In the early nineties, Undaria pinnatifida has been accidentally introduced to Nuevo Gulf (Patagonia, Argentina) where the environmental conditions would have favored its expansion. The effect of the secondary treated sewage discharge from Puerto Madryn city into Nueva Bay (located in the western extreme of Nuevo Gulf) is one of the probable factors to be taken into account. Laboratory cultures of this macroalgae were conducted in seawater enriched with the effluent. The nutrients (ammonium, nitrate and phosphate) uptake kinetics was studied at constant temperature and radiation (16?°C and 50 μE m?2 s?1 respectively). Uptake kinetics of both inorganic forms of nitrogen were described by the Michaelis–Menten model during the surge phase (ammonium: V max sur: 218.1 μmol h?1 g?1, K s sur: 476.5 μM and nitrate V max sur: 10.7 μmol h?1 g?1, K s sur: 6.1 μM) and during the assimilation phase (ammonium: V max ass: 135.6 μmol h?1 g?1, K s ass: 407.2 μM and nitrate V max ass: 1.9 μmol h?1 g?1, K s ass: 2.2 μM), with ammonium rates always higher than those of nitrate. Even though a net phosphate disappearance was observed in all treatments, uptake kinetics of this ion could not be properly estimated by the employed methodology.  相似文献

Uptake of 14C-labelled sucrose and glucose by isolated seed coat halves of pea (Pisum sativum L. cv. Marzia) seeds was measured in the concentration range <0.1 μM to 100 mM. The initial influx of sucrose was strictly proportional to the external concentration, with a coefficient of proportionality (k) of 6.2 μmol·(g FW)?1·min?1·M?1. Sucrose influx was not affected by 10 μM carbonylcyanide m-chlorophenylhydrazone (CCCP), but it was inhibited by 40% in the presence of 2.5 mM p-chloromercuribenzenesulfonic acid (PCMBS). Influx with diffusional kinetics was also observed for glucose (k = 4.8 μmol·(g FW)?1·min ?1·M ?1) and mannitol (k = 5.1 μmol·(g FW)?1·min?1·M?1). For glucose an additional saturable system was found (Km = 0.26 mM, V max = 4.2 nmol·(g FW)?1·min?1), which appeared to be completely inhibited by CCCP and partly by PCMBS. In contrast to the diffusional pathway, uptake by this saturable system was slightly pH-dependent, with an optimum at pH 5.5. The influx of sucrose appears to be by the same pathway as the efflux of endogenous sucrose, which was inhibited by 36% in the presence of 2.5 mM PCMBS (De Jong A, Wolswinkel P, 1995, Physiol Plant 94: 78–86). It is argued that passive transport may be the only mechanism for sucrose transport through the plasma membrane of seed coat parenchyma cells. The estimated permeability coefficient of the plasma membrane for sucrose (P = 3.5·10?7 cm·s?1) is more than 1 × 106-fold higher than that reported for artificial lipid membranes. This relatively high permeability is hypothesized to result from pore-forming proteins that allow the diffusion of sucrose. Furthermore, it is shown that a sucrose gradient across the plasma membrane of the seed coat parenchyma of only 22 mM will suffice to result in the net efflux of sucrose which is required to feed the embryo.  相似文献

Uptake of phosphate ions by 1 mm segments of isolated maize root cortex layers was studied. Cortex segments (from roots of 8 days old maize plants) absorb phosphate ions from 1 mM KH2PO4 in 0.2 mM CaSCO4 at the average rate of 34.3 ±3.2 μg Pi g?1 (fr. m.) h?1,i.e. 0.35± 0.02 μmol Pi g?1 (fr. m.) h?1. Phosphate uptake considerably increases after a certain period of “augmentation”,i.e. washing in aerated 0.2 mM CaSO4. This increase is completely blocked by the presence of 10 μg ml?1 cycloheximide. The relation of uptake rate to phosphate concentration in the medium was shown to have 3 phases in the concentration range of 0.02 - 40 mM. Transition points were found between 0.8–1 mM and 10–20 mM. Following Km and Vmax values were found: Km[mM] : 0.37 - 3.82 - 27.67 Vmax[μg Pi g?1 (fr. m.) h?1] : 3.33 - 39.40 - 66.67 We have found no sharp pH optimum for phosphate uptake. It proceeds at almost constant rate till pH 6.0 and then the uptake rate drops with increasing pH. At low phosphate concentrations (1 mM) the lowest uptake rate was found at 5 and 13 °C, while the uptake is higher at 5 °C than at 13 °C at phosphate concentrations higher than 1 mM. At these concentrations uptake rate at 35 °C is lower than at 25 °C. Phosphate uptake considerably decreased in anaerobic conditions. DNP and iodoacetate (0.1 mM) completely blocked phosphate uptake from 1 mM KH2PO4, while uptake from 5 and 10 mM KH2PO4 was left unaffected by these substances. The inhibitors of active - SH groups NEM and PCMB inhibited phosphate uptake: 10?3 M NEM by 81.6%, 104 M NEM by 42% and 10?4 M PCMB by 42%.  相似文献

