Stem segments taken from Avena sativa plants grown at 10°, 20° or 30° varied in their phospholipid composition depending on the growth temperature; as temperature was lowered, there was a shift towards a greater proportion of unsaturated fatty acids. A significant increase was observed in the concentration of linolenic acid (18:3) as growth temperature was lowered. Although prolonged treatment of oat plants with GA3 produced marked changes in phospholipid composition of stem segments, these changes did not always accompany the GA3-induced growth response of segments. Treatment of stem segments with GA3 for only 20 hr produced a significant growth response with little or no effect on phospholipid composition over this time. The data support the hypothesis that GA3-induced growth in Avena stem segments can occur without a concomitant change in phospholipid composition.  相似文献

Batch cultures (8–32 l.) of Chlorella vulgaris and Scenedesmus obliquus and of Anacystis nidulans and Microcystis aeruginosa were grown in media containing 0.001 % KNO3 and at several stages in growth sampled for biomass, total protein, chlorophylls, lipids and fatty acids. With increasing time and decreasing nitrogen concentrations, the biomass of all of the algae increased, whereas the total protein and chlorophyll content dropped. Green and blue-green algae, however, behaved differently in their lipid metabolism. In the green algae the total lipid and fatty acid content as well as the composition of these compounds changed considerably during one growth phase and was dependent on the nitrogen concentration in the media at any given day of growth. More specifically, during the initial stages of growth the green algae produced larger amounts of polar lipids and polyunsaturated C16 and C18 fatty acids. Towards the end of growth, however, these patterns changed in that the main lipids of the green algae were neutral with mainly saturated fatty acids (mostly 18:1 and 16:0). Such changes did not occur in the blue-green algae. These differences between prokaryotic and eukaryotic algae can possibly be explained by the ‘endosymbiont theory’.  相似文献

Of the three morphological subunits of Avena sativa stem segments (node, leaf-sheath and internode) examined, internodes constituted the richest source of phospholipids and sterols, yielding almost double the concentration of lipid found in the leaf-sheath. The phospholipid compositions of nodes and internodes were similar, comprising mostly phosphatidylcholine (PC) and phosphatidylethanolamine (PE), with linoleic, linolenic and palmitic acids as the predominant component fatty acids. Leaf-sheath tissue contained mainly PE, with equally high amounts of palmitic, linoleic and linolenic acids. β-Sitosterol and cholesterol were the major 4-desmethylsterols of the internode, while β-sitosterol was predominant in the node and leaf-sheath tissues. The growth temperature of segments prior to isolation produced its major effect on the concentration of stigmasterol, which decreased markedly with temperature. The sitosterol/stigmasterol ratio increased significantly as temperature decreased. Stem segments isolated from plants treated with gibberellic acid (GA3) for 3 weeks, showed a significant reduction in the amounts of 4-desmethyl sterols on a dry wt basis when compared with control segments. However, when stem segments were incubated with GA3 for 20 hr, no change in 4-desmethylsterol composition or concentration was observed, even though significant growth in response to GA3 occurred.  相似文献

The cyanobacteriumSynechocystis PCC 6803 was grown photoautotrophically in an inorganic medium at constant growth temperatures of 20, 38 (control) or 43°C for 9 h. The up and down-shift of cultivation temperature decreased the growth as measured by culture absorbance and chlorophylla content. However, high temperature slightly increased the oxygen evolution while temperature lower than control inhibited oxygen evolution during the whole incubation period. The protein synthesis studied by14C-labeled protein declined under low temperature by about 50%. The fatty acid pattern is characterized as lacking in C20/C22 acids but containing large amounts of C16 and C18 polyunsaturated fatty acids, 16:2 and 18:3 in particular. The lower temperature increased the percentage of monounsaturated fatty acids while higher temperature increased the saturated fatty acid content in total lipids and lipid classes studied.  相似文献

