^3H]Cortisol及[^3H]Dexamethasone在大鼠肝细胞膜上的特异结合位点

本研究应用［３Ｈ］ｃｏｒｔｉｓｏｌ和［３Ｈ］ｄｅｘａｍｅｔｈａｓｏｎｅ（ＤＥＸ）两种配基,观察到大鼠肝细胞膜上存在一类糖皮质激素（ＧＣ）特异结合位点。这些位点与ＧＣ的结合具有饱和性、高亲和力及低容量。其平衡解离常数（Ｋｄ）分别为１２．８４±６．５８ｎｍｏｌ／Ｌ和４０．２７±２３．４４ｎｍｏｌ／Ｌ;最大结合容量（Ｂｍａｘ）分别为２．５７±１．８４ｐｍｏｌ／ｍｇ蛋白质与０．６４±０．１８ｐｍｏｌ／ｍｇ蛋白质（ｃｏｒｔｉｓｏｌ,ｎ＝４;ＤＥＸ,ｎ＝３;±ＳＥ）。动力学实验数据所得的Ｋｄ值与Ｓｃａｔｃｈａｒｄ分析所得的Ｋｄ值结果基本一致。［３Ｈ］ｃｏｒｔｉｓｏｌ和［３Ｈ］ＤＥＸ饱和结合实验数据用Ｓｃａｔｃｈａｒｄ作图分析,均显示为直线。Ｈｉｌｌ系数则分别为０．９８８０和０．９９９０．竞争抑制实验结果表明,ｃｏｒｔｉｓｏｌ对［３Ｈ］ｃｏｒｔｉｓｏｌ的结合位点有高度特异性竞争,比其它几种类固醇（强的松、黄体酮、ＲＵ４８６、ＤＥＸ）的竞争力至少强４０倍以上．用放射自显影技术进行研究,也提供了［３Ｈ］ｃｏｒｔｉｓｏｌ特异结合银粒位于大鼠肝细胞膜上的依据。     

环磷酸鸟苷(cGMP)单克隆抗体杂交瘤细胞系的建立及其特性的鉴定

Kinetics of [3H]prazosin and [3H]dihydroalprenolol binding to α1- and β-adrenoreceptors in rat brain cortex membranes

The binding of nonselective α₁- and β-adrenoreceptor antagonists [³H]prazosin and [³H]dihydroalprenolol ([³H]DHA) to rat cerebral cortex synaptosomal membranes has been studied. It is found that ligand-receptor interactions of α₁-adrenoreceptors fit into a single receptor pool model, which assumes the binding of two ligand molecules to one receptor molecule. The parameters of [³H]prazosin binding to α₁-adrenoreceptors are as follows: K _d = 2.58 ± 0.20 nM; B _m = 2.95 ± 1.12 fmol/mg protein; Hill coefficient, n = 2. For β-adrenoreceptors, ligand-receptor interactions fit into a model assuming the presence of two receptor pools in the same effector system and binding of two ligand molecules to one receptor molecule. The corresponding parameters of the [³H]DHA binding to β-adrenoreceptors are as follows: K _d1 = 0.74 ± 0.09 nM; K _d2 = 7.63 ± 0.70 nM; B _m1 = 25 ± 2 fmol/mg, B _m2 = 48 ± 2 fmol/mg, n ₁ = 2; n ₂ = 2. We suggest that in rat cerebral cortex membranes α-and β-adrenoreceptors exist as dimers.     

[~3H]2-脱氧葡萄糖定量研究方法的介绍

2-脱氧葡萄糖(2-DG)方法是一种新的形态与功能结合的神经科学研究方法。我们在现有条件下对此方法的应用做了一些研究。在实验中,我们采用了股静脉注射[~3H]标记的2-DG,测定血糖、血液中放射性物质含量及用显微分光光度计测定放射自显影片光密度的方法,并采用 Sokoloff 的计算公式计算各脑区的葡萄糖代谢率。

Discovery of trans-N-[1-(2-fluorophenyl)-3-pyrazolyl]-3-oxospiro[6-azaisobenzofuran-1(3H),1′-cyclohexane]-4′-carboxamide,a potent and orally active neuropeptide Y Y5 receptor antagonist

A series of trans-3-oxospiro[(aza)isobenzofuran-1(3H),1′-cyclohexane]-4′-carboxamide derivatives were synthesized to identify potent NPY Y5 receptor antagonists. Of the compounds, 21j showed high Y5 binding affinity, metabolic stability and brain and cerebrospinal fluid (CSF) penetration, and low susceptibility to P-glycoprotein transporters. Oral administration of 21j significantly inhibited the Y5 agonist-induced food intake in rats with a minimum effective dose of 1 mg/kg. This compound was selected for proof-of-concept studies in human clinical trials.     