Suspensions of 2 to 5% rat thymocytes were incubated at 35 °C in buffered balanced salt solution (pH 7.3) with lactate and β-hydroxybutyrate as fuels. The dependence of 3-O-[Me-3H]methylglucose influx on external and internal 3-O-methylglucose concentrations was studied. Entry was almost rectilinear during the first minute. From the dependence of methylglucose entry (into sugar-free cells) on external methylglucose concentration, we judged the entry Km to be about 7.7 mm and the entry V to be about 0.64 μmol · min?1 · (ml of packed cell volume)?1. Methylglucose inside the cell enhanced influx, hence equilibrium exchange was faster than entry. The dependence of equilibrium exchange on methylglucose concentration (inside and outside being equal) indicated a Km of about 25 mm and a V of about 2.1 μmol · (min)?1 · (ml of cell volume)?1. This effect of internal sugar indicated that entry into sugar-free cells is limited mainly by the return of empty carrier to the outside surface and that loading the carrier on the inside enhances its outward mobility. The Km and V for influx into cells containing 21 mm methylglucose were 5.9 mm and 1.17 μmol · min?1 · (ml of packed cells)?1. The effect of 21 mm internal sugar on lowering the influx Km from about 7.7 mm to about 6 mm was reproducible and contributed to the evaluation of the constants of the transport rate law. It indicated that loading of the carrier at the external surface reduces its mobility, in contrast to the effect of loading on the inside. Mechanical explanations for this behavior are discussed.  相似文献

Measurements of bimodal oxygen uptake have been made in a freshwater air-breathing fish,Notopterus chitala at 29.0±1(S.D.)°C. xhe mean oxygen uptake from continuously flowing water without any access to air, was found to be 3.58±0.37 (S.E.) ml O2 · h?1 and 56.84+4.29 (S.E.) ml O2 · kg?1 · h?1 for a fish weighing 66.92 + 11.27 (S.E.) g body weight. In still water with access to air, the mean oxygen uptake through the gills were recorded to be 2.49 ± 0.31 (S.E.) ml O2 · h?1 and 38.78 ± 1.92 (S.E.) ml O2 · kg?1 · h?1 and through the accessory respiratory organs (swim-bladder) 6.04±0.87 (S.E.) ml O2 · h?1 and 92.32±2.91 (S.E.) ml O2 · kg?1 · h?1 for a fish averaging 66.92±11.27 (S.E.) g. Out of the total oxygen uptake (131.10 ml O2 · kg?1 · h?1), about 70% was obtained through the aerial route and the remainder 30% through the gills.  相似文献

This study examines the seasonal physiological response of the codominant, perennial brown algae Lobophora variegata (Lamour.) Womers. and Zonaria tournefortii (Lamour.) Mont. to experimental temperatures approximating the seasonal range of their North Carolina continental shelf habitat. Respiration rates of Lobophora over experimental temperatures (10, 21, 27, and 34°C) seasonally varied, suggesting an acclimation response. The respiration rates of both species at experimental temperatures were generally lower for plants collected in the winter than for those tested in summer, which might enhance winter survival, when seasonally low irradiance might limit photosynthesis. Photosynthetic performance for both species showed significant physiological acclimation throughout the study period. Notably, both species had distinctive seasons of optimal photosynthetic performance. For example, photosynthetic performance at temperatures closest to ambient ranged from 30 (in September) to about 65 (in January and May) nmol O2·nmol Chla?1·h?1 for Lobophora. In contrast, lowest photosynthetic rate (40 nmolO2·nmolChla?1 · h?1) for Zonaria occurred in January; highest photosynthetic performance occurred in May (78 nmolO2 · nmolChla?1·h?1). This difference in their photosynthetic performance may contribute to their continued coexistence in a habitat characterized by large seasonal temperature fluctuations.  相似文献