The total lipid and fatty acid content ofSpirulina platensis UTEX 1928 was 7.2 and 2.2% respectively of cellular dry weight under controlled conditions supporting high growth rates. With increases in irradiance from 170 to 870 μmol photon m?2 s?1, growth rate increased, total lipid decreased, and fatty acid composition was unaffected. At 1411 μmol photon m?2 s?1, total lipid increased slightly and percent composition of the fatty acid gamma linolenic acid increased. Growth and total lipid content ofS. platensis were affected by changes in growth temperature from 25 to 38 °C. With increased growth rate, total lipid content increased. This suggests that the storage of carbon increases at temperatures supporting high growth rates. The degree of saturation increased with temperature. Although the percent composition of gamma linolenic acid was higher at lower growth temperature, production was still primarily a function of growth rate. The effect of temperature on fatty acid content and degree of saturation was of secondary importance. Nitrogen starvation increased total lipid content but decreased fatty acid content as a percentage of dry weight; composition of the fatty acids was unaffected. N-starvation appeared to suspend synthesis of long chain fatty acids inS. platensis, suggesting that some other compound stores fixed carbon when nitrogen is limiting. It was concluded that fatty acid production inS. platensis is maximized by optimizing culture conditions for growth.  相似文献

Comparative study about the salt-induced oxidative stress and lipid composition has been realised in primary root tissues for two varieties of maize (Zea mays L.) in order to evaluate their responses to salt stress. The root growth, root water content (WC), hydrogen peroxide (H2O2) generation, lipid peroxidation, membrane stability index and the changes in the profile of fatty acids composition were investigated. Salinity impacts in term of root growth, water content, H2O2 generation, lipid peroxidation and membrane destabilisation were more pronounced in primary roots of Aristo than in those of Arper indicating more sensitivity of the first variety. It was confirmed by gas chromatography that the composition of fatty acids in roots of both varieties was constituted mainly by 16:0 and 18:0 as major saturated fatty acids and 18:1ω9, 18:2ω6 and 18:3ω3 as major unsaturated fatty acids. Total lipid extracts from the roots of both varieties showed that the lipid saturation level increased under salt stress, notwithstanding the increased proportion of polyunsaturated fatty acids. The changes in lipid saturation being predominantly due to decreases in oleic acid (18:1ω9) and increases in palmitic acid (16:0). However, Arper root extracts contained a lower proportion of saturated lipids than Aristo. The enhanced proportion of highly polyunsaturated fatty acids especially linolenic and eicosapentaenoic acids was considered to be the characteristic of the relatively salt tolerance in Arper roots.  相似文献

Chlorella kessleri cultivated in a deep tank contained 4.8% of non-polar lipid; 51% of this fraction represents saturated fatty acids, 7% unsaturated fatty acids. Our investigation of the fatty acids profile demonstrated even- and odd-numbered saturated and unsaturated fatty acids ranging from C12 to C20. Unlike in otherChlorella species, stearic acid was the dominant fatty acid found. Also shown was an elevated C16:0 fatty acid content and a reduced level of unsaturated fatty acids.  相似文献

Organisms adjust the order, or fluidity, of their cellular membranes in response to changes in their physiochemical environment by adjusting the lipid composition of their membranes. We investigated membrane fluidity using the phospholipid, fatty acid and cholesterol content of red blood cells (RBCs) from multiple sclerosis (MS) patients and correlated this with C-reactive protein (CRP) as well as with the severity of neurological outcome as measured by the Kurtzke Expanded Disability Status Scale (EDSS) and its Functional System Scores. The study group consisted of 31 patients with MS and 30 healthy control subjects. Phospholipids were determined using a colorimetric assay, fatty acids by gas chromatography, cholesterol by an enzymatic assay and CRP by a Beckman nephelometer. Cell membrane fluidity was calculated according to previously established formulae. RBC membrane fluidity as measured by the saturated to polyunsaturated fatty acid ratio was higher in patients than in controls (P = 0.04). The phosphatidylethanolamine saturated to polyunsaturated fatty acid ratio showed highly significant positive correlations with the EDSS and CRP < 5 μg/ml. CRP showed significant inverse correlations with the saturated nature but positive correlations with the ordered-crystalline-phase to liquid-crystalline-phase lipid ratio. In this study we show that membrane fluidity as measured by the relationship between membrane fatty acids, phospholipids and cholesterol is closely interrelated with inflammation and disease outcome in patients with MS. In conclusion, our findings suggest that the membrane lipid composition of patients with MS and, consequently, membrane fluidity are altered, which seems to be influenced by the inflammatory status.  相似文献

