Over the past 10 years, Ghusel VDC, Lalitpur District has moved from primarily subsistence agriculture into the wider cash economy aided by the Small Farmers' Development Program (SFDP), which provides credit to farmers mainly for the purchase of buffalo for milk production, and by the National Dairy Corporation, which supports local dairy cooperatives. Analysis reveals that buffalo-keeping and milk sales are increasing the well-being of many households, while at the same time creating new inequalities in gender roles and responsibilities, greater inequities between Brahmin and Tamang residents in Ghusel, and placing pressures on the ecosystem for increased supplies of fodder and fuelwood. Evidence suggests that there is critical, need for attention to the social, and particularly gender-based, implications of maintaining livestock for milk sales and to the ecological underpinnings of this livelihood system.  相似文献

Structure and function of S-adenosylhomocysteine hydrolase   总被引:6,自引:0,他引:6  
In mammals, S-adenosylhomocysteine hydrolase (AdoHcyase) is the only known enzyme to catalyze the breakdown of S-adenosylhomocysteine (AdoHcy) to homocysteine and adenosine. AdoHcy is the product of all adenosylmethionine (AdoMet)-dependent biological transmethylations. These reactions have a wide range of products, and are common in all facets of biometabolism. As a product inhibitor, elevated levels of AdoHcy suppress AdoMet-dependent transmethylations. Thus, AdoHcyase is a regulator of biological transmethylation in general. The three-dimensional structure of AdoHcyase complexed with reduced nicotinamide adenine dinucleotide phosphate (NADH) and the inhibitor (1′R, 2′S, 3′R)-9-(2′,3′-dihyroxycyclopenten-1-yl)adenine (DHCeA) was solved by a combination of the crystallographic direct methods program, SnB, to determine the selenium atom substructure and by treating the multiwavelength anomalous diffraction data as a special case of multiple isomorphous replacement. The enzyme architecture resembles that observed for NAD-dependent dehydrogenases, with the catalytic domain and the cofactor binding domain each containing a modified Rossmann fold. The two domains form a deep active site cleft containing the cofactor and bound inhibitor molecule. A comparison of the inhibitor complex of the human enzyme and the structure of the rat enzyme, solved without inhibitor, suggests that a 17° rigid body movement of the catalytic domain occurs upon inhibitor/substrate binding.  相似文献