Using time-course, natural-light incubations, we assessed the rate of carbon uptake at a range of light intensities, the effect of supplemental additions of nitrogen (as NH4+ or urea) on light and dark carbon uptake, and the rates of uptake of NH4+ and urea by phytoplankton from Vineyard Sound, Massachusetts from February through August 1982. During the winter, photoinhibition was severe, becoming manifested shortly after the start of an incubation, whereas during the summer, there was little to no evidence of photoinhibition during the first several hours after the start of an incubation. At light levels which were neither photoinhibiting nor light limiting, rates of carbon uptake normalized per liter were high and approximately equal during winter and summer (22–23 μg C·l?1 · h?1), and low during spring (<10 μgC·l?1· h?1). In contrast, on a chlorophyll a basis, rates of carbon fixation were as high during spring (15–20μg C·μg Chl a?1·h?1), when concentrations of chlorophyll a were at the yearly minimum (<0.5 μg · l?1) as during the summer, when chlorophyll a concentrations were substantially higher (0.8–1.3 μg · l?1). Highest rates of NH4+ and urea uptake were observed during summer, and at no time of the year was there evidence for severe nitrogen deficiency, although moderate nitrogen nutritional stress was apparent during the summer months.  相似文献

The uptake kinetics of ammonium and phosphate by Gracilaria tikvahiae McLachlan were studied under field conditions. Seaweeds, pulse fed once a week for 6 h over a 4-week period, had maximum uptake rates of 19 μmol·g fwt?1·h?1 for ammonium and 0.28 μmol·g fwt?1·h?1 for phosphate. For both nutrients there was a positive linear correlation between uptake rate (v) and concentration (S) over the entire range of concentration tested. In a nutrient depletion experiment, the phosphate uptake curve determined over a wide range of concentrations consisted of two stages of saturation at low concentrations, and a linear phase at high concentrations. Ash free dry weight, chlorophyll a, phycoerythrin, and protein content were higher in pulse fed plants than in control plants receiving no nutrient additions, while the reverse held true for carbohydrate contents and the C/N ratios. The C/N ratio inversely correlated with ammonium and phosphate uptake rate as well as protein and phycoerythrin content, and positively with carbohydrate content.  相似文献

We investigated copper (Cu) acquisition mechanisms and uptake kinetics of the marine diatoms Thalassiosira oceanica Hasle, an oceanic strain, and Thalassiosira pseudonana Hasle et Heimdal, a coastal strain, grown under replete and limiting iron (Fe) and Cu availabilities. The Cu‐uptake kinetics of these two diatoms followed classical Michaelis–Menten kinetics. Biphasic uptake kinetics as a function of Cu concentration were observed, suggesting the presence of both high‐ and low‐affinity Cu‐transport systems. The half‐saturation constants (Km) and the maximum Cu‐uptake rates (Vmax) of the high‐affinity Cu‐transport systems (~7–350 nM and 1.5–17 zmol · μm?2 · h?1, respectively) were significantly lower than those of the low‐affinity systems (>800 nM and 30–250 zmol · μm?2 · h?1, respectively). The two Cu‐transport systems were controlled differently by low Fe and/or Cu. The high‐affinity Cu‐transport system of both diatoms was down‐regulated under Fe limitation. Under optimal‐Fe and low‐Cu growth conditions, the Km of the high‐affinity transport system of T. oceanica was lower (7.3 nM) than that of T. pseudonana (373 nM), indicating that T. oceanica had a better ability to acquire Cu at subsaturating concentrations. When Fe was sufficient, the low‐affinity Cu‐transport system of T. oceanica saturated at 2,000 nM Cu, while that of T. pseudonana did not saturate, indicating different Cu‐transport regulation by these two diatoms. Using CuEDTA as a model organic complex, our results also suggest that diatoms might be able to access Cu bound within organic Cu complexes.  相似文献

Uptake of d-glucosamine by rat brain synaptosomes occurs via a saturable transport process (Km 2.1 mM, V 3.0 nmol/mg per min) which was clearly distinguishable from simple diffusion. This transport process is highly sensitive to cytochalasin (Ki = 7 · 10?5 mM. d-Glucose competitively inhibits d-glucosamine uptake with a Ki value of 8 · 10?1 mM.  相似文献