Clostridium pasteurianum total cellular saturated fatty acids increased through its growth cycle from 81% to 91% but varied significantly in the composition under nitrogen- and non-nitrogen-fixing conditions. During ammonia-assimilating growth, palmitic acid decreased from 67.7% to 43.5% by late log while marked increases in shorter chain saturated fatty acids (C15:0 and below) and a long chain saturated C22:0 occured. In contrast, under N2-fixing growth conditions, palmitic acid increased from 45.5% to 84.3% by late log, representing nearly the total amound of saturated fatty acids found inC. pasteurianum. The total cellular lipid concentration decreased as the culture aged. irrespective of the nitrogen sources; however, the phospholipid concentration increased significantly during N2-fixing growth as compared with a 50% decrease during ammonia-assimilating conditions. The implication of these differences and possible role of palmitic acid and phospholipids inC. pasteurianum nitrogen fixation process are discussed.  相似文献

Fatty acids of acyl steryl glycoside (acyl SG) of different plants producing both green and photosynthetically inactive tissue have been analyzed. The major components are in all cases 16:0, 18:2 and 18:3 acids. The fatty acid composition of acyl SG of green parts is very similar to that of etiolated, pale or storage tissue of the same plant. Generally the degree of saturation of acyl SG is higher than that of the corresponding total lipid. Acyl SG tends to be more saturated in green parts than in colorless tissues of the same plant. Conversely, total lipid of green tissue containing large amounts of galactolipids and 18:3 acid is much less saturated than that of photosynthetically inactive tissue. Though containing smaller amounts of 18:3, and in some cases unsaturated C16 acids, acyl SG does not reflect the drastic increase of these acids in the total lipid of green tissue. It is concluded that fatty acids of acyl SG originate mainly from an acyl donor other than chloroplast galactolipids.  相似文献

The composition of tissue and membrane fatty acids in ectothermic vertebrates is influenced by both temperature acclimation and diets. If such change in body lipid composition and thermal physiology were linked, a diet-induced change in body lipid composition should result in a change in thermal physiology. We therefore investigated whether the selected body temperature of the agamid lizardAmphibolurus nuchalis (body mass 20 g) is influenced by the lipid composition of dietary fatty acids and whether diet-induced changes in thermal physiology are correlated with changes in body lipid composition. The selected body temperature in two groups of lizards was indistinguishable before dietary treatments. The selected body temperature in lizards after 3 weeks on a diet rich in saturated fatty acids rose by 2.1 °C (photophase) and 3.3 °C (scotophase), whereas the body temperature of lizards on a diet rich in unsaturated fatty acids fell by 1.5 °C (photophase) and 2.0 °C (scotophase). Significant diet-induced differences were observed in the fatty acid composition of depot fat, liver and muscle. The lipids in callus cultures of Hydnocarpus anthelminthica were studied after 60, 160 and 460 days of growth. In each of the cultures the lipid classes usually found in plant tissue cultures were detected. With increasing age of the cultures the total lipid content as well as the proportions of triglycerides decreased. The major constituent fatty acids of the total lipids were palmitic and linoleic acids. Small amounts of cyclopentenyl fatty acids were also present. The proportions of saturated straight-chain fatty acids increased with the age of the cultures whereas the proportions of monounsaturated straight-chain fatty acids decreased. Only small changes were observed with polyunsaturated fatty acids. The content of cyclopentenyl fatty acids rose with the age of the cultures. The monounsaturated straight-chain fatty acids consisted of mixtures of isomers whose composition changed with the age of the cultures. In contrast, the polyunsaturated straight-chain fatty acids belonged exclusively to the Δ9 series, regardless of the age of the cultures.  相似文献