转基因香石竹中F3’5’H基因的克隆、表达和免疫学鉴定

在研究转基因香石竹品系月之霓裳（Moonshade）、月之伊人（Moonlite）中外源基因F3’5’H的表达中,本文克隆了F3’5’H全长基因1.5kb,构建获得工程菌株Escherichia coli BL21（DE3）（＋F3＇5＇H）。SDS-PAGE分析的结果显示,该菌株高效表达出F3’5’H重组蛋白,约占菌体总蛋白的30%。用经纯化的F3’5’H重组蛋白作为抗原,制备F3’5’H重组蛋白的抗血清,经ELISA免疫学分析表明,该抗血清的效价为1：25600。Western blot结果表明F3’5’H重组蛋白具有良好的IgG结合活性,且抗血清与转基因香石竹品系月之霓裳和月之伊人中的外源基因F3’5’H所表达的蛋白发生明显的抗原抗体反应。这样,月之霓裳和月之伊人用于评价转基因香石竹品系的环境安全性在我国也得到了验证。     

3¢-磷酸腺苷-5¢-磷酸硫酸的高效合成及其应用

硫酸化化合物广泛存在于胞浆、细胞表面及胞外基质中,在机体细胞发育、分化、免疫、解毒和信号传递等生命活动过程中起着不可替代的作用。3'-磷酸腺苷-5'-磷酸硫酸(3'-phosphoadenosine-5'-phosphosulfate,PAPS)是化合物硫酸化过程中最常用的硫酸基供体,但目前合成PAPS并最终实现其工业化应用还困难重重。文中主要综述过去10年内关于PAPS的生物合成及应用的研究进展,以期为PAPS的合成及其在芥子油苷、肝素、硫酸软骨素及羟胺硝喹等的生物合成中的应用提供参考。

饱和分析法测定3′,5′-环一磷酸腺苷(cAMP)

本文提供了一个改进的3',5'-cAMP饱和分析法。用膜片分离自由的和与蛋白激酶结合的cAMP,直接用膜片法测定氚的放射性强度,灵敏度在微微克分子水平。同时对于各种影响测定的因素进行了研究和讨论。     

Synthesis and X-Ray Crystal Structure of 3-Amino-1-β-D-Ribofuranosyl-s-Triazolo[5, 1-c]-s-Triazole

Abstract

The first chemical synthesis of 3-amino-1-β-D-ribofuranosyl-s-triazolo[5,1-c]-s-triazole (6) is described. Direct glycosylation of 3-amino-5(7)H-s-triazolo[5,1-c]-s-triazole (2) with 1-O-acetyl-2,3,5-tri-O-benzoyl-D-ribofuranose (3) in the presence of TMS-triflate gave 3-amino-1-(2,3,5-tri-O-benzoyl-β-D-ribofuranosyl)-s-triazolo[5, 1-c]-s-triazole (4) which, on ammonolysis, gave 6. The absolute structure of 6 is determined by X-ray diffraction techniques employing Mo Kα radiation. The structure is solved by direct methods and refined to the R value of 0.044 by using a full-matrix least-squares method. The sugar of 6 has a ³T₂ configuration. The torsion angles about the C5′–C4′ bond are both gauche and the torsion angle about the glycosidic bond is in the anti range. Each azole ring of the aglycon is planar and the dihedral angle between the planes of the rings is 3.6°.     

[B_1Ala,B_2Ala,B_3Lys]-胰岛素的制备和生物活性(英文)

本文报道了胰岛素分子中Ｂ１～３序列（Ｐｈｅ－Ｖａｌ－Ａｓｎ）为Ａｌａ－Ａｌａ－Ｌｙｓ取代的胰岛素类似物制备及其生物性质。［Ｂ１Ａｌａ,Ｂ２Ａｌａ,Ｂ３Ｌｙｓ］－胰岛素仍保留天然胰岛素的全部体内活性和受体结合能力,但体外促脂肪生成活性和免疫活性分别只为胰岛素的７０％和０．８８％。本文还就胰岛素Ｂ链Ｎ端肽段对其结构和功能的影响进行了讨论。     

阴离子交换树脂分离制备腺嘌呤核糖核苷(3′，5′-)环磷酸(cAMP)

腺嘌呤核苷3′,5′-环磷酸对癌细胞在特定条件下有抑制作用,对冠心病和牛皮癣等有缓解作用,临床上已证明。本文所介绍的合成、分离、纯化、鉴定等方法,简易而又可靠,比目前国外报道的方法有很大改进,特别是采用国产离子交换树脂,分离简便、容量大、效果好,为大量生产创造了有利条件。——编者