Ambient sea-water nitrate and tissue nitrogen (ethanol soluble nitrate and amino acids, as well as total nitrogen) of Macrocystis integrifolia Bory were monitored over a 2-yr period in Bamfield, Vancouver Island, British Columbia. Sea-water nitrate varied from a high of 12 μmol · 1?1 (individual values as high as 23 μmol · 1?1 were recorded) in late winter to below detection limits for most of the summer. Tissue nitrate and total nitrogen paralleled the ambient nitrate levels and showed summer minima and winter maxima (from 0 to 70 μmol · g fresh wt?1 for nitrate and from 0.8 to 2.9% of dry wt for total N). The nitrate uptake capacity was inversely proportional to tissue nitrate concentration and, furthermore, was much higher for subapical surface blades (60–70 nmol · cm?2 · h?1) than for older, deeper blades (5–10 nmol · cm?2 · h?1). Nitrate uptake by subapical blade disks in summer is apparently higher in dark (1.0–1.7 μmol · g fresh wt?1 · h?1) than in light (0.6–1.3 μmol · g fresh wt?1 · h?1) and the data obtained in 36–108 h experiments indicate nitrate pool sizes of 30–90 μmol · g fresh wt?1. These pools are 23 to nearly full in winter. Ammonium does not inhibit nitrate uptake. It is taken up and apparently utilized much faster than nitrate and it may well be an important source of nitrogen for marine macrophytes.  相似文献

In all larval stages of Carcinus maenas L. oxygen consumption was measured at three temperatures (12,18,25 °C). Values increased during development and were in the range of 0.037 ± 0.01 (zoea-1, 12°C, x? ± 95% CL) to 0.734 ± 0.047 μl O2 · h?1 · ind?1 (megalopa, 25 °C). Growing larvae showed temperature dependent trends in weight specific respiration rates (referred to dry wt; DW), with values between ≈2.4 and 9.4 μl O2· h?1·mg DW?1. Increase in oxygen consumption of megalops did not differ much at temperatures between 18 and 25 °C. This points to an exceptional physiological position of this stage. Fed zoea-1 of C. maenas (18 °C) revealed growth rates in terms of 40% DW, 20% carbon (C), 30% nitrogen (N) and 65% hydrogen (H). At the same time larvae gained individual energy by 13% (J · ind?1), while weight specific energy dropped by ≈ 19% (J · mg DW?1) during the first day and remained constant until the moult. Starved zoea-1 of C. maenas (18 ° C) gained ≈ 20 % in DW through the first day, probably caused by inorganic salts which enter the organism after the moult of the prezoea. DW dropped to ≈ 25 % of initial value, when starvation continued. Single components decreased by ≈50% (C), 54% (N), 57% (J · ind?1). Weight specific energy (J · mg DW?1) decreased by 40% during the first 4 days of starvation, remaining constant thereafter. Individual respiration rate (R) dropped by 61 %, weight specific respiration rate (QO2) by 55 %. Individual energy loss in starved zoea-1 was 0.077 J over a period of 11 days. In this period ≈ 9.3 μl O2·ind?1 were consumed. Thus effective oxygen capacity was lower than in growing larvae. It dropped to 5.3 J·mlO2?1 after 4 days and remained constant if starvation continued, i.e. 65 % of possible energy loss occurred during the first 4 days. Decrease in requirement for oxygen and its effective capacity were both recognized as independent components of survival during starvation. Partitioning of energy through individual larval development of C. maenas was investigated for all five larval stages. The cumulative budget could be calculated: consumption (C) = 28.23 J, growth (G) = 0.92 J, exoskeleton (Ex) = 0.20 J, metabolism (M) = 5.30 J, egestion and excretion (E) = 21.82 J. Mean gross and net growth efficiency were, K1 = 3.3% and K2 = 14.8%, respectively.  相似文献

Steady state Cl? flux across the Ehrlich mouse ascites cell membrane was studied when gluconate replaced Cl? in the external medium. Saturation behavior was observed; K12 was 23.9 mM Cl? and V was 758 μmol · g?1 dry weight · h?1. The cells lost K+, Cl? and H2O, consistent with relative impermeability to gluconate, and the Cl? efflux rate coefficient was elevated. The results indicate that a major portion of Cl? exchange occurs as a membrane transport process and suggest that the process is sensitive to intracellular Cl? levels.  相似文献