The fatty acid content and composition of the house cricket Acheta domesticus have been investigated in entire insects at different developmental stages and in selected organs of male and female adults. We have also determined the fatty acid composition of the various lipid classes within extracts of the organs of adult female insects. Fatty acids were analysed by capillary gas chromatography or mass spectrometry as their methyl esters (FAMEs) after direct transesterification of insect material or separated lipid classes.The major esterified fatty acids in all extracts were palmitate (C16:0), stearate (C18:0), oleate (C18:1) and linoleate (C18:2). Levels of esterified fatty acid varied considerably between organs but the fatty acid compositions showed only small variations. The levels of polyunsaturated fatty acids of the C18 series were considerably higher in phospholipid fractions than in other lipid classes. Triacylglycerols formed the major lipid class in ovaries, fat-body and newly-laid eggs, whereas diacylglycerols and phospholipid predominate in the haemolymph. Triacylglycerols, phospholipids, diacylglycerols and free fatty acids were all found in significant amounts in the gut tissue.  相似文献

The objective was to examine the effect of polyunsaturated fatty acid type (plant vs fish oil-derived n-3, compared to n-6 fatty acids in the presence of constant proportions of saturated, monounsaturated and polyunsaturated fatty acids) on obesity, insulin resistance and tissue fatty acid composition in genetically obese rats. Six-week-old fa/fa and lean Zucker rats were fed with a 10% (w/w) mixed fat diet containing predominantly flax-seed, menhaden or safflower oils for 9 weeks. There was no effect of dietary lipid on obesity, oral glucose tolerance (except t=60 min insulin), pancreatic function or molecular markers related to insulin, glucose and lipid metabolism, despite increased n-3 fatty acids in muscle and adipose tissue. The menhaden oil diet reduced fasting serum free fatty acids in both fa/fa and lean rats. These data suggest that n-3 composition does not alter obesity and insulin resistance in the fa/fa Zucker rat model when dietary lipid classes are balanced.  相似文献

Seven day old seedlings of Echinochloa crus-galli var. oryzicola (Vasing) had a higher total lipid content when germinated under N2 than in air, although ungerminated seeds contained more lipid than either seedling. The triacylglycerol pool was not depleted under anaerobiosis as it was in air and only air-grown seedlings showed a net increase in free fatty acids and polar lipids. Concentrations of most of the individual acids of the total fatty acid profile declined during germination in air and in the free acid and polar lipid fractions of these seedlings the relative proportion of polyunsaturated fatty acids increased. Compared to air-grown seedlings, ungerminated seeds and N2-grown seedlings had a similar qualitative and quantitative lipid composition. Our results show that mobilization of storage lipids was apparently severely inhibited under anoxia. The importance of lipid metabolism to the germination and growth of Echinochloa during anoxia is discussed in terms of maintaining membrane integrity and serving (indirectly) to reoxidize pyridine nucleotides.  相似文献

Increasing the content of polyunsaturated fat in the human diet is a priority for reducing cardiovascular disease and cancer risks. Beef has the potential to contribute to the polyunsaturated fat content in the human diet; however, ruminants cannot synthesise many long-chain fatty acids de novo; they require dietary supplementation. The objectives of the current study were to evaluate (i) the effect of a partially rumen protected n-3 long-chain polyunsaturated fatty acid (LC-PUFA) dietary supplement on the fatty acid composition of muscle (Longissimus dorsi), adipose and liver tissues of beef heifers and (ii) the usefulness of blood plasma as a predictor of tissue concentrations of specific fatty acids. Charolais crossbred heifers (n = 20) were assigned to one of two isolipid dietary treatments namely palmitic acid (control) or an n-3 LC-PUFA supplement for a 91-day period. Blood plasma and adipose tissue samples were taken to determine the temporal effect of these diets on fatty acid composition (days 0, 10, 35 and 91), while liver and muscle samples were taken following slaughter. Dietary lipid source did not influence animal growth rate or body condition score. At day 91, the percentage differences between control and n-3 LC-PUFA heifers in concentrations of eicosapentaenoic acid were +61, +176 and +133 % in liver, muscle and adipose, respectively. For docosahexaenoic acid, at the same time point, the percentage differences were +57, +73 and +138 % for liver, muscle and adipose, respectively. Medium-to-strong positive correlation coefficients were evident for liver and plasma fatty acids, in particular, there were positive relationships with concentrations of total saturated fatty acid (SFA), total n-6 PUFA and total n-3 PUFA. This trend also extended to both the ratio of PUFA to SFA (slope (β1) = 0.56 ± 0.167, intercept (β0) = 0.56, R2 = 0.61, P < 0.05) and the ratio of n-6 to n-3 PUFA (β1 = 0.15 ± 0.054, β0 = 0.24, R2 = 0.52, P < 0.05). A strong correlation was also detected in the ratio of n-6 to n-3 in plasma and muscle tissue of heifers fed the n-3 LC-PUFA diet (β1 = 0.53 ± 0.089, β0 = −0.31, R2 = 0.83, P < 0.001). The results of this study show that the n-3 LC-PUFA can be readily increased through targeted supplementation and that plasma concentrations of n-3 LC-PUFA are useful predictors of their concentrations in a number of economically important tissues.  相似文献