铜、镉对三种豆科植物生长及氮磷钾含量的影响   总被引:3,自引:0,他引:3  
为了解豆科植物在Cu、Cd单一污染土壤中的生长状况及对土壤养分的吸收利用特点,采用盆栽实验研究了Cu~(2+)、Cd~(2+)单一污染下紫花苜蓿、红三叶、沙打旺3种豆科植物的株高、根长、生物量和叶、茎部N、P、K、Cu和Cd含量的变化情况.Cu~(2+)、Cd~(2+)处理浓度分别为:0、400、800、1200 mg·kg~(-1)和0、1、10、20 mg·kg~(-1).结果表明,3种豆科植物对Cu和Cd均有较强的吸收能力,除红三叶叶片中Cu含量外,3种豆科植物根、茎、叶中Cd和Cu的含量均与土壤中重金属添加量呈显著正相关.3种豆科植物在严重Cd~(2+)污染的土壤中均能正常生长.在Cu~(2+)添加量≤1200 mg·kg~(-1)时,红三叶能正常生长,而紫花苜蓿的生长则受到显著抑制作用,沙打旺在Cu~(2+)添加量≥800 mg·kg~(-1)时生长受到抑制.土壤Cu~(2+)添加量≤1200 mg·kg~(-1)时,能促进紫花苜蓿对N、P、K的吸收;Cu~(2+)添加量≤800 mg·kg~(-1)时,对红三叶N、P、K含量没有明显影响;Cu~(2+)添加量≤400 mg·kg~(-1)时,可提高沙打旺中N、P、K含量,但当Cu~(2+)添加量≥800 mg·kg~(-1)时则显著降低.土壤Cd~(2+)添加量≤20 mg·kg~(-1)时,对紫花苜蓿和红三叶茎叶以及沙打旺茎部的N、P、K含量有促进作用,但对沙打旺叶片的N、P、K含量起抑制作用.总体来看,3种豆科植物对Cu~(2+)和Cd~(2+)均有一定的耐性,红三叶对Cu~(2+)的耐性较好,紫花苜蓿对Cd~(2+)的耐性较好.
Abstract:
Aimed to understand the growth status and nutrient uptake of leguminous plants under soil copper-or cadmium contamination, a pot experiment with Medicago sativa, Trifolium pre-tense, and Astragalus adsurgens was conducted, with their plant height, root length, plant bio-mass, and N, P, K, Cu and Cd contents in leaf and stem measured. The application amounts of Cu~(2+) and Cd~(2+) were 0,400, 800, and 1200 mg·kg~(-1), and 0, 1, 10, and 20 mg·kg~(-1), re-spectively. All the test three leguminous plants had strong capability of absorbing Cu and Cd.The Cu and Cd contents in their roots, stems, and leaves, except the Cu content in T. Pratease leaf, were significantly positively correlated with the application amounts of Cu~(2+) and Cd~(2+). Un-der the application of Cd~(2+), all the three leguminous plants grew normally. When the application amount of Cu~(2+) was≤1200 mg·kgM-1, T. Pratense grew normally, while the growth of M. Sativa was significantly inhibited. The growth of A. Adsurgens was inhibited when the application amount of Cu~(2+) was≥800 mg·kg~(-1). An apphcation amount of≤1200 mg·kg~(-1) of Cu~(2+) pro-moted the N, P and K absorption of M. Sativa, but applying≤800 mg·kg~(-1) of Cu~(2+) had little effects on the N, P and K absorption of T. Pretense. The N, P and K contents of A. Adsurgens in-creased when the application amount of Cu~(2+) was ≤400 mg·kg~(-1), but decreased significantly when the Cu~(2+) application amount was≥800 mg·kg~(-1). When the application amount of Cd~(2+)was≤20 mg·kg~(-1) , the N, P and K contents in M. Sativa and T. Pratense stems and leaves and in A. Adsurgens stem increased, while those in A. Adsurgens leaf decreased. As a whole, the three leguminous plants all had certain tolerance to Cu~(2+) and Cd~(2+) stresses, especially T. Prat-ease to Cu~(2+) and M. Sativa to Cd~(2+).  相似文献

Laccase is a promising biocatalyst with many possible applications, including bioremediation, chemical synthesis, biobleaching of paper pulp, biosensing, textile finishing and wine stabilization. The immobilization of enzymes offers several improvements for enzyme applications because the storage and operational stabilities are frequently enhanced. Moreover, the reusability of immobilized enzymes represents a great advantage compared with free enzymes. In this work, we discuss the different methodologies of enzyme immobilization that have been reported for laccases, such as adsorption, entrapment, encapsulation, covalent binding and self-immobilization. The applications of laccase immobilized by the aforementioned methodologies are presented, paying special attention to recent approaches regarding environmental applications and electrobiochemistry.  相似文献

目的探讨血清中谷氨酸、甘氨酸和丙氨酸含量变化与脑出血和脑梗死的关系,为今后临床深入研究提供参考资料。方法用氨基酸分析仪对23例急性脑出血和26例脑梗死病人谷氨酸、甘氨酸和丙氨酸进行测定,并与对照组进行比较。结果病人组的兴奋性氨基酸谷氨酸的含量明显高于对照组(P<0.01),病人组的抑制性氨基酸甘氨酸和丙氨酸的含量与对照组无显著差异(P>0.05)。结论提示兴奋性氨基酸谷氨酸可能作为损伤因素参与脑出血和脑梗死脑损伤的发生。  相似文献

