1. Immune priming refers to improved protection of the host after a second encounter with the same parasite or pathogen. This phenomenon is similar to that of adaptive immunity in vertebrates. 2. There is evidence to suggest that this improved protection can be species/strain‐specific and can protect organisms for a lifetime. These two attributes, along with a biphasic immune response, are essential characteristics of immune priming and form the basis for the effectiveness of resistance to parasites and pathogens. 3. This paper considers the effect of immune priming within and across generations, the influence of a heterologous challenge during immune priming and the importance of testing the immune response with natural pathogens. 4. The analysis presented takes into account the multifaceted nature of the invertebrate immune response. The lack of evidence suggesting that the bacterial microbiome plays a complementary role in the immune priming outcome is discussed. 5. Finally, the cost of immune priming is explored. This is a poorly investigated issue, which could help to explain why there is a paucity of evidence in support of immune priming.  相似文献

利用多种免疫学方法研究了中药升血汤对小鼠免疫功能的影响。结果证明,升血汤对正常小鼠的细胞免疫功能和体液免疫功能确有明显的促进作用。表现为,非特异地(多克隆)增强T、B淋巴细胞对丝裂原的增殖反应;特异地增强T细胞对异型抗原的MLR、DTH反应;特异地增强小鼠对SRBC的抗体反应;明显增强小鼠脾细胞产生ILD2的能力。  相似文献

Genetic alterations are responsible for the development of cancer in ductal cells of the pancreas. These genetic changes result in abnormal molecular expression of proteins that are involved in cell proliferation, cell cycle control and adhesion. Some of the genetic mutations result in aberrant proteins that can be recognized as novel or foreign by cells of innate and adaptive immune systems. These are appropriate targets for therapeutic intervention which may involve immunobiologic approaches. These approaches may be less effective because of immune escape mechanisms developed by tumor cells within the microenvironment of the tumor mass. Immunobiotherapy intervention of pancreas cancer must circumvent these obstacles and integrate effective immunotherapy with molecularly targeted approaches to pancreas cancer intervention.  相似文献

肝特殊的解剖结构及生理特征使其成为暴露肠源性抗原的主要器官。由于肝具有独特的固有免疫系统,在正常情况下,肝分布多种致耐受的抗原提呈细胞,对持续性表达或递呈于肝的肠源性抗原物质,诱发针对该抗原的系统性免疫耐受,避免肝受到不必要的免疫损伤。当炎症发生及肝脏固有免疫系统活化时,则通过免疫效应细胞及免疫效应因子对肠源性病原体发挥强烈地免疫应答以控制感染。该过程形成机制的研究对肝功能的理解及肝性疾病的预防与治疗至关重要。本文就肝固有免疫系统对肠源性感染的免疫应答与免疫耐受形成机制作一综述。  相似文献

O1/O139霍乱口服疫苗是一种新型疫苗。本文通过2种不同途径免疫动物,检测血清抗菌抗体和肠道分泌性IgA抗体以及保护力试验,并探讨了它们之间的关系。通过家兔肠袢试验和细胞四甲基偶氮唑盐(MTT)试验检测免疫后抗血清的被动保护作用。试验提示:在霍乱的保护性免疫应答中,除粘膜免疫可能占主导作用外,血清抗菌抗体也发挥了一定的作用。  相似文献

在长期进化过程中,昆虫形成了强大的天然免疫防御系统,即体液免疫和细胞免疫。体液免疫主要包括Toll、IMD和JAK/STAT 3条信号通路,通过信号转导及免疫途径调控免疫相关基因的表达,诱导产生抗菌肽和其他效应分子。细胞免疫由血细胞介导,主要完成对病原物的包裹、吞噬和集结等。近年来,昆虫基因组学快速发展,通过生物信息学等方法从昆虫基因组数据中已鉴定到大量免疫相关基因,对这些基因的研究加深了人们对昆虫天然免疫分子机制的认识和理解。根据基因功能,免疫相关基因分为识别、信号转导、调制器、效应分子、黑化反应、RNA干扰和其他基因等7类,这些基因通过互作来调控体液免疫和细胞免疫。本文对昆虫免疫相关基因的分类、功能及家族进化等方面的研究成果进行总结,并对今后昆虫免疫的研究重点进行了展望,以期为昆虫免疫分子机制的研究及开发新的害虫防治策略提供依据。  相似文献