Clones of Skeletonema costatum (Grev.) Cl. isolated from Narragansett Bay, R.I., during different seasons were grouped according to their electrophoretic banding patterns. The growth rates, pg chlorophyll · cell?1, carbon uptake · cell?1· h?1, and carbon uptake · pg chl?1· h?1 were measured at 20°C, in a 14:10 h L:D cycle at 180 μE · m?2· s?1. Statistically significant sources of variation were found among groups of clones in growth rate, pg chl · cell?1, and carbon uptake · pg chl?1· h?1. It was concluded that there is a significant relationship between the physiological characteristics of clones isolated from populations in different seasons and patterns of genetic variation inferred from the electrophoretic studies. However, genetic diversity detected by banding patterns tends to underestimate the total genetic diversity in natural populations. The groups of clones most common in summer bloom populations had significantly higher growth rates, lower values of pg chl · cell?1, and higher rates of carbon uptake · pg chl?1· h?1 at 20°C than did the group of clones most common in winter bloom populations. However, differences among groups in these parameters at 20°C alone cannot account for the seasonal cycling of genetically variable populations of Skeletonema in Narragansett Bay. The range of growth rates among clones of this species is 0.1–5.0 divisions · d?1 under a single set of temperature and light conditions. Chlorophyll concentrations range from 0.2–1.7 pg chl · cell?1 and carbon uptake · pg chl?1· h?1 varies by a factor of 7 among clones. The range of physiological variation in this species means that it is difficult to use laboratory studies of single clones to analyze the responses of natural populations of Skeletonema.  相似文献

When intact chloroplasts are incubated in the dark with dihydroxyacetone phosphate, an increase in fructose 1,6-bisphosphatase activity occurs which resembles the reductive activation observed in illuminated chloroplasts. Under optimum conditions, the activity increases to about 150 μmol · h?1 · mg?1 chlorophyll within 60 min. The dark activation of the enzyme is reversed by electron acceptors such as oxaloacetate, nitrite, and 3-phosphoglycerate plus ATP. Activation is most marked under strictly anaerobic conditions, being strongly inhibited by O2. It is concluded that NADPH, generated from dihydroxyacetone phosphate in situ in the reaction catalyzed by NADP+-dependent glyceraldehyde phosphate dehydrogenase, can provide electrons for the reductive activation of fructose 1,6-bisphosphatase in the dark.  相似文献

1. Dihydroxyacetone phosphate in concentrations ? 2.5 mM completely inhibits CO2-dependent O2 evolution in isolated intact spinach chloroplasts. This inhibition is reversed by the addition of equimolar concentrations of Pi, but not by addition of 3-phosphoglycerate. In the absence of Pi, 3-phosphoglycerate and dihydroxyacetone phosphate, only about 20% of the 14C-labelled intermediates are found in the supernatant, whereas in the presence of each of these substances the percentage of labelled intermediates in the supernatant is increased up to 70–95%. Based on these results the mechanism of the inhibition of O2 evolution by dihydroxyacetone phosphate is discussed with respect to the function of the known phosphate translocator in the envelope of intact chloroplasts.2. Although O2 evolution is completely suppressed by dihydroxyacetone phosphate, CO2 fixation takes place in air with rates of up to 65μ mol · mg?1 chlorophyll · h?1. As non-cyclic electron transport apparently does not occur under these conditions, these rates must be due to endogenous pseudocyclic and/or cyclic photophosphorylation.3. Under anaerobic conditions, the rates of CO2 fixation in presence of dihydroxyacetone phosphate are low (2.5–7 μmol · mg?1 chlorophyll · h?1), but they are strongly stimulated by addition of dichlorophenyl-dimethylurea (e.g. 2 · 10?7 M) reaching values of up to 60 μmol · mg?1 chlorophyll · h?1. As under these conditions the ATP necessary for CO2 fixation can be formed by an endogenous cyclic photophosphorylation, the capacity of this process seems to be relatively high, so it might contribute significantly to the energy supply of the chloroplast. As dichlorophenyl-dimethylurea stimulates CO2 fixation in presence of dihydroxyacetone phosphate under anaerobic but not under aerobic conditions, it is concluded that only under anaerobic conditions an “overreduction” of the cyclic electron transport system takes place, which is removed by dichlorophenyl-dimethylurea in suitable concentrations. At concentrations above 5 · 10?7 M dichlorophenyl-dimethylurea inhibits dihydroxyacetone phosphate-dependent CO2 fixation under anaerobic as well as under aerobic conditions in a similar way as normal CO2 fixation. Therefore, we assume that a properly poised redox state of the electron transport chain is necessary for an optimal occurrence of endogenous cyclic photophosphorylation.4. The inhibition of dichlorophenyl-dimethylurea-stimulated CO2 fixation in presence of dihydroxyacetone phosphate by dibromothymoquinone under anaerobic conditions indicates that plastoquinone is an indispensible component of the endogenous cyclic electron pathway.  相似文献

