The juvenoid hydroprene or altozar (ZR-0512), when applied to larvae and pupae of Corcyra cephalonica at doses of 100 μg and 10 μg per individual, produced abnormalities in the growth and differentiation of imaginal testicular structures. In all the resultant larval-imaginal intermediates each testis contained a compact mass of cysts at various developmental stages with a comparatively higher number of spermatid cysts denoting a potentiality for increased fertility. Delayed differentiation of spermatozoa may also be inferred from this finding. On rare occasions the two larval testes failed to become enclosed in a single sac. Conversely, in the adultoids obtained from pupal treatment, the production of a normal quantity of testicular cysts of different kinds was inhibited. Moreover, malformed and sometimes degenerating spermatozoan cysts were formed. In a few extreme cases differentiation of even the spermatid cysts had been almost entirely prevented. Spermatogonia and the degenerating cell masses were the chief components of the testis of such adultoids. All observations indicated a sort of male sterility induced by the juvenoid when applied to pupae, especially at 12 hours age. The effect of the juvenoid on spermatogenesis could not be shown to be direct.  相似文献

This study aimed to evaluate the efficiency of immunocastration in lambs using testicular morphometry. Thirty lambs were randomly divided into two treatments (subcutaneous administration of 1.0 mL and 0.5 mL of an anti-GnRH vaccine) and a control group (1.0 mL saline solution). The animals were vaccinated at four months of age, received a second dose 30 days later, and were slaughtered 90 days after the first vaccine dose. After slaughter, testicles were collected, and samples were removed for histological processing and evaluation of testicular morphometric parameters. Analysis of variance, Tukey’s test, and Kruskal–Wallis test were performed, with a 5% level of significance. There was a reduction in testicular weight, gonadosomatic index, seminiferous tubule diameter, germinal epithelium height, leydigosomatic index, and total tubule length. The total length per testicular gram increased in the immunocastrated group. Intrinsic spermatogenesis yield, Sertoli cell indices, and estimates of sperm and Sertoli cell production were reduced in the immunized groups (P < 0.05). The anti-GnRH vaccine in lambs at doses of 1.0 mL and 0.5 mL is sufficient to promote immunocastration, verified through severe changes in testicular morphometry from animals.  相似文献

An enhanced chemiluminescence immunoassay for the determination of serum human chorionic gonadotropin (HCG) in specimens from oncology patients has been assessed with respect to its cross-reactivity with the free HCG ββ-subunit (HCG-β). The assay, standardized against the First International Reference Preparation 75/537, had a crossreactivity with the free β-subunit of 625% (molar basis). Therefore this assay achieves high sensitivity for the detection of either intact HCG or free HCG-β in serum of patients with seminomatous or nonseminomatous testicular cancers. Results of both assays, the in-house immunoradiometric assay (+ HCG-β) and the Amerlite HCG-60 assay, showed a close correlation (R =0.854?0.960) when serum samples from tumour patients were analyzed. Moreover, the content of free β-subunit determined in a specific HCG-β assay, could be quantitatively measured in the enhanced chemiluminescence immunoassay. Thus, this assay is suitable for oncology use, but also highlights the limitations of measuring HCG in serum samples.  相似文献

Active immunization of ram lambs and bull calves against highly purified LH results in the production of antibodies which bind radiolabeled ovine LH. Antibody titers were not dependent upon the dose (0.1 or 1.0 mg) of antigen given but were found to be greater when the antigen consisted of LH conjugated to a carrier protein; in this case human serum globulin (hSG). The presence of LH antibodies in ram sera seemed of little consequence. The presence of antibodies in sera of immunized bulls, however, appeared to neutralize endogenous LH sufficiently to suppress serum testosterone and reduce testicular size. Androgen deficiency, produced by immunization against oLH-hSG, resulted in atrophy of the testes, seminal vesicles and epithelial cells lining both epididymides and seminal vesicles. Reduced weight gain and lack of development of the secondary sexual characteristics were at levels expected of surgical castrates (steers). In summary, active immunization of young beef bulls against LH and in particular, oLH-hSG, results in a castration-like response which has been compared with that achieved in other species by luteinizing hormone releasing hormone (LHRH) immunocastration. The success of LH immunocastration in field trials with young bulls and investigations aimed toward reversibility are yet to be determined.  相似文献

