Tremella mesenterica (TM) is a common food and folk medicine widely used in several Asian countries as a tonic for the lungs. In the present study, we compared the effects of extracellular polysaccharides (EPS), intracellular polysaccharides (IPS), and ethanol extract (EE) of Tremella mesenterica on the induction of apoptosis into human lung carcinoma A549 epithelial cells. The EE, but not the EPS or the IPS, almost completely inhibited the growth of A549 cells. The results of Annexin V-FITC/PI staining and flow cytometric analysis indicated that the percentage of Annexin V(+)/PI(-) cells in EE-treated cells increased to 32.8%. The results of further investigation showed a disruption of mitochondrial transmembrane potential (DeltaPsi(m)), the production of reactive oxygen species (ROS), and the activation of caspase-3 protein in EE-treated cells. These findings suggest that EE can decrease cell viability and induce apoptosis in A549 cell lines by activating a mitochondrial pathway.  相似文献

Summary Structural features of haustorial interface parenchyma of the root hemiparasiteOlax phyllanthi are described. Walls contacting host xylem are thickened non-uniformly with polysaccharides, not lignin, and show only a thin protective wall layer when abutting pits in walls of host xylem vessels or tracheids. Lateral walls of interface parenchyma exhibit an expanded middle layer of open fibrillar appearance, sometimes with, but mostly lacking adjoining layers of dense wall material. Free ribosomes and rough endoplasmic reticulum are prominent and occasional wall ingrowths present. Experiments involving transpirational feeding of the apoplast tracers lanthanum nitrate or uranyl acetate to host roots cut below haustorial connections, indicate effective apoplastic transfer from host to parasite root via the haustorium. Deposits of the tracers suggest a major pathway for water flow through host xylem pits, across the thin protective wall layer, and thence into the haustorium via the electronopaque regions of the terminal and lateral walls of the contact parenchyma. Graniferous tracheary elements and walls of parenchyma cells of the body of the haustorium appear to participate in tracer flow as do walls of cortical cells, stele parenchyma and xylem conducting elements of the parasite root, suggesting that both vascular and non-vascular routes are involved in extracytoplasmic transfer of xylem sap from host to parasite. The Casparian strip of the endodermis and the suberin lamella of the exodermis of theOlax root act as barriers to flow within the system.  相似文献

The yellow brain mushroom Tremella mesenterica possesses a wide spectrum of medicinal properties, including immunostimulating, protecting against radiation, antidiabetic, anti-inflammatory, hypocholesterolemic, hepatoprotective, and antiallergic effects. A unique feature of T. mesenterica is that most of the above mentioned medicinal properties depend on glucuronoxylomannan (GXM) contained in fruiting bodies or produced in pure culture conditions. We developed a new strain of T. mesenterica CBS 101939, which grows in submerged culture and offers superior yields of one-cell biomass rich in exocellular heteropolysaccharide GXM. The structure of the GXM was analyzed by NMR spectroscopy and chemical methods. The polysaccharide has a defined repeating unit structure, which is O-acetylated at several points: [structure: see text]. These results differ from previously published structure of Tremella extracellular polysaccharides, where mannan backbone was believed to be randomly glycosylated with xylan chains of different length.  相似文献

对金耳(Tremella aurantialba)的担子果、酵母状分生孢子培养物和菌丝培养物的核糖体DNA内部转录间隔区(ITS)序列进行PCR扩增和测序。结果表明金耳担子果的ITS区PCR产物均为碱基数不同的两条带,片段长度和序列与酵母状分生孢子培养物、菌丝培养物一致。通过对ITS1和ITS2联合进行系统发育分析表明金耳酵母状分生孢子培养物归属于银耳属的金耳,参与组成担子果的寄主菌丝为毛韧革菌(Stereum hirsutum)。结合GenBank中登录的金黄银耳、脑状银耳、橙黄银耳等近似种构建了的系统发育树,结果支持形态学证据,表明金耳是一个独立种。  相似文献

研究了两个居群的金耳Tremella aurantialba以及近似品的rDNAITS区碱基全序列的特征及其差异,首次报道了金耳的ITS和5.8SrDNA完整序列,序列总长度为467~468,长度变异较少。聚类分析表明两个居群金耳亲缘关系非常密切,金耳药材和近似品金黄银耳形成一个稳定的独立分支,近似品黄金银耳形成另一个分支。ITS序列的差异为金耳的鉴别提供了可靠的分子标记,为金耳菌类药材基原入药建立了遗传基础。  相似文献