The foetus can be regarded as a half-allograft implanted into the maternal body. In a successful pregnancy, the mother does not reject the foetus because of the immune tolerance mechanism at the maternal-foetal interface. The innate immune cells are a large part of the decidual leukocytes contributing significantly to a successful pregnancy. Although the contributions have been recognized, their role in human pregnancy has not been completely elucidated. Additionally, the accumulated evidence demonstrates that the immune checkpoint molecules expressed on the immune cells are co-inhibitory receptors regulating their activation and biological function. Therefore, it is critical to understand the immune microenvironment and explore the function of the innate immune cells during pregnancy. This review summarizes the classic immune checkpoints such as PD-1, CTLA-4 and some novel molecules recently identified, including TIM-3, CD200, TIGIT and the Siglecs family on the decidual and peripheral innate immune cells during pregnancy. Furthermore, it emphasizes the role of the immune checkpoint molecules in pregnancy-associated complications and reproductive immunotherapy.  相似文献

近二十多年,全球范围内先后爆发了由严重急性呼吸综合征冠状病毒(severe acute respiratory syndrome coronavirus,SARS-CoV)、中东呼吸综合征冠状病毒(middle east respiratory syndrome coronavirus,MERS-CoV)和严重急性呼吸综合征冠状病毒2(severe acute respiratory syndrome coronavirus 2,SARS-CoV-2)3种高致病性冠状病毒导致的疫情。这3种高致病性冠状病毒感染通常伴随着免疫系统功能失调,临床表现有淋巴细胞减少症、细胞因子风暴、急性呼吸系统窘迫综合征,甚至多器官衰竭而导致死亡。揭示高致病性冠状病毒在免疫应答中的作用机制,对于预防与控制冠状病毒感染具有重要意义。本文总结了SARS-CoV、MRES-CoV和SARS-CoV-2的进入机制和受体特征、固有免疫应答和适应性免疫应答失调方面的研究进展,强调了高致病性冠状病毒与宿主免疫应答之间的复杂相互作用,以期为防治冠状病毒感染提供参考。  相似文献

Ferretti G., Gabriele F., Palmas C. and Wakelin D. 1984. Interactions between Trichinella spiralis and Hymenolepis nana in the intestine of the mouse. International Journal for Parasitology14: 29–33. The rejection of the intestinal phase of Trichinella spiralis in outbred CD1 mice is associated with an inflammatory response. The effects of this inflammation were studied in mice concurrently infected with an unrelated parasite, Hymenolepis nana, which stimulates a slight inflammation but is also immunologically rejected by this strain of mice. Moreover though Cd(In)1 mice seem to be ‘slow responders’, in concurrent infections with T. spiralis and H. nana there was a marked effect upon the cesode survival. At the highest level of T. spiralis infection, H. nana failed to establish at all. On the other hand, if 300 T. spiralis were administered 10 weeks earlier, H. nana become established in all mice and the distribution of parasites suggests that immunodepression is involved. An accelerated loss of the nematode, when H. nana was present, was also demonstrated. There was no evidence of any reciprocal or specific cross-immunity between the two parasites.  相似文献

Parasites represent a major threat to all organisms which has led to the evolution of an array of complex and effective defence mechanisms. Common to both vertebrates and invertebrates are innate immune mechanisms that can be either constitutively expressed or induced on exposure to infection. In nature, we find that a combination of both induced and constitutive responses are employed by vertebrates, invertebrates and, to an extent, plants when they are exposed to a parasite. Here we use a simple within-host model motivated by the insect immune system, consisting of both constitutive and induced responses, to address the question of why both types of response are maintained so ubiquitously. Generally, induced responses are thought to be advantageous because they are only used when required but are too costly to maintain constantly, while constitutive responses are advantageous because they are always ready to act. However, using a simple cost function but with no a priori assumptions about relative costs, we show that variability in parasite growth rates selects for a strategy that combines both constitutive and induced defences. Differential costs are therefore not necessary to explain the adoption of both forms of defence. Clearly, hosts are likely to be challenged by variable parasites in nature and this is sufficient to explain why it is optimal to deploy both arms of the innate immune system.  相似文献

完善的先天免疫系统使得昆虫成为分布最广、适应性最强、物种多样性最丰富的动物类群。在长期的进化过程中,昆虫建立了一套安全有效的先天免疫系统,一方面在面对外界微生物攻击的时候及时有效的发生免疫应答反应;另一方面通过免疫抑制来调控适度免疫应答,避免对自身发动攻击和控制环境共生菌刺激引起的免疫应答信号通路的持续激活。泛素-蛋白酶体系统在昆虫先天免疫中具有重要的调控作用,在Toll和IMD信号通路中,通过对免疫应答通路中信号分子的泛素化修饰加工,促进或抑制抗菌肽的表达,从而使免疫反应达到一个平衡。本文通过对泛素-蛋白酶体系统在Toll和IMD信号通路中的免疫应答和免疫抑制方面的研究进行综述,阐明了该系统在昆虫先天免疫中的调控作用,将有助于开展农业害虫与其天敌之间相互关系的深入研究,揭示其免疫调控机理,为开发生物农药,进行生物防控提供理论依据。  相似文献