The fatty acid composition of the whole cell and cell wall ofAerobacter aerogenes was studied employing column and gas-chromatographic technique. The cell wall contained a greater percentage of total lipid, complex lipid, and free fatty acids compared to the whole cell. Palmitic acid and oleic acid were the most abundant fatty acids found in the free fatty acid and complex lipid fractions. A saturated C17 fatty acid and small quantities of a branched C16 and iso and anteiso C12 fatty acids were detected. The glyceride fractions of the whole cell and cell wall contained very few fatty acids.  相似文献

The fatty acid patterns of Euonymus europaeus callus cultures and cell suspension cultures were analysed at the beginning of stationary growth phase and compared with those from the respective differentiated tissues. The lipid and fatty acid patterns in cell cultures differed remarkably from those in the tissues of the mother plant. No glycerol triacetate was detected in the callus cultures derived from differentiated tissues whereas in seeds this lipid compound amounts to 29%. In addition to fatty acids normally occurring in differentiated tissues, lipids in cultured cells also contained short-chain (C12–C14) as well as very long-chain fatty acids (C20–C24). In tissue culture cells the major fatty acids were found to be saturated, whereas in the mother cells unsaturated fatty acids were predominant. Palmitic acid is the most abundant fatty acid in most of the cultures. Lauric, myristic and palmitic acid amount to 50% in lipids of cell suspension cultures.  相似文献

Microalgae biomass can be a feasible source of ω‐3 fatty acids due to its stable and reliable composition. In the present study, the Crypthecodinium cohnii growth and docosahexaenoic acid (DHA, 22:6ω3) production in a 100 L glucose‐fed batch fermentation was evaluated. The lipid compounds were extracted by supercritical carbon dioxide (SC‐CO2) from C. cohnii CCMP 316 biomas, was and their fatty acid composition was analysed. Supercritical fluid extraction runs were performed at temperatures of 313 and 323 K and pressures of 20.0, 25.0 and 30.0 MPa. The optimum extraction conditions were found to be 30.0 MPa and 323 K. Under those conditions, almost 50% of the total oil contained in the raw material was extracted after 3 h and the DHA composition attained 72% w/w of total fatty acids. The high DHA percentage of total fatty acids obtained by SC‐CO2 suggested that this extraction method may be suitable for the production of C. cohnii value added products directed towards pharmaceutical purposes. Furthermore, the fatty acid composition of the remaining lipid fraction from the residual biomass with lower content in polyunsaturated fatty acids could be adequate for further uses as feedstock for biodiesel, contributing to the economy of the overall process suggesting an integrated biorefinery approach.  相似文献

The lipid fraction of the green alga Botryococcuscultured in a batch mode was found to contain polar lipids (more than 50% of the total lipids), di- and triacylglycerols, sterols and their esters, free fatty acids, and hydrocarbons. In aging culture, the content of polar lipids somewhat decreased and that of triacylglycerols increased by more than four times. The content of hydrocarbons in the algal biomass did not exceed 0.9% and depended little on the culture age. Intracellular lipids contained saturated and unsaturated (mono-, di-, and trienoic) fatty acids. The maximum content of C16 : 3and -C18 : 3fatty acids (up to 35% of the total fatty acids) was detected in the phase of active growth. The extracellular and intracellular lipids of the alga differed in the proportion of particular lipids and in the fatty acid pattern.  相似文献