将Wistar大鼠暴露于3 780 m低氧环境,分别于24 h、2 wk及3 wk后采用酶联免疫法和硝酸还原酶法测定血液中的ET~(-1)和NO的含量,计算NO/ET~(-1)值,并与高原鼠兔比较,探讨低氧条件下大鼠与高原鼠兔血液中NO与ET~(-1)含量的变化趋势。结果表明,低氧24 h后,大鼠血液中NO和ET~(-1)的含量显著高于同海拔的高原鼠兔(P<0·01),而NO/ET~(-1)值无显著差异(P>0·05)。随着大鼠在高海拔停留时间的延长,血液中NO含量呈减少趋势,而ET~(-1)则有上升趋势,二者呈显著的负相关(r2=0·2416,P<0·01)。高原鼠兔NO/ET~(-1)值约为大鼠低氧2 wk和3 wk的2倍(P<0·01)。说明不同低氧暴露时间,高原鼠兔和大鼠的NO、ET~(-1)及NO/ET~(-1)值有显著差异,提示NO/ET~(-1)值可以作为有机体是否适应高原低氧环境的一个指标。  相似文献

Circular RNAs (circRNAs) are a novel class of endogenous noncoding RNAs that form covalently closed continuous loops without 3′ end poly (A) tails and 5′ end caps. circRNAs are more conservative and stable than linear RNA. circRNAs can specifically bind to microRNAs as competing endogenous RNA, thereby directly or indirectly regulating the expression of related genes. circRNAs have been implicated in several cancers including gastrointestinal (GI) cancers. Some circRNAs have the potential to become biological biomarkers and therapeutic targets of GI cancers. Extracellular vesicles are known as actual intermediaries of intercellular communications, such as biological signals and cargo transfer between different cells. A variety of cells release the exosomes as nanovesicular bodies. Exosomes contain different compounds such as several types of nucleic acids and proteins. In this study, we focused on exosomes in colorectal cancer as good tools that can be involved in various cancer-related processes. Furthermore, we summarize the advantages and disadvantages of exosome extraction methods and review related studies on the role of exosomes in colorectal cancer. Finally, we focus on reports available on relations between mesenchymal stem cell–derived exosomes and colorectal cancer. Several cancer-related processes such as cancer progression, metastasis, and drug resistance of colorectal cancer are related to the cargoes of exosomes. A variety of molecules, especially proteins, microRNAs, and long noncoding RNAs, play important roles in these processes. The microenvironment features, such as hypoxia, also have very important effects on the properties of the origin cell–derived exosomes. On the other hand, exosomes derived from colorectal cancer cells also interfere with cancer chemoresistance. Furthermore, today it is known that exosomes and their contents can likely be very effective in noninvasive colorectal cancer diagnosis and therapy. Thus, exosomes, and especially their cargoes, play different key roles in various aspects of basic and clinical research related to both progression and therapy of colorectal cancer.  相似文献

目的:研究眶额叶(OFC)的谷氨酸(Glu)和γ-氨基丁酸(GABA)含量变化对胃运动的影响及其调节神经机制。方法:实验采用了大鼠眶额叶微量注射给药,结合核团损毁的方法,以记录胃内压,统计胃收缩幅度作为胃运动变化的指标。结果:①OFC注射Glu可显著降低胃收缩幅度,损毁杏仁核后可反转该效应,胃收缩幅度显著增强;损毁蓝斑核后,Glu的作用无显著性变化。②OFC注射GABA可显著增强胃的收缩幅度,损毁蓝斑核后消除该效应;损毁杏仁核后,胃收缩幅度进一步增强。结论:外源性增加OFC区Glu含量导致的抑胃效应可能是通过增强了杏仁核的经常性抑胃作用而引起的;而增加OFC区GABA的含量引起的胃运动增强与蓝斑核密切相关。  相似文献