Oxygen consumption rates (QO2) of laboratory reared stage one zoeae of Pandalus borealis (Krøyer) at 1.5, 3, 4.5, 6, and 9°C were 1.5, 2.2, 2.6, 3.6 and 4.1μ O2 · mg?1 · h?1, respectively. These values of QO2 correspond to 0.26, 0.38, 0.44, 0.60, and 0.70 μl O2 · individual?1 · h?1. At 10.5 °C oxygen consumption rates decreased suggesting thermally induced respiratory stress.The equation log10QO2 = 0.55 log10T°C + 0.086 describes the relationship between QO2 (μl O2 · mg?1 · h?1) and sea-water temperature between 1.5 and 9°C. Corresponding values of QO2 for an individual (μl O2 · h?1) exhibited the relationship log10QO2 = 0.55 log10T°C ?0.686.The minimum daily metabolic caloric requirements for an individual zoea ranged from 0.04 at 3 °C to 0.07 calories per day at 8 °C. The number of calories ingested daily ranged from 0.4 to 0.5 at 3 to 8 °C.  相似文献

Photosynthesis-irradiance relationships were determined in the field for five species of littoral and shallow sublittoral marine benthic green algae (Chlorophyta) of differing morphologies. Each species exhibited a linear increase in photosynthetic rate with increasing irradiance up to a maximum light-saturated value. Full sunlight (1405 to 1956 μE·m?2·s?1) inhibited photosynthesis of all species except the thick, optically dense, Codium fragile (Sur.) Har. Compensation irradiances ranged from 6.1 μE·m?2·s?1 for Enteromorpha intestinalis (L.) Link to 11.4 μE·m?2·s?1 for Ulva lobata (Kütz) S. & G. and did not reveal a consistent relationship to seaweed morphology. Saturation irradiances were determined statistically (Ik) and visually from graphical plots. with the latter technique resulting in values three to eight times higher and different comparative rankings of species than the former. Ik saturation irradiances were highest for Chaetomorpha linum (Müll.) Kütz. (81.9 μE·m?2·s?1) and lowest for Codium fragile (49.6 μE·m?2·s?1) and did not reveal a relationship with seaweed morphology. Regression equations describing light-limited photosynthetic rates and the relative magnitudes of the maximal net photosynthetic responses both strongly suggested a relationship with seaweed morphology. Highest net photosynthetic rates were obtained for the thin, sheet-like algae Ulva lobata (9.2 mg C·g dry wt?1·h?1), U. rigida C. Ag. (6.5 mg C·g dry wt?1·h?1) and the tubular form, Enteromorpha intestinalis (7.3 mg C·g dry wt?1·h?1), while lowest rates occurred for Codium fragile (0.9 mg C·g dry wt?1·h?1). Similarly, steepest light-limited slopes were found for the algae of simpler morphology, while the most gradual slope was determined for Codium fragile, the alga with greatest thallus complexity.  相似文献

Green thallus cells of the aquatic liverwort, Riccia fluitans, are rapidly depolarized in the presence of 1–20 μM NH4Cl and 5–100 μM CH3NH3Cl, respectively. Simultaneously, the membrane conductance is increased from 0.41 to 1.2 S · m?2. Uptake of [14C]methylamine is stimulated by increasing [K+]o and inhibited by increasing [Na+]o or [H+]o, is highly voltage sensitive, and saturates at low amine concentrations.Double-reciprocal plots of (a) maximal membrane depolarization and (b) methylamine uptake vs. external amine concentration give apparent Km values of 2 ± 1 μM ammonia and 25–50 μM methylamine; Km values for changes in conductance and membrane current are greater and voltage dependent. Whereas the amine transport into the cell is strongly inhibited by CN?, the amine efflux is stimulated. The current-voltage characteristics of the ammonia transport are represented by a sigmoid curve with an equilibrium potential of ?60 mV, and this is understood as a typical carrier curve with a saturation current of about 70 mA · m?2. It is further concluded that the evidently carrier-mediated transport is competitive for the two amines tested, and that ammonia and methylamine are transported in the protonated form as NH4+ and CH3NH3+ into the cytoplasm.  相似文献