啮齿目动物的睾丸较肝脏对镉毒性更为敏感.为阐明不同组织细胞的镉毒性分子机制,对镉暴露大鼠睾丸支持细胞与肝脏金属硫蛋白基因(MT)的表达及镉蓄积进行了时相研究.成年雄性SD大鼠低剂量镉(4.0 μmol/kg)皮下注射后,立即进行睾丸支持细胞和肝脏组织的分离.采用RT-PCR技术分析mRNA,并用光密度扫描作半定量分析,蛋白质定量用ELISA方法,原子分光光度吸收法测定镉浓度.结果显示:镉暴露1 h后肝脏MT mRNA即有明显的诱导表达,3 h达高峰;支持细胞也有明显的诱导表达, 6 h达高峰.镉暴露后肝脏MT有明显的诱导表达,但睾丸支持细胞不但未见MT增加而且还稍有下降.提示:a.镉对MTmRNA的诱导表达具有时间依赖性和组织特异性.b.镉虽然能诱导睾丸MT的转录,但没有促进其MT的合成,这可能是睾丸对镉毒性与致癌作用较肝脏更敏感的重要原因.  相似文献

Bisphenol A (BPA) is an omnipresent environmental pollutant. Despite being restrictions in-force for its utilization, it is widely being used in the production of polycarbonate plastics and epoxy resins. Direct, low-dose, and long-term exposure to BPA is expected when they are used in the packaging of food products and are used as containers for food consumption. Occupationally, workers are typically exposed to BPA at higher levels and for longer periods during the manufacturing process. BPA is a known endocrine disruptor chemical (EDC), that causes male infertility, which has a negative impact on human life from emotional, physical, and societal standpoints. To minimize the use of BPA in numerous consumer products, efforts and regulations are being made. Despite legislative limits in numerous nations, BPA is still found in consumer products. This paper examines BPA's overall male reproductive toxicity, including its impact on the hypothalamic-pituitary-testicular (HPT) axis, hormonal homeostasis, testicular steroidogenesis, sperm parameters, reproductive organs, and antioxidant defense system. Furthermore, this paper highlighted the role of non-monotonic dose–response (NMDR) in BPA exposure, which will help to improve the overall understanding of the harmful effects of BPA on the male reproductive system.  相似文献

探讨氯化镧对小鼠精子质量及睾丸细胞酶的影响。40只成年昆明种雄性小鼠随机分成对照组、低(25mg·kg-1)、中(50mg·kg-1)、高(100mg·kg-1)剂量组,腹腔注射1次/4d,饲养35d。测定睾丸和附睾脏器指数,检测并计算精子数量、活精率、活动率和畸形率以及睾丸碱性磷酸酶(AKP)、酸性磷酸酶(ACP)、乳酸脱氢酶(LDH)、一氧化氮合酶(NOS)活力。结果显示,高剂量氯化镧降低了小鼠睾丸AKP活力,抑制了精子数量和质量;中剂量氯化镧能促进NOS活力,使精子数量减少,对精子质量造成损伤。  相似文献

Abstract

In this study, a new affinity gel for the purification of bovine testicular hyaluronidase (BTH) was synthesized. l-Tyrosine was added as the extension arm to the Sepharose-4B activated with cyanogen bromide. m-Anisidine is a specific inhibitor of BTH enzyme. m-Anisidine was clamped to the newly formed Sepharose-4B-l-tyrosine as a ligand. As a result, an affinity gel having the chemical structure of Sepharose-4B-l-tyrosine-m-anisidine was obtained. BTH purified by ammonium sulfate precipitation and affinity chromatography was obtained with a 16.95% yield and 881.78 degree of purity. The kinetic constants KM and VMax for BTH were determined by using hyaluronic acid as a substrate. KM and VMax values obtained from the Lineweaver–Burk graph were found to be 2.23?mM and 19.85?U/mL, respectively. In vitro effects of some chemicals were determined on purified BTH enzyme. Some chemically active ingredients were 1,1-dimethyl piperidinium chloride, β-naphthoxyacetic acid and gibberellic acid. Gibberellic acid showed the best inhibition effect on BTH.  相似文献