Lignin is an integral constituent of the primary cell walls of the dark-grown maize (Zea mays L.) coleoptile, a juvenile organ that is still in the developmental state of rapid cell extension. Coleoptile lignin was characterized by (i) conversion to lignothiolglycolate derivative, (ii) isolation of polymeric fragments after alkaline hydrolysis, (iii) reactivity to antibodies against dehydrogenative polymers prepared from monolignols, and (iv) identification of thioacidolysis products typical of lignins. Substantial amounts of lignin could be solubilized from the coleoptile cell walls by mild alkali treatments. Thioacidolysis analyses of cell walls from coleoptiles and various mesocotyl tissues demonstrated the presence of guaiacyl-, syringyl- and (traces of)p-hydroxyphenyl units besidesp-coumaric and ferulic acids. There are tissue-specific differences in amount and composition of lignins from different parts of the maize seedling. Electron-microscopic immunogold labeling of epitopes recognized by a specific anti-guaiacyl/syringyl antibody demonstrated the presence of lignin in all cell walls of the 4-d-old coleoptile. The primary walls of parenchyma and epidermis were more weakly labeled than the secondary wall thickenings of tracheary elements. No label was found in middle lamellae and cell corners. Lignin epitopes appeared first in the tracheary elements on day 2 and in the parenchyma on day 3 after sowing. Incubation of coleoptile segments in H2O2 increased the amount of extractable lignin and the abundance of lignin epitopes in the parenchyma cell walls. Lignin deposition was temporally and spatially correlated with the appearance of epitopes for prolinerich proteins, but not for hydroxyproline-rich proteins, in the cell walls. The lignin content of coleoptiles was increased by irradiating the seedlings with white or farred light, correlated with the inhibition of elongation growth, while growth promotion by auxin had no effect. It is concluded that wall stiffness, and thus extension growth, of the coleoptile can be controlled by lignification of the primary cell walls. Purpose: Pituitary adenylate cyclase-activating polypeptide (PACAP), a member of the secretin/glucagons/vasoactive intestinal peptide family, induces the expression of catecholamine-synthesizing enzymes in adrenal medullary cells. In addition, PACAP and its receptor have been detected in human pheochromocytoma tissues, though it is not yet known whether PACAP enhances the expression of genes encoding catecholamine-synthesizing enzymes. To address this question, we analyzed PACAP, PACAP receptor, and tyrosine hydroxylase (TH) and phenylethanolamine-N-methyltransferase (PNMT) mRNAs in pheochromocytomas. Methods: The levels of the mRNA for PACAP and vasoactive intestinal peptide (VIP), and their receptors, and for TH and PNMT were measured by RT-PCR or real-time PCR analysis, and the concentrations of catecholamines were measured by HPLC in 24 intra-adrenal and six extra-adrenal pheochromocytomas. Results: mRNA expression of PACAP and its receptor VPAC1R were detected in many pheochromocytomas (24/30 and 29/30, respectively), but mRNA expression of the PAC1R and VPAC2R receptor subtypes were detected in only one of six extra-adrenal pheochromocytomas. PACAP mRNA expression correlated with TH (p=0.0018) and PNMT (p=0.05) mRNA expression, as well as epinephrine (p=0.0342) levels in 16 intra-adrenal pheochromocytomas. Conclusion: Our findings support a possible role for PACAP in the regulation of expression of genes encoding catecholamine-synthesizing enzymes in intra-adrenal pheochromocytomas.  相似文献

The objective of this study was to assess the effects of soil pH adjustment and treatment with sludge on plant growth and cadmium (Cd) accumulation in Chinese cabbage (pak choi) grown in Cd-contaminated soils. Soil was artificially spiked to raise its final Cd concentration to 10 mg/kg, adjusted to different pH values, and then supplemented with 5% sludge: either biosolids (BS) from wastewater treatment or sludge from drinking water treatment (WTS). Pak choi seeds were sown and the seedlings potted in the treated soil, then grown in a greenhouse for 45 days. The added Cd was present mainly in the exchangeable fraction and its concentration correlated to Cd accumulation in the roots and shoots. Adjustment of pH and application of sludge affected the Cd concentration in different fractions. Addition of WTS and adjustment to pH 7.0 inhibited growth and Cd accumulation in the pak choi. Addition of BS increased the soil nutrient content and thus enhanced growth and Cd accumulation. If pak choi were the only form of vegetable intake, consumption of pak choi grown in the Cd-spiked soils used in this study would exceed the maximum acceptable intake (60 μg/day) proposed by the World Health Organization.  相似文献