应用电镜技术对小麦条锈菌吸器母细胞入侵自身菌丝的现象进行了研究,观察发现,在吸器母细胞与寄主细胞和菌丝细胞同时相接触的情况下,入侵栓可在与寄主细胞接触处形成,也可在与菌丝接触处形成;在菌落中心部位,吸器母细胞虽然未与寄主细胞接触,但同样可在与菌丝细胞接触处产生入侵栓;吸器母细胞在与菌丝接触处形成的人侵栓,其超微结构正常,并且可侵入到菌丝细胞壁内,但是未能穿透菌丝细胞壁。本文观察结果表明,小麦条锈菌吸器母细胞和人侵栓形成所需诱导因子可能是物理接触作用,而不涉及到化学作用,并且该病菌与寄主间的识别作用可能发生在入侵栓形成之后。  相似文献

The fine structure of the intercellular hyphae of the obligate parasite Albugo candida infecting radish does not differ markedly from that described previously for cells of Peronospora manshurica. The stalked, capitate haustoria do not contain nuclei and are packed with mitochondria and lomasomes. The fungal plasma membrane and cell wall are continuous from the intercellular hypha throughout the haustorium except that there is no evidence of fungal cell wall around a portion of the haustorial stalk proximal to the haustorial head. Within the vacuolate host mesophyll cell, the haustorium is always surrounded by host plasma membrane and with at least a thin layer of host cytoplasm. The host cell wall invaginates at the point of haustorial penetration to form a short sheath around the region of penetration, but normally there is no host cell wall around the balance of the haustorium. About 1% of the haustoria observed were necrotic, and these were invariably walled-off completely from host cytoplasm by host cell wall. An amorphous, moderately electron-dense encapsulation lies between the haustorium proper and the host plasma membrane and extends into the penetration region between the sheath and the fungal cell wall. Invaded host cells contain more ribosomal-rich ground cytoplasm than uninfected cells. Glandular-like systems of tubules and connecting vesicles are often numerous in host cytoplasm in the vicinity of haustorial heads. These tubules open into the encapsulation, their limiting unit membranes being continuous with the host plasma membrane. We suggest that these represent a secretory mechanism of the host specifically induced by the parasite.  相似文献

The parasitic plant Parentucellia viscosa has been introduced recently to the Japanese floodplain. Because of its parasitic nature and high fecundity, P. viscosa may well become a major plant invader with a significant impact on floodplain ecosystems. Thus, a knowledge of the host selectivity of P. viscosa will not only provide basic biological information, but will also contribute to the understanding of floodplain vegetation conservation. We evaluated the host selectivity by comparing the observed numbers of haustoria with those expected from the relative below‐ground biomass. In addition, we examined the haustorial anatomy to determine whether haustoria are functional, and compared the above‐ground biomass of three functional plant groups (grasses, legumes and nonleguminous forbs) in intact and parasite removal quadrats. We found that haustoria were nonrandomly distributed amongst host species, suggesting host preference for Poaceae and Fabaceae. In addition, haustoria attached to certain forbs did not penetrate into the stele. The above‐ground biomass of grasses and legumes was increased significantly by the removal of P. viscosa, but the biomass of forbs did not change significantly. These results suggest that host preference depresses the performance of Poaceae and Fabaceae, thus affecting the competitive relationships among plants, meaning that P. viscosa may pose a serious threat to indigenous endangered legumes and grasses. © 2012 The Linnean Society of London, Botanical Journal of the Linnean Society, 2012, 170 , 69–78.  相似文献

Anatomical observations were made on the structure and developmentof the primary haustorium of Alectra vogelii. Its developmentinvolves a mutual aggressive growth of both the host and parasitetissues resulting in the formation of a very large and complextuberous organ. One of the host tissues whose growth is stimulatedby parasite infection is the pericycle whose cells divide repeatedlyand grow around and within the parasite haustorial cortex. Fromvarious points of the proliferating host pericycle, roots becomeinitiated and eventually the entire surface of the haustoriumbecomes covered with these roots. We have referred to them as‘haustorial roots’, a term which we have re-examinedand redefined. True xylary connections are established not onlybetween the parasite and the host root but also between theparasite and these ‘haustorial roots’. The uniquedevelopment of primary haustorium and ‘haustorial roots’in A. vogelii is discussed in relation to the development ofprimary haustoria in other root parasites.  相似文献