Cotesia congregata is a gregarious larval endoparasitoid of the tobacco hornworm, Manduca sexta. Parasitized larvae exhibit a variety of physiological and developmental aberrations, the most obvious of which is the induction of developmental arrest characterized by the absence of wandering behavior and suppression of pupation. This arrest appears attributable to continued maintenance of an elevated titer of juvenile hormone and reduced levels of hemolymph juvenile hormone esterase activity. Injection of the wasp's polydnavirus into nonparasitized larvae also causes arrest and the larvae eventually form larval-pupal intermediates instead of normal pupae, indicating the virus may be partially responsible. Aside from causing arrested host development, parasitism also inhibits the normal development and differentiation of testes in male host larvae, so that the testes atrophy instead of growing synchronously with other larval tissues. Here we report that parasitism has pronounced disruptive cytological effects on the developing reproductive organs of male hosts, in addition to causing them to atrophy. Parasitism results in a reduction in testicular volume attributable to a reduction in the number of developing germ cells. Microscopy revealed that the structural integrity of the sheaths surrounding the testicular follicles also is disrupted, so that the tissues appear grossly abnormal compared to those of nonparasitized larvae. Intrahemocoelic injection of purified C. congregata polydnavirus in combination with venom into nonparasitized fourth instar larvae, or topical application of 100 μg of methoprene to fourth instar larvae, also alters sheath integrity and reduces the numbers of developing germ cells, but not to the same degree as the pattern observed in truly parasitized hosts. The objective of this study was to evaluate the testicular biometric, seminal, and plasma testosterone levels in lambs subjected to an anti-GnRH vaccine as a method of castration. Thirty entire, crossbred Santa Inês male lambs were randomly distributed into three treatment (T): T1 was the control group, with the administration of 1 mL of saline solution subcutaneously (SC); 1.0 and 0.5 mL of an anti-GnRH vaccine were administered SC in T2 and T3, respectively. Testicular biometric variables, physical and morphological variables of semen, and plasma testosterone concentrations were evaluated. At D60, there was a reduction in testicular length, width, thickness, and scrotal circumference of the immunocastrated animals regardless of the vaccine dose used (P < 0.05). A reduction in semen physical variables at both dosages (P < 0.05) was observed, with azoospermia, in 80% and 70% of animals in the T2 and T3 groups, respectively. At D60, the immunocastrated animals also showed an increase in spermatozoa defects (P < 0.05), whereas plasma testosterone concentration decreased (P < 0.05). Immunocastration of lambs using the Bopriva vaccine at doses of 1.0 and 0.5 mL is efficient in inducing azoospermia in up to 80% of animals, although two doses in a 30-day interval are necessary for it to be an effective and safe method. Efficacy was demonstrated through a reduction in serum testosterone levels, testicular biometry, and seminal fluid analysis. Considering the efficacy of both doses in this study, we recommend using the lower dose (0.5 mL), which will allow for a 50% reduction in vaccine costs.  相似文献