Numerous methods have been developed for immunogold labeling of thick, cryo-preserved biological specimens. However, most of the methods are permutations of chemical fixation and sample sectioning, which select and isolate the immunolabeled region of interest. We describe a method for combining immunogold labeling with cryo-electron microscopy (cryo-EM) and cryo-electron tomography (cryo-ET) of the surface proteins of intact mammalian cells or the surface glycoproteins of assembling and budding viruses in the context of virus-infected mammalian cells cultured on EM grids. In this method, the cells were maintained in culture media at physiologically relevant temperatures while sequentially incubated with the primary and secondary antibodies. Subsequently, the immunogold-labeled specimens were vitrified and observed under cryo-conditions in the transmission electron microscope. Cryo-EM and cryo-ET examination of the immunogold-labeled cells revealed the association of immunogold particles with the target antigens. Additionally, the cellular structure was unaltered by pre-immunolabeling chemical fixation and retained well-preserved plasma membranes, cytoskeletal elements, and macromolecular complexes. We think this technique will be of interest to cell biologists for cryo-EM and conventional studies of native cells and pathogen-infected cells.  相似文献

Mycobacterium tuberculosis (M.tb.), the causative agent of tuberculosis (TB), cannot synthesize GSH, but synthesizes two major low molecular weight thiols namely mycothiol (MSH) and ergothioneine (ERG). Gamma-glutamylcysteine (GGC), an intermediate in GSH synthesis, has been implicated in the protection of lactic acid bacteria from oxidative stress in the absence of GSH. In mycobacteria, GGC is an intermediate in ERG biosynthesis, and its formation is catalysed by EgtA (GshA). GGC is subsequently used by EgtB in the formation of hercynine-sulphoxide-GGC. In this study, M.tb. mutants harbouring unmarked, in-frame deletions in each of the fives genes involved in ERG biosynthesis (egtA, egtB, egtC, egtD and egtE) or a marked deletion of the mshA gene (required for MSH biosynthesis) were generated. Liquid chromatography tandem mass spectrometry analyses (LC-MS) revealed that the production of GGC was elevated in the MSH-deficient and the ERG-deficient mutants. The ERG-deficient ΔegtB mutant which accumulated GGC was more resistant to oxidative and nitrosative stress than the ERG-deficient, GGC-deficient ΔegtA mutant. This implicates GGC in the detoxification of reactive oxygen and nitrogen species in M.tb.  相似文献

The question arises as to whether oxidative stress has a primary role in neurodegeneration or is a secondary end-stage epiphenomenon. The aim of the present study was to determine oxidative stress parameters like malondialdehyde (MDA), carbonyl proteins (CP) and Albumin-disulphide (Alb-SSR) and relate these parameters to the immune parameter neopterin, folic acid and vitamin B12 as vitamins and homocysteine in patients with neuro-degenerative diseases (NDD), namely mild cognitive impairment (MCI) and Alzheimer's disease (AD) compared to an aged matched control group. MDA, CP and Alb-SSR were significantly increased in the NDD group compared to controls, but not vitamin B12, folic acid and neopterin. Significant correlations were found between CP and Alb-SSR, CP and MDA and between MDA and Alb-SSR including patients with NDD and the control group. These results support the hypothesis that oxidative damage to lipids and proteins is an important early event in the pathogenesis of neurodegenerative diseases.  相似文献

The variations in the coordination environment of Co(II), Cu(II) and Zn(II) complexes with the neutral, tridentate ligand bis[1-(cyclohexylimino)ethyl]pyridine (BCIP) are reported. Analogous syntheses were carried out utilizing either the M(BF4)2 · xH2O or MCl2 · xH2O metal salts (where M = Co(II), Cu(II) or Zn(II)) with one equivalent of BCIP. When the hydrated metal starting material was used, cationic, octahedral complexes of the type [M(BCIP)2]2+ were isolated as the tetrafluoroborate salt (4, 5). Conversely, when the hydrated chloride metal salt was used as the starting material, only neutral, pentacoordinate [M(BCIP)Cl2] complexes (1-3) formed. All complexes were characterized by X-ray diffraction studies. The three complexes that are five coordinate have distortions due mainly to the pyridine di-imine bite angle. The [Cu(BCIP)Cl2] (2) also exhibits deviations in the Cu(II)-Cl bond distances with values of 2.4242(9) and 2.2505(9) Å, which are not seen in the analogous Zn(II) and Co(II) structures. Similarly, the two six coordinate complexes (5, 6) are also altered by the ligand frame bite angle giving rise to distorted octahedral geometries in each complex. The [Cu(BCIP)2](BF4)2 (6) also exhibits Cu(II)-Nimine bond lengths that are on average 0.14 Å longer than those found in the analogous 5 coordinate complex, [Cu(BCIP)Cl2]. In addition to X-ray analysis, all complexes were also characterized by UV/Vis and IR spectroscopy with 1H NMR spectroscopy being used for the analysis of the Zn(II) analogue (3).  相似文献