Summary Ultrastructure of the association between the rust fungus, Melampsora lini, and a compatible variety of flax, Linum usitatissimum, was studied to clarify the structural relationships and interactions at the site of host penetration and at the host-parasite interface. Results of freeze-etching as well as a special section-staining procedure consisting of periodate-chromate-phosphotungstate (PACP) are shown with a host-parasite combination for the first time. The host plasma membrane is invaginated by the fungus and forms a continuous boundary around the fungal haustoria which penetrate the host cells. No morphological continuities are observed linking the protoplasts of host and fungus. With both freeze-etching and the PACP stain, the invaginated portion of the host plasma membrane at the host-parasite interface shows distinctive features that are not characteristic of the non-invaginated portion of the membrane. This localized specialization of host plasma membrane in response to the fungus appears as a significant and consistent feature of the host-parasite interaction. The host plasma membrane is separated from the haustorial wall by an amorphous layer of sheath material which covers the body but not the neck of the haustorium. This sheath provides the environment in which the haustorium exists and functions during the course of the host-parasite association. Occasionally, a collar of wall-like material derived from the host cell forms around the haustorial neck. The collar is continuous with the host wall and is distinct and discontinuous from the haustorial sheath. In fewer than 5% of the infected cells this wall material encases entire haustoria. The fungal wall is structurally specialized around the site of host penetration, and it becomes intimately associated with the host wall where the fungus penetrates into the lumen of the host cell. During penetration, the host and fungal walls appear to be fused so that the interface between them is not clearly delineated. The haustorial wall is continuous, via the haustorial neck, with the wall of the haustorial mother cell which lies outside the host cell. Different staining properties reveal this wall continuum to consist of several well-defined regions having different structure or composition. A ring of fungal wall material midway along the haustorial neck stains densely with lead citrate, but is preferentially etched away by periodic acid. The neck ring denotes a transition in the staining reaction of the fungal wall, from that present in the region of host penetration to that of the wall surrounding the haustorium. The findings demonstrate specialization of the fungal wall in the area of host penetration as well as specialization of the host plasma membrane at the host-parasite interface to a degree not previously realized from ultrastructural information.  相似文献

Morphological and phylogenetic relationships of the worldwide Mediterranean lichen forming fungus, Parmelina quercina , have been studied. Specimens from western Europe, western North America and southern Australia were analysed using molecular data (nuITS rDNA, nuLSU rDNA and mtSSU rDNA) and selected morphological features (upper cortex maculae, scanning electron microscopy examination of the epicortex, ascospores and conidia shape and size, and amphithecial retrorse rhizines). The results conclusively reveal that: (1) there is not one single species but four separate species in the Mediterranean or sub Mediterranean areas of the world. Parmelina quercina and Parmelina carporrhizans (Euroasiatic species), Parmelina coleae sp. nov. (North America) and Parmelina elixia sp. nov. (Australia); (2) largely debated P. carporrhizans is not a synonym of P. quercina but supported as a valid species circumscribed to Macaronesic relict sites; (3) the geographical isolation of the Australian population is correlated with a large genetic distance; (4) morphological characters (ascospores and conidial variability and thallus epicortex) correlate with the phylogenetic hypothesis; (5) the new or revalidated species within Parmelina quercina are not cryptic species but morphologically recognizable taxa.  © 2007 The Linnean Society of London, Biological Journal of the Linnean Society , 2007, 91 , 455–467.  相似文献

The external features of the subterranean organs of coastal Californian Pholisma depressum Greene are described. A dimorphic root system exists. Long, stout roots (pilot roots) serve for vegetative propagation in reaching other host roots. Short, unbranching, more slender roots originating from the pilot roots are haustorial in function. Shoots seem to arise only on pilot roots. Host attachment is achieved through the direct transformation of the apical meristem of haustorial roots into the haustorial organ. The basal part of the shoot is also able to produce roots when near host roots, and may form lateral branch shoots either directly or after new haustorial connections are formed. Both types of roots arise endogenously. The invaded host root undergoes considerable hypertrophy. The intrusive organ itself forms a massive but irregular body. More than one haustorial root may enter even a small host root. Self parasitism may occur but is not common.  相似文献