The elasmobranch testis consists of spherical spermatocysts, each housing a single germ cell stage and its own clone of Sertoli cells. Because of the simple diametrical arrangement of cysts in maturational order, the testes of Squalus acanthias, Scyliorhinus canicula, and Prionace glauca are classified as the diametric shark testis type. The aim of this study was to document histologically the spermatocyst composition in the blue shark stage‐by‐stage and to establish whether the diametric testis type confers any uniformity regarding the expression of spermatogenesis in all sharks with this testis type. Analysis of the testes of blue sharks breeding in summer revealed extensive cyst degeneration of various forms and degrees, cyst shrinkage, and cyst disorganization with or without evidence of cell death, initially at the spermatogonia—spermatocyte transition but predominantly in spermatocyte and spermatid cysts. Animals could be grouped into two categories based on the major degenerative phenomena observed, namely those with extensive multinucleate cell (MNC) formation, and those with pronounced vacuolation in cysts. A major finding was the significant (P < 0.001) predominance of MNC formation and vacuolation in late‐stage spermatogonial cysts in the respective categories of sharks. Spermatocyte cysts showed varying degrees of germ cell depletion, with or without evidence of degeneration. Normal‐looking, but clearly subnormal‐sized primary and secondary spermatocyte cysts with no evidence of degeneration were significantly the dominant spermatocyte cyst types in both categories. It is proposed that these subnormal‐sized spermatocyte cysts could proceed into spermiogenesis. Because neighboring spermatid cysts lacked ordered bundling of spermatid heads (disorganized), a morphology significantly correlated with the vacuolation category of sharks, these results suggest that further progression into spermiogenesis was halted in such cysts. The sex-linked recessive gene Tfm in the mouse produces a condition of testicular feminization (androgen insensitivity syndrome, AIS) in hemizygotes, comparable to the condition of the same name in humans. The murine mutant was originally believed to have no derivatives of the mesonephric duct system (MDS), and this absence was ascribed to dependence of these derivatives on androgens for survival. However, microscopical epi-didymides, retia testes, and vasa deferentia were identified in these animals in our laboratory. These micro-organs may play a role in meiosis induction in Tfm/Y animals. The present study was designed to determine whether survival of these organs is due to retention of an ability to respond to androgens, or whether they are unique amongst MDS derivatives in being independent of androgens. Previous studies in our laboratory demonstrated that the enzyme β-glucuronidase (βG) is androgen sensitive in the epididymis of the normal mouse. In the present investigation we used this enzyme as a marker to study androgen sensitivity in the microscopical epididymides of Tfm/Y hemizygotes and in the epididymides of control +/Y litter-mate brothers. Both mutant and control animals were studied with and without exogenous androgen stimulation. Tfm/Y hemizygotes demonstrated low levels of diffuse, cytoplasmic βG activity that appears to be unresponsive to exogenous androgen stimulation. In light of our previous studies, this distribution of βG reaction products suggests some degree of androgen sensitivity. The survival of these micro-organs and their partial androgen sensitivity may be related to the role of the MDS in inducing meiosis.  相似文献

Four pairs of captive crowned lemurs (Lemur coronatus) were studied during their annual breeding season in order to obtain baseline data on their reproductive biology for comparison with other Lemur species and to enhance their captive breeding success. Vaginal smears, testicular measurements, and records of the Duke University Primate Center provided the presented data. During a single breeding season, females cycled an average of three times, with an average cycle length of 34 days. Cycles were detected between November and March. Vaginal estrus and copulations were limited to one day per cycle. After 125 days of gestation females gave birth to one or two young. Both sexes attained sexual maturity at an age of about 20 months. Mean male testis size peaked in late December; at the same time, three of the females experienced their first estrus. Based on all available data, there was a significant positive correlation between cycle length and gestation length in the genus Lemur.  相似文献

In the species Pseudoplatystoma fasciatum the males are smaller than females. Thus, the objective of this study was to describe the process of testicular differentiation and provide tools for sex manipulation techniques aimed at improving the productivity of farming of sexually‐reversed females. Correlation between the length, age, sex rate and the stage of gonadal development were analyzed in this species from samples collected between 0 and 240 days post‐hatching. Testicular differentiation was divided into eight stages, based on the cellular and morphological characteristics of the gonad. The results showed that sexual differentiation has a greater correlation to the size than the age in this species. This study provides knowledge on the proliferation and distinctive arrangement of somatic cells, which enabled the early identification of the testis due to the presence of future fringes in the specimens. Testicular differentiation in P. fasciatum was histologically different from other species due to the proliferation and distribution of somatic cells in the regions that would originate the testicular tubules and sperm ducts. Meiosis began at a later stage in comparison with other species, therefore, it was not considered a criterion for early sexual characterization. It can be concluded that testicular differentiation in P. fasciatum occurs relatively early and this data can be used to improve sex inversion protocols and increase productivity in this species.  相似文献