Repetitive sequences are a major constituent of many eukaryote genomes and play roles in gene regulation, chromosome inheritance, nuclear architecture, and genome stability. The identification of repetitive elements has traditionally relied on in-depth, manual curation and computational determination of close relatives based on DNA identity. However, the rapid divergence of repetitive sequence has made identification of repeats by DNA identity difficult even in closely related species. Hence, the presence of unidentified repeats in genome sequences affects the quality of gene annotations and annotation-dependent analyses (e.g. microarray analyses). We have developed an enhanced repeat identification pipeline using two approaches. First, the de novo repeat finding program PILER-DF was used to identify interspersed repetitive elements in several recently finished Dipteran genomes. Repeats were classified, when possible, according to their similarity to known elements described in Repbase and GenBank, and also screened against annotated genes as one means of eliminating false positives. Second, we used a new program called RepeatRunner, which integrates results from both RepeatMasker nucleotide searches and protein searches using BLASTX. Using RepeatRunner with PILER-DF predictions, we masked repeats in thirteen Dipteran genomes and conclude that combining PILER-DF and RepeatRunner greatly enhances repeat identification in both well-characterized and un-annotated genomes.  相似文献

Trivalent chromium (Cr3+) is essential for animal and human health, whereas hexavalent Cr (CrO4 2−) is a potent carcinogen and extremely toxic to animals and humans. Thus, the accumulated Cr in food plants may represent potential health hazards to animals and humans if the element is accumulated in the hexavalent form or in high concentrations. This study was conducted to determine the extent to which various vegetable crops absorb and accumulate Cr3+ and CrO4 2− into roots and shoots and to ascertain the different chemical forms of Cr in these tissues. Two greenhouse hydroponic experiments were performed using a recirculating-nutrient culture technique that allowed all plants to be equally supplied with Cr at all times. In the first experiment, 1 mg L−1 Cr was supplied to 11 vegetable plant species as Cr3+ or CrO4 2−, and the accumulation of Cr in roots and shoots was compared. The crops tested included cabbage (Brassica oleracea L. var. capitata L.), cauliflower (Brassica oleracea L. var. botrytis L.), celery (Apium graveolens L. var. dulce (Mill.) Pers.), chive (Allium schoenoprasum L.), collard (Brassica oleracea L. var. acephala DC.), garden pea (Pisum sativum L.), kale (Brassica oleracea L. var. acephala DC.), lettuce (Lactuca sativa L.), onion (Allium cepa L.), spinach (Spinacia oleracea L.), and strawberry (Fragaria ×  ananassaDuch.). In the second experiment, X-ray absorption spectroscopy (XAS) analysis on Cr in plant tissues was performed in roots and shoots of various vegetable plants treated with CrO4 2− at either 2 mg Cr L−1 for 7 d or 10 mg Cr L−1 for 2, 4 or 7 d. The crops used in this experiment included beet (Beta vulgaris L. var. crassa (Alef.) J. Helm), broccoli (Brassica oleracea L. var. Italica Plenck), cantaloupe (Cucumis melo L. gp. Cantalupensis), cucumber (Cucumis sativus L.), lettuce, radish (Raphanus sativus L.), spinach, tomato (Lycopersicon lycopersicum (L.) Karsten), and turnip (Brassica rapa L. var. rapifera Bailey). The XAS speciation analysis indicates that CrO4 2− is converted in the root to Cr3+ by all plants tested. Translocation of both Cr forms from roots to shoots was extremely limited and accumulation of Cr by roots was 100-fold higher than that by shoots, regardless of the Cr species supplied. Highest Cr concentrations were detected in members of the Brassicaceae family such as cauliflower, kale, and cabbage. Based on our observations and previous findings by other researchers, a hypothesis for the differential accumulation and identical translocation patterns of the two Cr ions is proposed.