In a study of host-parasite interrelationship and the mechanism of parasitization, ribonuclease activity was determined in Cuscuta reflexa Roxb, and in infected and control host plants of Lantana camara L. In the haustoria-bearing region of Cuscuta , the concave half of the vine bearing the haustorial site, always showed significantly higher RNase activity than the convex half, irrespective of the differing enzyme activity of the parasite tissue growing on different hosts — Brassica campestris L., Helianthus animus L., Lantana camara L., Medicago saliva L. and Solatium nigrum L. The uninfected host branch of L. camara showed the maximum specific RNase activity in the apical region which decreased toward the base, while the infected host branch showed minimum specific RNase activity in the apical region, gradually increasing towards the infected region.  相似文献

Tremella mesenterica (TM), a yellow jelly mushroom, has been traditionally used as tonic food to improve body condition in Chinese society for a long time. We have previously demonstrated that TM reduced in vitro hCG-treated steroidogenesis in MA-10 mouse Leydig tumor cells without any toxicity effect. In the present study, the mechanism how TM suppressed hCG-treated steroidogenesis in MA-10 cells was investigated. MA-10 cells were treated with vehicle, human chorionic gonadotropin (hCG, 50 ng/ml), or different reagents with or without TM to clarify the effects. TM significantly suppressed progesterone production with the presences of forskolin (10 and 100 microM) or dbcAMP (0.5 and 1mM), respectively, in MA-10 cells (p<0.05), which indicated that TM suppressed steroidogenesis after PKA activation along the signal pathway. Beyond our expectation, TM induced the expression of steroidogenic acute regulatory (StAR) protein with or without hCG treatments. However, TM profoundly decreased P450 side chain cleavage (P450scc) and 3beta-hydroxysteroid dehydrogenase (3beta-HSD) enzyme activities without any influences on the expression of both enzymes. These inhibitions on steroidogenic enzyme activities might counteract the stimulation of StAR protein expression. In conclusion, results suggest that TM suppressed hCG-treated steroidogenesis in MA-10 cells by inhibiting PKA signal pathway and steroidogenic enzyme activities.  相似文献

以马铃薯晚疫病水平抗性品种LBr-12和感病品种费乌瑞它为材料,采用普通光学和电子显微镜技术,系统研究了马铃薯与晚疫病菌(致病疫霉)互作的组织细胞学反应特征。观察结果显示:(1)接种后,水平抗性材料LBr-12出现过敏反应,病菌被限制在侵染点的几个细胞中,菌丝产生较少的分支和吸器。(2)感病品种费乌瑞它被侵染细胞呈蔓延趋势,菌丝产生较多的分支和吸器。(3)电镜观察发现,抗病品种上病菌的胞间菌丝、吸器母细胞、吸器在细胞和亚细胞水平均发生了一系列异常变化,包括原生质的电子致密度加深、液泡增多变大、菌丝细胞壁不规则增厚、细胞器排列紊乱及解体、吸器母细胞及吸器形态异常、病菌最终畸形坏死,同时发现抗病品种受病菌侵染时可迅速产生结构防卫反应,形成的细胞壁沉积物使胞壁极度增厚或细胞膜上产生乳突状结构。  相似文献

This paper deals with the haustorium development of parasitic plant (Cassytha filiformis L.) parasitized on purple willow (Salix purpurea L.).The polarity occurred in the portion near the host; and the cushion-shaped haustorial plate formed, and then the haustorial primordium initiated in the cortex. Finally, the haustoria reached the cortex of pith of the host, penetrating through its own cortex and epidermis. Tracheary elements were differentiated from the base of the haustoria and the sieve elements were not observed in the haustoria. Histochemical studies revealed that there were starch grains in normal stem cortex. The starch grains were increased in the portion near the axis after twisting on host. After the haustorial plate was formed, the starch grains were richly accumulated in the central group of cells, which were followed by the haustorial development; The starch disappeared in the meristem, in which protein stained deeply; The dynamic change of protein turned oK to be contrary to the tendency of starch accumulation. The structure and parasitic mechanism and the dynamic change of starch and protein are discussed.  相似文献