Coronavirus (CoV) infections are commonly associated with respiratory and enteric disease in humans and animals. In 2012, a new human disease called Middle East respiratory syndrome (MERS) emerged in the Middle East. MERS was caused by a virus that was originally called human coronavirus-Erasmus Medical Center/2012 but was later renamed as Middle East respiratory syndrome coronavirus (MERS-CoV). MERS-CoV causes high fever, cough, acute respiratory tract infection, and multiorgan dysfunction that may eventually lead to the death of the infected individuals. The exact origin of MERS-CoV remains unknown, but the transmission pattern and evidence from virological studies suggest that dromedary camels are the major reservoir host, from which human infections may sporadically occur through the zoonotic transmission. Human to human transmission also occurs in healthcare facilities and communities. Recent studies on Middle Eastern respiratory continue to highlight the need for further understanding the virus-host interactions that govern disease severity and infection outcome. In this review, we have highlighted the major mechanisms of immune evasion strategies of MERS-CoV. We have demonstrated that M, 4a, 4b proteins and Plppro of MERS-CoV inhibit the type I interferon (IFN) and nuclear factor-κB signaling pathways and therefore facilitate innate immune evasion. In addition, nonstructural protein 4a (NSP4a), NSP4b, and NSP15 inhibit double-stranded RNA sensors. Therefore, the mentioned proteins limit early induction of IFN and cause rapid apoptosis of macrophages. MERS-CoV strongly inhibits the activation of T cells with downregulation of antigen presentation. In addition, uncontrolled secretion of interferon ɣ-induced protein 10 and monocyte chemoattractant protein-1 can suppress proliferation of human myeloid progenitor cells.  相似文献

汉滩病毒84Fli株DNA疫苗诱导小鼠免疫应答的初步研究   总被引:1,自引:0,他引:1  
为了加强我国病毒性出血热的防治,本研究将汉滩病毒84Fli株核蛋白S和糖蛋白M编码片段分别克隆至pcDNA3.0载体,构建了pcDNA3/84S和pcDNA3/84M重组质粒,等量混合采用肌肉注射途径免疫C57BL/6小鼠,免疫3次,每次间隔2周,同时与双价出血热病毒灭活疫苗进行对比。ELISA及免疫荧光(IFA)分别检测小鼠血清中汉滩病毒核蛋白及糖蛋白特异性抗体,流式细胞仪和ELISPOT方法分析小鼠免疫后的细胞免疫水平。微量中和试验检测小鼠血清抗体的的中和活性。结果显示,DNA疫苗免疫组C57BL/6小鼠在初次免疫2周后即能检测到汉滩病毒核蛋白与糖蛋白的特异性抗体,与灭活疫苗组相比,重组质粒诱导的抗体滴度高,产生时间早,产生的抗体具有中和活性;同时可诱导产生特异性细胞免疫应答。研究表明,汉滩病毒pcDNA3/84S和pcDNA3/84M重组质粒能有效刺激小鼠产生特异性体液免疫和细胞免疫应答。  相似文献

目的:探讨分析前列腺癌患者^89Sr治疗后机体免疫功能的变化及其临床意义。方法:87例前列腺癌患者经^89Srcl2治疗1月,采用流式细胞术检测治疗前后其外周血淋巴细胞亚群,速率散射免疫比浊分析法检测其免疫球蛋白的含量。结果:经^89Sr治疗后,患者CD3、CD19、CD4比例均明显升高,CD8比例明显降低,差异均有统计学意义畔0.05),而CD16^+CD56^+无明显变化;IgG和IgA明显升高,差异均有统计学意义(P〈0.05),而IgM无明显变化。结论:用^89Srcl2治疗前列腺癌可以有效提高患者的免疫水平,可通过监测机体免疫功能指标的变化^89Sr核素的疗效。  相似文献

The tumor immune microenvironment (TIME) is the cellular environment in which tumors exist. This includes: surrounding blood vessels, immune cells, fibroblasts, bone marrow-derived inflammatory cells, lymphocytes, signaling molecules, immune checkpoint proteins and the extracellular matrix (ECM). The TIME plays a critical role in cancer progression and regulation. Tumors can influence the microenvironment by releasing extracellular signals, promoting tumor angiogenesis and inducing peripheral immune tolerance, while the immune cells in the microenvironment can affect the growth and evolution of cancerous cells. The molecules and cells in the TIME influence disease outcome by altering the balance of suppressive versus cytotoxic responses in the vicinity of the tumor. Having a better understanding of the tumor immune microenvironment will pave the way for identifying new targets for immunotherapies that promote cancer elimination.  相似文献