Modeling the structure of the C-domain of bovine angiotensin-converting enzyme revealed two putative chloride-binding sites. The kinetic parameters, K(m) and k(cat), of hydrolysis of the substrate Cbz-Phe-His-Leu catalyzed by the testicular (C-domain) enzyme were determined over a wide range of chloride concentrations. Chloride anions were found to be enzyme activators at relatively low concentrations, but they inhibit enzymatic activity at high concentrations. A general scheme for the effect of chloride anions on activity of the C-domain of bovine angiotensin-converting enzyme accounting for binding the "activating" and "inhibiting" anions is suggested.  相似文献

Fertility preservation has been included in the management of childhood cancer treatment. Cryopreservation of immature testicular tissue is the only available solution for pre-pubertal boys. Different freezing protocols have been developed in several species but without a clearly identified procedure. We tried to evaluate several protocols for cryopreservation of rat immature testicular tissue. Twelve different freezing protocols using different (i) cryoprotectant (dimethylsulphoxide [DMSO] or 1,2-propanediol [PROH]), (ii) cryoprotectant concentration (1.5M or 3M), (iii) equilibration time (30 or 60 min), (iv) equilibration temperature (4 °C or room temperature), (v) size of testicular fragment (7.5mg or 15mg), (vi) package (straws or cryovials), were compared using cord morphological damage evaluation. A testicular tissue piece of 7.5mg cryopreserved in cryovial using 1.5M DMSO, an equilibration time of 30 min at 4 °C showed fewer morphological alterations than the other protocols tested. The selected freezing protocol was able to maintain rat immature testicular tissue architecture, functionality after testicular pieces organotypic culture, and could be proposed in a human application.  相似文献

The testicular efferent duct system of Leuciscus cephalus (Cyprinidae), is described for three phases of testicular development. Testicular main ducts were analyzed by means of conventional histology and transmission electron microscopy. Additional techniques were applied for lectin histochemistry to determine secretory activity, as well as immunohistochemistry for cell proliferation activity and for muscle actin to demonstrate the distribution and amount of contractile cells. The contribution of the main ducts' epithelia and of degenerating spermatocytes to seminal fluid composition was confirmed, with the former being a source of carbohydrates and the latter that of phospholipids. The apical glycocalyx of epithelial cells, which is important in cell recognition and potentially involved in sperm storage, was marked by RCA I, LCA, and WGA lectin. Higher numbers of proliferating epithelial cells were ascertained during spawning phase compared to pre- and postspawning phases. In the ducts' stroma, a large number of cells expressed muscle actin and tropomyosin, indicating the ducts' contractile potential for the transport of seminal fluid towards release. Adjacent to these contractile cells, numerous nerves were found, indicating neuronal control of sperm fluid flow.  相似文献

为探讨不同转染试剂(LipofectamineTM LTXPLUSTM、Lipofectamine2000和纳米化聚酰胺-胺型树枝状聚合物(PAMAM-D))和睾丸注射方法 (睾丸网注射、曲精细管注射和间质注射)对转基因小鼠生产效率的影响,将pEGFP-C1质粒分别与不同转染试剂混合后,按照不同的注射方法注入小鼠睾丸内,30 d后检测小鼠精子密度、活力、精子阳性率以及配种后仔鼠转基因阳性率。结果 3种转染试剂对小鼠繁殖性能影响由小到大依次为LipofectamineTM LTXPLUSTM、Lipofectamine 2000和PAMAM-D。转染后LipofectamineTM LTXPLUSTM、Lipofectamine 2000和PAMAM-D组精子的GFP阳性率分别为35.65%±0.69%、12.86%±0.35%和10.04%±0.20%。配种后仔鼠的PCR阳性率分别为29.17%、13.70%和5.88%。3种不同注射方法对小鼠睾丸都造成损伤,由小到大依次为睾丸网注射、曲精细管注射和睾丸间质注射,三者的阳性精子比例分别为35.13%±1.727%、15.13%±1.457%和0%,配种后仔鼠的PCR阳性率分别为33.3%、12.5%和0%。结果表明,LipofectamineTM LTXPLUSTM和睾丸网注射对小鼠睾丸的损伤最小,并能获得较高的转染效率。  相似文献