Plants employ multiple cell‐autonomous defense mechanisms to impede pathogenesis of microbial intruders. Previously we identified an exocytosis defense mechanism in Arabidopsis against pathogenic powdery mildew fungi. This pre‐invasive defense mechanism depends on the formation of ternary protein complexes consisting of the plasma membrane‐localized PEN1 syntaxin, the adaptor protein SNAP33 and closely sequence‐related vesicle‐resident VAMP721 or VAMP722 proteins. The Arabidopsis thaliana resistance to powdery mildew 8.2 protein (RPW8.2) confers disease resistance against powdery mildews upon fungal entry into host cells and is specifically targeted to the extrahaustorial membrane (EHM), which envelops the haustorial complex of the fungus. However, the secretory machinery involved in trafficking RPW8.2 to the EHM is unknown. Here we report that RPW8.2 is transiently located on VAMP721/722 vesicles, and later incorporated into the EHM of mature haustoria. Resistance activity of RPW8.2 against the powdery mildew Golovinomyces orontii is greatly diminished in the absence of VAMP721 but only slightly so in the absence of VAMP722. Consistent with this result, trafficking of RPW8.2 to the EHM is delayed in the absence of VAMP721. These findings implicate VAMP721/722 vesicles as key components of the secretory machinery for carrying RPW8.2 to the plant–fungal interface. Quantitative fluorescence recovery after photobleaching suggests that vesicle‐mediated trafficking of RPW8.2–yellow fluorescent protein (YFP) to the EHM occurs transiently during early haustorial development and that lateral diffusion of RPW8.2–YFP within the EHM exceeds vesicle‐mediated replenishment of RPW8.2–YFP in mature haustoria. Our findings imply the engagement of VAMP721/722 in a bifurcated trafficking pathway for pre‐invasive defense at the cell periphery and post‐invasive defense at the EHM.  相似文献

In parasitic angiosperms the haustorium, an organ specialized for attachment and penetration of host tissue, functions in the transport of water and nutrients from the host to the parasite. In Agalinis purpurea (L.) Raf. (Scrophulariaceae) these organs are initiated laterally along its roots, opposite a primary xylem pole. Analyses of haustoria distribution and cellular root profiles show that the portion of the root which is most sensitive to haustorial elicitor molecules is the area distal to the zone of elongation and near the root meristem. Sectioned material supports this finding and, further, indicates that the cells which are the first to respond to haustorial elicitors are located in the inner cortex. Haustoria develop rapidly in response to a host root or to isolated chemical elicitors (xenognosins) normally contained in host root exudate. By 6 hr, vacuolation and radial cellular enlargement are observed in the cortex, and a lateral swelling along the root is visible. By 12 hr, cells of the epidermis divide anticlinally to establish a group of densely cytoplasmic cells at the apex of the haustorial swelling. Accompanying these divisions is the differentiation of specialized hair cells which elongate from epidermal cells flanking the presumptive haustorial apex. Next, the internal, radially enlarged cortical cells divide periclinally. Periclinal divisions are subsequently initiated in the pericycle as early as 18 hr post-induction. Cellular division and enlargement continue so that by 24–36 hr a mature pre-contact haustorium is formed. There is a reduction in root elongation concomitant with haustorial initiation. Depending upon the number of haustoria produced, elongation typically returns to the preinduction level within 2 or 3 days.  相似文献

Cellular fatty acids of Erwinia species in the Amylovora group ( E. amylovora, E. nigrifluens, E. quercina, E. rubrifaciens, E. salicis and E. tracheiphila ), grown on King's medium B agar for 1,3, and 6 days at 28°C, were analyzed by gas-liquid chromatography, grouped by chemical class, and compared in order to differentiate the species. For the group in general, the average percentage in 1-day-old cells ranged 36.4–41.5% for the saturated straight-chain even-carbon fatty acids, (Class A), 0.3–0.7% for saturated straight-chain odd-carbons (Class B), 43.4–53.3% for unsaturated acids (Class C), 4.5–7.1% for hydroxy-substituted acids (Class D), 0.2–2.0% for branched-chain acids (Class E), and 0–7.5% for cyclopropane fatty acids (Class F). Species could be differentiated in 4 sequential steps based on: 1) percentage of the 12:0 fatty acid ( E. amylovora >3.6%, E. quercina (1%);2) sum of Classes E + F ( E. tracheiphila (0.3%);3) ratio Class A/(E + F) ( E. salicis <8); and percentage of 18:1 ( E. rubrifaciens >14%, E. nigrifluens < 14%). Physiological age significantly affected relative percentages of Classes B, C and F in E. amylovora and E. salicis ; of Classes A, C and F in E. quercina ; of Class A in E. tracheiphila ; and of Class C in E. nigrifluens.  相似文献
